Abstract:A transient continuum model of the ODEP chip containing single circular particle inside is constructed based on multi-physical field coupling. The dielectrophoresis force and liquid viscous resistance acting on particle are calculated by employing the full Maxwell stress tensor. The coupled flow field, electric field and particle are solved by the arbitrary Lagrange–Euler (ALE) method simultaneously. The throughout dynamic process of particle in the ODEP chip is demonstrated, and the effect of several critical parameters on particle electrodynamics is illuminated. The additional disturbing effect of the photoconductive layer on the electric field as well as the micro-channel wall on the flow field is presented to clarify the particle motion in the vertical direction. The results in this study provide a detailed understanding of the particle dynamics in the ODEP chip.

Abstract:Cell separation has become @important in biological and medical applications. Dielectrophoresis (DEP) is widely used due to the advantages it offers, such as the lack of a requirement for biological markers and the fact that it involves no damage to cells or particles. This study aimed to report a novel approach combining 3D sidewall electrodes and contraction/expansion (CEA) structures to separate three kinds of particles with different sizes or dielectric properties continuously. The separation was achieved through the interaction between electrophoretic forces and inertia forces. The CEA channel was capable of sorting particles with different sizes due to inertial forces, and also enhanced the nonuniformity of the electric field. The 3D electrodes generated a non-uniform electric field at the same height as the channels, which increased the action range of the DEP force. Finite element simulations using the commercial software, COMSOL Multiphysics 5.4, were performed to determine the flow field distributions, electric field distributions, and particle trajectories. The separation experiments were assessed by separating 4 μm polystyrene (PS) particles from 20 μm PS particles at different flow rates by experiencing positive and negative DEP. Subsequently, the sorting performances of the 4 μm PS particles, 20 μm PS particles, and 4 μm silica particles with different solution conductivities were observed. Both the numerical simulations and the practical particle separation displayed high separating efficiency (separation of 4 μm PS particles, 94.2%; separation of 20 μm PS particles, 92.1%; separation of 4 μm Silica particles, 95.3%). The proposed approach is expected to open a new approach to cell sorting and separating.

Ecological and Mutation-Order Speciation in Digital Organisms

Abstract:Reproductive isolation between populations often evolves as a by-product of independent adaptation to new environments, but the selective pressures of these environments may be divergent (“ecological speciation” or uniform (“mutation-order speciation.” In this study, we use an artificial life platform to directly compare the strength of reproductive isolation (specifically, postzygotic) generated by ecological and mutation-order processes. We also tested the effect of gene flow as well as the dimensionality (i.e., number of selective pressures) of the environments on the strength of postzygotic isolation. We found that ecological speciation generally formed stronger isolation than mutation-order speciation, mutation-order speciation was more sensitive to gene flow than ecological speciation, and environments with high dimensionality formed stronger reproductive isolation than those with low dimensionality. How various factors affect the strength of reproductive isolation has been difficult to test in biological organisms, but the use of artificial life, which provides its own genetic system that evolves, allowed us to computationally test the effect of these factors more easily.

Abstract:Statistically studied are the equilibrium characteristics of a subsystem of mobile charges of one sort, taking into account the subsystem of fixed charges of the opposite sign creating a compensating electric background. The distribution of these charges under the influence of the external field is invariable. To represent free energy of the subsystem of mobile charges in the form of a functional of their density and to calculate cell potentials of the mean forces by the method of conditional distributions, a cumulant expansion with respect to the renormalized Mayer functions is used. To take into account the screening effects, the results of the collective variables method are used. A system of integral equations for the potentials of mean forces is obtained that accounts for the effects of near- and long-range interactions. The calculations are made in the lattice approximation. The correlation component distinguished in the expression for the binary distribution function makes it possible to calculate the correlated and uncorrelated parts of the electric potential using the Poisson equation. In the case of sufficiently small electric fields, a linear expansion of the chemical potential in terms of deviation of the charge concentration from the homogeneous distribution is considered. In final calculations the correlation between particles is taken into account in the approximation of the first neighbors. In this approximation the potential and charge concentration distribution is described by a linear differential equation of the fourth order. The results of its analytical solution and subsequent numerical calculations for the characteristics of solid electrolyte are analyzed.

Abstract:This work presents a microscale approach for simulating the dielectrophoresis assembly of polarizable particles under an external electric field. The model is shown to capture interesting dynamical and topological features, such as the formation of chains of particles and their incipient aggregation into hierarchical structures. A quantitative characterization in terms of the number and size of these structures is also discussed. This computational model could represent a viable numerical tool to study the mechanical properties of particle-based hierarchical materials and suggest new strategies for enhancing their design and manufacture. Classical density functional theory and Monte Carlo simulation study of electric double layer in the vicinity of a cylindrical electrode

Abstract:We have performed extensive Monte-Carlo simulations and classical density functional theory (DFT) calculations of the electrical double layer (EDL) near a cylindrical electrode in a primitive model (PM) modified by incorporating interionic dispersion interactions. It is concluded that (i) in general, an unsophisticated use of the mean field (MF) approximation for the interionic dispersion interactions does not distinctly worsen the classical DFT performance, even if the salt ions considered are highly asymmetrical in size (3:1) and charge (5:1), the bulk molar concentration considered is high up to a total bulk ion packing fraction of 0.314, and the surface charge density of up to 0.5 C m−2. (ii) More specifically, considering the possible noises in the simulation, the local volume charge density profiles are the most accurately predicted by the classical DFT in all situations, and the co- and counter-ion singlet distributions are also rather accurately predicted; whereas the mean electrostatic potential profile is relatively less accurately predicted due to an integral amplification of minor inaccuracy of the singlet distributions. (iii) It is found that the layered structure of the co-ion distribution is abnormally possible only if the surface charge density is high enough (for example 0.5 C m−2); moreover, the co-ion valence abnormally influences the peak height of the first counter-ion layer, which decreases with the former. (iv) Even if both the simulation and DFT indicate an insignificant contribution of the interionic dispersion interaction to the above three ‘local’ quantities, it is clearly shown by the classical DFT that the interionic dispersion interaction does significantly influence a ‘global’ quantity like the cylinder surface-aqueous electrolyte interfacial tension, and this may imply the role of the interionic dispersion interaction in explaining the specific Hofmeister effects. We elucidate all of the above observations based on the arguments from the liquid state theory and at the molecular scale.

Abstract:We present a theory to investigate electrokinetic behavior, namely, electrorotation and dielectrophoresis under alternating current (ac) applied fields for a pair of touching inhomogeneous colloidal particles and biological cells. These inhomogeneous particles are treated as graded ones with physically motivated model dielectric and conductivity profiles. The mutual polarization interaction between the particles yields a change in their respective dipole moments, and hence in the ac electrokinetic spectra. The multipolar interactions between polarized particles are accurately captured by the multiple images method. In the point-dipole limit, our theory reproduces the known results. We find that the multipolar interactions as well as the spatial fluctuations inside the particles can affect the ac electrokinetic spectra significantly.
On the low‐frequency dispersion observed in dielectrophoresis spectra

Abstract:The foundation of dielectrophoresis (DEP) as a tool for biological investigation is the use of the Clausius–Mossotti (C–M) factor to model the observed behaviour of cells experiencing DEP across a frequency range. Nevertheless, it is also the case that at lower frequencies, the DEP spectrum deviates from predictions; there exists a rise in DEP polarisability, which varies in frequency and magnitude with different cell types and medium conductivities. In order to evaluate the origin of this effect, we have studied DEP spectra from five cell types (erythrocytes, platelets, neurons, HeLa cancer cells and monocytes) in several conditions including medium conductivity and cell treatment. Our results suggest the effect manifests as a low‐pass dispersion whose cut‐off frequency varies with membrane conductance and capacitance as determined using the DEP spectrum; the effect also varies as a logarithm of medium conductivity and Debye length. These together suggest that the values of membrane capacitance and conductance depend not only on the impedance of the membrane itself, but also of the surrounding double layer. The amplitude of the effect in different cell types compared to the C–M factor was found to correlate with the depolarisation factors for the cells' shapes, suggesting that this ratio may be useful as an indicator of cell shape for DEP modelling.

Abstract:This paper presents the design and optimization of a dielectrophoresis-based cell separator by numerically analyzing the effects of geometrical dimensions on the separation performance and device safety in biological use. The separator uses negative DEP force to gather cells so that all cells have the same initial condition, and employs positive DEP to separate different cells. Simulation results show that the high electric field at the electrode edges could cause cell lysis. Therefore, we propose the maximum electric field as the criterion for measuring the safety of this device. We firstly obtain the optimal ratio of the electrode width to electrode interval by making a trade-off between separation efficiency and cell integrity. This result could be used as a general rule for designing DEP-based separators. The effects of channel height to separation efficiency are also discussed

Abstract:Ionic-based dielectrophoretic microchips have attracted significant attention due to their wide-ranging applications in electro kinetic and biological experiments. In this work, a numerical method is used to simulate the dynamic behaviors of ionic droplets in a microchannel under the effect of dielectrophoresis. When a discrete liquid dielectric is encompassed within a continuous fluid dielectric placed in an electric field, an electric force is produced due to the dielectrophoresis effect. If either or both of the fluids are ionic liquids, the magnitude and even the direction of the force will be changed because the net ionic charge induced by an electric field can affect the polarization degree of the dielectrics. However, using a dielectrophoresis model, assuming ideal dielectrics, results in significant errors. To avoid the inaccuracy caused by the model, this work incorporates the electrode kinetic equation and defines a relationship between the polarization charge and the net ionic charge. According to the simulation conditions presented herein, the electric force obtained in this work has an error exceeding 70% of the actual value if the false effect of net ionic charge is not accounted for, which would result in significant issues in the design and optimization of experimental parameters. Therefore, there is a clear motivation for developing a model adapted to ionic liquids to provide precise control for the dielectrophoresis of multi-phase ionic liquids.

Abstract:This paper builds on two previous works, Lindgren et al. J. Comp. Phys. 371, 712-731 (2018) and Quan et al. <a class="link-https" data-arxiv-id="1807.05384" href="https://arxiv.org/abs/1807.05384">arXiv:1807.05384</a> (2018), to devise a new method to solve the problem of calculating electrostatic interactions in a system composed by many dielectric particles, embedded in a homogeneous dielectric medium, which in turn can also be permeated by charge carriers. The system is defined by the charge, size, position and dielectric constant of each particle, as well as the dielectric constant and Debye length of the medium. The effects of taking into account the dielectric nature of the particles is explored in selected scenarios where the presence of electrolytes in the medium can significantly influence the total undergoing interactions. Description of the mutual interactions between all particles in the system as being truly of many-body nature reveals how such effects can effectively influence the magnitudes and even directions of the resulting forces, especially those acting on particles that have a null net charge. Particular attention is given to a situation that can be related to colloidal particles in an electrolyte solution, where it's shown that polarization effects alone can substantially raise or lower---depending on the dielectric contrast between the particles and the medium---the energy barrier that divides particle coagulation and flocculation regions, when an interplay between electrostatic and additional van der Waals forces is considered. Overall, the results suggest that for an accurate description of the type of system in question, it is essential to consider particle polarization if the separation between the interacting particles are comparable to or smaller than the Debye length of the medium.

Anomalous, Dielectrophoretic Transport of Molecules in Non-Electrolytes

Abstract:The electric field dielectric polarization-based separations mechanism represents a novel method for separating solutions at small length scales. An electric field gradient with a maximum strength of $\mathrm{0.4~MV/m}$ applied across a $\mathrm{10~\mu m}$ deep channel is shown to increase the concentration inside the low electric field region by $\approx \mathrm{40}\%$ relative to the high electric field region. This concentration change is two orders of magnitude higher than the estimated change predicted using the classical equilibrium thermodynamics for the same electric field. The deviation between the predicted and the experimental results suggests that the change in volumetric electric field energy with solute concentration is insufficient to explain this phenomenon. The study also explores the effect of varying strength of electric field and frequency of supplied voltage on the dielectric polarization-based separation efficiency. While the increase in the former increases the separation efficiency, the increase in the latter reduces the degree of concentration change due to ineffective charging of the electrodes.

Abstract:A theoretical study of the dielectrophoretic (DEP) spectrum of a pair of touching colloidal particles or biological cells in a host fluid under the application of a nonuniform alternating current (ac) electric field is presented. The main objective of this work is to investigate the correlation effects due to the presence of mutual polarization on the DEP spectrum. In particular, we employ the multiple image method to account for the effect of multiple images, and obtain an analytic expression for the DEP force. It is found that, at low frequencies, the DEP force can be enhanced (reduced) significantly for the longitudinal (transverse) field case due to the presence of multiple images. The numerical results can be well understood in the spectral representation theory. To first order in the dipole interaction, our results reduce to the established formula derived by an alternative method. Our approach demonstrates the importance of the correlation effects in ac electrokinetic phenomena of colloidal suspensions.

Abstract:We use theory, simulation, and experiment to quantify the dielectrophoretic force produced on spherical colloidal particles exposed to an interdigitated electrode array in a microfluidic environment. Our analytical predictions are based on a simplified two-dimensional model and agree with numerical solutions based on a finite difference scheme. Theoretical predictions align with experimental results without any fitting parameters over a wide range of frequencies and applied voltages. A frequency–response function for negative-dielectrophoresis instruments is derived by developing an equivalent electrical circuit model to understand the system power requirements better. Finally, we investigate the role of electrode width and array spacing on the magnitude and distance dependence of the negative dielectrophoresis force. Our analyses show that the electrode width controls the lateral position of the particles, whereas the voltage controls the vertical position. The strongest forces are achieved when the array spacing is matched to the particle size and when the electrode width is ~1/3 of the array spacing. These results can improve the design and optimization of negative-dielectrophoresis microfluidic instruments for applications in the separation and purification of colloidal microparticles in microelectromechanical systems.

Abstract:Determining the physical and chemical properties of biologically important particles such as cells, organelles, viruses, exosomes, complexes, nucleotides, and proteins is needed to understand their function. These properties are determined with common analytical tools (mass spectrometry, cryo-EM, NMR, various spectroscopies, nucleotide sequencing, etc.) whose function can be improved when samples are pure and concentrated. Separations science plays a central role in conditioning samples, ranging from low-resolution benchtop operations like precipitations or extractions to higher-resolution chromatography and electrophoresis. In the last two decades, gradient insulator-based dielectrophoresis (g-iDEP) has emerged as a high-resolution separation technique capable of highly selective enrichment of cells, viruses, exosomes, and proteins. Specific evidence has been shown that pure homogeneous and concentrated fractions of cells and exosomes can be generated from complex mixtures. However, recovering those fractions for analysis has not been developed, limiting the technique to an analytical rather than a preparative one. Here, a finite element analysis was undertaken to identify geometries and operational parameters to efficiently remove the enriched fraction while retaining maximum concentration and providing total mass transfer. Geometric factors (e.g., side channel width and distance from the gradient-inducing gap) were studied, along with the addition of a second inlet side channel. Two flow-generating mechanisms-electroosmosis and hydrostatic pressure-were evaluated for semi-optimized device designs, including a comparison of the one- and two-inlet designs. Simulations indicate effectively one hundred percent mass transfer and a concentration increase by an order of magnitude for several device configurations and operational parameters.
Many-body Effects in Anisotropic Dielectric Colloids: Polarization, Electrostatic Interactions, and Van der Waals Forces

Abstract:We study the polarizability of colloidal particles, as well as their interparticle interactions in the presence or absence of an external electric field. To this end, we employ the Coupled Dipole Method (CDM), which models the particles as clusters of Lorentz atoms (i.e., inducible dipoles) and uses a Hamiltonian approach and large-matrix manipulation to account for all many-atom effects. In this way, we are able to numerically and, in some cases, analytically calculate London-Van der Waals forces, polarizability, orientational energy in an external electric field, and external electric field-induced interparticle interactions. We investigate the polarizability of cuboids, bowls and dumbbells and study their alignment properties in an external electric field. We also investigate the effect of the size and shape of the particles on their tendency to align. For spheres and cubes in an external electric field we study the local polarization in detail and compare the results from the CDM with those from continuum electrostatics to investigate the equivalence of the two approaches. We also study the pair interaction between various particles in an external electric field, taking into account the influence the particles have on each other's polarization and compare the CDM results with those given by techniques that model the particles as built up from permanent dipoles as opposed to induced ones. Using the CDM, we moreover study the London-Van der Waals interaction between various particle shapes in the absence of an external electric field, and whether many-body effects can account for experimentally observed spontaneous chain formation. Lastly, we study the density of states in a string of Lorentz atoms, the evolution of modes on a pair of squares of Lorentz atoms as a function of the gap between the squares, and the interaction of a rod with a conducting plate under the influence of an external electric field.

A 3-D Microelectrode System for Dielectrophoretic Manipulation of Microparticles

Abstract:This paper presents a microfluidic system for manipulation and separation of micron-sized particles based on the combined use of negative dielectrophoresis (DEP) and hydrodynamic forces. A 3-D microelectrode structure (so called paired electrode array) are constructed face to face on the top and bottom sides of the microchannel and driven with high-frequency AC voltage to generate dielectrophoretic gates. Depending on the relative strengths of the two forces, particles such as polystyrene beads or cells carrying by a laminar flow can either penetrate the gate or settle there. This gives rise to certain applications including selectively concentrating particles from the flow, separating particles depending on their sizes or dielectric properties, and automatically positioning particles to selective locations. For this purpose, a microfluidic device consisting of the paired electrode array sitting on the channel has been fabricated using microfabrication techniques. Polystyrene beads were used to study the performance of the device. Experimental results including the concentration and separation of particles are presented.

Abstract:The dielectric dispersion, dielectrophoretic (DEP) and electrorotational (ER) spectra of spheroidal biological cell suspensions with an intrinsic dispersion in the constituent dielectric constants are investigated. By means of the spectral representation method, we express analytically the characteristic frequencies and dispersion strengths both for the effective dielectric constant and the Clausius-Mossotti factor (CMF). We identify four and six characteristic frequencies for the effective dielectric spectra and CMF respectively, all of them being dependent on the depolarization factor (or the cell shape). The analytical results allow us to examine the effects of the cell shape, the dispersion strength and the intrinsic frequency on the dielectric dispersion, DEP and ER spectra. Furthermore, we include the local-field effects due to the mutual interactions between cells in a dense suspension, and study the dependence of co-field or anti-field dispersion peaks on the volume fractions.
