Abstract:Context: Use of herbal medicine throughout the world is increasing. Plants still remaining the primary source of supply of many important drugs used in modem medicine. Spinacia Oleracea i,e spinach leaves contain more number of anti-inflammatory phytochemcicals such as carbohydratie, tanins and phenolic compounds, saponins, flavinoids and steroid. Better collagenation seen under the influence of the flavinoids, phenolic compound and saponins which is responsible for the free radical scavenging activity and are believed to be some of the most important component for wound healing and antioxident activity. Spinacia Oleracea scavenges free oxygen redicals and increases the catalase level in granulation tissue. Plants still remaining the primary source of supply of many important drugs used in modem medicine. Considering its medicinal value and availability in our country this study was undertaken to evaluate the antiinflammatory effect of the Spinacia Oleracea leaf extract in rat models. Material and Methods: The experiments were carried out on 30 (thirty) Swiss male albino rats. They were collected from the ICDDRB, Dhaka. The rats were of male weighing between 150- 200gm which were divided randomly into 5 groups each having 6 rats. Groups were labeled as group-I, group-II, group-III, group-IV and group-V. The anti inflammatory effect of Spinacia Oleracea leaf extract in experiment rat were evaluated and compared with the anti inflammatory effects of aspirin and hydrocortisone. The study was prospective experimental type and was conducted in the department of Pharmacology, Dhaka Medical College, Dhaka, from July 2011 to June 2012. Result : Administration of mathanolic extract and water extract of Spinacia Oleracea leaf at a dose of 200mg/kg body weight orally produced a significant (P<0.05) anti-inflammatory effect, and the percentage of inhibition of oedema formation was 28.75% and 40.79% respectively. There were highly significant (P<0.05) percentage of inhibition of oedema formation was observed in aspirin (40.52%) and in hydrocortisone (47.71%). Abstract:Cissampelos pareira is a medicinal plant with conflicting local claims regarding its usage in the management of fertility disorder. This study investigates the effect of the ethanolic extract of C. pareira on fertility indices in female albino rats. Ethanolic extract of C. pareira was administered orally for 14 days to female rats placed in different groups. Levonorgestrel (0.14mg/g) was orally administered to certain rat groups for 7 days to induce transient infertility before other treatments. The rats were sacrificed at the completion of their respective oral administration route. Blood samples were collected through cardiac puncture for hormonal and lipid profiling, which were analysed using commercial standard enzyme-linked immunosorbent assay (ELISA) kit. Organs were harvested for the assessment of oxidative parameters. Phytochemical screening reveals the presence of alkaloid, saponin, glycoside, tannin, steroid and flavonoid in the extract. The extract has no significant effect on progesterone, significantly reduced testosterone (p<0.05), but increased prolactin and estradiol concentrations (p<0.05). No significant hormonal difference in groups pre-administered levonorgestrel. The extract significantly elevated serum triglyceride, but reduced cholesterol level (p<0.05), even in groups pre-administered with levonorgestrel. The extract significantly reduced H2O2 and SOD activities (p<0.05) in the ovary and kidney respectively. Following levonorgestrel pre- administration, the extract significantly reduced H2O2 and GSH in the liver and kidney, elevated SOD activities in the liver, ovary and brain (p<0.05). This study demonstrated the antifertility properties of the ethanolic extract of Cissampelos pareira, shown by its effects of the hormonal and lipid profile, with moderate antioxidant effect.

Abstract:The aim of this work was to evaluate diuretic activity of the ethanol extract of Euphorbia hirta leaves and elucidate the possible mechanism of its action. The diuretic activity was studied on male albino rats in comparison to furosemide as a standard drug. Two doses of E. hirta extract (300 and 600 mg/kg) were administered and the urinary volume and electrolyte (Na + , K + ) concentrations were measured. Results showed that administration of the ethanol extract of E. hirta led to significantly increased urinary volume and excretion of urinary electrolytes such as sodium, potassium and chloride in 24 h urine compared to that for normal animals. Among the two selected doses, 600 mg/kg body weight exhibited higher diuretic activity level than 300 mg/kg dose. Although both these levels were statistically significant when compared to control in respect of all parameters, these levels were lower compared to the standard drug. Hence, the ethanol extract of E. hirta exhibited a dose dependent diuretic activity. Upon the isolation and identification of active compounds from E. hirta ethanol extract, it was found that lupeol and quercetin were the major constituents responsible for the diuretic activity in rats. The present study confirmed validity of the ethnopharmacological use of E. hirta as a diuretic agent under experimental conditions studied.

Abstract:Objective: To examine the effect of Cissus populnea root on testicular function of male Wistar rats. Methods: Twenty albino male Wistar rats [(100.0 ±25.5) g] were selected into four groups randomly. The control group were administered 0.20 mL of distilled water while the other groups received 0.05 mL, 0.10 mL, and 0.20 mL/kg body weight of ethanol infusion. Oral administration was done using oropharyngeal cannula once daily for 14 days. Animals were thereafter sacrificed on day 15. Data obtained from this study were analysed using one-way analysis of variance (ANOVA) with multiple comparisons, post-hoc Tukey HSD was subsequently used and values at P<0.05 were considered statistically significant. Results: There were significant increases in serum testosterone, testes-body weight ratio, luteinising hormone and follicle stimulating hormone, respectively, compared with those in control group. Similarly, increase in acid phosphatase, alkaline phosphatase, protein, glycogen and cholesterol levels were more significant compared with those in control group. Conclusions: The findings of this study reveals enhanced secretions of the sex hormones and other indices at 0.05 mL/kg body weight administration of Cissus populnea root ethanol infusion. However, dosages exceeding 0.05 mL/kg body weight gradually depletes the level of reproductive indices assayed for in this current study. Therefore, caution must be taken when consuming ethanol infusion of Cissus populnea root above the dosages used in this current study.
Routine clinical outcome measures for patients with severe mental illness: CANSAS and HoNOS

Abstract:Background Two recently developed measures intended for clinical use are HoNOS (Health of the Nation Outcome Scales), measuring social functioning, and CANSAS (Camberwell Assessment of Need Short Appraisal Schedule), for measuring need. Aims We investigated the association between CANSAS and HoNOS. Results For some domains there were substantial associations, with high HoNOS rating correlated with CANSAS rating of the presence of a need. For other areas the agreement was less than might be expected. Seven factors were identified within the 22 CANSAS domains, and the presence of needs in the Drug/alcohol and Activities of daily living factors was associated with high scores in the related HoNOS domains. Conclusions CANSAS and HoNOS assessments differ. HoNOS can track changes in social functioning over time, but may be less useful for treatment planning, and should not be used to infer the level of morbidity in a case-load. CANSAS indicates when treatment should be commenced or continued. It can also be used as a case-load measure, but may be insufficiently sensitive to be used as an outcome measure at the individual level.

Abstract:The objective of the present investigation was to study the antidiabetic and diuretic potential of Anogeissus latifolia (A. latifolia) bark in experimental rats. The A. latifolia bark was extracted with hydro-alcoholic solvent by cold extraction method. Acute toxicity study was performed according to OECD 425 guidelines for hydro-alcoholic extracts of A. latifolia bark (ALBE). The dose of 150 mg/kg p.o. and 300 mg/kg, p.o. of ALBE was selected for further studies. Animals were prepared diabetic by administration of alloxan (120 mg/kg, i.p.). The albino rats were divided in to five groups for oral glucose tolerance test (OGTT) and alloxan induced anti diabetic model with six animals in each group. Diabetic animals were treated with hydro-alcoholic extract of A. latifolia bark for 20 days. The blood glucose level was estimated according to standard procedures. Diuretic activity hydro-alcoholic extracts of A. latifolia was evaluated by Lipshitz method. The result shows that hydro-alcoholic extract from bark of Anogeissus latifolia 300 mg/kg (ALBE-II) shown significant hypoglycemic activity as compared to glibenclamide and diabetic group. The ALBE does not exhibit significant diuretic activity which is considered as positive marker in diabetic phenomena. Hence in present study extract of A. latifolia posses antidiabetic activity. This study may be benchmark in future to use of this drug scientifically.

Abstract:The aim of our presented study is to assess the effect of Rhizome extract of Iris pseudacorus L. and seed extract of Dolichos biflorus L. as preventive agent in experimentally induced urolithiasis model in rats. Rats were administered Sodium Oxalate (70 mg/kg, i. p.) in drinking water for 28 days in drinking water. In addition to this, Saponin extract of Iris pseudacorus and Dolichos biflorus of low dose and high dose were administered along with Sodium Oxalate on 14-28th day. After the experimental period, blood samples were collected by cardiac puncture to analyse for Creatinine, Calcium, Blood Urea Nitrogen (BUN), Phosphorus, Uric acid, Alkaline Phosphate, Potassium, and Alanine Amino Transferases followed by various antioxidants and kidney histopathology. The ethylene glycol feeding resulted in an increased level of all parameters evaluated compared to normal rats. All these conditions were reversed with plant extract treatment. Histopathological analysis also showed that rats treated with Sodium Oxalate had large deposits of calcium oxalate crystals, and that deposits were reduced in rats treated with plant extract. Results were also compared with the marketed product cystone as a standard. These data suggest that Iris pseudacorus and Dolichos biflorus Saponin extracts has a protective activity against urolithiasis.
Phytochemical investigation and nephroprotective potential of Sida cordata in rat

Abstract:Background: Plants are an efficient source of natural antioxidant against free radicals causing kidney damages. Sida cordata ethyl acetate fraction has been reported for strong in vitro antioxidant potency, previously. In the present study, our objective was to evaluate its in vivo antioxidant potency against CCl4 induced nephrotoxicity and investigates the bioactive phytochemicals by HPLC-DAD analysis. Methods: Phytochemical analysis was performed by HPLC-DAD methodology. For in vivo study, 42 male Sprague-Dawley rats were treated with alternatively managed doses for 60 days. Group I animals were remained untreated. Group II animals were treated with vehicle (1 mL of olive oil) by intragastric route on alternate days. Group III was treated with 30% CCl4 (1 mL/kg b.w.) i.p. Group IV was treated with 30% CCl4 (1 mL/kg b.w.) i.p and silymarin intragastric. Group V and VI rats were treated with 30% CCl4 and SCEE (150 and 300 mg/kg b.w., respectively) intragastric. Group VII animals were treated with SCEE (300 mg/kg b.w.) intragastrically. Blood parameters, Serum proteins and urine profile were investigated. Activities of tissue enzyme i.e. catalase, peroxidase, superoxide dismutase, glutathione-S-transferase, glutathione reductase, GSH and γ-GT were evaluated. Histopathological observations, total protein contents, lipid peroxidation, DNA damage and relative weight were also analyzed. Results: Gallic acid, catechin and caffeic acid were identified in SCEE fraction by HPLC-DAD. Decrease in the count of red blood cells, neutrophils, eosinophils and concentration of hemoglobin whereas increase in lymphocyte count and estimation of sedimentation rate (ESR) with 1 mL CCl4 (30% in Olive oil) administration (30 doses in 60 days) was restored dose dependently with co-treatment of SCEE (150 and 300 mg/kg b.w.). Treatment of rats with CCl4 markedly (P < 0.01) increased the count of urinary red blood cells and leucocytes, concentration of urea, creatinine and urobilinogen and specific gravity whereas creatinine clearance was reduced. Serum level of total protein, albumin, globulin, nitrite, creatinine and blood urea nitrogen (BUN) was significantly increased (P < 0.01) by CCl4 treatment. The activity of antioxidant enzymes; catalase, superoxide dismutase, glutathione peroxidase, glutathione-S-transferase and glutathione reductase and content of reduced glutathione was decreased (P < 0.01) significantly. However, increased concentration (P < 0.01) of thiobarbituric acid reactive substances and histopathological injuries were noticed in the renal tissues of rats after the treatment with CCl4. Co-administration of SCEE, dose dependently, protected the alterations in the studied parameters of rats at 150 and 300 mg/kg b.w. The present study revealed that SCEE could be used as a possible remedy for renal toxicity abnormalities. Conclusion: These results are an evidence of the renal protective role of S.cordat ethyl acetate fraction against CCl4 induced nephrotoxicity in rats which may be due to its antioxidant compounds.

Abstract:Diabetes mellitus (DM) is a serious health issue for the general public. In this work, male diabetic rats were used to determine the impact of Citrullus colocynthis and Phyllanthus Emblica fruits ethanolic extract (EECC and EEPE). Six groups of thirty mature rats (male Sprague Dawley) were examined. In the first group, normal rats have been fed a balanced diet served as the negative control group (C-); in the second group, diabetic rats served as the control positive (C+). Diabetic rats in groups 3, 4, 5, and 6 received daily treatments of (200 and 400 mg kg-1 body weight (BW) EECC and EEPE, respectively. After four weeks of treatment, Biochemical indicators were examined in all blood samples. Streptozotocin was injected and resulted in a significant reduction in feed intake (FI), feed efficiency (FER) body weight gain (BWG), serum insulin, high-density lipoprotein (HDL-c), globulin, albumin, and total protein, as well as antioxidant enzymes, glutathione (GSH) and superoxide dismutase (SOD) in pancreatic tissue, whereas a significant increment was noted in serum glucose, triglyceride (TG), total cholesterol (TC), low & very low-density lipoprotein cholesterol (LDL-c and VLDL-c), atherogenic index (AI), Alkaline phosphatase (ALP), Aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, uric acid, creatinine, tumor necrosis factor-α, creatine phosphate kinase (CPK) and lactate dehydrogenase (LDH) and lipid peroxidation (MDA) in pancreatic tissue. The treatment of diabetic animals with different ethanolic extracts caused improvements in all previously measured parameters, with EEPE (400 mg kg-1 BW) showing the greatest improvement.

Abstract:Pericopsis laxiflora is among the medicinal plants used by the traditional practitioner in the treatment of many diseases. However, data on its diuretic power are lacking. Moreover, the effect of the preparation of traditional galenic formulations on the composition of phytocompounds and essential minerals and metals is unknown. The aim of this study was to show the effect of treatment on the phytocompounds and minerals composition and to evaluate diuretic effect of Pericopsis laxiflora aqueous axtracts. The tube characterization reactions revealed flavonoids, sterols, terpenes, catechin tannins polyphenols and saponins in aqueous extracts. Gallic tannins and quinones are absent, All compounds were generally present in same proportions in two aqueous extracts. Chemical analysis of all aqueous extracts by atomic absorption spectrometry (AAS) showed effect of model extraction on minerals. Copper (0.3116 mg / 100g), magnesium (260.7 mg /100 g), selenium (0.00904 mg /100 g), zinc (4.52 mg /100 g), manganese (75.029 mg /100g) are more concentrated in maceration than a decoction. For decoction of same plant, the situation is reversed with sodium (58.6mg /100g), calcium (12670 mg/100g), iron (18.51mg/100g) and potassium (2781 mg /100g). For pharmacological investigation, 32 rats Sprague Dawley male weighing 385-433g are isolated each in metabolic cages from 6 pm to 8am and treated with 100mg/kg of Pericopsis laxiflora aqueous extracts. From 8am to 2pm the animals received only tap water in metabolic cages. The volumes of water intake and urine were not significantly different in the two groups (treated and untreated).The diuretic effect was not observed with 100mg/kg (b/w) . In addition polyphenols, flavonoids, saponins and minerals such as potassium, magnesium and selenium are indicators of the vasodilatory and cardioprotective functions of Pericopis laxiflora aqueous extracts. The prescription of these extracts in traditional medicine must take into account the toxicity of the plant associated with a calcium and manganese content. Antidiabetic and hepato-renal protective effects of medicinal plants in STZ induced diabetic rats

Abstract:Abstract The antidiabetic and hepato-renal protective effects of Citrullus colocynthis and Momordica charantia ethanol extracts were investigated in streptozotocin (STZ) induced diabetic male albino rats. Diabetic rats were treated with C. colocynthis, M. charantia or C. colocynthis + M. charantia mixed extract at a dose of 250 mg /kg body weight per oral per day for 21 days. The mean body weight of all the diabetic rat groups on day 1 of treatment (day 10 of diabetes) was significantly lower than the normal control rat group (P<0.05). The blood glucose level of all the diabetic rat groups on day 1 of treatment (day 10 of diabetes) was significantly (P 200 mg/dl) than the normal control rat group (95.5 ± 2.7). At the end of treatment (day 21), the diabetic rats treated with plant extracts showed significant increase (P<0.05) in body weight and significant (P<0.05) reduction in blood glucose level when compared to diabetic control animals. Significant increase (< 0.05) was observed in the serum bilirubin, alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), urea and creatinine levels of diabetic control rat group. The serum levels of these liver and kidney-related parameters of diabetic rats treated with plant extract were significantly lower when compared to diabetic control rat group (p < 0.05). Photomicrographs of liver and kidney microsections from diabetic rats treated with these plant extracts showed amelioration in the hepato-renal histoarchitectures. It was concluded that the C. colocynthis and M. charantia methanol extracts are antidiabetic and hepato-renal protective in STZ induced diabetic male rats. Treatment of the diabetic rats with C. colocynthis + M. charantia mixed extract is more effective in the amelioration of diabetes and hepato-renal injuries in STZ induced diabetic male rats.

Abstract:Medicinal plants are efficient ameliorator of oxidative stress associated with diabetes mellitus. In this study, ethyl acetate fraction (SCEE) of Sida cordata was investigated for scientific validation of its folk use in diabetes. Antidiabetic effect of SCEE was confirmed by antihyperglycemic activity in normal glucose loaded and diabetic glucose loaded animals as well as normal off feed animals. Confirmation of antidiabetic activity and toxicity ameliorative role of S. cordata was investigated in a chronic multiple dose treatment study of fifteen days. A single dose of alloxan (120 mg/kg) produced a decrease in insulin level, hyperglycemia, elevated total lipids, triglycerides, and cholesterol and decreased the high-density lipoproteins. Concurrent with these changes, there was an increase in the concentration of lipid peroxidation (TBARS), H 2 O 2 , and nitrite in pancreas, liver, and testis. This oxidative stress was related to a decrease in glutathione content (GSH) and antioxidant enzymes. Administration of SCEE for 15 days after diabetes induction ameliorated hyperglycemia, restored lipid profile, blunted the increase in TBARS, H 2 O 2 , and nitrite content, and stimulated the GSH production in the organs of alloxan-treated rats. We suggested that SCEE could be used as antidiabetic component in case of diabetes mellitus. This may be related to its antioxidative properties.

Group 1 is the normal control group; Group 2 is the negative control group (paracetamol-induced but untreated); Group 3 received 140 mgkg-1 of silymarin; Groups 4, 5, and 6 received 100, 300, and 500 mgkg-1 body of the aqueous polyherbal extract respectively for seven days. Therefore, the blood samples were collected and evaluated for creatinine, urea, sodium, potassium, and chloride ions concentrations to measure kidney function. The results revealed that the aqueous extract of the polyherbal mixture significantly (p<0.05) ameliorated the kidney function test parameters that were analyzed by lowering their concentrations previously elevated by the paracetamol intoxication comparable with the normal control rats. Therefore, it can be concluded that the polyherbal mixture extract expressed an anti-nephrotoxic effect against renal toxicity induced by paracetamol in Wistar albino rats.
Effect of Flavonoid Rich Fraction of Coriandrum sativum Leaf on Lipid Profile, Nitric Oxide, Ang II and Cardio Histopathology in L-NAME Intoxicated Experimental Rats

Abstract:Coriandrum sativum has been used in traditional medicines as an anti-inflammatory, analgesic, and antibacterial agent. It is recently been shown to have antioxidant activities. This study evaluated the effects of Flavonoid Rich Fraction of Coriandrum sativum Leaf (FRFCSL) on Lipid Profile, Nitric Oxide, Ang II and Cardio Histopathology in L-NAME intoxicated Experimental Rats. Standard analytical method and DPPH radical scavenging activity were employed for the phytochemicals and antioxidant analysis respectively. The acute toxicity study showed the extract and flavonoids fraction are nontoxic at 5000mg/kg body weight. Thirty rats were divided into five groups of six (6) rats each. Group 1 was administered water and feed only, Group 2 was administered 40mg/kg N-Nitro-L-Arginine Methyl Ester (L-NAME) without treatment, Group 3 through 5 were administered 40mg/kg N-Nitro-L-Arginine Methyl Ester (L-NAME) with concomitant administration of Captopril (20mg/kg b.w), FRFCSL (200mg/kg b.w) and FRFCSL (400mg/kg b.w) respectively for 21 days. In phytochemical screening, presence of flavonoids, saponins, tannins, alkaloids, cardiac glycosides, steroids were confirmed as part of secondary metabolites in the extract. The flavonoid fraction showed increase % DPPH radical inhibition in concentration-dependent manner. FRFCSL treatment significantly (p<0.05) and dose-dependently lowered low density lipoprotein and triglycerides. Also It significantly (p<0.05) prevented L-NAME induced decrease in serum angiotensin II, high density lipoprotein and serum nitric oxide concentrations compared to the untreated rats. The Flavonoid Rich Fraction of Coriandrum Sativum Leaf showed great potential as antihypertensive, antihyperlipidemic and cardioprotective agent in rats thus confirming its usefulness in traditional health therapy and potential for antihypertensive drug development.

Abstract:Ethnopharmacological relevance: Saraca asoca (Roxb.) de Wilde, Ashok, is a popular traditional plant used for gynecological disorders. In India, the juice of Ashok flowers is traditionally consumed as a tonic by women in case of uterine disorders. But despite the use, its estrogenic potency is not yet evaluated and thus lacks the scientific recognition and acclaim. Aim of the study: This study is designed to investigate the estrogenic potential of standardized ethanolic extract of Saraca asoca flowers (SAF) using ovariectomized (OVX) female albino Wistar rat model. Materials and methods: Saraca asoca flowers were extracted in ethanol using hot maceration technique and the extract was standardized in terms of content of four phytoestrogens like quercetin, kaempferol, β-sitosterol and luteolin using HPTLC technique. Safety of the extract was evaluated at a dose of 2000mg/kg body weight in female albino Wistar rats as per the OECD guidelines. Bilateral ovariectomy surgery was performed for the excision of both the ovaries. The OVX animals were treated with the ethanolic extract of SAF at three dose levels- 100mg/kg, 200mg/kg and 400mg/kg body weight in distilled water as a vehicle, orally once a day for two weeks. Estradiol valerate was employed as a modern drug for comparative evaluation of the results. Estrogenic potency was studied by assaying the activities of serum and plasma marker enzymes and hormones viz. G6PDH, LDH, 17β-estradiol, progesterone along with cholesterol, triglycerides and HDL, and vaginal cornification. The uterotrophic effect was evaluated by studying the histoarchitecture of the uterus, effect on uterine weight and changes in the levels of uterine glycogen content. Results: HPTLC revealed the presence of markers like quercetin, kaempferol, β-sitosterol and luteolin from the ethanolic extract of SAF. The content of the four markers was found to be 1.543mg/g, 0.924mg/g, 4.481mg/g and 2.349mg/g, respectively. SAF extract was found to be safe at an oral dose of 2000mg/kg body weight in rats. Among the three doses administered to ovariectomized rats, treatment with high dose was found to be more efficacious when compared with ovariectomized rats. Conclusion: The findings of this study firmly support the estrogenic potency of ethanolic extract of SAF which may be by the reason of phytoestrogens.

Abstract:The present study was undertaken to evaluate antidiabetic and antioxidant activities of Cayratia trifolia root extract against streptozotocin induced diabetes in experimental rats to scientifically validate its use against diabetes in some parts of India. Ethanolic extract, showing the highest activity in in vitro experiments, was prepared in saline and administered orally to streptozotocin induced albino Wistar diabetic rats for 21 days. Biochemical parameters liver and muscles glycogen and in vivo antioxidant activity in normal, diabetic control, standard (metformin) and treated animals were determined and compared. Attempt was made to isolate, purify and characterize one of the major secondary metabolites in extract by range of chromatographic and spectroscopic techniques. Treatment of streptozotocin induced diabetic rats with ethanolic root extract (500 mg/kg) caused significant (P < 0.01) reduction in blood glucose (312–178 mg/dL), increase in body weight (181–219 g) and serum insulin (1.28–2.26 IU/dL). It also maintained lipid profile and tests of liver and kidney functions within normal range as compared to diabetic control rats and almost at par with standard drug metformin. The oxidative stress induced decline in glutathione and catalase in liver and kidney tissues showed recovery nearly to normal level as a function of treatment. The GC–MS profile of the extract showed relatively high concentration of β-sitosterol which was characterized by different spectroscopic and chromatographic techniques. The result scientifically and comprehensively validate the reported use of roots of this indigenous plant against diabetes. A strong antioxidant activity of the ethanolic root extract suitably compliments the antidiabetic effect.

Abstract:Traditional applications of the roots of Chlorophytum borivilianum Santapau & R.R.Fern. include spermatogenic potential. Using rats as test subjects, the potential of alcoholic extract (AE-SM) and n-Butanol fraction (NB-SM), which is rich in saponin, to alter the levels of testicular testosterone and cholesterol as well as mRNA expression of the 3β-Hydroxy-∆5- steroid dehydrogenase (3β-HSD) and Acute Steroid Regulatory Protein (StAR) were determined. Histological analyses of testicular tissues were performed, and the quantity of sperm in the seminal fluid was measured. The testosterone concentration increased in AE-SM and NB-SM in a dose-dependent and statistically significant manner. In-vivo mRNA expression of the StAR and 3β-HSD enzymes was also enhanced by the test samples. When compared to control animals, AE-SM and NB-SM increased testicular testosterone by 11.46 and 12.98 fold, respectively, while the extract and fractions revealed a 5.72 and 7.70 fold alteration in StAR mRNA expression. In testicular tissues, AE-SM and NB-SM increased 3β-HSD activity by 4.98 and 9.05 fold, respectively, while changing the expression of the 3β-HSD mRNA by 6.46 and 15.54 factor, respectively. The fluid sampled from the caudal region of the AE-SM and NB-SM indicated an increased sperm concentration, and histological examinations revealed dense Sertoli cell cytoplasm and an increase in the number of sperms in the luminal part of the seminiferous tubules. The components of AE-SM and NB-SM might stimulated testosterone biosynthesis in-vivo by modifying mRNA expression of selected steroidogenic enzymes. By modifying the mRNA expression of the StAR and 3β-HSD, AE-SM and NB-SM can raise the level of testosterone in testicular tissues. As evidenced by an elevated sperm concentration discovered within the seminiferous fluid and histological studies, overexpression of steroidogenic enzymes increased testicular testosterone levels and may have stimulated the spermatogenesis process.Saponin enriched n-butanol fraction might have interacted with CREB and additionally stimulated LH receptors to elevate the StAR and 3β-HSD mRNA expression.

Abstract:Background: Solanum surattense Burm. (Solanaceae) is traditionally used for management of various ailments. The study was conducted for provision of pharmacological justification for folkloric uses of Solanum surattense in the treatment of dysuria. Methods: Rats were randomly divided into 5 groups, each of (n = 6). Aqueous methanolic fruit extract of S. surattense were also administered intraperitoneally to the rats at doses of 50, 70 and 100 mg/kg. Furosemide (10 mg/kg i.p) was used as standard drug whereas controls were given saline solution (40 mL/kg i.p). The electrolytes in urine were measured using a flame photometer whereas serum sodium, potassium, calcium, bicarbonate and blood urea nitrogen (BUN) were determined by using an automatic analyzer. Urine osmolality was assayed by the micro-osmometer. Results: The extract S. surattense induced diuretic effects in a dose-dependent manner as compared with control. Upon administration of extract (70 and 100 mg/kg), we observed the prominent (p < 0.01) increase in the urine volume and osmolality in comparison to control group. However, plant extract (100 mg/kg) significantly increase the urinary electrolyte excretion especially calcium (p < 0.05) to that of the furosemide whereas level of magnesium remains constant. Moreover, our results showed a decrease in serum levels of sodium, potassium, calcium and blood urea nitrogen (BUN), but concentration dependent increase in bicarbonate was found in the test groups. There was no substantial change in the pH of urine samples of the extract-treated groups. Conclusion: These results indicate that S. surattense investigated exert its action by causing diuresis in the treatment of dysuria.

Abstract:Oxidative injury or cellular damage arising from free radicals or reactive oxygen species (ROS) has been implicated in the pathogenesis of many human diseases. These ROS are generated through normal metabolism of drugs, environmental chemicals and other xenobiotics as well as endogenous chemicals. Accumulated evidence suggests that ROS can be scavenged through chemoprevention utilizing natural antioxidant compounds present in foods and medicinal plants. The aim of this study is to evaluate antioxidant activities of aqueous extract of Bryophyllum pinnatum using carbon tetrachloride-induced albino rats as model. An in-vivo antioxidant activity was carried out on the CCl4 induced rats by administration of aqueous extract of Bryophyllum pinnatum at doses of 50, 100 and 200mg/Kg bw for 14 consecutive days, and one group was treated with Silymarin at a dose of 50 mg/Kg as a standard drug. The antioxidant activities were estimated by evaluating its effects on antioxidant parameters in the tissues of albino rats (Serum, Liver and Heart). The activity of catalase (CAT), Superoxide dismutase (SOD), Glutathione peroxidase (GPx) and the levels of MDA and GSH were evaluated in tissue homogenates. Results showed that the rats fed with the higher concentrations of Bryophyllum pinnatum had significant increase (p<0.05) in glutathione content (GSH), GPx, SOD and catalase activities relative to normal and standard control. However, the level of MDA was decreased (p<0.05) for the treated groups (100 and 200mg/kg). These results suggest that aqueous extracts of Bryophyllum pinnatum has active substances contributing to varied degrees of potent antioxidant activity and could serve as important source of antioxidants in food, cosmetics and pharmaceutical industries.
