Oral Ingestion of AP Collagen Peptide Leads to Systemic Absorption of Gly-Pro-Hyp, Alleviating H2O2-Induced Dermal Fibroblast Aging

Abstract:Collagen-derived dipeptides and tripeptides have various physiological activities. In this study, we compared the plasma kinetics of free Hyp, peptide-derived Hyp, Pro-Hyp, cyclo(Pro-Hyp), Hyp-Gly, Gly-Pro-Hyp, and Gly-Pro-Ala after ingestion of four different collagen samples: AP collagen peptide (APCP), general collagen peptide, collagen, and APCP and γ-aminobutyric acid (GABA) combination. Each peptide was measured by high-performance liquid chromatography and triple quadrupole mass spectrometer. We found that, among all the peptides that were analyzed, only Gly-Pro-Hyp was significantly increased after ingestion of APCP compared with that of general collagen peptides and collagen. In addition, ingestion of the APCP and GABA combination improved the absorption efficiency of Gly-Pro-Ala. Finally, we reveal that Gly-Pro-Hyp was effective for preventing H2O2-induced reduction in extracellular matrix (ECM)-related genes, COL1A, elastin, and fibronectin, in dermal fibroblasts. Taken together, APCP significantly enhances the absorption of Gly-Pro-Hyp, which might act as an ECM-associated signaling factor in dermal fibroblasts, and the APCP and GABA combination promotes Gly-Pro-Ala absorption. Ingestion of bioactive collagen hydrolysates enhance facial skin moisture and elasticity and reduce facial ageing signs in a randomised double-blind placebo-controlled clinical study

Abstract:Background: Several human studies have demonstrated occurrence of two major collagen peptides, prolyl-hydroxyproline (Pro-Hyp) and hydroxyprolyl-glycine (Hyp-Gly), in human peripheral blood. Some in vitro studies have demonstrated that Pro-Hyp and Hyp-Gly exert chemotaxis on dermal fibroblasts and enhance cell proliferation. Additionally, Pro-Hyp enhances the production of hyaluronic acid by dermal fibroblasts. These findings suggest that the amounts of Pro-Hyp and Hyp-Gly in blood are important factors to show the efficacy of collagen hydrolysates on skin health. Results: We conducted a randomised double-blind placebo-controlled clinical trial of ingestion of two types of collagen hydrolysates, which are composed of different amounts of the bioactive dipeptides Pro-Hyp and Hyp-Gly, to investigate their effects on the improvement of skin conditions. Improvement in skin conditions, such as skin moisture, elasticity, wrinkles, and roughness, were compared with a placebo group at baseline, and 4 and 8 weeks after the start of the trial. In addition, the safety of dietary supplementation with these peptides was evaluated by blood test. Collagen hydrolysate with a higher content of bioactive collagen peptides (H-CP) showed significant and more improvement than the collagen hydrolysate with a lower content of bioactive collagen peptides (L-CP) and the placebo, in facial skin moisture, elasticity (R2), wrinkles and roughness, compared with the placebo group. In addition, there were no adverse events during the trial. Conclusion: This study demonstrated that the use of the collagen hydrolysate with a higher content of Pro-Hyp and Hyp-Gly led to more improvement in facial skin conditions, including facial skin moisture, elasticity, wrinkles and roughness. © 2016 Society of Chemical Industry.
Orally administered collagen peptide protects against UVB-induced skin aging through the absorption of dipeptide forms, Gly-Pro and Pro-Hyp

Abstract:Collagen hydrolysate is a well-known nutritional supplement for the improvement of healthy skin. Here, collagen peptide NS (CPNS) from fish scale was prepared, and its physicochemical properties were investigated. Gly-Pro was revealed as a representative low molecular weight peptide of CPNS, by performing prep-HPLC and LC-MS/MS. CPNS treatment attenuated matrix metalloproteinase-1 production and increased the synthesis of type 1 procollagen in HDF cells. After orally administering CPNS to rats, the plasma concentrations of Gly-Pro and Pro-Hyp increased dramatically. To examine the protective effects of CPNS against ultraviolet B (UVB)-induced photoaging in vivo, the dorsal skins of hairless mice were exposed to UVB and supplemented with CPNS for 12 weeks. The CPNS consumption significantly attenuated UVB-induced wrinkle formation, transepidermal water loss, and epidermis thickness, and increased skin hydration. Collectively, these results suggest that bioactive peptides of CPNS, Gly-Pro and Pro-Hyp, exert beneficial effects on skin health.
Effects of Collagen Hydrolysates on UV-induced Photoaging Mice: Gly-Pro-Hyp as a Potent Anti-photoaging Peptide

Abstract:This work aimed to investigate the protective effects of collagen hydrolysates containing different contents of Gly-Pro-Xaa tripeptides on UV-induced photoaging mice and to identify potent anti-photoaging peptides. Results showed that oral ingestion of collagen hydrolysate with higher content of Gly-Pro-Xaa tripeptides (~11.4%, HCH) dramatically enhanced absorption of Pro-Hyp, Hyp-Gly, and Gly-Pro-Hyp into the body, which were 1.77-, 2.18-, and 65.07-fold higher in area under the concentration-time curve (AUC) values than that of collagen hydrolysate with lower content of Gly-Pro-Xaa tripeptides (~3.8%, LCH), respectively. Furthermore, protective effects of HCH on photo-aged skin of mice were significantly stronger than those of LCH in terms of increases in contents of hyaluronic acid and collagen, improvement in skin elasticity and epidermal thickness, alleviation in inflammation, and decreases in contents of matrix metalloproteinase-1 (MMP-1) and MMP-3. More importantly, Gly-Pro-Hyp displayed potent anti-photoaging activities comparable with HCH based on an equivalent amount of Hyp. Network pharmacology analysis for potential mechanisms further indicated that Gly-Pro-Hyp might interact with JUN and FOS, and regulate IL-17 and TNF signaling pathways. Collectively, our results suggested that HCH had great potential to be applied into functional foods for skin health and Gly-Pro-Hyp was a potent collagen-derived anti-photoaging peptide, which might contribute to the excellent anti-photoaging effects of HCH.

Abstract:Introduction: We aimed to investigate the effect of orally ingested collagen peptides (CPs) on skin condition and elucidate their mechanism of action. Methods: A randomized, placebo-controlled, double-blind trial was conducted in 99 healthy Japanese women, aged 35–50 years. The subjects were randomized into 3 groups (33 subjects/group) to receive 1 or 5 g of CP or placebo once daily for 12 weeks. Skin water content, transepidermal water loss (TEWL), skin elasticity, and skin thickness were evaluated before treatment and after 4, 8, and 12 weeks of treatment. The level of natural moisturizing factor (NMF) constituents in the stratum corneum (SC) was quantified before treatment and after 12 weeks of treatment. Results: Oral ingestion of CP increased the water content in the SC and epidermis and decreased TEWL. Furthermore, the NMF level in the SC was increased. However, skin elasticity and skin thickness remained unchanged. Conclusions: The improvement in skin water content following the oral ingestion of CP can be attributed to an increase in the level of NMF in the SC. Abstract:85 I AM SENDING THIS LETTER to comment on the paper about the hand-assisted surgical technique that was recently published.1 The paper showed some cases where the handassisted technique was used for the laparoscopic retroperitoneal dissection of lymph nodes in nonseminomatous testicular cancer, and the authors concluded that this procedure makes the laparoscopic surgery simpler, faster, and safe. This technique was first described by Tobias-Machado et al. and the results already published.2 We want to draw attention to a few points of the technique that seem very important and are built on our experience. Conventional retroperitoneal lymphadenectomy is a major surgical procedure, which confers considerable morbidity to patients with nonseminomatous testicular tumor. The search for less invasive techniques identified the video laparoscopic surgery as a procedure with lower morbidity and no impairment in the oncologic control of the disease.2 On the other hand, we must consider that preliminary series of the laparoscopic approach recommend chemotherapy for positive nodes patients. It is clear that this kind of surgery has a considerable risk of vascular damage, demanding surgeons with large experience in vascular sutures by the laparoscopic approach, using vascular clamps and/or clips. The manual conversion in cases of emergency or difficulties may be extremely useful, especially when vascular laparoscopic material is not available or in cases where postchemotherapy fibrosis prevented the safe dissection of the great vessels. The hand-assisted technique is a useful method to prevent the surgery conversion or, eventually, to convert the surgery with a tamponed lesion, reducing the blood loss. However, in stage I patients, the carefully dissection posterior of the great and lumbar vessels and clipping lymphatic offer safety and complete resection of a small surgical specimen without hand assistance. In October 2004, Tobias-Machado et al. were the first to report the possibility of the use of the hand-assisted technique in laparoscopic retroperitoneal lymphadenectomy for nonseminomatous testicular tumors. Due to this fact, our work was cited in an important review article of laparoscopic retroperitoneal lymphadenectomy for nonseminomatous germ-cell tumors in 2007.3
Effect of Orally Administered Collagen Peptides from Bovine Bone on Skin Aging in Chronologically Aged Mice

Abstract:Collagen peptides (CPs) have demonstrated to exert beneficial effects on skin photoaging. However, little has been done to evaluate their effects on chronologically aged skin. Here, the effects of CPs from bovine bone on skin aging were investigated in chronologically aged mice. 13-month-old female Kunming mice were administered with CPs from bovine bone (200, 400 and 800 mg/kg body weight/day) or proline (400 mg/kg body weight/day) for 8 weeks. Mice body weight, spleen index (SI) and thymus index (TI), degree of skin laxity (DSL), skin components, skin histology and antioxidant indicators were analyzed. Ingestion of CPs or proline had no effect on mice skin moisture and hyaluronic acid content, but it significantly improved the skin laxity, repaired collagen fibers, increased collagen content and normalized the ratio of type I to type III collagen in chronologically aged skin. CPs prepared by Alcalase performed better than CPs prepared by collagenase. Furthermore, CPs intake also significantly improved the antioxidative enzyme activities in skin. These results indicate that oral administration of CPs from bovine bone or proline can improve the laxity of chronologically aged skin by changing skin collagen quantitatively and qualitatively, and highlight their potential application as functional foods to combat skin aging in chronologically aged process.

Abstract:Collagen-peptide supplementation could be an effective remedy to improve hydration, elasticity, and wrinkling in human skin. The aim of this study was to conduct a double-blind, randomized, placebo-controlled trial to clinically evaluate the effect on human skin hydration, wrinkling, and elasticity of Low-molecular-weight Collagen peptide (LMWCP) with a tripetide (Gly-X-Y) content >15% including 3% Gly-Pro-Hyp. Individuals (n = 64) were randomly assigned to receive either placebo or 1000 mg of LMWCP once daily for 12 weeks. Parameters of skin hydration, wrinkling, and elasticity were assessed at baseline and after 6 weeks and 12 weeks. Compared with the placebo group, skin-hydration values were significantly higher in the LMWCP group after 6 weeks and 12 weeks. After 12 weeks in the LMWCP group, visual assessment score and three parameters of skin wrinkling were significantly improved compared with the placebo group. In case of skin elasticity, one parameter out of three was significantly improved in the LMWCP group from the baseline after 12 weeks, while, compared with the placebo group, two parameters out of three in the LMWCP group were higher with significance after 12 weeks. In terms of the safety of LMWCP, none of the subjects presented adverse symptoms related to the test material during the study period. These results suggest that LMWCP can be used as a health functional food ingredient to improve human skin hydration, elasticity, and wrinkling.

Abstract:Skin problems caused by aging have attracted much attention, and marine collagen peptides have been proved to improve these problems, while mammalian collagen peptides are rarely reported. In this study, fermented deer bone collagen peptide (FCP) and non-fermented deer bone collagen peptide (NCP) were extracted from fermented and non-fermented deer bone, respectively, and their peptide sequences and differential proteins were analyzed using LC-MS/MS technology. After they were applied to aging mice induced with D-gal, the skin hydration ability, antioxidant ability, collagen synthesis, and degradation ability of the mice were studied. The results show that FCP and NCP are mainly peptides that constitute type I collagen, and their peptide segments are different. In vivo experiments show that FCP and NCP can improve the richness of collagen fibers in the skin of aging mice; improve the hydration ability of skin; promote the activity of antioxidant-related enzymes; and also show that through the TGF-β and MAPK pathways, the synthesis and degradation of collagen in skin are regulated. These results show that deer bone collagen peptide can improve skin problems caused by aging, promote skin hydration and antioxidant capacity of aging mice, and regulate collagen synthesis and degradation through the MAPK pathway.

Patterns of relapse and survival following radical mastectomy. Analysis of 716 consecutive patients

Abstract:The records of 716 consecutive patients with breast cancer randomized from January 1964 to January 1968 between radical and extended radical mastectomy not followed by postoperative radiation therapy were reviewed. Relapse and survival rates were related to different patterns with the intent to define, on clinical basis, the high‐risk groups in whom systemic adjuvant treatment can be attempted. The 10‐year relapse rate was 52.9% for the whole series. In patients with negative axillary nodes (N‐) this finding was 27.9% compared to 75.5% for patients with positive nodes (N+ 1–3: 66.5%, N+>3: 83.6%). The relapse rate was also affected by concomitant involvement of internal mammary nodes (IMN): N‐ IMN+: 60%, N+ IMN+: 96.7%. The corresponding 10‐year survival rates were as follows: 59.7% (total), 81.9% (N‐), 39.6% (N+), 53.7% (1–3), 25.6% (>3), 45.8% (N‐ IMN+), 20.5% (N+ IMN+). In N+ patients relapse and survival rates were directly proportional to the size of primary tumor, while this trend was not present in N‐ patients. Location of primary tumor as well as menopausal status showed no direct effect on relapse and survival. In both nodal groups the sites of first relapse were documented to occur preferentially in distant organs and tissues.
Hen Collagen Hydrolysate Alleviates UVA-induced Damage in Human Dermal Fibroblasts

Abstract:Collagen hydrolysate (CH) and peptides may have anti-aging properties on skin. In this study, CH prepared from the skin of porcine, bovine, tilapia, and hen were supplemented to human dermal fibroblasts (HDF) with UVA-exposure. Bovine CH inhibited matrix metalloproteinases (MMP)-1 synthesis. Tilapia CH promoted cell viability and pro-collagen I production, while inhibited the generation of reactive oxygen species (ROS) and MMP-1 in UVA-exposed HDF cells. Hen CH improved viability and pro-collagen I production, alleviated the expression of apoptotic genes, reduced ROS, MMP-1, and MMP-9 production, induced discoidin domain receptor 2 (DDR2) phosphorylation, and inhibited UVA-induced Akt and ERK1/2 phosphorylation in HDF cells, an effect largely comparable with a collagen-derived tripeptide Gly-Pro-Hyp. These results indicate that hen skin CH is superior to porcine, bovine, and tilapia skin CH on the protection of UVA-induced damage in fibroblast, with DDR2 being at least partially involved.

Abstract:Background: Oral low-molecular-weight collagen peptide (LMWCP) hydrolyzed enzymatically is a fish-derived type I collagen hydrolysate with more than 15% of its content made up of tripeptides in the form of Gly-X-Y (X and Y are placed arbitrarily, but are often occupied by proline, hydroxyproline, and alanine) including 3% Gly-Pro-Hyp. LMWCP helps with skin hydration, wrinkles and elasticity via previous findings, has been recognized individually by the Ministry of Food and Drug Safety (MFDS notice No. 2013-30) as a functional food ingredient. In this study, to expand the scope of diversity in the efficacy of LMWCP, we evaluated hydration according to depth of the stratum corneum, facial lifting, dermal density, skin thickness, skin desquamation, and roughness of the nail plate surface. Moreover, the measurement timelines were considered including the early time intake and off-intake periods. Methods and materials: This study was designed as a double-blind, randomized, placebo-controlled for 14 weeks including oral intake for 12 weeks followed by 2 weeks of the off-intake period. Water content (depths of 0.1 mm, 0.5 mm), facial lifting, dermal density, skin thickness and skin desquamation were assessed at baseline, 2 weeks, 4 weeks, 8 weeks, 12 weeks after intake of the oral supplementation and after 2 weeks of off-intake (+2W). The roughness of the fingernail plate was measured at 0W, 8W and 12W. Results: The test group saw significant improvements compared to the placebo group. According to each measurement result, the skin moisture (depth of 0.1 mm) and skin desquamation were improved after 2 weeks of ingestion, and the skin moisture (depth of 0.5 mm), facial lifting, dermal density and skin thickness were improved after 4 weeks. For all measurement items, even after 2 weeks of off-intake, the test group showed a statistically significant improvement compared to the placebo group. In the roughness of the fingernail plate, it was found that the roughness was improved in the test group after 12 weeks compared to before ingestion. Discussion: These results demonstrated that the effects of LMWCP appear in early-intake and are maintained even after off-intake. This study suggests LMWCP as a safe and effective ingredient for anti-skin aging in the nutricosmetic market targeting both internal and external beauty and health.

Abstract:In this randomized, double-blind, placebo-controlled study, we investigated the effects of collagen peptides (CP) containing high concentrations of prolyl-hydroxyproline and hydroxyprolyl-glycine on advanced glycation end products (AGEs) levels in the skin and subcutaneous blood vessel walls. A total of 31 individuals aged 47-87 years were randomly assigned to receive either 5 g/day of fish-derived CP or a placebo for 12 weeks. Body and blood compositions and AGEs levels were measured at the beginning and end of the study. No adverse events were observed, and both groups' blood and body compositions did not change significantly. However, the CP group had significantly lower AGEs levels and a slightly lower insulin resistance index (homeostasis model assessment ratio [HOMA-R]) than the placebo group. In addition, the percentage changes in AGEs and HOMA-R levels were positively and strongly correlated in both groups. These findings suggest that fish-derived CP may be effective in reducing AGEs levels and improving insulin resistance.

Abstract:Skin aging is induced by exposure to extrinsic factors, causing various diseases and adversely affecting aesthetics. Studies have suggested that as the quality of life improves, demand for beauty and nutritional cosmetics increases. Here, the protective effects of collagen peptide against UV-induced skin damage were evaluated in vitro and in vivo. Collagen peptide inhibited water loss and UVB irradiation-induced HA degradation in the skin of SKH-1 mice. Additionally, collagen peptide dose-dependently inhibited UVB-induced wrinkle formation, epidermal thickness, and elastase activity. These results suggest that collagen peptide regulates collagen degradation through the MAPK and MAPKK pathway. In addition, collagen peptide administration did not affect changes in weight of the liver, spleen, and kidney, or enzymatic indicators of liver damage. Taken together, oral administration of collagen peptide improved the effects of UV-induced skin aging without toxicity. Therefore, this study supports the development of collagen peptide for skin aging prevention in nutricosmetic products.

Abstract:The collagen present in the dermis of the skin is a fibrous protein that fills the gaps between cells and helps maintain tissue flexibility. Effectively increasing the collagen present in the skin is an important goal for cosmetic research. Recent research has shown that soybean peptide (SP) has anti-fatigue activity, antioxidant activity, and the ability to increase type I collagen, while collagen peptide (CP) has the ability to enhance corneal moisture content and viscoelasticity, as well as to increase levels of hyaluronic acid synthesizing enzymes in human skin. Little documented research, however, has been conducted on collagen formation in relation to these peptides. Therefore, this research applied SP and CP with molecular weights primarily around 500 and preparations containing both SP and CP to normal human dermal fibroblasts together with magnesium ascorbyl phosphate (VC-PMg), and used real-time PCR to determine the gene expression of type I collagen (COL1A1), which contributes to collagen synthesis, and Smad7, which contribute to collagen breakdown. In addition, enzyme linked immuno sorbent assay (ELISA) was used to measure collagen content in the media. COL1A1 gene expression at 24 h after sample addition showed higher tendency in all samples and increased with time at 4, 8 and 24 h after addition. Smad7 gene expression was not substantially different at 4 h after addition. matrix metalloproteinase-1 gene expression was higher following SP addition, but was lower after the addition of CP and SP+CP. Medium collagen content was higher in all samples and increased with time at 8 h after addition. Collagen levels were higher when SP and CP were added together.

The Protective Effects of Long-Term Oral Administration of Marine Collagen Hydrolysate from Chum Salmon on Collagen Matrix Homeostasis in the Chronological Aged Skin of Sprague-Dawley Male Rats.

Abstract:To investigate the long-term effects of marine collagen hydrolysate (MCH) from Chum Salmon skin on the aberrant collagen matrix homeostasis in chronological aged skin, Sprague-Dawley male rats of 4-wk-old were orally administrated with MCH at the diet concentrations of 2.25% and 4.5% for 24 mo. Histological and biochemical analysis revealed that MCH had the potential to inhibit the collagen loss and collagen fragmentation in chronological aged skin. Based on immunohistochemistry and western blot analysis, collagen type I and III protein expression levels in MCH-treated groups significantly increased as compared with the aged control group. Furthermore, quantitative real-time polymerase chain reaction and western blot analysis showed MCH was able to increase the expressions of procollagen type I and III mRNA (COL1A2 and COL3A1) through activating Smad signaling pathway with up-regulated TGF-βRII (TβRII) expression level. Meanwhile, MCH was shown to inhibit the age-related increased collagen degradation through attenuating MMP-1 expression and increasing tissue inhibitor of metalloproteinases-1 expression in a dose-dependent manner. Moreover, MCH could alleviate the oxidative stress in chronological aged skin, which was revealed from the data of superoxide dismutase activity and the thiobarbituric acid reactive substances level in skin homogenates. Therefore, MCH was demonstrated to have the protective effects on chronological skin aging due to the influence on collagen matrix homeostasis. And the antioxidative property of MCH might play an important role in the process.
Low molecular polypeptide from oyster hydrolysate recovers photoaging in SKH-1 hairless mice

Abstract:When the human skin is chronically exposed to external stimuli such as ultraviolet (UV) radiation, the skin tissue suffers damage and the structure of the extracellular matrix (ECM) in the skin is disrupted. This eventually causes symptoms such as wrinkles loss of elasticity, skin sagging, and skin cancer. We previously found that hydrolysate extracted from pacific oyster (Crassostrea gigas) is effective in improving wrinkle formation. In this study, we selected a pentapeptide that was expected to have the most wrinkle reduction effect among the various peptides in oyster hydrolysate through preliminary in vitro screening and examined whether the pentapeptide derived from oyster hydrolysate (OHP) is effective in reducing wrinkles in vivo. We investigated the wrinkle-reducing effect of the OHP through 18-week SKH-1 hairless mice model. Our results showed that the OHP reduces wrinkles lengths, depths, and epidermal thickness which were increased by UVB radiation, and restores the amount of collagen. The OHP recovered the activity of antioxidant enzymes and regulated the expression of proinflammatory cytokines. We also found that OHP increases the expression of type I collagen through stimulating the TGFβ/Smad signaling pathway and inhibits the MMPs expression by regulating the MAPK/AP-1 signaling pathway. This study has shown that the OHP plays crucial roles in collagen production and wrinkle reduction in hairless mice and we proved the possibility of the OHP as a component for inhibiting wrinkle formation which was induced by photoaging.

Abstract:The effect of diet on skin aging has become an interesting research topic. Previous studies have mostly focused on the beneficial effects of collagen peptides derived from marine organisms on the aging skin when administered orally, while the beneficial effects of collagen peptides derived from poultry on aging skin have been rarely reported. In this study, collagen peptides were prepared from chicken bone by enzymatic hydrolysis, and the effect and mechanism of action of orally administered collagen peptides on alleviating skin aging induced by UV combined with D-galactose were investigated. The results showed that the chicken bone collagen had typical characteristics of collagen, and the chicken bone collagen peptides (CPs) were mainly small molecular peptides with a molecular weight of <3000 Da. In vivo experiments showed that CPs had a significant relieving effect on aging skin, indicated by the changes in the compostion and structure of the aging skin, improvement of skin antioxidant level, and inhibition of inflammation; the relieving effect was positively correlated with the dose of CPs. Further investigation showed that CPs first reduce the level of skin oxidation, inhibit the expression of the key transcription factor AP-1 (c-Jun and c-Fos), then activate the TGF-β/Smad signaling pathway to promote collagen synthesis, inhibit the expression of MMP-1/3 to inhibit collagen degradation, and inhibit skin inflammation to alleviate skin aging in mice. Moreover, the skin transcriptome found that lysosomes activated after oral administration of CPs may be an important pathway for CPs in anti-skin aging, and is worthy of further research. These results suggested that CPs might be used as a functional anti-aging nutritional component.
