Abstract:To determine the auditory event-related potential (AERP) correlates of phoneme perception in 17-month-old children at risk of dyslexia and controls, AERPs were elicited by standard (/bAk/) and deviant (/dAk/) word stimuli presented in an oddball paradigm. The at-risk children tended to show delayed P1 and P2 peaks for standard stimuli. Hemispheric group differences were observed for N2 amplitude and P1 latency, suggesting atypical processing of spoken words in at-risk children. Larger P2 and to a lesser extent P1 amplitudes following deviant words occurred in the control but not in the at-risk group, this may signal poorer phoneme discrimination in at-risk children. The present AERP results offer clues to where to look for a marker to identify children at high risk of reading and writing problems at an early age to facilitate early intervention.

Abstract:Identification of the hepatitis C virus (HCV) JFH1 isolate enabled the development of infectious HCV cell culture systems. However, the relatively low virus titres and instability of some chimeric JFH1 reporter viruses restricts some uses of this system. We describe a higher-titre JFH1-EGFP reporter virus where the NS5A V3 region was replaced with the EGFP gene and adapted by serial passage in Huh7.5 cells. Six adaptive mutants were identified: one each in E2, P7 and NS4B, plus three in the NS5A region. These adaptive mutants increased the reporter virus titres to 1×10(6) immunofluorescent focus-forming units ml(-1), which is the highest titre of JFH1-EGFP reporter virus reported to our knowledge. This chimeric virus did not lose EGFP expression following 40 days of passage and it can be used to test the activity of HCV antivirals by measuring EGFP fluorescence in 96-well plates. Moreover, this reporter virus allows living infected Huh7.5 cells in Matrigel three-dimensional (3D) cultures to be visualized and produces infectious viral particles in these 3D cultures. The chimeric NS5A-EGFP infectious JFH1 reporter virus described should enable new studies of the HCV life cycle in 3D cell cultures and will be useful in identifying antivirals that interfere with HCV release or entry.
Neural dysfunction in ADHD with Reading Disability during a word rhyming Continuous Performance Task

Abstract:Background: Attention-Deficit/Hyperactivity Disorder (ADHD) is a heterogeneous, neurodevelopmental disorder which co-occurs often with Reading Disability (RD). ADHD with and without RD consistently have higher inattentive ratings compared with typically developing controls, with co-occurring ADHD and RD also demonstrating impaired phonological processing. Accordingly, inattention has been associated with greater phonological impairment, though the neural correlates of the association are poorly understood from a functional neuroimaging perspective. It was postulated that only the co-occurring subgroup would demonstrate hypoactivation of posterior, left hemispheric, reading-related areas and, to a lesser extent, alterations in right hemispheric, attention areas compared with controls. Methods: A novel word rhyming Continuous Performance Task assesses functional activation differences in phonology- and attention-related areas between three groups: ten boys with ADHD and RD, fourteen boys with ADHD without RD, and fourteen typically developing controls. Subjects respond to words that rhyme with a target word as mono- and disyllabic, English words are visually presented over 90s blocks. Results: Behavioral performance was not different between groups. Some hypoactivation of left hemispheric, reading-related areas was apparent in ADHD and RD, but not ADHD without RD, compared with controls. Right hemispheric, attention areas showed alterations in both ADHD subgroups relative to controls; however, the differences for each subgroup were dissimilar. Conclusions: The dorsal decoding subnetwork may not be grossly compromised in ADHD with Reading Disability. The role of cognitive impairments, including the level of inattention, on phonology requires clarification from a neuroimaging perspective.
Bioelectrochemical changes during the early stages of chalcopyrite interaction with Acidithiobacillus Thiooxidans and Leptospirillum sp.

Abstract:A bioelectrochemical study of charge transfer in the biofilm–chalcopyrite interface was performed to investigate the effect of surficial reduced sulfur species (RSS), in the form of non-stochiometric compounds or polysulfides (Sn2−) and elemental sulfur (S0) on a biofilm structure, during the earliest stages (1, 12 and 24 h) of chalcopyrite biooxidation by Acidithiobacillus thiooxidans alone and adding Leptospirillum sp. The surface of massive chalcopyrite electrodes was exposed to the bacteria, which were analyzed electrochemically, spectroscopically, and microscopically. At the studied earlier times, charge transfer and significant differences in the biofilm structure were detected, depending on the presence of Leptospirillum sp. acting on A. thiooxidans biofilms. Such differences were a consequence of a continuous chalcopyrite pitting and promoting changes in biofilm hydrophobicity. A. thiooxidans modifies the reactive properties of RSS and favors an acidic dissolution, which shifts into ferric dissolution when Leptospirillum sp. is present. A. thiooxidans allows H+ and Fe3+ diffusion, and Leptospirillum sp. enables to surpass the charge transfer (reactivity) barrier between the mineral interface and the ions. The observed changes of hydrophobicity on the interface are associated to ions and electrons activity and transfer. Finally, a model of S0 biooxidation by A. thiooxidans alone or with Leptospirillum sp. is proposed.
Orthographic and phonological processing in Chinese dyslexic children: an ERP study on sentence reading.

Abstract:An event-related potential (ERP) experiment was conducted to explore the differences between Chinese-speaking dyslexic children and normal school children in orthographic and phonological processing during Chinese sentence reading. Participants were visually presented with sentences, word-by-word and were asked to judge whether the sentences were semantically acceptable. The crucial manipulation was on the sentence-final two-character compound words, which were either correct or incorrect. For the incorrect compounds, the second characters of the base words were replaced by homophonic or orthographically similar characters. It was found that, for the normal controls, the orthographic and phonological mismatches elicited more negative ERP responses, relative to the baseline, over a relatively long time course (including the time windows for P200 and N400) at the central–posterior scalp regions. In contrast, the dyslexic children in general showed no differences between experimental conditions for P200 and N400, although the more detailed time course analyses did reveal some weak effects for the N400 component between experimental conditions. In addition, the mean amplitude of N400 in the homophonic condition was less negative-going for the dyslexics than for the controls. These findings suggest that Chinese dyslexic children have deficits in processing orthographic and phonological information conveyed by characters and, compared with normal children, they rely more on phonological information to access lexical semantics in sentence reading.
Neural Signature of Rhyming Ability During Story Listening in Preschool-Age Children

Abstract:Purpose: Rhyming is a phonological skill that typically emerges in the preschool-age range. Prosody/rhythm processing involves right-lateralized temporal cortex, yet the neural basis of rhyming ability in young children is unclear. The study objective was to use functional magnetic resonance imaging (fMRI) to quantify neural correlates of rhyming abilities in preschool-age children. Method: Healthy pre-kindergarten child-parent dyads were recruited for a study visit including MRI and the Preschool and Primary Inventory of Phonological Awareness (PIPA) rhyme subtest. MRI included an fMRI task where the child listened to a rhymed and unrhymed story without visual stimuli. fMRI data were processed using the CONN functional connectivity (FC) toolbox, with FC computed between 132 regions of interest (ROI) across the brain. Associations between PIPA score and FC during the rhymed versus unrhymed story were compared accounting for age, sex, and maternal education. Results: In total, 45 children completed MRI (age 54 ± 8 months, 37-63; 19M 26F). Median maternal education was college graduate. FC between ROIs in posterior default mode (imagery) and right fronto-parietal (executive function) networks was more strongly positively associated with PIPA score during the rhymed compared with the unrhymed story [F(2,39) = 10.95, p-FDR = 0.043], as was FC between ROIs in right-sided language (prosody) and dorsal attention networks [F(2,39) = 9.85, p-FDR = 0.044]. Conclusions: Preschool-age children with better rhyming abilities had stronger FC between ROIs supporting attention and prosody and also between ROIs supporting executive function and imagery, suggesting rhyme as a catalyst for attention, visualization, and comprehension. These represent novel neural biomarkers of nascent phonological skills.

Abstract:The acquisition of letter-speech sound correspondences is a fundamental process underlying reading development, one that could be influenced by several linguistic and domain-general cognitive factors. In the current study, we mimicked the first steps of this process by examining behavioral trajectories of audiovisual associative learning in 110 7- to 12-year-old children with and without dyslexia. Children were asked to learn the associations between eight novel symbols and native speech sounds in a brief training and subsequently read words and pseudowords written in the artificial orthography. We then investigated the influence of auditory attention as one of the putative domain-general factors influencing associative learning. To this aim, we assessed children with experimental measures of auditory sustained selective attention and interference control. Our results showed shallower learning trajectories in children with dyslexia, especially during the later phases of the training blocks. Despite this, children with dyslexia performed similarly to typical readers on the post-training reading tests using the artificial orthography. Better auditory sustained selective attention and interference control skills predicted greater response accuracy during training. Sustained selective attention was also associated with the ability to apply these novel correspondences in the reading tests. Although this result has the limitations of a correlational design, it denotes that poor attentional skills may constitute a risk during the early stages of reading acquisition, when children start to learn letter-speech sound associations. Importantly, our findings underscore the importance of examining dynamics of learning in reading acquisition as well as individual differences in more domain-general attentional factors.

Abstract:Purpose: We examined the neurocognitive bases of lexical morphology in children of varied reading abilities to understand the role of meaning-based skills in learning to read with dyslexia. Method: Children completed auditory morphological and phonological awareness tasks during functional near-infrared spectroscopy neuroimaging. We first examined the relation between lexical morphology and phonological processes in typically developing readers (Study 1, N = 66, M age = 8.39), followed by a more focal inquiry into lexical morphology processes in dyslexia (Study 2, N = 50, M age = 8.62). Results: Typical readers exhibited stronger engagement of language neurocircuitry during the morphology task relative to the phonology task, suggesting that morphological analyses involve synthesizing multiple components of sublexical processing. This effect was stronger for more analytically complex derivational affixes ( like + ly ) than more semantically transparent free base morphemes ( snow + man ). In contrast, children with dyslexia exhibited stronger activation during the free base condition relative to derivational affix condition. Taken together, the findings suggest that although children with dyslexia may struggle with derivational morphology, they may also use free base morphemes' semantic information to boost word recognition. Conclusion: This study informs literacy theories by identifying an interaction between reading ability, word structure, and how the developing brain learns to recognize words in speech and print. Attentional modulation of neural sound tracking in children with and without dyslexia

Abstract:Auditory selective attention forms an important foundation of children's learning by enabling the prioritisation and encoding of relevant stimuli. It may also influence reading development, which relies on metalinguistic skills including the awareness of the sound structure of spoken language. Reports of attentional impairments and speech perception difficulties in noisy environments in dyslexic readers are also suggestive of the putative contribution of auditory attention to reading development. To date, it is unclear whether non-speech selective attention and its underlying neural mechanisms are impaired in children with dyslexia and to which extent these deficits relate to individual reading and speech perception abilities in suboptimal listening conditions. In this EEG study, we assessed non-speech sustained auditory selective attention in 106 7-to-12-year-old children with and without dyslexia. Children attended to one of two tone streams, detecting occasional sequence repeats in the attended stream, and performed a speech-in-speech perception task. Results show that when children directed their attention to one stream, inter-trial-phase-coherence at the attended rate increased in fronto-central sites; this, in turn, was associated with better target detection. Behavioural and neural indices of attention did not systematically differ as a function of dyslexia diagnosis. However, behavioural indices of attention did explain individual differences in reading fluency and speech-in-speech perception abilities: both these skills were impaired in dyslexic readers. Taken together, our results show that children with dyslexia do not show group-level auditory attention deficits but these deficits may represent a risk for developing reading impairments and problems with speech perception in complex acoustic environments. Abstract:OBJECTIVE: Previous studies have reported atypical delta phase in children with dyslexia, and that delta phase modulates the amplitude of the beta-band response via delta-beta phase-amplitude coupling (PAC). Accordingly, the atypical delta-band effects in children with dyslexia may imply related atypical beta-band effects, particularly regarding delta-beta PAC. Our primary objective was to explore beta-band oscillations in children with and without dyslexia, to explore potentially atypical effects in the beta band in dyslexic children.METHODS: We collected EEG data during a rhythmic speech paradigm from 51 children (21 control; 30 dyslexia). We then assessed beta-band phase entrainment, beta-band angular velocity, beta-band power responses and delta-beta PAC.RESULTS: We found significant beta-band phase entrainment for control children but not for dyslexic children. Furthermore, children with dyslexia exhibited significantly faster beta-band angular velocity and significantly greater beta-band power. Delta-beta PAC was comparable in both groups.CONCLUSION: Atypical beta-band effects were observed in children with dyslexia. However, delta-beta PAC was comparable in both dyslexic and control children.SIGNIFICANCE: These findings offer further insights into the neurophysiological basis of atypical rhythmic speech processing by children with dyslexia, suggesting the involvement of a wide range of frequency bands.

Urinothorax: A Rare Complication of Total Abdominal Hysterectomy

Abstract:BACKGROUND: Urinothorax is defined as the presence of urine in the pleural cavity. This condition is due to the leakage of urine from the peritoneal and retroperitoneal space into the pleural space. We report a case of urinothorax after total abdominal hysterectomy. CASE: A premenopausal woman underwent total abdominal hysterectomy and left salpingo-oophorectomy for pelvic pain, requiring extensive dissection. Her postoperative course was complicated by right urinothorax, which was diagnosed after a diagnostic thoracocentesis and resolved after a therapeutic thoracocentesis and the repair of the urinary bladder rent. CONCLUSION: Urinothorax can occur in cases of undiagnosed urinary tract injury. It presents as postoperative shortness of breath and pleural effusion and should be considered in the differential diagnosis.
The role of word knowledge in error detection: a challenge to the broken error monitor account of dyslexia

Abstract:Dyslexic children often fail to correct errors while reading aloud, and dyslexic adolescents and adults exhibit lower amplitudes of the error-related negativity (ERN)-the neural response to errors-than typical readers during silent reading. Past researchers therefore suggested that dyslexia may arise from a faulty error detection mechanism that interferes with orthographic learning and text comprehension. An alternative possibility is that comprehension difficulty in dyslexics is primarily a downstream effect of low-quality lexical representations-that is, poor word knowledge. On this view, the attenuated ERN in dyslexics is a byproduct, rather than a source, of underdeveloped orthographic knowledge. Because the second view implies a direct association of the error response with comprehension skill in populations of all ability levels, the present study evaluates these alternatives through a reanalysis of behavioral and neural data from 31 typical adult readers. If it is true that faulty error processing can manifest as dyslexia, a model in which error monitoring contributes directly to comprehension should outperform a model in which it does not. ERNs recorded during spelling judgments were used as a measure of error detection aptitude in path analyses of reading comprehension. The data were better fit by a model in which error detection aptitude was a consequence of word knowledge than a model in which it contributed directly to comprehension. The findings challenge the notion that comprehension difficulty in dyslexics is attributable to error processing deficits and are consistent with the hypothesis that comprehension difficulty in dyslexics is partially attributable to low-quality word knowledge.

Abstract:Auditory phonological processing skills are critical for successful reading development in English not only in native (L1) speakers but also in second language (L2) learners. However, the neural deficits of auditory phonological processing remain unknown in English-as-the-second-language (ESL) learners with reading difficulties. Here we investigated neural responses during spoken word rhyme judgments in typical and impaired ESL readers in China. The impaired readers showed comparable activation in the left superior temporal gyrus (LSTG), but reduced activation in the left inferior frontal gyrus (LIFG) and left fusiform and reduced connectivity between the LSTG and left fusiform when compared to typical readers. These findings suggest that impaired ESL readers have relative intact representations but impaired manipulation of phonology and reduced or absent automatic access to orthographic representations. This is consistent with previous findings in native English speakers and suggests a common neural mechanism underlying English impairment across the L1 and L2 learners.
Auditory temporal structure processing in dyslexia: processing of prosodic phrase boundaries is not impaired in children with dyslexia

Abstract:Reading disability in children with dyslexia has been proposed to reflect impairment in auditory timing perception. We investigated one aspect of timing perception—temporal grouping—as present in prosodic phrase boundaries of natural speech, in age-matched groups of children, ages 6–8 years, with and without dyslexia. Prosodic phrase boundaries are characterized by temporal grouping of functionally related speech elements and can facilitate syntactic processing of speech. For example, temporary syntactic ambiguities, such as early-closure structures, are processed faster when prosodic phrase boundaries are present. We examined children’s prosodic facilitation by measuring their efficiency of sentence processing for temporary syntactic ambiguities spoken with (facilitating) versus without (neutral) prosodic phrase boundaries. Both groups of children benefited similarly from prosodic facilitation, displaying faster reaction times in facilitating compared to neutral prosody. These findings indicate that the use of prosodic phrase boundaries for speech processing is not impaired in children with dyslexia.

Abstract:Developmental dyslexia is typically associated with difficulties in basic auditory processing and in manipulating speech sounds. However, the neuroanatomical correlates of auditory difficulties in developmental dyslexia (DD) and their contribution to individual clinical phenotypes are still unknown. Recent intracranial electrocorticography findings associated processing of sound amplitude rises and speech sounds with posterior and middle superior temporal gyrus (STG), respectively. We hypothesize that regional STG anatomy will relate to specific auditory abilities in DD, and that auditory processing abilities will relate to behavioral difficulties with speech and reading. One hundred and ten children (78 DD, 32 typically developing, age 7-15 years) completed amplitude rise time and speech in noise discrimination tasks. They also underwent a battery of cognitive tests. Anatomical MRI scans were used to identify regions in which local cortical gyrification complexity correlated with auditory behavior. Behaviorally, amplitude rise time but not speech in noise performance was impaired in DD. Neurally, amplitude rise time and speech in noise performance correlated with gyrification in posterior and middle STG, respectively. Furthermore, amplitude rise time significantly contributed to reading impairments in DD, while speech in noise only explained variance in phonological awareness. Finally, amplitude rise time and speech in noise performance were not correlated, and each task was correlated with distinct neuropsychological measures, emphasizing their unique contributions to DD. Overall, we provide a direct link between the neurodevelopment of the left STG and individual variability in auditory processing abilities in neurotypical and dyslexic populations.

Abstract:Objective We explored neural components in Electroencephalography (EEG) signals during a phonological processing task to assess (a) the neural origins of Baddeley's working-memory components contributing to phonological processing, (b) the unitary structure of phonological processing and (c) the neural differences between children with dyslexia (DYS) and controls (CAC). Methods EEG data were collected from sixty children (half with dyslexia) while performing the initial- and final- phoneme elision task. We explored a novel machine-learning-based approach to identify the neural components in EEG elicited in response to the two conditions and capture differences between DYS and CAC. Results Our method identifies two sets of phoneme-related neural congruency components capturing neural activations distinguishing DYS and CAC across conditions. Conclusions Neural congruency components capture the underlying neural mechanisms that drive the relationship between phonological deficits and dyslexia and provide insights into the phonological loop and visual-sketchpad dimensions in Baddeley's model at the neural level. They also confirm the unitary structure of phonological awareness with EEG data. Significance Our findings provide novel insights into the neural origins of the phonological processing differences in children with dyslexia, the unitary structure of phonological awareness, and further verify Baddeley's model as a theoretical framework for phonological processing and dyslexia.

Abstract:During information processing, individuals benefit from bimodally presented input, as has been demonstrated for speech perception (i.e., printed letters and speech sounds) or the perception of emotional expressions (i.e., facial expression and voice tuning). While typically developing individuals show this bimodal benefit, school children with dyslexia do not. Currently, it is unknown whether the bimodal processing deficit in dyslexia also occurs for visual-auditory speech processing that is independent of reading and spelling acquisition (i.e., no letter-sound knowledge is required). Here, we tested school children with and without spelling problems on their bimodal perception of video-recorded mouth movements pronouncing syllables. We analyzed the event-related potential Mismatch Response (MMR) to visual-auditory speech information and compared this response to the MMR to monomodal speech information (i.e., auditory-only, visual-only). We found a reduced MMR with later onset to visual-auditory speech information in children with spelling problems compared to children without spelling problems. Moreover, when comparing bimodal and monomodal speech perception, we found that children without spelling problems showed significantly larger responses in the visual-auditory experiment compared to the visual-only response, whereas children with spelling problems did not. Our results suggest that children with dyslexia exhibit general difficulties in bimodal speech perception independently of letter-speech sound knowledge, as apparent in altered bimodal speech perception and lacking benefit from bimodal information. This general deficit in children with dyslexia may underlie the previously reported reduced bimodal benefit for letter-speech sound combinations and similar findings in emotion perception.

Abstract:Dyslexia in alphabetic languages has been extensively investigated and suggests a central deficit in orthography to phonology mapping in the left hemisphere. Compared to dyslexia in alphabetic languages, the central deficit for Chinese dyslexia is still unclear. Because of the logographic nature of Chinese characters, some have suggested that Chinese dyslexia should have larger deficits in the semantic system. To investigate this, Chinese children with reading disability (RD) were compared to typically developing (TD) children using functional magnetic resonance imaging (fMRI) on a rhyming judgment task and on a semantic association judgment task. RD children showed less activation for both tasks in right visual (BA18, 19) and left occipito-temporal cortex (BA 37), suggesting a deficit in visuo-orthographic processing. RD children also showed less activation for both tasks in left inferior frontal gyrus (BA44), which additionally showed significant correlations with activation of bilateral visuo-orthographic regions in the RD group, suggesting that the abnormalities in frontal cortex and in posterior visuo-orthographic regions may reflect a deficit in the connection between brain regions. Analyses failed to reveal larger differences between groups for the semantic compared to the rhyming task, suggesting that Chinese dyslexia is similarly impaired in the access to phonology and to semantics from the visual orthography.

Abstract:The role of phonology in word recognition has previously been investigated using a masked lexical decision task and transposed letter (TL) nonwords that were either pronounceable (barve) or unpronounceable (brvae). We used event-related potentials (ERPs) to investigate these effects in skilled deaf readers, who may be more sensitive to orthotactic than phonotactic constraints, which are conflated in English. Twenty deaf and twenty hearing adults completed a masked lexical decision task while ERPs were recorded. The groups were matched in reading skill and IQ, but deaf readers had poorer phonological ability. Deaf readers were faster and more accurate at rejecting TL nonwords than hearing readers. Neither group exhibited an effect of nonword pronounceability in RTs or accuracy. For both groups, the N250 and N400 components were modulated by lexicality (more negative for nonwords). The N250 was not modulated by nonword pronounceability, but pronounceable nonwords elicited a larger amplitude N400 than unpronounceable nonwords. Because pronounceable nonwords are more word-like, they may incite activation that is unresolved when no lexical entry is found, leading to a larger N400 amplitude. Similar N400 pronounceability effects for deaf and hearing readers, despite differences in phonological sensitivity, suggest these TL effects arise from sensitivity to lexical-level orthotactic constraints. Deaf readers may have an advantage in processing TL nonwords because of enhanced early visual attention and/or tight orthographic-to-semantic connections, bypassing the phonologically mediated route to word recognition.
