Abstract:OBJECTIVE:The aim of this study was to determine the profiles of periodontopathogenic bacteria in a Chinese population using quantitative real-time polymerase chain reaction (qRT-PCR).MATERIALS AND METHODS:Twenty-four periodontally healthy Chinese subjects and 60 patients with chronic periodontitis (CP) were enrolled in this cross-sectional study. qRT-PCR was used to quantify Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum, Porphyromonas gingivalis, and Prevotella intermedia as well as total bacterial counts from 252 samples collected from the saliva, supragingival plaque, and subgingival plaque of all 84 subjects.RESULTS:The detection frequency of A. actinomycetemcomitans was less than 50%. F. nucleatum was detected in all subjects and CP patients had higher bacterial loads than healthy subjects. The median proportion of F. nucleatum was significantly higher in subgingival plaque than in supragingival plaque and saliva. P. gingivalis and P. intermedia had higher detection frequencies and bacterial loads in CP patients than in healthy subjects. The median proportion of P. gingivalis was significantly different among the three intraoral locations in the CP group and its proportion in subgingival plaque was 9.01%. Moreover, strong positive Spearman's correlations were found in A. actinomycetemcomitans, P. gingivalis, and P. intermedia counts across the three intraoral locations.CONCLUSION:The presence and bacteria loads of these four bacteria in this Chinese population are similar to those from other populations.CLINICAL RELEVANCE:Examination of bacterial detection frequency and loads in Chinese adults may assist microbial studies of periodontal disease and will shed light on periodontal disease diagnosis and treatment using antibiotics in the Chinese population.

Abstract:In the title compound C26H25NO7, the mean plane through the lactone-substituted ring of the pyrrolizidine moiety forms dihedral angles of 78.46 (6) and 58.28 (8)° with the acenaphthylene moiety and the sugar based-lactone ring, respectively. The sum of the angles at the the N atom of the pyrrolizidine ring (335.0°) is in accordance with sp 3 hybridization. Some atoms of the acetate group are disordered and were refined using a split model [occupancy ratio 0.673 (10):0.327 (10)].
qPCR Detection and Quantification of Aggregatibacter actinomycetemcomitans and Other Periodontal Pathogens in Saliva and Gingival Crevicular Fluid among Periodontitis Patients

Abstract:Objective: The detection of special bacterial species in patients with periodontitis is considered useful for clinical diagnosis and treatment. The aim of this study was to investigate the presence of specific periopathogens and investigate whether there is a correlation between the results of different bacterial species in whole saliva and pooled subgingival plaque samples (healthy and diseased sites) from individuals with periodontitis and periodontally healthy subjects. Materials and methods: In total, 52 patients were recruited and divided into two groups: non-periodontitis and periodontitis patients. For each group, the following periodontal pathogens were detected using real-time polymerase chain reaction: A. actinomycetemcomitans JP2 clone, A. actinomycetemcomitans non JP2 clone, Porphyromonasgingivalis, and total eubacteria. Results: Higher levels of the various studied bacteria were present in both saliva and plaque samples from the periodontitis group in comparison to non-periodontitis subjects. There were significant differences in P. gingivalis and A. actinomycetemcomitans JP2 clones in the saliva of periodontitis patient compared to the control group. Subgingival plaque of diseased sites presented a significant and strong positive correlation between A. actinomycetemcomitans and P. gingivalis. In saliva samples, there was a significant positive correlation between A. actinomycetemcomitans JP2 clone and P. gingivalis (p ≤ 0.002). Conclusion: Quantifying and differentiating these periodontal species from subgingival plaque and saliva samples showed a good potential as diagnostic markers for periodontal disease. Regarding the prevalence of the studied bacteria, specifically A. actinomycetemcomitans JP2 clone, found in this work, and the high rate of susceptibility to periodontal species in Africa, future larger studies are recommended.

Abstract:OBJECTIVE:To establish a 16S rDNA multiplex polymerase chain reaction (PCR) for simultaneously detecting P. gingivalis, A. actinomycetemcomitans and T. denticola in clinical specimens of chronic periodontitis and to study the correlation between different modes of infection and severity of the disease.METHODS:Periodontal pocket specimens from 152 patients with mild, moderate or advanced chronic periodontitis and gingival sulcus specimens from 30 periodontally healthy individuals were collected and placed in 200 microl lysis buffer. The specimens were incubated in 100 degrees C for 10 min and 10 microl of the supernatant was directly used as PCR template. DNAs from P. gingivalis strain ATCC33277, A. actinomycetemcomitans strain Y4, T. denticola strain FM and E. coli strain DH5alpha were used as positive and negative controls in PCR with all of which were prepared by routing phenol-chloroform method. A multiplex PCR assay, using three sets of primers specific to 16S rDNA genes of the three anaerobes, was developed to detect the specimens. The target amplification fragments from 3 cases of PCR products positive for all the three anaerobes were sequenced after T-A cloning. Chi-square test was applied to analyze the correlation between different coinfection of the three anaerobes and severity of the disease.RESULTS:The established 16S rDNA multiplex PCR assay was able to detect P. gingivalis, A. actinomycetemcomitans and T. denticola at a minimum of 10, 20 and 20 cells, respectively. In comparison with the reported corresponding sequences, similarities of the nucleotide sequences from the three anaerobes 16S rDNA amplification fragments were as high as 99.45%, 97.08% and 96.59%, respectively. Of the 30 periodontally healthy gingival sulcus specimens, only one (3.3%) positive for P. gingivalis and two (6.7%) for A. actinomycetemcomitans could be identified but all of the rest were negative. In the 152 CP periodontal pocket specimens, 147 cases (96.7%) were positive for P. gingivalis, A. actinomycetemcomitans and/or T. denticola, respectively, and 5 cases (3.3%) were negative for all the three anaerobes. The positive rate of P. gingivalis detection (91.5%, 139/152) was significantly higher than those of A. actinomycetemcomitans (72.4%, 110/152) and T. denticola (80.9%, 123/152) (chi(2) = 7.07, 18.67; P < 0.01). 89.8% of the specimens from patients showed coinfections with two (26.5%) or three anaerobes (63.3%), and the coinfection rates in the specimens from moderate and advanced CP were remarkably higher than that from mild CP (chi(2) = 10.43, P < 0.01).CONCLUSION:The 16S rDNA multiplex PCR established in this study showed high sensitivity and specificity, which could be used to detect P. gingivalis, A. actinomycetemcomitans and T. denticola in clinical specimens. CP was an disease caused by multiple pathogenic microbes while the synergistic pathopoiesis of the three microbes was closely related to the severity of the disease.
Comparison of subgingival bacterial sampling with oral lavage for detection and quantification of periodontal pathogens by real-time polymerase chain reaction

Abstract:Background: Saliva has been studied for the presence of subgingival pathogens in periodontitis patients. With the anaerobic culture technique, the discrepancy between salivary recovery and subgingival presence has been significant, which makes this approach not suitable for practical use in the microbial diagnosis of periodontitis patients. The real-time polymerase chain reaction (PCR) technique represents a very sensitive technique to detect and quantify bacterial pathogens. The aim of the study was to compare the presence and numbers of Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythensis, Prevotella intermedia, and Micromonas micros in subgingival plaque and mouthwash samples by the anaerobic culture and real-time PCR techniques. Methods: Pooled subgingival plaque samples and 10-ml mouthwash samples were collected from 21 adult patients with periodontitis and analyzed by quantitative anaerobic culture and real-time PCR for A. actinomycetemcomitans, P. gingivalis, T. forsythensis, P. intermedia, and M. micros. Results: The detection frequency of A. actinomycetemcomitans, P. gingivalis, and T. forsythensis in subgingival plaque was identical by culture and real-time PCR and was higher for P. intermedia and M. micros by real-time PCR. The highest detection frequencies for the target bacteria were found in mouthwash samples by real-time PCR. The additional value of the real-time PCR to detect target bacteria was 38% for P. gingivalis, 73% for T. forsythensis, 77% for P. intermedia, and 71% for M. micros. The sensitivity to detect target species in mouthwash by real-time PCR was 100% for all test species except for P. intermedia (93.8%). Conclusions: Rapid detection and quantification of periodontal pathogens in mouthwash samples are possible by real-time PCR. The procedure is significantly less time-consuming than subgingival sampling with paper points. This approach to detect major periodontal pathogens in mouthwash samples may simplify microbial diagnosis in periodontitis patients and may be used to monitor periodontal treatment.
[Detection of 3 Anaerobic Microorganisms in Saliva and Subgingival Plaque of Patients with Periodontitis].

Abstract:OBJECTIVE To investigate the prevalence of 3 anaerobic microorganisms including Porphyromonas gingivalis (Pg), Tannerella forsythia (Tf) and Treponema denticola (Td) in saliva and pooled subgingival plaque of different types of periodontitis and compare the detections of three microorganisms between saliva and pooled subgingival plaque sample from the same patient, and analyze the relationship between prevalence of these pathogens in saliva and periodontal parameters. METHODS Non-stimulated saliva samples and pooled subgingival plaque samples were collected from 50 patients with aggressive periodontitis (AgP), 48 patients with chronic periodontitis (CP) and 25 subjects with no periodontitis, and three microorganisms including Pg, Tf and Td were detected in these samples by PCR method. RESULTS Prevalence of three microorganisms in pooled subgingival plaque and saliva samples of AgP patients and CP patients were significantly higher than subjects with no periodontitis (P<0.01). The prevalence of Pg in Subgingival plaque and saliva was as follows: AgP, 100% vs 100%; CP, 93.8% vs 93.8%; Subjects with no periodontitis, 32% vs 48%. The prevalence of Tf in Subgingival plaque and saliva was as follows: AgP, 96% vs 88%; CP, 97.9% vs 89.6%; Subjects with no periodontitis, 32% vs 24%. The prevalence of Td in Subgingival plaque and saliva was as follows: AgP, 94% vs 86%; CP, 89.6% vs 70.8%; Subjects with no periodontitis, 12% vs 16%. The agreement between saliva and pooled subgingival plaque samples of these three microorganisms from the same patient was high. The presence of three microorganisms in saliva was all associated with bleeding index of gingiva, and odds ratio of Pg was 13.5 (P<0.01). CONCLUSION Pg, Tf and Td were widely distributed in saliva and subgingival plaque of patients with AgP and CP. Presence of Pg, Tf and Td in saliva was related to periodontal parameters of gingiva, and saliva could be used as a promising sample for oral detection of these 3 microorganisms.

Abstract:Oral health is maintained by a healthy microbiome, which can be monitored by state-of-the art diagnostics. Therefore, this study evaluated the presence and quantity of ten oral disease-associated taxa (P. gingivalis, T. forsythia, T. denticola, F. nucleatum, C. rectus, P. intermedia, A. actinomycetemcomitans, S. mutans, S. sobrinus, oral associated Lactobacilli) in saliva and their clinical status association in 214 individuals. Upon clinical examination, study subjects were grouped into healthy, caries and periodontitis and their saliva was collected. A highly specific point-of-care compatible dual color qPCR assay was developed and used to study the above-mentioned bacteria of interest in the collected saliva. Assay performance was compared to a commercially available microbial reference test. Eight out of ten taxa that were investigated during this study were strong discriminators between the periodontitis and healthy groups: C. rectus, T. forsythia, P. gingivalis, S. mutans, F. nucleatum, T. denticola, P. intermedia and oral Lactobacilli (p < 0.05). Significant differentiation between the periodontitis and caries group microbiome was only shown for S. mutans (p < 0.05). A clear distinction between oral health and disease was enabled by the analysis of quantitative qPCR data of target taxa levels in saliva.

Effective microbial molecular diagnosis of periodontitis-related pathogen Porphyromonas gingivalis from salivary samples using rgpA gene

Abstract:Importance of accurate molecular diagnosis and quantification of particular disease-related pathogenic microorganisms is highlighted as an introductory step to prevent and care for diseases. In this study, we designed a primer/probe set for quantitative real-time polymerase chain reaction (qRT-PCR) targeting rgpA gene, known as the specific virulence factor of periodontitis-related pathogenic bacteria 'Porphyromonas gingivalis', and evaluated its diagnostic efficiency by detecting and quantifying relative bacterial load of P. gingivalis within saliva samples collected from clinical subjects. As a result of qRT-PCR, we confirmed that relative bacterial load of P. gingivalis was detected and quantified within all samples of positive control and periodontitis groups. On the contrary, negative results were confirmed in both negative control and healthy groups. Additionally, as a result of comparison with next-generation sequencing (NGS)-based 16S metagenome profiling data, we confirmed relative bacterial load of P. gingivalis, which was not identified on bacterial classification table created through 16S microbiome analysis, in qRT-PCR results. It showed that an approach to quantifying specific microorganisms by applying qRT-PCR method could solve microbial misclassification issues at species level of an NGS-based 16S microbiome study. In this respect, we suggest that P. gingivalis-specific primer/probe set introduced in present study has efficient applicability in various oral healthcare industries, including periodontitis-related microbial molecular diagnosis field.
USING HOMOGENIZED MACROSCOPIC GROUP CROSS SECTIONS IN CONTINUOUS-ENERGY MONTE CARLO NEUTRON TRANSPORT CALCULATIONS WITH MCNP

Abstract:A method has been developed to include diffusion-theory-type (macroscopic) group cross sections into MCNP, while still running in continuous (point-wise) energy calculation mode. A good agreement is observed between results (keff, reaction rates and flux profiles) from a heterogeneous reference MCNP calculation and those from MCNP calculations based on homogenised macroscopic group cross sections, obtained from the heterogeneous MCNP calculation and ELNINJO post-processing. Also a good agreement is observed between results from MCNP transport-theory calculations and PANTHER diffusion-theory calculations, based on exactly the same macroscopic group cross sections, although effects of the fundamental differences between transportand diffusion theory are apparent as well. The presented results provide further evidence for the validity of the MCNP-ELNINJO method for the generation of homogenised (macroscopic) group cross sections for complicated geometries. Furthermore, the method presented opens the possibility to speed up continuous-energy MCNP calculations, as less important regions of a large, complicated geometry can be replaced by homogenised areas with appropriate group cross sections, while the details of the important sections are retained.
Real-Time PCR Method as Diagnostic Tool for Detection of Periodontal Pathogens in Patients with Periodontitis

Abstract:The most common type of periodontal disease is chronic periodontitis, an inflammatory condition caused by pathogenic bacteria in subgingival plaque. The aim of our study was the development of a real-time PCR test as a diagnostic tool for the detection and differentiation of five periodontopathogenic bacteria, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythia, Prevotella intermedia, and Treponema denticola, in patients with periodontitis. We compared the results of our in-house method with the micro-IDent® semiquantitative commercially available test based on the PCR hybridization method. DNA was isolated from subgingival plaque samples taken from 50 patients and then analyzed by both methods. Comparing the results of the two methods, they show a specificity of 100% for all bacteria. The sensitivity for A. actinomycetemcomitans was 97.5%, for P. gingivalis 96.88%, and for P. intermedia 95.24%. The sensitivity for Tannerella forsythia and T. denticola was 100%. The Spearman correlation factor of two different measurements was 0.976 for A. actinomycetemcomitans, 0.967 for P. gingivalis, 0.949 for P. intermedia, 0.966 for Tannerella forsythia, and 0.917 for T. denticola. In conclusion, the in-house real-time PCR method developed in our laboratory can provide information about relative amount of five bacterial species present in subgingival plaque in patients with periodontitis. It is likely that such a test could be used in dental diagnostics in assessing the efficacy of any treatment to reduce the bacterial burden.

Purification and characterization of phospholamban from canine cardiac sarcoplasmic reticulum.

Abstract:Phospholamban, a putative regulator of the Ca2+-dependent ATPase of cardiac sarcoplasmic reticulum (SR), was purified from canine cardiac SR membranes. Cardiac SR was extracted with deoxycholate and fractionated with ammonium sulfate followed by gel permeation high performance liquid chromatography in the presence of the nonionic detergent, octa-ethylene glycol mono-n-dodecyl ether (C12E8), and KI. Further purification was achieved with CM-Sepharose CL 6B column chromatography in the presence of C12E8. The purified phospholamban showed a single band of 22,000 daltons on neutral sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (Weber, K., and Osborn, M. (1969) J. Biol. Chem. 244, 4406-4412) and 27,000 daltons on alkaline SDS gels (Laemmli, U. K. (1970) Nature (Lond.) 227, 680-685). Boiling of phospholamban in 2% SDS produced total conversion into the lower molecular weight component on SDS gels (11,000 on Laemmli gel and 10,500 on Weber and Osborn gel). The apparent molecular weight of phospholamban on SDS gels was slightly increased by cAMP-dependent phosphorylation. The extent of phosphorylation catalyzed by cAMP-dependent protein kinase in the purified phospholamban preparations was about 42 nmol of phosphate/mg of protein when the protein concentration was determined by the method of Lowry et al. (Lowry, O. H., Rosebrough, N. J., Farr, A. L., and Randall, R. J. (1951) J. Biol. Chem. 193, 265-275), or 138 nmol/mg of protein based on the protein concentration estimated by the dye absorption method. Rabbit antisera were prepared against purified phospholamban. The obtained antisera were found to bind to purified phospholamban as well as that in cardiac SR. No reaction was detected in fast skeletal muscle SR by immunofluorescent staining of Western blots. The present preparation of purified phospholamban and the antisera should facilitate further understanding of the regulatory action of phospholamban on the calcium pump ATPase.
Highly accurate measurement of the relative abundance of oral pathogenic bacteria using colony-forming unit-based qPCR

Abstract:Purpose: Quantitative polymerase chain reaction (qPCR) has recently been employed to measure the number of bacterial cells by quantifying their DNA fragments. However, this method can yield inaccurate bacterial cell counts because the number of DNA fragments varies among different bacterial species. To resolve this issue, we developed a novel optimized qPCR method to quantify bacterial colony-forming units (CFUs), thereby ensuring a highly accurate count of bacterial cells. Methods: To establish a new qPCR method for quantifying 6 oral bacteria namely, Porphyromonas gingivalis, Treponema denticola, Tannerella forsythia, Prevotella intermedia, Fusobacterium nucleatum, and Streptococcus mutans, the most appropriate primer-probe sets were selected based on sensitivity and specificity. To optimize the qPCR for predicting bacterial CFUs, standard curves were produced by plotting bacterial CFU against Ct values. To validate the accuracy of the predicted CFU values, a spiking study was conducted to calculate the recovery rates of the predicted CFUs to the true CFUs. To evaluate the reliability of the predicted CFU values, the consistency between the optimized qPCR method and shotgun metagenome sequencing (SMS) was assessed by comparing the relative abundance of the bacterial composition. Results: For each bacterium, the selected primer-probe set amplified serial-diluted standard templates indicative of bacterial CFUs. The resultant Ct values and the corresponding bacterial CFU values were used to construct a standard curve, the linearity of which was determined by a coefficient of determination (r²) >0.99. The accuracy of the predicted CFU values was validated by recovery rates ranging from 95.1% to 106.8%. The reliability of the predicted CFUs was reflected by the consistency between the optimized qPCR and SMS, as demonstrated by a Spearman rank correlation coefficient (ρ) value of 1 for all 6 bacteria. Conclusions: The CFU-based qPCR quantification method provides highly accurate and reliable quantitation of oral pathogenic bacteria.

Abstract:Objective To analyze the effect of non-surgical periodontal treatment on the prevalence of putative periodontal pathogens in saliva and gingival crevicular fluids (GCF) in patients with aggressive periodontitis(AgP).Methods Sixteen patients with AgP received non-surgical periodontal treatment,and un-stimulated saliva and GCF were collected before treatment and 2 weeks,2 months,4 months,6 months post-treatment.Periodontal pathogens including Aggregatibacter actinomycetemcomitans (Aa),Porphyromonas gingivalis (Pg),Tannerella forsythia (Tf),Treponema denticola (Td),Prevotella intermedia (Pi) and Fusobacterium nucleatum (Fn) were detected by polymerase chain reaction.Results The Aa,Pg,Tf,Td,Pi and Fn in saliva were detected in 7,16,13,14,9 and 14 patients respectively at baseline,and in 3,9,5,4,4 and 7 patients 2 weeks post-treatment.The prevalence of the 6 periodontal pathogens showed a tendency to increase at 2 months post-treatment,and was almost the same as that of baseline at 6 months post-treatment.While in GCF the prevalence of Aa,Pg,Tf,Td,Pi and Fn were 17％ (11/64),95％ (61/64),84％ (54/64),72％ (46/64),41％ (26/64) and 56％ (36/64) at baseline,and reduced to 8％ (5/64),75％ (48/64),28％ (18/64),14％ (9/64),16％ (10/64) and 23％ (15/64) at 2 weeks post-treatment (P ＜ 0.05).The prevalence of the 6 periodontal pathogens remained a relatively low level [13％ (8/64),73％ (47/64),42％ (27/64),23％ (15/64),8％ (5/64) and 38％ (24/64)respectively] at 6 months post-treatment in GCF(P ＜ 0.05) compared with baseline except Aa.There was difference of periodontal pathogens detection between saliva and GCF.Conclusions Non-surgical periodontal treatment can reduce pocket depth as well as periodontal pathogens in saliva and GCF.Periodontal ruaintenance every 2 months can keep the periodontal pathogens in GCF at a relatively low level.The periodontal pathogen detection in saliva can not completely replace the site detection.

Abstract:Porphyromonas gingivalis is a key pathogen in the polymicrobial biofilm that is associated with the oral disease chronic periodontitis. A number of studies have shown that in humans the level of P. gingivalis in the polymicrobial biofilm is positively correlated with disease progression. The aim of this study was to develop a P. gingivalis diagnostic that has high specificity and sensitivity for P. gingivalis using a range of laboratory and clinical isolates and then compare the efficacy of the diagnostic with RTPCR using samples from chronic periodontitis patients and age- and sex-matched healthy controls. Key parameters for the kit were to use saliva as the biological fluid as this is a most convenient medium for chair-side sampling and to give a positive reading for the reported threshold for detection of 5×10(5) P. gingivalis cells/mL that indicates disease progression. We initially screened a range of monoclonal antibodies for recognition of the P. gingivalis conserved virulence factor RgpA-Kgp complex and identified two mAbs that could be used in a capture and detection ELISA system. These mAbs were used to formulate and manufacture the GC P. gingivalis saliva diagnostic kit used in the study. To validate the saliva kit, saliva (P. gingivalis free) was spiked with known concentrations of viable P. gingivalis whole cells of W50, 381, A7A1-28, and ATCC 33277; P. gingivalis clinical isolates; P. gingivalis vesicles; and the secreted form of the RgpA-Kgp complex. Laboratory findings indicated that the kit was able to detect all laboratory and clinical isolate strains of P. gingivalis at 5×10(4)/mL to 5×10(5)/mL. It was also able to detect the RgpA-Kgp complex and vesicles at 5×10(4) and 5×10(5) cell equivalent doses, respectively. Saliva and plaque were then collected from 50 subjects with moderate-severe chronic periodontitis and 50 age- and sex-matched subjects with healthy periodontium. Real-time PCR was utilised to analyse levels of P. gingivalis in both saliva and plaque. The saliva kit was found to give a positive result within 90 seconds. Using point bi-serial correlation analysis, a significant (p=0.04) correlation was found for detection of P. gingivalis using the saliva kit and P. gingivalis levels in saliva and plaque as determined by real-time PCR. A sensitivity of 92% and a specificity of 96% were found when compared to real-time PCR at a 10(5) P. gingivalis cell threshold.In conclusion, the P. gingivalis saliva kit was shown to be rapid and has a comparable detection capacity to real-time PCR. Thus, the P. gingivalis saliva diagnostic has the potential to be a simple and time-efficient chair-side diagnostic for the detection of P. gingivalis.

Abstract:Objective To develop PCR assays for detecting Porphyromonas gingivalis (Pg) and for understanding genotype difference of Pg strains in subgingival plaque samples from chronic periodontitis patients. Methods Culture method was used to isolate Pg strains were isolated in subgingival plaque samples from different foci of chronic periodontitis. Pg 16S rDNA gene, collagenase gene (prtC) and fimbrillin gene (fimA) of Pg strains in these samples were detected by PCR. Partial amplification products were sequenced after T-A cloning. Results Among 122 subgingival plaque samples from 61 patients, the positive rates of Pg 16S rDNA, prtC and fimA gene detection were 90.6%, 81.9% and 78.0% respectively. In the same samples, the positive rate of co-presence of the three genes was as high as 98.4% while the positive rate was lower as 31.1% by routine culture. 18 of 60 patients (30.0%) were found to be infected with different genotypes of Pg strains in different tooth sites of an individual. The homology of PCR products from Pg 16S rDNA, prtC and fimA genes ranged from 98.62% to 100% compared with the reported nucleotide sequences. Conclusion The PCR assays developed in this study show high sensitivity and specificity, and are suitable for clinical fast diagnosis of Pg in subgingival plaque samples from chronic periodontitis patients. Some patients can be simultaneously infected by Pg different genotypes of Pg strains.

Abstract:Porphyromonas gingivalis (P. gingivalis) constitutes an essential part of the subgingival dental plaque biofilm, serving as a significant factor in the development of periodontitis. Therefore, establishing a rapid and highly sensitive detection method for P. gingivalis is crucial to effectively manage periodontitis and its associated complications. In this study, droplet digital PCR (ddPCR) technology was employed for the detection of P. gingivalis, with a detection limit of 10 1 CFU/mL, exhibiting a 10-fold higher sensitivity compared to qPCR (with a sensitivity of 10 2 CFU/mL). Furthermore, no cross-reactivity was observed with four other bacterial species. In comparison to real-time quantitative PCR, ddPCR demonstrated enhanced sensitivity in detecting P. gingivalis at lower concentrations in 16 simulated samples, indicating its applicability for rapid detection of P. gingivalis.

Abstract:To the Editor. —In the January 1984Archives, Sloand and Taylor Thompson1presented a case of unknown pheochromocytoma without histologic evidence of heart disease, in which propranolol hydrochloride—induced pulmonary edema and shock, leading to death. It is known that propranolol may cause pulmonary edema in patients with pheochromocytoma without having a previous α-blockade.2As propranolol is a nonspecific betablocking drug, its administration in these patients may leave α-adrenergic effects unopposed.1,2This could result in arterial vasoconstriction causing afterload increase and heart failure, that may lead to pulmonary edema2and cardiogenic shock.1Sloand and Taylor Thompson1suggested that the use of a selective β1-blocker such as metoprolol tartrate, would be a theoretical alternative to propranolol. Genotyping of Actinobacillus Actinomycetemcomitans Isolated from Subgingival Plaques of Patients with Chronic Periodontitis

Abstract:Objective To develop PCR assays for detecting A.actinomycetemcomitans in subgingival plaque samples and to find the infection rate and dominant genotypes of A.actinomycetemcomitans in patients with chronic periodontitis at different tooth sites. Methods 122 subgingival plaque samples were collected from two different tooth sites from each of 61 patients suffering from chronic periodontitis. Culture method was used to isolate A.actinomycetemcomitans strains and PCR or multiplex PCR were applied to detect 16S rDNA, leukotoxin (lktA) and fimbria associated protein (fap) genes of A.actinomycetemcomitans in these samples. Parts of the amplification products were sequenced after cloning. Results 11 strains of A.actinomycetemcomitans were isolated from 11 subgingival plaque samples of 11 patients. In 122 subgingival plaque samples, the positive rates of 16S rDNA, lktA and fap genes by PCR were 84.4%, 75.4% and 50.0% respectively. Different A.actinomycetemcomitans genotypes were detected in different two tooth sites of 19 cases of 49 patients (38.8%). 4 genotypes of A.actinomycetemcomitans were found in these samples and the dominant genotypes were first 16S rDNA +/lktA +/fap + and then 16S rDNA +/lktA +/fap -. The homologies of nucleotide sequence of amplification products from partial samples compared with the reported sequences were from 93.75% to 100%. Conclusion The PCR assays developed in this study with high sensitivity and specificity are suitable for rapid diagnosis of A.actinomycetemcomitans in subgingival plaque samples. A high frequency of A.actinomycetemcomitans infection in the adult periodontitis patients was found, and dominant genotypes of the bacteria in the samples were identified. Some of the patients have been diagnosed as infected with different genotypes of A.actinomycetemcomitans . [
Quantitative real-time PCR for target periodontal bacteria in subgingival plaque before and after local delivery of periocline, scaling and root planning]

Abstract:To compare the copy number of Porphyromonas gingivalis (Pg) and Prevotella intermedia (Pi) in subgingival plaque before and after local delivery of periocline (2% minocycline hydrochloride ointment, MO), scaling and root planning (SRP) by quantitative real-time PCR (qRT-PCR) and evaluate the efficacy.Sixty-two adults with moderate to severe chronic periodontitis were selected in the study. Microbial samples were taken from pocket before and after MO and SRP(7d). The samples were evaluated by qRT-PCR for Pg and Pi. Microbiological effectiveness of treatments was assessed using Kruskal-Wallis and Wilcoxon rank-sum test. All tests were two-sided with a significance level of 0.05. All analyses were conducted with SAS 9.1.3 software package.The copy number of Pg and Pi in subgingival plaque was 10(3)-10(6) and 10(2)-10(6). Bacterial loads of Pg were reduced in SPR+ MO, SRP and MO site. The counts of Pi decreased in SRP+ MO sites compared with those in the MO or SRP alone sites significantly (P<0.05).Quantitative real-time PCR (qRT-PCR) is used as a powerful tool with high sensitivity and specificity to quantitatively assess target periodontal bacteria. The results show that subgingival administration of MO and SRP was effective for reducing pathogenic bacteria and improving clinical outcome.
