Abstract:Solar-induced chlorophyll (chl) fluorescence (SIF) has been shown to be positively correlated with vegetation photosynthesis, suggesting that it is a useful signal for understanding of environmental responses and spatial heterogeneity of photosynthetic activity at various scales from leaf to the globe. Photosynthesis is often inhibited in stressful environments (photoinhibition), but how photoinhibition influences the relationship between photosynthesis and chl fluorescence remains unclear. Here, I studied light energy allocation among photosynthesis, chl fluorescence and heat dissipation in photoinhibited leaves and tested whether photosynthesis in photoinhibited leaves can be evaluated from chl fluorescence and reflectance spectra in remote sensing. Chl fluorescence and reflection spectra were examined with the pulse amplified modulation (PAM) system and spectroradiometer, respectively. Photoinhibited leaves had lower photosynthetic rates and quantum yields of photochemistry (ΦP) and higher chl fluorescence yields. Consequently, photosynthesis was negatively correlated with chl fluorescence, which contrasts the positive relationships between photosynthesis and SIF observed in past remote sensing studies. This suggests that vegetation photosynthesis evaluated solely from chl fluorescence may be overestimated if the vegetation is dominated by severely photoinhibited leaves. When a model of energy allocation was applied, ΦP estimated from chl fluorescence and photochemical reflectance index (PRI) significantly correlated with the observed ΦP, suggesting that the model is useful to evaluate photosynthetic activities of photoinhibited leaves by remote sensing.

Abstract:Inactivation of the chlL gene in Synechocystis sp. PCC 6803 resulted in negligible chlorophyll content when the mutant was grown in darkness. Upon phycocyanin excitation at 580 nm, the 77K fluorescence spectrum of dark-grown cells showed three peaks at 648 nm, 665 nm, and 685 nm, this last being the largest. This reflects the functional presence of major components of phycobilisomes, including phycocyanin, allophycocyanin, and the terminal emitter, and efficient energy transfer between these components. As expected, no fluorescence emission peaks corresponding to chlorophyll in the photosystems were observed. Intact phycobilisomes could be isolated from the dark-grown chlL-deletion mutant. However, the phycobilisomes had a lower efficiency of energy transfer than did those isolated from the light-grown mutant, probably because of a decreased phycobilisome stability in the absence of chlorophyll. Exposing the dark-grown chlL-deletion mutant to light triggered the biosynthesis of chlorophyll. For the first 6 h in the light, upon phycocyanin excitation at 580 nm, the 77K fluorescence emission spectrum of greening cells was identical to that of dark-grown cells that lacked significant amounts of chlorophyll. With increased chlorophyll synthesis, gradual energy transfer from phycobilisomes to the two photosystems can be demonstrated.

Abstract:Chlorophyll fluorescence analysis is one of the most powerful and widely used techniques to study the effect of stresses on the photosynthetic process. From the first utilization, the F v/F m ratio has been largely used as a sensitive indicator of plant photosynthetic performance. Decreases of this index are indicative of the reduction of photosystem II (PSII) efficiency, namely photoinhibition. In the last 20 years, application of chlorophyll fluorescence has been largely improved, and many other informative parameters have been established to detect PSII photochemical efficiency and the partitioning of light energy to alternative dissipative mechanisms (qE, energy-dependent quenching; qZ, zeaxanthin-dependent quenching and qI, photoinhibitory quenching; qH, sustained photoprotective antenna quenching; qM, quenching dependent to chloroplast movement; qT, light harvesting complexes II-I state-transition) such as the recently developed "photoprotective power" of non-photochemical quenching (pNPQ). This review reports a brief description of the main chlorophyll fluorescence parameters and a wide analysis of the current bibliography on the use of different parameters which are useful to detect events of PSII photoinhibition. In addition, in view of the inherent differences in morpho-anatomical, physiological and biochemical features between C3 and C4 metabolism, possible differences in terms of photoinhibition between C3 and C4 plant species under stress conditions are proposed. The attempt is to highlight the limits of their comparison in terms of susceptibility to photoinhibition and to propose direction of future research which, assisted by chlorophyll fluorescence, should improve the knowledge of the different sensitivity of C3 and C4 to abiotic stressors.

Abstract:Fluctuating light (FL) can induce the preferential photoinhibition of photosystem I (PSI) in angiosperms. However, the underlying mechanisms have not been fully clarified. In this study, we examined the effects of low-light phases on FL-induced photoinhibition of PSI in Bletilla striata. We found that PSI was highly over-reduced within the first 20 s after transition from 59 to 1455 μmol photons m−2 s−1. However, such severe over-reduction of PSI was not observed in FL transition from 132 or 272–1455 μmol photons m−2 s−1. As a result, FL (59–1455) induced stronger PSI photoinhibition than FL (132–1455) and FL (272–1455). During low-light phase, a higher light intensity induced a higher proton gradient (ΔpH) across the thylakoid membranes, favoring the rapid formation of ΔpH after light increased and preventing an over-reduction of PSI. These results indicate that the low-light phase significantly affects the extent of PSI photoinhibition in FL. After transition from 59 or 132–1455 μmol photons m−2 s−1, CEF activity first increased and then decreased to the steady-state. However, such transient CEF stimulation was not observed during the transition from 272–1455 μmol photons m−2 s−1. Furthermore, the CEF activation in the high-light phase was highly dependent on the PSI reduction state. We conclude that CEF activation is finely regulated to optimize the trade-off between photoprotection and light use efficiency in FL.

PHOTOSYNTHESIS IN CHLORELLA DURING INTERM\IITTENT ILLUMINATION OF LONG PERIODICITY1

Abstract:Subsequent to a dark period of several minutes the rate of photosynthesis during the first seconds of illumination is generally less than that in the subsequent steady state. This phenomenon has been described by the general term "induction." Recently, Burk and Warburg (1), using a manometric method, determined pressure changes during light and dark for a suspension of Chlorella which was illuminated for periods of three minutes or less alternated with equal dark periods. The pressure changes were interpreted as demonstrating that under these conditions the rate of oxygen production and carbon dioxide consumption during the light periods exceeded that of photosynthesis during the steady state; similarly the oxygen consumption and carbon dioxide production in the dark were greater than the steady state rate of respiration. These experiments are subject to criticism because it appears improbable that during periods as short as one minute the manometer can respond sufficiently rapidly so that the observed pressure changes represent the metabolic rates without the complication of physical lag. Subsequently Damaschke, T6dt, Burk and Warburg (2) determined the consumption and production of oxygen using an electrochemical method. Few results have yet been published; these show that after a period of illumination from 5 to 30 seconds the oxygen consumption immediately following the cessation of illumination was greater than in the preceding or subsequent steady state. The effect was considerably smaller than that observed in the earlier manometric studies. The production of oxygen during the period of illumination corresponded to a quantum efficiency of 0.69 mole/mole quantum whereas measurements in the steady state gave a value of approximately 0.3. On the basis of these observations Warburg, Geleick and Briese (3) formulated a mechanism for photosynthesis which postulated that during illumination the photochemical process was accompanied by an accelerated combustion process. During the steady state the net effect of the combined processes is observed; the two can only be observed independently as transitory phenomena during periods of alternate light and dark. Brackett, Olson and Crickard (4), who also used a polarographic method to determine the time course of oxygen consumption and production by Chlorella, were unable to observe transient phenomena of the type described by Burk, Warburg and collaborators.

Abstract:Abstract Plants are constantly exposed to changing environments, sometimes leading to extreme conditions and stress. For example, sudden exposure to high light leads to excess absorbed light energy, causing reactive oxygen species (ROS) formation. ROS damage the photosynthetic machinery, particularly the D1 protein in photosystem II (PSII), which therefore needs to be continuously repaired and replaced. The effect of the damage inflicted by high light is a prolonged decrease in photosynthetic efficiency. Hence, it is not surprising that photoinhibition has been subject to numerous experimental studies investigating its effects in the context of crop productivity. However, it has become apparent that classical measures of photoinhibition, i.e., changes in the chlorophyll fluorescence parameter Fv/Fm, are not only determined by the loss of PSII core function but also by processes such as energy transfer and quenching. Mathematical models can help dissect the influences on such fluorescence signals and quantify the contributions of various interacting mechanisms. We present a mathematical model with a dynamic description of the photosynthetic electron transport chain (PETC), non- photochemical quenching, and photoinhibition. With our model, we investigate the interconnection between quenching, photoprotection, and fluorescence using simulations and experimental data. We found that different energy-dissipating properties of intact and damaged PSIIs, as well as energy transfer between PSIIs, are critical components that need to be included in the model to ensure a satisfactory fit to the experimental data. We envisage that our model provides a framework for future investigations of photoinhibition dynamics and its importance for plant growth and yield.

Abstract:The inhibitory effect of high photon flux densities (PFD) on the growth of Spirulina platensis and its sequent recovery were investigated using photoautotrophic and mixotrophic cultures. The light-dependent O-2 evolution rate and variable chlorophyll fluorescence were measured after the cultures had been exposed to high PFD. Marked decreases in these two parameters of both cultures were found but the dropping rate and also the extent of mixotrophic cultures were slower and smaller. In adding chloramphenicol (CAP), an inhibitor of protein synthesis, no significant difference between the two cultures was seen. Both cultures were able to recover when placed in low PFD but photoautotrophic cultures recovered in a lower extent and a slower rate. The maximal photosynthetic rate (P-max) and light saturation value (E-k) Of mixotrophic cultures were higher than those of the photoautotrophic cultures. Thus, mixotrophic cultures were less susceptible to high PFD stress. This can lead to a greater potential in using mixotrophic cultures for mass cultivation.

Abstract:The photosynthetic light reactions provide energy that is consumed and stored in electron sinks, the products of photosynthesis. A balance between light reactions and electron consumption in the chloroplast is vital for plants, and is protected by several photosynthetic regulation mechanisms. Photosystem I (PSI) is particularly susceptible to photoinhibition when these factors become unbalanced, which can occur in low temperatures or in high light. In this study we used the pgr5 Arabidopsis mutant that lacks ΔpH-dependent regulation of photosynthetic electron transport as a model to study the consequences of PSI photoinhibition under high light. We found that PSI damage severely inhibits carbon fixation and starch accumulation, and attenuates enzymatic oxylipin synthesis and chloroplast regulation of nuclear gene expression after high light stress. This work shows that modifications to regulation of photosynthetic light reactions, which may be designed to improve yield in crop plants, can negatively impact metabolism and signalling, and thereby threaten plant growth and stress tolerance. Abstract:To maintain CO2 fixation in the Calvin Benson-Bassham cycle, multi-step regulation of the chloroplast ATP synthase (CF1Fo) is crucial to balance the ATP output of photosynthesis with protection of the apparatus. A well-studied mechanism is thiol modulation; a light/dark regulation through reversible cleavage of a disulfide in the CF1Fo γ-subunit. The disulfide hampers ATP synthesis and hydrolysis reactions in dark-adapted CF1Fo from land plants by increasing the required transmembrane electrochemical proton gradient (ΔμH+). Here, we show in Chlamydomonas reinhardtii that algal CF1Fo is differently regulated in vivo. A specific hairpin structure in the γ-subunit redox domain disconnects activity regulation from disulfide formation in the dark. Electrochromic shift measurements suggested that the hairpin kept wild type CF1Fo active whereas the enzyme was switched off in algal mutant cells expressing a plant-like hairpin structure. The hairpin segment swap resulted in an elevated ΔμH+ threshold to activate plant-like CF1Fo, increased by ~1.4 photosystem (PS) I charge separations. The resulting dark-equilibrated ΔμH+ dropped in the mutants by ~2.7 PSI charge separation equivalents. Photobioreactor experiments showed no phenotypes in autotrophic aerated mutant cultures. In contrast, chlorophyll fluorescence measurements under heterotrophic dark conditions point to a reduced plastoquinone pool in cells with the plant-like CF1Fo as the result of bioenergetic bottlenecks. Our results suggest that the lifestyle of Chlamydomonas reinhardtii requires a specific CF1Fo dark regulation that partakes in metabolic coupling between the chloroplast and acetate-fueled mitochondria.

Abstract:Part of the chlL gene encoding a component involved in light-independent protochlorophyllide reduction was deleted in wild type and in a photosystem I-less strain of Synechocystis sp. PCC 6803. In resulting mutants, chlorophyll biosynthesis was fully light-dependent. When these mutants were propagated under light-activated heterotrophic growth conditions (in darkness except for 15 min of weak light a day) for several weeks, essentially no chlorophyll was detectable but protochlorophyllide accumulated. Upon return of the chlL- mutant cultures to continuous light, within the first 6 h chlorophyll was synthesized at the expense of protochlorophyllide at a rate independent of the presence of photosystem I. Chlorophyll biosynthesized during this time gave rise to a 685 nm fluorescence emission peak at 77 K in intact cells. This peak most likely originates from a component different from those known to be directly associated with photosystems II and I. Development of 695 and 725 nm peaks (indicative of intact photosystem II and photosystem I, respectively) required longer exposures to light. After 6 h of greening, the rate of chlorophyll synthesis slowed as protochlorophyllide was depleted. In the chlL- strain, greening occurred at the same rate at two different light intensities (5 and 50 μE m-2s-1), indicating that also at low light intensity the amount of light is not rate-limiting for protochlorophyllide reduction. Thus, in this system the rate of chlorophyll biosynthesis is limited neither by biosynthesis of photosystems nor by the light-dependent protochlorophyllide reduction. We suggest the presence of a chlorophyll-binding ‘chelator’ protein (with 77 K fluorescence emission at 685 nm) that binds newly synthesized chlorophyll and that provides chlorophyll for newly synthesized photosynthetic reaction centers and antennae.

Abstract:This review on chlorophyll a fluorescence starts with an overview of the primary photochemistry occurring at PSII and a characterization of the so-called “open” and “closed” states of its reaction centers. This provides the theoretical background for understanding the origin of PSII-emitted fluorescence and how its yield varies with the fraction of open reaction centers. The review proceeds to discuss the changes in fluorescence emission following illumination of a dark-adapted leaf and to define the PSII intrinsic quantum yield of photochemistry, which in turn provides an indication of PSII capacity. In light-adapted leaves, it is discussed how the use of modulated fluorometers and the double lighting technique allow an evaluation of photochemical and non-photochemical quenching, two parameters that give useful information about the plant’s photosynthetic performance under field conditions. Finally, it is described how the PSII operational efficiency can be used to calculate the photosynthetic electron transport rate and the conditions under which this is linearly related to the CO2 assimilation rate. Some requirements for a valid application of the technique as well as some limitations in interpreting its results are discussed.

Abstract:Photosynthesis involves the conversion of sunlight energy into stored chemical energy, which is achieved through electron transport along a series of redox reactions. Excess photosynthetic electron transport might be dangerous due to the risk of molecular oxygen reduction, generating reactive oxygen species (ROS) over-accumulation. Avoiding excess ROS production requires the rate of electron transport to be coordinated with the capacity of electron acceptors in the chloroplast stroma. Imbalance between the donor and acceptor sides of photosystem I (PSI) can lead to inactivation, which is called PSI photoinhibition. We used a light-inducible PSI photoinhibition system in Arabidopsis thaliana to resolve the time dynamics of inhibition and to investigate its impact on ROS production and turnover. The oxidation state of the PSI reaction center and rates of CO2 fixation both indicated strong and rapid PSI photoinhibition upon donor side/acceptor side imbalance, while the rate of inhibition eased during prolonged imbalance. PSI photoinhibition was not associated with any major changes in ROS accumulation or antioxidant activity; however, a lower level of lipid oxidation correlated with lower abundance of chloroplast lipoxygenase in PSI-inhibited leaves. The results of this study suggest that rapid activation of PSI photoinhibition under severe photosynthetic imbalance protects the chloroplast from over-reduction and excess ROS formation.

Chlorophyll Fluorescence as a Tool to Monitor Plant Response to the Environment

Abstract:Chlorophyll (Chi) fluorescence is an intriguing tool that can reveal information on plant performance and protective responses via rapid, non-intrusive measurements. Yet, if their limitations are not recognized, these measurements can be misleading and erroneous. A robust parameter obtained from Chi fluorescence in the field is the potential photochemical efficiency of (open) Photosystem II (PS II) units, either in darkness before sunrise (Fv/Fm, where Fv is variable Chi fluorescence, Fm – F0, and Fm and F0 are maximal and minimal fluorescence, respectively) or during exposure to light (F′v/F′m). Changes in this potential for photochemical utilization of light absorbed in PS II antennae can provide insight into changes in plants’ photosynthetic performance over the course of the day and between seasons. Decreases in Fv/Fm or F′v/F′m can be due to either increases in (the rate constant for) photoprotective energy dissipation or to decreases in (the rate constant for) photochemistry, and it is not possible to discern between these two possibilities without additional accompanying measures. Without a doubt, predawn depressions in PS II efficiency Fv/Fm suggest that the plant is experiencing stress, but the nature of the stress and the underlying mechanism responsible for the depression in PS II efficiency require additional investigation before any conclusions can be drawn. The determination of thermal energy dissipation activity from non-photochemical quenching (NPQ) is useful, but requires a control Fm value determined prior to sunrise that is fully relaxed. If the plant is under any stress at all, it is often impossible to obtain a fully relaxed Fm value. Photochemical quenching qp is sensitive to underestimation (overestimation of the degree to which PS II is reduced) at high levels of energy dissipation activity, and its utilization to estimate PS II electron transport rates should be viewed with caution.
The relationship between maximum tolerated light intensity and photoprotective energy dissipation in the photosynthetic antenna: chloroplast gains and losses

Abstract:The principle of quantifying the efficiency of protection of photosystem II (PSII) reaction centres against photoinhibition by non-photochemical energy dissipation (NPQ) has been recently introduced by Ruban & Murchie (2012 Biochim. Biophys. Acta 1817 , 977–982 ( doi:10.1016/j.bbabio.2012.03.026 )). This is based upon the assessment of two key parameters: (i) the relationship between the PSII yield and NPQ, and (ii) the fraction of intact PSII reaction centres in the dark after illumination. In this paper, we have quantified the relationship between the amplitude of NPQ and the light intensity at which all PSII reaction centres remain intact for plants with different levels of PsbS protein, known to play a key role in the process. It was found that the same, nearly linear, relationship exists between the levels of the protective NPQ component (pNPQ) and the tolerated light intensity in all types of studied plants. This approach allowed for the quantification of the maximum tolerated light intensity, the light intensity at which all plant leaves become photoinhibited, the fraction of (most likely) unnecessary or ‘wasteful’ NPQ, and the fraction of photoinhibited PSII reaction centres under conditions of prolonged illumination by full sunlight. It was concluded that the governing factors in the photoprotection of PSII are the level and rate of protective pNPQ formation, which are often in discord with the amplitude of the conventional measure of photoprotection, the quickly reversible NPQ component, qE. Hence, we recommend pNPQ as a more informative and less ambiguous parameter than qE, as it reflects the effectiveness and limitations of the major photoprotective process of the photosynthetic membrane.
Photoinhibition of Photosystem I Induced by Different Intensities of Fluctuating Light Is Determined by the Kinetics of ∆pH Formation Rather Than Linear Electron Flow

Abstract:Fluctuating light (FL) can cause the selective photoinhibition of photosystem I (PSI) in angiosperms. In nature, leaves usually experience FL conditions with the same low light and different high light intensities, but the effects of different FL conditions on PSI redox state and PSI photoinhibition are not well known. In this study, we found that PSI was highly reduced within the first 10 s after transition from 59 to 1809 μmol photons m−2 s−1 in tomato (Solanum lycopersicum). However, such transient PSI over-reduction was not observed by transitioning from 59 to 501 or 923 μmol photons m−2 s−1. Consequently, FL (59-1809) induced a significantly stronger PSI photoinhibition than FL (59-501) and FL (59-923). Compared with the proton gradient (∆pH) level after transition to high light for 60 s, tomato leaves almost formed a sufficient ∆pH after light transition for 10 s in FL (59-501) but did not in FL (59-923) or FL (59-1809). The difference in ∆pH between 10 s and 60 s was tightly correlated to the extent of PSI over-reduction and PSI photoinhibition induced by FL. Furthermore, the difference in PSI photoinhibition between (59-923) and FL (59-1809) was accompanied by the same level of linear electron flow. Therefore, PSI photoinhibition induced by different intensities of FL is more related to the kinetics of ∆pH formation rather than linear electron flow.

Effects of water stress on photosynthesis, chlorophyll fluorescence and photoinhibition in wheat plants

Abstract:Effects of water stress on photosynthesis, PS II photochemistry and photoinhibition were investigated in wheat plants (Tritium aestivum L.). To separate water stress effects from photoinhibition, water stress was imposed at low irradiance (180 μmol m-2 s-1). When water stress developed gradually, net CO2 assimilation rate and leaf stomatal conductance decreased significantly. However, water stress had no effects on the PS II photochemistry in dark-adapted leaves. There were no significant changes in the maximal efficiency of PS II photochemistry and no apparent damages in PS II reaction centre, its oxidising and acceptor sides, or its antennae system. However, PS II photochemistry in light-adapted leaves was modified in water-stressed plants. This was shown by the decrease in the efficiency of excitation energy capture by open PS II reaction centres and the quantum yield of PS II electron transport and a significant increase in non-photochemical quenching. In addition, water stress increased the susceptibility to photoinhibition. The extent of photoinhibition became more pronounced as water stress increased. It was found that water-stressed plants exhibited a much greater accumulation of the QB-non-reducing PS II reaction centres and a smaller increase in non- photochemical quenching during photoinhibition. Such changes might be responsible for the increased susceptibility to photoinhibition.

Response of Strawberry Leaves Photosynthesis to Strong Light and Its Mechanism

Abstract:In this paper, PAM-2000 portable chlorophyll fluorometer and HCM-1000 photosynthesis measurement system were applied to measure the apparent quantum efficiency (AQY), initial fluorescence (Fo), maximal photochemical efficiency of PSII (Fv/Fm), maximal fluorescence (Fm), photochemical quenching (qP), non-photochemical quenching (qN), actual quantum yield of PS II electron transport (phiPSII), electron transport rate (ETR), amount of inactive PS II reaction centers (Fi-Fo), Q(A) reduction rate, proportion of Q(B)-non-reducing PS II reaction centers [(Fi-Fo)/(Fp-Fo)], and energy-dependent quenching (qE), photoinhibitory quenching (qI) and state-transition quenching (qT) of non-photochemical quenching of strawberry leaves. The results showed that the Fv/Fm, Fm, Fi-Fo and Q(A) reduction rate decreased in strong light and increased during subsequent dark recovery, but (Fi-Fo)/(Fp-Fo) was in reverse. They changed drastically within the first 10 minutes in strong light or in subsequent darkness. In strong light, phiPSII, ETR and qP increased firstly and then decreased, while qN decreased drastically firstly and then increased slightly. After exposure to strong light for 4 hours, the AQY of two varieties "Toyonoka" and "Houkouwase" decreased by 20.9% and 37.5%, respectively, and qE was 89.1% and 87.1%, respectively in NPQ (qE + qI + qT). In strong light, "Toyonoka" showed less changes than "Houkouwase" in Fo, Fv/Fm, Fm, ETR, phiPSII and AQY. After treated with DTT, the Fv/Fm and Fm were lower, but Fo was much higher than control. It is deduced that in strong light, xanthophyll cycle-dependent non-radiative energy dissipation and pH-dependent heat dissipation could play an important protective role against photo-damage to the photosynthetic apparatus in strawberry leaves.
Role of Cytochrome B559 in Photoinhibition

Abstract:The aim of this research project was to obtain information on the role of the cytochrome b559 in the function of Photosystem-II (PSII) with special emphasis on the light induced photo inactivation of PSII and turnover of the photochemical reaction center II protein subunit RCII-D1. The major goals of this project were: 1) Isolation and sequencing of the Chlamydomonas chloroplast psbE and psbF genes encoding the cytochrome b559 a and b subunits respectively; 2) Generation of site directed mutants and testing the effect of such mutation on the function of PSII under various light conditions; 3) To obtain further information on the mechanism of the light induced degradation and replacement of the PSII core proteins. This information shall serve as a basis for the understanding of the role of the cytochrome b559 in the process of photoinhibition and recovery of photosynthetic activity as well as during low light induced turnover of the D1 protein. Unlike in other organisms in which the psbE and psbF genes encoding the a and b subunits of cytochrome b559, are part of an operon which also includes the psbL and psbJ genes, in Chlamydomonas these genes are transcribed from different regions of the chloroplast chromosome. The charge distribution of the derived amino-acid sequences of psbE and psbF gene products differs from that of the corresponding genes in other organisms as far as the rule of "positive charge in" is concerned relative to the process of the polypeptide insertion in the thylakoid membrane. However, the sum of the charges of both subunits corresponds to the above rule possibly indicating co-insertion of both subunits in the process of cytochrome b559 assembly. A plasmid designed for the introduction of site-specific mutations into the psbF gene of C. reinhardtii. was constructed. The vector consists of a DNA fragment from the chromosome of C. reinhardtii which spans the region of the psbF gene, upstream of which the spectinomycin-resistance-conferring aadA cassette was inserted. This vector was successfully used to transform wild type C. reinhardtii cells. The spectinomycin resistant strain thus obtained can grow autotrophically and does not show significant changes as compared to the wild-type strain in PSII activity. The following mutations have been introduced in the psbF gene: H23M; H23Y; W19L and W19. The replacement of H23 involved in the heme binding to M and Y was meant to permit heme binding but eventually alter some or all of the electron transport properties of the mutated cytochrome. Tryptophane W19, a strictly conserved residue, is proximal to the heme and may interact with the tetrapyrole ring. Therefore its replacement may effect the heme properties. A change to tyrosine may have a lesser affect on the potential or electron transfer rate while a replacement of W19 by leucine is meant to introduce a more prominent disturbance in these parameters. Two of the mutants, FW19L and FH23M have segregated already and are homoplasmic. The rest are still grown under selection conditions until complete segregation will be obtained. All mutants contain assembled and functional PSII exhibiting an increased sensitivity of PSII to the light. Work is still in progress for the detailed characterization of the mutants PSII properties. A tobacco mutant, S6, obtained by Maliga and coworkers harboring the F26S mutation in the b subunit was made available to us and was characterized. Measurements of PSII charge separation and recombination, polypeptide content and electron flow indicates that this mutation indeed results in light sensitivity. Presently further work is in progress in the detailed characterization of the properties of all the above mutants. Information was obtained demonstrating that photoinactivation of PSII in vivo initiates a series of progressive changes in the properties of RCII which result in an irreversible modification of the RCII-D1 protein leading to its degradation and replacement. The cleavage process of the modified RCII-D1 protein is regulated by the occupancy of the QB site of RCII by plastoquinone. Newly synthesized D1 protein is not accumulated in a stable form unless integrated in reassembled RCII. Thus the degradation of the irreversibly modified RCII-D1 protein is essential for the recovery process. The light induced degradation of the RCII-D1 protein is rapid in mutants lacking the pD1 processing protease such as in the LF-1 mutant of the unicellular alga Scenedesmus obliquus. In this case the Mn binding site of PSII is abolished, the water oxidation process is inhibited and harmful cation radicals are formed following light induced electron flow in PSII. In such mutants photo-inactivation of PSII is rapid, it is not protected by ligands binding at the QB site and the degradation of the inactivated RCII-D1 occurs rapidly also in the dark. Furthermore the degraded D1 protein can be replaced in the dark in absence of light driven redox controlled reactions. The replacement of the RCII-D1 protein involves the de novo synthesis of the precursor protein, pD1, and its processing at the C-terminus end by an unknown processing protease. In the frame of this work, a gene previously isolated and sequenced by Dr. Pakrasi's group has been identified as encoding the RCII-pD1 C-terminus processing protease in the cyanobacterium Synechocystis sp. PCC 6803. The deduced sequence of the ctpA protein shows significant similarity to the bovine, human and insect interphotoreceptor retinoid-binding proteins. Results obtained using C. reinhardtii cells exposes to low light or series of single turnover light flashes have been also obtained indicating that the process of RCII-D1 protein turnover under non-photoinactivating conditions (low light) may be related to charge recombination in RCII due to back electron flow from the semiquinone QB- to the oxidised S2,3 states of the Mn cluster involved in the water oxidation process.
Response of chlorophyll fluorescence to dynamic light in three alpine species differing in plant architecture

Abstract:A study was carried out to examine the effect of dynamic photosynthetically active photon flux density (PPFD) on photoinhibition and energy use in three herbaceous species, prostrate Saussurea superba, erect-leaved S. katochaete, and half-erect-leaved Gentiana straminea, from the Qinghai—Tibet Plateau. Chlorophyll fluorescence response was measured under each of three sets of high–low PPFD combinations: 1700–0, 1400–300, and 1200–500μmolm−2s−1, illuminating in four dynamic frequencies: 1, 5, 15, and 60 cycles per 2h. The total light exposure time was 2h and the integrated PPFD was the same in all treatments. The highest frequency of PPFD fluctuation resulted in the lowest photochemical activity, the highest level of non-photochemical quenching, and the greatest decrease of Fv/Fm (maximal photochemical efficiency of PSII). The 5 and 15 cycles per 2h treatments resulted in higher photochemical activity than the 1 cycle per 2h treatment. The 1700–0 PPFD combination led to the lowest photochemical activity and more serious photoinhibition in all species. S. superba usually exhibited the highest photochemical activity and CO2 uptake rate, the lowest reduction of Fv/Fm, and the smallest fraction of energy in thermal dissipation. With similar fractions of thermal dissipation, S. katochaete had relatively less photoinhibition than G. straminea owing to effective F0 quenching. The results suggest that high frequency of fluctuating PPFD generally results in photoinhibition, which is more serious under periods of irradiation with high light intensity.
Determining photosynthetic control, a probe for the balance between electron transport and Calvin–Benson cycle activity, with the DUAL-KLAS-NIR

Abstract:Photosynthetic Control is defined as the control imposed on photosynthetic electron transport by the lumen-pH-sensitive re-oxidation of plastoquinol (PQH 2 ) by cytochrome b 6 f. Photosynthetic Control leads at higher actinic light intensities to an electron transport chain with a (relatively) reduced photosystem (PS) II and PQ pool and a (relatively) oxidized PS I. Making Light Curves of more than 33 plant species with the recently introduced DUAL-KLAS-NIR (Chl a fluorescence + the redox states of plastocyanin (PC), P700, and ferredoxin (Fd)) the light intensity-dependent induction of Photosynthetic Control was probed and characterized. It was observed that PC became completely oxidized at light intensities ≤ 400 μmol photons m −2 s −1 (at lower light intensities in shade than in sun leaves). The relationship between qP and P700(red) was used to determine the extent of Photosynthetic Control. Instead of measuring the whole Light Curve, it was shown that a single moderate light intensity can be used to characterize the status of a leaf relative to that of other leaves. It was further found that in some shade-acclimated leaves Fd becomes again more oxidized at high light intensities indicating that electron transfer from the PQ pool to P700 cannot keep up with the outflow of electrons on the acceptor side of PS I. It was observed as well that for NPQ-induction a lower light intensity (less acidified lumen) was needed than for the induction of Photosynthetic Control. The measurements were also used to make a comparison between the parameters qP and qL, a comparison suggesting that qP was the more relevant parameter.
