Abstract:Sugar beet ( Beta vulgaris L.) is an economically important crop, which contributes significantly to the global sugar supply. The present study was performed to establish and optimize the parameters of Agrobacterium -mediated genetic transformation with the sugar beet blade–petiole transition zone as explants to provide a reliable and efficient genetic transformation method for studies of sugar beet. This study used beet pollination line 32467. In the explants screening test, the differentiation rate of the explant blast–petiole transition zone was 10.8%, which was significantly higher than those of hypocotyl segments (6.7%) and multiple shoot clumps (4.2%). In the medium selection test, the main medium components used in the transformation process were as follows: induced differentiation medium: MS medium, 1.2 mg kinetin, 0.7 mg 1-naphthylacetic acid (pH 5.8); rooting medium: MS medium, 1.2 mg 1-naphthylacetic acid (pH 5.8); infection medium: MS medium, 0.7 mg 1-naphthylacetic acid, 1.2 mg kinetin, 29 mg acetosyringone (pH 6.8); co-cultivation medium: MS medium, 29 mg acetosyringone, 0.7 mg 1-naphthylacetic acid, 1.2 mg kinetin, 388 mg d -galacturonic acid (pH 5.8); delayed selection medium: MS medium, 0.5 mg benzylaminopurine, 0.05 mg 1-naphthaleneacetic acid, 500 mg carboxypenicillin, 100 mg cefotaxime (pH 5.8); and selection medium: MS medium, 0.5 mg benzylaminopurine, 0.05 mg 1-naphthaleneacetic acid, 500 mg carboxypenicillin, 100 mg cefotaxime, 0.013 mg glyphosate (pH 5.8). The results of Agrobacterium infection experiments showed that the optimal Agrobacterium infection concentration was OD 600 = 0.4 with an infection time of 20 minutes.

Abstract:Background: Common bean (Phaseolus vulgaris L.) is a globally important grain and vegetable legume crop, providing a substantial portion of the diet protein and minerals for many people in the developing world. However, the genetic studies and improvement on this crop has long been impeded by its recalcitrance to Agrobacterium-mediated whole plant genetic transformation. Established Agrobacterium rhizogenes-based hairy root transformation in common bean heavily relies on the strain K599. Methods: In order to develop an efficient alternative protocol for hair transformation in common bean, the efficiency of Agrobacterium rhizogenes strain R1000 in inducing hairy roots from 6-day-old seedlings with root below cotyledons excised by the soaking and smearing method were tested. The binary plasmid pBI121 with the reporter gene GUS (pBI121-GUS) or eGFP (pBI121-eGFP) driven by the constitutive promoter was used for transformation and rapid identification of the transgenic hairy roots. Result: We established a strain R1000-based system for the induction of hairy roots in common bean. The plant receptor genotypes and infection methods were optimized, which led to a high transformation rate of hairy roots up to 60%. This method therefore provides a useful alternative means for functional genomic studies in common bean.

Study on Transformation of Cotton Pollen Using Agrobacterium Tumefaciens

Abstract:The pollen-mediated transformation approach could not only circumvent the tedious tissue culture procedures like in Agrobacterium infection, but also circumvent the incertitude of the pollen tube path-way. So,its transformants combined from the transformed pollen and eggs would not be chimera.At last this approach is simple, easy to operate, and could be widely used in practice. For the transformation happened in the pollen culture medium, the solution with a suitable solute has to be selected. This study showed the medium consisting of (0.1%) H_3BO_3 、(0.3%) Ca(NO_3)_2、(0.2%) MgSO_4·7H_2O、(0.1%) KNO_3 and 45% sucrose could maintain normal turgidity of the pollens and facilitate them to be germinated. There are more than 95% pollens don't dispose in this medium and the germinating rate was up to 58.5%. A transient GUS expression assay and the exploded pollen rate proved that the transformation method with 1‰ Tween-20 in the medium using vacuum-infiltration was a better method. Finally, four transformed seedlings were confirmed by Kanamycin selection, GUS expression and Northern blot. Therefore this transformation approach is available.

Abstract:A protocol was developed for Agrobacterium tumefaciens-mediated transformation of creeping bentgrass [Agrostis stolonifera L. var. palustris (Huds) Farw]. The transformation was performed using the vector pCAMBIA 1301 which contains the reporter (uidA) gene and the selectable marker hygromycin phosphotransferase (hph) gene. Embryogenic calli initiated from mature seeds were infected with A. tumefaciens strain EHA105 followed by hygromycin selection. Effects of some variables influencing efficiency of T-DNA delivery and selection were examined, and the protocol was successfully employed in the production of transgenic creeping bentgrass. Southern blot analysis demonstrated the integration of a single or two copies of foreign gene, and RT-PCR confirmed the transcription of the genes.

Abstract:Plant regeneration and dedifferentiation are known to be difficult for sugar beet (Beta vulgaris L.) due to its recalcitrant nature. This leads to low transformation efficiency and thus successful application of plant molecular techniques is limited in sugar beet for its genetic enhancement. A prolific regeneration method has been established by modulating several plant growth regulators on the in vitro regeneration of Beta vulgaris var. V6 KWS Serenada. Several types of explants excised from young seedlings of this variety were used for both direct and indirect regeneration of shoots. The highest response towards direct shoot formation and callus induction were obtained from cotyledonary nodes and hypocotyls, respectively. Explants were cultured on MS medium supplemented with different concentrations of BAP, GA3 and 2, 4 -D for callus induction as well as formation of shoots. Hypocotyls responded well for callus induction on MS medium containing 0.1 mg/l BAP + 2.0 mg/l 2, 4-D, while cotyledonary nodes exhibited the highest responses towards shoot formation on MS medium containing 1.0 mg/l BAP and 1.5 mg/l GA3. MS medium containing 2.0 mg/l IBA produced the highest number of roots per shoot. The in vitro raised rooted plantlets were successfully transferred to soil for acclimatization. Abstract:Snapdragon (Antirrhinum majus L.) is a popular ornamental and model plant species, and the recently released reference genome could greatly boost its utilization in fundamental research. However, the lack of an efficient genetic transformation system is still a major limiting factor for its full application in genetic and molecular studies. In this study, a simple method for quick regeneration and efficient Agrobacterium-mediated transformation of snapdragon was developed. Cotyledon petiole and hypocotyl explants derived from two-week-old seedlings were cultured on MS media supplemented with 2 mg/L zeatin (ZT), 0.2 mg/L 1-naphthaleneacetic acid (NAA), and 2 mg/L AgNO3, and adventitious shoots were regenerated through organogenesis with an average regeneration of 48.00% and 41.33%, respectively. By contrast, the regeneration frequency was only 22.67% for cotyledon petiole and 25.67% for hypocotyl explants in the absence of AgNO3. Moreover, the application of AgNO3 promoted indirect shoot organogenesis, while direct shoot organogenesis occurred in the absence of AgNO3 from both hypocotyl or cotyledon petiole explants. Agrobacterium-mediated genetic transformation systems were developed with this high-efficient regeneration system. The transformation efficiency has been improved from 0 to 1% through the direct shoot organogenesis to 3 to 4% via the indirect shoot organogenesis. This efficient regeneration and genetic transformation method could be important for future use of snapdragon as a model plant to address some fundamental questions which are hard to be solved by using other model plant species, and to accelerate the breeding process through CRISPR/Cas9 genome editing.

Abstract:Somatic hybridization between different plants through protoplast fusion represent an efficient experimental approach to produce genetically transformed plant species. Electrofution of mesophyll protoplasts in sugar beet was occurred to overcome the barriers faced breeding program of this economically industrial crop Protoplasts were successfully isolated from leave's mesophyll of two varieties of sugar beet (Beta vulgaris L.). Various enzyme solutions were assessed for the cell wall degrading ability. They express different efficiency in isolation of mesophyll protoplasts of var. Baraka. The protoplasts yield was 18 × 104 cell ml-1 using the mixture consisting of 0.5% Cellulase RS, 1.0% Hemicellulase and 0.1% Pectolyase Y-23 with 13% mannitol. A total of 16 hrs. for cell wall digestion, and protoplast viability approached 93%. Protoplasts were isolated from leaf mesophyll of var. Carola using the same enzymatic mixtures. High protoplasts yield 20 × 104 cell ml-1 was obtained, requiring the same period 16 hrs. to approach viability 96%. The protoplasts were spherical in shape, varied in chloroplast distribution, having size ranged 12 – 52 μm. The present study succeeded in electrofusion between Baraka × Carola mesophyll protoplasts, producing somatic hybrid cells under conditions of 1MHz, 1000 Vcm-1, 2 pulses, 1.5 msec./pulse with fusion percent of 73%.

Abstract:An efficient protocol for the establishment of transgenic high hariy root tea plant(Camellia sinensis) system induction infected by Agrobacterium rhizogenes is reported using two wild type strains of A.rhizogenes,three different co-cultivation media and six different explants.The highest induction frequency was obtained from stem segments of aseptic seedlings infected by A.rhizogenes and co-cultivated on YMB solid media with 100 mmol/L AS for 2 d.The hairy roots grew rapidly on LG0 medium without phytohormone and produced a plenty of branches and root hairs.PCR confirmed that rolA,rolB and rolC have inserted into the genomic DNA of hairy root induced by wild type A.rhizogenes.Hairy roots were also induced from explants of tea plant infected by A.rhizogenes 15834 carrying pCAMBIA2301 binary vector harboring Bt gene,transgenic root were confirmed by PCR and GUS histochemical staining that the Bt exogenous gene was inserted and expressed.
Highly efficient induction of sugar beet plant regeneration

Abstract:A method for high-frequency in vitro regeneration of adventitious shoots from petiole explants of a tetraploid breeding line 405 of Beta vulgaris L. was established. Plant regeneration was obtained by direct bud formation from petioles. Explants were precultured in a medium containing MS inorganic salts supplemented with Gamborg's vitamins, named MSB. Adventitious shoots were developed when petiole explants excised from 20-30-day-old seedlings grown on MSB containing 0.3 mg/L naphthalene acetic acid (NAA), 1.0 mg/L 6-benzylaminopurine (BA), and were then cultured on MS with 0.3 mg/L NAA and 1.0 mg/L BA. Shoots rooted on half-strength MS medium containing 0.5-1.0 mg/L NAA. This procedure of the culture method may be very useful both for multiplication of sugar beet and for the introduction of foreign genes into Beta vulgaris. The proliferation rate of induced plantlets is as high as 51.3%.

Abstract:Radish (Raphanus sativus) is a rich source of glucosinolates (GSLs) and their hydrolytic products such as isothiocyanates (ITCs). GSLs and ITCs enhance plant defense responses to biotic and abiotic stresses and are health promoting effect in human. The branched-chain aminotransferase 4 (BCAT4) gene encode an enzyme catalyzing the deamination of methionine in the first step in the chain elongation of aliphatic GSL biosynthesis. Previously, plant transformation in radish has been successfully performed using several methods such as floral dipping, vacuum infiltration and sonic infiltration, protoplast transformation and microspore culture. However, the recalcitrant of regeneration in radish affects the transformation efficiency remain relatively low. Therefore, there is still a need to improve the transformation methods for radish. In this study, we used a simple method for the efficient transformation of radish using Agrobacterium tumefaciens strain GV3101 and tested it with the radish BCAT4 (RsBCAT4) transgene. The PCR, RT-qPCR, Southern blot, GFP fluorescence, and HPLC analyses were used to confirm the transgene integration. Positive correlations between the expression of RsBCAT4 and downstream genes (i.e., CYP79F1, CYP83A1, and GRS1) were also observed in selected T2 transgenic lines. RsBCAT4 transgenic lines exhibited significantly increased levels of aliphatic GSLs compared to the levels in wild type plants, particularly glucoraphasatin. This needle perforation technique is simple in plant transformation method significantly enhancing transformation efficiency in radish, which could be utilized for molecular breeding of radish to improve its traits.
Biology of Hepatocellular Carcinoma are Restrained through PI 3 K / AKt Signaling Pathways and Targeted by Tumor Infiltrating Macrophages

Abstract:To study the biology of hepatocellular carcinoma are restrained through PI3K/AKt signaling pathways and targeted by tumor infiltrating macrophages.Methods: Mononuclear cells were isolated from healthy adult peripheral blood by density gradient centrifugation and induced with IL 4 for selective activation of macrophagesin vitro.PI3K siRNA and AKT – siRNA were transfected to hepatocellular carcinoma cell line HepG2 respectively by lipofectermine 3000, and then cultivate with selective activation of macrophages (M2) for 48 hour. PI3K and Akt mRNA expression level in liver cancer HepG2 cells were detected with real-time fluorescent quantitative PCR. Results: After transfection, PI3K, and Akt mRNA level were significantly decreased. In vitro microenvironment of liver cancer, M2 can significantly increase levels of PI3K, and Akt mRNA levels in HepG2 cell lines, and can significantly promote the HepG2 cell proliferation ability (P< 0.05) after co-culture with M2 for 24 hours. Conclusion: Tumor associated macrophages can promote liver cancer cell proliferation through PI3K/Akt signaling pathway. If changing the microenvironment of liver cancer or specific inhibition of PI3K/Akt signaling pathway can also control the malignant biological behavior of tumors.
An Efficient Hairy Root Transformation Method for Common Bean based on Petiole Explants

Abstract:Background: Common bean (Phaseolus vulgaris L.) is a major food legume with high nutritional value and economic importance globally. A remarkable attribute of this plant is its propensity to generate adventitious roots from petioles of detached leaves when moisture is appropriate. This distinctive feature presents promising prospects for harnessing petioles in the facilitation of transgenic hairy root production. Methods: We achieved the successful induction of transgenic hairy roots from common bean petioles through the utilization of Agrobacterium rhizogenes strain K599. Our experimentation encompassed a diverse array of common bean varieties and leaves of varying ages. Subsequently, we refined and optimized the procedure for hairy root induction. Result: Our investigations have revealed that the most conducive conditions for hairy root transformation with K599 are attained using 5-day-old leaves from the cultivar ‘Honghuaqingjia’, exhibiting a remarkable induction efficiency of 59%. The usefulness of this system was demonstrated through a subcellular localization analysis of the transcription factor PvTCP2 protein in combination with GFP (Green Fluorescent Protein).

Abstract:The efficient procedure for Agrobacterium mediated transformation of Medicago truncatula cv. R 108 1 was developed. Using the advantage of recycling embryo formation of system for direct somatic embryogenesis promoted by TDZ, transgenic plantlets were obtained for a short period of 60 days. Transformation was performed with bacterial strain—LBA 4404, containing binary vector pBI 121 carries two genes—uid A gene exprecing β- glucoronidase (GUS) and npt II gene for resistance to kanamicin. Different parameters of the gene transfer—pre-treatment of the embryo clusters, density of the bacterial suspension, time for inoculation and co-cultivation, type of selection were optimized. For evaluation of the transformation efficiency, histochemical GUS assay was performed and glucoronidase activity was detected in embryos, plantlets and seedlings of T1 progeny. Transgenic nature of plantlets was confirmed by npt II-specific PCR amplification and Southern hybridization. In the progeny transgenes segregated in Mendelian manner.

Effective Agrobacterium-mediated transformation protocols for callus and roots of halophyte ice plant (Mesembryanthemum crystallinum)

Abstract:BackgroundIce plant (Mesembryanthemum crystallinum L.) is a model plant for studying salt-tolerant mechanisms in higher plants. Many salt stress-responsive ice plant genes have been identified with molecular and biochemical approaches. However, no further functional characterization of these genes in host plant due to lack of easy and effective transformation protocols.ResultsTo establish efficient transformation system of ice plants, three types of ice plant materials, hypocotyl-derived callus, aseptically-grown seedlings and pot-grown juvenile plants, were used to develop Agrobacterium-mediated transformation protocols. The highest transient transformation efficiency was with 5-day-old ice plant callus co-incubated with an Agrobacterium tumefaciens at 2.5 × 109 cells mL−1 for 48 h. The 3-day-old ice plant seedlings with root tip removed were successfully infected with A. tumefaciens or A. rhizogenes, and obtained 85% and 33–100% transient transformation rates, respectively. The transient transformation assays in ice plant callus and seedlings demonstrated that the concentrations of Agrobacteria, the durations of co-incubation time, and the plant growth stages were three important factors affecting the transient transformation efficiencies. Additionally, pot-grown juvenile plants were syringe-injected with two A. rhizogenes strains A8196 and NCPPB 1855, to establish transformed roots. After infections, ice plants were grown hydroponically and showed GUS expressions in transformed roots for 8 consecutive weeks.ConclusionsOur Agrobacterium-mediated transformation protocols utilized hypocotyl-derived callus and seedlings as plant materials, which can be easily obtained in large quantity. The average successful transient transformation rates were about 2.4–3.0% with callus and 33.3–100.0% with seedlings. We also developed a rapid and efficient protocol to generate transgenic roots by A. rhizogenes infections without laborious and challenging tissue culture techniques. This protocol to establish composite ice plant system demonstrates excellent improvements in efficiency, efficacy, and ease of use over previous ice plant transformation protocols. These Agrobacterium-mediated transformation protocols can be versatile and efficient tools for exploring gene functions at cellular and organ levels of ice plants.

Genetic transformation of Gentiana macrophylla with Agrobacterium rhizogenes: growth and production of secoiridoid glucoside gentiopicroside in transformed hairy root cultures

Abstract:Hairy root cultures of Gentiana macrophylla were established by infecting the different explants four Agrobacterium rhizogenes strains namely A(4)GUS, R1000, LBA 9402 and ATCC11325, and hairy root lines were established with A. rhizogenes strain R1000 in 1/2 MS + B(5) medium. Initially, 42 independent hairy root clones were maintained and seven clones belongs to different category were evaluated for growth, morphology, integration and expression of Ri T-DNA genes, and alkaloid contents in dry root samples. On the basis of total root elongation, lateral root density and biomass accumulation on solid media, hairy root clones were separated into three categories. PCR and Southern hybridization analysis revealed both left and right T-DNA integration in the root clones and RT-PCR analysis confirmed the expression of hairy root inducible gene. GUS assay was also performed to confirm the integration of left T-DNA. The accumulation of considerable amounts of the root-specific secoiridoid glucosides gentiopicroside was observed in GM1 (T +/L and T +/R) and the GM2 (T +/L and T -/R DNA) type clones in considerably higher amount whether as two T -/L but T +/R callus-type clones (GM3) accumulated much less or only very negligible amounts of gentiopicroside. Out of four media composition the 1/2 MS + B(5) vitamin media was found most suitable. We found that initial establishment of root cultures largely depends on root:media ratio. Maximum growth rate was recorded in 1:50 root:media ratio. The maximum biomass in terms of fresh weight (33-fold) was achieved in 1/2 MS + B(5) media composition after 35 days in comparison to sixfold increase in control. The biomass increase was most abundant maximum from 15 to 30 days. Influence of A. rhizogenes strains and Ri plasmid of hairy root induction, the possible role of the T(L)-DNA and T(R)-DNA genes on growth pattern of hairy root, initial root inoculum:media ratio and effect of media composition is discussed.
Agrobacterium rhizogenes-mediated Transformation and Hairy Root Regeneration of Malus baccata(L.) Borkh.

Abstract:To enhance the rooting ability of Malus baccata (L.)Borkh., hairy roots were induced from cuttings of Malus baccata (L.)Borkh. explants co-cultured with Agrobacterium rhizogenes (8196 and R1601). Shoots were regenerated from hairy roots. Different strains of A. rhizogenes, medium, time of co-cultivation and usage of acetosyringone (AS) were researched. It indicated that the highest rooting rate (88.89%)was obtained when explants were infected by the 8196 bacterium suspension with AS(100 μmol·L-1), co-cultured for 3 days and cultured on 1/4MS medium. Regeneration system was established by screening different plant growth regulator recombination. Adventitious shoots were regenerated from calli that were induced from hairy roots in medium with 2.0 mg·L-1 BA, 0.1 mg·L-1 IBA and 0.1 mg·L-1 GA3. Regeneration frequency was 3%. Rooting ability of regeneration shoots increased significantly. It is a very promising method for M. baccata (L.)Borkh. transformation.

Abstract:Agrobacterium rhizogenes is a soil bacteria with extensive infectivity, which can infect almost all dicotyledonous plants and a few monocotyledonous plants to induce root nodules. This is caused by the root-inducing plasmid, which contains genes responsible for the autonomous growth of root nodules and crown gall base synthesis. Structurally, it is similar to the tumor-inducing plasmid in that it mainly contains the Vir region, the T-DNA region, and the functional region of crown gall base synthesis. Its T-DNA is integrated into the nuclear genome of the plant with the assistance of Vir genes, causing hairy root disease in the host plant and the formation of hairy roots. The roots produced by Agrobacterium rhizogenes-infested plants are characterized by a fast growth rate, high degree of differentiation, physiological, biochemical, and genetic stability, and ease of manipulation and control. In particular, the hairy root system is an efficient and rapid research tool for plants that have no affinity for transformation by Agrobacterium rhizogenes and low transformation efficiency. The establishment of germinating root culture system for the production of secondary metabolites in the original plants through the genetic transformation of natural plants mediated by root-inducing plasmid in Agrobacterium rhizogenes has become a new technology combining plant genetic engineering and cell engineering. It has been widely used in a variety of plants for different molecular purposes, such as pathological analysis, gene function verification, and secondary metabolite research. Chimeric plants obtained by induction of Agrobacterium rhizogenes that can be expressed instantaneously and contemporarily are more rapidly obtained, compared to tissue culture and stably inheritable transgenic strains. In general, transgenic plants can be obtained in approximately one month.
An Efficient Root Transformation System for Recalcitrant Vicia sativa

Abstract:Common vetch (Vicia sativa) is a multi-purpose legume widely used in pasture and crop rotation systems. Vetch seeds have desirable nutritional characteristics and are often used to feed ruminant animals. Although transcriptomes are available for vetch, problems with genetic transformation and plant regeneration hinder functional gene studies in this legume species. Therefore, the aim of this study was to develop a simple, efficient and rapid hairy root transformation system for common vetch to facilitate functional gene analysis. At first, we infected the hypocotyls of 5-day-old in vitro or in vivo, soil-grown seedlings with Rhizobium rhizogenes K599 using a stabbing method and produced transgenic hairy roots after 24 days at 19 and 50% efficiency, respectively. We later improved the hairy root transformation in vitro by infecting different explants (seedling, hypocotyl-epicotyl, and shoot) with R. rhizogenes. We observed hairy root formation at the highest efficiency in shoot and hypocotyl-epicotyl explants with 100 and 93% efficiency, respectively. In both cases, an average of four hairy roots per explant were obtained, and about 73 and 91% of hairy roots from shoot and hypocotyl-epicotyl, respectively, showed stable expression of a co-transformed marker β-glucuronidase (GUS). In summary, we developed a rapid, highly efficient, hairy root transformation method by using R. rhizogenes on vetch explants, which could facilitate functional gene analysis in common vetch.
