Abstract:In MRI, researchers have long endeavored to effectively visualize myelin distribution in the brain, a pursuit with significant implications for both scientific research and clinical applications. Over time, various methods such as myelin water imaging, magnetization transfer imaging, and relaxometric imaging have been developed, each carrying distinct advantages and limitations. Recently, an innovative technique named as magnetic susceptibility source separation has emerged, introducing a novel surrogate biomarker for myelin in the form of a diamagnetic susceptibility map. This paper comprehensively reviews this cutting-edge method, providing the fundamental concepts of magnetic susceptibility, susceptibility imaging, and the validation of the diamagnetic susceptibility map as a myelin biomarker that indirectly measure myelin content. Additionally, the paper explores essential aspects of data acquisition and processing, offering practical insights for readers. A comparison with established myelin imaging methods is also presented, and both current and prospective clinical and scientific applications are discussed to provide a holistic understanding of the technique. This work aims to serve as a foundational resource for newcomers entering this dynamic and rapidly expanding field.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to effectively visualize the myelin distribution in the brain in magnetic resonance imaging (MRI). Specifically, the paper mainly focuses on an emerging technology - Magnetic Susceptibility Source Separation (MSSS), which indirectly measures the myelin content by generating a new myelin biomarker - the paramagnetic susceptibility map. The paper aims to comprehensively review this cutting - edge method, providing the basic concepts of magnetic susceptibility, magnetic susceptibility imaging techniques, and the validation of the effectiveness of the paramagnetic susceptibility map as a myelin biomarker. In addition, the paper also explores the key aspects of data acquisition and processing, compares it with existing myelin imaging methods, and discusses the current and potential applications of this technology in clinical and scientific research. ### Main Objectives of the Paper: 1. **Introduce the Magnetic Susceptibility Source Separation Technology**: Explain the basic principles of magnetic susceptibility source separation and its application in myelin imaging. 2. **Verify the Effectiveness of the Paramagnetic Susceptibility Map as a Myelin Biomarker**: Prove through experimental data and image analysis that the paramagnetic susceptibility map can indirectly reflect the myelin content. 3. **Explore Data Acquisition and Processing Methods**: Provide practical operation guidelines to help researchers better apply this technology. 4. **Compare Different Myelin Imaging Methods**: Evaluate the advantages and limitations of the magnetic susceptibility source separation technology compared to other existing methods. 5. **Discuss Clinical and Scientific Applications**: Explore the potential value of this technology in neuroscience research and clinical diagnosis. ### Background: Myelin is a lipid bilayer membrane structure in the central nervous system. It wraps around axons to form a protective sheath, which is crucial for rapid and accurate nerve conduction. In MRI, the properties of myelin make it a key determinant of multiple MRI contrasts, such as T1 - weighted images, T2 - weighted images, and magnetic susceptibility imaging. Therefore, developing effective myelin imaging methods has always been an important goal in MRI research. ### Existing Methods: - **Myelin Water Imaging (MWI)** - **Magnetization Transfer Imaging (MTI)** - **Diffusion Imaging** - **Quantitative Susceptibility Mapping (QSM)** ### Emerging Technology: - **Magnetic Susceptibility Source Separation (MSSS)**: Utilize the paramagnetic properties of myelin to generate high - resolution quantitative myelin information. ### Main Contributions: - **Theoretical Basis**: Elaborately introduce the basic concepts of magnetic susceptibility and magnetic susceptibility imaging techniques. - **Experimental Verification**: Through in vitro and in vivo experiments, verify the strong correlation between the paramagnetic susceptibility map and myelin distribution. - **Application Prospects**: Discuss the potential applications of this technology in neuroscience research and clinical diagnosis, especially in the research of diseases such as multiple sclerosis (MS). In conclusion, this paper aims to provide a basic resource for researchers entering this rapidly developing field to help them understand and apply the magnetic susceptibility source separation technology.
