Abstract:Introduction of anthropogenic chemicals into soil may have lasting effects on soil microbial activities and thus soil health. This study was conducted with chlorothalonil to evaluate its effects on soil bacterial, fungal, and actinomycete populations and soil enzymes (acid phosphatase, alkaline phosphatase, urease, catalase, and invertase) after repeated applications. After the first addition of chlorothalonil, the soil bacterial and actinomycete populations were significantly reduced, whereas the population of soil fungi was unchanged. The most marked inhibition on soil microorganisms was observed after the second pesticide addition. However, after initial variations, soil bacteria, fungi, and actinomycetes adapted gradually to chlorothalonil, and the negative effects became transient and weaker following the third and fourth treatments. The inhibitory effect of repeated chlorothalonil applications on soil enzymes followed a similar trend to that on soil microorganisms. Repeated chlorothalonil applications did not result in significant changes in its persistence. Three bacterial strains capable of utilizing chlorothalonil as a sole source of carbon and energy for growth were isolated 21 days after the fourth treatment with chlorothalonil, which indicated that the capability of soil microorganisms for degrading chlorothalonil was formed during the experiment.

Abstract:The application of the herbicide chlorimuron-ethyl has a lasting toxic effect on some succession crops. Here, a bacterium capable of utilizing chlorimuron-ethyl as the sole source of nitrogen was isolated from the contaminated soil and was identified as Enterobacter ludwigii sp. CE-1, and its detoxification and degradation of the herbicide were then examined. The biodegradation of chlorimuron-ethyl by the isolate CE-1 was significantly accelerated with increasing concentration (1–10mg/l) and temperature (20–40°C). The optimal pH for the degradation of chlorimuron-ethyl by the isolate CE-1 was pH 7.0. A pathway for the biodegradation of chlorimuron-ethyl by the isolate CE-1 was proposed, in which it could be first converted into 2-amino-4-chloro-6-methoxypyrimidine and an intermediate product by the cleavage of the sulfonylurea bridge and then transformed into saccharin via hydrolysis and amidation. The plant height and fresh weight of corn that had been incubated in nutrient solution containing 0.2mg/l of chlorimuron-ethyl significantly recovered to 83.9% and 83.1% compared with those in the uninoculated control, although the root growth inhibition of chlorimuron-ethyl could not be alleviated after inoculation for 14 d. The results indicate that the isolate CE-1 is a promising bacterial resource for the biodegradation and detoxification of chlorimuron-ethyl.

Abstract:The effect of repeated carbendazim applications on functional diversity of culturable microorganisms and bacterial community composition was studied under field conditions. The functional diversity of soil culturable microbial community (Shannon index, H') reduced significantly (P<0.05) after the first introduction of carbendazim at levels of 0.94, 1.88 and 4.70 kg active ingredient (a.i.)ha(-1) and then recovered to that in the control with subsequent applications. An evident (P<0.01) difference in the bacterial community composition was observed after the second carbendazim application by Temperature Gradient Gel Electrophoresis (TGGE) analysis of 16S rRNA genes amplified from treated and control soils, which remained after the third and fourth treatments. Our results indicated that repeated carbendazim applications have a transient harmful effect on functional diversity of soil culturable microbial community and result in an alteration in bacterial community composition largely due to one species within the γ-proteobacterium.
Degradation of Chlorpyrifos in Laboratory Soil and Its Impact on Soil Microbial Functional Diversity.

Abstract:Degradation of chlorpyrifos at different concentrations in soil and its impact on soil microbial functional diversity were investigated under laboratory condition. The degradation half-live of chlorpyrifos at levels of 4, 8, and 12 mg/kg in soil were calculated to be 14.3, 16.7, and 18.0 d, respectively. The Biolog study showed that the average well color development (AWCD) in soils was significantly (P < 0.05) inhibited by chlorpyrifos within the first two weeks and thereafter recovered to a similar level as the control. A similar variation in the diversity indices (Simpson index 1/D and McIntosh index U) was observed, but no significant difference among the values of the Shannon-Wiener index H' was found in chlorpyrifos-treated soils. With an increasing chlorpyrifos concentration, the half-life of chlorpyrifos was significantly (P < or = 0.05) extended and its inhibitory effect on soil microorganisms was aggravated. It is concluded that chlorpyrifos residues in soil had a temporary or short-term inhibitory effect on soil microbial functional diversity.
Persistence of Repeated Triadimefon Application and Its Impact on Soil Microbial Functional Diversity

Abstract:The effects of repeated applications of the fungicide triadimefon in agricultural soil on the microbial functional diversity of the soil and on the persistence of the fungicide in the soil were investigated under laboratory conditions. The degradation half-lives of triadimefon at the recommended dosage, simulated by a first-order kinetic model, were 23.90, 22.95, and 21.52 days for the first, second, and third applications, respectively. Throughout this study, no significant inhibition of the Shannon-Wiener index (H′) was observed. However, the Simpson index (1/D) and the McIntosh index (U) were obviously reduced (p ≤ 0.05) during the initial 3 days after the first triadimefon application and thereafter, gradually recovered to or exceeded the level of the control soil. A similar trend in variation but with a faster recovery in the 1/D and U was observed after the second and third triadimefon applications, respectively. Taken together, the above results indicate that the repeated application of triadimefon enhanced the degradation rate of the fungicide and the recovery rate of the soil microbial functional diversity. It is concluded that repeated triadimefon applications in soil have a transient or temporary inhibitory effect on soil microbial communities.
Exploring Microbial Community Structure and Biological Function in Manured Soil During Ten Repeated Treatments with Chlortetracycline and Ciprofloxacin.

Abstract:The changes of enzyme activities, microbial community structure and function, and the diversity and resistance level of antibiotic-resistant bacteria (ARB) were studied in soil during ten repeated treatments with chlortetracycline (CTC) and/or ciprofloxacin (CIP) together with organic manure (OM) under laboratory conditions. The activities of neutral phosphatase (NPA) and catalase (CAT) displayed the suppression-recovery-stimulation trend in the OM&CTC treatment but the stimulation trend in the OM&CTC&CIP treatment. The NPA was stimulated but the CAT was little affected in the OM&CIP treatment. Soil microbial functional diversity displayed the suppression-recovery-stimulation trend in the OM&CTC and OM&CTC&CIP treatments and the stimulation-suppression trend in the OM&CIP treatment with the treatment frequency. Metagenomic analysis showed that the relative abundances of Actinobacteria and Firmicutes in the antibiotic treatment significantly increased by 0.5-235.6%, but that of Proteobacteria decreased by 0.2-27.3% compared to the control with the treatment frequency. Furthermore, the relative abundances of dominant bacterial genera including Streptomyces, Actinomadura, Mycobacterium, and Streptococcus in the antibiotic treatment significantly increased by 1.1-10433.3% compared to the control. Meanwhile, repeated antibiotic treatments induced a significant increase in the diversity and resistance level of ARB isolates, especially in the OM&CTC treatment. It is concluded that repeated treatments with CTC and/or CIP can alter enzyme activities, microbial community structure and function, and increase the diversity and resistance level of ARB isolates.
Repeated Treatments of Ciprofloxacin and Kresoxim-Methyl Alter Their Dissipation Rates, Biological Function and Increase Antibiotic Resistance in Manured Soil.

Abstract:The dissipation of ciprofloxacin (CIP, 1.0 and 10.0mg/kg) and kresoxim-methyl (KM, 1.0 and 2.0mg/kg) in manure-amended soil, the variations in soil enzyme activities and microbial functional diversities, and CIP-induced bacterial community tolerances were studied using a chromatographic analysis, enzyme colorimetric and titration analyses, and the BIOLOG EcoPlate method. Three successive treatments of individual and combined samples of CIP and KM at low and high concentrations were performed at 60d intervals. The dissipation half-life of CIP increased, but that of KM decreased in manured soil with treatment frequency; furthermore, the combined treatment altered the dissipation rates of CIP and KM. A stronger inhibitory effect on the activities of soil neutral phosphatase and urease was observed in the individual KM treatment than in the individual CIP treatment. A similar inhibitory trend was also found in soil neutral phosphatase activity in the combined treatment at high concentration compared to that at low concentration, but the activity of soil catalase was enhanced in the early stages of the KM or CIP treatments. Meanwhile, the inhibitory trend on the overall activity and functional diversity of soil microorganisms was observed in the individual KM or CIP treatment, and the combined treatment exerted a greater suppression effect than that in the individual treatment. Bacterial community resistance to CIP increased significantly with increasing treatment frequency and concentration, and furthermore antibiotic resistance developed faster in the combined treatment than in the individual treatment. It was concluded that the repeated treatments of CIP and KM could alter their dissipation rates and soil enzyme activities, suppress microbial functional diversity, and increase bacterial community resistance to CIP in manured soil.

Abstract:The degradation characteristics of carbendazim (CBD) and chlortetracycline (CTC) in individual and combined treatments, and dynamics of soil microbial structural and functional diversity as well as their potential relations were studied during three repeated treatments using different concentrations. The results showed that the degradation half-life of CBD at concentrations of 3mg/kg and 6mg/kg obviously increased, but that of CTC at levels of 1mg/kg and 10mg/kg decreased with increasing treatment frequency. Soil microbial activity and functional diversity displayed the suppression trend in CBD treatment and the suppression-recovery-stimulation trend in CTC and CBD+CTC treatments, which were consistent with the findings of decreased degradation rate of CBD and increased degradation rate of CTC. 16S amplicon sequencing analysis revealed five potentially dominant CTC-resistant microbial genera including Bacillus, Actinobacillus, Pseudomonas, Mycobacterium, and Corynebacterium, which may mainly carry major facilitator superfamily transporter protein, ribosomal protection protein, and other proteins encoded by tetA, tetB, tetC, tetH, tetL, tetM, tetO, tetV, tetW, tetX, tetZ, tet33, and tet39. These five dominant genera may jointly contribute to the elevated bacterial community resistance to CTC. Our findings provided a better understanding of microbial community structure and function changes in repeatedly treated soils with CBD and CTC.
Microbial Response to Repeated Treatments of Manure Containing Sulfadiazine and Chlortetracycline in Soil

Abstract:Substantive addition of antibiotic-contaminated manure to agricultural soil may lead to "persistent" residues of antibiotics and may affect soil health. Therefore, this study examines the effects of repeated manure treatments containing sulfadiazine (SDZ) and chlortetracycline (CTC) residues, both individually and combined, on the functional diversity and structure of soil microbial communities in the soils under laboratory conditions. The average well color development (AWCD), Simpson diversity index (1/D, dominant populations), Shannon-Wiener diversity index (H', richness), and McIntosh diversity index (U, evenness) in the antibiotics-treated soils decreased in the first 60-day treatment and then gradually recovered or even exceeded the initial level in the unamended soils with increasing treatment frequency. A total of 11 specific bands in temperature gradient gel electrophoresis (TGGE) profiles were observed and sequence analyzed for five repeated treatments, and most of them belonged to the phyla Firmicutes, Actinobacteria, and Proteobacteria. These results indicate that repeated treatments of manure containing SDZ and CTC residues can alter soil microbial community structure, although they have a temporary suppression effect on soil microbial functional diversity.
Changes in Soil Biological Properties with the Addition of Metsulfuron-Methyl Herbicide

Abstract:An incubation study was conducted to investigate the effects of metsulfuron-methyl herbicide on the soil microbial biomass in loamy sand soil. The herbicide was applied to the soil at four concentrations: control, 0.01, 0.10, and 1.00 μg·g −1 soil. Determinations of microbial biomass-C and microbial biomass-N contents were carried out 1, 3, 5, 7, 10, 15, 25, and 45 days after herbicide application. In comparison to untreated soil, the microbial biomass-C and biomass-N decreased significantly in soils treated with herbicide at concentrations of 0.1 and 1.0 μg·g −1 soil within the first 7 days of incubation. The application of metsulfuron-methyl herbicide to the soil reduced the C mic /C org and N mic /N total percentages, which decreased with increasing application rate of metsulfuron-methyl herbicide. Compared to the untreated control, a marked increase in the microbial biomass C: N ratio was observed in the herbicide treated soil. This effect was transitory and was significant only at the higher rates of metsulfuron-methyl.

Abstract:Long-term and excessive application of chlorimuron-ethyl has led to a series of environmental problems. Strain Hansschlegelia sp. CHL1, a highly efficient chlorimuron-ethyl degrading bacterium isolated in our previous study, was employed in the current soil bioremediation study. The residues of chlorimuron-ethyl in soils were detected, and the changes of soil microbial communities were investigated by phospholipid fatty acid (PLFA) analysis. The results showed that strain CHL1 exhibited significant chlorimuron-ethyl degradation ability at wide range of concentrations between 10μg kg-1 and 1000μg kg-1. High concentrations of chlorimuron-ethyl significantly decreased the total concentration of PLFAs and the Shannon-Wiener indices and increased the stress level of microbes in soils. The inoculation with strain CHL1, however, reduced the inhibition on soil microbes caused by chlorimuron-ethyl. The results demonstrated that strain CHL1 is effective in the remediation of chlorimuron-ethyl-contaminated soil, and has the potential to remediate chlorimuron-ethyl contaminated soils in situ.

Abstract:Effects of repeated applications of chlorpyrifos on its persistence and soil microbial functional diversity were studied under laboratory conditions. The results showed that the degradation rate of chlorpyrifos increased whereas its inhibitory effect on soil microbial communities gradually decreased with application frequency of chlorpyrifos. A bacterial strain DSP capable of utilizing chlorpyrifos as a sole source of carbon and energy was isolated 21 days after the third chlorpyrifos application, which indicated that the capability of soil microorganism for degrading chlorpyrifos was formed during the experiment. It could be concluded that repeated applications of chlorpyrifos had no lasting impact on soil health.

Abstract:Ding, Junnan; Li, Xin; Biolog Eco technology was used to investigate the effects of different chemical herbicide application methods, such as pre-emergence after sowing and post-emergence (stem and leaf spraying), on the characteristics of carbon source utilization by microbial communities in alfalfa rhizosphere soil. The averages of well color development (AWCD) and microbial metabolism diversity indices of post-emergence herbicide spraying on stems and leaves were significantly lower than those of pre-emergence herbicide and without herbicide treatments. Furthermore, pre-emergence after sowing herbicide treatment did not affect soil microorganisms. The principal component analysis (PCA) revealed that the microbial community diversities of the rhizosphere soils differed significantly between herbicide treatments. Carbohydrate was the carbon source type that was most sensitive to herbicide treatment changes, followed by amino acids and carboxylic acids. The herbicide application method significantly reduced the capacities of the microbial community to utilize the carbon sources, as mainly manifested in the ability to use polymers. As shown by a comprehensive analysis on the growth of alfalfa and the physiological and biochemical indices of its root system, the application of herbicide chlorimuron-ethyl reduced the alfalfa yield, and the two different application methods tested showed no significant difference. Herbicide application and different application methods showed significant impacts on alfalfa root activity and membrane permeability, suggesting that the application of herbicide damaged the membrane permeability and the activity of the alfalfa root system. Impacts of herbicide residues in the soil after stem and leaf post-emergence treatment on alfalfa growth and soil microorganisms should not be ignored.

Abstract:Chlorpyrifos is a widely used insecticide in recent years, and it will produce adverse effects on soil when applied on crops or mixed with soil. In this study, nested polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE) were combined to explore the bacterial and fungal community successions in soil treated with 5 and 20 mg/kg of chlorpyrifos. Furthermore, isolates capable of efficiently decomposing chlorpyrifos were molecular-typed using enterobacterial repetitive intergenic consensus-PCR (ERIC-PCR). Under the experimental conditions, degradation of chlorpyrifos in soil was interpreted with the first-order kinetics, and the half-lives of chlorpyrifos at 5 and 20 mg/kg doses were calculated to be 8.25 and 8.29 d, respectively. DGGE fingerprint and principal component analysis (PCA) indicated that the composition of the fungal community was obviously changed with the chlorpyrifos treatment, and that samples of chlorpyrifos treatment were significantly separated from those of the control from the beginning to the end. While for the bacterial community, chlorpyrifos-treated soil samples were apparently different in the first 30 d and recovered to a similar level of the control up until 60 d, and the distance in the PCA between the chlorpyrifos-treated samples and the control was getting shorter through time and was finally clustered into one group. Together, our results demonstrated that the application of chlorpyrifos could affect the fungal community structure in a quick and lasting way, while only affecting the bacterial community in a temporary way. Finally, nine typical ERIC types of chlorpyrifos-degrading isolates were screened.
Effect of Methamidophos and Urea Application on Microbial Communities in Soils As Determined by Microbial Biomass and Community Level Physiological Profiles.

Abstract:In this study, we evaluated the effect of the application by two agrochemicals, methamidophos (O,S-dimethyl phosphoroamidothioate) and urea, on microbial diversity in soil, using the combined approaches of soil microbial biomass analysis and community level physiological profiles (CLPPs). The results showed that both a low and a high level of methamidophos application (CS2 and CS3) and urea application (CS4) significantly decreased microbial biomass C (Cmic) by 41-83% compared with the control (CS1). The soil organic C (Corg) values of CS3 and CS4 were significantly higher and lower by 24% and 14%, respectively, than that of CS1. Similarly to Cmic, the values of Cmic/Corg of the three applied soils which decreased were lower by 31-84% than that of CS1. In contrast, the respiration activity of the three applied soils were significantly higher than the control. Agrochemical application also significantly increased the soil total of N and P (Ntol and Ptol) and decreased the Corg/Ntol and Corg/Ptol values. The CLPPs results showed that the AWCD (average well color development) of the three applied soils were significantly higher than that of CS1 during the incubation period. Substrate richness, Shannon and Simpson indices of microbial communities under chemical stresses, increased significantly. In addition, the CFU (colony-forming unit) numbers of methamidophos metabolized bacteria in CS2 and CS3 also increased significantly by 86.1% and 188.9% compared with that of CS1. The combined results suggest that agrochemicals reduce microbial biomass and enhance functional diversities of soil microbial communities; meanwhile, some species of bacteria may be enriched in soils under methamidophos stress.

Abstract:The degradation capacity of the four chlorimuron-ethyl-degrading strains, D310-1, LCY-2, LCY-3 and LCY-4, which identified as Rhodococcus sp.,Stenotrophomonas maltophilia sp,Bacillus subtilis sp., and Rhodotorula mucilaginosa sp., respectively, combined in different ways was examined in this study. Then the optimal degradation conditions and mitigation phytotoxicity of chlorimuron-ethyl on sensitive crop were investigated by both orthogonal test and pot culture experiment. The experimental results showed that the combination of four strains was adopted, and the removal rate of chlorimuron-ethyl reached the maximum of 90% in 30 days. Then the optimal degradation conditions were inoculum size 2.5%, temperature 28°C, pH 7.0 and the soil moisture 35%, and the mixed strains could eradicate 92.57% of chlorimuron-ethyl within 30 days. Furthermore pot culture experiment indicated that inoculation of mixed strains could also mitigate the phytotoxic effects of chlorimuron-ethyl on the growth of cucumber seedling.

Abstract:Imazethapyr (IM) is a chiral herbicide composed of an (-)-R-enantiomer and an (+)-S-enantiomer with differential herbicidal activity. In this study, the effects of microbial organisms, humidity, and temperature on the selective degradation of the (-)-R- and (+)-S-enantiomers of IM were determined in silty loam (SL) and clay loam (CL) soil with different pH values. The (-)-R-enantiomer of IM was preferentially degraded in two soils under different microorganism, humidity, and temperature conditions. The average half-lives of R-IM ranged from 43 to 66.1 days and were significantly shorter (P < 0.05) than those of S-IM, which ranged from 51.4 to 79.8 days. The enantiomer fraction (EF = (+)-S-enantiomer/((-)-R-enantiomer + (+)-S-enantiomer)) values were used to describe the enantioselectivity of degradation of IM were >0.5 (P < 0.05) in two unsterilized soils under different humidity and temperature conditions. The highest EF values were observed at unsterilized CL soil samples under 50% maximum water-holding capacity (MWHC) and 25 °C environmental conditions. The EF values of the IM enantiomers were significantly higher (P < 0.05) in CL soils (higher pH = 5.81) and were 0.581 (unsterilized) and 0.575 (50% MWHC; 25 °C) compared with those recorded in SL soil (lower pH = 4.85). In addition, this study revealed that microbial organisms preferentially utilized the more herbicidal active IM enantiomer.
Preparation of white rot fungal inoculum and its application to bioremediation of chlorimuron-ethyl-contaminated soil

Abstract:Chlorimuron-ethyl is currently the primary herbicide used for chemical weed control in a soybean field. In this study, a solid microbial inoculum (corn stalk-white rot fungus (W-1)) was prepared for the remediation of farmland soil contaminated by chlorimuron-ethyl. Firstly, the preparation method of the microbial inoculum was studied. Secondly, the degradation rate of the chlorimuron-ethyl in the ground by the solid microbial inoculum is improved by optimizing the proportion of the protective agent. Then the effects of applying solid microbial inoculum, free bacteria and corn straw on the degradation rate of chlorimuron-ethyl in soil were weighed. Finally, Illumina MiSeq sequencing was used to measure the composition and diversity of bacterial and fungal communities in the ground before and after using microbial inoculum. The degradation rate of chlorimuron-ethyl in soil by solid microbial inoculum was 84.87% after 20 d using corn straw as the support, room temperature drying, 4% Ca 3 (PO 4 ) 2 as the protective drying agent, and 1%(w) dextrin as the ultraviolet protective agent. Inoculation of white rot fungi could significantly affect the community structure of bacteria and fungi in the soil, making the chlorimuron-ethyl degrading communities become the dominant communities and playing an essential role in the degradation of chlorimuron-ethyl. The results showed that using solid microbial inoculum was an effective way to repair farmland soil polluted by chlorimuron-ethyl.

Abstract:The effect of repeated chlorothalonil applications on its persistence and soil microbial activities were investigated in planted soil under greenhouse conditions. The results revealed that suppressed dissipation of chlorothalonil and the consequent accumulation of chlorothalonil and its toxic metabolite, 4-hydroxychlorothalonil, resulted from four successive applications at rates of 2, 4, and 10 kg a.i./ha. Soil respiration for 7 h (SR7) was slightly affected by chlorothalonil applications at all three levels, whereas soil respiration for 24 h (SR24) at 10 kg a.i./ha was reduced by 17.6–59.1 %, depending upon the frequency. This harmful effect of chlorothalonil on SR24 persisted throughout the experiment although it gradually weakened with time. A persistent inhibitory effect on soil dehydrogenase activity was also observed with repeated applications of chlorothalonil at 10 kg a.i./ha. This study indicated that repeated chlorothalonil applications may lead to the accumulation of chlorothalonil and its metabolite in soil under greenhouse conditions and thereby alter soil microbial activity.
New insights into the effects of chlorpyrifos on soil microbes: Carbon and nitrogen cycle related microbes in wheat/maize rotation agricultural field

Abstract:Chlorpyrifos, a broad−spectrum organophosphorus insecticide, has been widely detected worldwide and is a potential neurotoxin and endocrine disruptor. Besides, chlorpyrifos has been proven that have a negative effect on soil microbes. In the present study, chlorpyrifos formulation (LORSBAN®, 45% emulsifiable concentrate) was applied in an agricultural field at the recommended dose (R dose, 270.0 and 337.5 g a.i. ha −1 for wheat and maize respectively) and double recommended (DR) dose. Chlorpyrifos residue level and effect on soil microbes related to soil carbon and nitrogen cycle function were analyzed. Results showed that the half−lives of chlorpyrifos in wheat and maize field soil were 7.23–8.23 and 1.45–1.77 d, respectively. Application of chlorpyrifos at even DR dose did not result in unacceptable residual chlorpyrifos, where the final residual chlorpyrifos in wheat/maize (leaf, stem, and grain) was meet the requirement of the maximum residual limit (0.5 mg kg −1 for wheat and 0.05 mg kg −1 for maize) in China. Chlorpyrifos enhanced the activity of β −glucosidase by increasing the relative abundance of Sphingosinicella and promoted the carbon cycle in wheat field. The changes of cbbLR and cbbLG gene abundance also confirmed that chlorpyrifos could affect the import and export of soil carbon pool. The effect of chlorpyrifos on soil N cycle was determined by changes in the abundance of the bacterial genus Gemmatimonas , which is associated with denitrification. Further analysis of N−cycle functional genes and urease activity showed that chlorpyrifos inhibited nitrogen fixation in wheat field, but promoted nitrogen fixation in maize field. In general, bacterial abundance, urease, and AOA− amoA gene could be early warning markers of chlorpyrifos contamination. The results demonstrated the negative effects of chlorpyrifos on soil microbes especially on soil C and N cycle in actual agricultural field. It provides new insights about chlorpyrifos environmental pollution and its effect on soil ecosystems.
