Abstract:Salt stress is a major abiotic stress factor affecting crop production, and understanding of the response mechanisms of seed germination to salt stress can help to improve crop tolerance and yield. The differences in regulatory pathways during germination in different salt-tolerant barley seeds are not clear. Therefore, this study investigated the responses of different salt-tolerant barley seeds during germination to salt stress at the proteomic and metabolic levels. To do so, the proteomics and metabolomics of two barley seeds with different salt tolerances were comprehensively examined. Through comparative proteomic analysis, 778 differentially expressed proteins were identified, of which 335 were upregulated and 443 were downregulated. These proteins, were mainly involved in signal transduction, propanoate metabolism, phenylpropanoid biosynthesis, plant hormones and cell wall stress. In addition, a total of 187 salt-regulated metabolites were identified in this research, which were mainly related to ABC transporters, amino acid metabolism, carbohydrate metabolism and lipid metabolism; 72 were increased and 112 were decreased. Compared with salt-sensitive materials, salt-tolerant materials responded more positively to salt stress at the protein and metabolic levels. Taken together, these results suggest that salt-tolerant germplasm may enhance resilience by repairing intracellular structures, promoting lipid metabolism and increasing osmotic metabolites. These data not only provide new ideas for how seeds respond to salt stress but also provide new directions for studying the molecular mechanisms and the metabolic homeostasis of seeds in the early stages of germination under abiotic stresses.
Comprehensive Transcriptome and Proteome Analyses Reveal a Novel Sodium Chloride Responsive Gene Network in Maize Seed Tissues During Germination

Abstract:Germination is a plant developmental process by which radicle of mature seeds start to penetrate surrounding barriers for seedling establishment and multiple environmental factors have been shown to affect it. Little is known how high salinity affects seed germination of C4 plant, Zea mays. Preliminary germination assay suggested that isolated embryo alone was able to germinate under 200 mM NaCl treatment, whereas the intact seeds were highly repressed. We hypothesized that maize endosperm may function in perception and transduction of salt signal to surrounding tissues such as embryo, showing a completely different response to that in Arabidopsis. Since salt response involves ABA, we analysed in vivo ABA distribution and quantity and the result demonstrated that ABA level in isolated embryo under NaCl treatment failed to increase in comparison with the water control, suggesting that the elevation of ABA level is an endosperm dependent process. Subsequently, by using advanced profiling techniques such as RNA sequencing and SWATH-MS-based quantitative proteomics, we found substantial differences in post-transcriptional and translational changes between salt-treated embryo and endosperm. In summary, our results indicate that these regulatory mechanisms, such as alternative splicing, are likely to mediate early responses to salt stress during maize seed germination.
[Efficacy and safety of coupled plasma filtration adsorption combined with continuous veno-venous hemofiltration for multiple organ dysfunction syndrome patients with acute liver failure].

Abstract:OBJECTIVE To evaluate efficacy and safety of coupled plasma filtration adsorption (CPFA) combined with continuous veno-venous hemofiltration (CVVH) for the treatment of multiple organ dysfunction syndrome (MODS) patients with acute liver failure (ALF), and to evaluate the effect of CPFA plus CVVF on inflammatory mediators in these patients. METHODS A total of 38 cases of 11 MODS patients with ALF (male 6, female 5) were treated with CPFA plus CVVH, and the following clinical indicators including changes in mean arterial pressure (MAP), oxygen index (PaO(2)/FiO(2)), tumor necrosis factor-alpha (TNF-alpha), interleukin-1 beta (IL-1 beta), IL-6, IL-8, biochemical parameters of liver and kidney function, parameters of systemic inflammatory response syndrome (SIRS) score, and acute physiology and chronic health evaluation II (APACHE II) score were determined before and after the treatment. The degree of improvement in clinical symptoms, feasibility, tolerance toward CPFA plus CVVH, therapy-related adverse reactions and security were simultaneously evaluated. RESULTS MAP increased by 12 mmHg (1 mmHg=0.133 kPa), and PaO(2)/FiO(2) increased by 40 mmHg after the application of CPFA plus CVVH (both P<0.05), along with significant decrease in TNF-alpha, IL-1 beta, IL-6, IL-8 and markedly lowered levels of serum total bilirubin (IBIL), direct bilirubin (DBIL), blood urea nitrogen (BUN), serum creatinine (SCr) and blood ammonia (all P<0.05). Besides, clinical symptoms, including urinary volume, mental disturbance, jaundice, debility, nausea, vomiting, fever, abdominal distention, anepithymia, and SIRS, APACHE II scores were improved significantly after the CPFA plus CVVH (all P<0.05). No therapy-related adverse reactions, including severe haemorrhage, shock, hypersensitivity, were noted, and patients tolerated well toward CPFA plus CVVH. The total survival rate of patients was 45.5% (5/11 cases) at the end of the treatment. CONCLUSION Our data indicate that CPFA combined with CVVH is an effective and safe method to improve the prognosis of MODS patients with ALF, the mechanism of which may be related to its effective removal of inflammatory cytokines.
Ameliorating effect of calcium on primary root elongation of soybean under sodium stress

Abstract:The present study was carried out to investigate the ameliorating effect of calcium on salt-induced inhibitory effects on primary root elongation. Following imbibition and germination, primary roots of soybean (Glycine max (L.) MERR.) cv. Dare, 40 +/- 0.4 mm in length, were exposed for 9 h to solutions of NaCl (10, 15, 25, 50, 100 mM) and 100 mM NaCl + 0.15, 1.0 or 5.0 mM CaCl2. The presence of Na+ significantly inhibited primary root elongation and Ca2+ significantly alleviated this inhibition. The NaCl treatments significantly increased Na+, decreased K+ and Ca2+ contents in elongation, middle and mature zones of primary roots compared with those of controls, respectively. Addition of Ca2+ significantly decreased Na+, increased K+ and Ca2+ contents in elongation, middle and mature zones of primary roots comparing to those deprived of Ca2+, respectively. Thus, significantly higher K/Na and Ca/Na ratios were maintained in the former than the later roots. The decreasing gradient of Na+ accumulation from root tip towards hypocotyl indicated that Na+ exclusion ability was increasing with cell maturation. Results of Xray microanalysis showed that the epidermis cells accumulated largest fraction of Ca2+ among the three ions, compared with those of cortex and stele cells. The Ca/Na and K/Na ratios were higher in epidermis than in cortex. These results suggest that Ca2+ interacted with Na+ mostly in the epidermis. As a result, the amount of Na+ entered the primary roots was reduced and hence the adverse effects of Na+ on primary root elongation were ameliorated.
Integrated Physiological, Transcriptomic, and Metabolomic Analyses Revealed Molecular Mechanism for Salt Resistance in Soybean Roots

Abstract:Salinity stress is a threat to yield in many crops, including soybean (Glycine max L.). In this study, three soybean cultivars (JD19, LH3, and LD2) with different salt resistance were used to analyze salt tolerance mechanisms using physiology, transcriptomic, metabolomic, and bioinformatic methods. Physiological studies showed that salt-tolerant cultivars JD19 and LH3 had less root growth inhibition, higher antioxidant enzyme activities, lower ROS accumulation, and lower Na+ and Cl- contents than salt-susceptible cultivar LD2 under 100 mM NaCl treatment. Comparative transcriptome analysis showed that compared with LD2, salt stress increased the expression of antioxidant metabolism, stress response metabolism, glycine, serine and threonine metabolism, auxin response protein, transcription, and translation-related genes in JD19 and LH3. The comparison of metabolite profiles indicated that amino acid metabolism and the TCA cycle were important metabolic pathways of soybean in response to salt stress. In the further validation analysis of the above two pathways, it was found that compared with LD2, JD19, and LH3 had higher nitrogen absorption and assimilation rate, more amino acid accumulation, and faster TCA cycle activity under salt stress, which helped them better adapt to salt stress. Taken together, this study provides valuable information for better understanding the molecular mechanism underlying salt tolerance of soybean and also proposes new ideas and methods for cultivating stress-tolerant soybean.

It has been widely suggested that calcium (Ca) application ameliorates salt stress, but characteristic changes in root cell wall due to Ca application under saline conditions are poorly documented. Our objectives were: (1) to determine the effect of Ca on root cell wall composition, using two soybean cultivars differing in sensitivity to salt stress and (2) to understand the relationship between the internal effects of sodium–calcium interaction on the root cell wall. Uniform seedlings were transplanted into mixed solutions of NaCl (0, 40 mM,) and CaCl2 (0, 0.5, 2 mM). Root lengths were measured after an exposure of 14, 24 and 40 h to the treatments and cell wall analysis performed for total sugars, uronic acid and ion contents. Without salinity stress, Ca application caused no significant changes in root growth and cell wall constituents in both cultivars. However, it did ameliorate the decrease in the amount of cell wall under stress, especially the pectin fraction. Both cell wall and cellular Ca2+ and K+ contents were significantly increased by additional Ca2+ under saline condition. Therefore, by applying Ca2+, the maintenance of pectin level and increase in cell wall Ca2+ may contribute to the restoration of root growth under salinity. Calcium application significantly increased the pectin level under salinity and soybean root growth also showed notable restoration. One way Ca ameliorates salt toxicity may be by maintaining the composition of the cell wall. This ameliorative effect was more conspicuous in the salt-tolerant cultivar, Dare, than the salt-sensitive cultivar, Touzan 69.

Abstract:To investigate the effects of exogenous Spd on proteomic changes under normal condition and NaCl stress of 3 days in cucumber seedling leaves, a 2-DE gel electrophoresis and MALDI-TOF/TOF MS was performed. A total of 63 differentially expressed proteins responded to salt stress or exogenous Spd treatments, and they were all successfully identified by MALDI-TOF/TOF MS. Many changes were observed in the levels of proteins involved in energy and metabolic pathways, protein metabolic, stress defense, and other functional proteins. Increased salt tolerance by exogenous Spd would contribute to higher expressions of proteins involved in the SAMs metabolism, protein biosynthesis, and defense mechanisms on antioxidant and detoxification. Meanwhile, the regulation of Calvin cycle, protein folding assembly and the inhibition of protein proteolysis by Spd might play important roles in salt tolerance. This study provides insight that may facilitate a better understanding of the salt resistance by Spd in cucumber seedlings.

Abstract:Salt-alkali stress is a key factor affecting plant development and production, altering essential physiological and metabolic processes within plants. Particularly, soybean ( Glycine max ) exhibits a wide range of saline-alkali tolerance in soybean germplasms. Hence, the objective of this research was to pinpoint potential genes that react to saline-alkali stress through a comparative transcriptome analysis of two distinct soybean varieties exposed to such stress conditions. The two soybean varieties, including salt-tolerant Heinong531 and salt-sensitive 20_1846, were used for RNA-seq analysis. Nine physiological parameters including malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), peroxidase (POD), proline, catalase (CAT), soluble protein, scavenging rate of hydroxyl radical, and soluble sugar activity were measured under the saline-alkaline treatment series of 0,1,2,3,4 days. The results showed that the levels of MDA, proline, and catalase contents decreased significantly, while levels of GSH and POD contents significantly increased. A total of 200 saline-alkali tolerance responsive differentially expressed genes (DEGs) were identified, comprising 93 up-regulated genes and 107 down-regulated. The functional gene ontology analysis for these 200 DEGs, where 29 key DEGs were found to be involved in the regulation of 14 GO pathways responding to salt-alkaline stress which have been previously considered as vital for salt resistance mechanisms in soybean. Remarkably, the association analysis between physiological parameters and Differentially Expressed Genes (DEGs) under salt-alkali stress conditions demonstrated that distinct soybean cultivars exhibited unique patterns of gene expression, ultimately resulting in alterations in physiological indices as a response to salt stress. This study offers a compilation of potential genes worthy of additional exploration concerning their role in enhancing soybean's resistance to salt-alkali stress.

Comparing transcriptome expression profiles to reveal the mechanisms of salt tolerance and exogenous glycine betaine mitigation in maize seedlings

Abstract:Salt stress is a common abiotic stress that limits the growth, development and yield of maize (Zea mays L.). To better understand the response of maize to salt stress and the mechanism by which exogenous glycine betaine (GB) alleviates the damaging effects of salt stress, the morphology, physiological and biochemical indexes, and root transcriptome expression profiles of seedlings of salt-sensitive inbred line P138 and salt-tolerant inbred line 8723 were compared under salt stress and GB-alleviated salt stress conditions. The results showed that under salt stress the growth of P138 was significantly inhibited and the vivo ion balance was disrupted, whereas 8723 could prevent salt injury by maintaining a high ratio of K+ to Na+. The addition of a suitable concentration of GB could effectively alleviate the damage caused by salt stress, and the mitigating effect on salt-sensitive inbred line P138 was more obvious than that on 8723. Transcriptome analysis revealed that 219 differentially expressed genes (DEGs) were up-regulated and 153 DEGs were down-regulated in both P138 and 8723 under NaCl treatment, and that 487 DEGs were up-regulated and 942 DEGs were down-regulated in both P138 and 8723 under salt plus exogenous GB treatment. In 8723 the response to salt stress is mainly achieved through stabilizing ion homeostasis, strong signal transduction activation, increasing reactive oxygen scavenging. GB alleviates salt stress in maize mainly by inducing gene expression changes to enhance the ion balance, secondary metabolic level, reactive oxygen scavenging mechanism, signal transduction activation. In addition, the transcription factors involved in the regulation of salt stress response and exogenous GB mitigation mainly belong to the MYB, MYB-related, AP2-EREBP, bHLH, and NAC families. We verified 10 selected up-regulated DEGs by quantitative real-time polymerase chain reaction (qRT-PCR), and the expression results were basically consistent with the transcriptome expression profiles. Our results from this study may provide the theoretical basis for determining maize salt tolerance mechanisms and the mechanism by which GB regulates salt tolerance.

γ-Aminobutyric Acid Imparts Partial Protection from Salt Stress Injury to Maize Seedlings by Improving Photosynthesis and Upregulating Osmoprotectants and Antioxidants

Abstract:Abstractγ-Aminobutyric acid (GABA) has high physiological activity in plant stress physiology. This study showed that the application of exogenous GABA by root drenching to moderately (MS, 150 mM salt concentration) and severely salt-stressed (SS, 300 mM salt concentration) plants significantly increased endogenous GABA concentration and improved maize seedling growth but decreased glutamate decarboxylase (GAD) activity compared with non-treated ones. Exogenous GABA alleviated damage to membranes, increased in proline and soluble sugar content in leaves, and reduced water loss. After the application of GABA, maize seedling leaves suffered less oxidative damage in terms of superoxide anion (O2·−) and malondialdehyde (MDA) content. GABA-treated MS and SS maize seedlings showed increased enzymatic antioxidant activity compared with that of untreated controls, and GABA-treated MS maize seedlings had a greater increase in enzymatic antioxidant activity than SS maize seedlings. Salt stress severely damaged cell function and inhibited photosynthesis, especially in SS maize seedlings. Exogenous GABA application could reduce the accumulation of harmful substances, help maintain cell morphology, and improve the function of cells during salt stress. These effects could reduce the damage to the photosynthetic system from salt stress and improve photosynthesis and chlorophyll fluorescence parameters. GABA enhanced the salt tolerance of maize seedlings.

Abstract:To characterize differences in soybean resistance to salt stress, two soybean species, the wild salt-tolerant soybean Glycine cyrtoloba (serial number ACC547) and the cultivated salt-sensitive soybean G. max (cv. Melrose) were treated with 0, 50, 100, or 150mmolL−1 NaCl for 5days. A series of physiological parameters were determined in both shoots and roots, including content of chlorophyll (Chl) and malondialdehyde (MDA); electrolyte leakage (EL); hydrogen peroxide (H2O2) concentration; superoxide oxygen radical (O2−) production rate; activities of several enzymes including superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD); and selective ion (Na+ and K+) accumulation. Our results showed that the relative salt tolerance of ACC547 was associated with lower loss of Chl content; lower MDA content, EL, H2O2 concentration, and O2− production rate in both shoots and roots; higher POD activity caused by new isoforms in roots; and higher K+ concentration and K+/Na+ ratio in shoots. These results suggested that relative lower membrane injury, efficient K+ vs. Na+ selective accumulation, and newly induced POD isoenzymes are mechanisms of salt tolerance in soybean.
[Physiological responses of symbiotic rhizobium pea to exogenous calcium under salt stress]

Abstract:To clarify the effects of exogenous calcium on salt tolerance of nodule symbiotic pea, a pot experiment was conducted under salt stress (170 mmol·L-1) to examine the effects of exogenous CaCl2(0, 5 and 15 mmol·L-1) on physiological parameters of two pea cultivars, 'Dingwan 8' (salt tolerant) and 'Longwan 6' (salt sensitive), inoculated with rhizobium strains 15657, 15735 and Ca66. The results showed that plant biomass, the activities of superoxide dismutase (SOD) and peroxidase (POD), and the contents of proline (Pro) and soluble sugar (SS) were enhanced, but malondialdehyde (MDA) content was decreased, with rhizobium inoculation, CaCl2 application or CaCl2 application after rhizobium inoculation. Plant biomass and activities of SOD and POD and Pro content were increased significantly by the application of 15 mmol·L-1 CaCl2 after rhizobium inoculation. The application of CaCl2 after inoculation of rhizobium strain 15735 suitable for pea had limited effects on physiological parameters of pea plants, whereas the application of CaCl2 after inoculation of rhizobium (15657, Ca66) unsuitable for pea had obvious effects. The comprehensive analysis of membership function showed that pea treated with CaCl2 after rhizobium inoculation exhibited stronger salt tolerance, and 'Dingwan 8' plants treated with 15 mmol·L-1 CaCl2 after inoculation of rhizobium strain 15735 showed the strongest ability of salt tolerance with the highest membership function value of 0.814. In summary, compared with CaCl2 application or rhizobium inoculation alone, CaCl2 application after rhizobium inoculation could effectively enhance the activities of antioxidant enzymes and osmotic regulation ability, reduce the damage of membrane lipid peroxidation, and consequently improve the salt tolerance of pea.
Physiological mechanisms of exogenous calcium on alleviating salinity-induced stress in rice (Oryza sativa L.)

Abstract:Being more sensitive to salt stress among the cereals, growth of rice (Oryza sativa L.) has been habitually affected by salinity. Although, several practices have evolved to sustain the growth of rice under salinity, the enormous role of calcium (Ca2+) as a signalling molecule in salt stress mitigation is still arcane. Considering this fact, an experiment was performed aiming to explicate the mechanism of salt-induced growth inhibition in rice and its alleviation by exogenous Ca2+. At germination stage, 10 mM and 15 mM CaCl2 primed rice (cv. Binadhan-10 & Binadhan-7) seeds were grown in petri dishes for 9 days under 100 mM NaCl stress. At seedling stage, 9-day-old rice seedlings grown on sand were exposed to 100 mM NaCl alone and combined with 10 mM and 15 mM CaCl2 for 15 days. This research revealed that salinity radically slowed down growth of rice seedlings and Ca2+ treatment noticeably improved growth performances. At germination stage, 10 mM CaCl2 treatment significantly increased the final germination percentage, germination rate index (in Binadhan-7), shoot, root length (89.20, 67.58% in Bindhan-10 & 84.72, 31.15% in Bindhan-7) and biomass production under salinity. Similarly, at seedling stage, 10 mM CaCl2 supplementation in salt-stressed plants enhanced shoot length (42.17, 28.76%) and shoot dry weight (339.52, 396.20%) significantly in Binadhan-10 & Binadhan-7, respectively, but enhanced root dry weight (36.76%) only in Binadhan-10. In addition, 10 mM CaCl2 supplementation on salt-stressed seedlings increased the chlorophyll and proline content, and oppressed the accretion of reactive oxygen species thus protecting from oxidative damage more pronouncedly in Binadhan-10 than Binadhan-7 as reflected by the elevated levels of catalase and ascorbate peroxidase activity. The 15 mM CaCl2 somehow also enhanced some growth parameters but overall was less effective than 10 mM CaCl2 to alleviate salt stress, and sometimes showed negative effect. Therefore, supplementary application of calcium-rich fertilizers in saline prone soils can be an effective approach to acclimatize salt stress and cultivate rice successfully.
Exogenous γ-Aminobutyric Acid Can Improve Seed Germination and Seedling Growth of Two Cotton Cultivars under Salt Stress

Abstract:Excessive salt content in soil has adverse effects on cotton production, especially during the germination and seedling stages. γ-aminobutyric acid (GABA) is an important active substance that is expected to improve the resistance of plants to abiotic stresses. This study focused on two cotton cultivars (Gossypium hirsutum L.: Tahe 2 and Xinluzhong 62) and investigated the impact of exogenous GABA (0, 1, 2, 3, and 4 mM) on seed germination, seedling growth, and related morphological, physiological, and biochemical indicators under salt stress (150 mM NaCl). The results showed that salt stress significantly reduced the germination rate and germination index of cotton seeds (decreased by 20.34% and 32.14% for Tahe 2 and Xinluzhong 62, respectively), leading to decreased seedling height and biomass and causing leaf yellowing. Salt stress induced osmotic stress in seedlings, resulting in ion imbalance (marked reduction in K+/Na+ ratio) and oxidative damage. Under salt stress conditions, exogenous GABA increased the germination rate (increased by 10.64~23.40% and 2.63~31.58% for Tahe 2 and Xinluzhong 62, respectively) and germination index of cotton seeds, as well as plant height and biomass. GABA treatment improved leaf yellowing. Exogenous GABA treatment increased the content of proline and soluble sugars, with varying effects on betaine. Exogenous GABA treatment reduced the Na+ content in seedlings, increased the K+ content, and increased the K+/Na+ ratio (increased by 20.44~28.08% and 29.54~76.33% for Tahe 2 and Xinluzhong 62, respectively). Exogenous GABA treatment enhanced the activities of superoxide dismutase and peroxidase, and reduced the accumulation of hydrogen peroxide and malondialdehyde, but had a negative impact on catalase activity. In conclusion, exogenous GABA effectively improved cotton seed germination. By regulating osmoprotectant levels, maintaining ion homeostasis, and alleviating oxidative stress, GABA mitigated the adverse effects of salt stress on cotton seedling growth.

Comparative Transcriptome Analysis of Salt-Tolerant and -Sensitive Soybean Cultivars under Salt Stress

Abstract:Soil salinity is a major limiting factor in soybean (Glycine max (L.) Merr.) yield in Xinjiang, China. Therefore, breeding soybean to tolerate highly saline soils is crucial to improve its yield. To explore the molecular mechanisms underlying the response of soybean to salt stress, we performed a comparative transcriptome analysis of root and leaf samples collected from two local soybean cultivars. The salt-tolerant cultivar 'Xin No. 9' (X9) showed higher photosynthetic activity than the salt-sensitive cultivar 'Xinzhen No. 9' (Z9) under salt stress. In total, we identified 13,180 and 13,758 differential expression genes (DEGs) in X9 and Z9, respectively, of which the number of DEGs identified in roots was much higher than that in leaves. We constructed the co-expression gene modules and conducted Gene Ontology (GO) term and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses. The results suggested there were distinct differences in the mechanisms of response to salt stress between the two soybean cultivars; i.e., the salt-tolerant cultivar X9 exhibited alterations in fundamental metabolism, whereas the salt-sensitive cultivar Z9 responded to salt stress mainly through the cell cycle. The possible crosstalk among phytohormone signaling, MAPK signaling, phenylpropanoid biosynthesis, starch and sucrose metabolism, and ribosome metabolism may play crucial roles in the response to salt stress in soybean. Our results offered a comprehensive understanding of the genes and pathways involved in the response to salt stress in soybean and provided valuable molecular resources for future functional studies and the breeding of soybean varieties with enhanced tolerance to salinity.
Phosphoproteomic Analysis of Two Contrasting Maize Inbred Lines Provides Insights into the Mechanism of Salt-Stress Tolerance

Abstract:Salinity is a major abiotic stress that limits maize yield and quality throughout the world. We investigated phosphoproteomics differences between a salt-tolerant inbred line (Zheng58) and a salt-sensitive inbred line (Chang7-2) in response to short-term salt stress using label-free quantitation. A total of 9448 unique phosphorylation sites from 4116 phosphoproteins in roots and shoots of Zheng58 and Chang7-2 were identified. A total of 209 and 243 differentially regulated phosphoproteins (DRPPs) in response to NaCl treatment were detected in roots and shoots, respectively. Functional analysis of these DRPPs showed that they were involved in carbon metabolism, glutathione metabolism, transport, and signal transduction. Among these phosphoproteins, the expression of 6-phosphogluconate dehydrogenase 2, pyruvate dehydrogenase, phosphoenolpyruvate carboxykinase, glutamate decarboxylase, glutamate synthase, l-gulonolactone oxidase-like, potassium channel AKT1, high-affinity potassium transporter, sodium/hydrogen exchanger, and calcium/proton exchanger CAX1-like protein were significantly regulated in roots, while phosphoenolpyruvate carboxylase 1, phosphoenolpyruvate carboxykinase, sodium/hydrogen exchanger, plasma membrane intrinsic protein 2, glutathione transferases, and abscisic acid-insensitive 5-like protein were significantly regulated in shoots. Zheng58 may activate carbon metabolism, glutathione and ascorbic acid metabolism, potassium and sodium transportation, and the accumulation of glutamate to enhance its salt tolerance. Our results help to elucidate the mechanisms of salt response in maize seedlings. They also provide a basis for further study of the mechanism underlying salt response and tolerance in maize and other crops.
Effects of Exogenous Calcium on Photosynthesis and Its Related Physiological Characteristics of Rice Seedlings Under Salt Stress

Abstract:Two rice (Oryza sativa L.) cultivars, Wuyujing No.3 (salt-tolerant) and IR36 (salt-sensitive), were used to investigate the effects of exogenous calcium on some photosynthetic and physiological characteristics of the leaves of rice grown under salt stress. The results showed that the photosynthesis and photosynthetic efficiency of the leaves of salt-stressed rice were significantly enhanced with the inclusion of Ca2+. The levels of net photosynthetic rate and stomatal conductance increased more significantly than the control without Ca2+, and the rate of increase or decrease of intercellular CO2 concentration and stomatal limitation value were faster than that of the controlwithout Ca2+ too. Addition of Ca2+ increased the contents of chlorophyll, proline and soluble sugar of the leaves of rice under salt stress. Furthermore, addition of Ca2+ reduced the content of MDA and the permeability of cell membrane, but increased the SOD activity.
Knockdown of β‐conglycinin α′ and α subunits alters seed protein composition and improves salt tolerance in soybean

Abstract:SUMMARY Soybean is an important plant source of protein worldwide. Increasing demands for soybean can be met by improving the quality of its seed protein. In this study, GmCG‐1, which encodes the β‐conglycinin α′ subunit, was identified via combined genome‐wide association study and transcriptome analysis. We subsequently knocked down GmCG‐1 and its paralogues GmCG‐2 and GmCG‐3 with CRISPR‐Cas9 technology and generated two stable multigene knockdown mutants. As a result, the β‐conglycinin content decreased, whereas the 11S/7S ratio, total protein content and sulfur‐containing amino acid content significantly increased. Surprisingly, the globulin mutant exhibited salt tolerance in both the germination and seedling stages. Little is known about the relationship between seed protein composition and the salt stress response in soybean. Metabonomics and RNA‐seq analysis indicated that compared with the WT, the mutant was formed through a pathway that was more similar to that of active salicylic acid biosynthesis; however, the synthesis of cytokinin exhibited greater defects, which could lead to increased expression of plant dehydrin‐related salt tolerance proteins and cell membrane ion transporters. Population evolution analysis suggested that GmCG‐1, GmCG‐2, and GmCG‐3 were selected during soybean domestication. The soybean accessions harboring GmCG‐1Hap1 presented relatively high 11S/7S ratios and relatively high salt tolerance. In conclusion, knockdown of the β‐conglycinin α and α′ subunits can improve the nutritional quality of soybean seeds and increase the salt tolerance of soybean plants, providing a strategy for designing soybean varieties with high nutritional value and high salt tolerance.

Abstract:Calcium ions function as key messengers in the context of intracellular signal transduction. The ability of plants to respond to biotic and abiotic stressors is highly dependent on the calcineurin B-like protein (CBL) and CBL-interacting protein kinase (CIPK) signaling network. Here, a comprehensive effort was made to identify all members of the soybean CBL gene family, leading to the identification of 15 total genes distributed randomly across nine chromosomes, including 13 segmental duplicates. All the GmCBL gene subfamilies presented with similar gene structures and conserved motifs. Analyses of the expression of these genes in different tissues revealed that the majority of these GmCBLs were predominantly expressed in the roots. Significant GmCBL expression and activity increases were also observed in response to a range of stress-related treatments, including salt stress, alkaline stress, osmotic stress, or exposure to salicylic acid, brassinosteroids, or abscisic acid. Striking increases in GmCBL1 expression were observed in response to alkaline and salt stress. Subsequent analyses revealed that GmCBL1 was capable of enhancing soybean salt and alkali tolerance through the regulation of redox reactions. These results offer new insight into the complex mechanisms through which the soybean CBL gene family regulates the responses of these plants to environmental stressors, highlighting promising targets for efforts aimed at enhancing soybean stress tolerance.

Abstract:Abstract Background Salt stress is a serious abiotic stress that caused crop growth inhibition and yield decline. Previous studies have reported on the the synthesis of gamma-aminobutyric acid (GABA) and its relationship with plant resistance under various abiotic stress. However, the relationship between exogenous GABA alleviating plant salt stress damage and ion flux, amino acid synthesis, and key enzyme expression remains largely unclear. We investigated plant growth, Na + transportation and accumulation, reactive oxygen species (ROS) metabolism and evaluated the effect of GABA on amino acids, especially SlGADs gene expression and the endogenous GABA content of tomato ( Solanum lycopersicum L.) seedlings treated with or without 5 mmol·L − 1 GABA under 175 mmol·L − 1 NaCl stress. Results Exogenous application of GABA significantly reduced the salt damage index and increased plant height, chlorophyll content and the dry and fresh weights of tomato plants exposed to NaCl stress. GABA significantly reduced Na + accumulation in leaves and roots by preventing Na + influx in roots and transportation to leaves. The transcriptional expression of SlGAD1–3 genes were induced by NaCl stress especially with GABA application. Among them, SlGAD1 expression was the most sensitive and contributed the most to the increase in glutamate decarboxylase (GAD) activity induced by NaCl and GABA application; Exogenous GABA increased GAD activity and amino acid contents in tomato leaves compared with the levels under NaCl stress alone, especially the levels of endogenous GABA, proline, glutamate and eight other amino acids. These results indicated that SlGADs transcriptional expression played an important role in tomato plant resistance to NaCl stress with GABA application by enhancing GAD activity and amino acid contents. GABA significantly alleviated the active oxygen-related injury of leaves under NaCl stress by increasing the activities of antioxidant enzymes and decreasing the contents of active oxygen species and malondialdehyde. Conclusion Exogenous GABA had a positive effect on the resistance of tomato seedlings to salt stress, which was closely associated with reducing Na + flux from root to leaves, increasing amino acid content and strengthening antioxidant metabolism. Endogenous GABA content was induced by salt and exogenous GABA at both the transcriptional and metabolic levels.
