Abstract:In recent years, research on nanoparticles (NPs) has brought a new phase in the development of medicine. The term "NPs" refers to nanoscale particles, typically between 10 and 1,000 nm in size, that are within the range of cellular and molecular structures. Compared with traditional ophthalmic drugs, NPs can carry drugs to overcome obstacles, target drug delivery, prolong drug release, reduce drug toxicity, and can be used in gene therapy. After a brief introduction to the classification of NPs, we will focus on the potential applications of different NPs in the diagnosis and treatment of different retinal diseases and introduce innovative methods derived from NPs.

Abstract:Near-infrared (NIR) imaging technology has been widely used for biomedical research and applications, since it can achieve deep penetration in biological tissues due to less absorption and scattering of NIR light. In our research, polymer nanoparticles with NIR fluorophores doped were synthesized. The morphology, absorption/emission features and chemical stability of the fluorescent nanoparticles were characterized, separately. NIR fluorescent nanoparticles were then utilized as bright optical probes for macro in vivo imaging of mice, including sentinel lymph node (SLN) mapping, as well as distribution and excretion monitoring of nanoparticles in animal body. Furthermore, we applied the NIR fluorescent nanoparticles in in vivo microscopic bioimaging via a confocal microscope. Under the 635 nm-CW excitation, the blood vessel architecture in the ear and the brain of mice, which were administered with nanoparticles, was visualized very clearly. The imaging depth of our one-photon microscopy, which was assisted with NIR fluorescent nanoprobes, can reach as deep as 500 μm. Our experiments show that NIR fluorescent nanoparticles have great potentials in various deep-tissue imaging applications.

Abstract:The employment of nanometric optical probes is powerful for bioimaging, disease diagnosis and therapy, es-pecially for the deep understanding of some dynamic biological processes at the molecular level. Optical probes such as gold nanorods (GNRs), upconversion nanoparticles (UCNPs) and graphene oxide (GO) can overcome many draw-backs of conventional optical agents. These nanoparticles possess high imaging contrast, stability and biocompatibility. Furthermor, they provide optical approaches for deep-tissue imaging and real-time dynamic imaging. In this review pa-per, the optical properties and advantages of these nanoparticles are briefly introduced, and some of their applications in optical bioimaging, diagnosis and therapy are summarized based on some of the research progress made by the authors and others. The prospects of their application are also discussed.

Abstract:Fluorescent imaging based on near-infrared (NIR) fluorophores has revolutionized the techniques employed for detecting biological events in depth owing to their advantages referring to diminished photon scattering, high signal-to-noise ratio and better light transparence through tissue. As for conventional luminogens, the nanofabrication of those innately hydrophobic π-conjugated architectures into water-dispersible nanoparticles (NPs) may result in attenuated fluorescent intensity deriving from the detrimental distribution of π-π interactions in the confined space. Oppositely, chromophores possessing aggregation-induced emission (AIE) characteristics emit boosted brightness at aggregate level according to the mechanism of restriction of intramolecular motion (RIM). In this review, we summarize the recent progresses of NIR emissive AIE NPs for multifarious biomedical applications from the viewpoint of different fabricated manners, mainly covering self-assembly and matrices assisted approaches. Furthermore, the current challenges and future research directions of NIR AIE NPs are briefly discussed.

Abstract:As the population ages, cancer has become a major health concern. Although surgical treatment, chemoradiotherapy, and other clinical treatments have become dominant, their strong invasiveness and intolerable toxic side effects seriously affect patients' quality of life. Therefore, developing non-invasive optical therapy with high safety is of great significance. In the last two decades of development, great progress has been made in lanthanide doped nanoparticles on biomedical applications under near-infrared (NIR) light due to their unique properties. These nanomaterials have been used in a wide range of applications, including bioluminescence marker detection, in vitro and in vivo imaging, photodynamic therapy (PDT), photothermal therapy (PTT), and other therapeutic applications. These therapies have been rapidly developed in the profession involving oncology due to their non-invasive, spatiotemporal controllability, and high efficiency. For this key review, the applications based on lanthanide nanoparticles have been listed as follows: bioimaging, biosensors, PDT and PTT. Some emerging biological applications of lanthanide-doped nanoparticles currently at the forefront of the nano/biological interface are also discussed. Finally, this review summarizes the future opportunities and challenges of nanotechnology research based on lanthanide-doped nanoparticles.

Abstract:Traditional wavelengths (400–700 nm) have made tremendous inroads in vivo fluorescence imaging. However, the ability of visible light photon penetration hampered the bio-applications. With reduced photon scattering, minimal tissue absorption and negligible autofluorescence properties, near-infrared light (NIR 700–1700 nm) demonstrates better resolution, high signal-to-background ratios, and deep tissue penetration capability, which will be of great significance for in-vivo determination in deep tissue. In this review, we summarized the latest novel NIR inorganic nanomaterials and the emission mechanism including single-walled carbon nanotubes, rare-earth nanoparticles, quantum dots, metal nanomaterials. Subsequently, the recent progress of precise noninvasive diagnosis in biomedicine and cancer therapy utilizing near-infrared inorganic nanomaterials are discussed. In addition, this review will highlight the concerns, challenges and future directions of near-infrared light utilization.

Abstract:Fluorescence imaging signal is severely limited by the quantum efficiency and emission wavelength. To overcome these challenges, novel NIR-emitting K5NdLi2F10 nanoparticles under NIR excitation was introduced as fluorescence imaging probe for the first time. The photostability of K5NdLi2F10 nanoparticles in the water, phosphate buffer saline, fetal bovine serum and living mice was investigated. The fluorescence signal was detected with depths of 3.5 and 2.0 cm in phantom and pork tissue, respectively. Fluorescence spectrum with a significant signal-to-background ratio of 10:1 was captured in living mice. Moreover, clear NIR images were virtualized for the living mice after intravenous injection. The imaging ability of nanoparticles in tumor-beard mice were evaluated, the enrichment of K5NdLi2F10 nanoparticles in tumor site due to the enhanced permeability and retention effect was confirmed. The systematic studies of toxicity, bio-distribution and in-vivo dynamic imaging suggest that these materials give high biocompatibility and low toxicity. These NIR-emitting nanoparticles with high quantum efficiency, high penetration and low toxicity might facilitate tumor identification in deep tissues more sensitively.

Abstract:This article discusses optically active nanomaterials and their optical biosensing applications. In addition to enhancing their sensitivity, these nanomaterials also increase their biocompatibility. For this reason, nanomaterials, particularly those based on their chemical compositions, such as carbon-based nanomaterials, inorganic-based nanomaterials, organic-based nanomaterials, and composite-based nanomaterials for biosensing applications are investigated thoroughly. These nanomaterials are used extensively in the field of fiber optic biosensing to improve response time, detection limit, and nature of specificity. Consequently, this article describes contemporary and application-based research that will be of great use to researchers in the nanomaterial-based optical sensing field. The difficulties encountered during the synthesis, characterization, and application of nanomaterials are also enumerated, and their future prospects are outlined for the reader's benefit.

Abstract:The introduction of nanometric contrast agents to optical imaging is helpful for the understanding of some biological processes at the molecular level, as well as the development of diagnostic tools and therapies. Optical imaging agents such as gold nanorods (GNRs), quantum dots (QDs), and organically modified silica (ORMOSIL) nanoparticles can overcome many drawbacks of conventional agents, such as poor contrast, photobleaching, and low chemical and optical stability in biological environment. These nanoparticles can also be developed for absorbance, emission, and scattering in the near-IR region, which allows optical approaches for deep-tissue real-time imaging. The synthesis methods and optical properties of GNRs, QDs, and ORMOSIL nanoparticles are briefly introduced, and some of their applications in optical bioimaging are demonstrated. Specific targeting, “green” synthesis methods, and optical signal demodulation are also introduced.

Abstract:Intrinsic shortcomings associated with conventional therapeutic strategies often compromise treatment efficacy in clinical ophthalmology, prompting the rapid development of versatile alternatives for satisfactory diagnostics and therapeutics. Given advances in material science, nanochemistry, and nanobiotechnology, a broad spectrum of functional nanosystems has been explored to satisfy the extensive requirements of ophthalmologic applications. In the present review, the recent progress in nanosystems, both conventional and emerging nanomaterials in ophthalmology from state-of-the-art studies, are comprehensively examined and the role of their fundamental physicochemical properties in bioavailability, tissue penetration, biodistribution, and elimination after interacting with the ophthalmologic microenvironment emphasized. Furthermore, along with the development of surface engineering of nanomaterials, emerging theranostic methodologies are promoted as potential alternatives for multipurpose ocular applications, such as emerging biomimetic ophthalmology (e.g., smart electrochemical eye), thus provoking a holistic review of "ocular nanomedicine." By affording insight into challenges encountered by ocular nanomedicine and further highlighting the direction of future studies, this review provides an incentive for enriching ocular nanomedicine-based fundamental research and future clinical translation.

Abstract:Optical diagnosis and treatment have paved the way for real-time, invasive, and efficient diagnosis and treatment of tumors. Functional nanomaterials, with their exceptional and precisely controllable optical properties, hold immense promise for various applications, including early tumor detection, interventional therapy, and prognosis prediction. In this paper, we present a comprehensive review of ongoing research endeavors focused on the development of functional inorganic nanomaterials for cancer detection, biosensing, and phototherapy. The integration of multifunctional inorganic nanomaterials with desirable optical properties has the potential to enable simultaneous achievement of early tumor diagnosis, real-time monitoring, and image-guided treatment. These nanomaterials possess tunable physical properties, allowing for innovative research directions and solutions in the diagnosis and treatment of major clinical diseases.

Abstract:The second near-infrared(NIR-Ⅱ,1000-1700 nm) window provides a superior optical platform with high resolution,deep penetration and high signal-to-noise ratios(SNRs),which results from the intrinsic low scattering and auto fluorescence in biological tissues.As one of the promising NIR-Ⅱ emitting probes,lanthanide based nanoparticles(LnNPs) exhibit high photo stability and chemostability,long photoluminescence lifetimes,low long-term cytotoxicity and narrow emission bandwidths.All these merits have spurred the evolution of related bio-optics and a variety of biomedical applications of LnNPs.This mini-review discusses the most recent advances in both the design-the composition and surface modifications-and the applications of NIR-Ⅱ emitting LnNPs in bioimaging,disease diagnosis and therapy.We also summarize the current limits and challenges facing the applications of LnNPs as well as discuss the directions of future development.

Abstract:Nanotechnology has become more and more potentially used in diagnosis or treatment of diseases. Advances in nanotechnology have led to new and improved nanomaterials in biomedical applications. Common nanomaterials applicable in biomedical applications include liposomes, polymeric micelles, graphene, carbon nanotubes, quantum dots, ferroferric oxide nanoparticles, gold nanoparticles (Au NPs), and so on. Among them, Au NPs have been considered as the most interesting nanomaterial because of its unique optical, electronic, sensing and biochemical properties. Au NPs have been potentially applied for medical imaging, drug delivery, and tumor therapy in the early detection, diagnosis, and treatment of diseases. Abstract:Au nanoparticles (NPs) possess unique physicochemical and optical properties, showing great potential in biomedical applications. Diagnostic spectroscopy utilizing varied Au NPs has become a precision tool of in vitro and in vivo diagnostic for cancer and other specific diseases. In this review, we tried to comprehensively introduce the remarkable optical properties of Au NPs, including localized surfaces plasmon resonance (LSPR), surface-enhanced Raman scattering (SERS), and metal-enhanced fluorescence (MEF). Then, we highlighted the excellent works using Au NPs for optical diagnostic applications. Ultimately, the challenges and future perspective of using Au NPs for optical diagnostic were discussed.

Abstract:The complexity of the human eye structure poses challenges in the diagnosis and treatment of ocular diseases, especially the low efficiency of ocular drug delivery. With the continuous advancements in nanomaterials, various ocular nano-drug delivery systems with extended drug delivery, long-term controlled release and targeted delivery have been developed to improve the flaws in traditional ophthalmology diagnosis and treatment. With many features, these novel nanomaterials are expected to provide controllability, precision, and individualization when treating eye disorders. In this review, the focus has been on various applications of nanomaterials in ophthalmic diseases from a clinical perspective, including abnormalities in eyelid, orbit, ocular surface, lens, vitreous body, uvea and fundus. We elaborate the advances of the emerging nanomaterials in ophthalmology in the past five years and summarize their prospects, providing new insights into the disease-specific development and clinical translation of ophthalmic nanomaterials.

Abstract:The last decade has witnessed the growing development of optical imaging using near-infrared (NIR) light (wavelength range: 700–1700 nm) in a wide range of applications, from fundamental research of live biological and physiological processes to clinical applications such as disease screening and image-guide surgical interventions. The low-energy NIR light offers reduced tissue scattering, absorption as well as autofluorescence background compared to traditional wavelengths in the visible (400–700 nm) range, thus dramatically enhancing imaging resolution, signal-to-noise ratio, and penetration depth. This Research Topic calls for advances in new fluorescent probes, imaging instrumentation, analytical methods and their applications for improvement of diagnosis and therapy in this “tissue transparent window,” aiming to provide a timely update of this continuously evolving filed. Imaging and diagnosis agents are the cornerstone of preclinical applications. In their study, Tan et al. report lycosin-I peptide modified gold nanoclusters (LGNCs), which exhibit strong NIR fluorescence, for imaging of specific cancer cell nuclei. The lycosin-I peptide not only possesses highly efficient and selective intracellular translocation against several cancer cells, but also induces aggregation of gold nanoclusters to further enhance their brightness. After uptake by tumor cells, the LGNCs aggregates can be depolymerized into ultrasmall nanoclusters by the high level GSH in the tumor microenvironment, facilitating nuclear targeting translocation in cancer cells. Another study by Huang et al. reports a novel biomimetic nanoprobe for photoacoustic (PA) imaging of cervical carcinoma. This probe is constructed by coating indocyanine green (ICG)-loaded mesoporous silica nanoparticles with the cancer cell membrane, providing small particle size, excellent dispersion, large loading efficiency, good biocompatibility and homologous targeting ability to Hela cells in vitro. The biomimetic nanoprobe can specifically accumulate into tumor and display a superior PA imaging efficacy compared with free ICG, allowing for non-invasive deep tissue imaging of cervical carcinoma in vivo. Another article by Gu et al. focuses on a particular type of NIR fluorophores, semiconducting polymer nanoparticles (SPNs). SPNs are constructed by encapsulating hydrophobic semiconducting polymers (SPs) with amphiphilic copolymers, and have shown great promise for phototheranostics of cancer. The authors review recent advances of thiadiazoloquinoxaline (TQ)-based SPNs from their synthesis and hydrophilic functionalization to NIR-II (1,000–1,700 nm) fluorescence imagingguided cancer photothermal therapy (PTT). To enhance the PTT efficiency, they also discuss methods for constructing SPNs with capability of multimodal imaging. Current limitations are summarized at the end that hamper clinical applications of the SPNs, such as relatively large size and low tumor targeting ability. Abstract:This study explores and reviews systemic nanoparticle delivery barriers and highlights nanoparticle design and biological modulation strategies to mitigate nanoparticle clearance by biological systems. A literature analysis systematically evaluating the in vivo performance of current nanoparticle design strategies is conducted to clarify how nanoparticle physicochemical properties affect their pharmacokinetics and biodistribution in preclinical mouse models. Most nanomedicines require efficient in vivo delivery to elicit meaningful diagnostic and therapeutic effects. However, en route to their intended tissues, systemically administered nanoparticles often encounter delivery barriers. To describe these barriers, the term "nanoparticle blood removal pathways" (NBRP) is proposed, which summarizes the interactions between nanoparticles and the body's various cell‐dependent and cell‐independent blood clearance mechanisms. Nanoparticle design and biological modulation strategies are reviewed to mitigate nanoparticle‐NBRP interactions. As these interactions affect nanoparticle delivery, the preclinical literature from 2011–2021 is studied, and the nanoparticle blood circulation and organ biodistribution data are analyzed. The findings reveal that nanoparticle surface chemistry affects the in vivo behavior more than other nanoparticle design parameters. Combinatory biological‐PEG surface modification improves the blood area under the curve by ≈418%, with a decrease in liver accumulation of up to 47%.
