Changes of Pathology and MRI in Rat Right Hepatic Lobe by Obstruction of Blood Supply

Abstract:Objective:To discuss the probability of detecting apoptosis and necrosis of hepatic cells in vivo by MRI new technique.Methods:90 rats with body weight 250~300g were randomly divided into three groups,with 30 rats in each group.Group A had ligation of the right portal vein for detecting apoptosis.Group B had ligation of both the right hepatic artety and portal vein for the aim of detecting necrosis.Group C had no obstruction of blood supply,just for control and comparison.Six rats from each group were observed at 3h,1d,3d,7d,14d after operation.All rats of each group were examined by MRI,DWI,MRS and PWI.The right hepatic lobes were sampled and examined for pathology and electronic microscopy.MRI and the results of pathology were analyzed by comparison.Results:For group A,cell apoptosis in rat hepatic right lobe was proved by pathology and electronic microscopy.For group B,coagulative necrosis was found in right hepatic lobe.There were some changes between group A and group B,including the intensity of T_1 higher,the intensity of T_2 lower at first and higher at other time,the intensity of DWI higher,the intensity of ADC lower,the peak of Cho,Cr and Glx a little lower,the peak of Lac and Lip higher in MRS,and the area under curve in PWI lower.For group B,there were some changes,including the intensity of T_1 weighted imaging lower,the intensity of T_2 higher,the intensity of DWI higher,the peak of Cho apparently decreasing,the peak of Cr and Glx a little lower,the peak of Lac apparently increasing in MRS and the curve in PWI flater.Conclusion:Hepatic cell apoptosis and necrosis can be gained by obstruction of blood supply of the right hepatic lobe.There are special changes in DWI,MRS,PWI,so cell apoptosis and necrosis can be detected in vivo by them.
Investigation of liver cell apoptosis following iigation of the right portal vein in rats

Abstract:Objective To appraise the pathological changes of liver cells after ligation of the right portal vein. Methods 78 S-D rats were randomly divided into control group,right hepatic artery and right portal vein ligated group(HA+PV group),right portal vein ligated group(PV group).Rats of each group were sacrificed and sam- pled at 3h,12h,24h,3d,7d,14d after operation.The apoptosis and proliferation of hepatic cells were ob- served and quantificational analysis was performed by HE staining,electron microscope,TUNEL and immunohis- tochemical techniques.Results In HA+PV group,coagulation necrosis was found in right hepatic lobe,with nec- rosis of liver cells found in each subsections,accompanied by a few apoptotie cells in the early periods.In PV group,right hepatic lobe atrophied,apoptosis was found at 3h,reaching peak at 24h and declined thereafter.The left lobe became hypertrophyied.PCNA-LI increased and reached the maximum at 14d.Conclusions Ligation of the right portal vein induces hepatocyte apoptosis in the right hepatic lobe and proliferation in the left lobe,which are more significant at 24h,14d respectively.As a result,we have developed an ideal animal model for investiga- tion by functional MRI to reveal hepatoeyte apoptosis and proliferation.
Diffusion weighted imaging features after hepatic right portal vein ligation in rats

Abstract:Objective:To study the diffusion weighted imaging (DWI) characters of hepatic cell focal apoptosis.Materials and Methods:Thirty SD rats about 300g are operated by hepatic right portal vein ligation and divided into 5 groups.T1WI,T2WI,DWI and apparent diffusion coefficient (ADC) image were obtained at 3 hours,1 day,3 days,7 days,14 days after the operation.After MRI examination,rats were sacrificed and the right hepatic lobes were sampled for pathology,electronic microscopy and TUNEL staining examination.Results:All right hepatic lobe focal apoptosis in 30 rats was certificated by pathology,electronic microscopy and TUNEL staining examination.Occurring from 3 hours after operation,apoptosis cell gradually increased with time.It was high signal at T1-weighted image in each group after operation,low signal at T2-weighted image after 3 hours and high signal in other groups,and slightly high signal at diffusion weighted image after 3 hours,and high signal in other periods.The ADC value of the right lobe was less than the left lobe in each group (P<0.05),the relative ADC value increased in all groups with time.The ADC value,signal-noise ratio (SNR) and sensitivity of DWI decreased with b value increasing,but the specificity increased.Conclusion:There are focal apoptosis in right hepatic lobes of rats after the portal vein ligation.What's more,its signal in DWI image and the ADC value is unique,so DWI is useful to judge the success of hepatic vessel ligation and detect focal apoptosis area in vivo.

Abstract:Objective To study the performance and value of MR spectroscopy(MRS) in detecting and characterizing hepatic necrosis and apoptosis in vivo by a comparison with pathological findings in animal model after ligation of hepatic blood-supplying vessels.Methods Seventy-two SD rats were randomly divided into three groups:a sham operated control group,a necrosis group with both right hepatic artery and right portal vein ligated(HA+PV group),and an apoptosis group with only right portal vein ligated(PV group).There were 4 sub-groups(6 rats each) in the HA+PV group or PV group.The rats in each group were examined by MRI and Single Voxel PRESS 35 sequence for spectroscopic analysis.The peak and area of metabolites were measured and the ratios between right and left lobes were calculated.Results At 3 h in both HA+PV group and PV group, compared with left lobe,the Lip-Lac peak of right hepatic lobe increased obviously with few necrotic and apoptic cells.Along with increasing necrotic and apoptic cells,the Lip-Lac peak of right hepatic lobe increased.Similarly,Cho of the right hepatic lobe decreased at 12 h after operation and more obviously thereafter.The ratios of Lip-Lac/Cho of right lobe increased in both HA+PV and PV groups at 3 h after operation,and more obviously in the PV 3 h sub-group.Conclusions There are an increase of Lip-Lac peak and a decrease of Cho in either necrotic or apoptic tissue.The ratio of Lip-Lac/Cho increases more obviously in apoptic tissue than in necrotic tissue in very early stage and has significance in differentiating acute necrosis and apoptosis.
Experimental MRI Study on Differentiation of Necrosis and Apoptosis of Hepatic Tissue

Abstract:A series of advanced MRI sequences including T 1 WI,T 2 WI,DWI,and ADC mapping were con-ducted on rat models of hepatic necrosis and apoptosis.The morphological appearances and signal characteristics were collected to demonstrate the diagnostic ability and the differential features of the advanced MRI technology for hepatic necrosis and apoptosis.Materials and Methods:Seventy-eight rats were randomly divided into13groups,6rats in each group.Rats in group A1,B1,C1,D1,E1,、F1were enrolled for the groups of hepatic necrosis where both right hepatic and portal vein were ligated.Rats in group A2,B2,C2,D2,E2,F2for hepatic apoptosis by ligating only the right portal vein.Six rats were sham-operated for control study.Rats in group A(including A1and A2),B,C,D,E,F were exmanined at3h,12h,24h,3d,7d,14d after the surgical operation of ligation.The rats were sacrificed immediately after MRI examination and the hepatic tissue was harvested.The hepatic samples were further examined by HE stain,TUNEL stain,Envision PCNA stain,and electronic microscopy.For each rat,conventional axial T 1 WI,T 2 WI,coronal T 2 WI sequences were conducted initially,followed by ADC mapping and DWI imaging.Results:The success rate of anesthesia and surgery is93.8%.Pathological studies showed hepatic coagulative necrosis after the ligation of both right hepatic artery and portal vein.When only right portal vein was ligated,apoptosis process can be seen at3hrs after the surgery,with a peak at24th hour.In the group of hepatic necrosis,DWI showed a significant high signal right lobe at early stage.On conventional images of T 1 WI,T 2 WI,the right lobe was long T 1 and long T 2 manifestations.Distinguishing high signal intensities on T1WI were observed in the right lobes of the rats in group A2 ,B2 , C2 . No signal changes were seen on T2WI . Blotch or patchy high signal was revealed on T2WI ingroup D2 and E2 .On DWI ,right lobes were low signal intensity in early stage and iso -density or spothigh signal intensity after 3d .Conclusion:There were significant different signal changes on conven-tional T1WI and T2WI on the groups of necrosis and apoptosis . Combined with the findings on DWI andADC mapping ,it is possible and practical to detect and differentiate the two pathological processes :hepatic necrosis and apoptosis .
The Effect of Hepatic Blood Inflow Occlusion on Hepatic Cancer Treated with Diode-Laser Thermocoagulation

Abstract:OBJECTIVE:To assess the effect of temporary occlusion of hepatic blood inflow on hepatic cancer treated with diode-laser induced thermocogation (LITT). METHODS:The carcinoma Walker-256 was implanted in 40 SD rat livers. Twelve days later, the animals were randomly divided into 4 groups. Group A received LITT alone; group B received hepatic artery temporary occlusion during LITT; group C received portal vein temporary occlusion during LITT; group D received hepatic artery and portal vein temporary occlusion during LITT. Tumors were exposed to 810 nm diode-laser light at 0.95 watts for 10 min from a scanner tip applicator placed in the tumor. At the same time, the intrahepatic temperature distribution in rats with liver tumors was measured per 2 min during thermocoagulation. Tumor control was examined immediately 7 and 14 d after thermocoagulation. RESULTS:There was significant difference of intrahepatic temperature distribution in rats with liver tumors among the 4 groups (P<0.05) except when group C samples were compared with group D samples at each time point, and group B samples were compared with group C samples at 120 s (P>0.05). Light microscopic examination of the histologic section samples revealed three separate zones: regular hyperthermic coagulation necrosis zone, transition zone and reference zone. Compared with the samples in group A and group B, group C and group D samples had more clear margin among the three zones. CONCLUSION:The hepatic blood inflow occlusion, especially portal vein hepatic blood inflow occlusion, or all hepatic blood inflow occlusion considerably increased the efficacy of LITT in the treatment of liver cancer.
Expression of Hedgehog Signaling Molecules in Rat Livers after Partial Hepatectomy

Abstract:Objective To investigate the effect of ischemia and reperfusion on expression of Hedgehog signaling in the liver after rat partial hepatectomy.Methods The model of rat partial hepatectomy was established by resecting the middle and left lobes of the liver.Eighteen Sprague-Dawley rats were randomly divided into 3 groups:sham operation (A),oartial hepatectomy (B),partial hepatectomy combined with hepatic ischemia and reperfusion (C).Blood and hepatic tissues were collected 48 h after the operation Plasma alanine transaminase (ALT) and the expression of proliferation cell nuclear antigen ( Ki-67 ),Sonic hedgehog ( Shh),Indian hedgehog (Ihh) and glioma-assocated oncogene-2 (Gli-2) in the hepatic tissue were detected.Results As compared with group A,the plasma ALT was increased in groups C (P ＜ 0.01 ) and B (P ＞0.05 ).As compared with group B,the plasma ALT was increased in group C (P＜0.01 ).Adipose degeneration and cells at different division stages were shown in hepatic tissue of groups B and C.As compared with group A,the expression of Ki-67,Shh,Ihh and Gli-2 in groups B and C was increased significantly ( P ＜ 0.01 ).and that was higher in group C than in group B ( P ＜ 0.05 ).Conclusion Hedgehog signaling may be activated after rat partial hepatectomy,which can promote the liver regeneration.Ischemia and reperfusion may enhance the activation of Hh signaling after partial hepatectomy.

Evaluation of MRI after Blood Supply Blockage at Right Hepatic Lobes of Rats

Abstract:Objective To evaluate the sensitivities of different MR imagings in revealing hepatic ischemia.Methods S-D rats were divided radomly into A,B and C groups.Right hepatic artery and right portal vein were ligated in A group; right portal vein was ligated only in B group; and C was control group.A and B were divided into 3 subsections respectively.MRI was examined after 12、24 h and 72 h,respectively.Results Two different ligations resulted in different changes in both morphology of hepatic roght lobe and signals on T1WI,T2WI and DWI.In Group A, hepatic right lobes resulted in coagulation necrosis with no significant morphological change,but distinct hyperintensity on T2WI and DWI appeared.In Group B, after 24h and 72 hof ligations, right lobes were atrophied and darkled, the rest lobes became hypertrophied.On T1WI and T2WI, right lobes showed isointensity or slight hypointensity, but distinct hypointensity on DWI.Conclusion In Groups A and B, hepatic lobes show different signals on T2WI and DWI.DWI is more sensitive and exact than T2WI.
Establishment and Evaluation of the Animal Models of Hepatic Apoptosis and Necrosis in Rats

Abstract:Objective:To establish rat models of hepatic cell apoptosis and necrosis by intervention of blood supply of the right hepatic lobe. Methods:90 SD rats with body weight 250-300g were randomly divided into three groups, with 30 rats in each group. Group A had ligation of both the right hepatic artery and portal vein for the aim of necrosis. Group B had ligation of the right portal vein for apoptosis. Group C had no intervention of blood supply for control and comparison. Six rats from each group were sacrificed at 3h,1d,3d,7d and 14d after operation,and the right hepatic lobes were sampled and examined using light microscopy and electronic microscopy. Results: All of the rats from group A had typical coagulative necrosis in right hepatic lobe under light microscopy and electron microscopy, and were negative on TUNEL staining. In group B rats,focal apoptosis in right hepatic lobe could be revealed. There were pyknosis under light microscope; karyon bound gathering and apoptosis small body under electron microscopy. Results of TUNEL staining were positive. In the right lobe,apoptosis could be seen at 3h,with a peak at 24h;focal necrosis accompanied apoptosis could be revealed at 7d after surgery. All thirty rats from group C showed no signs of necrosis and apoptosis in the right hepatic lobe. Conclusion: Right hepatic lobe coagulative necrosis can be obtained in rat models by ligation of right portal vein and hepatic artery,focal apoptosis can be obtained simply by right portal vein ligation in rat models.
[Preliminary Experimental Study on Treatment of Portal Hypertension with Auxiliary Partial Orthotopic Liver Transplantation].

Abstract:OBJECTIVE:To evaluate the therapeutic efficacy of auxiliary partial orthotopic liver transplantation (APOLT) on portal hypertension in liver cirrhosis with amelioration of portal vein congestion, changes in portal vein pressure and status of the graft.METHODS:The recipients were porcine model of biliary cirrhosis reproduced by ligation of the common bile duct. During transplantation, arterial blood pressure, central venous pressure were recorded. Buffer base, standard bicarbonate and pH of arterial blood samples were determined and analyzed in order to assess the impact of operation on the animals. The hemodynamics were also monitored. Color Doppler ultrasonographic examination was performed on recipients before operation, intra-operation and 7 days after operation, respectively. Portal vein pressure, blood bilirubin and variables of liver function were measured by using an autoanalyzer. Wedge biopsy specimens of each pig were obtained, stained with hematoxylin-eosin, and examined. Analysis of variance was performed. Otherwise non-parametric tests were used.RESULTS:Eight weeks after ligation of the common bile duct, biliary cirrhosis was reproduced in all the pigs, and histopathological examination of the liver specimen showed a large number of pseudo-lobules. In 6 pigs with hepatic cirrhosis liver transplantation was done. Five of the 6 (83.3%) animals survived for 7 days. One recipient died because of unsuccessful operation, the others showed stable hemodynamics during the operation. Seven days after transplantation, the blood flow of the two liver portal veins was observed by the use of color Doppler ultrasonography. It was found that the blood flow in the donor portal vein was much richer than that of native portal vein. The venous outflow of the graft was ample and smooth, and no thrombosis of the hepatic vein or portal vein was found. The variables, including alanine aminotransferase, aspartate aminotransferase, bilirubin and total bilirubin, were significantly improved 7 days after operation compared with pre-operative data. Portal pressure was found to be (20.76+/-2.42)cm H(2)O(1 cm H(2)O=0.098 kPa), (17.62+/-2.33)cm H(2)O and (14.72+/-2.25)cm H(2)O before the operation, during the operation, and 7 days after operation, respectively, and the difference was statistically significant(P<0.01). On the 7 days after transplantation, histopathological examination revealed evidence of damage with mild steatosis and sporadic necrotic hepatocytes and focal hepatic lobular degeneration in the graft, especially in the area around the central vein.CONCLUSION:APOLT is a hopeful option for the treatment of portal hypertension. This procedure can provide not only some improvement of the liver function but also decrease the pressure of portal vein.
Extended Ligation of the Hepatic Vein May Yield a Similar Effect to Liver Venous Deprivation in a Rat Model

Abstract:To validate the hypothesis that hepatic vein ligation (HVL) alone may produce similar results to liver venous deprivation (LVD or HVL + portal vein ligation [PVL]). Rats were assigned to five groups, namely, the control group; the R group in which the right median hepatic vein (RMHV) was ligated; the M group in which the middle median hepatic vein (MMHV) was ligated; the RM group in which both the RMHV and MMHV were ligated (R + MMHVL, extended ligation of the hepatic veins); and the LVD group in which both the right median portal vein and the RMHV were ligated. The liver hypertrophy effect and liver enzymes were determined. Methylene blue staining and retrograde pressurized perfusion assays were performed to investigate the hemodynamic changes. The RM and LVD groups exhibited similar significant hypertrophy in the future liver remnants when compared to that of the control group, and almost no additional hypertrophy effect was observed in the R and M groups. There was a remarkable elevation in serum transaminase levels in both groups. The methylene blue staining experiment indicated that pressure-dependent collaterals formed between the contiguous drainage areas, and the R + MMHVL procedure blocked the outflow of the right median lobe. Extended ligation of the hepatic vein (R + MMHVL) resulted in a similar hypertrophy effect and hepatic damage to those of LVD (HVL + PVL) treatment in a rat model, and intrahepatic venovenous collaterals play key roles.

Multiparameter magnetic resonance imaging of liver fibrosis in a bile duct ligation mouse model

Abstract:BACKGROUND: Bile duct ligation (BDL) in animals is a classical method for mimicking cholestatic fibrosis. Although different surgical techniques have been described in rats and rabbits, mouse models can be more cost-effective and reproducible for investigating cholestatic fibrosis. Magnetic resonance imaging (MRI) has made great advances for noninvasive assessment of liver fibrosis. More comprehensive liver fibrotic features of BDL on MRI are important. However, the utility of multiparameter MRI to detect liver fibrosis in a BDL mouse model has not been assessed.AIM: To evaluate the correlation between the pathological changes and multiparameter MRI characteristics of liver fibrosis in a BDL mouse model.METHODS: Twenty-eight healthy adult male balb/c mice were randomly divided into four groups: sham, week 2 BDL, week 4 BDL, and week 6 BDL. Multiparameter MRI sequences, included magnetic resonance cholangiopancreatography, T1-weighted, T2-weighted, T2 mapping, and pre- and post-enhanced T1 mapping, were performed after sham and BDL surgery. Peripheral blood and liver tissue were collected after MRI. For statistical analysis, Student's t-test and Pearson's correlation coefficient were used.RESULTS: Four mice died after BDL surgery; seven, six, five and six mice were included separately from the four groups. Signal intensities of liver parenchyma showed no difference on TI- and T2-weighted images. Bile duct volume, ΔT1 value, T2 value, and the rate of liver fibrosis increased steadily in week 2 BDL, week 4 BDL and week 6 BDL groups compared with those in the sham group (P < 0.01). Alanine aminotransferase and aspartate transaminase levels initially surged after surgery, followed by a gradual decline over time. Strong correlations were found between bile duct volume (r = 0.84), T2 value (r = 0.78), ΔT1 value (r = 0.62), and hepatic fibrosis rate (all P < 0.01) in the BDL groups.CONCLUSION: The BDL mouse model induces changes that can be observed on MRI. The MRI parameters correlate with the hepatic fibrosis rate and allow for detection of cholestatic fibrosis.

Abstract:BackgroundPartial bile duct ligation (PBDL) model is a reliable cholestatic fibrosis experimental model that showed complex histopathological changes. Magnetic resonance imaging (MRI) features of PBDL have not been well characterized. PurposeTo investigate the potential of MRI parameters in assessing fibrosis in PBDL and explore the relationships between MRI and pathological features. Animal ModelEstablished PBDL models. PopulationFifty-four mice were randomly divided into four timepoints PBDL groups and one sham group. Field Strength/Sequence3.0 T; MRI sequences included T1-weighted fast spin-echo (FSE), T2-weighted single shot FSE, variable flip angle T1 mapping, multi-echo SE T2 mapping, multi-echo gradient-echo T2* mapping, and multi-b-value diffusion-weighted imaging. AssessmentMRI examination was performed at the corresponding timepoints after surgery. Native T1, ΔT1 (T1native-T1post), T2, T2*, apparent diffusion coefficient (ADC) values, histogram parameters (skewness and kurtosis), intravoxel incoherent motion parameters (f, D, and D*) within the entire ligated (PBDL), non-ligated liver (PBDL), and whole liver (sham) were obtained. Fibrosis and inflammation were assessed in Masson and H&E staining slices using the Metavir and activity scoring system. Statistical TestsOne-way ANOVA, Spearman's rank correlation, and receiver operating characteristic curves were performed. P < 0.05 was considered statistically significant. ResultsFibrosis and inflammation were finally staged as F3 and A3 in ligated livers but were not observed in non-ligated or sham livers. Ligated livers displayed significantly elevated native T1, ΔT1, T2, and reduced ADC and T2* than other livers. Spearman's correlation showed better correlation with inflammation (r = 0.809) than fibrosis (r = 0.635) in T2 and both ΔT1 and ADC showed stronger correlation with fibrosis (r = 0.704 and r = −0.718) than inflammation (r = 0.564 and r = −0.550). Area under the curve (AUC) for ΔT1 performed the highest (0.896). When combined with all relative parameters, AUC increased to 0.956. Data ConclusionMultiparametric MRI can evaluate and differentiate pathological changes in PBDL. ΔT1 and ADC better correlated with fibrosis while T2 stronger with inflammation. Comparative Research of Right Portal Vein Embolization and Ligation in SD Rats

Abstract:Objective　To evaluate the morphological and functional changes after proximal complete block by ligation or distal embolization by Lipiodol-ethonal 2∶1 mixture at the right portal vein branch.Methods　Twenty-five SD rats were randomly divided into 5 groups,5 rats each. Right portal veins were distally embolized by Lipiodol-ethonal 2∶1 mixture in group 1、2 rats and were ligated in group 3 and 4. Five rats served as a control group. Rats of group 1、3 and rats of group 2、4 were sacrificed to inspect the morphological changes of each lobe at 14 and 21 s days after the procedure. Each lobe of the liver was dissected and weighted to obtain the weight ratios of the right lobe to the whole liver and the whole liver to the body. The results of four groups were analyzed and compared to each other and to those of the control group using statistical Student “t” test.Results　At 14 or 21 days after the procedure, the right lobes of the rats in group 1 and 2 were atrophied apparently with lots of spotty yellow scars in different size. In the rats from group 3 and 4, the right lobes were significantly atrophied but the morphology had no significant change. By statistical analysis, no significant difference was found between the embolization group and the ligation group (P>0.05). Conclusion　Right portal vein branch embolization using Lipiodol-ethanol 2∶1 mixture and proximal ligation can induce similar extent of atrophy of the lobe although the morphology was different. The rest lobes of the liver become hypertrophied and the weight of the whole liver remains the same although the right lobe is atrophied.
Assessment of Liver Regeneration after Associating Liver Partition and Portal Vein Ligation for Staged Hepatectomy: a Comparative Study with Portal Vein Ligation.

Abstract:Background: To investigate the diagnostic value of diffusion kurtosis imaging (DKI) and diffusion-weighted imaging (DWI) in assessing liver regeneration after associating liver partition and portal vein ligation for staged hepatectomy (ALPPS) compared with portal vein ligation (PVL). Methods: Thirty rats were divided into the ALPPS, PVL, and control groups. DKI and DWI were performed before and 7 days after surgery. Corrected apparent diffusion (D), kurtosis (K) and apparent diffusion coefficient (ADC) were calculated and compared, radiologic-pathologic correlations were evaluated. Results: The volume of the right median lobe increased significantly after ALPPS. There were larger cellular diameters after ALPPS and PVL (P = 0.0003). The proliferative indexes of Ki-67 and hepatocyte growth factor were higher after ALPPS (P = 0.0024/0.0433). D, K and ADC values differed between the groups (P = 0.021/0.0015/0.0008). A significant correlation existed between D and the hepatocyte size (r = -0.523), no correlations existed in ADC and K (P = 0.159/0.111). The proliferative indexes showed moderate negative correlations with ADC (r = -0.484/-0.537) and no correlations with D and K (P = 0.100-0.877). Discussion: Liver regeneration after ALPPS was effective and superior to PVL. DKI, especially the D map, may provide added value in evaluating the microstructure of liver regeneration after ALPPS, but this model alone may perform no better than the standard monoexponential model of DWI.
MRI Appearance of Focal Apoptosis in Rat Liver Tumor

Abstract:Purpose:To establish tumor apoptosis model in rat liver cell and to study its appearances on MRI.Methods:Forty-five SD rats were randomly divided into three groups:radiation group,injection group with alcohol and no therapy group.All the rats in each group were observed at 1D,3D,7D after operation and were examined with conventional MRI,DWI,and MRS.The ADC value and the peak value of the metabolites in MRS were measured and analyzed.After that,the rats were sampled for electronic microscopy and pathology exam.HE staining and TUNEL,PCNA,CD34 and VEGF staining were done. The results of MRI and pathology were compared and analyzed.Results:①Apoptosis tumor cells were detected by pathology examination in radiation group,necroses were found in injection group with alcohol.②There were some changes between radiation group and no therapy group in MRI,DWI,and MRS, including the decrease of tumor volume,the increase of signal of MRI,the decrease of intension in DWI, the increase of value of ADC,the decrease of peak value of Cho,Glx,Lac,Lip in MRS.③In injection group with alcohol,the tumor volumes were decreased,and accompany with the decrease of signal in central of tumor,the decrease of local intension in DWI,the decrease of peak value of Cho,Glx,Lac and Lip in MRS.Conclusion:The apoptosis model in rat liver tumor would be made by radiation.There were some characteristic appearances of apoptosis in MRI,especially with T_2WI,DWI and MRS.The apoptosis could be monitored in vivo by MRI.
Partial Portal Vein Arterialization Attenuates Acute Bile Duct Injury Induced by Hepatic Dearterialization in a Rat Model

Abstract:Hepatic infarcts or abscesses occur after hepatic artery interruption. We explored the mechanisms of hepatic deprivation-induced acute liver injury and determine whether partial portal vein arterialization attenuated this injury in rats. Male Sprague-Dawley rats underwent either complete hepatic arterial deprivation or partial portal vein arterialization, or both. Hepatic ischemia was evaluated using biochemical analysis, light microscopy, and transmission electron microscopy. Hepatic ATP levels, the expression of hypoxia- and inflammation-associated genes and proteins, and the expression of bile transporter genes were assessed. Complete dearterialization of the liver induced acute liver injury, as evidenced by the histological changes, significantly increased serum biochemical markers, decreased ATP content, increased expression of hypoxia- and inflammation-associated genes and proteins, and decreased expression of bile transporter genes. These detrimental changes were extenuated but not fully reversed by partial portal vein arterialization, which also attenuated ductular reaction and fibrosis in completely dearterialized rat livers. Collectively, complete hepatic deprivation causes severe liver injury, including bile infarcts and biloma formation. Partial portal vein arterialization seems to protect against acute ischemia-hypoxia-induced liver injury.

Abstract:Background To investigate the role and mechanism of liver parenchyma transection in accelerating the regeneration of future liver remnants in rats with portal vein ligation (PVL). Methods Rats were randomly divided into the PVL group (90% PVL at the caudate lobe, right lobe , left lateral lobe and left median lobe), associating liver partition and portal vein ligation for staged hepatectomy (portal vein ligation with complete liver parenchyma transection [ALPPS]) group (90% PVL with 80 to 90% liver parenchyma transection), PVL + partial liver partition (PLP) group (90% PVL with 30 to 50% liver parenchyma transection), PVL + partition in the ligated lobe (PLL) group (90% PVL with 40 to 60% liver parenchyma transection in the portal vein ligated lobe), PVL + partition in the remnant lobe (PRL) group (90% PVL with 40 to 60% liver parenchyma transection in the remnant lobe), PVL + radiofrequency ablation (RFA) group (90% PVL with splenic ablation) and sham operation (sham) group. The animals were killed at 4 time points of postoperative days 1, 3, 5, and 7. Six rats were killed at each time point, with 24 rats in each group. The weights of the future liver remnant and whole liver were measured. Serum alanine aminotransferase, aspartate aminotransferase, and total bilirubin were analyzed by using an automatic biochemical analyzer. Serum tumor necrosis factor-α, interleukin-6, and hepatocyte growth factor were measured by enzyme-linked immunosorbent assay. The expression of cell proliferating nuclear antigen (Ki67) and phosphorylated histone H3 was detected by immunohistochemistry, and the positive rate was calculated. Results The ALPPS group displayed the highest FLR weight to body weight ratio compared with that of the other groups ( P < .05), and the partial liver split (PVL + PLP) group also displayed higher remnant weight to body weight ratio than the ectopic liver split (PVL + PLL and PVL + PRL) groups ( P < .05). During the first 7 days after surgery the cytokine levels of the ALPPS, PVL + PLP, PVL + PLL and PVL + PRL groups were comparable ( P > .05). The PVL + PLP, PVL + PLL, PVL + PRL and PVL + RFA groups showed similar necrotic areas in the portal vein ligated lobe ( P > .05). A hemodynamic study revealed that a liver split along the demarcation line could further increase the portal pressure of the FLR and both the split site and completeness were associated with portal hemodynamic alternations and liver hypertrophy. Extrahepatic organ injury (eg, spleen ablation) also has a significant impact on portal hemodynamics and liver regeneration. Conclusion Complete liver splitting along the demarcation line induced higher portal vein pressure and more rapid FLR hypertrophy than partial or ectopic liver splitting after PVL. The portal hemodynamic alterations after liver split rather than inflammatory cytokine release may be the major cause of ALPPS-induced rapid liver hypertrophy.

Abstract:Objective To evaluate in vivo magnetic resonance imaging with a conventional 1.5-T system for tracking og intra-portal vein transplantation nanometer magnetically labeled BMSCs in rat liver. Methods BMSCs were isolated from 5 SD rats bone marrow with the density gradient centrifugation method. Then BMSCs were labeled with nanometer superpara-magnetic iron oxide and transfection agent. Cell labeling efficiency was assessed with determination of the percentage of Perls Prussian blue stain. Then BMSCs transplanted into normal rats′ livers via portal vein. The receipts were divided into 5 groups ,including sham control、2 h 、3 d、7 d and 2 w after transplantation. Follow-up serial T_1WI、T_2WI and T_2*-weighted gradient-echo MR imaging were performed at 1.5 T MRI system. MR imaging findings were compared with histology. Results Cell labeling efficiency was more than 95% by Perls Prussian blue stain. After transplantation of labeled BMSCs via portal vein, livers had diffuse granular signal intensity appearance in T_2*WI MRI. Cells were detected for up to 2 w in receipts′ livers. At histologic analysis, signal intensity loss correlated with iron-loaded cells. Conclusion MR imaging could aid in monitoring of magnetically labeled BMSCs administered via portal vein in vivo.
