Abstract:This study examines the involvement of GTP-binding proteins (Gps) in the regulation of Na+/H+ exchange and Ca2+ influx, which are increased in vascular smooth muscle cells from spontaneously hypertensive rats. Gp activity was modulated by fluoride, GTPgammaS, GDPbetaS, and antisense oligodeoxynucleotides complementary to conserved regions of the alpha- and beta-subunits of Gps (alpha-comm and beta-comm, respectively). Beta-adrenergic-induced Gs-mediated cAMP production was used as a positive control to estimate the efficiency of these compounds. Na+/H+ exchange, measured as ethylisopropyl amiloride-sensitive 22Na influx, was activated by 5- to 6-fold by a 30-minute preincubation of cells with 10 mmol/L NaF with a K0.5 for NaF of approximately 13 mmol/L. In contrast, no activation of 45Ca influx was observed under preincubation of vascular smooth muscle cells with NaF in Ca2+-free medium, whereas at [Ca2+]o >0.5 mmol/L, simultaneous addition of 45Ca and 10 mmol/L NaF led to sharply increased isotope uptake. NaF-induced 45Ca influx did not reach saturation up to 3 mmol/L [Ca2+]o and 20 mmol/L NaF and was correlated with the formation of calcium-fluoride complexes measured by light scattering. GTPgammaS increased basal cAMP production and Na+/H+ exchange, whereas GDPbetaS decreased isoproterenol-induced cAMP production and Na+/H+ exchange. Alpha-comm reduced whereas beta-comm augmented isoproterenol-induced cAMP production by 70%. Both oligodeoxynucleotides decreased basal Na+/H+ exchange by 40% to 50%. NaF-induced Na+/H+ exchange was not sensitive to alpha-comm but was inhibited by 60% in beta-comm-loaded cells. Neither basal nor NaF-induced 45Ca uptake was affected by GTPgammaS, GDPbetaS, and the oligodeoxynucleotides. Our results show that 45Ca uptake is activated by NaF in vascular smooth muscle cells by nonspecific accumulation of calcium-fluoride complexes and is not related to modification of Gps. On the contrary, the Na+/H+ exchanger is controlled by Gps, and Gp beta-subunits are involved in [Ca2+]o-independent activation of this carrier by NaF.
Exploiting the Cellular Actions of Skca and Ikca Channels to Manipulate Endothelial Function and Vascular Tone

Abstract:Growing evidence indicates that small‐ and intermediate‐conductance, Ca2+‐activated K+ channels (SKCa and IKCa channels, respectively) play pivotal roles in the cellular mechanisms contributing to stimulated vasorelaxation (i.e. nitric oxide (NO) synthesis, actions of EDHF). Using single cell patch clamp and fluorimetry, along with arteriolar pressure myography, we have obtained novel results demonstrating that activators of SKCa and IKCa channels (i.e. NS309 and DCEBIO, 0.01 ‐ 1 μM) enhance a number of cellular processes associated with stimulated vasorelaxation (i.e. cytosolic Ca2+ elevations, membrane hyperpolarization and NO synthesis). These observed effects of NS309 and DCEBIO were largely prevented by the SKCa and IKCa channel blockers apamin and TRAM‐34 in combination, but not when applied separately. In rat cremaster arterioles pressurized to 70 mmHg, NS309 and DCEBIO reduced myogenic tone and further augmented ACh‐induced vasodilation. The eNOS inhibitor L‐NAME (0.1 mM) decreased ACh‐evoked inhibition of myogenic tone, but did not affect the dilatory responses to NS309 and DCEBIO. The observed inhibition of NS309/DCEBIO action following endothelial denudation confirmed that these agents acted predominantly via the vascular endothelium. Finally, we will discuss initial efforts to discern the contribution of STIM1 to SKCa and IKCa activity and endothelial vasodilatory processes.
Development of Economical ASIC for PCS for water Quality Monitoring

Abstract:In this paper, the design of an ASIC is presented that implement a low-cost system for the supervision of water quality in urban areas or rivers. Photo catalytic sensor (PCS) estimates the parameter biological oxygen demand (BOD) which is generally used to estimate quality of water. The system proposed in this paper involves a simple potentiometric approach that provides a correlation in the input–output signals of low-cost sensors. This approach which is more users friendly and fast in operation is obtained by modeling and optimization of sensor for water quality monitoring. This is to overcome several drawbacks generally found in the previous flow injection analysis method of determining chemical oxygen demand (COD)-like complex designing, nonlinearity and long computation time. The system constitutes a significant cost reduction in the supervision of water quality monitoring. The main reason of employing a readout circuit to PCS circuitry, is the fact that the fluctuation of O2 influences the threshold voltage, which is internal parameter of the FET and can manifest itself as a voltage signal at output but as a function of the trans-conductance gain. The trans-conductance is a passive parameter and in order to derive voltage or current signal from its fluctuations the sensor has to be attached to readout circuit. This circuit provides high sensitivity to the changes in percentage of O2 in the solution. In this design simple potentiometric approach with few passive components are used to build a readout circuit. The paper focuses on the electronic implementation of the readout system for the PCS which optimize the circuit performance and increases reliability.
Small- and Intermediate-Conductance Ca2+-activated K+channels Directly Control Agonist-Evoked Nitric Oxide Synthesis in Human Vascular Endothelial Cells

Abstract:The contribution of small-conductance (SK(Ca)) and intermediate-conductance Ca(2+)-activated K(+) (IK(Ca)) channels to the generation of nitric oxide (NO) by Ca(2+)-mobilizing stimuli was investigated in human umbilical vein endothelial cells (HUVECs) by combining single-cell microfluorimetry with perforated patch-clamp recordings to monitor agonist-evoked NO synthesis, cytosolic Ca(2+) transients, and membrane hyperpolarization in real time. ATP or histamine evoked reproducible elevations in NO synthesis and cytosolic Ca(2+), as judged by 4-amino-5-methylamino-2',7'-difluorofluorescein (DAF-FM) and fluo-3 fluorescence, respectively, that were tightly associated with membrane hyperpolarizations. Whereas evoked NO synthesis was unaffected by either tetraethylammonium (10 mmol/l) or BaCl(2) (50 micromol/l) + ouabain (100 micromol/l), depleting intracellular Ca(2+) stores by thapsigargin or removing external Ca(2+) inhibited NO production, as did exposure to high (80 mmol/l) external KCl. Importantly, apamin and charybdotoxin (ChTx)/ triarylmethane (TRAM)-34, selective blockers SK(Ca) and IK(Ca) channels, respectively, abolished both stimulated NO synthesis and membrane hyperpolarization and decreased evoked Ca(2+) transients. Apamin and TRAM-34 also inhibited an agonist-induced outwardly rectifying current characteristic of SK(Ca) and IK(Ca) channels. Under voltage-clamp control, we further observed that the magnitude of agonist-induced NO production varied directly with the degree of membrane hyperpolarization. Mechanistically, our data indicate that SK(Ca) and IK(Ca) channel-mediated hyperpolarization represents a critical early event in agonist-evoked NO production by regulating the influx of Ca(2+) responsible for endothelial NO synthase activation. Moreover, it appears that the primary role of agonist-induced release of intracellular Ca(2+) stores is to trigger the opening of both K(Ca) channels along with Ca(2+) entry channels at the plasma membrane. Finally, the observed inhibition of stimulated NO synthesis by apamin and ChTx/TRAM-34 demonstrates that SK(Ca) and IK(Ca) channels are essential for NO-mediated vasorelaxation.

Abstract:Recent data have led us to hypothesize that selective activation of endothelial small- and/or intermediate-conductance, calcium-activated potassium channels (SKCa and IKCa channels, respectively) by the opener compounds NS309 and DCEBIO would augment stimulated nitric oxide (NO) synthesis and vasodilation in resistance arteries. Experimentally, ATP-evoked changes in membrane potential, cytosolic Ca2+, and NO synthesis were recorded by patch clamp and microfluorimetry in single human endothelial cells. Agonist-evoked inhibition of myogenic tone in isolated, pressurized arterioles from rat cremaster skeletal muscle was analyzed by video microscopy. NS309 and DCEBIO enhanced ATP-evoked membrane hyperpolarization and cytosolic Ca2+ transients, along with acute NO synthesis in isolated endothelial cells. The acetylcholine-mediated inhibition of myogenic tone (IC50 = 237 nM) was left-shifted in the presence of NS309 and DCEBIO (10, 100, and 1000 nM) to IC50 values of 101, 78, and 43 nM; endothelial denudation inhibited this drug effect. L-NAME attenuated the acetylcholine-induced inhibition of myogenic tone but did not interfere with NS309 and DCEBIO-evoked vasodilation. Collectively, our data demonstrate that drug-induced enhancement of endothelial SKCa and IKCa channel activities represents a novel cellular mechanism to increase vasodilation of small-resistance arterioles, thereby highlighting these channels as potential therapeutic targets in cardiovascular disease states associated with compromised NO signaling.-Sheng, J.S., Ella, S., Davis, M. Altered Vascular Reactivity And K-Atp Channel Currents In Vascular Smooth Muscle Cells From Deoxycorticosterone Acetate (Doca)-Salt Hypertensive Rats

Abstract:This study was designed to evaluate the contribution of ATP-dependent potassium (K-ATP) channels to the changes in vascular reactivity and spontaneous tone observed in vessels isolated from deoxycorticosterone acetate (DOCA)-salt hypertensive rats. In phenylephrine preconstricted aortic rings, cromakalim induced concentration-dependent, glibenclamide-sensitive relaxation. The concentration response curve to cromakalim was shifted to the right in DOCA-salt hypertensive rats (EC50: 0.850 +/- 0.100 muM) compared with SHAM-normotensive rats (0.108 +/- 0.005 muM), and the maximum relaxation (E-max) evoked by cromakalim was significantly lower in aortic rings from the DOCA group (68 +/- 2%) compared with the SHAM group (108 +/- 5%). The results were similar in endothelium-denuded rings. Spontaneous tone was observed in aortic rings (5 g preload) from DOCA-salt but not SHAM rats. Cromakalim abolished spontaneous tone and the effect was blocked by glibencamide. In whole cell patch clamp studies, increasing extracellular K+ concentrations from 5.4 to 140 mM and the administration of cromakalim evoked dramatic increases in K-ATP channel currents in aortic cells isolated from SHAM rats. In contrast, in aortic cells from DOCA-salt hypertensive rats, K-ATP channel currents were either absent or weak in response to challenges by elevated extracellular K+ and by cromakalim. These findings suggest that the function of K-ATP channels is impaired in smooth muscle cells from aorta of DOCA-salt hypertensive rats, which may contribute to the impaired vasodilatation and spontaneous tone observed in these rats.

Abstract:Cardiotonic steroids (CTS), used by certain insects, toads, and rats for protection from predators, became, thanks to Withering's trailblazing 1785 monograph, the mainstay of heart failure (HF) therapy. In the 1950s and 1960s, we learned that the CTS receptor was part of the sodium pump (NKA) and that the Na+/Ca2+ exchanger was critical for the acute cardiotonic effect of digoxin- and ouabain-related CTS. This "settled" view was upended by seven revolutionary observations. First, subnanomolar ouabain sometimes stimulates NKA while higher concentrations are invariably inhibitory. Second, endogenous ouabain (EO) was discovered in the human circulation. Third, in the DIG clinical trial, digoxin only marginally improved outcomes in patients with HF. Fourth, cloning of NKA in 1985 revealed multiple NKA α and β subunit isoforms that, in the rodent, differ in their sensitivities to CTS. Fifth, the NKA is a cation pump and a hormone receptor/signal transducer. EO binding to NKA activates, in a ligand- and cell-specific manner, several protein kinase and Ca2+-dependent signaling cascades that have widespread physiological effects and can contribute to hypertension and HF pathogenesis. Sixth, all CTS are not equivalent, e.g., ouabain induces hypertension in rodents while digoxin is antihypertensinogenic ("biased signaling"). Seventh, most common rodent hypertension models require a highly ouabain-sensitive α2 NKA and the elevated blood pressure is alleviated by EO immunoneutralization. These numerous phenomena are enabled by NKA's intricate structure. We have just begun to understand the endocrine role of the endogenous ligands and the broad impact of the ouabain-binding site on physiology and pathophysiology.

Abstract:Reduced smooth muscle (SM)‐specific α2 Na + pump expression elevates basal blood pressure (BP) and increases BP sensitivity to angiotensin II (Ang II) and dietary NaCl, whilst SM‐α2 overexpression lowers basal BP and decreases Ang II/salt sensitivity. Prolonged ouabain infusion induces hypertension in rodents, and ouabain‐resistant mutation of the α2 ouabain binding site (α2 R/R mice) confers resistance to several forms of hypertension. Pressure overload‐induced heart hypertrophy and failure are attenuated in cardio‐specific α2 knockout, cardio‐specific α2 overexpression and α2 R/R mice. We propose a unifying hypothesis that reconciles these apparently disparate findings: brain mechanisms, activated by Ang II and high NaCl, regulate sympathetic drive and a novel neurohumoral pathway mediated by both brain and circulating endogenous ouabain (EO). Circulating EO modulates ouabain‐sensitive α2 Na + pump activity and Ca 2+ transporter expression and, via Na + /Ca 2+ exchange, Ca 2+ homeostasis. Inhibition of the Na+/Mg2+ exchanger decreases blood pressure and modulates vascular ERK1/2 and p38 MAP kinase phosphorylation in Ang II-induced hypertensive rats

Abstract:Abnormal regulation of Mg2+ in vascular smooth muscle cells (VSMC) has been implicated to play a role in vascular dysfunction in hypertension. This has been attributed, in part, to increased activation of the Na+/Mg2+ exchanger. We investigated whether inhibition of the Na+/Mg2+ exchanger by quinidine and imipramine influences development of hypertension in Ang II-infused rats. 16 week-old Sprague Dawley rats were randomly divided into 4 groups (6/group): Control (vehicle), Ang II (Ang II, 150 ng/kg/min by osmotic pump), Ang II+quinidine (5 mg/kg/day), Ang II+imipramine (5 mg/kg/day). Rats were treated for 4 weeks. Systolic blood pressure (SBP) and serum cations were measured weekly. Rats were killed by decapitation. Kidneys and mesenteric arteries were collected for determination of MAP kinase expression. [Mg2+]i and [Na+]i were measured in platelets by Mag-fura and SBFI methodology respectively. Phosphorylation of ERK1/2, p38 MAP kinase and JNK was assessed by Western blots using phosphospecific antibodies. SBP increased significantly in the Ang II group (111±1 to 208±6 mmHg). In the quinidine and imipramine groups, SBP increased but the magnitude of increase was significantly less than the Ang II group (quinidine 173±6 mm Hg, imipramine 159±2 mmHg). Serum Mg2+, Ca2+, Na+ and K+ levels were not influenced by treatment. [Mg2+]i was significantly increased and [Na+]i was significantly decreased in the treated groups compared with Ang II group (p<0.01). Phosphorylation of ERK1/2, p38 MAP kinase and JNK was increased 3-5 fold in arteries and kidneys from Ang II rats compared with controls. Quinidine and imipramine significantly reduced (p<0.05) expression of ERK and p38 MAP kinase. Phosphorylation of JNK was not influenced by treatment. Our data demonstrate that quinidine and imipramine attenuate the development of hypertension in Ang II-infused rats. This is associated with increased [Mg2+]i and decreased [Na+]i and reduced phosphorylation of vascular and renal ERK1/2 and p38 MAP kinase. These findings suggest that the Na+/Mg2+ exchanger plays a role in Ang II-dependent hypertension.

Abstract:The effects of captopril (Cap) and verapamil (Ver) alone and in combination on intracellular Na+ concentration ([Na+]i) in cultured aortic smooth muscle cells (ASMC) of rabbits was evaluated by a direct measurement of [Na+]i with fluorescent dye sodium-binding benzofuran isophthalate (SBFI) combined with digital image. [Na+]i in resting cells was found to be 11.9 �� 0. 7 mmol/L. Angiotensin II (Ang-II, 0.1-10 ��mol/L) induced an increase of [Na+]i in concentration-dependent manner. Ver (0.1-10 ��mol/L) inhibited Ang-II (1 ��mol/L)-induced increase in [Na+]i, while Cap enhanced Ang-II-induced increase in [Na+]i at 10 ��mol/L but not at 0.1-1 ��mol/L. Ver (0.1-1 ��mol/L)abolished enhancement of Ang-II-induced increase in [Na+]i by Cap. Thus, the inhibition of Capenhanced [Na+]i by Ver may provide a new hypothesis for the underlying molecular mechanism of synergistic effect of the combination of Ca2+ antagonists and angiotensin-converting enzyme inhibitors in controlling blood pressure.

A New View of K + -Induced Contraction in Rat Aorta: the Role of Ca 2+ Binding

Abstract:Strong, K + -induced contractions of rat aorta in Ca-free, Mg-free media were not accompanied by increased intracellular calcium concentration, [Ca 2+ ] i , whereas such contractions in the presence of the divalent cations were correlated with rising [Ca 2+ ] i as assessed by fura-2. At the same time, calcium channel blockers, a modulator of Ca 2+ -binding proteins, and a modulator of actin polymerization, inhibited all types of K + -induced contractions. Increasing the K + in isotonic medium evoked a rise of 45 Ca 2+ binding to the plasma membrane of freshly isolated aortic cells. Although Ca 2+ -dependent events underlie the mechanism of K + -induced vascular contractions in both the presence and absence of Ca 2+ , in contrast to the view that [Ca 2+ ] i is a key regulator of excitation-contraction coupling in smooth muscle, we suggest that the modulation of Mg 2+ -dependent Ca 2+ binding, probably within/at the L-type calcium channel by K + , is a trigger for aortic contraction. This Ca 2+ binding may then activate actin-myosin interaction.
Comparison of susceptibility patterns using commercially available susceptibility testing methods performed on prevalent Candida spp.

Abstract:The rising rates of invasive fungal infections caused by non-albicans Candida and the increasing emergence of antifungal resistance complicate the management of invasive candidiasis. Accurate and timely antifungal susceptibility testing is critical to targeting antifungal therapy. The purpose of this study was to compare commercially available susceptibility testing methods using prospectively collected Candida isolates. Susceptibility testing was performed on 74 Candida isolates collected from July 2014 to March 2015 using broth microdilution according to the Clinical and Laboratory Standards Institute method, Etest, Vitek 2 (YS-05) and Sensititre. Essential agreement and categorical agreement (CA) were assessed using the reference method. Of the 34 total blood isolates collected, Candida albicans comprised only 38 % (13) of the Candida spp. with Candidaglabrata being nearly as prevalent (29 %, 10). CA using Etest was 86 % for fluconazole, 72 % for caspofungin, 98 % for micafungin and 97 % for anidulafungin. Vitek 2 CA was 90 % for fluconazole and 98 % for caspofungin. Sensititre CA was 93 % for fluconazole, 98 % for caspofungin, 98 % for micafungin and 100 % for anidulafungin. Although our study tested a small population of Candida isolates, our results were variable by method. When implementing antifungal susceptibility testing, clinicians should be aware of the strengths and limitations of each testing method.
Mechanisms of Sphingosine-1-phosphate-mediated Vasoconstriction of Rat Afferent Arterioles.

Abstract:AimSphingosine-1-phosphate (S1P) influences resistance vessel function and is implicated in renal pathological processes. Previous studies revealed that S1P evoked potent vasoconstriction of the pre-glomerular microvasculature, but the underlying mechanisms remain incompletely defined. We postulated that S1P-mediated pre-glomerular microvascular vasoconstriction involves activation of voltage-dependent L-type calcium channels (L-VDCC) and the rho/rho kinase pathway. MethodsAfferent arteriolar reactivity was assessed in vitro using the blood-perfused rat juxtamedullary nephron preparation, and diameter was measured during exposure to physiological and pharmacological agents. ResultsExogenous S1P (10(-9)-10(-5) mol L-1) evoked concentration-dependent vasoconstriction of afferent arterioles. Superfusion with nifedipine, a L-VDCC blocker, increased arteriolar diameter by 39 18% of baseline and significantly attenuated the S1P-induced vasoconstriction. Superfusion with the rho kinase inhibitor, Y-27632, increased diameter by 60 +/- 12% of baseline and also significantly blunted vasoconstriction by S1P. Combined nifedipine and Y-27632 treatment significantly inhibited S1P-induced vasoconstriction over the entire concentration range tested. In contrast, depletion of intracellular Ca2+ stores with the Ca2+-ATPase inhibitors, thapsigargin or cyclopiazonic acid, did not alter the S1P-mediated vasoconstrictor profile. Scavenging reactive oxygen species (ROS) or inhibition of nicotinamide adenine dinucleotide phosphate oxidase activity significantly attenuated S1P-mediated vasoconstriction. ConclusionExogenous S1P elicits potent vasoconstriction of rat afferent arterioles. These data also demonstrate that S1P-mediated pre-glomerular vasoconstriction involves activation of L-VDCC, the rho/rho kinase pathway and ROS. Mobilization of Ca2+ from intracellular stores is not required for S1P-mediated vasoconstriction. These studies reveal a potential role for S1P in the modulation of renal microvascular tone.

Abstract:Objective: The human radial artery has demonstrated superior long-term results as a graft in coronary bypass surgery, but undesirable post-surgical spasm limits its clinical application. Few have examined its excitatory properties, especially the underlying ion channel mechanisms. In this study, we investigated the kinetic and pharmacological properties of the smooth muscle membrane potassium currents of this important artery. Methods and results: Using whole cell patch-clamp techniques, we found the K+ current to be voltage-dependent and outwardly rectifying. Voltage-dependent inactivation was observed, being half-maximal at +28.0 mV but incomplete even at +40 mV. The K+ currents were predominantly sensitive to the K-Ca blocker tetraethylammonium (TEA; 63.9 +/- 12.1% inhibition, p < 0.05), less sensitive to the Kv blocker 4-aminopyridine (4-AP; 32.8 +/- 4.4% inhibition, p < 0.05), and the K-ATP blocker glibenclamide (28.7 +/- 8.5% inhibition), at -20 mV testing potential. Resting membrane potential was -52.0 +/- 6.8 mV (n=5), and suppression of K+ currents by TEA and iberiotoxin (IbTx) caused membrane depolarization. Western blot analysis with channel-specific antibodies confirmed the presence of K-Ca and Kv channel proteins. TEA evoked 20.7 +/- 9.9% of the contractile response to 60 mM KCl, whereas IbTx caused about 10% of the above response at 10(-7) M. The nitric oxide donor SNAP augmented membrane K+ currents in a concentration-dependent fashion; the augmentation was completely suppressed by TEA, but was relatively insensitive to the guanylate cyclase inhibitor ODQ. Conclusions: The radial artery manifests mainly Ca2+-dependent K+ currents at rest; this current is augmented by nitric oxide through a cGMP- and protein kinase G-independent action. The relatively depolarized membrane potential, as well as its muscular structure, predisposes the radial artery to spasm. Agents that activate the Ca2+-dependent K+ current could be of therapeutic value in preventing post-surgical vasospasm. (c) 2006 European Society of Cardiology. Published by Elsevier B.V. Mechanistic Distinction Between Activation and Inhibition of (Na++k+)-Atpase-mediated Ca2+ Influx in Cardiomyocytes

Abstract:(Na(+)+K(+))-ATPase (NKA) mediates positive inotropy in the heart. Extensive studies have demonstrated that the reverse-mode Na(+)/Ca(2+)-exchanger (NCX) plays a critical role in increasing intracellular Ca(2+) concentration through the inhibition of NKA-induced positive inotropy by cardiac glycosides. Little is known about the nature of the NCX functional mode in the activation of NKA-induced positive inotropy. Here, we examined the effect of an NKA activator SSA412 antibody on (45)Ca influx in isolated rat myocytes and found that KB-R7943, a NCX reverse-mode inhibitor, fails to inhibit the activation of NKA-induced (45)Ca influx, suggesting that the Ca(2+) influx via the reverse-mode NCX does not mediate this process. Nifedipine, an L-type Ca(2+) channel (LTCC) inhibitor, completely blocks the activation of NKA-induced (45)Ca influx, suggesting that the LTCC is responsible for the moderate increase in intracellular Ca(2+). In contrast, the inhibition of NKA by ouabain induces 4.7-fold (45)Ca influx compared with the condition of activation of NKA. Moreover, approximately 70% of ouabain-induced (45)Ca influx was obstructed by KB-R7943 and only 30% was impeded by nifedipine, indicating that both the LTCC and the NCX contribute to the rise in intracellular Ca(2+) and that the NCX reverse-mode is the major source for the (45)Ca influx induced by the inhibition of NKA. This study provides direct evidence to demonstrate that the activation of NKA-induced Ca(2+) increase is independent of the reverse-mode NCX and pinpoints a mechanistic distinction between the activation and inhibition of the NKA-mediated Ca(2+) influx path ways in cardiomyocytes.
In vivo regulation of Na/Ca exchanger expression by adrenergic effectors

Abstract:The Na/Ca exchanger encoded by the NCX1 gene plays an important role in calcium homeostasis in cardiac muscle. We previously identified three in vitro signaling pathways that are of major importance in the regulation of Na/Ca exchanger gene expression in neonatal cardiac myocytes, the protein kinase A (PKA) and protein kinase C (PKC) pathways, and intracellular Ca2+. To determine whether these pathways are important in vivo, we stimulated the PKA and PKC pathways and examined functional expression of the Na/Ca exchanger in adult rat heart. After a 3- and 7-day treatment, norepinephrine (200 mug . kg(-1) . h(-1)), isoproterenol (150 mug . kg(-1) . h(-1)), and phenylephrine (200 mug.kg(-1) . h(-1)) each stimulated a significant increase in NCX1 mRNA levels (35-85%, P< 0.05). Norepinephrine also stimulated a 35% increase in protein abundance (P< 0.05), a 20% decrease in relaxation duration (P< 0.05), and a 25% reduction in the fluorescence decay constant (P< 0.05) after a 7-day treatment. We conclude that a 7-day treatment of alpha- and beta -adrenergic agonists increases the expression of functional Na/Ca exchangers in adult rat heart.
Simultaneous Increases in Intracellular Sodium and Tonicity Boost Antimicrobial Activity of Macrophages

Abstract:Inflamed and infected tissues can display increased local sodium (Na+) levels, which can have various effects on immune cells. In macrophages, high salt (HS) leads to a Na+/Ca2+-exchanger 1 (NCX1)-dependent increase in intracellular Na+ levels. This results in augmented osmoprotective signaling and enhanced proinflammatory activation, such as enhanced expression of type 2 nitric oxide synthase and antimicrobial function. In this study, the role of elevated intracellular Na+ levels in macrophages was investigated. Therefore, the Na+/K+-ATPase (NKA) was pharmacologically inhibited with two cardiac glycosides (CGs), ouabain (OUA) and digoxin (DIG), to raise intracellular Na+ without increasing extracellular Na+ levels. Exposure to HS conditions and treatment with both inhibitors resulted in intracellular Na+ accumulation and subsequent phosphorylation of p38/MAPK. The CGs had different effects on intracellular Ca2+ and K+ compared to HS stimulation. Moreover, the osmoprotective transcription factor nuclear factor of activated T cells 5 (NFAT5) was not upregulated on RNA and protein levels upon OUA and DIG treatment. Accordingly, OUA and DIG did not boost nitric oxide (NO) production and showed heterogeneous effects toward eliminating intracellular bacteria. While HS environments cause hypertonic stress and ionic perturbations, cardiac glycosides only induce the latter. Cotreatment of macrophages with OUA and non-ionic osmolyte mannitol (MAN) partially mimicked the HS-boosted antimicrobial macrophage activity. These findings suggest that intracellular Na+ accumulation and hypertonic stress are required but not sufficient to mimic boosted macrophage function induced by increased extracellular sodium availability.
Resveratrol Stimulates the Na+–Ca2+ Exchanger on the Plasma Membrane to Reduce Cytosolic Ca2+ in Rat Aortic Smooth Muscle Cells

Abstract:<span> Resveratrol is well known to exhibit vascular relaxant and antihypertensive effects. In this study, we determined the effects of resveratrol on the modulation of cytosolic [Ca2+] level and adenosine 5′-triphosphate–induced Ca2+ release from the sarcoplasmic reticulum (SR) in rat aortic smooth muscle cells (ASMCs) and explored its underlying mechanisms. In this article, cytosolic [Ca2+] and SR [Ca2+] in ASMCs were determined by Fluo-4/acetoxymethyl and Mag-Fluo-4/acetoxymethyl respectively. Resveratrol (20, 50, and 100 µM) caused a rapid and substantial reduction in cytosolic [Ca2+] in ASMCs bathed in normal Hank's Balanced Salt Solution or Ca2+-free Hank's Balanced Salt Solution. Pretreatment with resveratrol reduced adenosine 5′-triphosphate–induced SR Ca2+ release and SR Ca2+ content. In the cells bathed in Na+-free physiological saline, which favors the reverse mode of the Na+–Ca2+ exchanger (NCX), resveratrol induced an increase in cytosolic [Ca2+] and SR [Ca2+]. However, its effect on cytosolic [Ca2+] was inhibited by the selective NCX inhibitor, SEA0400. Our findings suggest that resveratrol reduces cytosolic [Ca2+] and SR [Ca2+] in ASMCs in normal physiological saline, which might be, at least in part, mediated by the NCX.</span>

Abstract:Ca 2+ signals in smooth muscle cells (SMCs) contribute to vascular resistance and control blood pressure. Increased vascular resistance in hypertension has been attributed to impaired SMC Ca 2+ signaling mechanisms. In this regard, transient receptor potential vanilloid 4 (TRPV4 SMC ) ion channels are a crucial Ca 2+ entry pathway in SMCs. However, their role in blood pressure regulation has not been identified. We used SMC-specific TRPV4 −/− (TRPV4 SMC −/− ) mice to assess the role of TRPV4 SMC channels in blood pressure regulation. We determined the contribution of TRPV4 SMC channels to the constrictor effect of α1 adrenergic receptor (α1AR) stimulation and elevated intraluminal pressure: 2 main physiologic stimuli that constrict resistance-sized arteries. The contribution of spatially separated TRPV4 SMC channel subpopulations to elevated blood pressure in hypertension was evaluated in angiotensin II–infused mice and patients with hypertension. We provide first evidence that TRPV4 SMC channel activity elevates resting blood pressure in normal mice. α1AR stimulation activated TRPV4 SMC channels through PKCα (protein kinase Cα) signaling, which contributed significantly to vasoconstriction and blood pressure elevation. Intraluminal pressure–induced TRPV4 SMC channel activity opposed vasoconstriction through activation of Ca 2+ -sensitive K + (BK) channels, indicating functionally opposite pools of TRPV4 SMC channels. Superresolution imaging of SMCs revealed spatially separated α1AR:TRPV4 and TRPV4:BK nanodomains in SMCs. These data suggest that spatially separated α1AR–TRPV4 SMC and intraluminal pressure–TRPV4 SMC –BK channel signaling have opposite effects on blood pressure, with α1AR–TRPV4 SMC signaling dominating under resting conditions. Furthermore, in patients with hypertension and a mouse model of hypertension, constrictor α1AR–PKCα–TRPV4 signaling was upregulated, whereas dilator pressure–TRPV4–BK channel signaling was disrupted, thereby increasing vasoconstriction and elevating blood pressure. Our data identify novel smooth muscle Ca 2+ -signaling nanodomains that regulate blood pressure and demonstrate their impairment in hypertension.
