Abstract:Three diets containing either borage oil (BO) and southern hemisphere fish oil Marinol (MO), or BO and tuna orbital oil (TO), or a northern hemisphere fish oil (FO) were fed to duplicate groups of turbot (Scophthalmus maximus) of initial mean weight 1.2 g for a period of 12 weeks. The BO/MO and BO/TO diets were enriched in gamma-linolenic (18:3n-6, GLA) and eicosapentaenoic (20:5n-3, EPA) acids, and GLA and docosahexaenoic acid (22:6n-3, DHA), respectively. No differences were observed in final weights or growth rates, either between duplicate tanks or between dietary treatments. Half of the FO-fed fish sampled showed a histopathological lesion indicative of lipoid liver degeneration while the other treatments only showed a slight incidence of the same pathology. The fatty acid compositions of carcass and tissues broadly reflected the dietary input. In general, fish fed the BO/MO diet had increased levels of 18:2n-6, 18:3n-6, 20:3n-6 and 20:5n-3, but a lower level of 22:6n-3, compared to fish fed FO. In fish fed the BO/TO diet, levels of 18:2n-6, 18:3n-6, 20:3n-6 and 20:4n-6 were increased while levels of 20:5n-3 and 22:5n-3 were reduced, compared to fish fed FO. Concentrations of thromboxanes B (TXB) and leukotrienes B (LTB), derived from 20:4n-6 and 20:5n-3, were measured in plasma and stimulated blood cells. Levels of TXB2 were greatest in fish fed the BO/TO diet compared to both other treatments, while LTB4 was decreased in fish fed the BO/MO diet compared to both other treatments. In a stress test which involved anaesthesia followed by measurement of recovery times, fish fed the BO/MO diet had significantly lower recovery times compared to fish fed the FO diet.
Effects of purified diets containing different combinations of arachidonic and docosahexaenoic acid on survival, growth and fatty acid composition of juvenile turbot (Scophthalmus maximus)

Abstract:The objective of this study was to determine the relative essential fatty acid (EFA) growth-promoting activities of pure arachidonic (AA, 20:4n−6) and docosahexaenoic (DHA, 22:6n−3) acids and various concentrations of these two acids in the diet of juvenile turbot (Scophthalmus maximus). Casein-based, semi-purified diets containing 15% fish oil or 14% hydrogenated coconut oil/oleic acid (1:1) supplemented with 1% 20:4n−6, 1% 22:6n−3 or 1% of various combinations of these two acids were fed to duplicate groups of 26 juvenile turbot for 11 weeks. In this trial, feeding the diet containing 20:4n−6 as the only highly unsaturated fatty acid (HUFA) resulted in higher growth and survival than any of the mixtures of the two fatty acids or 22:6n−3 alone. The diet containing 22:6n−3 as the sole HUFA resulted in the lowest growth and survival of all dietary treatments. The control diet with 15% fish oil resulted in a greater growth rate than any of the pure HUFA-supplemented diets. There was a significant effect of dietary lipid on the somatic index of the brain but not heart, kidney or liver. The percentage of lipid in the liver, but not of heart, brain, eyes, gills or kidney, was influenced by dietary lipid, with the highest percentage in fish supplemented with DHA alone. After 11 weeks, the 20:4n−6 and 22:6n−3 levels in whole-body total lipids were strongly influenced by the content of these fatty acids in the diets. The relative effect of dietary levels of these two fatty acids on their content in fish lipids varied considerably among the various organs and tissues of the fish that were analyzed. Brain and eye lipids were generally highest in 22:6n−3 while gill and kidney lipids were consistently higher in 20:4n−6 than the other organs analyzed. The effect of dietary 20:4n−6 on the content of that HUFA in organ lipid was greatest in gill and liver. The greatest impact of dietary 22:6n−3 level on content of that acid in organ lipid was seen in gill and kidney. There were also significant effects of dietary HUFA content on organ lipid levels of saturated, mono-unsaturated fatty acids and other members of the n−3 and n−6 PUFA, and HUFA series. The present study suggests that the EFA growth-promoting activity of arachidonic acid provides strong support for the contention that dietary 20:4n−6 is essential for juvenile turbot.

[Factors affecting C-peptide level during the first year of type 1 diabetes in children].

Abstract:BACKGROUND C-peptide level is the most reliable factor evaluating the endogenous insulin secretion in patients with type 1 diabetes. OBJECTIVES The aim of the study was to investigate whether the age at onset, gender, presence of autoantibodies and ketoacidosis at diagnosis and insulin requirement, HbA1c levels could be applied to predict the C-peptide levels in the first year of type 1 diabetes in children. MATERIAL AND METHODS 122 type 1 diabetic children, aged: 2-18 years (average 11.2), 44 female and 78 male were studied. Fasting C-peptide levels were examined by radioimmunoassay at diagnosis, after 10 days and after 1, 2, 3, 6 and 12 months of disease. At diagnosis islet cell antibodies (ICA) were detected by indirect immunofluorescence, antibodies to glutamic acid decarboxylase (GADA) and tyrosine phosphatase antibodies (IA2A) were measured by microradioimmunoprecipitation assay. RESULTS Age at onset was positively correlated to C-peptide levels at each evaluated point of the disease (r=0.3-0.46, p<0.0001). One year after diagnosis C-peptide levels decreased in ICA(+) (p<0.04) and GADA(+) (p<0.002) patients but not in ICA(-) or GADA(-) children. There was no significant difference between the IA2A-positive and negative subjects in the C-peptide levels at 12th month of disease. C-peptide level was also related to ketoacidosis at diagnosis, insulin requirement and HbA1c levels during the first year of type 1 diabetes. Logistic regression analysis showed that male, younger age, low pH, higher HbA1c and insulin requirement at onset were associated with decreased C-peptide level at diagnosis (p<0.00002). CONCLUSIONS Young age, presence of diabetes-related autoantibodies and hyperglycaemia with severe acidosis at the disease onset may be associated with a decreased residual insulin secretion in type 1 diabetes in children.

Abstract:Three 56-day feeding trials were conducted to evaluate dietary lipid requirements and effects of dietary lipid levels on growth, digestive enzyme activities, antioxidant capacity, and lipid metabolism in turbot (Scophthalmus maximus L.) at three growth stages. The initial mean body weight of turbot was 9.13 ± 0.17 g, 50.10 ± 0.38 g, and 80.05 ± 0.76 g (small, medium, and large turbot), respectively. Five practical diets were formulated to contain 68.4, 93.9, 120.7, 147.8, and 171.2 g/kg lipid (L68.4, L93.9, L120.7, L147.8, and L171.2), respectively. Results of three trials showed that the weight gain rate and specific growth rate of turbot fed with L120.7, L147.8, and L171.2 diets were all significantly higher compared with turbot fed with L68.4 and L93.9 diets except for large turbot fed with the L147.8 diet. Activities of intestinal trypsin, lipase and hepatic catalase, superoxide dismutase, and total antioxidant capacity firstly increased and then decreased as dietary lipids increased. Meanwhile, malondialdehyde content decreased firstly but then increased with the elevation of dietary lipids in small and medium turbot, and it was significantly higher in the L171.2 group than the rest in large turbot. With increasing levels of dietary lipid, contents of whole-body lipid, liver lipid, serum total cholesterol, triglyceride, and low-density lipoprotein cholesterol were markedly increased at three stages. In addition, serum high-density lipoprotein cholesterol contents increased firstly and then decreased over the L120.7 group. Transcriptional levels of lipolysis-related genes lipin1 and lpl were significantly upregulated firstly and subsequently downregulated with increasing dietary lipid levels except lipin1 in medium fish. Meanwhile, a significant increase in lipogenesis-related genes lxr and pparγ expressions was detected in all fish. Based on the specific growth rate, the dietary lipid level of 130.1, 120.1, and 107.7 g/kg was optimal for the growth performance of turbot cultured at three phases, respectively. Additionally, the results indicated that high lipid diets may lead to abnormal lipid deposition and affect the physiological health of turbot.

Abstract:An eight-week feeding trial was conducted to examine the impact of dietary supplementation with glycerol monolaurate (GML) on juvenile black sea bream. A basal diet was formulated containing 24% fish meal, while five additional diets were prepared, each supplemented with varying levels of GML: GML1 (0.01%), GML2 (0.02%), GML3 (0.04%), GML4 (0.08%), and GML5 (0.16%). Triplicate tanks were randomly allocated to each diet, each containing 20 fish with an initial weight of 1.55 ± 0.05 g. By the trial’s end, the GML3 group displayed a notably higher final body weight (FBW), weight gain (WG), specific growth rate (SGR), and protein efficiency ratio (PER) compared to the other groups (p < 0.05), but the FCR was significantly higher in the control group. However, no significant differences were observed in the MFI, PPV, CF, HSI, IPF, VSI, or SR among the groups (p > 0.05). Regarding the proximate compositions of the dorsal muscle and whole body, no substantial differences were observed across the groups (p > 0.05). Additionally, there were no significant variations in digestive enzyme activity (p > 0.05), serum immune, or biochemical parameters in the midgut and hindgut among the treatment groups. But in the serum immune response IgM, C3 and C4 were significantly higher in the GML3 group as compared to the other groups (p < 0.05). However, the GML3 group exhibited significantly greater fore-intestinal villus height, crypt depth, villus height per crypt depth, and the number of goblet cells per villus compared to the other groups (p < 0.05). Overall, GML supplementation, particularly GML3, significantly improved growth indicators like the final body weight and intestinal morphology. While certain parameters remained unaffected, these findings suggest GML’s potential as a beneficial dietary supplement in fish diets.

Abstract:A 56-day feeding trial was conducted in a flow-through seawater system to investigate the effects of lysophosphatidylcholine (LPC) on growth performance and lipid metabolism of turbot. Four experimental diets were prepared, differing only in the LPC supplementation, namely, 0 (LPC-0, control), 0.1 (LPC-0.1), 0.25 (LPC-0.25), and 0.5% (LPC-0.5) of dry matter. Each diet was randomly fed to triplicate tanks. LPC-0.1 and LPC-0.25 led to significantly higher weight gain than the control diet, and the highest weight gain was observed in LPC-0.1. Compared to the control group, the LPC-supplemented groups had higher survival and lower hepatosomatic index and viscerosomatic index. LPC-0.25 led to significantly lower contents of crude lipid and ash in whole fish. Dietary LPC supplementation led to a basic decrease in the lipid metabolism-related biochemical parameters in serum but had only very minor influence on the fatty acid composition in the liver and subcutaneous tissue around the fin (STF). High LPC levels upregulated the mRNA expression of BSAL and ApoEα in both the liver and STF. In conclusion, dietary LPC supplementation (0.1-0.25%) enhanced the growth, lowered the lipid accumulation in juvenile turbot, and significantly regulated the lipid metabolism. However, it seldom influenced the fatty acid composition.

Fish oil replacement by beef tallow in juvenile turbot diets: Effects on growth performance, body composition and volatile flavor compounds in the muscle

Abstract:A 12-week feeding trial was conducted to evaluate the effects of dietary fish oil (FO) replacement by beef tallow (BT) on the growth performance and body composition of juvenile turbot (average initial weight, 19.6 g). The control diet contained 6% added FO (of dry matter, Diet FO-C). Beef tallow was supplemented into Diet FO-C, replacing added FO at different levels, i.e., 25%, 50%, 75%, and 100%, formulating the other 4 experimental diets, designated as 25BT, 50BT, 75BT, and 100BT, respectively. The feeding trial was conducted in a flow-through seawater system. Each diet was randomly fed to triplicate tanks (30 fish in each tank). The results showed that a cubic regression relationship was observed between weight gain and dietary FO replacement level. Although total FO replacement by BT significantly reduced the growth of juvenile turbot, no significant difference was observed between FO-C and 75BT. Diet BT decreased the whole-body crude lipid content but increased the whole-body moisture and ash contents. Dietary BT regulated the proximate composition of fish tissues in a dose dependent manner. High dietary BT levels did not reduce the n-3 long-chain polyunsaturated fatty acid content (% total fatty acid) in the muscle. Partial FO replacement (25 and 50%) by BT even significantly increased the muscle DHA content. The fatty acid composition of the liver, subcutaneous adipose tissue around the fin, and intestine generally reflected those of the diets. No significant difference was observed in muscle and liver mitochondrial DNA copy number among groups, indicating the similar general energy supply status. Dietary BT significantly reduced the concentration of triacylglycerol, cholesterol and malondialdehyde in the serum. The volatile flavor compound in muscle was analyzed with gas chromatography-ion migration spectrometry (GC-IMS). Dietary BT resulted in lower number and abundance of flavor aldehydes and alcohols in the muscle. In conclusion, beef tallow can replace 75% added fish oil (appr. 45% of total fish oil including the residual fish oil in fishmeal) in the diets of turbot, without reducing fish growth. Dietary beef tallow significantly affected the body composition and volatile flavor compound composition in the muscle.

Abstract:The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, and lipid metabolism in juvenile hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lancolatus ♂). Five isonitrogenous and isolipidic diets with graded levels of phospholipids (14.10, 32.10, 52.50, 70.20, and 91.30 g/kg) were fed to triplicate groups of juvenile grouper (initial body weight 13.00 ± 0.02 g) for 61 days. Results showed that dietary phospholipids produced no statistical influence on growth performance of grouper. The fatty acids profiles of experimental fish are correlated well with dietary fatty acids composition. The increase of dietary phospholipids significantly elevated the crude lipid content in whole fish body, liver and muscle. Meanwhile, the increased dietary phospholipids significantly promoted hepatic triglyceride (TG) accumulation. The results of gene expression analysis revealed that dietary phospholipids did not significantly affect the expression of lipogenesis related genes, FAS, ACC and DGAT2. In addition, high dietary phospholipids inclusion significantly decreased the expression of fatty acids oxidation related genes, PPARɑ and CPT1. In summary, juvenile grouper don’t have a specific requirement for dietary phospholipids which inclusion aggravated lipid deposition through the regulation of fatty acids oxidation at the transcriptional level.
Integrated Response of Growth Performance, Fatty Acid Composition, Antioxidant Responses and Lipid Metabolism to Dietary Phospholipids in Hybrid Grouper (epinephelus Fuscoguttatus ♀ × E. Lanceolatus ♂) Larvae

Abstract:Dietary phospholipids supplementation is essential for normal development of fish larvae due to their limited phospholipid biosynthetic ability. The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, antioxidant capacity and lipid metabolism of larval hybrid grouper. The larvae (40 day after hatching, 0.37 +/- 0.01 g) were fed the diet with graded level of phospholipids (1.87, 3.61, 5.53, 7.25 and 9.69%) four times daily for 23 days. Results showed that the specific growth rate of grouper larvae was significantly improved with the increase of dietary phospholipids, and optimal dietary phospholipids was estimated to be 9.17%. Fatty acids composition of whole fish body and liver was highly reflective of dietary fatty acid profile. In addition, the increase of dietary phospholipids resulted in a significantly elevated hepatic crude lipid and triglyceride content in a dose-dependent manner. The analysis of gene expression revealed that increased dietary phospholipids did not produce any statistical effect on the expression of lipogenesis genes including LXRalpha, FAS and ACC, but significantly facilitated the expression of DGAT2, a rate-limiting enzyme involved in triglyceride synthesis. The expression of genes, CPT1, involved in I3-oxidation was not significantly influenced as dietary phospholipids increased. However, phospholipids inclusion significantly reduced the expression of ATGL, an rate-limiting lipolytic enzyme. Additionally, the supplementation of dietary phospholipids significantly elevated the activity of antioxidant relate enzymes, CAT, SOD and T-AOC, but significantly decreased the MDA content, an end product of fatty acid peroxidation. In summary, dietary phospholipids supplementation was beneficial in improving growth performance and antioxidant capacity of grouper larvae, and meanwhile, promoted hepatic lipid deposition through regulating the synthesis and hydrolysis of triglyceride.
Dietary salt concentrations influence growth, nutrient utilization, and fatty acid profiles of turbot (Scophthalmus maximus) reared in brackish water

Abstract:Expansion of economically viable turbot (Scophthalmus maximus) aquaculture depends on access to brackish-cold ground water sources in various parts of the world. Since brackish water sources can adversely affect the physiology and zoo technical performance of fish due to the burden of osmoregulation, dietary salt inclusion can alleviate the negative impacts of low-saline waters in several aquaculture species. This study investigated the effects of increasing dietary salt levels on the growth, feed utilization, body composition, and tissue fatty acid composition of juvenile turbot (initial live weight 120.3 ± 0.03 g/fish). Fish were fed five experimental diets supplemented with varying levels of sodium chloride (1.8-6.4%) or a control diet without salt. Each diet was tested in triplicate tanks for 9 weeks. Results showed that increasing dietary salt intake negatively impacted turbot performance, with significant reductions in weight gain, specific growth rate, and feed conversion ratio. Dry matter and ash content in the whole body and filet increased quadratically with increasing salt levels, whereas gill moisture and protein content decreased linearly. Furthermore, the nitrogen, lipid, and energy utilization efficiencies decreased with their respective intake and gain levels. Dietary salt significantly influenced the fatty acid profiles of gill, liver, and filet tissues. In the gill, monounsaturated fatty acids (16:1n-7, ΣMUFA) and n-6 PUFA (20:2n-6) increased, whereas EPA and DHA decreased. Liver ΣSFA (16:0, 18:0) increased, and n-3 PUFA (18:3n-3, 20:5n-3) decreased with increasing dietary salt. Filet saturated fatty acids (14:0, 15:0, 17:0) and n-6 PUFA (20:2n-6, 20:4n-6) increased, while n-3 PUFA (18:3n-3, EPA) decreased with dietary salt. DHA levels in filets showed a quadratic increase. Overall, this study shows that increasing dietary salt negatively impacts turbot growth, feed utilization, and tissue fatty acid composition in brackish water, highlighting the need for further studies on salinity management strategies for turbot aquaculture.
EPA and DHA promote cell proliferation and enhance activity of the Akt-TOR-S6K anabolic signaling pathway in primary muscle cells of turbot (Scophthalmus maximus L.)

Abstract:Fish growth and health are predominantly governed by dietary nutrient supply. Although the beneficial effects of omega-3 polyunsaturated fatty acids supplementation have been shown in a number of fish species, the underlying mechanisms are still mostly unknown. In this study, we conducted an investigation into the effects of EPA and DHA on cell proliferation, nutrient sensing signaling, and branched-chain amino acids (BCAA) transporting in primary turbot muscle cells. The findings revealed that EPA and DHA could stimulate cell proliferation, promote protein synthesis and inhibit protein degradation through activation of target of rapamycin (TOR) signaling pathway, a pivotal nutrient-sensing signaling cascade. While downregulating the expression of myogenin and myostatin, EPA and DHA increased the level of myogenic regulatory factors, such as myoD and follistatin. Furthermore, we observed a significant increase in the concentrations of intracellular BCAAs following treatment with EPA or DHA, accompanied by an upregulation of the associated amino acid transporters. Our study providing valuable insights into the mechanisms underlying the growth-promoting effects of omega-3 fatty acids in fish.
Black Soldier Fly Oil in Different Lipid Diets Could Regulate Tissue Lipid Metabolism and Fatty Acid Composition of Juvenile Mirror Carp

Abstract:In the present study, our aim was to assess the effect of dietary black soldier fly oil (BSFO) with different lipid contents on the growth performance, body composition, lipid metabolism, and related gene expression in juvenile mirror carp (Cyprinus carpio var. specularis). An 8-week feeding trial was conducted using four isonitrogenous diets (32.0% crude protein) containing two lipid levels: 6% (control (CT) group) and 9% (high lipid (HL) group), both using soybean oil as the oil source. The other two diets contained BSFO on the basis of 6% and 9% lipid, referred to as (CT + BSFO) and (HL + BSFO) groups. The results showed that final body weight, weight gain rate, specific growth rate, protein retention ratio, and feed utilization were significantly increased by using higher dietary lipid levels ( P < 0.05 ). Additionally, the gene expression of lipid synthesis markers was significantly up-regulated in intra-peritoneal fat but significantly down-regulated in the hepatopancreas due to increased dietary lipid level ( P < 0.05 ). No negative effects on feed utilization or growth performance were observed in fish fed diets containing BSFO. However, the intra-peritoneal fat index, adipocyte size, and hepatopancreas lipid content were significantly decreased in the CT + BSFO group compared to the CT group ( P < 0.05 ). Furthermore, BSFO also up-regulated the expression of lipid lipolysis markers in the hepatopancreas and intra-peritoneal fat ( P < 0.05 ). Moreover, the use of BSFO also increased the content of n-3 polyunsaturated fatty acid (PUFA) while reducing the content of n-6 PUFA in the muscle ( P < 0.05 ). In conclusion, HL diets improved the growth of the fish and increased the lipid deposition. BSFO in the diet resulted in a reduction of lipid deposition in the hepatopancreas and intra-peritoneal fat, likely due to an increase in lipid oxidation.

Abstract:The aim of this study was to investigate the effects of feeding alternative dietary oils to hybrid grouper fish (♀Epinephelus fuscoguttatus × ♂E. lanceolatu) on their growth, histological morphology of hepatocytes, disease resistance, immune response, and expression of immune-related genes. Seven experimental fish meal-based isonitrogenous and isolipidic diets were formulated containing 5% fish oil (FO; acting as controls) and various vegetable oils (VOs): corn oil (CO), sunflower oil (SO), tea oil (TO), olive oil (OO), rice oil (RO), and mixed oil (MO); comprising equal amounts of these oils). Each diet was fed to triplicate groups of 40 fish (initial mean body weight ± standard error = 15.09 ± 0.01 g) for eight weeks. The results show that 1) alternative dietary oils had no significant effects on weight gain rate, specific growth rate, protein efficiency ratio, and survival rate compared with controls (P > 0.05). The weight gain rate (WGR) and specific growth rate (SGR) of the SO group were lower than in the CO and OO groups. 2) These were no differences in morphological indexes among groups; except for the CO group, in which the condition factor and hepatosomatic index were lower than those in other groups. 3) Compared with controls, the whole-body moisture and crude protein contents in the VO groups were higher, while their crude lipid contents were lower. 4) The fatty acid contents in liver and muscle were affected by lipid type, and the contents of eicosapentaenoic acid and docosahexaenoic acid in liver and muscle in the VO groups were markedly lower than in controls. 5) Compared with control group, VO groups damaged the histological morphology of hepatocytes. 6) After a challenge with the Vibrio parahaemolyticus bacterium, there were no differences in mortality among groups. However, VO enhanced the activity of non-specific immune enzymes while down-regulating the expression of Nrf2 and inducing the expression of pro-inflammatory factors (IL1β, TNFα, TLR22, and MyD88) in the kidney. It can be concluded that dietary VO substitution does not affect the growth of fish but damaged the histological morphology of hepatocytes and induced the expression of pro-inflammatory factors in tissues. Finally, OO and CO were recommended as the appropriate lipid replacement for FO.
The Importance of Fatty Acid Precision Nutrition: Effects of Dietary Fatty Acid Composition on Growth, Hepatic Metabolite, and Intestinal Microbiota in Marine Teleost Trachinotus ovatus

Abstract:Our recent study demonstrated that diet with blend oil (named BO1) as lipid, which is designed on the base of essential fatty acid requirement of Trachinotus ovatus, achieved good performance. Here, to confirm its effect and investigate the mechanism, three isonitrogenous (45%) and isolipidic (13%) diets (D1-D3) only differing in dietary lipids, which were, respectively, fish oil (FO), BO1, and blend oil 2 (BO2) consisting of FO and soybean oil at 2 : 3, were formulated and used to feed the T. ovatus juveniles (average initial weight: 7.65 g) for 9 weeks. The results showed that the weight gain rate of fish fed D2 was higher than that of fish fed D3 (P < 0.05) and had no significant difference from that of fish fed D1 (P > 0.05). Correspondingly, compared with the D3 group, fish of the D2 group exhibited better oxidative stress parameters such as lower serum malondialdehyde content and inflammatory indexes in the liver such as the lower expression level of genes encoding four interleukin proteins and tumor necrosis factor α, as well as higher hepatic immune-related metabolites such as valine, gamma-aminobutyric acid, pyrrole-2-carboxylic acid, tyramine, l-targinine, p-synephrine, and butyric acid (P < 0.05). Furthermore, the intestinal probiotic (Bacillus) proportion was significantly higher, while the pathogenic bacteria (Mycoplasma) proportion was significantly lower in the D2 group than that in the D3 group (P < 0.05). The main differential fatty acids of diet D2 were close to those of D1, while the levels of linoleic acid and n-6 PUFA, as well as the ratio of DHA/EPA of D3, were higher than those of D1 and D2. These results indicated that the better performance of D2 such as enhancing growth, reducing oxidative stress, and improving immune responses and intestinal microbial communities in T. ovatus may be mainly due to the good fatty acid composition of BO1, which indicated the importance of fatty acid precision nutrition.

Abstract:Marine oils are important to human nutrition as the major source of docosahexaenoic acid (DHA), a key omega-3 long-chain (≥C20) polyunsaturated fatty acid (n-3 LC-PUFA) that is low or lacking in terrestrial plant or animal oils. The inclusion of fish oil as main source of n-3 LC-PUFA in aquafeeds is mostly limited by the increasing price and decreasing availability. Fish oil replacement with cheaper terrestrial plant and animal oils has considerably reduced the content of n-3 LC-PUFA in flesh of farmed Atlantic salmon. Novel DHA-enriched oils with high alpha-linolenic acid (ALA) content will be available from transgenic oilseeds plants in the near future as an alternative for dietary fish oil replacement in aquafeeds. As a preliminary validation, we formulated an oil blend (TOFX) with high DHA and ALA content using tuna oil (TO) high in DHA and the flaxseed oil (FX) high in ALA, and assessed its ability to achieve fish oil-like n-3 LC-PUFA tissue composition in Atlantic salmon smolts. We applied proteomics as an exploratory approach to understand the effects of nutritional changes on the fish liver. Comparisons were made between fish fed a fish oil-based diet (FO) and a commercial-like oil blend diet (fish oil + poultry oil, FOPO) over 89 days. Growth and feed efficiency ratio were lower on the TOFX diet. Fish muscle concentration of n-3 LC-PUFA was significantly higher for TOFX than for FOPO fish, but not higher than for FO fish, while retention efficiency of n-3 LC-PUFA was promoted by TOFX relative to FO. Proteomics analysis revealed an oxidative stress response indicative of the main adaptive physiological mechanism in TOFX fish. While specific dietary fatty acid concentrations and balances and antioxidant supplementation may need further attention, the use of an oil with a high content of DHA and ALA can enhance tissue deposition of n-3 LC-PUFA in relation to a commercially used oil blend.

Abstract:Clostridium autoethanogenum meal (CAM) is a novel single-cell protein, which is produced from bacteria using carbon monoxide (CO) as sole carbon source. To evaluate the efficiency of CAM as an alternative for dietary fish meal, a 56-days growth experiment was performed on juvenile turbot ( Scophthalmus maximus L.) with initial average weight of 9.13 ± 0.02 g. Six iso-nitrogenous (crude protein, 51.0%) and iso-lipidic (crude lipid, 11.5%) diets were formulated with 0%, 15%, 30%, 45%, 60% and 80% dietary fish meal protein substituted by CAM protein, which were designated as CAM0 (the control group), CAM15, CAM30, CAM45, CAM60 and CAM80, respectively. Results showed that no significant differences were observed in survival rate (over 97.50%) among different dietary treatments ( p > 0.05). The specific growth rate (SGR) was not significantly affected when replacement levels of dietary fish meal with CAM were less than 45% ( p > 0.05). The feed intake (FI) was significantly linear reduced with increasing dietary CAM ( p 0.05). With increasing dietary CAM, lipid retention (LR) and carcass lipid tended to be increased in both significantly linear and quadratic patterns ( p < 0.05). The apparent digestibility coefficient (ADC) of crude protein and some essential amino acids, including threonine, valine, lysine, histidine and arginine, showed significantly linear increase with increasing dietary CAM ( p < 0.05). Furthermore, with the increase of dietary CAM, the gene expression of intestinal peptide and amino acids transporters was first up-regulated and then down-regulated with significantly quadratic pattern ( p < 0.05), peaking in fish fed with diets CAM30 or CAM45, which was similar to the expression of genes related protein degradation in muscle. For genes related to protein metabolism in liver and muscle, the expression of mammalian target of rapamycin ( mtor ) was not significantly affected by dietary CAM, while the general control nonderepressible 2 ( gcn2 ) tended to be first up-regulated and then down-regulated with significantly quadratic pattern ( p < 0.05). Apart from that, the lipid metabolism of turbot was also affected by high dietary CAM, evidenced by increased expression of hepatic genes related to lipogenesis as well as reduced expression of genes related to lipid oxidation and lipid transport. In conclusion, CAM can replace up to 45% fish meal protein in diet for juvenile turbot without significantly adverse effects on growth performance. But excessive dietary CAM would result in significant growth reduction, and excessive lipid deposition may also occur in fish fed diets with high levels of CAM.

In this review, we describe the basis of this interaction, as well as its clinical relevance to such problems as acute respiratory distress syndrome, percutaneous transluminal coronary angioplasty, and transplant allograft survival.

Abstract:An 8-week feeding trial was conducted to investigate the effects of dietary docosahexaenoic to eicosapentaenoic acid ratio (DHA/EPA) on growth performance, fatty acid profiles, antioxidant capacity, hematological characteristics and expression of some lipid metabolism related genes of juvenile black seabream (Acanthopagrus schlegelii) of initial weight 9.47 ± 0.03 g. Five isonitrogenous and isolipidic diets (45% crude protein and 14% crude lipid) were formulated to contain graded DHA/EPA ratios of 0.65, 1.16, 1.60, 2.03 and 2.67. There were no differences in growth performance and feed utilization among treatments. Fish fed higher DHA/EPA ratios had higher malondialdehyde (MDA) contents in serum than lower ratios. Serum triacylglycerol (TAG) content was significantly higher in fish fed the lowest DHA/EPA ratio. Tissue fatty acid profiles reflected the diets despite down-regulation of LC-PUFA biosynthesis genes, fatty acyl desaturase 2 (fads2) and elongase of very long-chain fatty acids 5 (elovl5), by high DHA/EPA ratios. Expression of acetyl-CoA carboxylase alpha (accα) and carnitine palmitoyl transferase 1A (cpt1a) were up-regulated by high DHA/EPA ratio, whereas sterol regulatory element-binding protein-1 (srebp-1) and hormone-sensitive lipase (hsl) were down-regulated. Fatty acid synthase (fas), 6-phosphogluconate dehydrogenase (6pgd) and peroxisome proliferator-activated receptor alpha (pparα) showed highest expression in fish fed intermediate (1.16) DHA/EPA ratio. Overall, this study indicated that dietary DHA/EPA ratio affected fatty acid profiles and significantly influenced lipid metabolism including LC-PUFA biosynthesis and other anabolic and catabolic pathways, and also had impacts on antioxidant capacity and hematological characteristics.

Abstract:The impact of replacing circa 70 % fish oil (FO) by a vegetable oil (VO) blend (rapeseed, linseed, palm oils; 20:50:30) in diets for European sea bass juveniles (IBW 96 ± 0.8 g) was evaluated in terms of activities of digestive enzymes (amylase, lipase, alkaline phosphatase, trypsin and total alkaline proteases) in the anterior (AI) and posterior (PI) intestine and tissue morphology (pyloric caeca—PC, AI, PI, distal intestine—DI and liver). For that purpose, fish were fed the experimental diets for 36 days and then liver and intestine were sampled at 2, 6 and 24 h after the last meal. Alkaline protease characterization was also done in AI and PI at 6 h post-feeding. Dietary VO promoted higher alkaline phosphatase activity at 2 h post-feeding in the AI and at all sampling points in the PI. Total alkaline protease activity was higher at 6 h post-feeding in the PI of fish fed the FO diet. Identical number of bands was observed in zymograms of alkaline proteases of fish fed both diets. No alterations in the histomorphology of PC, AI, PI or DI were noticed in fish fed the VO diets, while in the liver a tendency towards increased hepatocyte vacuolization due to lipid accumulation was observed. Overall, and with the exception of a higher intestine alkaline phosphatase activity, 70 % FO replacement by a VO blend in diets for European sea bass resulted in no distinctive alterations on the postprandial pattern of digestive enzyme activities and intestine histomorphology.

Abstract:A nine-week feeding trial was conducted to investigate changes in the intestinal microbiota of turbot in response to alternate feeding between terrestrially sourced oil (TSO)- and fish oil (FO)-based diets. The following three feeding strategies were designed: (1) continuous feeding with the FO-based diet (FO group); (2) weekly alternate feeding between soybean oil (SO)- and FO-based diets (SO/FO group); and (3) weekly alternate feeding between beef tallow (BT)- and FO-based diets (BT/FO group). An intestinal bacterial community analysis showed that alternate feeding reshaped the intestinal microbial composition. Higher species richness and diversity of the intestinal microbiota were observed in the alternate-feeding groups. A PCoA analysis showed that the samples clustered separately according to the feeding strategy, and among the three groups, the SO/FO group clustered relatively closer to the BT/FO group. The alternate feeding significantly decreased the abundance of Mycoplasma and selectively enriched specific microorganisms, including short-chain fatty acid (SCFA)-producing bacteria, digestive bacteria (Corynebacterium and Sphingomonas), and several potential pathogens (Desulfovibrio and Mycobacterium). Alternate feeding may maintain the intestinal microbiota balance by improving the connectivity of the ecological network and increasing the competitive interactions within the ecological network. The alternate feeding significantly upregulated the KEGG pathways of fatty acid and lipid metabolism, glycan biosynthesis, and amino acid metabolism in the intestinal microbiota. Meanwhile, the upregulation of the KEGG pathway of lipopolysaccharide biosynthesis indicates a potential risk for intestinal health. In conclusion, short-term alternate feeding between dietary lipid sources reshapes the intestinal microecology of the juvenile turbot, possibly resulting in both positive and negative effects.
