Abstract:A whole-cell bioreporter, Acinetobacter baylyi ADPWH_ recA , was used to estimate the genotoxicity and bioavailability of chromium (VI) [Cr(VI)] in contaminated soils. Upon direct exposure to pre-sonicated soil samples, ADPWH_ recA gave the highest response to the genotoxicity of Cr(VI) within 5 h with a detection limit of 2 µM Cr(VI). Investigations on sites contaminated with Cr(VI) revealed that soil-associated Cr(VI) was bioavailable to the bioreporter although it could not be extracted into the aqueous phase. The physical and chemical properties of soil might influence the bioavailability of Cr(VI), and higher genotoxicity was found in soils with a lower pH. This whole cell bioreporter approach makes it feasible to evaluate the bioavailability and genotoxicity of Cr(VI)-contaminated soils to uncover their potential impact on human health.

Abstract:A whole-cell bacterial bioreporter Acinetobacter baylyi strain ADP1_recA_lux that responds to genotoxins was employed to directly assess the adverse effects of the bioavailable fraction of mitomycin C (MMC), benzo[a]pyrene (BaP), chromium (VI) and lead (II) in amended soils and soil samples from two fragile areas in China without soil pre-treatment. The amended soils containing pollutants with the concentrations as low as 0.4 mg/kg MMC, 0.5 mg/kg BaP, 520 mg/kg Cr (VI) and 2072 mg/kg Pb (II) were found to be toxic. Soil particle-associated pollutants accounted for 86%, 100%, 29%, and 92% of the genotoxicity in the MMC, BaP, Cr (VI), and Pb (II) amended soil, respectively. The soils from contaminated sites were also valid to be genotoxic. The results suggest both free and soil particle-associated pollutants are bioavailable to soil organisms and a solid-phase contact bioreporter assay to soil contamination could provide a rapid screening tool for environmental risk assessment.
A Whole-Cell Bioreporter Assay for Quantitative Genotoxicity Evaluation of Environmental Samples

Abstract:Whole-cell bioreporters have emerged as promising tools for genotoxicity evaluation, due to their rapidity, cost-effectiveness, sensitivity and selectivity. In this study, a method for detecting genotoxicity in environmental samples was developed using the bioluminescent whole-cell bioreporter Escherichia coli recA::luxCDABE. To further test its performance in a real world scenario, the E. coli bioreporter was applied in two cases: i) soil samples collected from chromium(VI) contaminated sites; ii) crude oil contaminated seawater collected after the Jiaozhou Bay oil spill which occurred in 2013. The chromium(VI) contaminated soils were pretreated by water extraction, and directly exposed to the bioreporter in two phases: aqueous soil extraction (water phase) and soil supernatant (solid phase). The results indicated that both extractable and soil particle fixed chromium(VI) were bioavailable to the bioreporter, and the solid-phase contact bioreporter assay provided a more precise evaluation of soil genotoxicity. For crude oil contaminated seawater, the response of the bioreporter clearly illustrated the spatial and time change in genotoxicity surrounding the spill site, suggesting that the crude oil degradation process decreased the genotoxic risk to ecosystem. In addition, the performance of the bioreporter was simulated by a modified cross-regulation gene expression model, which quantitatively described the DNA damage response of the E. coli bioreporter. Accordingly, the bioluminescent response of the bioreporter was calculated as the mitomycin C equivalent, enabling quantitative comparison of genotoxicities between different environmental samples. This bioreporter assay provides a rapid and sensitive screening tool for direct genotoxicity assessment of environmental samples.

Abstract:Bioremediation is a sustainable and cost-effective means of contaminant detoxification. Although Cr(VI) is toxic at high concentrations, various microbes can utilise it as an electron accepter in the bioremediation process, and reduce it to the less toxic form Cr(III). During remediation, it is important to monitor the level of toxicity and effectiveness of Cr(VI) reduction in order to optimize the conditions. This study employed a whole-cell bioreporter Acinetobacter baylyi ADPWH-recA to access the degree of toxicity of different species of Cr over a range of initial concentrations. It also investigated whether Cr isotope fractionation factors were impacted by different levels of Cr toxicity (related to its concentration) and Cr(VI) reduction rates by Cr resistant bacteria Pseudomonas fluorescens LB 300. The results show that, of both Cr2O72- and CrO42-, the whole-cell bioreporter was efficient in indicating the level of genotoxicity of Cr(VI) at low concentrations and cytotoxicity at high concentrations via variations of bioluminescence. High concentrations (> 100 mg/L) of Cr(III) could also strongly induce the luminescence in the bioreporter, indicating DNA damage at such abundance. Pseudomonas fluorescens LB 300 was found to be effective in reducing Cr(VI) even when the concentration was high (40 mg/L); however, complete Cr(VI) reduction was only observed at low concentrations (< 5 mg/L), since the toxicity of high concentrations of Cr(VI) impacted the effectiveness of reduction by the bacteria. During reduction, the Cr-53/Cr-52 ratio of remaining Cr(VI) increased from its initial value, and the calculated fractionation factor by bacterial Cr(VI) reduction (epsilon) was -3.1 +/- 0.3 parts per thousand. The fractionation factor was independent of the initial Cr(VI) concentration. Therefore, a single Cr isotope fractionation factor can be effectively applied in indicating the extent of bioremediation processing of Cr(VI) over a wide range of concentrations. This significantly simplified monitoring of Cr(VI) depletion in bioremediation, since variations of e normally indicate a change in the reduction mechanism and therefore would complicate the elucidation of processes driving the remediation. The Influence of Carbon Sources on the Expression of the Reca Gene and Genotoxicity Detection by an Acinetobacter Bioreporter

Abstract:Bacterial whole-cell bioreporters are practical and reliable analytical tools to assess the toxicity and bioavailability of environmental contaminants, yet evidence has shown that their performance could be affected by different carbon sources. This paper evaluated the influence of carbon sources on the recA gene (ACIAD1385) in a DNA damage-inducible recA::luxCDABE Acinetobacter bioreporter and optimized the induction conditions for its practical application in environmental monitoring. Different carbon sources, including LB, potassium acetate (MMA), sodium citrate (MMC), sodium pyruvate (MMP), and sodium succinate (MMS), significantly influenced (p < 0.05) the bioluminescence intensity of the genotoxicity bioreporter. A reverse transcription quantitative PCR (RT-qPCR) showed the different expression levels of the DNA damage-inducible gene recA (p < 0.05), suggesting that carbon sources influenced the DNA damage response in the Acinetobacter bioreporter at the transcriptional level. Additionally, proteomic analysis identified 122 proteins that were differentially expressed after exposure to mitomycin C in defined media and LB, and 5 of them were related to the DNA damage response, indicating the effects of carbon sources on the DNA damage response in Acinetobacter at the translational level. The repression effect caused by the rich medium, LB, was possibly related to the mechanism of carbon catabolite repression. Our results suggest that the practical application of Acinetobacter bioreporters to the genotoxicity assessment of polycyclic aromatic hydrocarbon (PAH)-contaminated soils could be significantly improved by using a standard medium of defined composition, as this could increase their sensitivity.
Deciphering the impacts of chromium contamination on soil bacterial communities: A comparative analysis across various soil types

Abstract:Addressing the widespread concern of chromium (Cr) pollution, this study investigated its impacts on bacterial communities across eight soil types, alongside the potential Cr transformation-related genes. Utilizing real-time PCR, 16S rRNA gene sequencing and gene prediction, we revealed shifts in bacterial community structure and function at three Cr exposure levels. Our results showed that the bacterial abundance in all eight soil types was influenced by Cr to varying extents, with yellow‒brown soil being the most sensitive. The bacterial community composition of different soil types exhibited diverse responses to Cr, with only the relative abundance of Proteobacteria decreasing with increasing Cr concentration across all soil types. Beta diversity analysis revealed that while Cr concentration impacted the assembly process of bacterial communities to a certain extent, the influence on the compositional structure of bacterial communities was primarily driven by soil type rather than Cr concentration. The study also identified biomarkers for each soil type under three Cr levels, offering a basis for monitoring changes in Cr pollution. By predicting crucial functional genes related to Cr transformation, it was observed that the relative abundance of chrA (chromate transporter) in yellow‒brown soil significantly exceeded that in all other soil types, suggesting its potential for Cr adaptation. The study also revealed correlations among soil physicochemical properties, Cr concentration, and these functional genes, providing a foundation for future research aimed at more precise functional analysis and the development of effective soil remediation strategies.

Abstract:Oil contamination has been one of the most common environmental pollution due to its threat to both human health and ecosystems. It is difficult for the application of conventional chemical analysis to detect oil contamination in-situ, estimate bioavailability and rapidly assess the environmental risk. In this study, two bioreporters were employed for the rapid and direct detection of oil content and genotoxicity in a cutting oil contaminated site in China. Acinetobacter baylyi ADPWH_alk and ADPWH_recA are chromosomally-based bioreporter for the detection of alkanes and genotoxicity separately. The reporter gene cluster luxCDABE enabled cells to express bioluminescence in the presence of contaminants. Calibration curve of bioreporter response to oil in the soils illustrated a log-log linear relationship between oil content and bioluminescence expression, indicating that whole cell bioreporter can be used as a semi-quantitative analysis method to estimate the oil contamination. The oil contamination in soils detected by ADPWH_alk (5760 and 1090 mg/kg) were comparable with those obtained by GC/MS (7070 and 1490 mg/kg). Furthermore, the genotoxicity bioreporter ADPWH_recA showed that the ecological genotoxicity of the soil samples were equivalent to 33-36 mg mitomycin C per kilogram soils. The results suggested that bioreporter sensing could be a rapid, simple, low-costly and in-situ detection method with unique ability for the detection of toxicity and bioavailability, and it can be a useful and complementary tool to chemical analysis.

Abstract:In a study to optimize bacterial whole cell biosensors (bioreporters) for the detection of environmental contaminants, we constructed a toxicity sensing strain Acinetobacter bay/yi ADP1_recA_lux. The ADP1_recA_lux is a chromosomally based bioreporter which makes the sensing system stable and negates the need for antibiotics to maintain the trait. The ADP1_recA_lux is activated to express bioluminescence when it is exposed to DNA damaging toxicants. Since the ADP1_recA_lux constantly expresses a baseline level of bioluminescence, false negative results are avoided. The host strain, A. bay/yi ADP1, is an ideal model strain typical of water and soil bacteria occurring in the natural environment and it is more robust than E. coli in terms of viability, maintenance, and storage. The expression of reporter genes - luxCDABE cloned from Photorhabdus luminescens - is robust in a broad range of temperature (10-40 degrees C), The ADP1_recA_lux was used to detect a variety of toxic or potentially toxic compounds including mitomycin C (MMC), methyl methanesulfonate, ethidium bromide, H2O2, toluene, single-wall nanocarbon tubes (SWNCT), nano Au colloids (20 nm), pyrene, beno[a]pyrene, and UV light. These exposures revealed that the ADP1_recA_lux was able to detect both genotoxicity and cytoxicity, qualitatively and quantitatively. The optimal induction time of the ADP1_recA_lux bioreporter was 3 h, and the detection limits for MMC and benezo[a]pyrene were 1.5 nM and 0.4 nM, respectively. The ADP1_recA_lux was also used to detect toxicity of groundwater contaminated by a mixture of phenolic compounds, and the bioreporter toxicity detection was in a good agreement with chemical analysis. The optimized whole cell bioreporter ADP1_recA_lux could be valuable in providing a simple, rapid, stable, quantitative, robust, and costly efficient approach for the detection of toxicity in environmental samples.

Abstract:Hexavalent chromium (Cr(VI)) waste produced by chrome plating activities pollutes the surrounding environment and harms human health. However, information about the chromium (Cr) pollution characteristics of actual electroplating sites is still lacking. In this study, the concentration, leachability and speciation of Cr in soils from a typical chrome plating site were analyzed. Our results showed that this site was severely contaminated by Cr (7.2 to 7735.2 mg/kg) and Cr(VI) reached the mean concentration of 138.7 mg/kg. The spatial distribution of Cr(VI) was related to the plating processes. Chrome plating and sewage treatment areas could be considered as the hot spots of contaminated sites. The vertical distribution of Cr(VI) was mainly affected by soil properties, where the loam layer retained and reduced a large amount of Cr(VI) due to its high content of iron minerals and finer particle fractions. Additionally, the chemical extraction results showed that Cr was mainly in non-residual fractions and the existence of Cr(VI) led to a high leaching toxicity based on the toxicity characteristic leaching procedure (TCLP) results. Moreover, X-ray photoelectron spectroscopy (XPS) results revealed the speciation of Cr in the long-term contaminated soils. A large amount of Cr(VI) was reduced into Cr(III) and mainly existed as Cr(OH)3 and Cr2O3. Furthermore, Cr(VI) tended to precipitate as CaCrO4 and persisted in soils. Therefore, it is necessary to find appropriate strategies to remediate these contaminated soils. Overall, these findings strengthen our understanding of Cr(VI) behaviors and lay a foundation for the future pollution investigation, ecological remediation and risk assessment of sites contaminated by electroplating.

Abstract:Soil chromium (Cr) contamination poses a significant threat to the environment and public health. A phosphate solubilizing bacterial strain Bacillus megaterium PMW-03 was used to enhance the phytoremediation of Cr(VI) contamination by Pteris vittata in phosphate mining wasteland in this study. The results showed that the strain PMW-03 could promote the solubilization of phosphate rock (PR) and the removal of Cr(VI) by Pteris vittata in the phosphate mining wasteland. Compared with the groups not inoculated with PMW-03, the available phosphate content in the rhizosphere soil of the inoculated groups increased by 17.6% and 24.9%, respectively (in the group with Cr(VI) added and the group with Cr(VI) and PR added), and the available Cr(VI) content decreased by 28.2%, 25.2% and 33.4%, respectively. In addition, the strain PMW-03 also promoted the growth of Pteris vittata by secreting indole acetic acid (IAA) and siderophore, promoting Pteris vittata to produce chlorophyll, increasing the concentration of soluble phosphate and reducing the concentration of Cr(VI) in root and shoot. The results of metagenomic analysis showed that changes in biological community structure, enrichment of related functional abundances, and activation of genes (FTH1, FTL, HSP90A, glgP, htpG, glnA, and GLUD1_2) located in microbes contributed to the microbial-phytoremediation of Cr(VI) contamination. These results provide a reference for future research on the microbial-phytoremediation of Cr(VI) contamination in phosphate mining wasteland.

Abstract:Heavy metal contaminations may cause severe toxic impacts to ecological systems and human health. Mea-surements of metals' bioavailable concentrations and toxicities simultaneously and in-situ in environments can advance the understanding of their hazardous effects. The diffusive gradients in thin-films (DGT) is an in-situ technique can measure metal speciation and labile concentrations, but cannot yet provide the direct toxicity information. The whole-cell bioreporter Acinetobacter baylyi ADPWH_recA was successfully incorporated into the DGT device to develop a novel technique, Bio-DGT, for assessing the toxicity of metals at the same time of measuring their labile concentrations. The bioassay used in Bio-DGT can sense the mixture toxicity from multiple contaminants and the DGT can assist in identifying which toxicants may be causing the toxicity. Cadmium was used as the model metal to test the performance of Bio-DGT in waters and soils. The masses of Cd accumulated in Bio-DGT increased linearly and theoretically with time for 7 days deployment, indicating little influences from bioreporter cells on DGT performance. A positive relationship between bioluminescent signals towards Cd demonstrated the sensitive and active bioreporters' response. The sensitive of Bio-DGT, indicated by Cd con-centrations causing the response, is 0.01 mg/L. The stable response from Bio-DGT under various conditions (pH 4-8, ionic strengths 0.01-0.5 M) and 30 days storage time suggest the applicability of the technique in real environments. The deployment of Bio-DGT in contaminated soils demonstrated that Cd toxicity was regulated by labile concentration, showing its potential application for the risk assessment of heavy metal contaminations, and its further feasibility in using Bio-DGT for measuring integration of multiple contaminants' effects and simul-taneously determine the main toxicity driver(s).
Chromium-Resistant Bacteria Promote the Reduction of Hexavalent Chromium in Soils

Abstract:Knowledge of the chromium (Cr) redox process in soil is important in addressing Cr bioavailability and risk assessment of contaminated soils. In this study, seven representative agricultural soils with different physicochemical properties were used to investigate the importance of microbially mediated Cr(VI) reduction and the response of soil bacterial community to Cr contamination. Chromium application increased soil bacterial diversity in Periudic Argosols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols, with an exception of Udic Ferrisols. The soil bacterial community responded to Cr contamination through changes in bacterial community structure, with Cr-resistant bacteria becoming the dominant species, and the percentage of Cr-resistant bacteria of total cultivable bacteria was 89.9, 75.2, 92.8, 65.3, 72.8, 77.3, and 65.4%, respectively, for Periudic Argosols, Udic Ferrisols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols. , , , , , , , , , and were identified as the Cr-resistant bacteria. Moreover, our results demonstrated that microbial reduction was an important Cr(VI) reduction pathway, and the relative contribution of microorganisms to Cr(VI) reduction was 14.4, 44.0, 20.6, 34.9, 21.9, 21.7, and 22.0%, respectively for Periudic Argosols, Udic Ferrisols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols. Soil properties, especially Fe(II) and soil particle distribution, affected the microbially mediated Cr(VI) reduction. These results provide useful information for the bioremediation of Cr-contaminated soils under a wide range of environmental conditions.
Isolation and Characterization of Chromium(vi)-Reducing Bacillus Sp. FY1 and Arthrobacter Sp. WZ2 and Their Bioremediation Potential

Abstract:Two native bacterial strains, FY1 and WZ2, that showed high chromium(VI)-reducing ability were respectively isolated from electroplating and tannery effluent-contaminated sites and identified as Bacillus and Arthrobacter. The objective of the present study was to evaluate their potential for future application in soil bioremediation. The results showed that both Bacillus sp. FY1 and Arthrobacter sp. WZ2 were tolerant to 1000mg L-1 Cr(VI) and capable of reducing 78-85% and 75-82% of Cr(VI) (100-200mg L-1) within 24h, respectively. The Cr(VI) reduction rate decreased with increasing levels of Cr(VI) concentration (200-1000mg L-1). The optimum pH, temperature, and inoculum concentration for Cr(VI) reduction were found to be between pH 7.0 and 8.0; 30 and 35 degrees C; and 1x 10(8) cells ml(-1), respectively. Further evidence for the bioremediation potential of Bacillus sp. FY1 and Arthrobacter sp. WZ2 was provided by the high capacity to reduce 100, 200, and 500mg kg(-1) Cr(VI) in contaminated soil by 83-91%, 78-85%, and 71-78% within 7days, respectively. These findings demonstrated the high potential of Bacillus sp. FY1 and Arthrobacter sp. WZ2 for application in future soil bioremediation.

Abstract:The understanding of bacterial resistance to hexavalent chromium [Cr(VI)] are crucial for the enhancement of Cr(VI)-polluted soil bioremediation. However, the mechanisms related to plant-associated bacteria remain largely unclear. In this study, we investigate the resistance mechanisms and remediation potential of Cr(VI) in a plant-associated strain, AN-B15. The results manifested that AN-B15 efficiently reduced Cr(VI) to soluble organo-Cr(III). Specifically, 84.3 % and 56.5 % of Cr(VI) was removed after 48 h in strain-inoculated solutions supplemented with 10 and 20 mg/L Cr(VI) concentrations, respectively. Transcriptome analyses revealed that multiple metabolic systems are responsible for Cr(VI) resistance at the transcriptional level. In response to Cr(VI) exposure, strain AN-B15 up-regulated the genes involved in central metabolism, providing the reducing power by which enzymes (ChrR and azoR) transformed Cr(VI) to Cr(III) in the cytoplasm. Genes involved in the alleviation of oxidative stress and DNA repair were significantly up-regulated to neutralize Cr(VI)-induced toxicity. Additionally, genes involved in organosulfur metabolism and certain ion transporters were up-regulated to counteract the starvation of sulfur, molybdate, iron, and manganese induced by Cr(VI) stress. Furthermore, a hydroponic culture experiment showed that toxicity and uptake of Cr(VI) by plants under Cr(VI) stress were reduced by strain AN-B15. Specifically, strain AN-B15 inoculation increased the fresh weights of the wheat root and shoot by 55.5 % and 18.8 %, respectively, under Cr(VI) stress (5 mg/L). The elucidation of bacterial resistance to Cr(VI) has an important implication for exploiting microorganism for the effective remediation of Cr(VI)-polluted soils.

Abstract:We investigated the quantification of bioavailable arsenic in contaminated soils and evaluation of soil-washing processes in the aspect of bioavailability using a novel bacterial bioreporter developed in present study. The whole-cell bioreporter (WCB) was genetically engineered by fusing the promoter of nik operon from Escherichia coli and green fluorescent protein as a sensing domain and reporter domain. Among eight well-known hazardous heavy metals and metalloid, this system responded specifically to arsenic, thereby inferring association of As(III) with NikR inhibits the repression. Moreover, the response was proportional to the concentration of As(III), thereby it was capable to determine the amount of bioavailable arsenic quantitatively in contaminated soils. The bioavailable portion of arsenic was 5.9 (3.46-10.96) and 0.9 (0.27-1.74) % of total from amended and site soils, respectively, suggesting the bioavailability of arsenic in soils was related to the soil properties and duration of aging. On the other hand, only 1.37 (0.21-2.97) % of total arsenic was extracted into soil solutions and 19.88 (11.86-28.27) % of arsenic in soil solution was bioavailable. This result showed that the soluble arsenic is not all bioavailable and most of bioavailable arsenic in soils is water non-extractable. In addition, the bioavailable arsenic was increased after soil-washing while total amount was decreased, thereby suggesting the soil-washing processes release arsenic associated with soil materials to be bioavailable. Therefore, it would be valuable to have a tool to assess bioavailability and the bioavailability should be taken into consideration for soil remediation plans.

Abstract:The simulated in-vitro gastrointestinal method provides a simple way to evaluate the health risk of human body exposed to soil contaminants. Several in-vitro methods have been successfully established for soil As, Pb, and Cd. However, the method development for soil Cr failed up to now, which could be resulted from alteration in the species of Cr (e.g., Cr(VI)/Cr(III)) caused by the gastrointestinal digestion components, ultimately affecting the accessibility of Cr. This study explored the transformation and bioaccessibility of Cr in two Cr-contaminated soils during the physiologically based extraction test. The water-soluble and exchangeable Cr in soil was dissolved in gastrointestinal tract, accompanied with reduction of Cr(VI) into Cr(III), and the reduction occurred after the chemical extraction in two soils rather than during the extraction. Pepsin and organic acids in gastric phase could reduce Cr(VI) into Cr(III) and reduction efficiency were 20.4%- 53.0%, while in intestinal phase, pancreatin and bile salt had little effect on the Cr(VI) reduction, instead, more Cr(VI) was released from soil. In the gastric solution, Cr(VI) was mainly present as HCrO4- and Cr(III) as free Cr3+ ion. In the intestinal phase, Cr(VI) mainly occurred as CrO42- and Cr(III) as Cr(OH)3 (aq). Cr in the soil solid phase was dominated as the precipitates of Cr-Fe oxide, which was hardly extracted. Bioaccessibility of Cr in gastric phase increased as extraction duration increased and decreased in the intestinal phase, the contrary trend was observed for the hazard quotient of Cr in two phases due to Cr(VI)/Cr(III) transformation. This study indicates that the gastrointestinal components could influence the Cr transformation and subsequently affect the Cr bioaccessibility, which would help for a successful establishment of in vitro determination method for soil Cr bioaccessibility.

Whole-cell bioreporters and risk assessment of environmental pollution: A proof-of-concept study using lead

Abstract:As the world burden of environmental contamination increases, it is of the utmost importance to develop streamlined approaches to environmental risk assessment in order to prioritize mitigation measures. Whole-cell biosensors or bioreporters and speciation modeling have both become of increasing interest to determine the bioavailability of pollutants, as bioavailability is increasingly in use as an indicator of risk. Herein, we examine whether bioreporter results are able to reflect expectations based on chemical reactivity and speciation modeling, with the hope to extend the research into a wider framework of risk assessment. We study a specific test case concerning the bioavailability of lead (Pb) in aqueous environments containing Pb-complexing ligands. Ligands studied include ethylene diamine tetra-acetic acid (EDTA), meso-2,3 dimercaptosuccinic acid (DMSA), leucine, methionine, cysteine, glutathione, and humic acid (HA), and we also performed experiments using natural water samples from Lake Tai (Taihu), the third largest lake in China. We find that EDTA, DMSA, cysteine, glutathione, and HA amendment significantly reduced Pb bioavailability with increasing ligand concentration according to a log-sigmoid trend. Increasing dissolved organic carbon in Taihu water also had the same effect, whereas leucine and methionine had no notable effect on bioavailability at the concentrations tested. We find that bioreporter results are in accord with the reduction of aqueous Pb2+ that we expect from the relative complexation affinities of the different ligands tested. For EDTA and HA, for which reasonably accurate ionization and complexation constants are known, speciation modeling is in agreement with bioreporter response to within the level of uncertainty recognised as reasonable by the United States Environmental Protection Agency for speciation-based risk assessment applications. These findings represent a first step toward using bioreporter technology to streamline the biological confirmation or validation of speciation modeling for use in environmental risk assessment.
