Abstract:Cryo-electron microscopy (cryo-EM) has achieved near-atomic level resolution of biomolecules by reconstructing 2D micrographs. However, the resolution and accuracy of the reconstructed particles are significantly reduced due to the extremely low signal-to-noise ratio (SNR) and complex noise structure of cryo-EM images. In this paper, we introduce a diffusion model with post-processing framework to effectively denoise and restore single particle cryo-EM images. Our method outperforms the state-of-the-art (SOTA) denoising methods by effectively removing structural noise that has not been addressed before. Additionally, more accurate and high-resolution three-dimensional reconstruction structures can be obtained from denoised cryo-EM images.

What problem does this paper attempt to address?
The paper primarily addresses the issues of low signal-to-noise ratio (SNR) and complex noise structures in single-particle cryo-electron microscopy (cryo-EM) images by proposing a method that combines a diffusion model with a post-processing framework to effectively remove noise and restore images. Specifically, the paper tackles the following key problems: 1. **Effective Denoising**: Due to the typically extremely low SNR in cryo-EM images, the resolution and accuracy of reconstructed particles are significantly reduced. To address this, the researchers propose a diffusion model-based method to improve image quality. 2. **Structural Noise Removal**: In addition to background noise, cryo-EM images also contain structural noise caused by carbon, ice background, and molecular conformation changes. This noise interferes with protein structure analysis. The method in this study can effectively remove such structural noise. 3. **Improving 3D Reconstruction Accuracy**: By denoising the images, more accurate and high-resolution 3D structural reconstruction results can be obtained. In summary, the main goal of this study is to develop a new method that can effectively remove noise (including structural noise) from cryo-EM images under extremely low SNR conditions, thereby improving the resolution and accuracy of single-particle cryo-EM images and ultimately obtaining higher quality 3D structural information. Experimental results on both simulated and real datasets demonstrate the excellent performance of this method.
