Abstract:Two neutral Glycyrrhiza polysaccharide (GP), named GP-LA and GP-HA, with molecular weights of 3.023 × 105 and 1.291 × 105, respectively, were extracted from Glycyrrhiza residues by hot acid extraction (HA) and low-temperature acid extraction (LA) and purified by column chromatography. Comprehensive analysis showed that the backbone of GP was composed of →4)-β-D-Glcp-(1→. For GP-LA, the side chain probably composed of β-L-Araf-(1→, →5)-β-L-Araf-(1→, →4)-β-D-Xylp-(1→, β-D-Glcp-(1→, →3,4)-α-D-Glcp-(1→, →4,6)-α- D-Glcp-(1→, and for GP-LA, the side chain probably composed of→6)-β-D-Galp-(1→, -α-D-Glcp-(1→, →3,4)-α-D-Glcp-(1→, →2,4)-α-D-Glcp-(1→, →4,6)- D-Glcp-(1→. The GP had a triple helix conformation and numerous irregular spherical particles with smooth surfaces. Atomic force microscopy (AFM) confirmed that GP-HA possessed single-branched structures and GP-LA possessed cross-linked network structures. X-ray diffraction (XRD) analysis revealed that GP-LA had both crystalline and amorphous structures, but GP-HA had no crystal structure. Furthermore, the results indicated that GP-LA and GP-HA exhibited certain inhibition activity on α-glucosidase. CD spectra and fluorescence intensity measurements confirmed that the secondary structure of α-glucosidase changed with the addition of GP.

Abstract:A branched ionic polysaccharide isolated from the sap of the Chinese lac tree (Rhus vernicifera) was chemically modified by sulfation using sulfur trioxide–pyridine (SO3·Py) complex as a reagent. Effects of molar ratio of SO3·Py complex to sugar unit, reaction time and reaction temperature on degree of sulfation (DS) and molecular weights of products were studied. Solvent was another important factor affecting sulfation reaction. In different solvents, when the other conditions remained constant, DS and molecular weights were in the following order: DMSO>DMF>FA (formamide) and DMSO<DMF<FA, respectively. The results depended on the nucleophilicity of the solvents used. The structures of sulfated polysaccharides were investigated by GPC, UV–Vis, FT-IR and FT-Raman spectroscopy. The results indicated that unsaturated bonds and anhydrogalactose residues were formed in lacquer polysaccharides due to the strong dehydration of SO3·Py complex. Based on these, we deduced that degradation of polysaccharide in the sulfation reaction process involved both dehydrolysis and hydrolytic degradation.

Abstract:Litchi polysaccharides are a kind of macromolecular polymers with various biological activities and a wide range of molecular weights. In this study, two separate fractions, with average molecular weights of 378.67 kDa (67.33%) and 16.96 kDa (6.95%), which were referred to as LP1 and LP2, respectively, were separated using an ultrafiltration membrane. Their physicochemical properties, and immunomodulatory and prebiotic activity were compared. The results revealed that LP2 contained more neutral sugar, arabinose, galactose and rhamnose, but less uronic acid, protein, mannose and glucose than LP1. Compared with LP1, LP2 possessed higher solubility and lower apparent viscosity. LP2 exhibited stronger stimulation on macrophage secretion of NO, TNF-α and IL-6, as well as better proliferation of Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus casei and Bifidobacterium adolescentis. These results suggest that an ultrafiltration membrane might be used to prepare a highly-active polysaccharide fraction from litchi pulp that may be used for food or drug development.
Structural differences and conformational characterization of five bioactive polysaccharides from Lentinus edodes

Abstract:Five polysaccharide fractions (LST1, LST2, LJT1, LJT2 and LJT3) from Lentinus edodes were isolated and purified. Their molecular weights (Mw) were estimated as 65.5~697kDa by using high-performance gel permeation chromatography (HPGPC). The results from monosaccharide analysis indicated that all of them contained glucose as main sugar component. The chemical and spectroscopic analyses implied that these fractions were composed of a main chain consisting of several (2~6) β-(1→3)-d-glucose residues with branching at C6 position. The predicted structural units of the purified polysaccharides were established with different degrees of branching (DB). Congo-red test, X-ray diffraction (XRD) and atomic force microscope (AFM) were used to verify that the natural lentinan contained triple helical spatial conformation. Furthermore, the results from anti-tumor tests in vitro and in vivo revealed that these polysaccharide fractions could have dual anti-tumor effects, including killing tumor cells directly and immunoregulation with low toxicity to the host. Interestingly, the high Mw polysaccharides had smaller DB values and displayed greater anti-tumor effects.

Physicochemical Properties and Anticoagulant Activity of Purified Heteropolysaccharides from Laminaria japonica

Abstract:Laminaria japonica is widely consumed as a key food and medicine. Polysaccharides are one of the most plentiful constituents of this marine plant. In this study, several polysaccharide fractions with different charge numbers were obtained. Their physicochemical properties and anticoagulant activities were determined by chemical and instrumental methods. The chemical analysis showed that Laminaria japonica polysaccharides (LJPs) and the purified fractions LJP0, LJP04, LJP06, and LJP08 mainly consisted of mannose, glucuronic acid, galactose, and fucose in different mole ratios. LJP04 and LJP06 also contained minor amounts of xylose. The polysaccharide fractions eluted by higher concentration of NaCl solutions showed higher contents of uronic acid and sulfate group. Biological activity assays showed that LJPs LJP06 and LJP08 could obviously prolong the activated partial thromboplastin time (APTT), indicating that they had strong anticoagulant activity. Furthermore, we found that LJP06 exerted this activity by inhibiting intrinsic factor Xase with higher selectivity than other fractions, which may have negligible bleeding risk. The sulfate group may play an important role in the anticoagulant activity. In addition, the carboxyl group and surface morphology of these fractions may affect their anticoagulant activities. The results provide information for applications of L. japonica polysaccharides, especially LJP06 as anticoagulants in functional foods and therapeutic agents.

Abstract:Congenital Trypanosoma cruzi infection is a highly pathogenic and underreported condition. Early recognition is essential for effective treatment. Umbilical chord blood from newborns (n = 302) to infected mothers was analyzed with microhematocrit, hemoculture, and PCR methods. Each subject was then followed serologically. In calibrated suspensions of T. cruzi in blood, the sensitivity of PCR was 27-fold higher than hemoculture. However, this advantage was not reflected during routine testing of samples from maternities, partly because of the uneven distribution of few parasites in small samples. Levels of detection of congenital infection were 2.9% (8/272) for microhematocrit, 6.3% (18/287) for hemoculture, 6.4% (15/235) for PCR, and 8.9% (27/302) for cumulated results. Evaluation against the standard of delayed serology indicates that the regular application of PCR, hemoculture, and microhematocrit to blood samples allows the rapid detection of about 90% of the congenitally infected newborns, in samples that can be obtained before the mother and child leave the maternity ward.
Comparative Study on the Structural Properties and Bioactivities of Three Different Molecular Weights of Lycium barbarum Polysaccharides

Abstract:The molecular weight, the triple-helix conformation, the monosaccharide content, the manner of glycosidic linkages, and the polysaccharide conjugates of polysaccharides all affect bioactivity. The purpose of this study was to determine how different molecular weights affected the bioactivity of the Lycium barbarum polysaccharides (LBPs). By ethanol-graded precipitation and ultrafiltration membrane separation, one oligosaccharide (LBPs-1, 1.912 kDa) and two polysaccharides (LBPs-2, 7.481 kDa; LBPs-3, 46.239 kDa) were obtained from Lycium barbarum. While the major component of LBPs-1 and LBPs-2 was glucose, the main constituents of LBPs-3 were arabinose, galactose, and glucose. LBPs-2 and LBPs-3 exhibited triple-helix conformations, as evidenced by the Congo red experiment and AFM data. Sugar residues of LBPs-2 and LBPs-3 were elucidated by NMR spectra. The polysaccharides (LBPs-2 and LBPs-3) exhibited much higher antioxidant capacities than oligosaccharide (LBPs-1). LBPs-3 showed higher oxygen radical absorbance capacity (ORAC) and superoxide dismutase (SOD) activity than LBPs-2, but a lower capability for scavenging ABTS+ radicals. In zebrafish, LBPs-2 and LBPs-3 boosted the growth of T-lymphocytes and macrophages, enhanced the immunological response, and mitigated the immune damage generated by VTI. In addition to the molecular weight, the results indicated that the biological activities would be the consequence of various aspects, such as the monosaccharide composition ratio, the chemical composition, and the chemical reaction mechanism.

Abstract:A novel acidic heteropolysaccharide (LCP-90-1) was isolated and purified from a traditional "heat-clearing" Chinese medicine, Lysimachia christinae Hance. LCP-90-1 ( Mw , 20.65 kDa) was composed of Man, Rha, GlcA, Glc, Gal, and Ara, with relative molar ratios of 1.00: 3.00: 11.62: 1.31: 1.64: 5.24. The backbone consisted of 1,4- alpha -D-Glc p A, 1,4- alpha -D-Glc p , 1,4- beta -L-Rhap , and 1,3,5- alpha -L-Ara f , with three branches of beta -D-Gal p -(1 -> 4)- beta -L-Rha p -(1 ->, alpha -L-Ara f -(1 -> and alpha -D-Man p -(1 -> attached to the C-5 position of 1,3,5- alpha -L-Ara f . LCP-90-1 exhibited potent anticomplement activity (CH 50 : 135.01 +/- 0.68 mu g/mL) in vitro , which was significantly enhanced with increased glucuronic acid (GlcA) content in its degradation production (LCP-90-1-A, CH50 : 28.26 +/- 0.39 mu g/mL). However, both LCP-90-1 and LCP90-1-A were inactivated after reduction or complete acid hydrolysis. These observations indicated the important role of GlcA in LCP-90-1 and associated derivatives with respect to anticomplement activity. Similarly, compared with LCP-90-1, the antioxidant activity of LCP-90-1-A was also enhanced. Thus, polysaccharides with a high content of GlcA might be important and effective substances of L. christinae .

Abstract:Polysaccharides were extracted by hot water and alkali in sequence from Dolichos lablab L. hull, and further purified by ion-exchange and gel columns. Hot water extracted D. lablab hull polysaccharide (DLHP) was rich in glucuronoxylan and pectin, and alkali extracted polysaccharide (DLHAP) mostly embraced glucuronoxylan. The structures of purified glucuronoxylans from DLHP and DLHAP were mainly analyzed by HPAEC-PAD, methylation combined with GC–MS, NMR and SEC-MALLS. DLHP-1 was identified as acetylated glucuronoxylan containing →4)-β-Xylp-(1→ backbone with substitution at O-2 site by α-GlcpA/4-O-methyl-α-GlcpA. The molar ratio of β-Xylp to α-GlcpA was 6.9:1, and acetylation was mainly at O-3 site of β-Xylp with acetylation degree of 21.5%. DLHP-1 and DLHP-2 had similar physicochemical properties, except for molecular weight (Mw). DLHAP-1 was the non-methylated glucuronoxylan almost without acetylation, and it had the molar ratio of β-Xylp to α-GlcpA of 5.6:1. Besides, DLHP-1 (Mw of 20.0 × 103 g mol−1) adopted semi-flexible chain, while DLHAP-1 (Mw of 15.4 × 103 g mol−1) showed flexible chain. These results provided a structural basis for study on polysaccharides from D. lablab hull, which was benefit for understanding biological activities and developing functional food or pharmaceuticals of D. lablab.

Abstract:Intracellular polysaccharides (iPs) were separated and purified from Coriolus versicolor LH1 mycelia and characterized for their α-glucosidase inhibitory properties. Three iP fractions (iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1) were extracted, separated, and purified from LH1 mycelia using microwave extraction technology, a DEAE-Sepharose CL-6B column, a Diaion HP20 macroporous adsorption column, and a Sephadex™ G-50 gel-permeation column. The principal constituents of iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1 were saponins and polyphenoic compound mixtures. The enzyme inhibition activity, IC(50) values, of these three fractions were 1.7, 1.8, and 0.8 mg/mL, respectively. The α-glucosidase inhibitory properties were related to the presence of α-(1,4) glycosidic linkages in the polysaccharide structure and the total relative percentage of d-glucose and d-galactose in the structure of polysaccharides, other than triterpenoids.

Abstract:At present, the structure-activity relationship of polysaccharides is a common and important focus in the fields of glycobiology and carbohydrate chemistry. To better understand the effect of specific polysaccharide structures on bioactive orientation, four homogeneous polysaccharides from Lycii fructus, one neutral along with three acidic polysaccharides, were purified, structurally characterized and comparatively evaluated on the antioxidative and anti-aging activities. The GC–MS-based monosaccharide composition analysis and methylation results showed that the LFPs had similar glycosyl types but varied proportions. Nuclear magnetic resonance (NMR) spectroscopy showed that LFPs consisted of arabinogalactan, rhamnogalacturonan and homogalacturonan structural domains. The results of the structure-activity relationship indicated that the antioxidative activity was positively correlated with the galacturonic acid (GalA) content, while the neutral multi-branched chains might be responsible for the anti-aging activity. This study is the first time to compare the principal structures and multiple biological activities of LFPs, which provided a reference for the industrial development and deep excavation of the health value of LFPs.

Abstract:The effect of partial acid hydrolysis on the physical and chemical properties of galactomannan, arabinoxylan, and xyloglucan was investigated. Polysaccharides were treated at 50 °C with hydrochloric acid for 3–48 h. Portions of isopropanol (i-PrOH) were added sequentially to the hydrolyzates, resulting in fractions that were collected by centrifugation. As expected, a significant reduction of weight-average molecular weight (Mw) was observed with increasing hydrolysis time. Fractional precipitation was successfully applied to collect at least one polymer fraction with dispersity (Đ) close to one for each polysaccharide. The monosaccharide composition analysis showed that the partial hydrolysis usually lowered the relative amount of side chains, with the exception of galactomannan, where the composition remained largely unaffected. Estimation of the polymer conformation in solution, through evaluation of the Mark-Houwink parameter coefficient (α), confirmed that acid hydrolysis influenced the polysaccharides’ conformation. It was demonstrated that acid treatment in dilute solution followed by fractional isopropanol precipitation is a method, extendible to a variety of polysaccharides, to obtain materials of decreased molecular weight and low dispersity with slightly altered overall composition and conformation.

Abstract:Currently, most of the polysaccharide fractions are purified using the methods usually limited to the laboratory scale or easily raising question of residual reagent, which may obstruct the future application of polysaccharide. In our study, we employed sequential ultrafiltration with different membranes combined with HPGPC to purify polysaccharide fractions from the water extract of Lentinus edodes directly. The chemical composition, morphology and structure characteristic were analyzed by chemical methods and instrumental analysis including automatic amino acid analyzer, Elemental Analyzer, SEM, HPGPC, HPAEC-PAD, FT-IR and NMR. Results indicated that three polysaccharide fractions named as LE-UF-1, LE-UF-2 and LE-UF-3 were successfully purified using ultrafiltration membranes with MWCO of 2.5, 5 and 10 kDa in sequence. Mw and polysaccharide content of LE-UF-1 (~136 kDa, 86.11%) were the highest, followed by LE-UF-2 (14–61 kDa, 49.82%) and LE-UF-3 (14–35 kDa, 22.44%). Surprisingly, LE-UF-2 had 11.73% amino acid, and LE-UF-3 had 19.64% amino acid. The polysaccharide in all the three fractions was heteropolysaccharide that mainly consisted of Glc, Gal and Man. There might be more β-sugar residues in LE-UF-1 and LE-UF-2, while more α-sugar residues in LE-UF-3. Our study provides a greener and more efficient potential procedure for large-scale polysaccharide purification, and is able to prepare the polysaccharide fractions from Lentinus edodes that may be used for food or drug development.

Modulation of the baroreceptor reflex by the dorsomedial hypothalamic nucleus and perifornical area.

Abstract:Neurons within the dorsomedial hypothalamic nucleus (DMH) and perifornical area (PeF), which lie within the classic hypothalamic defense area, subserve the cardiovascular response to psychological stress. Previous studies have shown that electrical stimulation of the hypothalamic defense area causes inhibition of the cardiac and (in some cases) sympathetic components of the baroreceptor reflex. In contrast, naturally evoked psychological stress does not appear to be associated with such inhibition. In this study, we tested the effect of specific activation of neurons within the DMH and PeF on the baroreflex control of renal sympathetic nerve activity and heart rate in urethane-anesthetized rats. Microinjection of bicuculline (a GABA(A) receptor antagonist) into the DMH caused dose-dependent increases in heart rate and renal sympathetic activity, shifted the baroreflex control of both variables to higher levels (i.e., increased the upper and lower plateaus of the baroreflex function curves, and increased the threshold, midpoint, and saturation levels of mean arterial pressure). The maximum gain of the sympathetic component of the baroreflex was also increased, while that of the cardiac component was not significantly changed. Increases in the midpoint were very similar in magnitude to the evoked increases in baseline mean arterial pressure. Microinjection of bicuculline into the PeF evoked very similar effects. The results indicate that disinhibition of neurons in the DMH/PeF region not only increases sympathetic vasomotor activity and heart rate but also resets the baroreceptor reflex such that it remains effective, without any decrease in sensitivity, over a higher operating range of arterial pressure.
Effect of Salts on Laccase‐Catalyzed Polymerization of Lignosulfonate

Abstract:Enzymatic polymerization of lignosulfonate (LS) has a high potential for various applications. Here, the effect of different ions in low concentrations on enzymatic polymerization of LS was investigated, including salt solutions consisting of mono‐ and dicarboxylic acids, sulfate, phosphate and chloride with sodium as counter ion. LS polymerization was followed by viscometry and size exclusion (SEC) chromatography. Interestingly, there was only a small effect of ions on the activity of the laccase on standard substrate ABTS, while the effect on polymerization of LS was substantially different. The presence of acetate led to a 39% higher degree of polymerization (DP) for LS. Small angle X‐ray scattering (SAXS) revealed that the structure of the enzyme was largely unaffected by the ions, while the determination of the zeta potential showed that those ions conveying higher negative surface charges onto LS particles showed lower DPs, than those not affecting the surface charge. Further, electron paramagnetic resonance (EPR) spectroscopy showed 5‐times higher intensity in phenoxyl radicals for the monovalent ions compared to the divalent ones. It was concluded that the DPs of LS could be tuned in the presence of certain ions, by facilitating the interaction between the laccase substrate‐binding site and the LS molecules.

Polysaccharide metabolism in Anacardiaceae (Asian lacquer) cross-linked polymers elucidated using in situ trimethylsilylation pyrolysis-gas chromatography-mass spectrometry

Abstract:Carbohydrates from polysaccharides in natural thermoset Anacardiaceae polymers of Gluta usitata, Toxicodendron succedaneum and Toxicodendron vernicifluum were identified using pyrolysis-gas chromatography-mass spectrometry with in situ trimethylsilylation. Pyrolysates resulting from the pyrolytic intermolecular chain scission of the polysaccharides were used to elucidate monomeric units. Polysaccharides, dispersed in the phenolic lacasse catalysed cross-linked macromolecules, showed to be metabolised through various catabolic and anabolic routes. Galactose functionalities, abundantly present in the polysaccharides were determined to be enzymatically converted to glucose-6-phosphate, followed by conversion via glycolysis and pentose phosphate pathways. Determination of specific routes of carbohydrate modification via glycolysis and pentose phosphate pathways facilitated differentiating G. usitata, T. succedaneum and T. vernicifluum polymers, based on the carbohydrate content. It was also found that uronic type acids, present as end groups of the branched polysaccharide structure, were biochemically converted to aldonic acids. Following the pentose phosphate and glycolysis routes, carbohydrates in G. usitata and T. vernicifluum polymers showed to be further modified via shikimate and cinnamate pathways to produce phenylpropanoid compounds. Parent molecules and pyrolysis products thereof were verified using analytical standards of high purity. The mass spectra and Kovats retention indices were compiled in an AMDIS library, which can be made available on request.
Structural Characterization and Anticomplement Activity of an Acidic Heteropolysaccharide from Lysimachia christinae Hance

Abstract:A novel acidic heteropolysaccharide (LCP-90-1) was isolated and purified from a traditional "heat-clearing" Chinese medicine, Lysimachia christinae Hance. LCP-90-1 ( Mw , 20.65 kDa) was composed of Man, Rha, GlcA, Glc, Gal, and Ara, with relative molar ratios of 1.00: 3.00: 11.62: 1.31: 1.64: 5.24. The backbone consisted of 1,4- α -D-Glc p A, 1,4- α -D-Glc p , 1,4- β -L-Rha p , and 1,3,5- α -L-Ara f , with three branches of β -D-Gal p -(1 → 4)- β -L-Rha p -(1→, α -L-Ara f -(1→ and α -D-Man p -(1→ attached to the C-5 position of 1,3,5- α -L-Ara f . LCP-90-1 exhibited potent anticomplement activity (CH 50 : 135.01 ± 0.68 μg/mL) in vitro , which was significantly enhanced with increased glucuronic acid (GlcA) content in its degradation production (LCP-90-1-A, CH 50 : 28.26 ± 0.39 μg/mL). However, both LCP-90-1 and LCP90-1-A were inactivated after reduction or complete acid hydrolysis. These observations indicated the important role of GlcA in LCP-90-1 and associated derivatives with respect to anticomplement activity. Similarly, compared with LCP-90-1, the antioxidant activity of LCP-90-1-A was also enhanced. Thus, polysaccharides with a high content of GlcA might be important and effective substances of L. christinae . Abstract:Summary In this study, three litchi glycans (Lzp1‐Lzp3) with molecular weights (Mw) of 2.25×106 Da, 6.00×103 Da and 3.24×102 Da were purified, and their structural characteristics, biological activities and the structure‐activity relationships were investigated. The results showed that Lzp1‐Lzp3 had similar inhibitory effects on Escherichia coli and Staphylococcus aureus, as evidenced by the fact that there were no significant correlations between the Mw, active groups, monosaccharide compositions of Lzp1‐Lzp3 and their antimicrobial activities. Lzp1 consisted of more mannose, galactose and arabinose, and less of ribose and xylose, thus leading to its higher antioxidant activities of DPPH, ABTS, hydroxyl and superoxide radical scavenging and ferric‐ion reducing with the values of 78.0%, 99.0%, 90.2%, 1.08 and 3.93, respectively. Lzp2 contained the more of sulfonic acid and uronic acid, and the least of glucose possessed the best effect on the promotion of EPS production (24.77 mg/mL) by Weissella confusa, and the proliferation of Weissella confusa, Lacticaseibacillus rhamnosus, Bifidobacterium bifidum and Bifidobacterium breve with the OD600 ranging from 0.278‐1.082. This study can provide a reference for the preparation of litchi glycans possessed specific biological activity.

[Effects of different extraction methods on chemical compositions and biological activities of polysaccharides from Lycium barbarum]

Abstract:In this study, five polysaccharides from Lycium barbarum(LBPs)(LBP-1-LBP-5) were selectively extracted by different extraction methods, and the chemical composition, structural characteristics, and biological activities of LBPs were explored. The results of chemical composition analysis showed that alkaloids were not detected in the five LBPs. The total polysaccharide content was(81.95%±1.6%)-(92.96%±0.76%), the uronic acid content was(8.26%±0.46%)-(24.81%±0.46%), and the protein content was(0.06%±0.03%)-(1.35%±0.13%). The monosaccharide compositions of the five LBPs were basically same, mainly including glucose, xylose, and galactose. However, there was significant difference in the content ratio of different monosaccharide. The results of infrared spectra analysis indicated that the five LBPs had typical infrared spectral characteristics of polysaccharides. The results of nuclear magnetic resonance characteristic spectrum analysis revealed that the five LBPs had two configurations of α and β. Meanwhile, there were triple helix structures in LBP-2, LBP-3, and LBP-4, which enhanced the activities of polysaccharides. The results of activities screening suggested that the biological activities of the five LBPs were significantly different. LBP-3 showed the highest lipid oxidation clearance rate, and its antioxidant activity was equivalent to that of the positive control group. The inhibitory rate of LBP-4 on α-amylase and its activation rate of alcohol dehydrogenase were better than those of other fractions, and the inhibitory rate of LBP-4 on α-amylase was slightly higher than that of the positive control group when the mass concentration was 10 g·L~(-1). LBP-2 showed stronger inhibitory activity against α-glucosidase and hyaluronidase. This study provides references for the precise development and utilization of LBPs.
Study of antioxidant activities of sulfated polysaccharides from Laminaria japonica

Abstract:The composition, molecular weight and in vitro antioxidant activity of various sulfated polysaccharides obtained by anion exchange chromatography, acid hydrolysis and radical process degradation of the crude sulfated polysaccharide extracted from Laminaria japonica were compared. The low sulfated F-A2, with a peak-molecular weight (Mp) of 5–15 kDa, 14.5% sulfated ester and 21.8% glucuronic acid, exhibited a very strong antioxidant activity on superoxide and hydroxyl radicals, with activity even higher than that of large molecular weight fractions F-A and F-B. However, highly sulfated fractions with a peak-molecular weight below 15 kDa had much lower antioxidant activities than other fractions. These results indicated that the sulfate group of the low molecular weight fractions represents a physical block for the reaction with oxygen radicals. The chemical properties and antioxidant activities of sulfated polysaccharide fractions obtained by radical process degradation of crude sulfated polysaccharide were quite different from those obtained by acid hydrolysates. By radical process degradation, the high molecular weight was decreased to give LM2 (Mp 8 kDa) and LM1 (Mp 1.5 kDa), with a yield of 40% and 15%, respectively. LM2 was enriched with fucose and sulfated ester, while containing low amounts of glucuronic acid. The antioxidant activity showed that LM2 was unable to scavenge either superoxide or hydroxyl radical, which suggested that radical process degradation targeted mainly ascopyllan-like species rich in glucuronic acid, while the fraction rich in sulfated l-fucose remained unchanged. However, LM1 with Mp 1.5 kDa still retained apparent scavenging ability for superoxide radical, although it contained no glucuronic acid and certain amounts of galactose and mannose as main neutral sugars. These result suggest that the antioxidant activity of sulfated polysaccharides is apparently related not only to molecular weight and sulfated ester content, as previously determined, but also to glucuronic acid and fucose content.
