Abstract:Recent magnetic resonance (MR)-based electrical impedance tomography (MREIT) of in vivo animal and human subjects enabled the imaging of electromagnetic properties, such as conductivity and permittivity, on tissue structure and function with a few millimeter pixel size. At those resolutions, the conductivity contrast might be sufficient to distinguish different tissue type for certain applications. Since the precise measurement of electrical conductivity under the tissue levels can provide alternative information in a wide range of biomedical applications, it is necessary to develop high-resolution MREIT technique to enhance its availability. In this study, we provide the experimental evaluation of sub-millimeter resolution conductivity imaging method using a 3T MR scanner combined with a multi-echo MR pulse sequence, multi-channel RF coil, and phase optimization method. From the phantom and animal imaging results, sub-millimeter resolution exhibited similar signal-to-noise ratio of MR magnitude and noise levels in magnetic flux density comparing to the existing millimeter resolution. The reconstructed conductivity images at sub-millimeter resolution can distinguish different brain tissues with a pixel size as small as 350 μm.

Abstract:Magnetic resonance electrical impedance tomography (MREIT) aims at producing high-resolution cross-sectional conductivity images of an electrically conducting object such as the human body. Following numerous phantom imaging experiments, the most recent study demonstrated successful conductivity image reconstructions of postmortem canine brains using a 3 T MREIT system with 40 mA imaging currents. Here, we report the results of in vivo animal imaging experiments using 5 mA imaging currents. To investigate any change of electrical conductivity due to brain ischemia, canine brains having a regional ischemic model were scanned along with separate scans of canine brains having no disease model. Reconstructed multi-slice conductivity images of in vivo canine brains with a pixel size of 1.4 mm showed a clear contrast between white and gray matter and also between normal and ischemic regions. We found that the conductivity value of an ischemic region decreased by about 10-14%. In a postmortem brain, conductivity values of white and gray matter decreased by about 4-8% compared to those in a live brain. Accumulating more experience of in vivo animal imaging experiments, we plan to move to human experiments. One of the important goals of our future work is the reduction of the imaging current to a level that a human subject can tolerate. The ability to acquire high-resolution conductivity images will find numerous clinical applications not supported by other medical imaging modalities. Potential applications in biology, chemistry and material science are also expected.

Abstract:Multi-frequency electrical impedance tomography (MFEIT) reconstructs the image of conductivity inside the human body based on the dependence of tissue conductivity on frequency. As there exist differences in the conductivity over frequency between blood, ischemic cortical tissue and normal cortical tissue, MFEIT has potential application in the detection of acute stroke. However, because the conductivity distribution of the human head is highly inhomogeneous and the conductivities of normal head tissue and stroke lesion tissue both change with frequency, the anomaly and normal head tissues are often mixed together in the reconstructed image, which makes it difficult to discern the anomaly. Here we present a spectral decomposition frequency-difference (SD-FD) imaging algorithm in an attempt to address this issue: firstly, we reconstruct so-called EIT spectral images according to the conductivity spectra of tissues; secondly, we obtain the EIT image of the anomaly from the spectral images by using independent component analysis. The results show that the proposed algorithm is capable of detecting the anomaly in a numerical head phantom, as well as in a realistic human head tank with frequency-dependent and heterogeneous conductivities distribution. The proposed SD-FD algorithm may support MFEIT use for human stroke imaging in the future.

Abstract:Low-frequency conductivity of an anisotropic tissue is associated with its cellular structure. Imaging of the conductivity tensor inside the human body could prove invaluable to analyses of interactions between electromagnetic fields and biological systems, such as predictions of current pathways during electrical stimulation. Since the muscle is the most abundant anisotropic tissue in the human body, in vitro and in vivo muscle conductivity tensor assessment has been attempted. In this study, we conducted phantom imaging of biological tissues using conductivity tensor imaging (CTI) to validate its in vivo usefulness. We constructed phantoms using bovine and/or porcine muscles and performed a CTI experiment using MRI. High-frequency conductivity was first obtained using B1 mapping with a multi-echo spin-echo pulse sequence. Information about the cellular space was obtained with a multi-b diffusion tensor imaging sequence. We combined the data from these separate scans to reconstruct the conductivity tensor images of the phantoms. The low-frequency and high-frequency conductivities of the muscle tissues in the phantoms were compared with the corresponding values measured by an impedance analyzer. The anisotropy of each muscle tissue was quantified as an anisotropy ratio (AR), defined as the ratio of the eigenvalues of a conductivity tensor along the longitudinal direction to those along the transversal directions. The isotropic conductivity and conductivity tensor in bovine muscles were less than those of porcine muscles. However, the anisotropy was stronger in bovine muscles based on the AR values by fiber directions. Current CTI is a promising noninvasive tool for evaluation of the muscle microstructure.

Abstract:Magnetic resonance electrical properties tomography (MREPT) uses the B1 mapping technique to provide the high-frequency conductivity distribution at Larmor frequency that simultaneously reflects the intracellular and extracellular effects. In biological tissues, the electrical conductivity can be described as the concentration and mobility of charge carriers. For the water molecule diffusivity, diffusion weighted imaging (DWI) measures the random Brownian motion of water molecules within biological tissues. The DWI data can quantitatively access the mobility of microscopic water molecules within biological tissues. By measuring multi-b-value DWI data and the recovered high-frequency conductivity at Larmor frequency, we propose a new method to decompose the conductivity into the total ion concentration and mobility in the extracellular space (ECS) within a routinely applicable MR scan time. Using the measured multi-b-value DWI data, a constrained compartment model is designed to estimate the extracellular volume fraction and extracellular mean diffusivity. With the extracted extracellular volume fraction and water molecule diffusivity, we directly reconstruct the low-frequency electrical properties including the extracellular mean conductivity and extracellular conductivity tensor. To demonstrate the proposed method by comparing the ion concentration and the ion mobility, we conducted human experiments for the proposed low-frequency conductivity imaging. Human experiments verify that the proposed method can recover the low-frequency electrical properties using a conventional MRI scanner.

Abstract:The catalytic domain of metalloelastase (matrix metalloproteinase-12 or MMP-12) is unique among MMPs in exerting high proteolytic activity upon fibrils that resist hydrolysis, especially elastin from lungs afflicted with chronic obstructive pulmonary disease or arteries with aneurysms. How does the MMP-12 catalytic domain achieve this specificity? NMR interface mapping suggests that α-elastin species cover the primed subsites, a strip across the β-sheet from β-strand IV to the II–III loop, and a broad bowl from helix A to helix C. The many contacts may account for the comparatively high affinity, as well as embedding of MMP-12 in damaged elastin fibrils in vivo. We developed a strategy called BINDSIght, for bioinformatics and NMR discovery of specificity of interactions, to evaluate MMP-12 specificity without a structure of a complex. BINDSIght integration of the interface mapping with other ambiguous information from sequences guided choice mutations in binding regions nearer the active site. Single substitutions at each of ten locations impair specific activity toward solubilized elastin. Five of them impair release of peptides from intact elastin fibrils. Eight lesions also impair specific activity toward triple helices from collagen IV or V. Eight sites map to the “primed” side in the III–IV, V–B, and S1′ specificity loops. Two map to the “unprimed” side in the IV–V and B–C loops. The ten key residues circumscribe the catalytic cleft, form an exosite, and are distinctive features available for targeting by new diagnostics or therapeutics.
Validation of conductivity tensor imaging against diffusion tensor magnetic resonance electrical impedance tomography

Abstract:Diffusion tensor magnetic resonance electrical impedance tomography (DT-MREIT) and electrodeless conductivity tensor imaging (CTI) are two emerging modalities that can quantify low-frequency tissue anisotropic conductivity properties by assuming similar properties underlie ionic mobility and water diffusion. While both methods have potential applications to estimating neuro-modulation fields or formulating forward models used for electrical source imaging, a direct comparison of the two modalities has not yet been performed in-vitro or in-vivo. Therefore, the aim of this study was to test the equivalence of these two modalities. We scanned a tissue phantom and the head of human subject using DT-MREIT and CTI protocols and reconstructed conductivity tensor and effective low frequency conductivities. We found both gray and white matter conductivities recovered by each technique were equivalent within 0.05 S/m. Both DT-MREIT and CTI require multiple processing steps, and we further assess the effects of each factor on reconstructions and evaluate the extent to which different measurement mechanisms potentially cause discrepancies between the two methods. Finally, we discuss the implications for spectral models of measuring conductivity using these techniques. The study further establishes the credibility of CTI as an electrodeless non-invasive method of measuring low frequency conductivity properties.

Abstract:Abstract Conventional magnetic resonance electrical impedance tomography (MREIT) reconstruction methods require administration of two linearly independent currents via at least two electrode pairs. This requires long scanning times and inhibits coordination of MREIT measurements with electrical neuromodulation strategies. We sought to develop an isotropic conductivity reconstruction algorithm in MREIT based on a single current injection, both to decrease scanning time by a factor of two and enable MREIT measurements to be conveniently adapted to general transcranial- or implanted-electrode neurostimulation protocols. In this work, we propose and demonstrate an iterative algorithm that extends previously published MREIT work using two-current administration approaches. The proposed algorithm is a single-current adaptation of the harmonic B z algorithm. Forward modeling of electric potentials is used to capture changes of conductivity along current directions that would normally be invisible using data from a single-current administration. Computational and experimental results show that the reconstruction algorithm is capable of reconstructing isotropic conductivity images that agree well in terms of L 2 error and structural similarity with exact conductivity distributions or two-current-based MREIT reconstructions. We conclude that it is possible to reconstruct high quality electrical conductivity images using MREIT techniques and one current injection only.

Imaging Intra- and Extracellular Conductivity using MR based Conductivity Tensor Imaging

Abstract:Imaging electrical conductivity may reveal relationships between biological tissues, cellular structures, and physiological processes. Biological tissues are primarily composed of major ions such as Na+ and K+, with varying concentrations and mobility within the cellular structures. These tissues consist of intracellular and extracellular fluids separated by cell membranes, and their electrical conductivity can be expressed as a function of ion concentration and mobility. This study introduces Conductivity Tensor Imaging (CTI) to independently reconstruct the electrical conductivity of intra- and extracellular compartments in biological tissues using MRI. We validated this method using a conductivity phantom with three compartments filled with electrolytes and/or giant vesicle suspensions. These vesicles mimic cell-like materials with thin insulating membranes, providing a realistic model for cellular structures. Measurements showed that high-frequency conductivity closely matched low-frequency conductivity in normal electrolytes. However, in the giant vesicle compartment, the conductivity of extracellular (EC) and intracellular (IC) regions correlated with cell volume fraction. In vivo human brain imaging using CTI revealed significant EC and IC conductivity variations across different brain regions, corresponding to underlying cellular compositions and structures. CTI introduces a novel MR contrast mechanism to distinctly measure IC and EC conductivities. Our findings highlight the potential of CTI to enhance our understanding of brain microstructure and its physiological processes through detailed conductivity mapping. This method signifies a notable advancement in non-invasive imaging, providing novel insights into the electrical properties of biological tissues and their implications for biophysical properties.

Abstract:This work was inspired by the observation that a majority of MR-electrical properties tomography studies are based on direct comparisons with ex vivo measurements carried out on post-mortem samples in the 90's. As a result, the in vivo conductivity values obtained from MRI in the megahertz range in different types of tissues (brain, liver, tumors, muscles, etc.) found in the literature may not correspond to their ex vivo equivalent, which still serves as a reference for electromagnetic modelling. This study aims to pave the way for improving current databases since the definition of personalized electromagnetic models (e.g. for Specific Absorption Rate estimation) would benefit from better estimation. Seventeen healthy volunteers underwent MRI of both brain and thorax/abdomen using a three-dimensional ultrashort echo-time (UTE) sequence. We estimated conductivity (S/m) in several classes of macroscopic tissue using a customized reconstruction method from complex UTE images, and give general statistics for each of these regions (mean-median-standard deviation). These values are used to find possible correlations with biological parameters such as age, sex, body mass index and/or fat volume fraction, using linear regression analysis. In short, the collected in vivo values show significant deviations from the ex vivo values in conventional databases, and we show significant relationships with the latter parameters in certain organs for the first time, e.g. a decrease in brain conductivity with age.

Abstract:The electroporation effect on tissue can be assessed by measurement of electrical properties of the tissue undergoing electroporation. The most prominent techniques for measuring electrical properties of electroporated tissues have been voltage-current measurement of applied pulses and electrical impedance tomography (EIT). However, the electrical conductivity of tissue assessed by means of voltage-current measurement was lacking in information on tissue heterogeneity, while EIT requires numerous additional electrodes and produces results with low spatial resolution and high noise. Magnetic resonance EIT (MREIT) is similar to EIT, as it is also used for reconstruction of conductivity images, though voltage and current measurements are not limited to the boundaries in MREIT, hence it yields conductivity images with better spatial resolution. The aim of this study was to investigate and demonstrate the feasibility of the MREIT technique for assessment of conductivity images of tissues undergoing electroporation. Two objects were investigated: agar phantoms and ex vivo liver tissue. As expected, no significant change of electrical conductivity was detected in agar phantoms exposed to pulses of all used amplitudes, while a considerable increase of conductivity was measured in liver tissue exposed to pulses of different amplitudes.

Abstract:Magneto-acousto-electrical tomography (MAET) is a hybrid imaging method that combines the high spatial resolution of ultrasonography with the high contrast of electrical impedance tomography (EIT). While most previous studies on MAET have focused on two-dimensional imaging, our recent research proposed a novel three-dimensional (3D) MAET method using B-mode and translational scanning. This method has been the first to reconstruct a 3D volume image of conductivity interfaces. However, this method has its limitations in mapping irregular shapes of conductivity. To address this challenge, we propose a 3D magneto-acousto-electrical computed tomography (3D MAE-CT) method utilizing an ultrasound linear array transducer in this work. Both phantom and in vitro experiments were conducted to validate our proposed method. The results from the phantom experiments demonstrate that our method can map the 3D volume conductivity with high spatial resolution. The oblique angles extracted from the 3D image closely match practical value, with the relative error ranging between -2.80% and 4.07%. Furthermore, the in vitro experiment successfully obtained a 3D image of a chicken heart, marking the first MAET 3D conductivity image of a tissue sample to date.
Magneto-acousto-electrical Measurement Based Electrical Conductivity Reconstruction for Tissues.

Abstract:OBJECTIVE:Based on the interaction of ultrasonic excitation and magnetoelectrical induction, magneto-acousto-electrical (MAE) technology was demonstrated to have the capability of differentiating conductivity variations along the acoustic transmission. By applying the characteristics of the MAE voltage, a simplified algorithm of MAE measurement based conductivity reconstruction was developed.METHODS:With the analyses of acoustic vibration, ultrasound propagation, Hall effect, and magnetoelectrical induction, theoretical and experimental studies of MAE measurement and conductivity reconstruction were performed. The formula of MAE voltage was derived and simplified for the transducer with strong directivity. MAE voltage was simulated for a three-layer gel phantom and the conductivity distribution was reconstructed using the modified Wiener inverse filter and Hilbert transform, which was also verified by experimental measurements.RESULTS:The experimental results are basically consistent with the simulations, and demonstrate that the wave packets of MAE voltage are generated at tissue interfaces with the amplitudes and vibration polarities representing the values and directions of conductivity variations. With the proposed algorithm, the amplitude and polarity of conductivity gradient can be restored and the conductivity distribution can also be reconstructed accurately.CONCLUSION:The favorable results demonstrate the feasibility of accurate conductivity reconstruction with improved spatial resolution using MAE measurement for tissues with conductivity variations, especially suitable for nondispersive tissues with abrupt conductivity changes.SIGNIFICANCE:This study demonstrates that the MAE measurement based conductivity reconstruction algorithm can be applied as a new strategy for nondestructive real-time monitoring of conductivity variations in biomedical engineering.
Noninvasive imaging of bioimpedance distribution by means of current reconstruction magnetic resonance electrical impedance tomography

Abstract:We have developed a novel magnetic resonance electrical impedance tomography (MREIT) algorithm-current reconstruction MREIT algorithm-for noninvasive imaging of electrical impedance distribution of a biological system using only one component of magnetic flux density. The newly proposed algorithm uses the inverse of Biot-Savart Law to reconstruct the current density distribution, and then, uses a modified J-substitution algorithm to reconstruct the conductivity image. A series of computer simulations has been conducted to evaluate the performance of the proposed current reconstruction MREIT algorithm with simulation settings for breast cancer imaging applications, with consideration of measurement noise, current injection strength, size of simulated tumors, spatial resolution, and position dependency. The present simulation results are highly promising, demonstrating the high spatial resolution, high accuracy in conductivity reconstruction, and robustness against noise of the proposed algorithm for imaging electrical impedance of a biological system. The present MREIT method may have potential applications to breast cancer imaging and imaging of other organs.
Direct reconstruction of tissue conductivity with deconvolution in magneto-acousto-electrical tomography (MAET): theory and numerical simulation

Abstract:Magneto-acousto-electrical tomography (MAET), a combination of ultrasound imaging and electrical impedance tomography (EIT), offers both high resolution (in comparison to EIT) and high contrast (in comparison to ultrasound imaging). It is used to map the internal conductivity distribution of an imaging object. However, conductivity reconstruction in MAET is a challenge, so conventional MAET is mainly devoted to mapping the conductivity interface. This is primarily because integration byparts is used in the theory derivation, and the simplified measurement formula suggests the voltage is proportional to the conductivity gradient, which leads to an error in the measurement formula. In this study, the measurement signal is expressed as the convolution of acoustic velocity and conductivity distribution without using integration by parts, which retains the low-frequency term in the measurement signal. Based on the convolution formula, we subsequently propose a direct conductivity reconstruction scheme with deconvolution by utilizing the low-frequency component. We verify the proposed method based on two two-dimension models and quantify the L2 errors of reconstructed conductivity. Besides, we analyze factors influencing the reconstructed accuracy such as reconstructed regularization parameter ultrasound frequency, and noise. We also demonstrate that the spatial resolution is not influenced by the duration of excitation ultrasound. With the contributions of the proposed method, conductivity imaging appears to be feasible for application to the early diagnosis in the future.

Radiofrequency ablation lesion detection using MR-based electrical conductivity imaging: A feasibility study of ex vivo liver experiments

Abstract:PURPOSE: The aim of this study was to show the potential of magnetic resonance electrical impedance tomography (MREIT) conductivity imaging in terms of its capability to detect ablated lesions and differentiate tissue conditions in liver radiofrequency (RF) ablation.MATERIALS AND METHODS: RF ablation procedures were performed in bovine livers using a LeVeen RF needle electrode. Ablation lesions were created using a power-controlled mode at 30, 50, and 70 W for 1, 3, and 5 min of exposure time, respectively. After the ablation, the liver was cut into several blocks including the ablated lesion, and positioned inside a phantom filled with agarose gel. Electrodes were attached on the side of the phantom and it was placed inside the MRI bore. For MREIT imaging, multi-spin-echo pulse sequence was used to obtain the magnetic flux density data according to the injection currents.RESULTS: The conductivity of ablation lesions was significantly changed with the increase of exposure time (pKW < 0.01, Kruskal-Wallis test). With RF powers of 30 and 50 W, significant differences between the coagulation necrosis and hyperaemic rim were observed for more than 5 min and 3 min, respectively (pMW < 0.01, Mann-Whitney test). At 70 W, all cases showed significant differences except 3 min (pMW < 0.01). The positive correlation between the exposure time and tissue conductivity was observed in both two ablation areas (pSC < 0.01, Spearman correlation).CONCLUSIONS: This ex vivo feasibility study demonstrates that current MREIT conductivity imaging can detect liver RF ablation lesions without using any contrast media or additional MR scan.

Abstract:Imaging of electrical conductivity is a promising technique in biomedical field, which can reveal the impedance distribution within the region of interest. However, the contact measurement of traditional electrical impedance tomography (EIT) results in some challenging practical limitations on its applications. This paper introduces a novel capacitively coupled EIT to the biomaterial/biomedical field for resistivity imaging, and studies various aspects of this new contactless technique on practical applications. A 12-electrode experimental phantom is developed and the corresponding computational model is established to obtain the sensitivity matrix of the phantom. A hybrid image reconstruction method, which combines the Tikhonov regularization method and the simultaneous iterative reconstruction technique, is introduced to solve the inverse problem. In biomedical applications, the frequency-dependent conductivity aspect is very critical. Therefore, both the time-difference and frequency-difference imaging methods are investigated. A background calibration approach is proposed for the frequency-difference capacitively coupled EIT to overcome the frequency dependence of the background signal. Experiments were carried out with three kinds of biomaterials and three backgrounds with different conductivities. Results show the working principles and potential of the capacitively coupled EIT on biomaterial and biomedical applications.

Abstract:This article presents a series of three-dimensional results for Magneto-acousto-electrical Tomography (MAET), which is a hybrid imaging modality combining the merits of high contrast and high resolution. The ultrasound field and electromagnetic field are coupled to generate current density distribution inside the sample. However, three-dimensional images have not yet been realized for MAET through voltage signals detected by electrodes. In this paper, the mathematical model of MAET is analyzed, and a new image reconstruction scheme is proposed, which is verified by a numerical framework. One three-dimensional numerical model of normal and tumor tissues is formed to analyze the multiphysics problems. In the model, vibration velocity is obtained for pulse type ultrasound excitations with the duration of 0.5 <span class="equationTd inline-formula"><math> μ</math></span>s. The static magnetic field produced by two permanent magnets is about 300 mT. In order to measure the voltage caused by the MAET effect, two electrodes are attached on the surface of the sample. A series of MAET signals are obtained when the transducer locates in different positions. The distribution of three-dimensional conductivity is recovered by the new image reconstruction scheme. The reconstructed conductivity images are consistent with the original distribution, thus verifying the effectiveness of the new image reconstruction scheme. In addition, the experiment has also validated the mathematical model and simulation results. The simulation and the experimental results indicate that the MAET has the potential to become a new tool to study the electrical properties of tumors in humans.

Abstract:Purpose To evaluate the performance of various MR electrical properties tomography (MR‐EPT) methods at 3 T in terms of absolute quantification and spatial resolution limit for electrical conductivity. Methods Absolute quantification as well as spatial resolution performance were evaluated on homogeneous phantoms and a phantom with holes of different sizes, respectively. Ground‐truth conductivities were measured with an open‐ended coaxial probe connected to a vector network analyzer (VNA). Four widely used MR‐EPT reconstruction methods were investigated: phase‐based Helmholtz (PB), phase‐based convection‐reaction (PB‐cr), image‐based (IB), and generalized‐image‐based (GIB). These methods were compared using the same complex images from a 1 mm‐isotropic UTE sequence. Alternative transceive phase acquisition sequences were also compared in PB and PB‐cr. Results In large homogeneous phantoms, all methods showed a strong correlation with ground truth conductivities (r > 0.99); however, GIB was the best in terms of accuracy, spatial uniformity, and robustness to boundary artifacts. In the resolution phantom, the normalized root‐mean‐squared error of all methods grew rapidly (>0.40) when the hole size was below 10 mm, with simplified methods (PB and IB), or below 5 mm, with generalized methods (PB‐cr and GIB). Conclusion VNA measurements are essential to assess the accuracy of MR‐EPT. In this study, all tested MR‐EPT methods correlated strongly with the VNA measurements. The UTE sequence is recommended for MR‐EPT, with the GIB method providing good accuracy for structures down to 5 mm. Structures below 5 mm may still be detected in the conductivity maps, but with significantly lower accuracy.

In rats more than 5 weeks after induction, the fibrous region showed an increased conductivity of ≥12% compared to that of the corresponding normal control rats. From regional comparisons in the same liver, the fibrous region showed an increased conductivity of ≥11% compared to the opposite, less induced region of rats more than 5 weeks after induction. Liver samples from the fibrous region represent tissue damages such as diffuse centrilobular congestion with marked dilatation of central veins from the histological findings. Immunohistochemistry revealed significant levels of attenuated fibrosis and increased inflammatory response.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Data Conclusion</h3><p>The increased conductivity in the fibrous region is related to the changes of the extracellular space.
