Abstract:New mononuclear copper(II), nickel(II) and cobalt(II) hydrazone complexes of 2-cyano-N′-((4-oxo-4H-chromen-3-yl)methylene)acetohydrazide have been synthesized. Structure identification of the prepared complexes was carried out by physicochemical and spectroscopic techniques including elemental analyses, infrared, electronic, mass and electron spin resonance spectra, thermal analysis, conductivity and magnetic susceptibility measurements, powder x-ray diffraction and transmission electron microscope. The isolated binary complexes are neutral mononuclear complexes (with 1:1 and 1:2; M:L stoichiometries) and ternary complexes (with 1:1:1; M:L:L′ stoichiometry). The analytical and spectroscopic techniques illustrated the monoanionic tridentate behavior of the hydrazone ligand with γ-pyrone oxygen, enolic oxygen and azomethine nitrogen coordinating sites. The metal complexes displayed square planar, tetrahedral and octahedral geometries. Density Functional Theory calculations were carried out for the chelating agent and its copper(II) chelates. The phenoxazinone synthase activity of the binary copper(II) complexes was studied and discussed. The antimicrobial activity of the hydrazone ligand and its complexes was evaluated toward some sorts of Gram–positive bacteria, Gram–negative bacteria, yeast and fungus.

Abstract:A set of seventeen metal complexes with 1-(5-acetyl-2,4-dihydroxyphenyl)ethylidene) hydrazono)ethyl)-2,4-dihydroxyphenyl)ethylidene)hydrazono)-3-(hydroxyimino)pentan-2-one have been elaborated. The characterization of the synthesized hydrazonic chelator and its complexes have been carried out by physical, analytical and spectral techniques such as 1 H-, 13 C-NMR, FT-IR, UV–Vis mass and ESR spectra, in addition to elemental, thermal, conductivity and magnetic moments measurements. It was found that the hydrazonic chelator attached with the central ions via ON donors as a mononegative or neutral bidentate fashion in complexes (2-17) bonded with the central ions through the nitrogen of protonated/deprotonated oximato and azomethine groups. All metal complexes are nonelectrolytic in nature and have various geometries. The ESR showed that the complexes of copper(II) have a d 9 configuration with an axial symmetry type of a d (x2-y2) ground state with significant covalency features. The TG study verified that the complexes had been disintegrated in two, three or four phases beginning with the libration of crystallized, coordinated water molecules or anions exclusion and was finished with the full decomposition of the complexes, leaving the oxide of the metal. The microbicide screening data showed that the prepared compounds have a greater fungicide activity against A. flavus than S. cerevisiae , while all compounds have low activity against bacteria E. coli , and B. subitilis.

“Streamers” in liquids: Relation with practical high voltage insulation and testing of liquids

Abstract:This paper discuss the relations between laboratory studies of breakdown precursors in liquids (usually called "streamers"), and practical applications such as high voltage insulation of power transformers and testing of liquids. The conditions existing within high voltage power transformers (ac voltage, moderately divergent electric field, presence of insulating solids, particles in the oil, etc.) are rather different from those frequently used in the laboratory to study streamers (very divergent field -point-plane geometry-, and impulse voltage). Parameters such as field geometry, voltage waveform, liquid conditioning, can influence either streamer inception or propagation. After a short presentation of streamer phenomena in liquids, the most important effects of these parameters are recalled in this paper. Many features previously observed empirically by the measurement of breakdown voltage can be explained by the behaviour of streamers in conditions close to those actually present in transformers. It remains rather difficult today to model and predict streamer properties (initiation fields, propagation length, velocity, ...) when the liquid nature is changed. The large variety of physical processes involved during initiation and propagation, and the non-linearity of many processes make the study of breakdown in liquids complex. However, knowledge about streamers is helpful to define and measure parameters of practical importance (initiation and propagation conditions), providing a more scientific basis to test and compare the properties of liquids.
Synthesis, molecular docking and computational studies of novel hydrazone complexes

Abstract:In this study, a hydrazone ligand complexes based on p-anisidine and acetylacetone derivatives with transition metals like Ni, Cu, Fe, and Zr were prepared. The structure of the prepared compounds was confirmed using Nuclear magnetic resonance (1H-NMR and 13C-NMR), Distortionless enhancement by polarization transfer (DEPT), heteronuclear single quantum coherence (HSQC-NMR), Correlated Spectroscopy (COSY-NMR), Fourier-transform infrared spectroscopy (FT-IR) and Electrospray ionization mass spectrometry (ESI-MS). The x-ray single crystal of the ligand was studied and the results showed that an intramolecular N – H … .O = C hydrogen bond (1.87Å) exists that stabilizes the geometrical structure of the ligand. Also, the magnetic properties for the prepared complexes were studied using Gouy’s method of susceptibility measurement. In addition, there in silico docking and In vitro antibacterial activities were investigated and the results showed that Fe complex has the highest antimicrobial activity. The expected structures and conformers for the prepared ligands and complexes were examined and fully optimized using the level B3LYP/6-31G*.

Abstract:Four dioxidomolybdenum(VI) complexes, [MoO 2 (L)] 2 ·H 2 O ( 1 ), [MoO 2 (L)(py)] ( 2 ), [MoO 2 (L)(3-pic)]·H 2 O ( 3 ) and [MoO 2 (L)(4-pic)] ( 4 ) of a dibasic tridentate ONO donor aroylhydrazone, H 2 L (where H 2 L = 3-methoxy-2-hydroxybenzaldehyde-2-furoic acid hydrazone) have been synthesized and well characterized. The stoichiometric reaction of aroylhydrazone with MoO 2 (acac) 2 in methanolic medium yielded phenoxobridged binuclear complex 1 whereas monomeric complexes, 2 , 3 and 4 were formed as a result of incorporating different monodentate heterocyclic bases. The tridentate aroylhydrazone coordinates to the MoO 2 2+ core through phenolate oxygen, azomethine nitrogen and iminolate oxygen atoms. Single crystal XRD studies established the coordination geometry of mononuclear dioxidomolybdenum(VI) complexes as distorted octahedron. The crystal structures and various solid state interactions were also investigated here. DFT investigations were conducted to explore the reactivity parameters of the studied aroylhydrazone and complexes. Furthermore, molecular docking analyses unveiled the superior putative binding energy of the investigated compounds in contrast to cisplatin. These findings were particularly pronounced in relation to the selected target proteins, which are indicative of their potential efficacy against lung and breast cancer cell lines.

Abstract:A new chelating ligand, 2-(2-(5-tert-butylisoxazol-3-yl)hydrazono)-N-(2,4-dimethylphenyl)-3-oxobutanamide (HL), and its four binuclear transition metal complexes, M(2)(L)(2) (micro-OCH(3))(2) [M=Ni(II), Co(II), Cu(II), Zn(II)], were synthesized using the procedure of diazotization, coupling and metallization. Their structures were postulated based on elemental analysis, (1)H NMR, MALDI-MS, FT-IR spectra and UV-vis electronic absorption spectra. Smooth films of these complexes on K9 glass substrates were prepared using the spin-coating method and their absorption properties were evaluated. The thermal properties of the metal(II) complexes were investigated by thermogravimetry (TG) and differential scanning calorimetry (DSC). Different thermodynamic and kinetic parameters namely activation energy (E*), enthalpy of activation (DeltaH*), entropy of activation (DeltaS*) and free energy change of activation (DeltaG*) are calculated using Coats-Redfern (CR) equation.

Abstract:Different metal complexes through reaction of sodium 4‐hydroxy‐3‐((2‐nicotinoylhydrazineylidene)methyl) benzenesulfonate with Fe(III), Co(II), and Mn(II) chloride in a 1:1 ratio in excellent yield were synthesized and characterized by spectral analysis investigation. The redox behavior of Co(II) and Mn(II) ions was studied by cyclic voltammetry. Furthermore, antioxidant, anticancer, antimicrobial activities, and docking simulation are also investigated. Photoactivity of the ligand was studied in presence and absence of Co(II) and Mn(II) ions. A water‐soluble hydrazone ligand bearing 2‐nicotinoylhydrazineylidene moiety, sodium 4‐hydroxy‐3‐([2‐nicotinoylhydrazineylidene]methyl) benzenesulfonate (NaH2NH), and its complexes with Co(II), Fe(III), and Mn(II) have been synthesized and characterized. The structure of the ligand and its complexes were analyzed using various spectroscopic techniques such as FT‐IR, 1H‐ and 13C‐NMR, EI‐mass, powder X‐ray diffraction, UV–visible spectra, and magnetic measurements. The infrared spectra indicated that the ligand behaves as a negatively charged bidentate ligand (ON) through the deprotonation of the enolic (OH) group and the azomethine (C=N) group in the [Co (NaHNH)2(H2O)4Cl2].3H2O complex. In the [Fe (NaNH)2(H2O)Cl].2H2O complex, the ligand behaves as bi‐negative tetradentate (O2N2) through the protonation of the enolic oxygen and the nitrogen atom of the azomethine group in two nicotinic hydrazone molecules. Another coordination type was observed for [Mn (NaNH)(H2O)3] complex in which NaH2NH acts as bi‐negative tridentate ONO through the nitrogen atom of the azomethine group, the protonated of enolic and phenolic groups. The fluorescence spectrum of the NaH2NH was found to be quenched upon addition of these Co(II) and Mn(II) ions. The antimicrobial results indicated that the Mn(II) complex exhibited the highest antibacterial potency against all targeted organisms. The results of antioxidants demonstrated that the complexes exhibited higher activity compared to the free ligand, indicating an enhancement of antioxidant activity through complexation. The tested substances established strong nuclease‐like activity at a high concentration of 7.5 mM, leading to the complete degradation of DNA as observed in agarose gel electrophoresis. This suggests that these compounds have the potential to serve as antitumor agents in vivo by inhibiting DNA replication in cancer cells and preventing tumor growth. The interaction of the isolated compounds with 1c14 and 6vj3 targets was discussed using molecular docking.

Abstract:The binuclear Pd(II) and mononuclear Zn(II), Ru(III) and Pt(IV) chelates were synthesized based on new hydrazone ligand derived from 2-(phenylamino)acetohydrazide and 3-Acetyl-2-hydroxy-6-methyl-4 H -pyran-4-one (dehydroacetic acid DHA) (H 2 L). Variable characterization tools were used to confirm the exact structures. Spectral data show that (H 2 L) ligand acts as monobasic tridentate with ONO donor form. UV–Vis absorption spectroscopy and magnetic moment measurements demonstrated that the Ru(III) and Pt(IV) chelates possess an octahedral environment while Zn(II) and Pd(II) chelates possess square planar environment. The geometry of each complex was optimized using DFT calculation. The stability of the compounds was elucidated by the TGA technique. Antimicrobial activity of the compounds was evaluated and the results showed that the complexes demonstrated superior inhibitory action to ligand and display excellent antimicrobial activities against the tested microorganisms. Zn(II) mononuclear chelate showed the greatest effect against both types of bacterial strains. Antiviral activity was evaluated against Coxsackie virus (Cox B4), Herpes simplex (HSV1) and Hepatitis A Virus (HAV). The binuclear Pd(II) chelate revealed high activity against the three studied viruses while the mononuclear Ru(III) chelate revealed similar activity against the two types only. The cytotoxicity results supported the greatest effect of Zn(II) and Pd(II) against MCF-7 type. Zn(II) chelate revealed the best result against Hep-G2 type. Finally, we can conclude that binuclear Pd(II) chelate was active against all studied disease types, while not all mononuclear chelates are active. The microbial and tumor inhibition activity was compared with the theoretical molecular docking results with the specification of the active species in the selected proteins.

Synthesis, Characterization and Biological Studies of Some Transition Metal Complexes with Pyrazine Schiff Base Hydra zone Ligand

Abstract:New pyrazine carbohydra zone ligand N'-(1-(5-chloro-2-hydroxyphenyl) ethylidene) pyrazine-2-carbohydrazide (H2L), prepared by the condensation of equimolar amounts of pyrazine-2-carbohydrazide with 2-hydroxy-5-chloroacetophenone in methanol, reacts with suitable metal salt precursors to give complexes of two general formulae: [M(HL)(Cl)(H2O)2] {M = Mn(II), Co(II), Ni(II) and Cu(II)} and [M(L)(H2O)] {M = Zn(II) and Cd(II)}. Structure of ligand was confirmed by elemental analysis, IR, 1H and 13C NMR and mass spectroscopy, while synthesized complexes were additionally characterized by magnetic susceptibility measurements, molar conductivity measurements, XRD, ESR (for Cu(II)), SEM and thermogravimetric analysis. Spectroscopic studies confirmed a tridentate ONO donor behavior of the ligand towards the central metal ion. The molar conductance (12–17 W–1 cm2 mol–1) measurements in DMSO indicated non-electrolytic nature. Thermal behavior of the complexes suggests their extended stability and the thermal decomposition generally proceeds via partial loss of the organic moiety and ends with the formation of respective metal oxide as a final product. Various kinetic and thermodynamic parameters were evaluated using the Coats-Redfern method. The solid-state electrical conductivity of the complexes measured in the temperature range 303-463 K suggested their semiconducting behavior. The ligand and its metal complexes were screened in vitro for their antibacterial activity against the Gram-positive bacteria S. aureus and B. subtilis, the Gram-negative bacteria E. coli and S. typhi and the fungi C. albicans and A. niger. The obtained results indicated improved activity of the complexes compared to the free ligand against all studied bacterial and fungal species.

Abstract:Spectral study, DFT and some biological applications of HL and its complexes. A novel Schiff base ligand, ((4‐oxo‐4H‐chromen‐3‐yl)methylene)isonicotinohydrazone (HL), in addition to its VO(II), Cd(II), Mn(II), and Pd(II) complexes, were prepared and identified. Various analytical techniques such as elemental, TGA, and magnetic susceptibility measurements, additionally, nuclear magnetic resonance (NMR) spectroscopy of hydrogen and carbon‐13, MS, FT‐IR, EPR, and electronic (UV‐VIS) spectroscopy, are utilized and proposed that HL has a mono‐negative tridentate mode of chelation with VO(II) and Mn(II) complexes, whereas it displays a mono‐negative bidentate with the Cd(II) complex and a neutral bidentate with the Pd(II) chelate. The crystallographic structures and atomic spacing for both HL and its complexes were examined through powder X‐ray diffraction analysis. Moreover, electrochemical analyses were conducted on Cd(II) and Mn(II) metal ions with and without the agent that chelates for the purpose of foreseeing the consequences of complex interactions in the case of the liquid phase. Furthermore, photoluminescence study was applied on VO(II) and Pd(II) complexes and showed a quenching effect for both. The ligand's geometry and its complexes were examined through calculations based on density functional theory (DFT) using B3LYP/LANL2DZ computational method and showed molecular electrostatic potential, chemical reactivity, and FMOs. The Cd(II) complex showed promising results against various bacterial strains, displaying excellent activity that exceeded that of the standard, Gentamicin (108% against K. pneumoniae, 100% against Salmonella, and 90.47% against A. fungi). In contrast, it exhibited no cytotoxicity toward the MCF‐7 cell line. However, in a DNA binding study, the compound demonstrated significant activity as its concentration increased up to 6 mM, suggesting its potential for use in cancer therapy due to its ability to inhibit DNA replication.

Abstract:An aroylhydrazone compound (HL) was prepared and explored as a ligand (HL = N' -(2-hydroxy-4-methoxybenzylidene)-3-hydroxy-4-methoxybenzohydrazide). Reaction of the aroylhydrazone ligand with copper bromide and copper chloride afforded two new copper(II) complexes, [Cu 2 Br 2 L 2 ] ( 1 ) and [CuL(DMF)]Cl ( 2 ), respectively. The aroylhydrazone ligand and copper complexes were characterized by CHN elemental analyses and infrared spectroscopy. Structures were further confirmed by single crystal X-ray diffraction experiments. The aroylhydrazone compound coordinates to Cu atoms through its phenolate oxygen, imino nitrogen and carbonyl oxygen. The Cu atom in 1 adopts a square pyramidal coordination, with three donor atoms of one L and one phenolate oxygen of a second L in the basal plane, and with one Br ligand at the apical position. The Cu atom in 2 is square pyramidal, with three donor atoms of one L and one oxygen of a DMF ligand in the basal plane, and with one Cl ligand at the apical position. Crystal structures of the aroylhydrazone and the copper complexes are stabilized by hydrogen bonds. The complexes possess antibacterial activities against B. subtilis , S. aureus and E. coli .

Abstract:The present study deals with design and synthesis of new mononuclear Cu(II), Co(II), Ni(II) and Zn(II) metal complexes of hydrazone of 2-(2-(2,4-dihydroxybenzylidene)hydrazinecarbonyl)pyridine-1-oxide (H 2 L), in a view to study their potential relevance for the biological applications. Structural aspects of the title compound (H 2 L) and metal complexes synthesized were evaluated by spectral and analytical methods viz. 1 H NMR, 13 C NMR, LC-MS, FT-IR, UV–Visible, SEM, EDX, Powder XRD, ESR, TGA and DTA. Ligational properties of H 2 L were explored by employing pH metric equilibrium studies for recognizing metal ion binding potential donor sites, and further HyperChem 7.5 software for computing properties of frontier molecular orbitals to ascertain orientation of highest occupied molecular orbitals from which presumably electrons are donated to metal ion in complex formation. The affinity of all title compounds to bind with CT-DNA and the type of interaction, therein have been studied by conducting UV–VIS absorption and fluorescence emission titrations. The protein-binding ability of all metal complexes with two serum proteins (BSA and HSA) were explored by absorption titrations, and further fluorescence titrations for BSA binding were also performed. Antioxidant activity of the title compounds was carried out by the method of free radical scavenging using spectrophotometric technique. Additionally, cytotoxicity of all metal complexes and ligand was measured by CTG cell proliferation assay after treating them with MDA-MB-231 and SKOV-3 cell lines. The cell cycle arrest and apoptosis assay in above cell lines after treatment with title compounds were also investigated by flow cytometry. In addition, molecular docking studies at target CDK2 protein inferred binding affinity of the title compounds through non covalent bonding interactions.

Abstract:In the search of potential multi-target medicinal agents, new NOO-tridentate hydrazone ligands (H 2 L 1 –H 2 L 4 ) ( 1 – 4 ) and their transition metal(II) complexes ( 5 – 20 ) were synthesized from salicylaldehyde derivatives and xanthene-9-carboxylic acid hydrazide. The synthesized compounds were well characterized by different techniques, such as FT-IR, ( 1 H, 13 C) NMR, mass spectrometry, UV–Vis, ESR, TG–DTA, powder-XRD, SEM-EDAX and molar conductivity measurements. The spectral techniques confirmed the tridentate nature of hydrazones and coordination of ligands with metal ion via N azomethine , O phenolic and O enolic (NOO) which suggested octahedral stereochemistry of the complexes. The complexes were obtained in good yield, non-electrolytic in nature and stable up to 150 °C. The compounds ( 1 – 20 ) were screened for in vitro anti-malarial activity against Plasmodium falciparum 3D7 strain and the Co(II) complex ( 5 ) (IC 50 = 0.94 ± 0.03 nM) exhibited comparable potency to quinine (IC 50 = 0.826 ± 0.02 nM), whereas, Cu(II) complex ( 19 ) (IC 50 = 0.65 ± 0.05 nM) have excellent potency to control malarial infection. Furthermore, the in vitro antimicrobial activity against six strains revealed that the compounds 5 and 19 demonstrated greater efficacy to inhibit the growth of microbial strains among the tested compounds ( 1 – 20 ). The most potent anti-malarial compounds ( 1 , 5 , 19 ) were examined by molecular docking study against Dihydroorotate dehydrogenase protein receptor (PDB ID: 1TV5) to accomplish the acquired biological results through binding energy and interaction mode.

Abstract:An appropriate conventional method has been utilised to synthesise new tridentate Schiff base ligand 4-chloro-2-((2,4-dihydroxybenzylidene)amino)benzoic acid with Co (II), Ni (II), Cu (II), and Zn (II) complexes. Several spectroscopic(FT-IR, 1 H NMR, 13 C NMR, UV–Vis, Mass), TGA-DTA, powder X-ray diffraction, and DFT calculations, were used to characterise these complexes.According to these spectroscopic investigations, the ligand coordinated with the metals via carboxylic oxygen, azomethine nitrogen, and hydroxyl oxygen, acting as a tridentate ligand. Appropriate geometry for every complex has been proposed based on spectroscopic and analytical data. In tetrahedral geometry with Co(II), Ni(II), Zn(II), and square planner with Cu(II) ion, the ligand functions as a tridentate via ONO donors.The optimal structure (bond length and bond angle) and chemical reactivity of the ligand and its metal complexes from their Frontier Molecular Orbitals (FMO) and Molecular electrostatic potential (MESP) calculations have been investigated using Density Functional Theory (DFT/B3LYP,6-31G**/6-311G**(d,p), and LanL2DZ basis sets) calculations. In vitro DPPH radical scavenging techniques were used to assess the antioxidant activity of the ligand and its metal complexes. The results indicated that all of the complexes exhibited good antioxidant activity, with Cu (II) complex being the most potent with the lowest IC 50 values.The ligand and complexes' in vitro antibacterial activities were further explored by screening them against Gram(+ve) bacteria (Bacillus subtilis) and Gram(−ve) bacteria (E. coli) . Significant activity was demonstrated by all synthesised compounds against the tested strains of bacteria. The antifungal activity of complexes against Aspergillus niger and Candida albicans was determined. The results indicated that Cu (II) complex exhibited good antifungal activity against Candida albicans , but not against Aspergillus niger . The fungal strain Aspergillus niger was not significantly inhibited by complexes. The complexes have greater antimicrobial activity than the Schiff base ligand, as indicated by the MIC values.

Abstract:Solid complexes (1–4) of Schiff base ((o‐tolylimino)methyl)phenol (HL) with manganese(II), cobalt(II), nickel(II), and copper(II) were synthesized in methanol under reflux. Schiff base (HL) was synthesized through condensation reaction of 2‐hydroxybenzaldehyde with o‐toluidine. Ligand (HL) and synthesized complexes (1–4) were characterized by using different spectroscopic techniques (UV–visible, FTIR, 1H NMR, mass spectrometry, powder X‐ray diffractometer [XRD]), magnetic measurements, and thermal gravimetric analysis. Spectroscopic analysis demonstrated that ligand being bidentate, in all the complexes, coordinates with metals through the N atom of azomethine group and O atom of the hydroxyl group. Molecular ion peaks (m/z) appear in the mass spectra of complexes confirm the proposed stoichiometry. TGA data of complexes 1 and 2 exhibited water molecules coordinated with the central metal ions (Mn2+ and Co2+). Magnetic Susceptibility analysis of metal complexes proposed octahedral geometry for Mn(II) and Co(II) complexes. Conductivity analysis was in good agreement with non‐electrolyte nature of metal complexes. Global reactivity data exhibited that metal(II) complexes are soft as compared to HL. Moreover, frontier molecular orbital (FMO) findings revealed that among the metal complexes, [Mn(L)2(H2O)2] 1 was having large energy gap showing greater stability and less reactivity, while [Ni(L)2] 3 was found most reactive among all of them. Values of chemical reactivity descriptors obtained denoted that synthesized metal complexes can be proved as efficient biological candidates owing to their reactivity patterns. In vitro antibacterial and antifungal activity of complexes and ligand were evaluated against different strains of bacteria (Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, and Bacillus subtilis) and fungi (Candida albicans, Aspergillus niger, Trichoderma harzianum, and Aspergillus flavus) via disc diffusion method. The bacterial and fungal growth more efficiently inhibited by the use of metal complexes (1–4) than ((o‐tolylimino)methyl)phenol (HL).

Abstract:The tridentate Schiff base ligand was prepared via condensation of 4,6-diacetylresorcinol and 1,8-naphthalenediamine in 1:1 molar ratio using conventional method. Additionally, a new series of transition metal complexes of Cr(III), Mn(II), Fe(III), Co(II), Ni(II), Cu(II), Zn(II) and Cd(II) metal ions were prepared and fully characterized by elemental analysis, FT-IR, 1H NMR, UV–vis, conductivity and magnetic susceptibility measurements and theoretically through density function theory (DFT). Spectral studies suggested that, the Schiff base coordinated to the metal ions through the azomethine N, N-amine and phenolic oxygen atoms. From the elemental, magnetic susceptibility and spectroscopic data, the complexes were suggested to have octahedral geometry. The Schiff base and its metal complexes were screened for their in vitro antimicrobial activities and showed that Cd(II), Cu(II) and Cr(III) complexes have high activity against Escherichia coli, while Ni(II) and Cd(II) complexes have the highest activity against Bacillus Subtilis, Co(II) complex has the highest activity against Staphylococcus aureus and Cd(II) complex has the highest activity against Pseudomonas aeruginosa. In addition, the molecular docking study was performed to explore the possible ways for binding to crystal structure of Staphylococcus aureus nucleoside diphosphate kinase complexed with ADP (PDB ID: 3Q8U).

Abstract:Unsymmetrical Schiff base hydrazone complexes of cobalt(II), nickel(II) and zinc(II) were synthesized from oxalic dihydrazide, salicylaldehyde and ethyl acetoacetate. The ligand and its metal(II) complexes were characterized by elemental analysis, UV-visible, FT-IR, molar conductivity. To determine the biological activity, in vivo antipyretic as well as in vitro antibacterial activities were conducted along with molecular docking simulation, HOMO-LUMO and MEP calculations. The findings from the experimental and molecular docking investigations demonstrated that the ligand and all the metal(II) complexes has an ability to interact with E. coli 24kDa domain protein (PDB: 1KZN). Furthermore, these interactions were accompanied by favourable docking score values, indicating potential therapeutic implications. The quantum parameters and geometric optimization of the ligand and complexes were performed using DFT simulations.

Abstract:The diorganotin(IV) complexes ( 5–20 ) were synthesized in the present research from 4-fluorophenoxyacetic hydrazide and salicylaldehyde derivatives-based hydrazone ligands (1–4) to get an effective biological agent to combat microbial and oxidant deformities. Numerous spectral techniques such as ( 1 H, 13 C, 119 Sn) NMR, UV–Vis, IR, and mass spectrometry were executed to illuminate the composition of complexes. These techniques ascertained tridentate chelation of hydrazone ligands with tin metal through enolic, phenolic oxygens and imine nitrogen, revealing pentacoordinated geometry of the complexes. The single crystal XRD of complex ( 5) confirmed distorted trigonal bipyramidal geometry. The TGA studies showed thermal stability up to 180 °C of the complexes, whereas the low conductance observed pointed to the non-electrolytic nature of the compounds. Furthermore, serial dilution assay was implemented to uncover the microbial inhibition efficacy (against six strains) of the compounds using ciprofloxacin and fluconazole. Among the synthesized compounds, ( 1, 8 ) exhibited comparable MIC value to standard. The compound (8) was reported as four times more potent than the fluconazole against C. albicans. Using DPPH assay, the antioxidant efficiency was examined which advocates enhanced efficacy of complexes than the ligands. The potency of complex (8) against C. albicans makes it a point of interest for molecular docking investigation, so, complex (8) and its ligand (1) were studied against protein of C. albicans (5TZ1), revealing the more efficacy of complex (binding energy-11.6 kcal/mol) than ligand Further, the compounds were analysed for ADME prediction which concluded the efficacy of compounds as orally efficient pharmaceuticals.

Abstract:Four mixed-ligand oxidovanadium(IV) complexes, [V IV O(L 1-4 )(phen)], 1-4 , incorporating tridentate dibasic ONO donor hydrazone ligands (H 2 L 1-4 , general abbreviation H 2 L) derived from the condensation of 2-picolinyl hydrazide with 2-hydroxy acetophenone and its 5-substituted derivatives as primary ligands and 1,10-phenanthroline (phen) as auxiliary ligand have been synthesized in excellent yields. The complexes were characterized by elemental analyses, magnetic susceptibility measurements, and various spectral ( e.g. IR, UV–Vis and EPR) studies. The molecular structure of one of the four complexes has been determined by single-crystal X-ray diffractometry. In 1 , the hydrazone ligand is meridionally disposed of, coordinating through one enolic-O, one phenolic-O, and one imine-N. The other basal position is occupied by one pyridine-N of the phen moiety. The axial positions are occupied by one oxido-O atom and the other pyridine-N atom of the phen moiety. The complexes are paramagnetic with one unpaired electron ( μ eff ≈ 1.73 BM), suggesting no interaction with the neighboring molecules and are EPR active. The complexes display two d-d transitions, one near 725 and the other near 840 nm due to dxy→dxz,dyz and dxy→dx2−y2 transitions, respectively. The other d-d transition associated with dxy→dz2 transition was not observed as it falls in the UV region, which is masked by highly intense intra-ligand transitions. The complexes display one-electron oxidation peaks in the +0.75 to +0.89 V vs. Ag/AgCl region in the CH 3 CN solution that show a linear relationship with the Hammett parameter (σ) of the substituents in the aryloxy ring.
