Abstract:Human milk contains a lot of components (i.e. proteins, carbohydrates, lipids, inorganic elements) which provide basic nutrients for infants during the first period of their lives. Qualitative composition of milk components of healthy mothers is similar, but their levels change during lactation stages. Colostrum is the fluid secreted during the first days postpartum by mammary epithelial cells. Colostrum is replaced by transitional milk during 5-15 days postpartum and from 15 days postpartum mature milk is produced. Human milk, apart from nutritional components, is a source of biologically active molecules, i.e. immunoglobulins, growth factors, cytokines, acute phase proteins, antiviral and antibacterial proteins. Such components of human milk are responsible for specific biological activities of human milk. This secretion plays an important role in growth and development of newborns. Bioactive molecules present in the milk support the immature immune system of the newborn and also protect against the development of infection. In this article we describe the pathways involved in the production and secretion of human milk, the state of knowledge on the proteome of human milk, and the contents of components of milk during lactation. Moreover, some growth factors and proteins involved in innate and specific immunity, intercellular communication, immunomodulation, and inflammatory processes have been characterized.
Immunomodulatory Effects of L. Helveticus WHH2580 Fermented Milk on an Immunosuppressed Murine Model

Abstract:Several bacterial strains from traditional fermented foods were screened on a large scale to identify novel candidate probiotics that can be used as starters. Two in vitro cellular models were used to identify strains with immunomodulatory activity. Three strains with pro-inflammatory activity were selected, and their recovery effect was tested in vivo in an immunosuppressed murine model. Only Lactobacillus helveticus WHH2580 enhanced natural killer cell activity and splenic lymphocyte proliferation. Furthermore, milk fermented using the strain WHH2580 exerted an immunomodulatory effect with enhanced resistance to gastrointestinal conditions, including strong adhesion to HT-29 cells. Whole genome sequencing revealed the presence of a circular chromosome and two circular plasmids; no antibiotic resistance or putative virulence genes were discovered. These findings indicate the safety and positive role of L. helveticus WHH2580 in maintaining host health in mice and suggest that milk fermented with this strain can deliver this strain and thus promote immunity.

Abstract:Breastfeeding has been associated with many benefits, both in the short and in the long term. Infants being breastfed generally have less illness and have better cognitive development at 1 year of age than formula-fed infants. Later in life, they have a lower risk of obesity, diabetes and cardiovascular disease. Several components in breast milk may be responsible for these different outcomes, but bioactive proteins/peptides likely play a major role. Some proteins in breast milk are comparatively resistant towards digestion and may therefore exert their functions in the gastrointestinal tract in intact form or as larger fragments. Other milk proteins may be partially digested in the upper small intestine and the resulting peptides may exert functions in the lower small intestine. Lactoferrin, lysozyme and secretory IgA have been found intact in the stool of breastfed infants and are therefore examples of proteins that are resistant against proteolytic degradation in the gut. Together, these proteins serve protective roles against infection and support immune function in the immature infant. α-lactalbumin, β-casein, κ-casein and osteopontin are examples of proteins that are partially digested in the upper small intestine, and the resulting peptides influence functions in the gut. Such functions include stimulation of immune function, mineral and trace element absorption and defense against infection.

Abstract:There is no doubt that milk proteins provide excellent nutrition for the suckling. However, apart from that, milk proteins can also exert numerous physiological activities benefiting the suckling in a variety ofways. These activities include enhancement of immune function, defense against pathogenic bacteria, viruses, and yeasts, and development of the gut and its functions. Besides the naturally occurring, biologically active proteins present in milk, a variety of bioactive peptides are encrypted within the sequence of milk proteins that are released upon suitable hydrolysis of the precursor protein. A large range of bioactivities has been reported for milk protein components, with some showing more than one kind of biological activity (Korhonen&Pihlanto, 2006). This chapter reviews the most important antimicrobial and antitumor peptides derived from milk proteins, especially those that may have a physiological significance to the suckling neonate. Antimicrobial peptides present in milk that are not derived frommilk proteins are also considered. Special attention is given to the generation of these peptides by the action of different proteolytic enzymes and the origin of these enzymes since, if present in the digestive tract, it is likely that the peptides might play a role in the host defense system. Finally, the most relevant in vivo studies carried out with this kind of bioactive peptides are discussed.

Abstract:The objective of this study was to evaluate the nutritional quality of reconstituted milk by determining concentrations of several important proteins. Difference in protein between raw milk and reconstituted milk was determined with spectrophotometer, HPLC, fluorometry, ELISA, photon correlation spectroscopy. Total undenatured whey protein, β-lactoglobulin, α-lactalbumin, chemically available lysine, insulin-like growth factor-Ⅰ (IGF-Ⅰ), epidermal growth factor (EGF), immunoglobulin G (IgG) contents and casein micelle size were analyzed for both raw milk and reconstituted milk. Results showed that reconstituted milk had a significant decrease (P<0.01) in total undenatured whey protein, β-lactoglobulin, α-lactalbumin, chemically available lysine, IGF-Ⅰ, EGF and IgG. The decrease of casein micelle particle size confirmed that most of the whey protein denatured and associated with the casein micelle occurred in the processing of milk powder manufacture. It is concluded that reconstituted milk has a lower nutritional value than that of raw milk.

Abstract:Bioactive peptides encrypted within the native milk proteins can be released by enzymatic proteolysis, food processing, or gastrointestinal digestion. These peptides possess a wide range of properties, including immunomodulatory properties. The first months of life represent a critical period for the maturation of the immune system because a tolerance for nutrient molecules should be developed while that for pathogen-derived antigens is avoided. Evidence has accumulated to suggest that milk peptides may regulate gastrointestinal immunity, guiding the local immune system until it develops its full functionality. Our data using the weaning piglet as the model suggest that several milk peptides can downregulate various immune properties at a time (one to two weeks after weaning) that coincides with immaturity of the immune system. The protein kinase A system and/or the exchange protein directly activated by cyclic AMP (Epac-1) are implicated in the mechanism through which milk peptides can affect immune function in the early postweaning period. Despite the fact that the research in this field is in its infancy, the evidence available suggests that milk protein peptides may promote development of neonatal immune competence.Milk contains a variety of components that provide immunological protection and facilitate the development of neonatal immune competence. Two main categories of milk compounds are thought to be associated with immunological activity. The first category includes cytokines, which neonates do not produce efficiently. Cytokines present in milk are thought to be protected against intestinal proteolysis and could alleviate immunological deficits, aiding immune system maturation (Kelleher&Lonnerdal, 2001; Bryan et al., 2006). The second category of milk compounds includes milk protein peptides. Milk peptides may affect mucosal immunity possibly by guiding local immunity until it develops its full functionality (Baldi et al., 2005). This chapter focuses on the effects of milk peptides on immune function and attempts to provide an overview of the knowledge available in this field.

Abstract:Milk powder, a nutrient-rich dairy product, lacks comprehensive information summarizing its specific properties when produced by spray- and freeze-dried technologies from different sources. Therefore, this study investigated the differences in physicochemical properties, microstructure, and proteome of spray- and freeze-dried milk powders from bovine, goat, and horse sources. The results revealed that spray-dried milk powder exhibited a smaller particle size, lower air content within the powder particles, inferior reconstitution properties, and lower lactose crystallinity compared with freeze-dried milk powder. Additionally, among the studied varieties, horse milk powder showed the lowest flowability but the most effective reconstitution properties. Proteomic analysis indicated that freeze-dried milk powder exhibited higher levels of immune-related proteins, including complement C3, C7, and complement factor B, and antimicrobial enzymes such as lysozyme and lactoperoxidase compared with spray-dried milk powder. Furthermore, specific milk powders contained more immune-related proteins such as serum amyloid A, myeloid antimicrobial peptide-28, polymeric immunoglobulin receptor, and mucin-1 compared with bovine milk powder. These findings contribute to a deeper understanding of the differences in the physical-chemical properties and potential biological functions of spray- and freeze-dried milk powders from various sources, which may help in further optimizing specific milk powder processing technologies.

Abstract:Nowadays, consumers choose food products, pay attention not only to their nutritional value and taste, but also to health-promoting properties. Milk from various animal species is a rich source of health-enhancing components present in the fat, protein and water fractions. They exert a multidirectional impact on the human organism and limit the risk of development of many lifestyle diseases. There are differences in the content of bioactive ingredients in milk from various animal species. In comparison with cow milk, which is of key importance in the world production, sheep and donkey milk contains higher amounts of whey proteins (mainly β-Lg) and polyunsaturated acids. Camel milk deserves special attention as well due to its high content of antibacterial substances, i.e. lactoferrin and lysozyme, as well as vitamins C and E. Importantly, milk and dairy products are a rich source of essential amino acids and minerals (mainly calcium), indispensable for normal functioning of the human organism. Due to the presence of antioxidants, i.e. β-Lg, lactoferrin, CLA, and vitamins E and C, they are classified as natural antioxidants.

Abstract:Milk contains components that provide critical nutritive elements, immunological protection, and biologically active substances to both neonates and adults. Milk proteins are currently the main source of a range of biologically active peptides. Concentrates of these peptides are potential health-enhancing nutraceuticals for food and pharmaceutical applications. Several bioactive peptides may be used as nutraceuticals, for example, in the treatment of diarrhea, hypertension, thrombosis, dental diseases, as well as mineral malabsorption, and immunodeficiency. Minor whey proteins, such as lactoferrin, lactoperoxidase, lysozyme, and immunoglobulins, are considered antimicrobial proteins. Milk also contains some natural bioactive substances. These include oligosaccharides, fucosylated oligosaccharides, hormones, growth factors, mucin, gangliosides, and endogenous peptides, which are present in milk at secretion. Most of the claimed physiological properties of milk bioactive components have been carried out in vitro or in animal model systems, and these hypothesized properties remain to be proven in humans. Whether these milk bioactive components will replace drugs entirely in the immediate future is still unclear, but the increasing appreciation of "drug foods" or nutraceuticals plays a complementary rather than a substitutional role to the synthetic pharmacological drugs.

Abstract:The milk proteins from samples of 13 different animals were identified utilizing 4D-Label-Free proteomics technology, leading to the identification of a substantial number of proteins. Among the various samples, Chinese people (CHP) milk proteins exhibited the highest count, with 1149 distinct proteins. Simultaneously, we identified common proteins present in these animal milk. It's notable presence in goat milk contributes to enhancing infant infection resistance, showcasing the beneficial role of lactoperoxidase. Galectin-3 binding protein (Gal-3BP) and tetraspanin in human milk are significantly higher than those in other animals, which determine the prominent antiviral effect of human milk and the important processes related to cell transduction. Furthermore, human milk, camel milk, goat milk and sheep milk proved to be rich sources of milk fat globule membrane (MFGM) proteins. The insights obtained from this study can serve as a foundational framework for exploring the role of different animal milk proteins in disease treatment and the composition of infant formula.

Abstract:Milk is a source of many valuable nutrients, including minerals, vitamins and proteins, with an important role in adult health. Milk and dairy products naturally containing or with added probiotics have healthy functional food properties. Indeed, probiotic microorganisms, which beneficially affect the host by improving the intestinal microbial balance, are recognized to affect the immune response and other important biological functions. In addition to macronutrients and micronutrients, biologically active peptides (BPAs) have been identified within the amino acid sequences of native milk proteins; hydrolytic reactions, such as those catalyzed by digestive enzymes, result in their release. BPAs directly influence numerous biological pathways evoking behavioral, gastrointestinal, hormonal, immunological, neurological, and nutritional responses. The addition of BPAs to food products or application in drug development could improve consumer health and provide therapeutic strategies for the treatment or prevention of diseases. Herein, we review the scientific literature on probiotics, BPAs in milk and dairy products, with special attention to milk from minor species (buffalo, sheep, camel, yak, donkey, etc.); safety assessment will be also taken into consideration. Finally, recent advances in foodomics to unveil the probiotic role in human health and discover novel active peptide sequences will also be provided.

Abstract:Proteins and peptides play vital roles in different biological processes in vivo . As a dynamic hydrolysis system, milk is rich in proteins and proteases and provides a constant supply of endogenous bioactive peptides to newborn mammals. Previous studies have primarily focused on researching bioactive peptides by adding exogenous enzymes to milk samples. However, such an approach overlooks the significance of endogenous peptides and parent proteins that naturally exist in milk. Herein, we analyzed and compared parent proteins and their releasing peptides in human colostrum (HC), bovine colostrum (BC), and donkey colostrum (DC). The predominant proteins and hydrolyzed peptides in the three types of milk were identified. Among them, peptides were found to possess common bioactivities, including ACE inhibitory, antioxidant, antibacterial and immunomodulatory properties in HC, BC, and DC. Furthermore, the biological functions of these parent proteins were clarified using bioinformatics. These insights offer a novel perspective on natural bioactive peptides and the potential utilization of specific parent proteins and peptides to develop infant formulae derived from diverse milk sources.

Abstract:Milk has long been recognized as a source of macro- and micronutrients, immunological components, and biologically active substances, which not only allow growth but also promote health in mammalian newborns. Many milk lipids, lipid-soluble substances, and their digested products are bioactive, including vitamins and vitamin-like substances. Vitamins A, E, D, and K and carotenoids are known as highly lipophilic food microconstituents (HLFMs), and all occur in milk. HLFMs also include phytosterols, which, although they are not vitamins, are nevertheless biologically active and present in milk. Fat-soluble micronutrients, including fat-soluble vitamins, are embedded in the milk fat fraction, and this has important implications for their bioaccessibility and bioavailability from milk. In fact, the fat component of milk is an effective delivery system for highly lipophilic microconstituents. The vitamin content of animal products can be enhanced by increasing the feed content of synthetic or natural vitamins or precursors. An advantage of augmenting milk microconstituents by animal nutrition rather than milk fortification is that it helps safeguard animal health, which is a primary factor in determining the quality, safety, and wholesomeness of animal-origin foods for human consumption. The milk fat delivery system offers numerous possibilities for exploitation by nutritionists. For example, the payload could consist of enhanced levels of several micronutrients, opening possibilities for synergic effects that are as yet incompletely understood.

Improving INS/GPS Navigation Accuracy through Compensation of Kalman Filter Errors

Abstract:The Kalman filter is often used to integrate satellite navigation systems with inertial navigation systems. Such integrated systems are especially useful for navigation of vehicles in urban environments where satellite signals are frequently blocked by tall buildings. The filter weights the measurements of both navigation systems to provide an overall optimal solution. Unfortunately, an optimal solution is only achieved when the filter has been supplied with ideal a priori information such as proper measurement noise characteristics and system dynamics. If such parameters are not perfect they can be detected and compensated for using an intelligent navigation scheme which is adaptable to different sensors. As dynamics are encountered, satellite signal blockages are simulated to test the optimality of the filter. A neural network is then trained to learn any residual deterministic errors which are then removed from future system drifts during signal blockages.

Abstract:The provision of essential and non-essential amino acids for breast-fed infants is the major function of milk proteins. In addition, breast-fed infants might benefit from bioactivities of milk proteins, which are exhibited in the intestine during the digestive phase and by absorption of intact proteins or derived peptides. For lactoferrin, osteopontin and milk fat globule membrane proteins/lipids, which have not until recently been included in substantial amounts in infant formulas, in vitro experiments and animal models provide a convincing base of evidence for bioactivities, which contribute to the protection of the infant from pathogens, improve nutrient absorption, support the development of the immune system and provide components for optimal neurodevelopment. Technologies have become available to obtain these compounds from cow´s milk and the bovine compounds also exhibit bioactivities in humans. Randomized clinical trials with experimental infant formulas incorporating lactoferrin, osteopontin, or milk fat globule membranes have already provided some evidence for clinical benefits. This review aims to compare findings from laboratory and animal experiments with outcomes of clinical studies. There is good justification from basic science and there are promising results from clinical studies for beneficial effects of lactoferrin, osteopontin and the milk fat globule membrane complex of proteins and lipids. Further studies should ideally be adequately powered to investigate effects on clinically relevant endpoints in healthy term infants.

Abstract:Colostrum contains numerous bioactive proteins, required to develop the passive immune system of neonates. The composition of colostrum varies with lactations and the number of bioactive components also changes in due course. Most of the colostrum and milk proteins contain glycosylated components that affect the functionality of these proteins. The glycosylation pattern of these proteins also changes with lactations from colostrum to the mature milk. Glycosylation has a crucial effect on many biological activities of colostrum proteins. The glycosylated components of these proteins avoid pathogenic infections by reducing the cell adhesion and internalization abilities of bacteria and viruses. Exploring the changes in glycosylation of colostrum proteins during lactation can help to understand the pathogenic inhibitory activity of these proteins. The pathogen–host cell interactions are the basic key to starting the infection, internalization and proliferation of pathogens. The colostrum contains higher amount of glycosylated components as compared to mature milk which decreases significantly in the first week of lactation. The colostrum glycoproteins have antiviral and antibacterial properties against many strains. The variation in glycosylation pattern with lactations could be utilized to modify these interactions between host cells and pathogens to avoid the infection to enhance the immune system. These interactions can also help to develop novel drugs to cure different infections and diseases.

Abstract:Background/Objectives: Human milk is the optimal source of nutrition and protection against infection for infants. If breastfeeding is not possible, standard and hydrolyzed infant formulas (IF) are an alternative. Extensively hydrolyzed IFs (eHFs) contain bioactive peptides, but their activities have rarely been evaluated. The aim of this study was to characterize and compare the bioactive peptide profiles of different eHFs and standard IFs before and after in vitro digestion. Methods: Two forms, liquid and powder, of intact protein formula (iPF) and eHF were subjected to in vitro gastrointestinal digestion, mimicking a young infant's gut (age 0–4 months) and an older infant's gut (>6 months). Bioactive peptides of in vitro digested and undigested formulas were analysed with Liquid Chromatography–Mass Spectrometry (LC–MS). Results: In all samples, a variety of peptides with potential bioactive properties were found. Immuno-regulatory peptides, followed by antimicrobial and antioxidative peptides were most frequent, as were peptides promoting wound healing, increasing mucin secretion, regulating cholesterol metabolism, and preventing bacterial infection. Peptides typically found in yoghurt and colostrum were identified in some formula samples. Conclusions: The high amounts of bioactive peptides with various properties in eHFs and iPFs indicate a possible contribution to infection protection, healthy gut microbiomes, and immunological development of infants. eHFs showed similar compositions of bioactive peptides to iPFs, with intermittently increased peptide variety and quantity.

Colostrum and milk are excellent sources of antibacterial components that help to strengthen the immunity of the offspring and accelerate the maturation of the immune system. It is possible to study these important defenses of milk and colostrum, such as lactoferrin, lysozyme, immunoglobulins, oligosaccharides, etc., as biotherapeutic agents for the prevention and treatment of numerous infections caused by microbes. Each of these components has different mechanisms and interactions in various places. The compound's mechanisms of action determine where the antibacterial activity appears. The activation of the antibacterial activity of milk and colostrum compounds can start in the infant's mouth during lactation and continue in the gastrointestinal regions. These antibacterial properties possess potential for therapeutic uses. In order to discover new perspectives and methods for the treatment of bacterial infections, additional investigations of the mechanisms of action and potential complexes are required.
