Abstract:Bacterial membrane lipids have been traditionally defined as fatty acids (FAs) bilayers linked through ester bonds, while those of Archaea as ether-linked isoprenoids forming bilayers or monolayers of membrane spanning lipids (MSLs) known as isoprenoidal glycerol dialkyl glycerol tetraethers (isoGDGTs). This paradigm has been challenged with the discovery of branched GDGTs (brGDGTs), membrane spanning ether-bound branched alkyl FAs, that are of bacterial origin but whose specific producers in the environment are often unknown. The limited number of available microbial cultures restricts the knowledge of the biological sources of membrane lipids, which in turn limits their potential applicability as biomarkers. To address this limitation, we detected membrane lipids in the Black Sea using high resolution accurate mass/mass spectrometry and inferred their potential producers by targeting lipid biosynthetic pathways encoded on the metagenome, in metagenome-assembled genomes and unbinned scaffolds. We also detected brGDGTs and overly branched GDGTs in the suboxic and euxinic waters, which are potentially attributed, to members of the Planctomycetota, Cloacimonadota, Desulfobacterota, Chloroflexota, Actinobacteria and Myxococcota, all anaerobic microorganisms. These results open a new chapter in the use of specific brGDGTs as biomarkers of anoxic conditions in marine settings and of the role of these membrane lipids in microbial adaptation.

Abstract:The composition of membrane lipids varies in a number of ways as adjustment to growth conditions. Variations in head group composition and carbon skeleton and degree of unsaturation of glycerol-bound acyl or alkyl groups results in a high structural complexity of the lipidome of bacterial cells. Three CDL synthases were identified; one catalyzes the condensation of two PGs, the other two are probably involved in the condensation of a PE with a PG. A heterologous gene expression experiment showed the production of dialkylglycerols upon anaerobic expression of the glycerol ester reductase enzyme of in . Reduction of the ester bonds probably occurs first at the -1 and subsequently at the -2 position after the formation of PEs and PGs, since PGs possess a much higher percentage of ether bonds than PEs.

Abstract:Solvent extracted sediments from El Junco Lake, Galapagos were sequentially hydrolyzed with base and acid. Some of the "bound'' lipids thus generated were not detected among the "free'' lipids obtained from the initial solvent extract of the freeze dried sediment. These include alpha-hydroxy acids, beta-hydroxy acids, methyl beta-hydroxy acids and cutin acids. Other bound lipids differed from the free lipids in their chain length, such as alkanols, sterols, omega-hydroxy acids and (omega-1)-hydroxy acids. A suite of biomarkers with unique ions of m/z 341 were identified in the acid hydrolysates that are most likely C-40 cyclic biomarkers derived from the L race of Botryococcus braunii.Free lipids and their isotopic composition are likely to be sufficient for the purpose of paleoenvironmental reconstructions, and therefore base or acid hydrolysis of solvent extracted sediment is not necessary. Nevertheless, the additional lipid biomarkers liberated by base and acid hydrolysis can provide information on microbial inputs to the sediment. (C) 2014 Elsevier Ltd. All rights reserved.
The origins and implications of glycerol ether lipids in China coastal wetland sediments

Abstract:Coastal wetlands are terrestrial-marine transition zones harboring diverse active microbial communities. The origins of diverse glycerol ether lipids preserved in coastal wetlands are rarely investigated. 16 surface sediments were collected from the coastal wetland at Guangrao (GR), Changyi (CY) and Xiamen (XM), where both climate and sedimentary environment show significant differences. Ten groups of glycerol ether lipids, including isoprenoidal and branched glycerol dialkyl glycerol tetraethers (iGDGTs and bGDGTs), isoprenoidal and branched glycerol dialkanol diethers (iGDDs and bGDDs), hydroxylated isoprenoidal GDGTs and GDDs (OH-GDGTs and OH-GDDs), overly branched GDGTs (OB-GDGTs), sparsely branched GDGTs (SB-GDGTs), hybrid isoprenoid/branched GDGTs (IB-GDGTs) and a tentatively assigned H-shaped branched GDGTs (H-B-GDGTs) were detected and quantified. Sediments collected in the north (Guangrao and Changyi) contain, in general, a lower abundance of GDGT (3.7-55.9 ng/g sed) than samples from south (Xiamen; 251-1020 ng/g sed). iGDGTs and bGDGTs are the predominant components at all sites and account for 17.2-74.3% and 16.1-75.1% of total ether lipids, respectively. The relative abundance of iGDGTs decreases but that of bGDGTs increases with the distance from sea, suggesting a marine vs. terrestrial origin of iGDGT and bGDGTs, respectively. In addition, the methylation index (MIOB/B/SB) of branched GDGTs shows a significant inverse correlation with water content, suggesting that marine waters have a major influence on the microbial communities inhabiting wetland sediment. Such an assumption was confirmed by the distinct lipid pattern of three low water content (<5%) samples collected in an area isolated from tidal flushing. The other isoprenoidal ether lipids, such as iGDDs, OH-GDGTs and OH-GDGTs, have a similar distribution as iGDGTs, indicating a common biological source, so do the corresponding non-isoprenoidal ether lipid series with bGDGTs. The BIT value increases with increasing distance from the sea, which implies that the BIT index can be probably applied to trace past sea level change in costal wetland settings. The reconstructed temperature from TEX86 shows significant offset from observed data, but only little deviation for the MBT/CBT calculated temperature. This suggests that the MBT/CBT has the potential to reconstruct past temperatures in coastal wetland settings.

Abstract:Lipid biomarkers in lacustrine sediments provide valuable information about lake history and environmental change. Here we report the occurrence of a unique set of lipid biomarkers from a freshwater crater lake, El Junco, in the Galápagos. In addition to previously reported alkadienes, botryococcenes and lycopadienes indicative of Botryococcus braunii A, B and L races, we find highly branched C25 isoprenoids (HBIs) from diatoms, monomethyl alkanes likely from insect epicuticular waxes, long chain alkenols, diols and a triol, keto-ols, hydroxy acids and keto acids. Saturated and mono-unsaturated long chain diols from C30–C36 had terminal hydroxyl groups and hydroxyl groups between the ω16 and ω20 positions. Vicinal diols with hydroxyl groups at ω9 and ω10 were likely from the floating fern Azolla. C30–C36 keto-ols, mid-chain hydroxy and keto acids had mid-chain functional groups at similar positions to the diols, suggesting common origins. The predominance of ω20-hydroxy acids and diols, together with 20,21-dihydroxy-nonacosanoic acid is indicative of an Azolla source, while ω16 and ω18 hydroxy acids and diols imply a microalgal source.
Hydrogen Isotope Fractionation in Freshwater Algae: I. Variations among Lipids and Species

Abstract:Five species of freshwater green algae, including three strains of Botryococcus braunii (two A Race, one B Race), Eudorina unicocca and Volvox aureus, were cultured under controlled conditions in media containing different concentrations of deuterium. The hydrogen isotopic ratios of lipids in the algae, including alkadienes, botryococcenes, heptadecenes, fatty acids, and phytadiene, were measured by gas chromatography–isotope ratio-mass spectrometry (GC–IRMS) and found to closely track water δD values. While correlation coefficients (R2) in excess of 0.99 for all lipids in all species suggest that lipid δD values can be used to determine water δD values, hydrogen isotope fractionation was found to vary systematically between lipids and lipid homologues within a single alga, as well as for the same lipid between species of algae. Under similar growth conditions, two species of Chlorophyceae (Eudorina unicocca and Volvox aureus) and three species of Trebouxiophyceae (Botryococcus braunii) produced palmitic acid (C16 fatty acid) that differed by 90–100‰ relative to water. Ubiquitous lipids such as palmitic acid, with a multitude of aquatic and terrestrial sources, are therefore not good targets for D/H-based paleohydrologic reconstructions. In addition to the use of source-specific biomarkers that derive unambiguously from a single family or species, paleohydrologic applications of lipid D/H ratios will need to consider the as yet unstudied potential influence that environmental parameters such as nutrients, light and temperature, etc., may have on D/H fractionation during lipid synthesis.

Abstract:The different fractions of glycolipids were separated by HPLC column. Lipase XI from Rhizopus arrhizus was used to hydrolyze the fatty acids esterified in sn-1. The main fatty acids in MGDG and DGDG were C16:0, C18:2n-6, and C18:3n-6; those in SQDG were C16:0 and C18:2n-6 with a small amount of C18:3n-6. The molecular species analysis of the glycolipids showed that unsaturated fatty acids, such as C18:2n-6 and C18:3n-6, mainly occupied the sn-1 position, while saturated fatty acid C16:0 occupied the sn-2 position. MGDG contained 43.6% (18:3, 16:0), 13.9% (16:0, 16:0), 9.9% (18:2,16:0), and 8.9% (18:3, 16:1). DGDG contained 37.6% (18:3, 16:0), 8.7% (16:0, 16:0), and 8.7% (18:2, 16:0). Molecular species composition of SQDG differed from those of MGDG and DGDG, and it contained 52.5% (18:2, 16:0), 11.0% (16:0, 16:0), 7.1% (16:1, 16:0), and 7.7% (18:1, 16:0).

Abstract:Unique archaeal lipid membrane composition, including membrane‐spanning, tetraether lipids, presumably contribute to survival in extreme pH, temperature, and salinity. The biosynthesis of tetraether lipids in the model species Thermococcus kodakarensis promotes survival at high temperatures, and we demonstrate that alternative lipid species are produced when tetraether lipid biosynthesis is compromised. The sole unifying feature of the incredibly diverse Archaea is their isoprenoid‐based ether‐linked lipid membranes. Unique lipid membrane composition, including an abundance of membrane‐spanning tetraether lipids, impart resistance to extreme conditions. Many questions remain, however, regarding the synthesis and modification of tetraether lipids and how dynamic changes to archaeal lipid membrane composition support hyperthermophily. Tetraether membranes, termed glycerol dibiphytanyl glycerol tetraethers (GDGTs), are generated by tetraether synthase (Tes) by joining the tails of two bilayer lipids known as archaeol. GDGTs are often further specialized through the addition of cyclopentane rings by GDGT ring synthase (Grs). A positive correlation between relative GDGT abundance and entry into stationary phase growth has been observed, but the physiological impact of inhibiting GDGT synthesis has not previously been reported. Here, we demonstrate that the model hyperthermophile Thermococcus kodakarensis remains viable when Tes (TK2145) or Grs (TK0167) are deleted, permitting phenotypic and lipid analyses at different temperatures. The absence of cyclopentane rings in GDGTs does not impact growth in T. kodakarensis, but an overabundance of rings due to ectopic Grs expression is highly fitness negative at supra‐optimal temperatures. In contrast, deletion of Tes resulted in the loss of all GDGTs, cyclization of archaeol, and loss of viability upon transition to the stationary phase in this model archaea. These results demonstrate the critical roles of highly specialized, dynamic, isoprenoid‐based lipid membranes for archaeal survival at high temperatures.

Structure-Function Relationships in Pure Archaeal Bipolar Tetraether Lipids

Abstract:Archaea, the third fundamental domain of life are distinguished from bacteria and eukaryotes due to the presence of unique lipids in their cell membranes. Archaeal bipolar lipids are among the most unusual lipids occurring in nature because of their presumed ability to span the entire membrane. They are challenging to extract in pure form from natural sources or synthesize chemically, and as a result, prior studies on pure lipids have been limited. Here we have utilized synthesis to enable in-depth biophysical investigations on a series of archaeal glycerol dialkyl glycerol tetraether (GDGT) lipids having symmetric or unsymmetric combinations of polar head groups. We showed that these lipids self-assemble to form membrane-bound vesicles in aqueous media, encapsulate polar molecules, and reconstitute a functional integral membrane protein. Membrane thicknesses and electron density profiles were investigated by performing small-angle X-ray scattering (SAXS) studies and cryogenic electron microscopy (cryo-EM) imaging on unilamellar vesicles of GDGT lipids. SAXS studies on bulk aqueous dispersions of GDGT lipids over a large temperature range (10-90 °C) allowed us to identify lamellar and non-lamellar phases and their interconversions under various buffer conditions. We also studied how the propensity to form various mesophases is reflected in the functional behavior of the GDGT membranes. Specifically, we asked whether vesicles overwhelmingly composed of GDGTs can undergo fusion as it is difficult to conceptualize such behavior with the assumption that such membranes have a monolayer structure. Interestingly, we observed that GDGT vesicles undergo fusion with influenza virus with lipid mixing kinetics comparable to that with vesicles composed of typical monopolar phospholipids. Our results suggest that GDGT membranes may consist of regions with a bilayer structure which facilitates fusion and thus offer insight into how archaea may perform important physiological functions that require dynamical membrane behavior.

Lipids of extremely halophilic archaeobacteria from saline environments in India: a novel glycolipid in Natronobacterium strains

Abstract:Several strains of extremely halophilic archaeobacteria, both non-alkaliphilic and alkaliphilic, including Halobacterium, Haloferax and Natronobacterium species, were isolated from salt locales in India. The major phospholipids in these strains were the C20-C20-glycerol diether analogues of phosphatidylglycerolmethylphosphate (PGP-Me), phosphatidylglycerol (PG) and phosphatidic acid (PA). In addition, the Halobacterium strains possessed the characteristic glycolipids, sulfated triglycosyl and tetraglycosyl diethers (S-TGD-1 and S-TeGD, respectively) and the unsulfated triglycosyl diether (TGD-1); and the Haloferax strains had the characteristic sulfated and unsulfated diglycosyl glycerol diethers (S-DGD-1 and DGD-1, respectively). The PGP-Me, and PG components of the haloalkaliphiles each occurred as two molecular species with C20-C20- and C20-C25-(isopranoid) glycerol diether lipid cores. In contrast to previous reports of the absence of glycolipids in natronobacteria, the Natronobacterium strains from India were found to contain small amounts of a novel glycolipid identified as glucopyranosyl-1-->6-glucopyranosyl-1-->1-glycerol diether (DGD-4). The lipid cores of DGD-4 also contained mainly unhydroxylated or hydroxylated C20-C20, C20-C25 and C25-C25 molecular species with unsaturated (isoprenoid) chains. Hydroxylated lipid cores have previously been identified only in some methanogenic archaeobacteria.
Characteristics of isoprenoid and branched tetraether lipids in the deep ocean: implications from surface sediments in the seamount area of the Western Pacific Ocean

Abstract:The isoprenoid glycerol dialkyl glycerol tetraethers (isoGDGTs) and branched glycerol dialkyl glycerol tetraethers (brGDGTs) in marine sediments have been widely used for paleoceanographic reconstructions. However, their distributions in the deep ocean (> 4000 m) have not been well characterized. This study analyzed GDGTs in surface sediments from the seamount area of Marcus-Wake and Magellan, western Pacific Ocean (4431-5756 m). The results were further compared with published data of GDGTs in the South China Sea (SCS), equatorial Pacific Ocean, and Mariana Trench to reveal the provenance of GDGTs in the deep ocean and their suitability for paleoceanographic reconstruction. The ratios of GDGT-0/crenarchaeol (0.59–1.38) and GDGT-2/3 (10.65–14.78) indicate the contribution from benthic Thaumarchaeota in the seamount area. The isoGDGTs of the surface sediments from the seamount and the Mariana Trench regions were clustered for a redundancy analysis. They are characterized by the similar relative abundance of each isoGDGT, i.e. higher fractional abundance of GDGT-0 (26–49%) than those in the SCS (12–23%) and the equatorial Pacific Ocean (22–28%). This suggests isoGDGTs in the deep ocean (> 4000 m) are for both planktonic and benthic Thaumarchaeota and the benthic Thaumarchaeota likely produce excessive GDGT-0 in the seamount area. For brGDGTs, the higher IIIa/IIa ratios in the seamount area (1.67–4.83) than the marine-derived values (0.92), indicating in-situ production of brGDGTs. The discrepancy (0.7 to 5.0 °C) between sea surface temperature, deduced by TetraEther indeX (TEX86), and the observed sea surface temperature is likely due to less input of isoGDGTs from the pelagic Thaumarchaeota but more from the benthic Thaumarchaeota.

Abstract:Branched glycerol dialkyl glycerol tetraethers (brGDGTs) are bacterial membrane lipids that are frequently employed as paleoenvironmental proxies because of the strong empirical correlations between their relative abundances and environmental temperature and pH. Despite the ubiquity of brGDGTs in modern and paleoenvironments, the source organisms of these enigmatic compounds have remained elusive, requiring paleoenvironmental applications to rely solely on observed environmental correlations. Previous laboratory and environmental studies have suggested that the globally abundant bacterial phylum of the Acidobacteria may be an important brGDGT producer in nature. Here, we report on experiments with a cultured Acidobacterium, Solibacter usitatus, that makes a large portion of its cellular membrane (24 ± 9% across all experiments) out of a structurally diverse set of tetraethers including the common brGDGTs Ia, IIa, IIIa, Ib, and IIb. Solibacter usitatus was grown across a range of conditions including temperatures from 15 to 30°C, pH from 5.0 to 6.5, and O2 from 1% to 21%, and demonstrated pronounced shifts in the degree of brGDGT methylation across these growth conditions. The temperature response in culture was in close agreement with trends observed in published environmental datasets, supporting a physiological basis for the empirical relationship between brGDGT methylation number and temperature. However, brGDGT methylation at lower temperatures (15 and 20°C) was modulated by culture pH with higher pH systematically increasing the degree of methylation. In contrast, pH had little effect on brGDGT cyclization, supporting the hypothesis that changes in bacterial community composition may underlie the link between cyclization number and pH observed in environmental samples. Oxygen concentration likewise affected brGDGT methylation highlighting the potential for this environmental parameter to impact paleotemperature reconstruction. Low O2 culture conditions further resulted in the production of uncommon brGDGT isomers that could be indicators of O2 limitation. Finally, the production of brGTGTs (trialkyl tetraethers) in addition to the previously discovered iso-C15-based mono- and diethers in S. usitatus suggests a potential biosynthetic pathway for brGDGTs that uses homologs of the archaeal tetraether synthase (Tes) enzyme for tetraether synthesis from diethers.

Abstract:Glycerol dialkyl diethers (GDDs) are membrane lipids and a variation of the more commonly known glycerol dialkyl glycerol tetraethers (GDGTs). GDGTs include both archaeal and bacterial membrane lipids that are both frequently used for paleoclimate reconstruction in a range of terrestrial and aquatic environments. In contrast to GDGTs, GDDs lack one of the terminal glycerol moieties. Although both isoprenoidal (iso) and branched (br) GDDs have been found, this study focuses on isoGDDs. These lipids have been found in a few terrestrial and aquatic environments. However, the origin of isoGDDs is debated and the extent of their distribution across the surface of the Earth is poorly constrained. Based on a few single site studies, previous authors hypothesised that isoGDDs are degradation products of isoGDGTs, but more recent studies that isolated isoGDDs from cultured nitrososphaerota (formerly thaumarchaeota) proposed a biological source through direct archaeal biosynthesis. Here we compiled a global dataset of isoGDD and isoGDGT abundance in environmental samples to thoroughly investigate the distribution of isoGDDs and the correlation with isoGDGTs on a global scale and across a variety of environments (peat, mineral soils, lake sediments, and marine sediments). We find that isoGDDs are present in most samples that we analysed. Their abundance is strongly proportional to isoGDGT abundance (r 2 = 0.85), dominated by the GDGT-crenarchaeol/GDD-crenarchaeol ratio (r 2 = 0.94) and supported by individual compound isoGDGT/isoGDD ratios (r 2 = 0.56–0.94). In addition, the degree of cyclisation of isoGDDs, reflected in the ring index, is positively correlated (r 2 = 0.84) with that of isoGDGTs across all environments. We conclude that isoGDDs are abundant on the surface of the Earth and predominantly originate from the degradation of isoGDGTs.

Abstract:The Black Sea is a permanently anoxic, marine basin serving as model system for the deposition of organic‐rich sediments in a highly stratified ocean. In such systems, archaeal lipids are widely used as paleoceanographic and biogeochemical proxies; however, the diverse planktonic and benthic sources as well as their potentially distinct diagenetic fate may complicate their application. To track the flux of archaeal lipids and to constrain their sources and turnover, we quantitatively examined the distributions and stable carbon isotopic compositions (δ13C) of intact polar lipids (IPLs) and core lipids (CLs) from the upper oxic water column into the underlying sediments, reaching deposits from the last glacial. The distribution of IPLs responded more sensitively to the geochemical zonation than the CLs, with the latter being governed by the deposition from the chemocline. The isotopic composition of archaeal lipids indicates CLs and IPLs in the deep anoxic water column have negligible influence on the sedimentary pool. Archaeol substitutes tetraether lipids as the most abundant IPL in the deep anoxic water column and the lacustrine methanic zone. Its elevated IPL/CL ratios and negative δ13C values indicate active methane metabolism. Sedimentary CL‐ and IPL‐crenarchaeol were exclusively derived from the water column, as indicated by non‐variable δ13C values that are identical to those in the chemocline and by the low BIT (branched isoprenoid tetraether index). By contrast, in situ production accounts on average for 22% of the sedimentary IPL‐GDGT‐0 (glycerol dibiphytanyl glycerol tetraether) based on isotopic mass balance using the fermentation product lactate as an endmember for the dissolved substrate pool. Despite the structural similarity, glycosidic crenarchaeol appears to be more recalcitrant in comparison to its non‐cycloalkylated counterpart GDGT‐0, as indicated by its consistently higher IPL/CL ratio in sediments. The higher TEX86, CCaT, and GDGT‐2/‐3 values in glacial sediments could plausibly result from selective turnover of archaeal lipids and/or an archaeal ecology shift during the transition from the glacial lacustrine to the Holocene marine setting. Our in‐depth molecular‐isotopic examination of archaeal core and intact polar lipids provided new constraints on the sources and fate of archaeal lipids and their applicability in paleoceanographic and biogeochemical studies.

Abstract:As one of the least-explored regions on Earth, the hadal zone remains poorly understood in terms of sedimentary organic geochemistry. This study examined the composition of isoprenoid glycerol dialkyl glycerol tetraethers (Iso-GDGTs) and hydroxy GDGTs (OH-GDGTs) in sediment cores with water depths ranging from 5058 to 10,911 m from the Mariana and Yap Trenches. The results showed a higher proportion of GDGT-0 (25–73%, mean = 49%) in the trench Iso-GDGTs pool than those from shallower ocean counterparts (<5000 m). Properties, including higher values of the [GDGT-2/GDGT-3] ratio (7.17–20.03, mean = 13.02) and the [GDGT-0/Crenarchaeol] ratio (0.49–3.94, mean = 1.48), as well as a dominance of OH-GDGT-0 in the OH-GDGTs, are similar to characteristics observed in other cold deep marine environments rather than tropical shallow ocean. Ring Index decreased with increasing water depth of the sampling sites, corresponding with increased fractional abundance of acyclic Iso-GDGTs and OH-GDGTs. Paleothermometer TEX86 in the surface sediments (>5000 m) revealed a significant negative linear correlation with water depth (TEX86H: R2 = 0.89, p < 0.01; TEX86L: R2 = 0.89, p < 0.01), leading to a bias toward lower proxy-derived temperatures. This is likely due to the increased incorporation of cold- and pressure- adapted species of archaea in dark ocean. Overall, this study provided implications of the extreme water depth effect on the distribution of isoprenoid and hydroxy tetraether lipids, as well as the proxy application in hadal environments.

Abstract:Glycerol dibiphytanyl glycerol tetraether (GDGT) lipids are part of the cellular membranes of Thaumarchaeota, an archaeal phylum composed of aerobic ammonia oxidizers, and are used in the paleotemperature proxy TEX86 . GDGTs in live cells possess polar head groups and are called intact polar lipids (IPL-GDGTs). Their transformation to core lipids (CL) by cleavage of the head group was assumed to proceed rapidly after cell death, but it has been suggested that some of these IPL-GDGTs can, just like the CL-GDGTs, be preserved over geological timescales. Here, we examined IPL-GDGTs in deeply buried (0.2-186 mbsf, ~2.5 Myr) sediments from the Peru Margin. Direct measurements of the most abundant IPL-GDGT, IPL-crenarchaeol, specific for Thaumarchaeota, revealed depth profiles, which differed per head group. Shallow sediments (<1 mbsf) contained IPL-crenarchaeol with both glycosidic and phosphate head groups, as also observed in thaumarchaeal enrichment cultures, marine suspended particulate matter and marine surface sediments. However, hexose, phosphohexose-crenarchaeol is not detected anymore below 6 mbsf (~7 kyr), suggesting a high lability. In contrast, IPL-crenarchaeol with glycosidic head groups is preserved over timescales of Myr. This agrees with previous analyses of deeply buried (>1 m) marine sediments, which only reported glycosidic and no phosphate-containing IPL-GDGTs. TEX86 values of CL-GDGTs did not markedly change with depth, and the TEX86 of IPL-derived GDGTs decreased only when the proportions of monohexose- to dihexose-GDGTs changed, likely due to the enhanced preservation of the monohexose GDGTs. Our results support the hypothesis that in situ GDGT production and differential IPL degradation in sediments is not substantially affecting TEX86 paleotemperature estimations based on CL-GDGTs and indicates that likely only a small amount of IPL-GDGTs present in deeply buried sediments is part of cell membranes of active archaea. The amount of archaeal biomass in the deep biosphere based on these IPLs may have been substantially overestimated.

Abstract:Typically, glycerophospholipids are represented with two esterified fatty acids. However, by up to 20%, a significant proportion of this lipid class carries an ether-linked fatty alcohol side chain at the sn -1 position, generally referred to as ether lipids, which shape their specific physicochemical properties. Among those, plasmalogens represent a distinct subgroup characterized by an sn -1 vinyl-ether double bond. The total loss of ether lipids in severe peroxisomal defects such as rhizomelic chondrodysplasia punctata indicates their crucial contribution to diverse cellular functions. An aberrant ether lipid metabolism has also been reported in multifactorial conditions including Alzheimer's disease. Understanding the underlying pathological implications is hampered by the still unclear exact functional spectrum of ether lipids, especially in regard to the differentiation between the individual contributions of plasmalogens (plasmenyl lipids) and their non-vinyl-ether lipid (plasmanyl) counterparts. A primary reason for this is that exact identification and quantification of plasmalogens and other ether lipids poses a challenging and usually labor-intensive task. Diverse analytical methods for the detection of plasmalogens have been developed. Liquid chromatography–tandem mass spectrometry is increasingly used to resolve complex lipid mixtures, and with optimized parameters and specialized fragmentation strategies, discrimination between ethers and plasmalogens is feasible. In this review, we recapitulate historic and current methodologies for the recognition and quantification of these important lipids and will discuss developments in this field that can contribute to the characterization of plasmalogens in high structural detail.

Abstract:Compared with other lipid biomarkers,glycerol diether membrane lipids generally bear more specific implications for provenance and environment.Up to date,comprehensive reviews on this topic are rare in both domestic and international literatures.In this paper,we made a brief review on the analytical methods of glycerol diethers,the composition characteristics of archaeal diethers and bacte rial diethers,with emphasis on Archaea (e.g.,methanogens,methanotrophs,halophiles) and bacteria (e.g.,sulfate-reducing bacteria,Aquificales,and some thermophiles).Special attention is paid to the application of glycerol diether membrane lipids and possible biogeochemical processes to the study of extreme environment,such as cold seep,hot spring and hydrothermal systems.The impacts of environmental parameters on the distribution of glycerol diether membrane lipids are briefly introduced and future application of diethers and other lipid biomarkers are discussed.
