Abstract:[17-13C,3H]Gibberellin A4 (GA4) was injected into the shoots of tall (W23/L317), dwarf-1 (d1), and dwarf-5 (d5) Zea mays L. (maize); tall (cv Nipponbare), dwarf-x (dx), and dwarf-y (dy) Oryza sativa L. (rice); and tall (ecotype Landsberg erecta), ga4, and ga5 Arabidopsis thaliana (L.) Heynh. [13C]GA4 and its metabolites were identified from the shoots by full-scan gas chromatography-mass spectrometry and Kovats retention indices. GA4 was metabolized to GA1 in all nine genotypes. GA4 was also metabolized in some of the genotypes to 3-epi-GA1, GA2, 2[beta]-OH-GA2, 3-epi-GA2, endo-GA4, 16[alpha], 17-H2-16, 17-(OH)2-GA4, GA34, endo-GA34, GA58, 15-epi-GA63, GA71, and 16-epi-GA82. No evidence was found for the metabolism of GA4 to GA7 or of GA4 to GA3. The bioactivities of GA4 and GA1 were determined using the six dwarf mutants for assay. GA4 and GA1 had similar activities for the maize and rice mutants. For the Arabidopsis mutants, GA4 was more active than GA1 at low dosages; GA4 was less active than GA1 at higher dosages.

Abstract:The development of successful immunotherapeutic strategies requires the identification and characterisation of immunogenic cancer antigens that will be recognised by the host immune system, leading to tumour rejection. The concept of immunotherapy is based on the assumption that antigenic structures expressed in tumours can be used for therapeutic approaches employing the autologous immune system or by the application of immunotherapeutic reagents. Based on this concept, there is a great need to gain profound knowledge of the actual protein/antigen expression and its distribution pattern within normal tissues and cancerous tissues. Cancer testis (CT) antigens represent a unique class of tumour antigens, which are expressed in a variety of cancerous tissues and are silent in normal tissues, except for the testis. Owing to their restricted gene expression in the testis and various malignancies, CT antigens represent potential defined targets for antigen-based vaccination and antigen-directed immunotherapy to control cancer growth. Moreover, the analysis of humoral and cellular immune responses to CT antigens has proved useful for identifying novel cancer serum biomarkers with potential implications in early diagnosis of cancer.
Adenosine-elicited accumulation of adenosine 3', 5'-cyclic monophosphate in the chick embryo retina.

Abstract:The cyclic AMP level of 17-day-old chick embryo retina increased from 20 to 331 pmol/mg protein when the tissue was incubated for 20 min in the presence of 4-(3-butoxy-4-methoxybenzyl-2-imidozolinone) (RO 20-1724). The addition of 0.5 mM-3-isobutyl-1-methylxanthine (IBMX) or 0.5 units/ml of adenosine deaminase (EC 3.5.4.4) to the medium reduced the increase of cyclic AMP content from 20 to 100 pmol/mg protein. Dipyridamole did not interfere with the rise of the retinal cyclic AMP level observed with RO 20-1724. The EC50 of 6-amino-2-chloropurine riboside (2-chloroadenosine)-elicited accumulation of cyclic AMP of retinas incubated in the presence of RO 20-1724 plus adenosine deaminase was approximately 1 microM. When retina incubation was carried out in the presence of 0.5 mM-IBMX, the 2-chloroadenosine dose-response curve was shifted to the right two orders of magnitude. Maximal stimulation of the cyclic AMP level of 17-day-old chick embryo retina incubated in the presence of 0.5 mM-IBMX was observed at 1 mM-adenosine concentration. This effect was not blocked by dopamine antagonists. Guanosine and adenine did not affect the retinal cyclic AMP level. AMP and ATP had a slight stimulatory effect. Adenosine response of embryonic retina increased sharply from the 14th to the 17th embryonic day. A similar, but not identical adenosine effect was observed in cultured retina cells.

Abstract:Gibberellin A9(1) is metabolised by re-suspended cultures of Gibberella fujikuroi, mutant B1-41a, principally to gibberellins A20(17) and A40(18). Other metabolites, detected by g.l.c.–mass spectrometry include gibberellin A10(2) and A11(24); two didehydrogibberellin A9 derivatives (12) and (14); 16α,17-dihydro-16,17-dihydroxy-gibberellin A9(26); 3-epi-gibberellin A4(4); and a monohydroxygibberellin A9, possibly (19).Gibberellin A9 is partially metabolised by cultures of Rhizopus nigricans to give only gibberellin A10(2). Gibberellin A9 methyl ester, however, is converted into the methyl esters of the 16α- and 16β-epimers (26) and (27) of 16,17-dihydro-16,17-dihydroxygibberellin A9, gibberellin A20(17), gibberellin A40(18), gibberellin A45(20), and a monohydroxygibberellin A9, possibly (19).

Abstract:It has been almost a century since biologically active gibberellin (GA) was isolated. Here, we give a historical overview of the early efforts in establishing the GA biosynthesis and catabolism pathway, characterizing the enzymes for GA metabolism, and elucidating their corresponding genes. We then highlight more recent studies that have identified the GA receptors and early GA signaling components (DELLA repressors and F-box activators), determined the molecular mechanism of DELLA-mediated transcription reprogramming, and revealed how DELLAs integrate multiple signaling pathways to regulate plant vegetative and reproductive development in response to internal and external cues. Finally, we discuss the GA transporters and their roles in GA-mediated plant development.

Abstract:A series of novel C-3-OH substituted gibberellin derivatives bearing an amide group were designed and synthesized from the natural product gibberellic acid (GA₃). Their activities on the plant growth regulation of rice and Arabidopsis were evaluated in vivo. Among these compounds, 10d and 10f exhibited appreciable inhibitory activities on rice (48.6% at 100 μmol/L) and Arabidopsis (41.4% at 100 μmol/L), respectively. These results provide new insights into the design and synthesis of potential plant growth regulators.

Abstract:It has previously been demonstrated that a thiourea derivative, namely 1-phenethyl-3-[3-(trifluoromethyl)phenyl]thiourea (Y21), exhibits high gibberellin-like activity. In this work, the influence of the alkyl chain length and various benzene ring substituents on the biological activity of Y21 was investigated. 1-Substituted 3-[3-(trifluoromethyl)phenyl]thiourea derivatives were designed, synthesized, and comprehensively evaluated. The biological activity analysis demonstrated that at a dose of 0.1 μM, compound S8, i.e., 1-(3-bromophenethyl)-3-[3-(trifluoromethyl)phenyl]thiourea), exhibited significantly higher promoting activity with respect to Arabidopsis thaliana hypocotyl elongation and rice germination than did gibberellin A3 (GA3) and Y21. Moreover, the physiological response of the A. thaliana mutant ga1-1 to treatment with S8 suggested that this compound possessed GA-like function. The molecular docking analysis indicated that S8 formed hydrogen bonding interactions with residues Phe238 and Ser191 of the gibberellin insensitive DWARF1 (GID1) receptor, which resulted in stronger binding with GID1 than Y21. The bioactivity of derivatives S1–S22 exhibited a clear negative monotonic association with the molecular docking-based score values. Overall, it was determined that S8 displayed excellent GA-like bioactivity and facilitated hypocotyl elongation and germination; therefore, it could be applied as a potential plant growth regulator.

Abstract:Gibberellins (GAs) A17, A19, A20, A29, A44, 2βOH-GA44 (tentative) and GA29-catabolite were identified in 21-day-old seeds of Pisum sativum cv. Alaska (tall). These GAs are qualitatively similar to those in the dwarf cultivar Progress No. 9 with the exception of GA19 which does not accumulate in Progress seeds. There was no evidence for the presence of 3-hydroxylated GAs in 21 day-old Alaska seeds. Dark-grown shoots of the cultivar Alaska contein GA1, GA8, GA20, GA29, GA8-catabolite and GA29-catabolite. Dark-grown shoots of the cultivar Progress No.9 contain GA8, GA20, GA29 and GA29-catabolite, and the presence of GA1 was strongly indicated. Quantitation using GAs labelled with stable isotope showed the level of GA1 in dark-grown shoots of the two cultivars to be almost identical, whilst the levels of GA20, GA29 and GA29-catabolite were significantly lower in Alaska than in Progress No. 9. The levels of these GAs in dark-grown shoots were 10(2)- to 10(3)-fold less than the levels in developing seeds. The 2-epimer of GA29 is present in dark-grown-shoot extracts of both cultivars and is not thought to be an artefact.

Abstract:Gibberellic acid (GA), a plant hormone stimulating plant growth and development, is a tetracyclic di-terpenoid compound. GAs stimulate seed germination, trigger transitions from meristem to shoot growth, juvenile to adult leaf stage, vegetative to flowering, determines sex expression and grain development along with an interaction of different environmental factors viz., light, temperature and water. The major site of bioactive GA is stamens that influence male flower production and pedicel growth. However, this opens up the question of how female flowers regulate growth and development, since regulatory mechanisms/organs other than those in male flowers are mandatory. Although GAs are thought to act occasionally like paracrine signals do, it is still a mystery to understand the GA biosynthesis and its movement. It has not yet confirmed the appropriate site of bioactive GA in plants or which tissues targeted by bioactive GAs to initiate their action. Presently, it is a great challenge for scientific community to understand the appropriate mechanism of GA movement in plant's growth, floral development, sex expression, grain development and seed germination. The appropriate elucidation of GA transport mechanism is essential for the survival of plant species and successful crop production.

Identification and Characterisation of A Gibberellin-Related Dwarf Mutant in Pumpkin (cucurbita Moschata)

Abstract:'Non-vine I' is a dwarf variety of pumpkin (Cucurbita moschata) with short vines, while 'Yellow wolf' is a normal variety with long vines. The phenotypes of 'Non-vine I' and 'Yellow wolf' pumpkin were compared, and changes in the length of the vine in'Non-vine F were observed after treatment with gibberellin A(3) (GA(3)), naphthylacetic acid (NAA) or brassinolide (BL). Cell sizes in the internode in'Non-vine F were then observed and compared after application of 0.6 mM GA(3) or water (control). The results obtained showed that: (1) the morphological characteristics of 'Non-vine F plants were similar to normal pumpkins except for the vine trait; (2) 'Non-vine F plants were sensitive to exogenous GA(3) which significantly promoted the elongation of vines and petioles, but did not respond to exogenous auxin (NAA) or brassinolide (BL); (3) internode cells of 'Non-vine I' plants were shorter than those of normal pumpkins, and GA(3) promoted internode cell elongation in 'Non-vine I' plants. We propose that 'Non-vine I' is a GA-related mutant, and the dwarf trait results from the failure of normal internode cell elongation.

Abstract:Gibberellins are considered to play an important role in modulating plant–pathogen interactions. In order to find this, we studied the effect of GA4 on the interactions of Bipolaris sorokiniana (Scc.) (BIPOL) with Triticum aestivum. After inoculating the roots of hydroponically grown T. avestivum with BIPOL, the GA4 level shot up by 26%. The increased level of GA4 elicited a hypersensitive response (HR) in the T. aestivum seedlings. The elicitation in HR was confirmed through measurement of HR-inducing signaling compounds (i.e. c-di-GMP, cAMP, phosphatidic acid, pure and esterified oxo-phytodeinoic acid, and jasmonic acid). The concentration of HR-inducing signaling compounds in the treated T. aestivum seedlings was 31% higher compared to the control seedlings. Moreover, the induced HR response lowered the indole-3-acetic acid (IAA) level by 15% and the transzeatin (TZn) level by 22%. However, the application of 10 mM uniconazole through aerial spray inhibited GA4 biosynthesis in T. aestivum seedlings. Also, BIPOL colonized the roots of T. aestivum seedlings after uniconazole treatment and promoted seedling growth through increased relative growth rate (14%) and net assimilation rate (27%).

Abstract:Significance Growth hormones are mobile chemicals that exert considerable influence over how multicellular organisms like animals and plants take on their shape and form. Of particular interest is the distribution of such hormones across cells and tissues. In plants, one of these hormones, gibberellin (GA), is known to regulate cell multiplication and cell expansion to increase the rate at which roots grow. In this work, biosensor measurements were combined with theoretical models to elucidate the biochemical mechanisms that direct GA distribution and how these patterns relate to root growth. Our detailed understanding of how GA distributions are controlled in roots should prove a valuable model for understanding the makings of the many other hormone distributions that influence how plants grow.

Abstract:Gibberellins (GAs), a class of phytohormones, act as an essential natural regulator of plant growth and development. Many studies have shown that GA is related to rhizobial infection and nodule organogenesis in legume species. However, thus far, GA metabolism and signaling components are largely unknown in the model legume Medicago truncatula. In this study, a genome-wide analysis of GA metabolism and signaling genes was carried out. In total 29 components, including 8 MtGA20ox genes, 2 MtGA3ox genes, 13 MtGA2ox genes, 3 MtGID1 genes, and 3 MtDELLA genes were identified in M. truncatula genome. Expression profiles revealed that most members of MtGAox, MtGID1, and MtDELLA showed tissue-specific expression patterns. In addition, the GA biosynthesis and deactivation genes displayed a feedback regulation on GA treatment, respectively. Yeast two-hybrid assays showed that all the three MtGID1s interacted with MtDELLA1 and MtDELLA2, suggesting that the MtGID1s are functional GA receptors. More importantly, M. truncatula exhibited increased plant height and biomass by ectopic expression of the MtGA20ox1, suggesting that enhanced GA response has the potential for forage improvement.

Changes in peroxidase and IAA oxidase activities during cell elongation in Phaseolus hypocotyls

Abstract:Cytoplasmic and salt-extracted peroxidase and IAA oxidase activities were studied in Phaseolus vulgaris hypocotyls treated with gibberellic acid (GA, 200 µM), naphthyl acetic acid (NAA, 100 µM) and distilled water control (DW). Peroxidase activity was assayed with four hydrogen donors during the initial phase of hypocotyl elongation. Though peroxidase activity showed a decreasing trend with time in all the hydrogen donors studied; considerable variation with different hydrogen donors was observed. NAA had maximum peroxidase activity as compared to DW or GA treatment. The activity showed a clear inverse correlation with hypocotyl growth. IAA oxidase activity showed a similar trend with growth as peroxidase activity. A highly significant correlation was observed between peroxidase and IAA oxidase activities and high molecular weight xyloglucan content (P<0.001). Finally, the possible role of peroxidase and IAA oxidase activities in hypocotyl elongation growth is discussed.

Biosynthetic Origin of Gibberellins A(3) and A(7) in Cell-Free Preparations from Seeds of Marah macrocarpus and Malus domestica

Abstract:Cell-free preparations from seeds of Marah macrocarpus L. and Malus domestica L. catalyzed the conversion of gibberellin A(9) (GA(9)) and 2,3-dehydroGA(9) to GA(7); GA(9) was also metabolized to GA(4) in a branch pathway. The preparation from Marah seeds also metabolized GA(5) to GA(3) in high yield; GA(6) was a minor product and was not metabolized to GA(3). Using substrates stereospecifically labeled with deuterium, it was shown that the metabolism of GA(5) to GA(3) and of 2,3-dehydroGA(9) to GA(7) occurs with the loss of the 1beta-hydrogen. In cultures of Gibberella fujikuroi, mutant B1-41a, [1beta,2beta-(2)H(2)]GA(4), was metabolized to [1,2-(2)H(2)]GA(3) with the loss of the 1alpha- and 2alpha-hydrogens. These results provide further evidence that the biosynthetic origin of GA(3) and GA(7) in higher plants is different from that in the fungus Gibberella fujikuroi.

Abstract:The effect of gibberellins on tea leaf chemical composition and quality was investigated. The results showed that application of gibberellins was beneficial to green tea quality. Content of amino acids increased by 9.8% and that of vitamin C by 17.8% and tea catechins index increased by 12.9%. The content of tea polyphenols and ratio of tea polyphenols to amino acids decreased by 9.9% and 11.5%, respectively. Differences of all the indicators were statistically significant (P < 0.05). Quality scores of the treated leaf increased significantly (P < 0.05). It was also revealed that tea plant response to gibberellins was dependent not only on genetics, but also on shoot development stage.

Abstract:2',3'-Benzoabscisic acid 4a is significantly more active than (±)-ABA and can be potentially used as a plant growth regulator for agriculture. In this study, six 4a analogs were designed and synthesized. Bioassay showed that 4a displayed greater activity than (±)-ABA and the six analogs produced less inhibition than 4a itself. Specially, some analogs displayed markedly different activities to different physiological and biochemical process, which were largely different from ABA and 4a. Compared to (±)-ABA, 4b and 4c were more effective germination inhibitors for lettuce, but less effective inhibitors for rice elongation. Five-membered analog 5 was higher or slightly weaker in inhibiting Arabidopsis seed germination and rice elongation, respectively, but at least 10 times less effective than (±)-ABA in lettuce seed germination. Dual acid 6 and alkyne acid 20 nearly produced no inhibitory activity for Arabidopsis seed germination, but displayed excellent activity in inhibiting rice seedling growth. The preference of the analogs to different physiology process indicated that they might provide a strategy to develop novel ABA agonists or antagonist and be used as probe to investigate the function of different ABA receptors.

Abstract:The iso-abscisic acid (iso-ABA) analogs 11, 12, 13 and 14 were synthesized via regioselective addition of acetylides to enediones, selective reduction, Dess-Martin oxidation and Lindgren oxidation. The racemic mixtures 11 were resolved by chiral HPLC. These iso-ABA analogs and ABA were tested for their inhibitory activity in four bioassays. (+)-iso-ABA 11 exhibited high ABA-like activity, including the ability to effectively inhibit seed germination and seedling growth. The analogs 12, 13, 14 were also prepared and biological activities were investigated. Comparison of four bioassays, the iso-abscisic acid analogs had better activity in lettuce seed germination.

Abstract:The levels of endogenous gibberellin A1 (GA1), GA3, GA4, GA9 and a cellulase-hydrolysable GA9-conjugate in needles and shoot stems of Sitka spruce [Picea sitchensis (Bong.) Carr.] grafts with different coning or flowering histories were estimated by combined gas chromatography-mass spectrometry selected ion monitoring using deuterated GA3, GA4 and GA9 as internal standards. The samples were taken at the approximate time of the start of flower-bud differentiation, i.e. when the shoots had elongated approx. 95% of the final length. The needles of the good-flowering clones contained 11-12 ng per g fresh weight (FW) and 15-28 ng· (g FW) (-1) of GA9-conjugate and GA9, respectively. The shoot stems of the same material contained no detectable amounts of GA9-conjugate and 11-15 ng-(g FW)(-1) of GA9. The amounts of GA9-conjugate and GA9 were apparently lower in the poor-flowering clones, the needles containing 4-9 ng-(g FW)(-1) and 7-17 ng·(g FW)(-1), respectively. Also in this material the shoot stems contained no detectable amounts of GA9-conjugate. The amounts of GA4 were very small in both materials, ranging from 1-1.6 ng-(g FW)(-1). The good-flowering clones contained no detectable amounts of the more polar gibberellins, GA1 and GA3. The poor-flowering clones, on the other hand, contained high levels of GA15 17-19ng·(gFW)(-1) in the needles and 10-13 ng·(g FW) (-1) in the shoot stems, and also smaller amounts of GA3, 2-3 ng·(g FW)(-1) in the needles and approx. 1 ng·(g FW)(-1) in the shoot stems. The results demonstrate differences in GA-metabolism between the poor- and the good-flowering clones. The higher amounts of GA9-conjugate and GA9 might indicate a higher capacity for synthesizing GA4 in the good-flowering material. This synthesis does not, however, result in a build-up of the GA4-pool, maybe because of a high rate of turnover. Gibberellin A4 was apparently neither hydroxylated to GA1 nor converted to GA3 in the goodflowering material, as was the case in the poor-flowering material. This might indicate that gibberellin metabolism in the poor-flowering material is directed towards GA1 and GA3, GAs preferentially used in vegetative growth.

Abstract:The global agricultural system has been badly affected by adverse environmental changes in the past few years. These changes, including the rise of abiotic and biotic stressors negatively altered the growth and physiology of crop plants. Abiotic stresses, such as salinity, temperature extremes, drought, and heavy metals/metalloids, are major environmental constraints limiting crop growth, productivity, and sustainability worldwide. These stresses adversely affect plant metabolic activities and redox homeostasis, eventually leading to a reduction in plant growth and development. Plant growth regulators (PGRs) play a key role in regulating plants developmental processes and defensive responses under adverse environmental conditions. Among PGRs, gibberellic acid ( GA 3 ), an endogenous tetracyclic diterpenoid plant hormone, regulates many growth and developmental aspects of crop plants. GA 3 plays a pivotal role in mitigating abiotic stresses induced-perturbations in plants by modulating various physio-biochemical and molecular processes. Based on recent reports, this review article describes the role of exogenously applied GA 3 in improving seed germination, phenotypic characteristics, metabolic processes, yield and quality components, and post-harvest life of fruits, vegetables, and flowers. In this article, we summarize research concerning GA 3 biosynthesis and signaling and discuss the potential role of GA 3 in mediating tolerance to various abiotic stresses. Moreover, the present article enlightens the current research concerning the signaling pathway in gibberellin and gibberellin-mediated crosstalk with other plant hormones.
