Abstract:Phase behavior and micellization of dextran-graft-poly (N-isopropylacrylamide) (PNIPAAm) polymers in aqueous solution are investigated in this paper using DSC and AFM methods. It is found that with the increase of grafting (G%) of the copolymers the endothermic enthalpy during the phase transition increases significantly and the transition temperature decreases slightly. The phase transition behavior of the copolymers is scanning rate dependent. Micelles are formed whenever the solution temperature is raised above the LCST of the copolymers. It is proposed that by using this thermal responsive property of the copolymers, drugs could be incorporated into the micelles without employing any organic solvent.
Tetraphenylethene Cross-Linked Thermosensitive Microgels Via Acylhydrazone Bonds: Aggregation-Induced Emission in Nanoconfined Environments and the Cononsolvency Effect

Abstract:We studied the aggregation-induced emission (AIE) phenomenon in a nanoconfined environment, where the AIE-active molecule, namely, 1,1,2,2-tetrakis(4-methanoylphenyl)ethane (TPE-4ALD), was held in space via four acylhydrazone bonds within the thermosensitive microgel networks. The thermosensitive microgels, namely N-AH-TPE, were synthesized via the copolymerization of N-isopropylacrylamide (NIPAM) and 4-acylhydrazine-(2-hydroxy-3-(methacryloxypropyl)pyridine hydrochloride (AH monomer) with TPE-4ALD as cross-linker via surfactant free emulsion polymerization (SFEP) in aqueous solution at 70 degrees C. Acylhydrazone-bonded tetraphenylethene (TPE-4AH) moieties were thus constructed and worked as the fluorophore in N-AH-TPE microgels. The aqueous suspensions of N-AH-TPE microgels exhibit strongly bluish-green fluorescence under ultraviolet excitation because the four arms of TPE-4AH moieties were held and their intramolecular motions are strongly restricted. It is estimated that there is one TPE-4AH moiety per about 394 nm3 for the swollen N-AH-TPE microgels. The fluorescent properties of N-AH-TPE microgels can be modulated via the change of hydrophilic and hydrophobic environments of TPE-4AH moieties exerted by external stimuli, like addition of various good solvents for TPE-based structures, i.e., N,N-dimethylformamide (DMF), methanol, ethanol, tetrahydrofuran (THF), and N,N-dimethyl sulfoxide (DMSO), varying the solution temperature as well as the counteranions of the microgels. An unusual enhancement in the fluorescent intensity is observed when specific amounts of organic solvent are added into the aqueous suspensions of N-AH-TPE microgels, which can be attributed to the cononsolvency of the polyNIPAM network chains. The shrinkage of N-AH-TPE microgels caused by the cononsolvency effect further strengthens the confinement of TPE-4AH moieties and hence enhances the fluorescent emission of the microgels even though the organic solvents added are good solvents for TPE-4AH. Increasing the solution temperature of N-AH-TPE microgels or introducing hydrophobic counteranions into the microgels also significantly enhances the fluorescent emission of the microgels.
Tuning the Morphology, Network Structure, and Degradation of Thermo-Sensitive Microgels by Controlled Addition of Degradable Cross-Linker

Abstract:Thermo-sensitive degradable poly(N-isopropylacrylamide) (PNIPAm)-based microgels were prepared by surfactant-free emulsion polymerization with a redox initiator pair of potassium persulfate (KPS) and N,N,N′,N′-tetramethylethylenediamine at 50 °C. NIPAm, sodium 2-acrylamido-2-methyl-1-propanesulfonate (AMPS-Na), and N,N′-bis(acryloyl)cystamine (BAC) were used as main monomer, anionic comonomer, and degradable cross-linker, respectively. It was found that the morphology and network structure of the resultant microgels could be tuned via the controlled addition of BAC and AMPS-Na, which, in turn, strongly affected the corresponding thermo-sensitivity, stability, and degradation behavior of the microgels. The inhomogeneous network structures of the microgels could be improved by increasing the time period t BAC between KPS initiation and the addition of BAC. The morphology of microgels changed from spherical into hollow interior spherical morphology. The stability of the microgels in physiological condition could be enhanced by the controlled addition of comonomer AMPS-Na. The extent of microgel degradation increased with increasing t BAC.

Abstract:To explore the potential biomedical application of thermoresponsive nanosystems, it is important to enhance their thermosensitivity to improve the controllability in delivery of therapeutic agents. The present work develops multifunctional nanogels with enhanced thermosensitivity through copolymerization of N-isopropylacrylamide (NIPAM) and acrylic acid (AA) in the presence of β-cyclodextrin (β-CD), using N,N'-bis(acryloyl)cystamine (BAC) as a biodegradable crosslinker. The resulting nanogels display significantly improved sensitivity in deswelling (swelling) behavior upon temperature increase (decrease) around body temperature. The nanogels can effectively encapsulate doxorubicin (DOX), which can be released in an accelerated way under microenvironments that mimic intracellular reductive conditions and acidic tumor tissues. Release can also be remotely manipulated by increasing temperature. In vitro study indicates that the nanogels are quickly taken up by KB cells (a human epithelial carcinoma cell line), exerting improved anticancer cytotoxicity, showing their potential for delivery of therapeutic agents beyond anticancer drugs.

Thermo-sensitive ionic microgels via post quaternization cross-linking: fabrication, property, and potential application

Abstract:A post quaternized cross-linking strategy was reported to fabricate thermo-sensitive ionic microgels from thermo-sensitive linear copolymer poly( N -isopropylacrylamide- co -1-vinylimidazole) [P(NIPAm- co -VIM)] or poly( N -isopropylacrylamide- co -4-vinylpyridine) [P(NIPAm- co -4VP)] in aqueous solution at elevated temperature above its critical transition temperature ( T c ). 1,4-dibromobutane, 1,5-dibromopentane, or 1,6-dibromohexane were chosen as the quaternization cross-linker to quaternize the imidazole or pyridine moieties, leading to the formation of cross-linking and ionic network. Transmission electron microscopy (TEM), dynamic and static light scattering (DLS & SLS), electrophoretic light scattering (ELS), and UV-visible spectroscopy were used to systematically investigate the sizes, morphologies, and properties of the obtained microgels. The sizes, swelling properties, and uniformity of microgels could be tuned by selecting proper quaternization cross-linking reaction temperature and cross-linkers. The obtained ionic microgels showed capability to adsorb the heavy metal salts like K 2 Cr 2 O 7 in aqueous solution via the anion exchange reactions. It was also demonstrated that other functional groups could be introduced into the microgel networks if the unquaternized moieties were available.

Abstract:A thermoresponsive copolymer, trimethyl chitosan-g-poly(N-isopropylacrylamide) (TMC-g-PNIPAAm), was synthesized by coupling PNIPAAm-COOH to TMC. Their molecular structures were characterized by 1HNMR. The lower critical solution temperature (LCST) of TMC-g-PNIPAAm in PBS was measured as 32°C by dynamic light scattering (DLS) and UV–vis spectroscopy, regardless of the grafting ratios. Upon mixing with DNA, TMC/DNA particles were formed, whose size and morphology were investigated by DLS and transmission electron microscopy, respectively. The particle size ranged from 200 to 900nm depending on the N/P ratio and was less influenced by the temperature variation. The majority of the particles have spherical morphology. The zeta potentials of these particles were increased along with the N/P ratio. At a given N/P ratio, the zeta potentials were almost constant at 25°C regardless of the existence of serum proteins. However, the values were significantly decreased at 37°C in a solution containing serum protein. The affinity between DNA and TMC was examined by ethidium bromide competitive binding assay. TMC-g-PNIPAAm has stronger ability to combine with DNA at 40°C when the PNIPAAm chain is collapsed. Gel electrophoresis results reveal that the vectors/DNA complexes can be formed regardless of the incubation temperature. HEK293 cell line was chosen as a model to study cellular uptake of the TMC-g-PNIPAAm/DNA particles, gene transfection and cytotoxicity. The grafting of PNIPAAm will not affect cellular uptake of the particles at 37°C. The level of gene transfection could be thermally controlled. By using a temperature variation protocol, i.e. incubation of the cultured cells at 25°C for a while, the gene transfection efficiency was significantly improved. Finally, the optimized gene transfection efficiency achieved by TMC-g-PNIPAAm is comparable to Lipofectamine 2000. No obvious cytotoxicity was detected for the TMC-g-PNIPAAm/DNA particles. These results suggest that TMC-g-PNIPAAm is an effective thermoresponsive gene carrier with minimal cytotoxicity, which has great promise for practical applications.

Abstract:A type of thermosensitive ionic microgel was successfully prepared via the simultaneous quaternized cross-linking reaction during the surfactant-free emulsion copolymerization of N-isopropylacrylamide (NIPAm) as the main monomer and 1-vinylimidazole or 4-vinylpyridine as the comonomer. 1,4-Dibromobutane and 1,6-dibromohexane were used as the halogenated compounds to quaternize the tertiary amine in the comonomer, leading to the formation of a cross-linking network and thermosensitive ionic microgels. The sizes, morphologies, and properties of the obtained ionic microgels were systematically investigated by using transmission electron microscopy (TEM), dynamic and static light scattering (DLS and SLS), electrophoretic light scattering (ELS), thermogravimetric analyses (TGA), and UV-visible spectroscopy. The obtained ionic microgels were spherical in shape with narrow size distribution. These ionic microgels exhibited thermosensitive behavior and a unique feature of poly(ionic liquid) in aqueous solutions, of which the counteranions of the microgels could be changed by anion exchange reaction with BF4K or lithium trifluoromethyl sulfonate (PFM-Li). After the anion exchange reaction, the ionic microgels were stable in aqueous solution and could be well dispersed in the solvents with different polarities, depending on the type of counteranion. The sizes and thermosensitive behavior of the ionic microgels could be well tuned by controlling the quaternization extent, the type of comonomer, halogenated compounds, and counteranions. The ionic microgels showed superior swelling properties in aqueous solution. Furthermore, these ionic microgels also showed capabilities to encapsulate and release the anionic dyes, like methyl orange, in aqueous solutions.

Polymer structure was confirmed by FT-IR, (1)H NMR, elemental analysis, and GPC. The thermosensitivity of PNIPAm-g-PPP aqueous solution was investigated by turbidity method. The lower critical solution temperature (LCST) of PNIPAm-g-PPP was observed to be approximately 30 degrees C in water, while it was 24 degrees C in 0.1M PBS (pH 7.4). Micellization behavior of PNIPAm-g-PPP in aqueous solution was characterized by fluorescence probe technique, TEM, and DLS. The critical micelle concentration (CMC), thus, determined was 0.0187 g/L. Both TEM and DLS measurement suggested that the diameter of micelles was approximately 190 nm at 20 degrees C. Diflunisal (DIF)-loaded micelles were prepared by dialysis method. In vitro release test at various temperatures was also performed to study the effect of temperature on the drug release profiles. It was demonstrated that DIF release from PNIPAm-g-PPP micelles was slower at the temperature of 37 degrees C than that at 4 degrees C.

Abstract:In this paper, novel thermosensitive poly(N-isopropylacrylamide) (PNIPAM) nanocapsules with temperature-tunable diameter and permeability are reported. Firstly, the core-shell composite microparticles were synthesized by precipitation polymerization with isothiocyanate fluorescein (FITC) entrapped SiO2 as core and cross-linked PNIPAM as shell. Then, the SiO2 core was etched by hydrofluoric acid at certain condition and the pre-trapped FITC molecules remained within the inner cavity. The FITC release profile and TEM studies clearly indicate that the release behavior of FITC could be controlled effectively by the external temperature. Above the LCST of PNIPAM (32°C), the dehydrated PNIPAM shell inhibited the release of FITC from the internal cavity while below its LCST, the fluorophore could permeate the swollen shell easily.

Abstract:Targeted drug delivery in response to external stimuli is therapeutically desirable, but long-term drug retention at the target site after stimulation is turned off remains a challenge. Herein, we present a targeted-delivery strategy via irreversible aggregation of drug carriers in response to mild external heating. We constructed two types of polymeric micelles, DBCO-TRM and Az-TRM, having a thermo-responsive polymer shell based on N -isopropylacrylamide (NIPAAm) and incorporating alkyne and azide moieties, respectively. Upon heating at 42 °C, the micelles aggregated through hydrophobic interaction between their dehydrated shells. Further, the azide moieties of Az-TRM become exposed on the surface due to the thermally shrinkage of the shells, thereby enabling crosslinking between the two types of micelles via azide-alkyne click chemistry to form irreversible aggregates. These aggregates were efficiently accumulated at tumor sites in mice by local heating after intravenous administration of a mixture of the micelles, and were well retained after cessation of heating due to their increased size. As proof of concept, we show that delivery of doxorubicin in this heat-guided drug delivery system dramatically improved the anti-tumor effect in a mouse model after a single treatment. Our results suggest that this platform could be an efficient tool for on-demand drug delivery.

Abstract:Thermoresponsive phospholipid-poly(N-isopropylacrylamide) (PL-PNIPAM) conjugates were synthesized via reversible addition fragmentation chain transfer polymerization mediated by a phospholipid-modified trithiocarbonate. Temperature triggered the micellization of the PL-PNIPAM conjugate to form phosphate group-decorated micelles in the aqueous solution. Driven by the chelation of phospholipids and Ca2+, the PL-PNIPAM conjugate and Ca2+ ions formed size-tunable nanoclusters at a temperature beyond the lower critical solution temperature. To fabricate cross-linked nanogels, NIPAM was copolymerized with N-succinimidyl acrylate (NSA) to obtain the PL-P(NIPAM-co-NSA) conjugate bearing pendent cross-linkable functionalities. Subsequently, the size-controllable nanogels containing disulfide linkages were generated at 37 °C by cross-linking the PL-P(NIPAM-co-NSA)/Ca2+ nanoclusters with cystamine through modulation of Ca2+ concentrations. These negatively charged nanogels demonstrate temperature/pH/reduction triple responsiveness. The nanogels can be efficiently loaded with doxorubicin (DOX) and proteins with various isoelectric points. The DOX-loaded nanogels exhibited a temperature/pH/reduction triple-responsive release profile. The immobilized RNase A, BSA, and GOx retained the protein bioactivity. The release of RNase A-loaded nanogels possesses a temperature-responsive profile. The immobilization of Lys and cytochrome C in nanogels inhibited protein bioactivity. However, the addition of NaCl triggered the recovery of bioactivity. These multistimuli-responsive nanogels can provide a versatile platform applicable in biotechnology and drug/protein delivery.

Abstract:Three diblock copolymers (PTPEE-PNns) with controllable polymer structures were synthesized via reversible addition-fragmentation chain transfer (RAFT) polymerization, combining 1-ethenyl-4-(1,2,2-triphenylethenyl)-benzene (TPEE) and N-isopropylacrylamide (NIPAM). All PTPEE-PNns copolymers showed remarkable aggregation-induced emission (AIE) characteristic. Due to the amphiphilic structure of the copolymers, PTPEE-PNn micelles (mainly hollow) were prepared with either "precipitation" or "dialysis" method, respectively. Scanning electron microscopy, transmission electron microscopy, dynamic light scattering and fluorescence methodology were performed to investigate the morphology, size distribution and stability of the micelles, revealing the effects of molecular weight and micelle preparation method on the nanostructure of the micelles. The PTPEE-PN2 hollow micelles prepared by the "dialysis" method had the advantages of small particle size, extremely narrow particle size distribution, remarkable thermal sensitivity, excellent particle stability, and showed good biocompatibility and fluorescence emission capability in cell imaging. This work would inspire more approaches to prepare novel AIE-active, thermoresponsive and biocompatible polymers for biomedical application.

Abstract:The improvement of the capability of poly(N-isopropylacrylamide) (PNIPAAm) hydrogel coating in cell adhesion and detachment is critical to efficiently prepare cell sheets applied in cellular therapies and tissue engineering. To enhance cell response on the surface, the amine group-modified PNIPAAm (PNIPAAm-APTES) nanohydrogels were synthesized and deposited spontaneously on tannic acid (TA)-modified polyethylene (PE) plates. Subsequently, TA was introduced onto PNIPAAm-APTES nanohydrogels to fabricate coatings composed of TA-modified PNIPAAm-APTES (PNIPAAm-APTES-TA). Characterization techniques, including TEM, SEM, XPS, and UV-Vis spectroscopy, confirmed the effective deposition of hydrogels of PNIPAAm as well as the morphologies, content of chemical bonding-TA, and stability of various coatings. Importantly, the porous hydrogel coatings exhibited superhydrophilicity at 20 °C and thermo-responsive behavior. The fluorescence measurement demonstrated that the coating's stability effectively regulated protein behavior, influencing cell response. Notably, cell response tests revealed that even without precise control over the chain length/thickness of PNIPAAm during synthesis, the coatings enhanced cell adhesion and detachment, facilitating efficient cell culture. This work represented a novel and facile approach to preparing bioactive PNIPAAm for cell culture.

Abstract:Poly(N-isopropylacrylamide-co-acrylic acid) hydrogel submicrometer particles were prepared by free radical copolymerization of N-isopropylacrylamide and acrylic acid in the presence of a crosslinker above the lower critical solution temperature (LCST). They exhibited a reversible swelling and deswelling behavior: ca. 200-nm diameter below the LCST and ca. 100-nm diameter above the LCST. The hydrogel particles were tagged with fluorescent dye (FITC) in order to monitor the extent of cellular uptake and were further modified with galactose moieties to evaluate the extent of receptor-mediated endocytosis against HepG2 cells. Flow cytometry and confocal microscopy were used to investigate cellular uptake behaviors of the submicrometer particles. It was found that the extent of cellular uptake of submicrometer particles was far greater above the LCST than below the LCST, suggesting that smaller particles were taken up more readily within cells. When the submicrometer particles were galactosylated, the extent of cellular uptake increased dramatically due to receptor-mediated endocytosis. This study proposes a new possibility of controlling intracellular events such as protein and gene expression by a thermally modulated endocytosis process using thermo-sensitive microgel beads.

Abstract:Hydrogels can offer many opportunities for drug delivery strategies. They can be used on their own, or their benefits can be further exploited in combination with other nanocarriers. Intelligent hydrogels that react to changes in the surrounding environment can be utilized as gatekeepers and provide sustained on-demand drug release. In this study, a hybrid nanosystem for temperature- and pH-sensitive delivery was prepared from MCM-41 nanoparticles grafted with a newly synthesized thermosensitive hydrogel (MCM-41/AA-g-PnVCL). The initial particles were chemically modified by the attachment of carboxyl groups. Later, they were grafted with agar (AA) and vinylcaprolactam (VCL) by free radical polymerization. Doxorubicin was applied as a model hydrophilic chemotherapeutic drug. The successful formulation was confirmed by FT-IR and TGA. Transmission electron microscopy and dynamic light scattering analysis showed small particles with negative zeta potential. Their release behaviour was investigated in vitro in media with different pH and at different temperatures. Under tumour simulating conditions (40 °C and pH 4.0), doxorubicin was almost completely released within 72 h. The biocompatibility of the proposed nanoparticles was demonstrated by in vitro haemolysis assay. These results suggest the possible parenteral application of the newly prepared hydrogel-functionalized mesoporous silica nanoparticles for temperature-sensitive and pH-triggered drug delivery at the tumour site.

Abstract:Constructing noncovalent interactions has been a benign method to tune the stimuli responsivity and assembled structure of polymers in solution; this is essential for controlling the functions and properties of stimuli-responsive materials. Herein, we demonstrate a novel supramolecular strategy to manipulate the cloud point (T-cp) and assembled structure of thermoresponsive polymers in solution by using H-bonding interactions. We use poly(lactide-co-glycolide)-b-poly(ethylene glycol)-b- poly(lactide-co-glycolide) (PLGA-PEG-PLGA) as a model thermoresponsive polymer and functionalize its chain terminals by the self-complementary quadruple H-bonding motif, 2-ureido-4[1H]-pyrimidinone (UPy). UPy end functionalization and increasing PLGA block length decrease the T-cp of copolymer. Both UPy- and nonfunctionalized copolymers form the spherical micelles at low temperature. They undergo the intermicellar aggregation and form large compound micelles during heating; this thermally induced structural transition causes the presence of T-cp. Due to the UPy-UPy H-bonding interactions, UPy end functionalization leads to more copolymer chains to associate in one micelle, thus, enhancing the hydrodynamic, gyration radii, core size, as well as the packing density of PLGA in micelle core and grafting density of PEG on core-shell interface. The decreased T-cp of UPy-functionalized copolymer stemmed from the stronger intermicellar attractions at high temperature. Furthermore, UPyfunctionalized copolymers exhibit higher drug loading content, slower drug release rate, and better separation efficiency in removing the hydrophobic substances from water than PLGA-PEG-PLGA precursors.
