Abstract:Women of the Yakima Indian Nation, a northwest Native American population, are known to have an increased prevalence of a rheumatoid arthritis-like disease characterized by erosive arthritis, frequent involvement of metacarpophalangeal and wrist joints, and positive rheumatoid factor. These patients are frequently positive for antinuclear antibodies and often demonstrate adverse reactions to gold therapy. HLA antigens were determined for 29 Yakima Indians with this disease, but there was no increased frequency of either HLA-Dw4 or HLA-DR4, in contrast to other populations with rheumatoid arthritis. There was, however, a trend toward an increase in HLA-B40 and a decrease in HLA-DR8. The relative risk for rheumatoid arthritis in Yakima Indians was 2.53 in the presence of B40 and 0.28 in the presence of DR8.

Abstract:Heavy metals such as Cd pose environmental problems and threats to a variety of organisms. The effects of cadmium (Cd) on the growth and activities of photosystem I (PSI) and photosystem II (PSII) of Chlorella pyrenoidosa were studied. The growth rate of cells treated with 25 and 100 µM of Cd for longer than 48 h were significantly lower than the control, accompanying with the inhibition of photosynthesis. The result of quantum yields and electron transport rates (ETRs) in PSI and PSII showed that Cd had a more serious inhibition on PSII than on PSI. Cd decreased the efficiency of PSII to use the energy under high light with increasing Cd concentration. In contrast, the quantum yield of PSI did not show a significant difference among different Cd treatments. The activation of cyclic electron flow (CEF) and the inhibition of linear electron flow (LEF) due to Cd treatment were observed. The photochemical quantum yield of PSI and the tolerance of ETR of PSI to Cd treatments were due to the activation of CEF around PSI. The activation of CEF also played an important role in induction of non-photochemical quenching (NPQ). The binding features of Cd ions and photosystem particles showed that Cd was easier to combine with PSII than PSI, which may explain the different toxicity of Cd on PSII and PSI.
Effect of cadmium in the microalga Chlorella sorokiniana: A proteomic study

Abstract:Cadmium is one of the most common heavy metals in contaminated aquatic environments and one of the most toxic contaminants for phytoplankton. Nevertheless, there are not enough studies focused on the effect of this metal in algae. Through a proteomic approach, this work shows how Cd can alter the growth, cell morphology and metabolism of the microalga Chlorella sorokiniana. Using the sequential window acquisition of all theoretical fragment ion spectra mass spectrometry (SWATH-MS), we concluded that exposure of Chlorella sorokiniana to 250 μM Cd2+ for 40 h caused downregulation of different metabolic pathways, such as photosynthesis, oxidative phosphorylation, glycolysis, TCA cycle and ribosomal proteins biosynthesis. However, photorespiration, antioxidant enzymes, gluconeogenesis, starch catabolism, and biosynthesis of glutamate, cysteine, glycine and serine were upregulated, under the same conditions. Finally, exposure to Cd also led to changes in the metabolism of carotenoids and lipids. In addition, the high tolerance of Chlorella sorokiniana to Cd points to this microalga as a potential microorganism to be used in bioremediation processes.
Toxicity of Cu (II) to the green alga Chlorella vulgaris: a perspective of photosynthesis and oxidant stress

Abstract:The toxic effects of Cu (II) on the freshwater green algae Chlorella vulgaris and its chloroplast were investigated by detecting the responses of photosynthesis and oxidant stress. The results showed that Cu (II) arrested the growth of C. vulgaris and presented in a concentration- and time-dependent trend and the SRichards 2 model fitted the inhibition curve best. The chlorophyll fluorescence parameters, including qP, Y (II), ETR, F v /F m , and F v /F 0, were stimulated at low concentration of Cu (II) but declined at high concentration, indicating the photosystem II (PSII) of C. vulgaris was destroyed by Cu (II). The chloroplasts were extracted, and the Hill reaction activity (HRA) of chloroplast was significantly decreased with the increasing Cu (II) concentration under both illuminating and dark condition, and faster decline speed was observed under dark condition. Activities of superoxide dismutase (SOD) and catalase (CAT) and malondialdehyde (MDA) content were also significantly decreased at high concentration Cu (II), companied with a large number of reactive oxygen species (ROS) production. All these results indicated a severe oxidative stress on algal cells occurred as well as the effect on photosynthesis, thus inhibiting the growth of algae, which providing sights to evaluate the phytotoxicity of Cu (II).

Abstract:Cadmium, which is widely used in electroplating industry, chemical industry, electronic industry and nuclear industry, is harmful to human health and ecological environment. The effects of Cd at different initial concentrations on biomass, antioxidant enzyme activity and ultrastructure of Chlorella vulgaris were analysed in the present study. The results showed that C. vulgaris maintained a slow-growth trend at 3.0 mg/L Cd, and the peroxidase (POD) enzyme activity reached the highest at this concentration, which indicated that C. vulgaris could resist the oxidative damage of cells by increasing the enzyme activity, so as to improve the tolerance of C. vulgaris to Cd. When the concentration of Cd was 5.0 mg/L, although the activity of the superoxide dismutase enzyme was still very high, POD enzyme could not remove the hydrogen peroxide produced in cells in time, leading to cell damage and even death. Therefore, when the concentration reached 5.0 mg/L, the growth of C. vulgaris began to decline after four days of stress, and the cell structure was significantly damaged after six days of stress. And the higher concentration of Cd caused more Cd accumulation in cells and a serious damage to C. vulgaris. C. vulgaris can be used as an early warning indicator of Cd pollution, and it can be used for bioremediation of Cd contaminated water through tolerant subculture.GRAPHICAL

Abstract:Chlorella vulgaris was tested to assess their toxicities in freshwater contaminated by the metal compounds of copper (Cu) and cadmium (Cd), both singly and combined. Exposure to 0.5 and 1.5μM Cu or 1.0 and 2.0μM Cd alone significantly decreased algal growth and chlorophyll content and increased reactive oxygen species (ROS) content. Two-way ANOVA analysis shows that the combination of these two metal compounds decreased cell growth, chlorophyll content and increased ROS content synergistically. The highest algal cell inhibition was 78.55%, the lowest levels of chl a, chl b and total-chl were 10.59%, 33.33% and 17.94% of the control, respectively. The highest increase in ROS was 9.15-fold greater than that of the control when exposed to Cu(1.5)+Cd(2.0). Real-time PCR shows that Cu and Cd reduced the transcript abundance of psbA and rbcL, but without a synergistic interaction, whereas Cu and Cd increased the transcript abundance of psaB synergistically. These results demonstrate that Cu and Cd independently inhibit PSII activity and CO2 assimilation, but synergistically increase ROS content to disrupt chlorophyll synthesis and inhibit cell growth.

Abstract:Individual differences in working memory (WM) predict principled variation in tasks of reasoning, response time, memory, and other abilities. Theoretically, a central function of WM is keeping task-relevant information easily accessible while suppressing irrelevant information. The present experiment was a novel study of mental control, using performance in the game Taboo as a measure. We tested effects of WM capacity on several indices, including perseveration errors (repeating previous guesses or clues) and taboo errors (saying at least part of a taboo or target word). By most measures, high-span participants were superior to low-span participants: High-spans were better at guessing answers, better at encouraging correct guesses from teammates, and less likely to either repeat themselves or produce taboo clues. Differences in taboo errors occurred only in an easy control condition. The results suggest that WM capacity predicts behavior in tasks requiring mental control, extending this finding to an interactive group setting.

Abstract:Cadmium (Cd) is one of the most toxic pollutants found in aquatic ecosystems. Although gene expression in algae exposed to Cd has been studied at the transcriptional level, little is known about Cd impacts at the translational level. Ribosome profiling is a novel translatomics method that can directly monitor RNA translation in vivo. Here, we analyzed the translatome of the green alga Chlamydomonas reinhardtii following treatment with Cd to identify the cellular and physiological responses to Cd stress. Interestingly, we found that the cell morphology and cell wall structure were altered, and starch and high-electron-density particles accumulated in the cytoplasm. Several ATP-binding cassette transporters that responded to Cd exposure were identified. Redox homeostasis was adjusted to adapt to Cd toxicity, and GDP-L-galactose phosphorylase (VTC2), glutathione peroxidase (GPX5), and ascorbate were found to play important roles in maintaining reactive oxygen species homeostasis. Moreover, we found that the key enzyme of flavonoid metabolism, i.e., hydroxyisoflavone reductase (IFR1), is also involved in the detoxification of Cd. Thus, in this study, translatome and physiological analyses provided a complete picture of the molecular mechanisms of green algae cell responses to Cd.
Alleviation of cadmium-induced photoinhibition and oxidative stress by melatonin in Chlamydomonas reinhardtii

Abstract:As one of the most threatening challenges to the natural environment and human health, cadmium (Cd) pollution has seriously impacted natural organisms. Green algae, such as Chlamydomonas reinhardtii (C. reinhardtii), can provide a safer, lower cost, and more effective ecological approach to the treatment of heavy metal ions in wastewater due to their sorption properties. However, heavy metal ions affect C. reinhardtii when adsorbed. Melatonin is able to protect the plant body from damage when the plant is under biotic/abiotic stress. Therefore, we investigated the effects of melatonin on the cell morphology, chlorophyll content, chlorophyll fluorescence parameters, enzymatic activity of the antioxidant system, gene expression, and the ascorbic acid (AsA)-glutathione (GSH) cycle of C. reinhardtii under the stress of Cd (13 mg/L). Our results indicated that Cd significantly induced photoinhibition and overaccumulation of reactive oxygen species (ROS). By application with the concentration of 1.0 μM melatonin, the algal solute of C. reinhardtii under the Cd stress gradually regained its green color, the cell morphology became intact, and the photosynthetic electron transport function was retained. However, in the melatonin-silenced strain, there was a significant decrease in all of the above indicators. In addition, the use of exogenous melatonin or the expression of endogenous melatonin genes could enhance the intracellular enzyme activities of catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), ascorbate peroxidase (APX), and glutathione reductase (GR). It also upregulated the expression of active enzyme genes such as SOD1, CAT1, FSD1, GSH1, GPX5, and GSHR1. These results indicate that the presence of melatonin effectively protects the activity of photosynthetic system II in C. reinhardtii, enhances antioxidant activity, upregulates gene expression in the AsA-GSH cycle, and reduces the level of ROS, thereby alleviating the damage caused by Cd toxicity.

Abstract:lytD, the structural gene of the Bacillus subtilis 168 N‐acetylglucosaminidase was localized at 310°, next to the tagABC operon. Sequence analysis revealed a monocistronic operon encoding a 95.6 kDa protein endowed with an export signal, the cleavage of which yields the monomer polypeptide (92.8 kDa) of the dimeric active form of the enzyme. Transcription is initiated at a sigma‐D (σD)‐dependent promoter and ends at a terminator common to lytD and the divergently transcribed tagABC operon. In addition, we report the sequence of the adjacent upstream ORF, transcribed in the same direction as lytD, which shows significant homology to phosphomannose isomerase‐encoding genes. Cell separation, motility, autolysis, cell wall turnover and growth were not affected in strains devoid of the N‐acetylglucosaminidase. A mutant deficient in the two most abundant autolysins, i.e. the LytC amidase and the glucosaminidase, exhibited the phenotype of the amidase‐deficient strains, revealing their non‐requirement for growth. This conclusion raises two fundamental questions: how does the cell undo the highly cross‐linked peptidoglycan so as to be able to grow, and what is the rote of the considerable amount of autolysin normally present? Possible answers to these questions are discussed.
Cadmium Toxicity to Two Marine Phytoplankton under Different Nutrient Conditions

Abstract:Cd accumulation and toxicity in two marine phytoplankton (diatom Thalassiosira weissflogii and dinoflagellate Prorocentrum minimum) under different nutrient conditions (nutrient-enriched, N- and P-starved conditions) were examined in this study. Strong interactions between the nutrients and Cd uptake by the two algal species were found. Cd accumulation as well as N and P starvation themselves inhibited the assimilation of N, P, and Si by the phytoplankton. Conversely, N starvation strongly inhibited Cd accumulation but no influence was observed under P starvation. However, the Cd accumulation difference between nutrient-enriched and N-starved cells was smaller when [Cd(2+)] was increased in the medium, indicating that net Cd accumulation was less dependent on the N-containing ligands at high-Cd levels. As for the subcellular distribution of the accumulated Cd, most was distributed in the insoluble fraction of T. weissflogii while it was evenly distributed in the soluble and insoluble fractions of P. minimum at low-Cd levels. A small percentage of cellular Cd (<15%) was adsorbed on the cell surface for both algae at the lowest [Cd(2+)], which increased when the [Cd(2+)] increased. Cd toxicity in phytoplankton was quantified as depression of growth and maximal photosynthetic system II quantum yield, and was correlated with the [Cd(2+)], intracellular Cd concentration, and Cd concentrations in the cell-surface-adsorbed, soluble, and insoluble fractions. According to the estimated median inhibition concentration (IC50) based on the different types of Cd concentration, the toxicity difference among the different nutrient-conditioned cells was the smallest when the Cd concentration in the soluble fraction was used, suggesting that it may be the best predictor of Cd toxicity under different nutrient conditions.

Abstract:The investigation of heavy metal wastewater treatment utilizing microalgae adsorption has been extensively demonstrated. However, the response mechanism based on metabolomics to analyze the time-series changes of microalgae under Cd stress has not been described in detail. In this study, SEM/TEM demonstrated that Cd accumulated on the cell surface of microalgae and was bioconcentrated in the cytoplasm, vesicles, and chloroplasts. Carbonyl/quinone/ketone/carboxyl groups (O C O), membrane polysaccharides ( OH), and phospholipids ( PO) were involved in the interaction of Cd ions, and the chlorophyll content underwent a process of decreasing in the early stage (1.62 mg/g at 48 h) and recovering to the normal level in the late stage, and the contents of MDA, GSH, and SOD were all increased (29.7 nmol/g, 0.23 mg/g, and 30.01 u/10 6 cells) and then gradually returned to the steady state. The results of EPS content and fluorescent labeling showed that Cd induced the overexpression and synthesis of extracellular polysaccharides and proteins, which is one of the defense mechanisms participating in the reduction of cellular damage by complexed Cd. Metabolomics results indicated that the malate synthesis pathway was activated after Cd-20 h, and the microalgal cells began to shift the metabolic pathway to storage lipid or polysaccharide biosynthesis. In the Calvin cycle, the expression of D-Sedoheptulose 7-phosphate in Cd-20 h_vs_ck and Cd-72 h_vs_Cd-20 h firstly declined and then increased, and the photosynthesis system was suppressed at the beginning, and then gradually returned to normal to maintain the successful development of the dark reaction. The results of time series analysis revealed that the response of microalgae to Cd was categorized into fast response and slow response to regulate cell adsorption and growth metabolism.

Abstract:Copper sulfate (CuSO4) is widely used in the control of algal blooms. Cu can promote or inhibit algal growth, while also affecting trace element uptake, therefore, the response mechanisms of algae cells under Cu2+ interference should be studied. In this study, wild-type Chlamydomonas reinhardtii (C. reinhardtii) and wall-less mutant C. reinhardtii were selected as the research objects. Except for the cell wall, these two algae were physiologically the same. While manipulating the concentration of Cu, the accumulation of Cu, Fe, Zn, and Mn by the two algal cell types was studied. The cell wall hindered the accumulation of Cu by cells and alleviated the toxicity of Cu to C. reinhardtii. The addition of Cu increased the accumulation of Fe by both cell types. In an environment with excess Cu, the total amount of Zn and Mn accumulated by cells also increased. On the one hand, this may be due to the synergistic and antagonistic effects of trace elements in the adsorption and uptake process, and on the other hand, it may be due to the changes in metal speciation in the culture medium. In addition, the difference in the total accumulation of various trace elements between wild-type and wall-less-type C. reinhardtii may be due to the structure and function differences between cell wall and cell membrane. At the same time, by measuring the changes in the levels of glutathione (GSH) in algal cells, the relevant mechanisms underlying the algae's uptake of trace elements by algae were further explored.

Psychology of Addictive Behaviors

Abstract:Alcoholics Anonymous (AA) attendance is predictive of increased abstinence for many problem drinkers and treatment referral to AA is common. Strong encouragement to acquire an AA sponsor is likewise typical, and findings about the benefits associated with social support for abstinence in AA support this practice, at least indirectly. Despite this widespread practice, however, prospective tests of the unique contribution of having an AA sponsor are lacking. This prospective study investigated the contribution of acquiring an AA sponsor using a methodologically rigorous design that isolated the specific effects of AA sponsorship. Participants were recruited from AA and outpatient treatment. Intake and follow-up assessments included questionnaires, semi-structured interviews, and urine toxicology screens. Eligibility criteria limited prior treatment and AA histories to clarify the relationship of interest while, for generalizability purposes, broad substance abuse criteria were used. Of the 253 participants, 182 (72%) provided complete data on measures central to the aims of this study. Overall reductions in alcohol, marijuana, and cocaine use were found over 12-months and lagged analyses indicated that AA attendance significantly predicted increased abstinence. During early AA affiliation but not later logistic regressions showed that having an AA sponsor predicted increased alcohol-abstinence and abstinence from marijuana and cocaine after first controlling for a host of AA-related, treatment, and motivational measures that are associated with AA exposure or are generally prognostic of outcome.
Cadmium-effect on performance and symbiotic relationship of microalgal-bacterial granules

Abstract:So far, the microalgal-bacterial granular sludge process has attracted growing interest as an emerging wastewater treatment technology. Cadmium ion (Cd2+) commonly found in wastewater is toxic to microorganisms, thus its effect on microalgal-bacterial granules was investigated in this study. Results showed that Cd2+ at the concentration above 1 mg/L could compromise the performances of microalgal-bacterial granules. The removal efficiency of chemical oxygen demand decreased from about 70% in the control to 42.2% and 25.0% after 30-day operation at the respective Cd2+ concentrations of 5 and 10 mg/L, while the ammonia-nitrogen removal also declined from 70.4% to 30.5% with the increase of the Cd2+ concentration from 1 to 10 mg/L, indicating that nitrifying bacteria were susceptive to the presence of Cd2+. It was further revealed that Cd2+ could stimulate the production of extracellular polymeric substances, e.g. 190.19 ± 7.04 mg/g VSS in the presence of 10 mg/L of Cd2+ versus 100.26 ± 3.82 mg/g VSS in the control after 10-day operation. More importantly, about 84.1%–94.8% of Cd2+ was found to bind to the extracellular proteins in microalgal-bacterial granules at the Cd2+ concentrations studied. In addition, Chlorococcum and Cyanobacteria in microalgal-bacteria granules were withered in the presence of 10 mg/L of Cd2+, suggesting uncoupled symbiosis between microalgae and bacteria induced by Cd2+. Consequently, this study showed that Cd2+ could negatively impact on the microbial structures and metabolisms of microalgal-bacterial granular sludge, leading to a compromised process performance in terms of organic and nitrogen removal.

Abstract:Contamination by heavy metals has become a serious environmental pollution issue today due to its potential threat to plant, wildlife, and human health. Photosynthesis, a process in which light energy is used to produce sugar and other organic compounds, is sensitive to heavy metals. In the present study, the response of photosynthetic process and carbon assimilation of Schima superba was investigated under cadmium (Cd) stress. Three Cd concentrations (0, 300, and 600 mg kg−1) were used designated as control (CK), low Cd (L1), and high Cd treatment (L2) of plants. Results showed that photosystem II (PSII) acceptor and donor side electron transport were more easily blocked in treatment compared to control, and L2 have more significant changes than L1. A substantial decrease of 820 nm reflection curve absorption was observed both in L1 and L2 treatments. Special energy fluxes showed significant difference between the control group and the treated group, which indicated that low concentration Cd stress can cause decrease in quantum yield of PSII in plants studied. Non-stomatal factors resulted in a decrease in net photosynthetic rate and a decrease in photosystem activity. Our results suggested that Cd can damage structure and function of the photosynthesis of S. superba young plants.
