Abstract:1040 Objectives: Modulation of phosphodiesterase 10A (PDE10A) has shown potential application in the treatment of neurodegenerative and neuropsychiatric disorders such as schizophrenia and Huntington’s disease. A PET tracer for PDE10A would offer non-invasive quantification of its density and distribution in vivo, which could enable target engagement studies for drug discovery. In this study, we aim to identify an aryl 18F-labeled PDE10A PET tracer ([18F]P10A-1910) with facile labeling procedures, high brain uptake, excellent in vivo stability and specificity. Methods: The PDE10A inhibitor P10A-1910 (2-(2-(3-(4-fluorophenyl)-7-methyl-4-oxo-3,4-dihydroquinazolin-2-yl)ethyl)-4-isopropoxyisoindoline-1,3-dione) and its labeling precursor were prepared in house. [18F]P10A-1910 was labeled by Cu-mediated radiofluorination. Lipophilicity was tested by the shake-flask method. The stability and biodistribution studies were evaluated on CD-1 mice. Dynamic PET scans (0-90 min) were performed in the brain of mature cynomolgus. Results: P10A-1910 and its corresponding precursor were synthesized as colorless solids, with overall yields of 11% and 9%, respectively. [18F]P10A-1910 was produced in 28 ± 3% non-decay-corrected radiochemical yields, with high radiochemical purity (>99%) and high molar activity (>1 Ci/μmol). The LogD value was determined as 3.50, which is appropriate for brain permeability. Ex vivo biodistribution revealed high radioactivity levels in small intestine and liver (>50 %ID/g at 5 min), indicating possible enterohepatic circulation. Radiometabolism analysis showed that [18F]P10A-1910 maintained 80% of parent fraction in mice brain at 30 min post-injection. PET imaging in cynomolgus showed rapid brain uptake and heterogeneous distribution. Specifically, the uptake in the striatum was increased to 1.6 SUV at 3 min, and then slowly decreased to 0.9 SUV at the end of scan, which was consistent with the expression of PDE10A in brain, indicating good specific binding. Conclusions: We have successfully synthesized a new PDE10A PET tracer [18F]P10A-1910 in good radiochemical yields and high molar activities. The biodistribution study, stability testing and preliminary NHP PET imaging data indicated that [18F]P10A-1910 is promising for further evaluation.
Spirocyclic Iodonium Ylide (Scidy)-Mediated Radiofluorination of a Novel PDE10A PET Ligand for Neuroimaging

Abstract:150 Objectives: Phosphodiesterase-10 (PDE10A), one of the dual-substrate specific enzymes of PDE superfamily, represents a potential biomarker for neurodegenerative and neuropsychiatric diseases. However, a number of outcomes in clinical trials with PDE10A inhibitors so far are disappointing. Imaging of PDE10A in vivo with effective PET tracers would considerably accelerate drug discovery and clinical application for disease diagnosis. Here we report a novel PDE10A radioligand 18F-P10A1910, including the radiolabeling, ex vivo biodistribution (bioD) and metabolism, and preliminary PET imaging on rats and rhesus by dose-dependent blocking study for evaluating specificity and receptor occupancy. Methods: The PDE10A inhibitor P10A1910 and the SCIDY precursor Pre-1 were synthesized according to the literature [1,2,3].The binding affinity and selectivity of P10A1910 to PDE10A and other fourteen PDE family enzymes were examined in vitro. The radiolabeling of 18F-P10A1910 was realized by one-step 18F incorporation via the commercially automatic GE FX2 N module. Pharmacokinetic properties were tested via bioD studies in ICR mice. In addition, dynamic PET imaging studies of 18F-P10A1910 were conducted on SD rats and rhesus for evaluating in vivo brain penetration, specificity, and target engagement. Results: The inhibitor P10A1910 and SCIDY precursor Pre-1 were synthesized in 4-5 steps with 11% and 8% overall yields, respectively. P10A1910 exhibited good binding affinity for PDE10A (IC50 = 2.05 nM) and excellent selectivity over other PDE subtypes.18F-P10A1910 was produced from Pre-1 in 15 ± 5% n.d.c RCYs (n = 30) in less than 70 min from EOB with more than 99% radiochemical purity and around 3 Ci/μmol molar radioactivity. The radioactivity at 5 min post-injection was mainly accumulated in the liver and small intestine (more than 50 %ID/g), which indicated that the excretion of radiotracer occurred primarily through the hepatobiliary clearance. Blood clearance of 18F-P10A1910 was fairly fast (activity ratio is more than 20). No obvious bone uptake was observed. PET imaging on rats showed a high brain uptake (SUVpeak = 1.5 SUV at 2 min post-injection) and a rapid washout (SUV1.4min/60 min = 3). Furthermore, the pre-blocking and chase-blocking PET study on rats demonstrated the binding of 18F-P10A1910 to PDE10A was reversible and specific. Metabolite analysis demonstrated the first-class stability of 18F-P10A-1910 in rat brain (more than 96% of intact tracer retained at 60 min post injection). Furthermore, blocking experiments with escalating doses of PF-02545920 revealed a dose-dependent decrease of striatal tracer uptake in the rhesus brain. Target engagement studies unveiled that dose of 2.09 μmol/kg PF-02545920 occupied 50% of the target receptor. In addition, our results showed that the recently reported therapeutically active PF-02545920 doses of 0.21 mg/kg and 1.33 mg/kg occupy 22% and 63% of PDE10A in the monkey brain, respectively. Accordingly, our results imply that a relatively low receptor occupancy already elicits neuroprotective activity. Further, we hypothesize that higher doses might lead to improved therapeutic efficacy by occupying a higher degree of PDE10A. These results underline the potential utility of 18F-P10A1910 for in vivo visualization and quantification of PDE10A-based target engagement. Conclusion: Based on the designed SPIAd precursor Pre-1, we successfully synthesized a novel PDE10A radioligand 18F-P10A1910 in good radiochemical yields with high purities and molar activities. In vitro binding affinity and selectivity testing, ex vivo bioD studies in mice, as well as dynamic neuroPET imaging on rats and rhesus were performed, and the results demonstrated 18F-P10A1910 is a promising candidate with adequate brain penetration, good stability and excellent specificity.
Discovery of a Highly Specific 18F-Labeled PET Ligand for Phosphodiesterase 10A Enabled by Novel Spirocyclic Iodonium Ylide Radiofluorination.

Abstract:As a member of cyclic nucleotide phosphodiesterase (PDE) enzyme family, PDE10A is in charge of the degradation of cyclic adenosine (cAMP) and guanosine monophosphates (cGMP). While PDE10A is primarily expressed in the medium spiny neurons of the striatum, it has been implicated in a variety of neurological disorders. Indeed, inhibition of PDE10A has proven to be of potential use for the treatment of central nervous system (CNS) pathologies caused by dysfunction of the basal ganglia-of which the striatum constitutes the largest component. A PDE10A-targeted positron emission tomography (PET) radioligand would enable a better assessment of the pathophysiologic role of PDE10A, as well as confirm the relationship between target occupancy and administrated dose of a given drug candidate, thus accelerating the development of effective PDE10A inhibitors. In this study, we designed and synthesized a novel 18F-aryl PDE10A PET radioligand, codenamed [18F]P10A-1910 ([18F]9), in high radiochemical yield and molar ac-tivity via spirocyclic iodonium ylide-mediated radiofluorination. [18F]9 possessed good in vitro binding af-finity (IC50 = 2.1 nmol/L) and selectivity towards PDE10A. Further, [18F]9 exhibited reasonable lipophilicity (logD = 3.50) and brain permeability (Papp > 10 x 10-6 cm/s in MDCK-MDR1 cells). PET imaging studies of [18F]9 revealed high striatal uptake and excellent in vivo specificity with reversible tracer kinetics. Preclinical studies in rodents revealed an improved plasma and brain stability of [18F]9 when compared to the current reference standard for PDE10A-targeted PET, [18F]MNI659. Further, dose-response experiments with a series of escalating doses of PDE10A inhibitor 1 in rhesus monkey brains confirmed the utility of [18F]9 for evaluating target occupancy in vivo in higher species. Abstract:Phosphodiesterase-4 (PDE4), which metabolizes the second messenger cyclic adenosine monophosphate (cAMP), has 4 isozymes: PDE4A, PDE4B, PDE4C, and PDE4D. PDE4B and PDE4D have the highest expression in the brain and may play a role in the pathophysiology and treatment of depression and dementia. This study evaluated the properties of the newly developed PDE4B-selective radioligand 18F-PF-06445974 in the brains of rodents, monkeys, and humans. Methods: Three monkeys and 5 healthy human volunteers underwent PET scans after intravenous injection of 18F-PF-06445974. Brain uptake was quantified as total distribution volume (V T) using the standard 2-tissue-compartment model and serial concentrations of parent radioligand in arterial plasma. Results: 18F-PF-06445974 readily distributed throughout monkey and human brain and had the highest binding in the thalamus. The value of V T was well identified by a 2-tissue-compartment model but increased by 10% during the terminal portions (40 and 60 min) of the monkey and human scans, respectively, consistent with radiometabolite accumulation in the brain. The average human V T values for the whole brain were 9.5 ± 2.4 mL ⋅ cm-3 Radiochromatographic analyses in knockout mice showed that 2 efflux transporters-permeability glycoprotein (P-gp) and breast cancer resistance protein (BCRP)-completely cleared the problematic radiometabolite but also partially cleared the parent radioligand from the brain. In vitro studies with the human transporters suggest that the parent radioligand was a partial substrate for BCRP and, to a lesser extent, for P-gp. Conclusion: 18F-PF-06445974 quantified PDE4B in the human brain with reasonable, but not complete, success. The gold standard compartmental method of analyzing brain and plasma data successfully identified the regional densities of PDE4B, which were widespread and highest in the thalamus, as expected. Because the radiometabolite-induced error was only about 10%, the radioligand is, in the opinion of the authors, suitable to extend to clinical studies.

Abstract:a-Amino-3-hydroxyl-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are implicated in the pathology of neurological diseases such as epilepsy and schizophrenia. As pan antagonists for this target are often accompanied with undesired effects at high doses, one of the recent drug discovery approaches has shifted to subtype-selective AMPA receptor (AMPAR) antagonists, specifically, via modulating transmembrane AMPAR regulatory proteins (TARPs). The quantification of AMPARs by positron emission tomography (PET) would help obtain insights into disease conditions in the living brain and advance the translational development of AMPAR antagonists. Herein we report the design, synthesis and preclinical evaluation of a series of TARP γ-8 antagonists, amenable for radiolabeling, for the development of subtype-selective AMPAR PET imaging agents. Based on the pharmacology evaluation, molecular docking studies and physiochemical properties, we have identified several promising lead compounds 3, 17-19 and 21 for in vivo PET studies. All candidate compounds were labeled with [11C]COCl2 in high radiochemical yields (13-31% RCY) and high molar activities (35-196 GBq/μmol). While tracers 30 ([11C]17) &32 ([11C]21) crossed the blood-brain barrier and showed heterogeneous distribution in PET studies, consistent with TARP γ-8 expression, high nonspecific binding prevented further evaluation. To our delight, tracer 31 ([11C]3) showed good in vitro specific binding and characteristic high uptake in the hippocampus in rat brain tissues, which provides the guideline for further development of a new generation subtype selective TARP γ-8 dependent AMPAR tracers.
In Vivo Characterization of Two 18F-Labeled PDE10A PET Radioligands in Nonhuman Primate Brains.

Abstract:Positron emission tomography (PET) with phosphodiesterase 10A (PDE10A) specific radioligands provides a noninvasive and quantitative imaging tool to access the expression of this enzyme in vivo under normal and diseased conditions. We recently reported two potent F-18-labeled PDE10A radioligands (F-18-TZ19106B and F-18-TZ8110); initial evaluation in rats and nonhuman primates indicated stable metabolic profiles and excellent target-to-nontarget ratio (striatum/cerebellum) for both tracers. Herein, we focused on in vivo characterization of F-18-TZ19106B and F-18-TZ8110 to identify a suitable radioligand for imaging PDE10A in vivo. We directly compared microPET studies of these two radiotracers in adult male Macaca fascicularis nonhuman primates (NHPs). F-18-TZ19106B had higher striatal uptake and tracer retention in NHP brains than F-18-TZ8110, quantified by either standardized uptake values (SUVs) or nondisplaceable binding potential (BP (ND) ) estimated using reference-based modeling analysis. Blocking and displacement studies using the PDE10A inhibitor MP-10 indicated the binding of F-18-TZ19106B to PDE10A was specific and reversible. We also demonstrated sensitivity of F-18-TZ19106B binding to varying number of specific binding sites using escalating doses of MP-10 blockade (0.3, 0.5, 1.0, 1.5, and 2.0 mg/kg). Pretreatment with a dopamine D2-like receptor antagonist enhanced the striatal uptake of F-18-TZ19106B. Our results indicate that F-18-TZ19106B is a promising radioligand candidate for imaging PDE10A in vivo and it may be used to determine target engagement of PDE10A inhibitors and serve as a tool to evaluate the effect of novel antipsychotic therapies.
Comparison of Two Pde10a Radiotracers, [11c]tz1964b and [18f]mni659 in the Brains of Non-Human Primates

Abstract:289 Objectives Phosphodiesterase 10A (PDE10A) is a potential therapeutic target for neurological diseases with striatal pathology, including Parkinson disease, Huntington disease and schizophrenia. PET radiotracers for PDE10A have been developed for in vivo assessing the PDE10A level and for target engagement for relevant therapeutic interventions. The aim of this study is to in vivo compare two potent and selective PDE10A radiotracers, [11C]TZ1964B and [18F]MNI659, in quantifying the expression of PDE10A in non-human primate (NHP) brain. Methods [11C]TZ1964B and [18F]MNI659 were used for double scans in the same NHP (totally 4 scans in 2 adult male cynomolgus monkeys, weighing 7-8 kg). The microPET was performed using a Focus 220 scanner, with an injection dose of ~10 mCi for each tracer. Binding potential (BP) of striatum was calculated to assess the radiotracer uptake using reference-based modeling (Logan Reference (LoganREF) model and simplified reference tissue model (SRTM)), with cerebellum as the reference. The striatum-to-cerebellum ratio of regional area under the time tissue-activity curve (AUC) was also calculated as a modeling-independent parameter for evaluating tracer uptake. HPLC metabolite analysis of NHP plasma was also performed for both the tracers. Results Both radiotracers showed high accumulation in NHP striatum. Striatal uptake of [11C]TZ1964B reached the max SUV value around 35 min post-injection (p.i.), while the peak of [18F]MNI659 appeared at ~10 min p.i.. Compared to [18F]MNI659, the tracer clearance of [11C]TZ1964B in the striatum was slower, probably due to differences in in vivo tracer metabolism. However, the washout in other (non-target) brain regions was similarly fast, indicating the target specificity and regional selectivity for both two tracers. The BP estimates of [11C]TZ1964B were 3.72 by LoganREF and 4.39 by SRTM; the values for [18F]MNI659 were 5.08 by BP-LoganREF and 5.33 by BP-SRTM. AUC ratios of these two tracers were 5.87 for [11C]TZ1964B and 7.60 for [18F]MNI659. Based on the BP-LoganREF and BP-SRTM data in NHP striatum, coefficients of variation were ~10% for [11C]TZ1964B and ~30% for [18F]MNI659. Additionally, the in vivo tracer metabolism study showed, at 60 min p.i., the percentage of parent compounds were ~70% for [11C]TZ1964B and ~50% for [18F]MNI659. Conclusions Both [11C]TZ1964B and [18F]MNI659 could serve as suitable PET radiotracers for assessment of PDE10A in the brain, regardless of differences in in vivo tracer characteristics. Due to the half-life of radiotracers, the C-11 tracer could be utilized for multiple injections for patients in the same day, while the F-18 tracer have commercial distribution advantage that allows delivery to multiple sites with 3-4 hrs drive distance. Abstract:As a subclass of ionotropic glutamate receptors (iGluRs), α-amino-3-hydroxyl-5-methyl-4-isoxazolepropionic acid (AMPA) receptors have been implicated in various neurological disorders and neurodegenerative diseases. To further our understanding of AMPA receptor-related disorders in the central nervous system (CNS), it is important to be able to image and quantify AMPA receptors in vivo. In this study, we identified a novel F-containing AMPA positive allosteric modulator (PAM) 6 as a potential lead compound. Molecular docking studies and CNS PET multi-parameter optimization (MPO) analysis were used to predict the absorption, distribution, metabolism, and excretion (ADME) characteristics of 6 as a PET probe. The resulting PET probe, [ 18 F] 6 (codename [ 18 F]AMPA-2109), was successfully radiolabeled and demonstrated excellent blood-brain barrier (BBB) permeability and high brain uptake in rodents and non-human primates. However, [ 18 F] 6 did not show substantial specific binding in the rodent or non-human primate brain. Further medicinal chemistry efforts are necessary to improve specific binding, and our work may serve as a starting point for the design of novel 18 F-labeled AMPA receptor-targeted PET radioligands aimed for clinical translation.

Abstract:Our previous work showed that [18F]P10A-1910 was a potential radioligand for use in imaging phosphodiesterase 10A (PDE10A). Specifically, it had high brain penetration and specific binding that was demonstrated in both rodents and non-human primates. Here, we present the first automatic cGMP-level production of [18F]P10A-1910 and translational PET/MRI study in living human brains. Successful one-step radiolabeling of [18F]P10A-1910 on a GE TRACERlab FX2N synthesis module was realized via two different methods. First, formulated [18F]P10A-1910 was derived from heating spirocyclic iodonium ylide in a tetra-n-butyl ammonium methanesulfonate solution. At the end of synthesis, it was obtained in non-decay corrected radiochemical yields (n.d.c. RCYs) of 12.4 ± 1.3%, with molar activities (MAs) of 90.3 ± 12.6 μmol (n = 7) (Method I). The boronic pinacol ester combined with copper and oxygen also delivered the radioligand with 16.8 ± 1.0% n. d.c. RCYs and 77.3 ± 20.7 GBq/μmol (n = 7) MAs after formulation (Method II). The radiochemical purity, radionuclidic purity, solvent residue, sterility, endotoxin content and other parameters were all validated for human use. Consistent with the distribution of PDE10A in the brain, escalating uptake of [18F]P10A-1910 was observed in the order of cerebellum (reference region), substantial nigra, caudate and putamen. The non-displaceable binding potential (BPND) was estimated by simplified reference-tissue model (SRTM); linear regressions demonstrated that BPND was well correlated with the most widely used semiquantitative parameter SUV. The strongest correlation was observed with SUV(50–60 min) (R2 = 0.966, p < 0.01). Collectively, these results indicated that a static scan protocol could be easily performed for PET imaging of PDE10A. Most importantly, that [18F]P10A-1910 is a promising radioligand to clinically quantify PDE10A.
Synthesis and Characterization of [18F]JNJ-46356479 as the First 18F-Labeled PET Imaging Ligand for Metabotropic Glutamate Receptor 2

Abstract:Purpose: Metabotropic glutamate receptor 2 (mGluR2) has been implicated in various psychiatric and neurological disorders, such as schizophrenia and Alzheimer's disease. We have previously developed [11C]7 as a PET radioligand for imaging mGluR2. Herein, [18F]JNJ-46356479 ([18F]8) was synthesized and characterized as the first 18F-labeled mGluR2 imaging ligand to enhance diagnostic approaches for mGluR2-related disorders. Procedures: JNJ-46356479 (8) was radiolabeled via the copper (I)-mediated radiofluorination of organoborane 9. In vivo PET imaging experiments with [18F]8 were conducted first in C57BL/6 J mice and Sprague-Dawley rats to obtain whole body biodistribution and brain uptake profile. Subsequent PET studies were done in a cynomolgus monkey (Macaca fascicularis) to investigate the uptake of [18F]8 in the brain, its metabolic stability, as well as pharmacokinetic properties. Results: JNJ-46356479 (8) exhibited excellent selectivity against other mGluRs. In vivo PET imaging studies showed reversible and specific binding characteristic of [18F]8 in rodents. In the non-human primate, [18F]8 displayed good in vivo metabolic stability, excellent brain permeability, fast and reversible kinetics with moderate heterogeneity across brain regions. Pre-treatment studies with compound 7 revealed time-dependent decrease of [18F]8 accumulation in mGluR2 rich regions based on SUV values with the highest decrease in the nucleus accumbens (18.7 ± 5.9%) followed by the cerebellum (18.0 ± 7.9%), the parietal cortex (16.9 ± 7.8%), and the hippocampus (16.8 ± 6.9%), similar to results obtained in the rat studies. However, the volume of distribution (VT) results derived from 2T4k model showed enhanced VT from a blocking study with compound 7. This is probably because of the potentiating effect of compound 7 as an mGluR2 PAM as well as related non-specific binding in the tissue data. Conclusions: [18F]8 readily crosses the blood-brain barrier and demonstrates fast and reversible kinetics both in rodents and in a non-human primate. Further investigation of [18F]8 on its binding specificity would warrant translational study in human.

Abstract:Muscarinic acetylcholine receptors (mAChR), including M4, draw attention as therapeutic targets for several neurodegenerative diseases including Alzheimer's disease (AD). PET imaging of M4 positive allosteric modulator (PAM) allows qualification of the distribution as well as the expression of this receptor under physiological conditions and thereby helps to assess the receptor occupancy (RO) of a drug candidate. In this study, our aims were (a) to synthesize a novel M4 PAM PET radioligand [11C]PF06885190 (b) to evaluate the brain distribution of [11C]PF06885190 in nonhuman primates (NHP) and (c) to analyze its radiometabolites in the blood plasma of NHP. Radiolabeling of [11C]PF06885190 was accomplished via N-methylation of the precursor. Six PET measurements were performed using two male cynomolgus monkeys, where three PET measurements were at baseline, two after pretreatment with a selective M4 PAM compound CVL-231 and one after pretreatment with donepezil. The total volume of distribution (VT) of [11C]PF06885190 was examined using Logan graphical analysis with arterial input function. Radiometabolites were analyzed in monkey blood plasma using gradient HPLC system. Radiolabeling of [11C]PF06885190 was successfully accomplished and the radioligand was found to be stable in the formulation, with radiochemical purity exceeding 99% 1 h after the end of the synthesis. [11C]PF06885190 was characterized in the cynomolgus monkey brain where a moderate brain uptake was found at the baseline condition. However, it showed fast wash-out as it dropped to half of the peak at around 10 min. Change of VT from baseline was around −10% after pretreatment with a M4 PAM, CVL-231. Radiometabolite studies showed relatively fast metabolism. Although sufficient brain uptake of [11C]PF06885190 was observed, these data suggest that [11C]PF06885190 might have too low specific binding in the NHP brain to be further applied in PET imaging.

Abstract:The cyclic nucleotide phosphodiesterase 2A is an intracellular enzyme which hydrolyzes the secondary messengers cAMP and cGMP and therefore plays an important role in signaling cascades. A high expression in distinct brain areas as well as in cancer cells makes PDE2A an interesting therapeutic and diagnostic target for neurodegenerative and neuropsychiatric diseases as well as for cancer. Aiming at specific imaging of this enzyme in the brain with positron emission tomography (PET), a new triazolopyridopyrazine-based derivative (11) was identified as a potent PDE2A inhibitor (IC50, PDE2A = 1.99 nM; IC50, PDE10A ~2000 nM) and has been radiofluorinated for biological evaluation. In vitro autoradiographic studies revealed that [18F]11 binds with high affinity and excellent specificity towards PDE2A in the rat brain. For the PDE2A-rich region nucleus caudate and putamen an apparent KD value of 0.24 nM and an apparent Bmax value of 16 pmol/mg protein were estimated. In vivo PET-MR studies in rats showed a moderate brain uptake of [18F]11 with a highest standardized uptake value (SUV) of 0.97. However, no considerable enrichment in PDE2A-specific regions in comparison to a reference region was detectable (SUVcaudate putamen = 0.51 vs. SUVcerebellum = 0.40 at 15 min p.i.). Furthermore, metabolism studies revealed a considerable uptake of radiometabolites of [18F]11 in the brain (66% parent fraction at 30 min p.i.). Altogether, despite the low specificity and the blood−brain barrier crossing of radiometabolites observed in vivo, [18F]11 is a valuable imaging probe for the in vitro investigation of PDE2A in the brain and has potential as a lead compound for further development of a PDE2A-specific PET ligand for neuroimaging.

Abstract:AMPA glutamate receptors (AMPARs) play a pivotal role in excitatory neurotransmission, particularly in the hippocampus where the TARP γ‐8 subunit is enriched and serves as a target for emerging anti‐epileptic drugs. To enable in vivo visualization of TARP γ‐8 distribution and expression by positron emission tomography (PET), this study focuses on the development of novel 18F‐labeled TARP γ‐8 inhibitors and their corresponding precursors, stemming from the azabenzimidazole scaffold. The resulting radioligands [18F]TARP‐2204 and [18F]TARP‐2205 were successfully synthesized with acceptable radiochemical yield, high molar activity, and excellent radiochemical purity. In vitro autoradiography demonstrates high level of specific binding of [18F]TARP‐2205 to TARP γ‐8 in both rat and nonhuman primate brain tissues. However, unexpected radiodefluorination in PET imaging studies of rodents emphasizes the need for further structural refinement. This work serves as an excellent starting point for the development of future 18F‐labeled TARP γ‐8 PET tracers, offering valuable insights into medicinal chemistry design, radiosynthesis and subsequent PET evaluation.

Abstract:The sigma 1 receptor (S1R) is widely expressed in the CNS and is mainly located on the endoplasmic reticulum. The S1R is involved in the regulation of many neurotransmission systems and, indirectly, in neurodegenerative diseases. The S1R may therefore represent an interesting neuronal biomarker in neurodegenerative diseases such as Parkinson's (PD) or Alzheimer's diseases (AD). Here we present the characterisation of the S1R-specific 18F-labelled tracer 18F-IAM6067 in two animal models and in human brain tissue. Methods: Wistar rats were used for PET-CT imaging (60 min dynamic acquisition) and metabolite analysis (1, 2, 5, 10, 20, 60 min post-injection). To verify in vivo selectivity, haloperidol, BD1047 (S1R ligand), CM398 (S2R ligand) and SB206553 (5HT2B/C antagonist) were administrated for pre-saturation studies. Excitotoxic lesions induced by intra-striatal injection of AMPA were also imaged by 18F-IAM6067 PET-CT to test the sensitivity of the methods in a well-established model of neuronal loss. Tracer brain uptake was also verified by autoradiography in rats and in a mouse model of PD (intrastriatal 6-hydroxydopamine (6-OHDA) unilateral lesion). Finally, human cortical binding was investigated by autoradiography in three groups of subjects (control subjects with Braak ≤2, and AD patients, Braak >2 & ≤4 and Braak >4 stages). Results: We demonstrate that despite rapid peripheral metabolism of 18F-IAM6067, radiolabelled metabolites were hardly detected in brain samples. Brain uptake of 18F-IAM6067 showed differences in S1R anatomical distribution, namely from high to low uptake: pons-raphe, thalamus medio-dorsal, substantia nigra, hypothalamus, cerebellum, cortical areas and striatum. Pre-saturation studies showed 79-90% blockade of the binding in all areas of the brain indicated above except with the 5HT2B/C antagonist SB206553 and S2R ligand CM398 which induced no significant blockade, indicating good specificity of 18F-IAM6067 for S1Rs. No difference between ipsi- and contralateral sides of the brain in the mouse model of PD was detected. AMPA lesion induced a significant 69% decrease in 18F-IAM6067 uptake in the globus pallidus matching the neuronal loss as measured by NeuN, but only a trend to decrease (-16%) in the caudate putamen despite a significant 91% decrease in neuronal count. Moreover, no difference in the human cortical binding was shown between AD groups and controls. Conclusion: This work shows that 18F-IAM6067 is a specific and selective S1R radiotracer. The absence or small changes in S1R detected here in animal models and human tissue warrants further investigations and suggests that S1R might not be the anticipated ideal biomarker for neuronal loss in neurodegenerative diseases such as AD and PD.

Abstract:Chemogenetic tools are designed to control neuronal signaling. These tools have the potential to contribute to the understanding of neuropsychiatric disorders and to the development of new treatments. One such chemogenetic technology comprises modified Pharmacologically Selective Actuator Modules (PSAMs) paired with Pharmacologically Selective Effector Molecules (PSEMs). PSAMs are receptors with ligand-binding domains that have been modified to interact only with a specific small-molecule agonist, designated a PSEM. PSAM 4 is a triple mutant PSAM derived from the α7 nicotinic receptor (α7 L131G,Q139L,Y217F ). Although having no constitutive activity as a ligand-gated ion channel, PSAM 4 has been coupled to the serotonin 5-HT 3 receptor (5-HT 3 R) and to the glycine receptor (GlyR). Treatment with the partner PSEM to activate PSAM 4 -5-HT 3 or PSAM 4 -GlyR, causes neuronal activation or silencing, respectively. A suitably designed radioligand may enable selective visualization of the expression and location of PSAMs with positron emission tomography (PET). Here, we evaluated uPSEM792, an ultrapotent PSEM for PSAM 4 -GlyR, as a possible lead for PET radioligand development. We labeled uPSEM792 with the positron-emitter, carbon-11 ( t 1/2 = 20.4 min), in high radiochemical yield by treating a protected precursor with [ 11 C]iodomethane followed by base deprotection. PET experiments with [ 11 C]uPSEM792 in rodents and in a monkey transduced with PSAM 4 -GlyR showed low peak radioactivity uptake in brain. This low uptake was probably due to high polarity of the radioligand, as evidenced by physicochemical measurements, and to the vulnerability of the radioligand to efflux transport at the blood–brain barrier. These findings can inform the design of a more effective PSAM 4 based PET radioligand, based on the uPSEM792 chemotype.

Abstract:There is an accumulating body of evidence implicating the muscarinic acetylcholine receptor 4 (M 4 ) in schizophrenia and dementia with Lewy bodies, however, a clinically validated M 4 positron emission tomography (PET) radioligand is currently lacking. As such, the aim of this study was to develop a suitable M 4 PET ligand that allows the non-invasive visualization of M 4 in the brain. Structure–activity relationship studies of pyrazol-4-yl-pyridine derivates led to the discovery of target compound 12 ― a subtype-selective positive allosteric modulator (PAM). The radiofluorinated analogue, [ 18 F] 12 , was synthesized in 28 ± 10% radiochemical yield, >37 GBq/μmol and an excellent radiochemical purity >99%. Initial in vitro autoradiograms on rodent brain sections were performed in the absence of carbachol and showed moderate specificity as well as a low selectivity of [ 18 F] 12 for the M 4 -rich striatum. However, in the presence of carbachol, a significant increase in tracer binding was observed in the rat striatum, which was reduced by >60% under blocking conditions, thus indicating that orthosteric ligand interaction is required for efficient binding of [ 18 F] 12 to the allosteric site. Remarkably, however, the presence of carbachol was not required for high specific binding in the non-human primate (NHP) and human striatum, and did not further improve the specificity and selectivity of [ 18 F] 12 in higher species. These results pointed towards significant species-differences and paved the way for a preliminary PET study in NHP, where peak brain uptake of [ 18 F] 12 was found in the putamen and temporal cortex. In conclusion, we report on the identification and preclinical development of the first radiofluorinated M 4 PET radioligand with promising attributes. The availability of a clinically validated M 4 PET radioligand harbors potential to facilitate drug development and provide a useful diagnostic tool for non-invasive imaging.

18 F-Labeled PET Tracers Specific for Adenosine A 2A Receptor: Design, Synthesis, and Biological Evaluation

Abstract:By modifying the structures of targeted A2AR antagonists and tracers, novel compounds 3, 7a, 9, 12c, and BIBD-399 were designed and synthesized. In vitro inhibition experiments demonstrated that 3, 12c, and BIBD-399 have high affinity for A2AR. [18F]3 and [18F]BIBD-399 were successfully synthesized. In terms of biological distribution, the brain uptake of [18F]MNI-444 exhibits greater than that of [18F]3 and [18F]BIBD-399. PET imaging shows that [18F]3 is off-target in the brain, while [18F]BIBD-399 and [18F]MNI-444 can be specifically imaged in regions with high A2AR expression. Differently, [18F]BIBD-399 could quickly reach equilibrium in the targeted region within 10 min after administration, while [18F]MNI-444 shows a slowly increasing trend within 2 h of administration. [18F]BIBD-399 is mainly metabolized by the liver and kidney, and there is no obvious defluorination in vivo. Additional in vitro autoradiography showed that the striatal signals of [18F]BIBD-399 and [18F]MNI-444 were inhibited by the A2AR antagonist SCH442416 but not by the A1R antagonist DPCPX, demonstrating the high A2AR binding specificity of [18F]BIBD-399. Molecular docking further confirms the high affinity of MNI-444 and BIBD-399 for A2AR. Further tMCAo imaging showed that [18F]BIBD-399 can sensitively distinguish between infarcted and noninfarcted sides, a capability not observed with [18F]MNI-444. Given its pharmacokinetic properties and the ability to identify lesion regions, [18F]BIBD-399 has potential advantages in monitoring A2AR changes, meriting further clinical investigation.

Abstract:Backgroundα-Amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptor is a primary mediator of fast glutamatergic excitatory signaling in the brain and has been implicated in diverse neuropsychiatric diseases. We recently developed a novel positron emission tomography (PET) ligand, 2-(1-(3-([11C]methylamino)phenyl)-2-oxo-5-(pyrimidin-2-yl)-1,2-dihydropyridin-3-yl) benzonitrile ([11C]HMS011). This compound is a radiolabelled derivative of perampanel, an antiepileptic drug acting on AMPA receptors, and was demonstrated to have promising in vivo properties in the rat and monkey brains. In the current study, we performed a human PET study using [11C]HMS011 to evaluate its safety and kinetics.Four healthy male subjects underwent a 120-min PET scan after injection of [11C]HMS011. Arterial blood sampling and metabolite analysis were performed to obtain parent input functions for three of the subjects using high-performance liquid chromatography. Regional distribution volumes (VTs) were calculated based on kinetic models with and without considering radiometabolite in the brain. The binding was also quantified using a reference tissue model with white matter as reference.ResultsBrain uptake of [11C]HMS011 was observed quickly after the injection, followed by a rapid clearance. Three hydrophilic and one lipophilic radiometabolites appeared in the plasma, with notable individual variability. The kinetics in the brain with apparent radioactivity retention suggested that the lipophilic radiometabolite could enter the brain. A dual-input graphical model, an analytical model designed in consideration of a radiometabolite entering the brain, well described the kinetics of [11C]HMS011. A reference tissue model showed small radioligand binding potential (BP*ND) values in the cortical regions (BP*ND = 0–0.15). These data suggested specific binding component of [11C]HMS011 in the brain.ConclusionsKinetic analyses support some specific binding of [11C]HMS011 in the human cortex. However, this ligand may not be suitable for practical AMPA receptor PET imaging due to the small dynamic range and metabolite in the brain.

Abstract:Fatty acid amide hydrolase (FAAH) is one of the principle enzymes for metabolizing endogenous cannabinoid neurotransmitters such as anandamide, and thus regulates endocannabinoid (eCB) signaling. Selective pharmacological blockade of FAAH has emerged as a potential therapy to discern the endogenous functions of anandamide-mediated eCB pathways in anxiety, pain, and addiction. Quantification of FAAH in the living brain by positron emission tomography (PET) would help our understanding of the endocannabinoid system in these conditions. While most FAAH radiotracers operate by an irreversible ("suicide") binding mechanism, a FAAH tracer with reversibility would facilitate quantitative analysis. We have identified and radiolabeled a reversible FAAH inhibitor, 7-(2-[(11)C]methoxyphenyl)-1-(5-(pyridin-2-yl)oxazol-2-yl)heptan-1-one ([(11)C]MPPO) in 13% radiochemical yield (nondecay corrected) with >99% radiochemical purity and 2 Ci/μmol (74 GBq/μmol) specific activity. The tracer showed moderate brain uptake (0.8 SUV) with heterogeneous brain distribution. However, blocking studies with a potent FAAH inhibitor URB597 demonstrated a low to modest specificity to the target. Measurement of lipophilicity, metabolite, and efflux pathway analysis were also performed to study the pharmacokinetic profile of [(11)C]MPPO. In all, we reported an efficient radiolabeling and preliminary evaluation of the first-in-class FAAH inhibitor [(11)C]MPPO with α-ketoheterocyclic scaffold.
