Abstract:Objective: Elevated levels of the hormone aldosterone are associated with increased risk of myocardial infarction and stroke in humans and increased progression and inflammation of atherosclerotic plaques in animal models. Aldosterone acts through the mineralocorticoid receptor (MR) which is expressed in vascular smooth muscle cells (SMCs) where it promotes SMC calcification and chemokine secretion in vitro. The objective of this study is to explore the role of the MR specifically in SMCs in the progression of atherosclerosis and the associated vascular inflammation in vivo in the apolipoprotein E knockout (ApoE-/-) mouse model. Methods and Results: Male ApoE-/- mice were bred with mice in which MR could be deleted specifically from SMCs by tamoxifen injection. The resulting atheroprone SMC-MR-KO mice were compared to their MR-Intact littermates after high fat diet (HFD) feeding for 8 or 16 weeks or normal diet for 12 months. Body weight, tail cuff blood pressure, heart and spleen weight, and serum levels of glucose, cholesterol, and aldosterone were measured for all mice at the end of the treatment period. Serial histologic sections of the aortic root were stained with Oil Red O to assess plaque size, lipid content, and necrotic core area; with PicroSirius Red for quantification of collagen content; by immunofluorescent staining with anti-Mac2/Galectin-3 and anti-smooth muscle α-actin antibodies to assess inflammation and SMC marker expression; and with Von Kossa stain to detect plaque calcification. In the 16-week HFD study, these analyses were also performed in sections from the brachiocephalic artery. Flow cytometry of cell suspensions derived from the aortic arch was also performed to quantify vascular inflammation after 8 and 16 weeks of HFD. Deletion of the MR specifically from SMCs did not significantly change plaque size, lipid content, necrotic core, collagen content, inflammatory staining, actin staining, or calcification, nor were there differences in the extent of vascular inflammation between MR-Intact and SMC-MR-KO mice in the three experiments. Conclusion: SMC-MR does not directly contribute to the formation, progression, or inflammation of atherosclerotic plaques in the ApoE-/- mouse model of atherosclerosis. This indicates that the MR in non-SMCs mediates the pro-atherogenic effects of MR activation.

Abstract:Menopause is associated with dyslipidemia and an increased risk of cardiovascular disease, the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen. In this study, we find that estrogen mediates an atherosclerotic-protective action via estrogen receptor alpha/SREBP-1 signaling. Increased lipid accumulation and low-density lipoprotein (LDL)-uptake in HepG2 cells and THP-1 macrophages were induced by treatment of mixed hyperlipidemic serum from postmenopausal women; 17β-estradiol [estrogen (E2)] (10 nM) administration significantly improved hyperlipidemic profiles, relieved fatty-liver damage and attenuated the plaque area in the heart chamber of high-fat diet (HFD)-fed ovariectomized (OVX) Apo E –/ – mice. Expression of sterol regulatory element-binding protein (SREBP)-1 mRNA of circulating leukocytes in postmenopausal women was strongly correlated to the serum E2 level. Exploration of data from the Gene Expression Profiling Interactive Analysis (GEPIA) database revealed that expression of SREBP-1 protein correlated to expression of estrogen receptor (ESR)α protein in the liver, blood and in normal tissue. Genetic overexpression/inhibition of ESRα resulted in increased/decreased SREBP-1 expression as well as attenuated/deteriorated lipid deposition in vitro . An inhibitor of the protein kinase B/mammalian target of rapamycin (AKT/mTOR) pathway, AZD8055, abolished ESRα-induced SREBP-1 expression in HepG2 cells. Moreover, E2 and statin co-treatment significantly reduced lipid accumulation in vitro and hindered the progression of atherosclerosis and fatty-liver damage in OVX Apo E –/ – mice. Collectively, our results suggest that estrogen could exerted its atherosclerotic-protective action via ESRα/SREBP-1 signaling. E2 might enhance the cellular sensitivity of statins and could be used as a novel therapeutic strategy against atherosclerotic disorders in postmenopausal women.

Effect of Estrogen Replacement Therapy in the Early Phase on Atherosclerosis in Rats

Abstract:Objective:To understand the effect of estrogen replacement therapy(ERT) in the early phase on atherosclerosis in rats fed by high fat diet.Methods:28 rats were divided into 4 groups:group A(sham operation),group B(ovariectomized without estradiol),group C(ovariectomized with low-dose estradiol) and group D(ovariectomized with high-dose estradiol).All the rat in 4 groups were given 1% cholesterol diet for 12 weeks,the levels of blood lipids,estradiol and serum superoxide dismutase(SOD) activity were detected before the operation and on the fourth and twelfth weeks.Results:After 12 weeks,the level of estradiol in group B was significantly lower than that in group A(P0.05),the level of estradiol in group D was significantly higher than that in group A(P0.05),there was no difference in the level of estradiol between group C and group A(P0.05);the levels of TC and LDL-C in group B were significantly higher than those in group A(P0.05);the levels of TC and LDL-C in group C and group D were significantly lower than those in group A(P0.05),and levels of TC and LDL-C in group B were the lowest among four groups.The aortic plaque area in group B was the largest among four groups(P0.05),the aortic plaque areas in group C and group D were smaller than that in group A(P0.05).The serum SOD activity of group B decreased significantly.Conclusion:Estrogen replacement therapy in the early phase can improve the metabolism of the serum lipids,increase the SOD activity and inhibit the formation of atherosclerosis.

The effects of sustained delivery of estrogen on bone strength and cardiovascular panels in osteoporotic female rats

Abstract:Osteoporosis and cardiovascular disease (CVD) are common age-related conditions, which are major public health problems leading to an increase in mortality, morbidity, and disability. There have been several connections found between CVD and osteoporosis such as common genetic factors, risk factors, and pathological mechanisms. There is a direct effect of estrogen on CVD and osteoporosis that is demonstrated by the manifestation of estrogen receptors on osteoblasts, osteoclasts, and vascular endothelial and smooth muscle cells. Loss of estrogen has been found to be involved in the pathogenesis of atherosclerosis and bone loss through modulation of other factors including cytokines and oxidized lipids. The goal of this proposed research was to determine if sustained delivery of estrogen is capable of regulating bone cell function while improving cardiovascular panels. Ovariectomized Sprague Dawley rats were administered estradiol at a rate of 5ng/day over an eight-week period. Body weights, estradiol levels, cholesterol levels, and bone strength were determined at 2, 4, and 8 weeks following sustained delivery of estradiol and compared with intact control and ovariectomized control animals. Estrogen replacement resulted in improved cholesterol panels without significant changes in bone flexural strength or improvements in bone porosity. Additional long term studies are needed to determine if the benefits of estrogen replacement outweigh the inherent risks associated with hormone replacement therapies.
Estrogen prevent atherosclerosis by attenuating endothelial cell pyroptosis via activation of estrogen receptor α-mediated autophagy

Abstract:Excessive inflammation and the pyroptosis of vascular endothelial cells caused by estrogen deficiency is one cause of atherosclerosis in post-menopausal women. Because autophagy is highly regulated by estrogen, we hypothesized that estrogen can reduce vascular endothelial cell pyroptosis through estrogen receptor alpha (ERα)-mediated activation of autophagy to improve atherosclerosis in post-menopausal stage. Aortic samples from pro-menopausal and post-menopausal women with ascending aortic arteriosclerosis were analyzed, and bilateral ovariectomized (OVX) female ApoE-/- mice and homocysteine (Hcy)-treated HUVECs were used to analyze the effect of estrogen supplementation therapy. The aortic endothelium showed a decrease in ERα expression and autophagy, but presented an increase in inflammation and pyroptosis in female post-menopausal patients. Estrogen treatment accelerated autophagy and ameliorated cell pyroptosis in the cardiac aortas of OVX ApoE-/- mice and Hcy-treated HUVECs. Estrogen had therapeutic effect on atherosclerosis and improved the symptoms associated with lipid metabolism disorders in OVX ApoE-/- mice. Inhibition and silencing of ERα led to a reduction in the autophagy promoting ability of estrogen and aggravated pyroptosis. Moreover, the inhibition of autophagy promoted pyroptosis and abolished the protective effect of estrogen, but had no influence on ERα expression. Thus, the results of the present study demonstrated that post-menopausal women present decreased autophagy and ERα expression and excessive damage to the ascending aorta. Abstract:This study addresses the increasing prevalence of obesity, especially among postmenopausal. Estrogen plays a crucial role in regulating adipose tissue in women, with its absence after menopause associated with metabolic complications. The study aimed to determine the lipolytic activity in different adipose tissue depots of ovariectomized rats submitted to a high-fat diet. Also, to analyze the expression of estrogen receptors in adipose tissues and perform histological and morphometric analyzes of these deposits. Female rats were ovariectomized (O) or sham operated (S). The animals were divided into groups: ovariectomized with high-fat diet (OF), sham-operated with high-fat diet (SF), ovariectomized with control diet (OC) or sham-operated with control diet as the control group (SC). After 24 weeks of consuming the diets, rats were killed and adipose tissue deposits were removed. Polymerase chain reaction was performed to analyze the expression of estrogen receptors in adipose tissues, lipolysis assay and histological analysis. Both the high-fat diet and ovariectomy increased body weight and adiposity. There was hypertrophy of adipocytes. Estrogen replacement therapy modulate lipolytic activity in different adipose depots, with different responses in relation to estrogen receptors. Estrogen receptor expression varied between fat depots. Mesenteric adipose tissue showed greater sensitivity to estrogen compared with others. Estrogen increased lipolytic activity in some fat depots, reducing in others. Expression of ERs depends of hormonal status and adipose tissue location, which may explain distinct actions of estrogen on the metabolism of adipose tissue and on the production of adipokines by them.
Abstract 528: Endothelial Estrogen Receptor Alpha Deletion Promotes Abdominal Aortic Aneurysm In Male Mice

Abstract:Introduction: Although men have a greater incidence of abdominal aortic aneurysm (AAA) compared with women, the role of sex-hormones in mediating these differences is not well understood. Given the known effect of estrogen receptor alpha (ERα) signaling in maintaining vascular wall homeostasis, the present study examines its role in the pathogenesis of AAA. Specifically, we scrutinize whether endothelial cell (EC) ERα protects against AAA via increased lysyl oxidase (LOX) activation and regulation of vascular smooth muscle cell (VSMC) phenotype. Methods: AAA was induced in male and female ECERα knockout (ECERαKO) mice and their littermates (n=6-11) by administering β-Aminopropionitrile (BAPN) via drinking water (1 mg/ml; 0.15 g/kg/d) from 3 days prior to angiotensin II (Ang II; 1 μg/kg/day) infusion for 2 weeks. The development of AAA was assessed by ultrasound measurement of aortic luminal dilation and histology. Relevant molecular mechanisms were examined in abdominal aorta and in human aortic endothelial cells (HAECs). Results: BAPN Ang II resulted in an increased incidence of AAA in male ECERαKO mice (72%), whereas female ECERαKO mice did not develop aneurysms. No difference in AAA incidence was observed between male and female littermate control mice (12.5%). Elastin fragmentation was significantly higher only in male ECERαKO mice compared to littermates, while female ECERαKO and their littermates had comparable elastin fragmentation. Ang II receptor type 1a mRNA expression was higher in male ECERαKO, whereas female ECERαKO mice had higher Ang II receptor type 1b mRNA expression. Gene expression of smooth muscle alpha-actin and smooth muscle 22 (contractile VSMC markers) was significantly higher in female aortas compared to males irrespective of genotype, whereas, osteopontin, synthetic VSMC marker was only upregulated in male ECERαKO. HAECs treated with estradiol (10 nM, 24 h) significantly increased LOX activity. Conclusions: Our findings identify a critical role of endothelial ERα in protection against AAA in male mice likely through increased LOX activity and maintenance of VSMC phenotype. Further studies are needed to delineate the crosstalk between endothelial ERα and VSMCs, and their impact in the pathogenesis of AAA.

Abstract:AIM: Menopause causes arterial senescence and atherosclerotic development through decrease of estrogen. Recently, histone deacetylase SIRT1 has been reported to have protective effects against arterial senescence and atherosclerosis. However, the relationship between estrogen and SIRT1 in the context of menopause-induced arterial senescence is not well understood. The present study aims to investigate whether SIRT1 is involved in the etiology of menopause-induced arterial senescence and atherosclerotic development.METHODS: Twelve-week old female apolipoprotein E-knockout (ApoE-KO) mice underwent ovariectomy (OVX) or sham surgery.RESULTS: SIRT1 expression and endothelial nitric oxide synthase (eNOS) activation in the aorta were significantly lower in OVX mice than they were in sham mice (OVX vs. sham, n=5 per group). Senescence-associated β-galactosidase activity, protein expression of Ac-p53 and PAI-1, and aortic atherosclerosis lesions were significantly greater in OVX mice than they were in sham mice. Administration of 17β-estradiol (E2) for eight weeks to OVX mice restored aortic SIRT1 expression, activated eNOS, and retarded OVX-induced arterial senescence and atherosclerotic development (E2 vs. control, n=5 per group). The effects of E2 on SIRT1 upregulation, anti-senescence and anti-atherosclerosis were attenuated by administration of a SIRT1 inhibitor, sirtinol. In vitro experiment using human endothelial cells demonstrated that E2 also increased SIRT1 expression and retarded oxidized low density lipoprotein-induced premature senescence, which were also abolished by sirtinol. These results suggested that estrogen modulated arterial senescence and atherosclerosis through SIRT1. Additionally a selective estrogen receptor modulator (SERM), bazedoxifene, also augmented SIRT1 and inhibited arterial senescence and atherosclerotic development (SERM vs. control, n=3 per group).CONCLUSIONS: Downregulation of SIRT1 causes OVX-induced arterial senescence and atherosclerosis in ApoE-KO mice. Administration of estrogen or SERM enables OVX mice to restore these alterations by SIRT1 induction.

Atherosclerosis and Bone Loss in Humans-Results From Deceased Donors and From Patients Submitted to Carotid Endarterectomy

Abstract:Background and Aims: Atherosclerosis and osteoporosis share common risk factors, as well as inflammatory mechanisms. Our aim was to understand how atherosclerotic lesions are related with disturbances in bone. Methods: Gene expression of pro-inflammatory and bone metabolism related proteins (IL-1β, IL-6, IL-17A, TNF, RANKL, OPG, COL1, CTSK, OCL, TRAP, CBFA1, DKK1, SOST, ADIPOQ, and ADIPOR1) were analyzed in arteries and bones from 45 deceased donors and adipose tissue was used as control. Additionally, in 139 patients with advanced atherosclerosis submitted to carotid endarterectomy we compared calcium content (Alizarin red) and plaque inflammatory scores (CD3+, CD68+, and adiponectin) of patients with normal bone mineral density (BMD) with those with low BMD and explored the associations between gene expression in atherosclerotic plaques and BMD. Serum levels of pro-inflammatory and bone related proteins were measured both in donors and patients. Associations were investigated by the Pearson or Spearman correlation tests, and multivariate regression analyzes were performed when justified. Results: Gene expression of bone remodeling and pro-inflammatory proteins correlated positively in bone and aorta, independently of age and sex of donors, but not in adipose tissue. The expression of bone formation genes was significantly higher in atheroma plaques from endarterectomized patients with normal vs. low BMD as well as inflammatory CD68+ scores, regardless of patients' age and sex, but not of body mass index. No relationship was observed between serum levels and gene expression levels of pro-inflammatory or bone remodeling proteins. Conclusions: Our results suggest that the relationship between bones and vessels in the context of atherosclerotic disease and osteoporosis may rely on the intrinsic connection between the tissues involved, independently of disease stage. Serum measurements of pro-inflammatory and bone-remodeling proteins do not accurately translate tissue pathologic processes.

Abstract:The temperature-dependent DC characteristics and noise performance of an interesting InGaP/GaAs heterojunction bipolar transistor (HBT) with emitter-edge-thinning structure are demonstrated. Experimentally, due to the emitter-edge-thinning structure, higher current gains and wider collector current operation regime over the measured temperature range (300 ~ 400 K) are observed as compared to the conventional device. In addition, the studied device exhibits lower base current ideality factors, better thermal stabilities on DC current gains, lower base surface recombination current densities, and improved device reliability. Therefore, the studied device is suitable for low-power and high-temperature electronic applications.

Abstract:Objectives: Postmenopausal women with metabolic disease are predisposed to develop cardiovascular disease. Our previous experiments have shown that these women experience purinergic/adenosinergic signaling alteration resulting in diastolic dysfunction. We hypothesize that these signaling alterations are reversible and to demonstrate we used exogenous CD39 agonist (apyrase) and an ENT-1 inhibitor (dipyridamole), in a murine model simulating postmenopausal women. Methods: We fed 23 C57BL/6 female mice a HFD (40% kcal) from 5 weeks of age and performed ovariectomy at 12 weeks. Intraperitoneal (IP) injections were given daily to three groups for 10 days at 28 weeks. The following groups and dosages were used: apyrase (9 mice, 200U/kg), dipyridamole (9 mice, 20mg/kg), and saline (5 mice). At 30 weeks, mice were assessed with an EchoMRI for body composition, and transthoracic echocardiography (TTE) used for evaluation of cardiac function while mice were anesthetized with 2.5% isoflurane. Results: Our apyrase-treated mice, had significantly lowered values in E/E’, E/A, with increased Ejection Fraction and Fraction Shortening when compared to the saline treated groups, analyzed with t-test. Apyrase also had lower body weight, lean mass, fat mass, and heart/body weight percentage, when compared to saline (p<0.05). Dipyridamole had significant increase in LV diastolic volume and IVRT when compared to our saline group. Conclusion: Our findings could suggest that increased ectonucleotidase activity may ameliorate diastolic dysfunction experienced in our murine model. Future studies are needed to confirm its effect in postmenopausal women.

Abstract:Estrone (E1) constitutes the primary component in oral conjugated equine estrogens (CEEs) and serves as the principal estrogen precursor in the female circulation in the post-menopause. E1 induces endothelium-dependent vasodilation and activate PI3K/NO/cGMP signaling. To assess whether E1 mitigates vascular dysfunction associated with postmenopause and explore the underlying mechanisms, we examined the vascular effects of E1 in ovariectomized (OVX) rats, a postmenopausal experimental model. Blood pressure was measured using tail-cuff plethysmography, and aortic rings were isolated to assess responses to phenylephrine, acetylcholine (ACh), and sodium nitroprusside. Responses to ACh in rings pre-incubated with superoxide dismutase (SOD), catalase (CAT), or apocynin were also evaluated. Protein expression of SOD, CAT, NOX1, NOX2, and NOX4 was determined by Western blotting. E1 treatment resulted in decreased body weight and retroperitoneal fat, increased uterine weight, and prevented elevated blood pressure in the OVX group. Furthermore, E1 improved endothelium-dependent ACh vasodilation, activated compensatory antioxidant mechanisms – i.e. increased SOD and CAT antioxidant enzymes activity, and decreased NOX4 expression. This, in turn, helped prevent oxidative stress and endothelial dysfunction in OVX rats. Additionally, E1 treatment reversed the increased total LDL cholesterol observed in the OVX group. The findings underscore protective effects of E1 on the cardiovascular system, counteracting OVX-related oxidative stress and endothelial dysfunction in Wistar rats. E1 exhibits promising therapeutic benefits for managing cardiovascular health, particularly in postmenopausal conditions.

[The Expression of Estrogen Receptor Alpha and Beta in the Intervention of Different Estrogens in Rat Bone Metabolism].

Abstract:In this present study, female rats were ovariectomized (OVX) as the models of osteoporosis. The aim is to determine the different mechanisms of estrogen receptor(ER) alpha and beta pathway in mediating estrogen to participate in trabecular bone metabolism, and to further explore the distinction of modulation on ER alpha or ER beta between estrogens with different components. Mature female Sprague-Dawley rats (n=40) were randomly divided into four groups: group Control (sham operated), group OVX (only ovariectomized), group CEE (OVX rats treated with conjugated equine estrogens) and group EV (OVX rats treated with estradiol valerate). Sham operation and OVX were performed 48 days (12 estrums) before different liquid diet. The rats in group Control and group OVX were orally administrated with physiological saline solution and the rats in group CEE or group EV were orally administrated with CEE or EV for 12 days (3 estrums) before sacrifice. Relative quantitative reverse transcription- polymerase chain reaction (RT-PCR) and western blot techniques were utilized to compare the levels of ER alpha and ER beta mRNA and proteins in trabecular bone among groups. The results showed that in rat trabecular bone of group Control, the expression of ER alpha protein (1.433 +/- 0.250) was significantly higher than that of ER beta(0.687 +/- 0.120), whereas the ER alpha mRNA (0.285 +/- 0.033) was much lower than ERbeta mRNA(0.590 +/- 0.044). Following OVX, the levels of ER alpha protein (0.685 +/- 0.103) declined significantly, whereas mRNA levels (0.405 +/- 0.036) markedly increased. Both the protein (1.091 +/- 0.078) and mRNA (0.729 +/- 0.030) levels of ER beta significantly increased after OVX. After treatment with CEE, the expression of ER beta protein (0.583 +/- 0.129) and mRNA (0.618 +/- 0.043) were markedly down-regulated compared with group OVX. After treatment with EV, the ER alpha protein expression (1.272 +/- 0.247) was markedly up-regulated, while ERa mRNA (0.277 +/- 0.040) and ER beta protein (0.620 +/- 0.174) were markedly down-regulated compared with group OVX. On the other hand, the loss of bone mass in rat trabecular bone by OVX (0.192 +/- 0.010) was markedly gained after CEE (0.228 +/- 0.012) and EV (0.259 +/- 0.007) treatment. These results demonstrate that ERalpha may be the predominant subtype expressed in female rat trebecular bone and play a key role in the bone metabolism. This study also shows that estrogens with different components protect bone through variant ERs pathway.
Comparative effects of long-term continuous release of 16 alpha-hydroxyestrone and 17 beta-estradiol on bone, uterus, and serum cholesterol in ovariectomized adult rats

Abstract:16Alpha-hydroxyestrone (16alpha-OHE(1)), an endogenous estrogen metabolite, is associated with increased bone density in postmenopausal women. This study was designed to evaluate the long-term activity of this metabolite on bone, uterus, and serum cholesterol in an animal model for postmenopausal bone loss. A preliminary dose-response study performed in weanling rats determined 2000 microg/kg/day to be the optimal dose of 16alpha-OHE(1) for studying estrogenic effect on bone. The long-term experiment was performed in 6-month-old animals that were either sham-operated or OVX. The OVX rats were implanted sc with 60-day continuous-release carrier, 17beta-estradiol (E(2)) (33 microg/kg/day) or 16alpha-OHE(1) pellets (2000 microg/kg/day). OVX decreased uterine weight, increased body weight, serum cholesterol, and all dynamic bone histomorphometric measurements in cortical and cancellous bone, and resulted in a 54% bone loss at the tibial metaphysis. E(2) completely prevented OVX-induced bone loss, suppressed bone turnover, and induced uterine hypertrophy and hypercholesterolemia. 16alpha-OHE(1) acted as an E(2) agonist on bone, suppressing bone formation and resorption. However, the estrogen metabolite lowered serum cholesterol and was only a partial E(2) agonist on uterine weight and epithelial cell height. These results suggest that 16alpha-OHE(1) is an estrogen agonist on bone and may be responsible, in part, for the cholesterol-lowering activity attributed to estrogen. As a consequence of its skeletal effects, older women who produce high levels of 16alpha-OHE(1) could have a lower risk for developing postmenopausal osteoporosis than women who produce less-active estrogen metabolites.

Abstract:Purpose. To observe the effect of estrogen on endothelin (ET) and the expression of ET-1 mRNA in experimental post-menopause atherosclerosis rabbits. Methods. Post-merlopause atherosclerosis rabbit model was established by feeding high cholesterol diet after the castration operation and injected with bovine serum albumin. The escrogen had been given for 12 weeks. Results. Estrogen can decrease ET (P < 0.05), and the express of ET-1 (P < 0.05). Conclusions. Estrogen possesses the effect of decreasing the level of ET, and the express of ET-1 mRNA, and thus preserves the function of enothelial cell as well.
Estrogen inhibits vascular calcification in rats via hypoxia-induced factor-1α signaling

Abstract:Objective Calcification serves as a surrogate for atherosclerosis-associated vascular diseases, and coronary artery calcification is mediated by multiple pathogenic factors. Estrogen is a known factor that protects the arterial wall against atherosclerosis, but its role in the coronary artery calcification development remains largely unclear. This study tested the hypothesis that estrogen inhibits coronary artery calcification via the hypoxia-induced factor-1α pathway. Methods Eight-week-old healthy female Sprague–Dawley rats were castrated, and vitamin D 3 was administered orally to establish. Hypoxia-induced factor-1 inhibitor was administered to test its effect on vascular calcification and expression of bone morphogenetic protein 2 and runt-related transcription factor-2. Vascular smooth muscle cell calcification was induced with CaCl 2 in rat aortic smooth muscle cells in the presence or absence of E2(17β-estradiol) and bone morphogenetic protein 2 siRNA intervention. Results The estrogen levels in ovariectomized rats were significantly decreased, as determined by ELISA. Expression of hypoxia-induced factor-1α mRNA and protein was significantly increased in vascular cells with calcification as compared to those without calcification ( p < 0.01). E2 treatment decreased the calcium concentration in vascular cell calcification and cell calcium nodules in vitro ( p < 0.05). E2 also lowered the levels of hypoxia-induced factor-1α mRNA and protein ( p < 0.01). Oral administration of the hypoxia-induced factor-1α inhibitor dimethyloxetane in castrated rats alleviated vascular calcification and expression of osteogenesis-related transcription factors, bone morphogenetic protein 2 and RUNX 2 ( p < 0.01). Finally, bone morphogenetic protein 2 siRNA treatment decreased the levels of p-Smad1/5/8 in A7r5 calcification cells ( p < 0.01). Conclusion Estrogen deficiency enhances vascular calcification. Treatment with estrogen reduces the expression of hypoxia-induced factor-1α as well as vascular calcification in rats. The estrogen effects occur in a fashion dependent on hypoxia-induced factor-1α regulation of bone morphogenetic protein-2 and downstream Smad1/5/8.

Abstract:Estradiol is an important regulator of bone accumulation and maintenance. Circulating estrogens are primarily produced by the gonads. Aromatase, the enzyme responsible for the conversion of androgens to estrogen, is expressed by bone marrow cells (BMCs) of both hematopoietic and nonhematopoietic origin. While the significance of gonad-derived estradiol to bone health has been investigated, there is limited understanding regarding the relative contribution of BMC derived estrogens to bone metabolism. To elucidate the role of BMC derived estrogens in male bone, irradiated wild-type C57BL/6J mice received bone marrow cells transplanted from either WT (WT(WT)) or aromatase-deficient (WT(ArKO)) mice. MicroCT was acquired on lumbar vertebra to assess bone quantity and quality. WT(ArKO) animals had greater trabecular bone volume (BV/TV p = 0.002), with a higher trabecular number (p = 0.008), connectivity density ( p = 0.017), and bone mineral content ( p = 0.004). In cortical bone, WT(ArKO) animals exhibited smaller cortical pores and lower cortical porosity (p = 0.02). Static histomorphometry revealed fewer osteoclasts per bone surface (Oc.S/BS%), osteoclasts on the erosion surface (ES(Oc+)/BS, p = 0.04) and low number of osteoclasts per bone perimeter (N.Oc/B.Pm, p = 0.01) in WT(ArKO). Osteoblast-associated parameters in WT(ArKO) were lower but not statistically different from WT(WT). Dynamic histomorphometry suggested similar bone formation indices' patterns with lower mean values in mineral apposition rate, label separation, and BFR/BS in WT(ArKO) animals. Ex vivo bone cell differentiation assays demonstrated relative decreased osteoblast differentiation and ability to form mineralized nodules. This study demonstrates a role of local 17β-estradiol production by BMCs for regulating the quantity and quality of bone in male mice. Underlying in vivo cellular and molecular mechanisms require further study.

Abstract:Both estrogen and hydrogen sulfide (H2S) are involved in the regulation of atherosclerosis development. We previously reported that male mice with cystathionine gamma-lyase (CSE) gene knockout (CSE-KO) exhibited reduced vascular H2S production and increased adhesion molecule expression, which led to early development of atherosclerosis. Estrogen offers significant protection against proliferative cardiovascular diseases in females. The objective of this study is to investigate the impact of estrogen on the anti-atherogenic effect of H2S. Plasma estrogen (17β-estradiol) levels were significantly decreased in female CSE-KO mice in comparison with female wild-type (WT) mice. Age-matched female CSE-KO and WT mice were fed with control or atherogenic diet for 12-weeks. Plasma lipid levels, except triglyceride levels, were significantly increased in atherogenic diet-fed WT and CSE-KO mice when compared with control diet-fed groups. However, the extent of lipid change was greater in CSE-KO mice than in WT mice. Atherogenic diet fed female CSE-KO mice, but not WT mice, developed early atherosclerotic lesion. Atherogenic diet induced more atherosclerotic lesion, oxidative stress and intra-cellular adhesion molecule-1 (ICAM-1) expression and nuclear factor kappa B (NFκB) pathway in female CSE-KO mice when compared with female WT mice. To explore the role of estrogen in early development of atherosclerosis, female WT and CSE-KO mice fed with atherogenic diet were administered subcutaneously with 17β-estradiol at 80 μg/kg/day for 12 weeks. This treatment suppressed atherogenic factors such as hypercholesterolemia, oxidative stress, ICAM-1 expression and NFκB pathway in female WT mice, but not in female CSE-KO mice. Deficiency of CSE/H2S pathway lowered plasma estrogen levels in female mice and nullified the effects of estrogen on hypercholesterolemia, oxidative stress, ICAM-1 expression and atherosclerosis development. Thus, H2S is irreplaceable for these vascular effects of estrogen. The impact of this H2S dependency of estrogen on atherosclerosis development and the underlying mechanisms remain to be clarified.
