Abstract:We herein describe a nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides. Both aryl/alkenyl iodines and bromides were found to be competent electrophiles that reacted with alkyl isocyanides, affording nitrile compounds in moderate to good yields. A range of functional groups including halogens as well as hydroxyl, formyl, and acetamino groups were fairly compatible with the nickel catalysis. This protocol featured broad functional group tolerance, simple reaction conditions, and gram-scale synthesis.

Abstract:A highly efficient and simple route for the synthesis of 2-substituted benzo[d]thiazoles has been developed by nickel catalyzed the cross-coupling reaction of 2-methylthiobenzo[d]thiazoles with aryl and alkenylaluminum reagents. Various 2-(hetero)aryl and 2-alkenyl substituted benzo[d]thiazoles derivatives can be obtained with 31-94% isolated yields using 4 mol% NiCl2(dppf)/4 mol% 2,2'-bipyridine as the catalyst under mild reaction conditions. The coupling reaction can be carried out smoothly whether the electron donor group or electron withdrawing group on the aromatic ring of organic aluminum reagents or 2-methylthiobenzo[d]thiazoles derivatives. Furthermore, the broad substrates scope and the typical maintenance of vigorous efficiency on gram scale make this protocol a potentially practical method to synthesize 2-substituted benzo[d]thiazoles derivatives. On the basis of the experimental results, a possible catalytic cycle has been proposed.

Abstract:Nickel-catalyzed cross-coupling of unactivated tertiary alkyl electrophiles with alkylmetal reagents is still a challenge. We report herein a nickel-catalyzed Negishi cross-coupling of alkyl halides, including unactivated tertiary halides, with boron-stabilized organozinc reagent BpinCH2ZnI, yielding versatile organoboron products with high functional-group tolerance. Importantly, the Bpin group was found to be indispensable for accessing the quaternary carbon center. The synthetic practicability of the prepared quaternary organoboronates was demonstrated by their conversion to other useful compounds.

Abstract:Abstract In the Kumada reaction, commercially accessible nickel catalysts facilitate the synthesis of 2-arylbenzothiazoles from various aryllithium compounds and 2-chlorobenzothiazole. This catalytic process is effective irrespective of the presence of electron-withdrawing or electron-donating groups on the benzene ring of aryllithium. Among these zero-valent, divalent nickel, and divalent nickel complexes, divalent nickel complexes have the best catalytic effect. NiCl2(PCy3)2 (Cy = cyclohexyl) has the best catalytic performance. The highest yield of 2-C6H4benzopyrazole from C6H4Li and 2-chlorobenzopyrazole was 92.1% with the use of an NiCl2(PCy3)2 catalyst. this method eliminates the need for expensive palladium reagents and nickel complexes that require synthesis.Graphical Abstract

Abstract:Two different types of coupling patterns for the Pd(0)-catalyzed coupling reaction of allenic/propargylic zinc reagents with organic halides or propargylic carbonates (acetate) with the corresponding organometallic reagents were observed. After studying the controlling factors on the regioselectivity of this reaction, we demonstrated that the steric hindrance of both reactants and the types of organic halides determine the regioselectivity of this coupling reaction. By subtle choosing of the substrates, the regioselectivity can be tuned. On the basis of these results, new methodologies for the highly regio- and stereoselective synthesis of 6-substituted hex-5-yn-2-enoates and 4,6-dialkylhexa-2,4,5-trienoates have been developed. Some of the products synthesized by the carbonate protocol cannot be prepared by the lithiation protocol because the regioselectivity of lithiation of dialkyl-substituted internal alkynes is an intrinsic problem.

Abstract:The alkyne motif is present in several naturally occur‐ring molecules and specialty chemicals. The Sonogashira reaction is recognized as one of the most exploited methods for the synthesis of substituted alkynes. While palladium catalysts have been traditionally employed, recent efforts have been directed towards developing alternatives based on Earth‐abundant metals such as nickel. However, nickel‐catalyzed Sonogashira couplings still present important substrate limitations. While more reactive aryl iodides have been well documented, the coupling of more challenging yet cost‐effective aryl bromides and chlorides lags significantly behind. Herein, we present a method for the nickel‐catalyzed Sonogashira coupling of aryl bromides and chlorides, whose discovery was accelerated thanks to high‐throughput experimentation. Fine‐tuning of the bipyridine ligand was essential to activate chemoselectively C( sp 2 )–Br or C( sp 2 )–Cl bonds. A wide range of functionally diverse alkynes, including examples of active ingredients and their intermediates, were accessed in a single step and with yields up to 98%. The developed methodology is expected to be widely adopted by academia and industry, marking a step forward in the field of base metal catalysis.

Abstract:We describe the first nickel-N-heterocyclic carbene catalyzed Negishi cross-coupling reaction of a variety of unactivated aryl chlorides, heterocyclic chlorides, aryl dichlorides, and vinyl chloride. Abstract:A novel methodology for the synthesis of trisubstituted allenes is reported. Lithiation of 1-aryl-3-alkylpropadienes and subsequent transmetalation with zinc bromide followed by Pd-catalyzed Negishi coupling reactions with halides afforded the corresponding trisubstituted allenes in a highly regioselective fashion with moderate to excellent yields. A plausible regioselective lithiation mechanism was proposed on the basis of deuterium labeling experiments.

Abstract:Due to the smaller size of nickel compared to palladium, the C–S cross-coupling of sterically challenging aryl electrophiles with alkyl thiols under nickel catalysis remained elusive. Herein, we report the nickel-catalyzed cross-coupling of alkyl thiols with aryl triflates bearing functional groups in ortho-position relative to the leaving group using Ni(cod)2/DPEphos (L1) or dppbz (L2) as the catalytic system. For substrates featuring non-coordinating ortho-substituents, the reaction operates under mild conditions using L1, while for electrophiles bearing coordinating groups, the ligand L2 and elevated temperatures are required. The synthetic utility could be demonstrated on numerous examples, including biologically relevant compounds, and on larger scale. Instead of Ni(cod)2, more cost-efficient Ni(OAc)2 can also be employed in the presence of zinc as reductant. Furthermore, insights into the reaction mechanism were obtained by competition experiments, isolation of organometallic intermediates and computations.

Abstract:This report describes a detailed study of Ni phosphine catalysts for the Suzuki-Miyaura coupling of dichloropyridines with halogen-containing (hetero)aryl boronic acids. With most phosphine ligands these transformations afford mixtures of mono- and diarylated cross-coupling products as well as competing oligomerization of the boronic acid. However, a ligand screen revealed that PPh2Me and PPh3 afford high yield and selectivity for monoarylation over diarylation as well as minimal competing oligomerization of the boronic acid. Several key observations were made regarding the selectivity of these reactions, including: (1) phosphine ligands that afford high selectivity for monoarylation fall within a narrow range of Tolman cone angles (between 136° and 157°); (2) more electron-rich trialkylphosphines afford predominantly diarylated products, while less-electron rich di- and triarylphosphines favor monoarylation; (3) diarylation proceeds via intramolecular oxidative addition; and (4) the solvent (MeCN) plays a crucial role in achieving high monoarylation selectivity. Experimental and DFT studies suggest that all these data can be explained based on the reactivity of a key intermediate: a Ni0-π complex of the monoarylated product. With larger, more electron-rich trialkylphosphine ligands, this π complex undergoes intramolecular oxidative addition faster than ligand substitution by the MeCN solvent, leading to selective diarylation. In contrast, with relatively small di- and triarylphosphine ligands, associative ligand substitution by MeCN is competitive with oxidative addition, resulting in selective formation of monoarylated products. The generality of this method is demonstrated with a variety of dichloropyridines and chloro-substituted aryl boronic acids. Furthermore, the optimal ligand (PPh2Me) and solvent (MeCN) are leveraged to achieve the Ni-catalyzed monoarylation of a broader set of dichloroarene substrates.

Abstract:The cadmium(II) chloride‐catalyzed dehydrative C−P cross‐coupling reaction of propargyl alcohols with diarylphosphine oxides is reported. Several propargyl alcohols including those bearing the sterically demanding tert‐butyl group at the triple bond terminus can be used as good substrates in the reaction to produce the corresponding allenylphosphine oxides in good to high yields in acetonitrile at 100 °C. The reaction can also be easily scaled up to a gram‐scale synthesis. A mechanism study indicates that the reaction may proceed through a process of propargylic substitution to generate phosphonite intermediates followed by [2,3] sigmatropic rearrangement to produce the allenyl products, rather than through a common allenylative substitution resulting from P‐nucleophilicity.

Abstract:A novel and efficient method for the palladium-catalysed and highly regioselective synthesis of trisubstituted allenes via deaminative coupling of tertiary propargylamines with arylsiloxanes is reported. Both unactivated propargylamines and arylsiloxanes were activated in situ using Et 3 N·3HF, followed by cross-coupling to facilitate C–C bond formation through C–N bond cleavage. The method demonstrates exceptional functional group compatibility, high yields, and readily accessible coupling partners.
