Abstract:Essential oils have been tested as insect repellents with moderate results. The most important characteristics of these oils are that they possess a high chemical diversity of compounds, are cheap, and are easily accessible. Recently, mixtures of essential oils have shown a higher repellent activity than a single essential oil, probably by increasing the effectiveness with the number of chemicals in the mixture. The repellent effect of essential oils from citronella (Cymbopogon nardus), mint (Mentha arvensis), rosemary (Rosmarinus officinalis L.), clove (Eugenia caryophyllata), and their mixture was evaluated against Aedes aegypti. The mixture of citronella, mint, and clove essential oils (1:1:1) was also evaluated in an adulticidal bioassay. Additionally, chemical identification was performed using these commercial essential oils. The 1:1:1 mixture of citronella, mint, and clove essential oils was as effective as the commercial repellent DEET at lower doses (5%), and it is a good candidate for an adulticide at higher doses (20–25%). The chemical constituents of the essential oils were mostly terpenes and oxygenated terpenoids. These findings were discussed in terms of economic and environmental frameworks.

Abstract:This study was performed to provide data on the chemical composition, skin irritant potential and mosquito repellency of Torreya grandis aril essential oil extracted by hydro distillation. Fifty nine compounds were identified through comparison with reference compounds, tentatively identified by comparing mass spectra and Kovats Index with those reported in mass spectra libraries and literature. Monoterpenes and oxygenated monoterpenes represented more than 80 % of the oil. The oil was characterized by high contents of alpha-pinene (34.20 %), D-limonene (27.58 %), limonene glycol (3.64 %), (E)-carveol (2.50 %) and isoborneol (2.36 %). No positive skin irritant reaction was found within the 15 volunteers exposed to undiluted oil (4-h patch test). Furthermore, the ethanol solutions of the essential oil at several concentrations were tested with arm-in-cage assay for mosquito (Aedes aegypti) repellency. Results showed that the ethanol solutions with essential oil at 12.5 %, 25 %, 50 %, and 100 % (the original undiluted essential oil) provided complete protection of 1.0 +/- 0.5, 5.0 +/- 0.5, 7.0 +/- 1.0, and 7.5 +/- 0.5h for human beings, respectively. These results suggest the promising application of the essential oil in natural mosquito repellent production.

Abstract:An on-line electrochemistry/electrospray mass spectrometry system (EC/MS) is described that allows fast analysis of the oxidation products of peptides. A range of peptides was oxidized in an electrochemical cell by application of a potential ramp from 0 to 1.5 V during passage of the sample. Electrochemical oxidation of peptides was found to occur readily when tyrosine was present. Tyrosine was found to be oxidized between 0.5 and 1.0 V to various oxidation products, including peptide fragments formed by hydrolysis at the C-terminal side of tyrosine. The results confirm earlier knowledge on the mechanisms and reaction products of chemical and electrochemical peptide oxidation. Methionine residues are also readily oxidized, but do not induce peptide cleavage. At potentials higher than about 1.1 V, additional oxidation products were observed in some peptides, including loss of 28 Da from the C-terminus and dimerization. The tyrosine-specific cleavage reaction suggests a possible use of the EC/MS system as an on-line protein digestion and peptide mapping system. In addition, the system can be used to distinguish phosphorylated from unphosphorylated tyrosine residues. Four forms of the ZAP-70 peptide ALGADDSYYTAR with both, either or neither tyrosine phosphorylated were subjected to a 0-1.5 V potential ramp. Oxidation of, and cleavage adjacent to, tyrosine was observed exclusively at unphosphorylated tyrosine residues.
Synergistic Repellent and Irritant Effects of a Mixture of β-Caryophyllene Oxide and Vetiver Oil against Mosquito Vectors

Abstract:Repellents play a major role in reducing the risk of mosquito-borne diseases by preventing mosquito bites. The present study evaluated the mosquito-repellent activity of β-caryophyllene oxide 1% (BCO), vetiver oil 2.5% (VO), and their binary mixtures (BCO + VO (1:1), BCO + VO (2:1), BCO + VO (1:2)) against four laboratory-colonized mosquito species, Aedes aegypti (L.), Aedes albopictus (Skuse), Anopheles minimus Theobald, and Culex quinquefasciatus Say, using an excito-repellency assay system. In general, the compound mixtures produced a much stronger response in the mosquitoes than single compounds, regardless of the test conditions or species. The greatest synergetic effect was achieved with the combination of BCO + VO (1:2) in both contact and noncontact trials with An. minimus (74.07–78.18%) and Cx. quinquefasciatus (55.36–83.64%). Knockdown responses to the binary mixture of BCO + VO were observed for Ae. albopictus, An. minimus, and Cx. quinquefasciatus, in the range of 18.18–33.33%. The synergistic repellent activity of BCO and VO used in this study may support increased opportunities to develop safer alternatives to synthetic repellents for personal protection against mosquitoes.

Abstract:Mosquitoes of the genera Aedes , Anopheles and Culex vector a wide range of pathogens seriously affecting humans and livestock on a global scale. Over-reliance on insecticides and repellents has driven research into alternative, naturally-derived compounds to fulfil the same objectives. Steam distilled extracts of four plants with strong, yet attractive, volatile profiles were initially assessed for repellency in a dual-port olfactometer using Aedes aegypti as the model species. Picea sitchensis was found to be the most repellent, proving comparable to leading products when applied at 100% (p = 1.000). Key components of conifer-derived volatile profiles were then screened via electroantennography before those components eliciting an electrophysiological response were assayed individually in the olfactometer; according to WHO protocol. The most promising 5 were selected for reductive analyses to produce an optimised semiochemical blend. This combination, and a further two variations of the blend, were then progressed to a multi-species analysis using the BG-test whereby bite-attempt frequency on hands was assessed under different repellent treatments; assays were compared between Aedes aegypti , Anopheles gambiae and Culex quinquefasciatus . Efficacy was found against all three species, although it was found that Ae. aegypti was the most susceptible to the repellent, with An. gambiae being the least. Here, a novel, naturally-derived blend is presented with weak spatial repellency, as confirmed in laboratory assays. Further work will be required to assess the full extent of the potential of the products, both in terms of field application and species screening; however, the success of the products developed demonstrate that plant metabolites have great capacity for use in the repellent sector; both to improve upon known compounds and to reduce the usage of toxic products currently on the market.

Abstract:AbstractInsecticidal plant-based compound(s)in combinations may show synergistic or antagonistic interactions against insect pest. Considering the rapid spread of theAedesborne diseases and increasing resistance amongAedespopulation against conventional insecticides, twenty-eight combinations of plant essential oil-based terpene compounds were prepared and tested against larval and adult stages ofAedes aegypti. Initially five plant essential oils (EOs) were assessed for their larvicidal and adulticidal efficacy and two of their major compounds from each EO were identified from GC-MS results. Identified major compounds namely Diallyldisulfide, Diallyltrisulfide, Carvone, Limonene, Eugenol, Methyl Eugenol, Eucalyptol, Eudesmol and α-pinene were purchased and tested individually againstA.aegypti. Binary combinations of these compounds were then prepared using sub-lethal doses, tested and their synergistic and antagonistic effects were determined. The best larvicidal compositions were obtained while Limonene was mixed with Diallyldisulfide and the best adulticidal composition was obtained while Carvone was mixed with Limonene. Commercially used synthetic larvicide “Temephos” and adulticide “Malathion” were tested individually and in binary combinations with the terpene compounds. The results revealed that the combination of Temephos and Diallyldisulfide and combination of Malathion and Eudesmol were the most effective combination. These effective combinations bear potential prospect to be used againstAedes aegypti.

Abstract:Mosquitoes represent one of the most important vectors and are responsible for the transmission of many arboviruses that affect human and animal health. The chemical method using synthetic insecticides disturbs the environmental system and promotes the appearance of resistant insect species. Therefore, this study investigated the insecticidal effect of some binary monoterpene combinations (1,8 cineole + α-pinene and carvone + R (+)-pulegone) using a mixture design approach. The fumigant toxicity was evaluated against Culex pipiens female adults using glass jars. The results show that the toxicity varies according to the proportions of each compound. Indeed, Mixture 1 (1,8-cineole + α-pinene) displayed a strong toxic effect (51.00 ± 0.86% after 24 h and 100.00 ± 0.70% after 48 h) when the pure compounds were tested at 0.25/0.75 proportions of 1,8-cineole and α-pinene, respectively. Nevertheless, the equal proportion (0.5/0.5) of carvone and R (+)-pulegone in Mixture 2 exhibited a toxic effect of 54.35 ± 0.75% after 24 h and 89.96 ± 0.14% after 48 h, respectively. For Mixture 1, the maximum area of mortality that the proposed model indicated was obtained between 0/1 and 0.25/0.75, while the maximum area of mortality in the case of Mixture 2 was obtained between 0.25/0.75 and 0.75/0.25. Moreover, the maximum possible values of mortality that could be achieved by the validated model were found to be 51.44% (after 24 h) and 100.24% (after 48 h) for Mixture 1 and 54.67% (after 24 h) and 89.99% (after 48 h) for Mixture 2. It can be said that all purev molecules tested through the binary mixtures acted together, which enhanced the insecticide's effectiveness. These findings are very promising, as the chemical insecticide (deltamethrin) killed only 19.29 ± 0.01% and 34.05 ± 1.01% of the female adults after 24 h and 48 h, respectively. Thus, the findings of our research could help with the development of botanical insecticides that might contribute to management programs for controlling vectors of important diseases.

Abstract:Plectranthus amboinicus(PA) is a highly aromatic perennial shrub that belongs to the Lamiaceae family. It has thick, fleshy and succulent leaves and stem. The margins of the leaves contain numerous glandular hairs that produce essential oil. The essential oil of PA(PAEO) exhibits therapeutical, nutritional, antioxidant, antimicrobial, insecticidal, and insect repellent potentials. The objective of this study was to evaluate and compare the repellent properties of PAEO and DEET against Aedes aegypti mosquitoes, which are known carriers of dengue fever. The extraction of PAEO was carried out using hydrodistillation method while the chemical profile was analysed by gas chromatography-mass spectrometry (GC-MS) technique. The bioefficacy test was conducted according to the household insecticide products personal mosquito repellent evaluation method for biological efficacy (First revision) (MS1497:2007) provided by the Standard and Industrial Research Institute of Malaysia (SIRIM) with slight modification. 10% PAEO was used as test solution while 10% DEET was used as positive control. Fabric and patches treated with 10% PAEO were tested on human volunteers. Both the fabric and the patch, treated with 10% PAEO, exhibit excellent repellency (97 - 100%) against Ae. aegypti mosquitoes for a duration of up to 240 min (4 h) post-application. In contrast, 10% DEET, which initially showed a comparable repellency (99%) 30 min after application, experiences a gradual decline in repellency over time, reaching a 51% repellency rate at the end of the test (240 min post-application). A two-way ANOVA analysis indicates significant effect for type of treatment (p = 0.016) but with no significant difference between the exposure time. Tukey's test showed significant differences between 10% PAEO-treated patches and 10% DEET (p = 0.016), and between 10% PAEO-treated fabric and 10% DEET (p = 0.026). These findings suggest that PAEO is potentially an effective natural repellent against Ae. aegypti.

Abstract:Plant essential oils have been widely used as fragrance in perfumes or flavouring agents in food products due to their unique characteristic scent of the active constituents. The objective of this study is to qualitatively and quantitatively analyse the repellent activity, and the properties of the bioactive constituents in nutmeg, peppermint, and basil oils. The repellent activity of these essential oils was tested against Aedes aegypti and analysed using gas chromatography-flame ionisation (GC-FID) method. α-pinene, terpinen-4-ol and safrole in nutmeg oil, α-pinene, β-pinene, and menthol in peppermint oil, and methyl chavicol, geraniol, and methyl eugenol in basil oil, were selected for further analysis. Selected constituents were tested for accuracy, intra-and interday precision, recovery, limit of detection (LOD), and limit of quantification (LOQ). Nutmeg oil contributed to the highest percentage of repellence with 70.2%, with the accuracy values ranged from 99.25 - 99.71%. The precision for intra- and interday measurements for all tested constituents ranged between 1.083 - 7.296% and 1.308 - 7.924% respectively. The range of recovery was 97.43 - 100.86%. LOD ranged from 0.005 - 0.4 mol/L, and LOQ (with signal to noise ratio of 10:1) ranged from 0.0002 - 0.025 mol/L. In conclusion, the proposed method is simple, precise, and accurate, for qualitative and quantitative analysis of constituents present in essential oils. Nutmeg oil is an alternative natural mosquito repellent with α-pinene as the main active constituent.

Roles of α-Farnesene in the Behaviors of Codling Moth Females

Abstract:Reproduction and olfactory behavioral responses of codling moth, Cydia pomonella (L.), females to synthetic α-farnesene were observed in the laboratory as well as their reproduction behaviors in an apple orchard. Calling levels were lifted and ovipositional peaks were advanced in codling moth females at presence of 1μg and 0.1 μg of α-farnesene, respectively. Mated females of codling moth more actively responded to 0.01 μg α-farnesene with walking and wing-fanning while walking than to other doses (0.001, 0.1, 1, 10 μg ) and control. The results show that α-farnesene plays important roles in the behaviors of codling moth females. However, the differences between responses to α-farnesene and those to apple volatiles by codling moth females indicate that components other than α-farnesene in apple volatiles also have biological activities
Combinations of Lemongrass and Star Anise Essential Oils and Their Main Constituent: Synergistic Housefly Repellency and Safety against Non-Target Organisms

Abstract:The present study evaluated the housefly repellency of single-component formulations and combinations of lemongrass and star anise essential oils (EOs) and their main constituents. The efficacies of the combinations were compared against those of single-component formulations and DEET. Safety bioassays of all formulations and DEET on non-target species—guppy, molly, dwarf honeybee, and stingless bee—were conducted. GC–MS analysis showed that the main constituent of lemongrass EO was geranial (46.83%) and that of star anise EO was trans-anethole (92.88%). All combinations were highly synergistic compared to single-component formulations, with an increased repellent value (IR) of 34.6 to 51.2%. The greatest synergistic effect was achieved by 1.0% lemongrass EO + 1.0% trans-anethole combination, with an IR of 51.2%. The strongest, 100% repellent rate at 6 h was achieved by 1.0% geranial + 1.0% trans-anethole. They were twice as effective as DEET and caused obvious damage to housefly antennae under microscopic observation. All single-component formulations and combinations were benign to the four tested non-target species. In contrast, DEET was highly toxic to them. The synergistic repellency and biosafety of these two combinations are compellingly strong support for developing them into an effective green repellent.

Abstract:In this study, the chemical composition, repellent, and oviposition deterrent effects of five plant essential oils (EOs) extracted from Lantana camara (Verbenaceae), Schinus terebinthifolia (Anacardiaceae), Callistemon viminalis (Myrtaceae), Helichrysum odoratissimum (Asteraceae), and Hyptis suaveolens (Lamiaceae) were evaluated against Aedes aegypti, Anopheles gambiae, and Culex quinquefasciatus. When tested at 33.3 μg/cm2, L. camara, S. terebinthifolia, C. viminalis, and H. odoratissimum were effective repellents against Ae. aegypti (89%, 91%, 90%, and 51% repellency, respectively), but they were less repellent against An. gambiae (66%, 86%, 59%, and 49% repellency, respectively). Interestingly, L. camara, S. terebinthifolia, C. viminalis, and H. odoratissimum exhibited 100% repellency against Cx. quinquefasciatus at 33.3 μg/cm2. In time-span bioassays performed at 333 μg/cm2, the EO of L. camara exhibited 100% repellence against Ae. aegypti and An. gambiae for up to 15 min and against Cx. quinquefasciatus for 75 min. The oviposition bioassays revealed that L. camara exhibited the highest activity, showing 85%, 59%, and 89% oviposition deterrence against Ae. aegypti, An. gambiae, and Cx. quinquefasciatus, respectively. The major compounds of L. camara, S. terebinthifolia, and C. viminalis were trans-β-caryophyllene (16.7%), α-pinene (15.5%), and 1,8-cineole (38.1%), respectively. In conclusion, the L. camara and S. terebinthifolia EOs have the potential to be natural mosquito repellents.

Abstract:Repellents are effective personal protective means against outdoor biting mosquitoes. Repellent formulations composed of EOs are finding increased popularity among consumers. In this study, after an initial screening of 11 essential oils (EOs) at the concentration of 33 μg/cm2, five of the most repellent EOs, Perovskia atriplicifolia, Citrus reticulata (fruit peels), C. reticulata (leaves), Mentha longifolia, and Dysphania ambrosioides were further investigated for repellent activity against Aedes aegypti mosquitoes in time span bioassays. When tested at the concentrations of 33 μg/cm2, 165 μg/cm2 and 330 μg/cm2, the EO of P. atriplicifolia showed the longest repellent effect up to 75, 90 and 135 min, respectively, which was followed by C. reticulata (peels) for 60, 90 and 120 min, M. longifolia for 45, 60 and 90 min, and C. reticulata (leaves) for 30, 45 and 75 min. Notably, the EO of P. atriplicifolia tested at the dose of 330 μg/cm2 showed complete protection for 60 min which was similar to the commercial mosquito repellent DEET. Gas chromatographic-mass spectrometric analyses of the EOs revealed camphor (19.7%), limonene (92.7%), sabinene (24.9%), carvone (82.6%), and trans-ascaridole (38.8%) as the major constituents of P. atriplicifolia, C. reticulata (peels), C. reticulata (leaves), M. longifolia, and D. ambrosioides, respectively. The results of the present study could help develop plant-based commercial repellents to protect humans from dengue mosquitoes.

Abstract:The management of mosquito resistance to chemical insecticides and the biting behaviour of some species are motivating the search for complementary and/or alternative control methods. The use of plants is increasingly considered as a sustainable biological solution for vector control. The aim of this study was to evaluate the biological effects of the essential oil (EO) of Lippia alba harvested in Abidjan (Côte d'Ivoire) against Anopheles gambiae and Aedes aegypti mosquitoes. Phytochemical compounds were identified by GC–MS. Knockdown and mortality were determined according to the WHO test tube protocol. Contact irritancy was assessed by observing the movement of mosquitoes from a treated WHO tube to a second untreated tube. Non-contact repellency was assessed using a standardised high-throughput screening system (HITSS). Blood meal inhibition was assessed using a membrane feeding assay treated with EO. The EO was identified as the citral chemotype. The EO gave 100% KD60 in both species at a concentration of 1%. Mortalities of 100% were recorded with An. gambiae and Ae. aegypti at concentrations of 1% and 5% respectively. The highest proportions of females escaping during the contact irritancy test were 100% for An. gambiae at 1% concentration and 94% for Ae . aegypti at 2.5% concentration. The 1% concentration produced the highest proportions of repelled mosquitoes in the non-contact repellency tests: 76.8% ( An. gambiae ) and 68.5% ( Ae. aegypti ). The blood meal inhibition rate at a dose of 10% was 98.4% in Ae . aegypti but only 15.5% in An. gambiae . The citral chemotype of L. alba EO has promising biological effects in both species that make it a potentially good candidate for its use in mosquito control. The results obtained in this study encourage the further evaluation of L. alba EOs from other localities and of different chemotypes, under laboratory and field conditions.

Moderate Acute Isovolemic Hemodilution Alters Myocardial Function in Patients with Coronary Artery Disease

Abstract:BACKGROUND: Although moderate hemodilution is usually well tolerated in coronary artery surgery patients, this may not be the case when myocardial oxygen demand is increased. We hypothesized that, in these patients, hemodilution in the presence of an increased heart rate could be associated with an impairment of myocardial function. METHODS: Forty coronary surgery patients were randomly assigned to two groups (n = 20), according to the rate of atrioventricular pacing [70 bpm (Group 70) or 90 bpm (Group 90)]. While paced at the fixed heart rate, hemodilution was performed before the start of cardiopulmonary bypass. Data were obtained from a pulmonary artery, a PiCCO catheter and a left ventricular pressure catheter. Measurements were obtained in steady-state conditions before and after isovolemic hemodilution. RESULTS: Hemodilution from 40% ± 2% to 30% ± 1% in Group 70, and from 39% ± 4% to 30% ± 2% in Group 90 resulted in a decrease in systemic vascular resistance and an increase in end-diastolic volume in both groups. This was associated with an increase in stroke volume in Group 70 but not in Group 90. In this latter group, the maximal rate of pressure development decreased significantly after hemodilution [from 856 ± 93 to 716 ± 80 mm Hg/s (P < 0.01)], whereas it remained unchanged in Group 70 (843 ± 86 mm Hg/s before and 832 ± 79 mm Hg/s after hemodilution). CONCLUSIONS: In the conditions of the present study, increased heart rate during moderate hemodilution was associated with a depression of myocardial function.
The Efficacy of Some Commercially Available Insect Repellents for Aedes aegypti (Diptera: Culicidae) and Aedes albopictus (Diptera: Culicidae)

Abstract:Reducing the number of host-vector interactions is an effective way to reduce the spread of vector-borne diseases. Repellents are widely used to protect humans from a variety of protozoans, viruses, and nematodes. DEET (N,N-Diethyl-meta-toluamide), a safe and effective repellent, was developed during World War II. Fear of possible side effects of DEET has created a large market for "natural" DEET-free repellents with a variety of active ingredients. We present a comparative study on the efficacy of eight commercially available products, two fragrances, and a vitamin B patch. The products were tested using a human hand as attractant in a Y-tube olfactometer setup with Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse), both major human disease vectors. We found that Ae. albopictus were generally less attracted to the test subject's hand compared with Ae, aegypti. Repellents with DEET as active ingredient had a prominent repellency effect over longer times and on both species. Repellents containing p-menthane-3,8-diol produced comparable results but for shorter time periods. Some of the DEET-free products containing citronella or geraniol did not have any significant repellency effect. Interestingly, the perfume we tested had a modest repellency effect early after application, and the vitamin B patch had no effect on either species. This study shows that the different active ingredients in commercially available mosquito repellent products are not equivalent in terms of duration and strength of repellency. Our results suggest that products containing DEET or p-menthane-3,8-diol have long-lasting repellent effects and therefore provide good protection from mosquito-borne diseases.

Abstract:Plant essential oil (EO) is a natural alternative to synthetic chemical insecticides for mosquito control. EOs from Citrus aurantium L., Cymbopogon citratus (Stapf.), and Cinnamomum verum (J. Presl.) were selected for topical assay of their ovicidal activity against Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse). Their efficacy was compared to that of 1% ( w/w ) temephos. In addition, their non-toxicity against aquatic mosquito predators, Poecilia latipinna and Poecilia reticulata , was tested. Found by GC–MS analysis, the major constituent of C . verum EO was trans -cinnamaldehyde, of C . aurantium EO was d -limonene, and of C . citratus EO was geranial. Both C . verum EO and trans -cinnamaldehyde at a high concentration (30,000 ppm) exhibited high ovicidal activity against Ae . aegypti and Ae . albopictus eggs after 48 h of incubation with an inhibition rate of 91.0–93.0% for C . verum EO and 96.7–95.2% for trans -cinnamaldehyde. The combination of C . verum EO + geranial exhibited the strongest synergistic inhibition activity (100%) against the two mosquito vectors and was five times more effective than temephos. Moreover, they were not toxic to the non-target fishes. As a safe ovicidal agent for mosquito egg control, the combination of C . verum EO + geranial has excellent potential.

Mosquito Repellent Potential of Carpesium abrotanoides Essential Oil and Its Main Components Against a Dengue Vector, Aedes aegypti (Diptera: Culicidae)

Abstract:Disease vectoring mosquitoes are a serious threat to humans. However, till today only few mosquito repellents have been identified. The current study was conducted to evaluate the repellent potential of Carpesium abrotanoides essential oil against Aedes aegypti females by human bait technique. Essential oil was extracted by steam distillation process while the identification of chemical constituents was carried out by gas chromatography-mass spectrometry. Time span repellent bioassays of C. abrotanoides essential oil in comparison to DEET were performed at three different doses (33 μg/cm2, 165 μg/cm2, and 330 μg/cm2) under laboratory conditions. Highest repellency periods for essential oil and DEET were observed at the tested dose of 330 μg/cm2 with 315 min and 720 min, respectively. Lowest repellency period of 45 min for essential oil and 105 min for DEET was recorded at the tested dose of 33 μg/cm2. Major constituents caryophyllene (24.3%) and trans-nerolidol (12.0%) of C. abrotanoides essential oil were also evaluated as repellents at three different doses (330 μg/cm2, 165 μg/cm2, and 33 μg/cm2) against Ae. aegypti. Surprisingly, trans-nerolidol completely inhibited Ae. aegypti landings for 45 min when tested at 330 μg/cm2. However, caryophyllene did not completely inhibit Ae. aegypti landing even after immediate application at the tested dose of 330 μg/cm2. At the tested dose of 330 μg/cm2, the mixture (trans-nerolidol + caryophyllene) completely inhibited Ae. aegypti landing for 60 min indicating the synergistic effect of caryophyllene. Hence, C. abrotanoides as well as its major constituent, especially trans-nerolidol, have potential to formulate as mosquito repellent comparable of DEET.
Insecticidal activity of essential oils from American native plants against Aedes aegypti (Diptera: Culicidae): an introduction to their possible mechanism of action

Abstract:Abstract Searching for new bioactive molecules to design insecticides is a complex process since pesticides should be highly selective, active against the vector, and bio-safe for humans. Aiming to find natural compounds for mosquito control, we evaluated the insecticidal activity of essential oils (EOs) from 20 American native plants against Aedes aegypti larvae using bioassay, biochemical, and in silico analyses. The highest larvicide activity was exhibited by EOs from Steiractinia aspera (LC 50 = 42.4 μg/mL), Turnera diffusa (LC 50 = 70.9 μg/mL), Piper aduncum (LC 50 = 55.8 μg/mL), Lippia origanoides (chemotype thymol/carvacrol) (LC 50 = 61.9 μg/mL), L. origanoides (chemotype carvacrol/thymol) (LC 50 = 59.8 μg/mL), Hyptis dilatata (LC 50 = 61.1 μg/mL), Elaphandra quinquenervis (LC 50 = 61.1 μg/mL), and Calycolpus moritzianus (LC 50 = 73.29 μg/mL) after 24 h. This biological activity may be related to the disruption of the electron transport chain through the mitochondrial protein complexes. We hypothesized that the observed EOs' effect is due to their major components, where computational approaches such as homology modeling and molecular docking may suggest the possible binding pose of secondary metabolites that inhibit the mitochondrial enzymes and acetylcholinesterase activity (AChE). Our results provided insights into the possible mechanism of action of EOs and their major compounds for new insecticide designs targeting the mitochondria and AChE activity in A. aegypti for effective and safe insecticide.
