Abstract:<h3 class="a-plus-plus">Objectives</h3>In this study, genome sequencing and metabolic analysis were used to identify and verify the key metabolic pathways for glucose and xylose utilization and fatty acid synthesis in the walnut endophytic bacterium (WEB) Bacillus subtilis HB1310.<h3 class="a-plus-plus">Results</h3>The genome sequence of WEB HB1310 was generated with a size of 4.1 Mb and GC content of 43.5%. Genome annotation indicated that the Embden–Meyerhof–Parnas, pentose phosphate, and fatty acid synthesis pathways were mainly involved in mixed sugar utilization and lipid production. In particular, diverse and abundant fatty acid synthesis genes were observed in a higher number than in other Bacillus strains. The tricarboxylic acid cycle competitively shared the carbon flux flowing before 48 h, and the acetic acid fermentation competed after 72 h. Moreover, fatty acid synthase activity was highly correlated with lipid titer with a correlation coefficient of 0.9626, and NADPH might be more utilized for the lipid synthesis within 48 h.<h3 class="a-plus-plus">Conclusions</h3>This study is the first attempt to explain the metabolic mechanism of mixed sugar utilization and lipid production based on genomic information, which provides a theoretical basis for the metabolic regulation of bacterial lipid production from lignocellulosic hydrolysates.

Abstract:Cost-effective conversion of lignocellulose hydrolysate to optically pure lactic acid is commercially attractive but very challenging. Bacillus coagulans JI12 was isolated from natural environment and used to produce L-lactic acid (optical purity > 99.5 %) from lignocellulose sugars and acid hydrolysate of oil palm empty fruit bunch (EFB) at 50 °C and pH 6.0 without sterilization of the medium. In fed-batch fermentation with 85 g/L initial xylose and 55 g/L xylose added after 7.5 h, 137.5 g/L lactic acid was produced with a yield of 98 % and a productivity of 4.4 g/L h. In batch fermentation of a sugar mixture containing 8.5 % xylose, 1 % glucose, and 1 % L-arabinose, the lactic acid yield and productivity reached 98 % and 4.8 g/L h, respectively. When EFB hydrolysate was used, 59.2 g/L of lactic acid was produced within 9.5 h at a yield of 97 % and a productivity of 6.2 g/L h, which are the highest among those ever reported from lignocellulose hydrolysates. These results indicate that B. coagulans JI12 is a promising strain for industrial production of L-lactic acid from lignocellulose hydrolysate.

Abstract:Bacillus coagulans JI12 was used to produce L-lactic acid from both cellulose and hemicellulose sugars of oil palm empty fruit bunch hydrolysate at 50 C without sterilization prior to fermentation. In fermentation of mixed glucose and xylose (10 g/L:100 g/L or 50 g/L:53 g/L), both sugars were simultaneously converted to L-lactic acid. B. coagulans JI12 was tolerant against up to 4 g/L of furfural and 20 g/L of acetate and able to metabolize furfural to 2-furoic acid. After acid hydrolysis, both the hemicellulosic and cellulosic fractions of oil palm empty fruit bunch were fermented to lactic acid in a simultaneous detoxification, saccharification and co-fermentation process supplemented with 25 FPU Celtic (R) CTec2 cellulase per g cellulose, yielding 80.6 g/L of lactic acid with a productivity of 3.4 g/L/h. Neither pre-detoxification nor separation of fermentable sugars from lignin was required. These results indicate that B. coagulans JI12 is a promising strain for L-lactic acid production from lignocellulosic biomass. (C) 2013 Elsevier B.V. All rights reserved.
Exploring an Isolate of the Oleaginous Alga Micractinium Inermum for Lipid Production: Molecular Characterization and Physiochemical Analysis under Multiple Growth Conditions

Abstract:Molecular characterization based on the internal transcribed spacer 2 (ITS2) sequence identified Micractinium inermum JL1, a local isolate of green alga capable of growing robustly under photoautotrophic, mixotrophic, and heterotrophic culture modes. Physiochemical analyses of the alga revealed great variations in growth, lipid content, oleic acid abundance, and productivities of lipids and triacylglycerol (TAG) under various culture conditions including nitrogen concentrations, phosphorus concentrations, light intensities, salinity levels, and glucose concentrations with or without light illumination. The carbon shunt from starch and to a lesser extent from membrane lipids likely contributed to the TAG accumulation. Noteworthily, salt supplementation promoted TAG content and productivity by ~ 88% and ~ 55%, respectively. The alga could accumulate ~ 52% lipids per dry weight and achieved a high lipid productivity of 0.68 g L−1 day−1. C18:1 percentage, a biodiesel quality indicator, reached ~ 50%, and correlated well with TAG contents in a positive manner, demonstrating the feasibility of using C18:1 percentage for TAG quantification thereby avoiding time- and labor-intensive analysis. These results together indicate the potential of M. inermum JL1 as a lipid producer for future exploration.
Orthogonal Optimization Of A Newly Isolated Microalga Desmodesmus Sp.-Chx1 As A Potential Candidate For Biodiesel Production

Abstract:This study aimed to optimize lipid productivity of an indigenous microalgal isolate Desmodesmus sp.-CHX1 when grown in modified BG-11 medium. An orthogonal L-9(3)(4) test was applied to select the optimum cultivation parameters including glucose concentration, nitrate concentration, seeding cell concentration and initial pH. Results showed that glucose concentration and seeding cell concentration were the main variables that influenced both biomass and lipid productivities. 160.8mg L-1 d(-1) of lipid productivity was much higher than those reported previously in the literatures, when Desmodesmus sp.-CHX1 was grown under the optimized conditions of 3g L-1 of glucose concentration, pH=10, 10% of seeding cell concentration and 2g L-1 of nitrate concentration. The presence of C16/C18 fatty acids accounted for 70% of total fatty acid as the major constituents. Both of high lipid productivity and ratio of C16/C18 fatty acids indicated that Desmodesmus sp.-CHX1 was a suitable feedstock for biodiesel production.

Abstract:Given the escalating demand for renewable biofuels amidst the continual consumption of fossil energy, the exploration and identification of microalgal strains for biodiesel production have become crucial. In this study, a microalgal strain named HDMA-12 was isolated from Lake Chenjiadayuan in China to evaluate its biodiesel potential. Phylogenetic analysis of its internal transcribed spacer sequences revealed HDMA-12 as a new molecular record in the genus Coelastrum . When cultivated in BG11 basal medium, HDMA-12 achieved a biomass of 635.7 mg L −1 and a lipid content of 26.4%. Furthermore, the fatty acid methyl ester profile of HDMA-12 exhibited favorable combustion characteristics. Subjected to 200 mM NaCl stress, HDMA-12 reached its maximum biomass of 751.5 mg L −1 and a lipid content of 28.9%. These findings indicate the promising prospects of HDMA-12 as a promising microalgal strain for further advancements in biodiesel production.

Abstract:Cost-effective production of optically pure lactic acid from lignocellulose sugars is commercially attractive but challenging. Bacillus coagulans C106 was isolated from environment and used to produce l-lactic acid from xylose at 50°C and pH 6.0 in mineral salts medium containing 1-2% (w/v) of yeast extract without sterilizing the medium before fermentation. In batch fermentation with 85g/L of xylose, lactic acid titer and productivity reached 83.6g/L and 7.5g/Lh, respectively. When fed-batch (120+80+60g/L) fermentation was applied, they reached 215.7g/L and 4.0g/Lh, respectively. In both cases, the lactic acid yield and optical purity reached 95% and 99.6%, respectively. The lactic acid titer and productivity on xylose are the highest among those ever reported. Ca(OH)2 was found to be a better neutralizing agent than NaOH in terms of its giving higher lactic acid titer (1.2-fold) and productivity (1.8-fold) under the same conditions.
Consolidated Bioprocessing Performance of Penicillium Sp P-2 in Cellulose-Based Substrates for Low-Cost Oil Production

Abstract:Summary Consolidated bioprocessing (CBP) is a convenient and cost‐efficient strategy to produce single cell oils ( SCOs ) from cellulose‐based substrates . In this study , Penicillium P‐2 having both effective cellulose degradation and lipid accumulation was isolated from 20 cellulolytic fungi . The SCOs of the strain P‐2 were mainly composed of palmitic acid ( C16 :0 , 21 .05% ) , oleic acid ( C18 :1 , 22 .43% ) and linoleic acid ( C18 :2 , 27 .78% ) . Fermentation experiments of the strain P‐2 showed that maximum lipid yields of 0 .65 g/L and 40 .13 mg/g ( per gram dry mass of initial solid substrate) could be obtained by submerged fermentation ( SmF) from cellulose and solid‐state fermentation (SSF) from wheat straw and bran mixture , respectively . The results from the two CBP indicated that the strain P‐2 had a potential to be a promising low‐cost oil producer using cellulose‐based substrates . The further analysis for lipid accumulation and cellulase secretion of the strain P‐2 suggested that the cellulase secretion of the strain P‐2 might play a key role in the lipid production . The submerged fermentation with exogenous cellulase addition by the strain P‐2 demonstrated that the higher exogenous cellulase dosages could lead to higher lipid production and the lipid yield would increase up to 0 .83 g/L with the addition of 24 IU /g cellulase . A highly significant positive correlation between the filter paper activity ( FPA) and the lipid yield in the strain P‐2 was further observed on the data pooled from the SSF parameter treatments ( P < 0 .01) . When the detected FPA in SSF system increased from 1 .0 IU /g to 3 .5 IU /g , the lipid yield was also increased from 26 .24 mg/g to 40 .13 mg/g with the increment of 52 .93% . All these analyses suggest that the weak cellulase secretion of the strain P‐2 is an important cause of the low lipid yield , so the regulation of endogenous cellulase secretion might be a feasible strategy for enhancing lipid production of the strain P‐2 .

Abstract:Alkane-based biodiesel is considered the next generation of biodiesel owing to its potential environmental benefits and the fact that it exhibits much higher specific caloric values than traditional biodiesel. However, the formidable obstacle impeding the commercialization of this cutting-edge fuel alternative lies in the cost associated with its production. In this study, an engineered strain Escherichia coli ( E. coli ) showcasing harmonized coexpression of a lipase (from Thermomyces lanuginosus , TL L) and a fatty acid photodecarboxylase (from Chlorella variabilis , Cv FAP) was first constructed to transform triglycerides into alkanes. The potential of E. coli BL21 (DE3)/pRSFDuet-1- TL L- Cv FAP for alkane synthesis was evaluated with tripalmitin as a model substrate under various process conditions. Following a comprehensive examination of the reaction parameters, the scope of the biotransformation was expanded to 'real' substrates (vegetable oils). The results showed that bioderived oils can be transformed into alkanes with high yields (0.80–10.20 mmol·L –1 ) under mild conditions (35 °C, pH 8.0 and 36 h) and blue light illumination. The selected processes were performed on an increased lab scale (up to 100 ml) with up to 24.77 mmol·L –1 tripalmitin, leading to a yield of 18.89 mmol·L –1 pentadecane. With the employment of a method for efficiently producing alkanes under mild conditions alongside a simple procedure to isolate alkanes from the reaction system, the utilization of sustainable biomass as a fundamental feedstock emerges as the primary solution to lower the cost of alkane-based biodiesel. Thus, this study proposes a readily implementable and highly effective approach for alkane-based biodiesel production.

Abstract:Nowadays, finding a talented lipase with high potency in biodiesel production has been attracted to researchers. In this study, an extracellular salt tolerant, cold-active and organic solvent-stable lipase producing bacterium was isolated from oil-contaminated environments located in Kerman province (Iran) and identified as Bacillus licheniformis KM12. Lipase was purified by 36.0% recovery and molecular weight was estimated about 33 kDa by SDS-PAGE. The optimal pH and temperature for lipase activity were found to be 8.0 and 35 °C, respectively. Lipase activity was remarkably increased about 37% after 20 min incubation at 20 °C. Km value of KM12 lipase was 0.53 mM for p-nitrophenyl palmitate (p-NPP) and maximum activity was detected for pNP-decanoate and glyceryl-decanoate substrates. KM12 lipase displayed significant stability in different organic solvents after 7 and 21 days of incubation, especially in polar organic solvents. Biodiesel production with 78% yields was achieved with the one-step addition of methanol at around 18 h by using Myrtus oil (as a non-edible oils feedstocks). These unique properties of KM12 lipase make it talented as a potential biocatalyst for biodiesel production.
Comparative lipid production by oleaginous yeasts in hydrolyzates of lignocellulosic biomass and process strategy for high titers

Abstract:Oleaginous yeasts can convert sugars to lipids with fatty acid profiles similar to those of vegetable oils, making them attractive for production of biodiesel. Lignocellulosic biomass is an attractive source of sugars for yeast lipid production because it is abundant, potentially low cost, and renewable. However, lignocellulosic hydrolyzates are laden with byproducts which inhibit microbial growth and metabolism. With the goal of identifying oleaginous yeast strains able to convert plant biomass to lipids, we screened 32 strains from the ARS Culture Collection, Peoria, IL to identify four robust strains able to produce high lipid concentrations from both acid and base-pretreated biomass. The screening was arranged in two tiers using undetoxified enzyme hydrolyzates of ammonia fiber expansion (AFEX)-pretreated cornstover as the primary screening medium and acid-pretreated switch grass as the secondary screening medium applied to strains passing the primary screen. Hydrolyzates were prepared at ∼18-20% solids loading to provide ∼110 g/L sugars at ∼56:39:5 mass ratio glucose:xylose:arabinose. A two stage process boosting the molar C:N ratio from 60 to well above 400 in undetoxified switchgrass hydrolyzate was optimized with respect to nitrogen source, C:N, and carbon loading. Using this process three strains were able to consume acetic acid and nearly all available sugars to accumulate 50-65% of cell biomass as lipid (w/w), to produce 25-30 g/L lipid at 0.12-0.22 g/L/h and 0.13-0.15 g/g or 39-45% of the theoretical yield at pH 6 and 7, a performance unprecedented in lignocellulosic hydrolyzates. Three of the top strains have not previously been reported for the bioconversion of lignocellulose to lipids. The successful identification and development of top-performing lipid-producing yeast in lignocellulose hydrolyzates is expected to advance the economic feasibility of high quality biodiesel and jet fuels from renewable biomass, expanding the market potential for lignocellulose-derived fuels beyond ethanol for automobiles to the entire U.S. transportation market. Abstract:Lignocellulosic materials such as corn cob are an inexpensive, natural and the most abundant renewable resource in the nature. In this study, it was found that an oleaginous yeast-like fungal strain Aureobasidium melanogenum P10 could grow well on corncob-derived xylose and accumulate 38.27% (w/w) of lipids from it within 120 h. Removal of an oxidoreductase pathway and overexpression of an isomerase pathway from A. melanogenum P10 made the XI-15 strain accumulate 53.74% (w/w) of lipids from the same xylose within 108 h. All the properties of the prepared biodiesel obtained from the crude lipids produced by both P10 and XI-15 strains could totally meet the requirements of US biodiesel ASTM D6751 and EU biodiesel EN 14214. Especially, the cetane numbers (56.82 ± 1.2 and 57.32 ± 1.3 min) of the prepared biodiesels were much higher than those of US biodiesel ASTM D6751 and EU biodiesel EN 14214 and their iodine numbers (80.20 ± 2.1 and 74.71 ± 1.9) were much lower than those of US biodiesel ASTM D6751 and EU biodiesel EN 14214. These meant that the properties of the prepared biodiesels in this study were better than those of the standard ones and the prepared biodiesels had high potential applications in renewable fuel industries. At the same time, the prepared biodiesel was found to be burnt well.

Morphological and cellular changes within embryonic striatal grafts associated with enriched environment and involuntary exercise

Abstract:Environmental enrichment (EE) and exercise have been implicated in influencing behaviour and altering neuronal processes associated with cellular morphology in both ‘normal’ and injured states of the CNS. Using a rodent model of Huntington's disease, we investigated whether prolonged EE or involuntary exercise can induce morphological and cellular changes within embryonic striatal transplants. Adult rats were trained on the Staircase test – requiring fine motor control to reach and collect reward pellets – prior to being lesioned unilaterally in the dorsal neostriatum with quinolinic acid. The lesioned animals received E15 whole ganglionic eminence cell suspension grafts followed by housing in EE or standard cages. Half of the animals in standard cages received daily forced exercise on a treadmill. The grafted animals showed significant functional recovery on both the Staircase test and in drug‐induced rotation. Neither the housing conditions nor the training had an impact on the behaviour, with the exception of the treadmill reducing the ipsilateral drug‐induced rotation observed amongst the lesioned animals. However, the animals housed in the EE had significantly increased striatal brain‐derived neurotrophic factor (BDNF) levels, and graft neurons in these animals exhibited both greater spine densities and larger cell volumes. Animals on forced exercise regime had reduced BDNF levels and grafted cells with sparser spines. The study suggests that the context of the animal can affect the plasticity of transplanted cells. Appropriately exploiting the underlying, and yet unknown, mechanisms could lead the way to improved anatomical and potentially functional integration of the graft.

Abstract:Biocomponents (such as lipids) accumulate in oleaginous microorganisms and could be used for renewable energy production. Oleaginous microbes are characterized by their ability to accumulate high levels of lipids, which can be converted into biodiesel. The oleaginous microbes (including microalgae, bacteria, yeast, and fungi) can utilize diverse substrates. Thus, in this study, commercially viable oleaginous microorganisms are comparatively summarized for their growth conditions, substrate utilization, and applications in biotechnological processes. Lipid content is species-dependent, as are culture conditions (such as temperature, pH, nutrients, and culture time) and substrates. Lipid production can be increased by selecting suitable microorganisms and substrates, optimizing environmental conditions, and using genetic engineering techniques. In addition, the emphasis on downstream processes (including harvesting, cell disruption, lipid extraction, and transesterification) highlights their critical role in enhancing cost-effectiveness. Oleaginous microorganisms are potential candidates for lipid biosynthesis and could play a key role in meeting the energy needs of the world in the future.

Abstract:Microbial lipids with fatty acid compositional profiles similar to vegetable oils are of great potential for the production of biofuels and oleochemicals. However, the costs of microbial lipids remain prohibitively high for broader applications. Microalgae known as the third-generation biomass resources may be explored to reduce the feedstock costs of lipid production. Here, hydrolysates of the microalga Arthrospira platensis were converted into lipids by the oleaginous yeast Cryptococcus curvatus ATCC 20509. Results indicated that hydrothermal pretreatment at 120 °C followed by enzymatic hydrolysis led to recovery of up to 97% total reducing sugar from microalgal biomass. While the as-prepared hydrolysates were ineffective for lipid production due to its nitrogen-rich nature, a high lipid content of 64.5% and a lipid yield of 0.233 g/g were obtained upon phosphate removal with calcium hydroxide. The lipid samples were found similar to palm oils in terms of fatty acid compositional profiles. This work demonstrated that A. platensis biomass can be readily converted into hydrolysates for high-yield lipid production, suggesting an alternative feedstock to cellulosic residues for microbial lipid technology and biorefinery.This article has not yet been cited by other publications.

Abstract:Conversion of acetates, the by-products generated in industrial processes into microbial lipids by using oleaginous yeasts offers a promising alternative for the economic utilization of waste acetates. In this study, two newly isolated oleaginous yeasts Cryptococcus podzolicus DSM 27192 and Trichosporon porosum DSM 27194 were found to be able to grow with acetate as the sole carbon source and tolerate up to 30 g/L of sodium acetate in shake flask fermentation. Furthermore, an automated fed-batch system using acetic acid as both carbon source and buffering agent was designed. By supplementing glucose as the stimulant for microbial growth, lipids contents of 38.8% and 39.7% were obtained by using C. podzolicus DSM 27192 and T. porosum DSM 27194, corresponding to lipids concentrations of 10.4 g/L and 11.5 g/L, respectively. Moreover, the fatty acid compositional profiles of the acetate-derived lipids were similar to those of vegetable oil, suggesting their potential for biodiesel production. Economical analysis showed that acetic acid derived from volatile fatty acids (VFAs) could be potential substrates for industrial microbial lipids production by using C. podzolicus DSM 27192 and T. porosum DSM 27194.

Abstract:Single cell oil (SCO) prepared from biodiesel-derived crude glycerol (BCG) and lignocellulosic biomass (LCB) via oleaginous yeasts is an intriguing alternative precursor of biodiesel. Here, a novel strategy combining alkaline BCG pretreated corn stover and dilute acid pretreated water hyacinth for SCO overproduction was developed. The mixed pretreatment liquors (MPLs) were naturally neutralized and adjusted to a proper carbon-to-nitrogen ratio beneficial for SCO overproduction by Cutaneotrichosporon oleaginosum . The toxicity of inhibitors was relieved by dilution detoxification. The enzymatic hydrolysate of solid fractions was suitable for SCO production either separately or simultaneously with MPLs. Fed-batch fermentation of the MPLs resulted in high cell mass, SCO content, and SCO titer of 80.7 g/L, 75.7 %, and 61.1 g/L, respectively. The fatty acid profiles of SCOs implied high-quality biodiesel characteristics. This study offers a novel BCG&LCB-to-SCO route integrating BCG-based pretreatment and BCG/LCB hydrolysates co-utilization, which provides a cost-effective technical route for micro-biodiesel production.

Abstract:As a potential feedstock for biofuel production, a high-cell-density continuous culture for the lipid production by Cryptococcus albidus was investigated in this study. The influences of dilution rates in the single-stage continuous cultures were explored first. To reach a high-cell-density culture, a single-stage continuous culture coupled with a membrane cell recycling system was carried out at a constant dilution rate of 0.36/h with varied bleeding ratios. The maximum lipid productivity of 0.69g/L/h was achieved with the highest bleeding ratio of 0.4. To reach a better lipid yield and content, a two-stage continuous cultivation was performed by adjusting the C/N ratio in two different stages. Finally, a lipid yield of 0.32g/g and lipid content of 56.4% were obtained. This two-stage continuous cultivation, which provided a higher lipid production performance, shows a great potential for an industrial-scale biotechnological production of microbial lipids and biofuel production.
