Abstract:The skeletal muscles of teleost fish encompass heterogeneous muscle types, termed slow-twitch muscle (SM) and fast-twitch muscle (FM), characterized by distinct morphological, anatomical, histological, biochemical, and physiological attributes, driving different swimming behaviors. Despite the central role of metabolism in regulating skeletal muscle types and functions, comprehensive metabolomics investigations focusing on the metabolic differences between these muscle types are lacking. To reveal the differences in metabolic characteristics between the SM and FM of teleost, we conducted an untargeted metabolomics analysis using Pseudocaranx dentex as a representative model and identified 411 differential metabolites (DFMs), of which 345 exhibited higher contents in SM and 66 in FM. KEGG enrichment analysis showed that these DFMs were enriched in the metabolic processes of lipids, amino acids, carbohydrates, purines, and vitamins, suggesting that there were significant differences between the SM and FM in multiple metabolic pathways, especially in the metabolism of energy substances. Furthermore, an integrative analysis of metabolite contents, enzymatic activity assays, and gene expression levels involved in ATP-PCr phosphate, anaerobic glycolysis, and aerobic oxidative energy systems was performed to explore the potential regulatory mechanisms of energy metabolism differences. The results unveiled a set of differential metabolites, enzymes, and genes between the SM and FM, providing compelling molecular evidence of the FM achieving a higher anaerobic energy supply capacity through the ATP-PCr phosphate and glycolysis energy systems, while the SM obtains greater energy supply capacity via aerobic oxidation. These findings significantly advance our understanding of the metabolic profiles and related regulatory mechanisms of skeletal muscles, thereby expanding the knowledge of metabolic physiology and ecological adaptation in teleost fish.

Hypoxia-acclimation adjusts skeletal muscle anaerobic metabolism and burst swim performance in a marine fish

Abstract:Red drum, Sciaenops ocellatus, are a marine teleost native to the Gulf of Mexico that routinely experiences periods of low oxygen (hypoxia). Recent work has demonstrated this species has the capacity to improve aerobic performance in hypoxia through respiratory acclimation. However, it remains unknown how hypoxia acclimation impacts anaerobic metabolism in red drum, and the consequences of exhaustive exercise and recovery. Juvenile fish were acclimated to normoxia (n = 15, DO 90.4 ± 6.42 %) or hypoxia (n = 15, DO 33.6 ± 7.2 %) for 8 days then sampled at three time points: at rest, after exercise, and after a 3 h recovery period. The resting time point was used to characterize the acclimated phenotype, while the remaining time points demonstrate how this phenotype responds to exhaustive exercise. Whole blood, red muscle, white muscle, and heart tissues were sampled for metabolites and enzyme activity. The resting phenotype was characterized by lower pHe and changes to skeletal muscle ATP. Exhaustive exercise increased muscle lactate, and decreased phosphocreatine and ATP with no effect of acclimation. Interestingly, hypoxia-acclimated fish had higher pHe and pHi than control in all exercise time points. Red muscle ATP was lower in hypoxia-acclimated fish versus control at each sample period. Moreover, acclimated fish increased lactate dehydrogenase activity in the red muscle. Hypoxia acclimation increased white muscle ATP and hexokinase activity, a glycolytic enzyme. In a gait-transition swim test, hypoxia-acclimated fish recruited anaerobic-powered burst swimming at lower speeds in normoxia compared to control fish. These data suggest that acclimation increases reliance on anaerobic metabolism, and does not benefit recovery from exhaustive exercise.

Abstract:ABSTRACT Poleward winters commonly expose animals, including fish, to frigid temperatures and low food availability. Fishes that remain active over winter must therefore balance trade-offs between conserving energy and maintaining physiological performance in the cold, yet the extent and underlying mechanisms of these trade-offs are not well understood. We investigated the metabolic plasticity of brook char (Salvelinus fontinalis), a temperate salmonid, from the biochemical to whole-animal level in response to cold and food deprivation. Acute cooling (1°C day−1) from 14°C to 2°C had no effect on food consumption but reduced activity by 77%. We then assessed metabolic performance and demand over 90 days with exposure to warm (8°C) or cold winter (2°C) temperatures while fish were fed or starved. Resting metabolic rate (RMR) decreased substantially during initial cooling from 8°C to 2°C (Q10=4.2–4.5) but brook char exhibited remarkable thermal compensation during acclimation (Q10=1.4–1.6). Conversely, RMR was substantially lower (40–48%) in starved fish, conserving energy. Thus, the absolute magnitude of thermal plasticity may be masked or modified under food restriction. This reduction in RMR was associated with atrophy and decreases in in vivo protein synthesis rates, primarily in non-essential tissues. Remarkably, food deprivation had no effect on maximum oxygen uptake rates and thus aerobic capacity, supporting the notion that metabolic capacity can be decoupled from RMR in certain contexts. Overall, our study highlights the multi-faceted energetic flexibility of Salvelinus spp. that likely contributes to their success in harsh and variable environments and may be emblematic of winter-active fishes more broadly.

Abstract:The concept of behavioural syndromes (i.e. correlations between behavioural traits) has provided an important framework for understanding individual variation in animal behaviour and its link to individual variation in physiology and life-history traits. The pace-of-life syndrome concept posits that behavioural, physiological and life-history traits coevolve in response to correlated selection pressures, and therefore predicts a positive correlation between boldness (i.e. exploration and risk taking) and metabolic capacity for locomotor performance in individuals. We tested for a pace-of-life syndrome linking boldness and metabolic capacity for locomotor activity in juvenile bluegill sunfish, Lepomis macrochirus. Individual fish were screened and classified as bold or shy using an established refuge emergence test. Subsequently, the aerobic and anaerobic metabolisms of bold and shy individuals were quantified using respirometry and by measuring the metabolic by-products of white muscle anaerobic glycolysis following exhaustive exercise, respectively. Bold fish demonstrated 25% greater metabolic scope for activity (i.e. aerobic capacity) than shy fish, which was attributable to a 15% greater maximum metabolic rate. However, there was no significant difference in resting metabolic rate or anaerobic energy expenditure (i.e. anaerobic capacity) between bold and shy fish. These results partially support a pace-of-life syndrome linking boldness and aerobic metabolism in juvenile bluegill sunfish, but did not reveal a link between boldness and anaerobic metabolism. Our findings suggest that aerobic and anaerobic capacities may be subject to different selection pressures, and that physiological processes governing maximum anaerobic performance in fishes are independent from behavioural and physiological traits related to boldness.

Abstract:The physiological consequences of the activation of the immune system in skeletal muscle in fish are not completely understood. To study the consequences of the activation of the immune system by bacterial pathogens on skeletal muscle function, we administered lipopolysaccharide (LPS), an active component of Gram-negative bacteria, in rainbow trout and performed transcriptomic and proteomic analyses in skeletal muscle. We examined changes in gene expression in fast and slow skeletal muscle in rainbow trout at 24 and 72 h after LPS treatment (8 mg/kg) by microarray analysis. At the transcriptional level, we observed important changes in metabolic, mitochondrial and structural genes in fast and slow skeletal muscle. In slow skeletal muscle, LPS caused marked changes in the expression of genes related to oxidative phosphorylation, while in fast skeletal muscle LPS administration caused major changes in the expression of genes coding for glycolytic enzymes. We also evaluated the effects of LPS administration on the fast skeletal muscle proteome and identified 14 proteins that were differentially induced in LPS-treated trout, primarily corresponding to glycolytic enzymes. Our results evidence a robust and tissue-specific response of skeletal muscle to an acute inflammatory challenge, affecting energy utilization and possibly growth in rainbow trout.

Abstract:Fishes' skeletal muscles are crucial for swimming and are differentiated into slow-twitch muscles (SM) and fast-twitch muscles (FM) based on physiological and metabolic properties. Consequently, mitochondrial characteristics (number and morphology) adapt to each fiber type's specific functional needs. However, the mechanisms governing mitochondrial adaptation to the specific bioenergetic requirements of each fiber type in teleosts remain unclear. To address this knowledge gap, we investigated the mitochondrial differences and mitochondrial homeostasis status (including biogenesis, autophagy, fission, and fusion) between SM and FM in teleosts using Takifugu rubripes as a representative model. Our findings reveal that SM mitochondria are more numerous and larger compared to FM. To adapt to the increased mitochondrial number and size, SM exhibit elevated mitochondrial biogenesis and dynamics (fission/fusion), yet show no differences in mitochondrial autophagy. Our study provides insights into the adaptive mechanisms shaping mitochondrial characteristics in teleost muscles. The abundance and elongation of mitochondria in SM are maintained through elevated mitochondrial biogenesis, fusion, and fission, suggesting an adaptive response to fulfill the bioenergetic demands of SM that rely extensively on OXPHOS in teleosts. Our findings enhance our understanding of mitochondrial adaptations in diverse muscle types among teleosts and shed light on the evolutionary strategies of bioenergetics in fishes.

Abstract:Although physiology ensures homeostasis and fitness in a particular environment, and ecological shifts cannot be realized without physiological changes, metabolic transformations during animal adaptive radiations still remain unexplored. We present a study of energy reserve storage in the salmonid assemblage inhabiting a cold-water Lake Kronotskoe. This assemblage diversified from Salvelinus malma and includes eight distinct ecomorphs with contrasting lifestyles and trophic specializations. We hypothesized that ecomorphs differ in energy storage and expenditure, and that their metabolic phenotypes should be among the primary targets of natural selection. To test this hypothesis, we compared the stored amount and ratio of carbohydrates, lipids, circulating peptides that supply the citric acid cycle, as well as proxy indicators of metabolic rate, the blood levels of plasma proteins (including albumin) and hemoglobin. Among ecomorphs, numerous significant differences in physiological parameters were found, closely related to the composition of food, the depth of habitat, and determined by internal factors, probably genetics. Each ecomorph has a specific metabolic phenotype corresponding to its tropho-ecological specialization and lifestyle. Metabolically advanced predators accumulate lipids; littoral insectivorous morphs grow slower and accumulate glycogen; amphipod feeders do not accumulate spare substances; the deepwater consumer of silt benthos differs in the most divergent physiological characteristics. We assume a specific selection on endocrine regulators of energy metabolism during the adaptive radiation of the assemblage, among which the most plausible candidates are thyroid hormones and leptin.

Abstract:There is increasing interest in documenting and explaining the existence of marked intraspecific variation in metabolic rate in animals, with fishes providing some of the best-studied examples. After accounting for variation due to other factors, there can typically be a two to three-fold variation among individual fishes for both standard and maximum metabolic rate (SMR and MMR). This variation is reasonably consistent over time (provided that conditions remain stable), and its underlying causes may be influenced by both genes and developmental conditions. In this paper, current knowledge of the extent and causes of individual variation in SMR, MMR and aerobic scope (AS), collectively its metabolic phenotype, is reviewed and potential links among metabolism, behaviour and performance are described. Intraspecific variation in metabolism has been found to be related to other traits: fishes with a relatively high SMR tend to be more dominant and grow faster in high food environments, but may lose their advantage and are more prone to risk-taking when conditions deteriorate. In contrast to the wide body of research examining links between SMR and behavioural traits, very little work has been directed towards understanding the ecological consequences of individual variation in MMR and AS. Although AS can differ among populations of the same species in response to performance demands, virtually nothing is known about the effects of AS on individual behaviours such as those associated with foraging or predator avoidance. Further, while factors such as food availability, temperature, hypoxia and the fish's social environment are known to alter resting and MMRs in fishes, there is a paucity of studies examining how these effects vary among individuals, and how this variation relates to behaviour. Given the observed links between metabolism and measures of performance, understanding the metabolic responses of individuals to changing environments will be a key area for future research because the environment will have a strong influence on which animals survive predation, become dominant and ultimately have the highest reproductive success. Although current evidence suggests that variation in SMR may be maintained within populations via context-dependent fitness benefits, it is suggested that a more integrative approach is now required to fully understand how the environment can modulate individual performance via effects on metabolic phenotypes encompassing SMR, MMR and AS.

Abstract:We evaluated the effects of body size, water temperature, and sustained swimming activity on swimming performance and the effects of exhaustive exercise on mortality of fasted juvenile rainbow trout. Fasting caused swimming performance to decline more rapidly for small fish than large fish, and warmer water temperatures and sustained swimming activity further decreased swimming performance. Exhaustive exercise increased mortality among fasted fish. Our observations suggest that juvenile rainbow trout with little or no food intake during winter can swim for long periods of time with little effect on mortality, but swimming to exhaustion can enhance mortality, especially among the smallest juveniles.

Abstract:Aquaculture has greater potential for seafood production than wild capture fisheries. To meet the growing demand for seafood, China's marine aquaculture industry has begun building deep-water cages in the open sea. However, under these conditions, fish encounter strong currents and waves, and ensuring their healthy growth is key to the farming process. To address these issues, it is necessary to study the sustained swimming abilities of cultured fish species. Blackhead seabream (Acanthopagrus schlegelii) and red drum (Sciaenops ocellatus) are traditional economic fish species in China; however, their sustained swimming ability under a constant current has been underexplored. Therefore, we examined the endurance swimming ability of three size classes of blackhead seabream and red drum at 20 °C. The fish were then subjected to swimming tests of 0, 30, 60, 90, 120, and 150 min at a constant swimming speed of 0.55 m/s (0.80 m/s), 0.65 m/s (0.90 m/s), and 0.70 m/s (0.98 m/s). The fish were then dissected to obtain muscle, blood, and liver samples; sample metabolite concentrations were measured at six time points, each of which guaranteed five sets of valid data. The results indicated that red drum has a significantly stronger swimming ability, and can be cultured in waters with a short-term flow rate not exceeding 0.75 m/s or 3.5 BL/s. Further, blackhead seabream can be cultured in waters with a flow velocity lower than 0.55 m/s or 2.5 BL/s. The species-related metabolic differences were mainly reflected in the hepatic glycogen and blood glucose concentrations, and those in swimming ability caused by body length were mainly reflected by the hepatic glycogen concentration. The hepatic glycogen concentration had the most significant effect on fish with body lengths >28 cm (P < 0.05). Overall, the experimental results indicated that the liver plays a major role in the physiological level of fish swimming fatigue, providing a direction for further research.

Abstract:The relationship between behavioral thermoregulation and physiological recovery following exhaustive exercise is not well understood. Behavioral thermoregulation could be beneficial for exercise recovery; for example, selection of cooler temperatures could reduce maintenance metabolic cost to preserve aerobic scope for recovery cost, or selection of warmer temperatures could accelerate recovery of exercise metabolites. While post-exercise behavioral thermoregulation has been observed in lizards and frogs, little is known about its importance in fish. We examined the influence of post-exercise recovery temperature on metabolic rate, thermal preference, and metabolite concentrations in juvenile brook char (Salvelinus fontinalis). Fish were acclimated to and exercised at 15 °C, then recovered at either 15 °C or 10 °C while their metabolic rate was measured via respirometry. Metabolite concentrations were measured in fish after exercise at 15 °C and recovery under one of three thermal treatments (to simulate various behavioral thermoregulation scenarios): (i) 6 h recovery at 15 °C, (ii) 6 h recovery at 10 °C, or (iii) 3 h recovery at 10 °C followed by 3 h recovery at 15 °C. Thermal preference was quantified using a static temperature preference system (15 °C vs. 10 °C). Metabolic rates returned to resting faster at 10 °C compared with 15 °C, although at 10 °C there was a tradeoff of delayed metabolite recovery. Specifically, post-exercise plasma osmolality, plasma lactate, and muscle lactate remained elevated for the entire period in fish recovering at 10 °C, whereas these parameters returned to resting levels by 6 h in fish from the other two recovery groups. Regardless, fish did not exhibit clear behavioral thermoregulation (i.e., fish overall did not consistently prefer one temperature) to prioritize either physiological recovery process. The advantage of metabolic rate recovery at cooler temperatures may balance against the advantage of metabolite recovery at warmer temperatures, lessening the usefulness of behavioral thermoregulation as a post-exercise recovery strategy in fish.
Correlates of prolonged swimming performance in F2 hybrids of migratory and non-migratory threespine stickleback

Abstract:Determining which underlying traits contribute to differences in whole-animal performance can be difficult when many traits differ between individuals with high and low capacities. We have previously found that migratory (anadromous marine) and non-migratory (stream-resident) threespine stickleback (Gasterosteus aculeatus) populations have genetically based differences in prolonged swimming performance (U(crit)) that are associated with divergence of a number of candidate morphological and physiological traits (pectoral fin size and shape, body shape, pectoral muscle and heart size, and pectoral muscle metabolic enzyme activities). Here, we use F2 hybrid crosses to determine which traits are correlated with U(crit) when expressed in a largely randomized genetic background and a range of trait values for other divergent traits. We found that four of our 12 candidate traits were positively correlated with U(crit) in F2 hybrids and that the combined effects of ventricle mass, pectoral adductor mass and adductor citrate synthase activity accounted for 17.9% of the variation in U(crit). These data provide additional support for a causal role of muscle and heart size in mediating intraspecific differences in U(crit), but indicate that many candidate morphological and biochemical traits do not have a strong effect on U(crit) when disassociated from other divergent traits. However, the limited variation in U(crit) in our F2 hybrid families may have decreased our ability to detect correlations among these candidate traits and U(crit). These data suggest that many traits, interactions among traits and traits not measured in this study affect prolonged swimming performance in threespine stickleback.

Abstract:The role of blood-borne glucose in the restoration of white muscle glycogen following exhaustive exercise in the active, pelagic rainbow trout (Oncorhynchus mykiss) and the more sluggish, benthic winter flounder (Pseudopleuronectes americanus) were examined. During recovery from exhaustive exercise, the animals were injected with a bolus of universally labelled [14C]glucose via dorsal aortic (trout) or caudal artery (flounder) catheters. The bulk of the injected label (50-70%) remained as glucose in the extracellular fluid in both species. The major metabolic fates of the injected glucose were oxidation to CO2 (6-8%) and production of lactate (6-8%), the latter indicative of continued anaerobic metabolism post-exercise. Oxidation of labelled glucose could account for up to 40% and 15% of the post-exercise MO2 in trout and flounder, respectively. Exhaustive exercise resulted in a reduction of muscle glycogen stores and accumulation of muscle lactate. Glycogen restoration in trout began 2-4h after exercise, whereas in flounder, glycogen restoration began within 2h. Despite a significant labelling of the intramuscular glucose pool, less than 1% of the infused labelled glucose was incorporated into muscle glycogen. This suggests that blood-borne glucose does not contribute significantly to the restoration of muscle glycogen following exhaustive exercise in either trout or flounder and provides further evidence against a prominent role for the Cori cycle in these species.

The effects of reduced food, as a putative environmental cue, were examined in two populations: one that exhibits high rates of anadromy (sea-migration) in nature, and one that does not exhibit anadromy in nature. Juveniles derived from wild-caught parents were reared for 2 years under four environmental treatments: low food in years 1 and 2 (Low-Low); high food in years 1 and 2 (High-High), low food in year 1 and high in year 2 (Low-High), and vice versa (High-Low). Food restriction had a significant effect on migratory tactics, with the frequency of smolts (juveniles choosing migration) highest in the Low-Low treatment in both populations. No individuals became smolts in the High-High treatment, and intermediate smolting rates were observed in the Low-High and High-Low treatments. Higher overall smolting rates in the naturally anadromous population suggested an inherited component to anadromy/migration decisions, but both populations showed variability in migratory tactics. Importantly, some fish from the naturally non-anadromous population became smolts in the experiment, implying the capacity for migration was lying "dormant," but they exhibited lower hypo-osmoregulatory function than smolts from the naturally anadromous population. Tactic frequencies in the naturally anadromous population were more affected by food in the 2nd year, while food in the 1st year appeared more important for the naturally non-anadromous population. Migratory tactics were also related to sex, but underpinned in both sexes by growth in key periods, size, and energetic state. Collectively these results reveal how migration decisions are shaped by a complex interplay between extrinsic and intrinsic factors, informing our ability to predict how facultatively migratory populations will respond to environmental change.

Abstract:Bioenergetics models for fishes are useful for understanding ecological processes (e.g., survivorship, growth, and reproduction) and can also inform fisheries management. Yet, current bioenergetics models are unable to ascertain direct energetic costs associated with standard and active metabolism for wild, free-swimming fishes. The use of telemetry with accelerometer sensors, calibrated in the laboratory using swim tunnel respirometers, have made it possible to estimate field metabolic activity in wild fish. Our objectives were to determine seasonal thermal habitat use and habitat-dependent metabolic costs associated with standard, active, and maximum metabolism in a ~1400 ha multibasin lake in Québec, Canada. We implanted 47 wild, free-swimming Lake Trout ( Salvelinus namaycush ) with either acoustic transmitters equipped with temperature and depth sensors or an acceleration sensor. Three sets of water temperature loggers (one set for each basin) were deployed at 2, 4, 6, 10, and 18 or 20 m to measure seasonally available thermal habitat. Thermal profiles of lake water temperature varied among basins with the thermocline being ~5 m in the smallest basin (north basin) and ~7.5 m in the largest basin (east basin). Thermal habitat used by Lake Trout varied seasonally, coupled with seasonal and basin differences in standard and maximum metabolism. Daily active metabolism loosely followed seasonal changes in thermal habitat use but was largely unaffected by differences in thermal habitat use among capture basins. The theoretical scope-for-activity followed seasonal trends and was estimated to range between 47% and 74% of theoretical aerobic scope. Our observations suggest that available thermal habitats influence Lake Trout thermal habitat use, and thus metabolic costs associated with swimming. These changes in thermal habitats could have metabolic consequences for individuals in a population, resulting in altered fitness metrics (i.e., survival, growth, and/or reproduction). Reductions in the volume and availability of optimal thermal habitats for Lake Trout are likely to occur under climate change scenarios. Our study indicates that the species can adjust metabolic costs throughout the year even when thermal habitats appear to be limiting, which may inform future evidence-based management decisions.
