Abstract:Sphingomonas sp.CJ-5 strain produces both chitinase and chitosanase into surrounding medium in response to chitin induction.The physiological function of CJ-5 fermented broth was also tested.The fermentor fuild showed antifungal activity against Fusarium oxysporum f.sp.lycopersici and growth-stimulation of Oryza sativa seedings to a certain extent.These results revealed a potential role for chitinase in biological control of plant pathogenic fungi and chitin oligosaccharide in stimulation growing of plant.
Peanut Rhizosphere Achromobacter xylosoxidans Inhibits Aspergillus flavus Development and Aflatoxin Synthesis by Inducing Apoptosis through Targeting the Cell Membrane

Abstract:Contamination of crop seeds and feed with Aspergillus flavus and its associated aflatoxins presents a significant threat to human and animal health due to their hepatotoxic and carcinogenic properties. To address this challenge, researchers have screened for potential biological control agents in peanut soil and pods. This study identified a promising candidate, a strain of the nonpigmented bacterium, Achromobacter xylosoxidans ZJS2-1, isolated from the peanut rhizosphere in Zhejiang Province, China, exhibiting notable antifungal and antiaflatoxin activities. Further investigations demonstrated that ZJS2-1 active substances (ZAS) effectively inhibited growth at a MIC of 60 μL/mL and nearly suppressed AFB1 production by 99%. Metabolomic analysis revealed that ZAS significantly affected metabolites involved in cell wall and membrane biosynthesis, leading to compromised cellular integrity and induced apoptosis in A. flavus through the release of cytochrome c. Notably, ZAS targeted SrbA, a key transcription factor involved in ergosterol biosynthesis and cell membrane integrity, highlighting its crucial role in ZJS2-1's biocontrol mechanism. Moreover, infection of crop seeds and plant wilt caused by A. flavus can be efficiently alleviated by ZAS. Additionally, ZJS2-1 and ZAS demonstrated significant inhibitory effects on various Aspergillus species, with inhibition rates ranging from 80 to 99%. These findings highlight the potential of ZJS2-1 as a biocontrol agent against Aspergillus species, offering a promising solution to enhance food safety and protect human health.
Identification of a Novel Antifungal Strain and Its Biocontrol Effect on Plant Pathogenic Fungi

Abstract:With the increase in the incidence of plant pathogenic fungi, the discovery and development of efficient, low-toxic and security antifungal compounds became even more important. A strain KZ001, antagonising plant pathogenic fungi, was isolated from a soil sample of Zhejiang Province, China. The strain was identified as Bacillus subtilis by sequencing 16S rRNA gene and BLAST searching GenBank. One of its metabolites from the microorganism was purified and identified as a macrolide compound with molecular weight of 402 Da by ESI/MS,(1)H-NMR and (13)C-NMR. The macrolide compound from Bacillus subtilis KZ001 showed strong inhibition against the fungal pathogens Aspergillus niger, Fusarium graminearu, Afternaria alternata, Exserohilum turcium, etc. As a result, the compound has the potential application value in biocontrol of phytopathogens in agriculture.

Abstract:The alteration of safety-secured yeast is a crucial step before scale applications. Based on previous studies, Pichia pastoris showed great potential and value to improve its biocontrol ability. The original sequences of antimicrobial peptide Ac-AMP2 and MiAMP1 were optimized according to the preference of Pichia pastoris and ligated into pPICZ alpha A plasmid which is emerged as a high-performance vector for transformation and expression. The results of RT-qPCR and Western blotting could imply that pPICZ alpha A/Ac-AMP2 and pPICZ alpha A/MiAMP1 were successfully overexpressed in Pichia pastoris GS115. The peptide concentration of GS115/Ac-AMP2 reached a maximum value of 210 mg L-1 at 60 h while GS115/MiAMP1 was 220 mg L-1 at 96 h. The biocontrol experiment indicated that the recombinant strain GS115/Ac-AMP2 and GS115/MiAMP1 could highly suppress the pathogen Penicillium expansum in vivo, which was respectively 42 % (GS115/Ac-AMP2) and 29.2 % (GS115/MiAMP1) of incidence disease lower than the sterile distilled water treatment. In the case of the experimental results considered, the modified GS115/Ac-AMP2 and GS115/MiAMP1 might be promising biological agent in postharvest applications.
Enhancing usability using automated security interface adaptation (ASIA)

Abstract:This paper investigates users’ understanding of security features and application and examines perceptions relating to usability. The study made use of an online survey consisting of five sections and recruited a total of 564 participants. Respondents were presented with a range of questions designed to measure their experience and knowledge of security. In addition, 2 scenarios were presented to respondents which examined their understanding of security warnings and potential threats, including email phishing and a potentially fraudulent attack through downloading an application. The survey results revealed that end-users are still experiencing significant difficulties with understanding and reacting to current state-of-the-art security applications, messages and potential threats. Furthermore, evidence suggests there is a corresponding need for a novel approach to improve perception and usability of information security.

Abstract:The Gram-negative rod-shaped bacterium Serratia marcescens is an opportunistic pathogen of many organisms, including insects. We report the identification and optimal in vitro chitinase production conditions of a novel chitinolytic S. marcescens strain TC-1 isolated from a naturally infected white grub (Anomala corpulenta) collected from a peanut field at Nanyang city, Henan province, China. Strain identification was conducted by morphological, physiological, biochemical and molecular analyses. The amplified 16S rRNA gene of TC-1 showed a similarity greater than 99% with multiple strains of S. marcescens. Based on Neighbor-joining phylogenetic tree analysis of bacterial 16S rRNA gene sequences, TC-1 formed a clade with S. marcescens, clearly separated from other Serratia spp. The strain TC-1 showed larvicidal activities against five insect species (A. corpulenta, Plutella xylostella, Spodoptera exigua, Helicoverpa armigera, Bombyx mori) and the nematode Caenorhabditis elegans, but not against S. litura. The operating parameters of chitinase production by TC-1 were optimized by response surface methodology using a three-factor, three-level Box-Behnken experimental design. The effects of three independent variables i.e. colloidal chitin concentration (7-13 g l-1), incubation time (24-72 h) and incubation temperature (24-32 °C) on chitinase production by TC-1 were investigated. A regression model was proposed to correlate the independent variables for an optimal chitinase activity predicted as 20.946 U ml-1, using a combination of colloidal chitin concentration, incubation time and incubation temperature of 9.06 g l-1, 63.83 h and 28.12 °C, respectively. The latter agreed well with a mean chitinase activity of 20.761 ± 0.102 U ml-1 measured in the culture supernatants of TC-1 grown under similar conditions with a colloidal chitin concentration, incubation time and incubation temperature of 9 g l-1, 64 h and 28 °C, respectively. Our study revealed the S. marcescens strain TC-1 with potential as a biocontrol agent of insect pests and nematodes and demonstrated the proposed regression model's potential to guide chitinase production by this strain.
Application of Non-Aflatoxigenic Aspergillus flavus for the Biological Control of Aflatoxin Contamination in China

Abstract:Biological control through the application of competitive non-aflatoxigenic Aspergillus flavus (A. flavus) to the soil during peanut growth is a practical method for controlling aflatoxin contamination. However, appropriate materials need to be found to reduce the cost of biocontrol products. In this study, a two-year experiment was conducted under field conditions in China, using a native non-aflatoxigenic strain to explore its effect. After three months of storage under high humidity, aflatoxin levels remained low in peanuts from fields treated with the biocontrol agent. Three types of substrates were tested with the biocontrol agent: rice grains, peanut meal (peanut meal fertilizer) and peanut coating. Compared to untreated fields, these formulations resulted in reductions of 78.23%, 67.54% and 38.48%, respectively. Furthermore, the ratios of non-aflatoxigenic A. flavus recovered in the soils at harvest in the treated fields were between 41.11% and 96.67% higher than that in untreated fields (25.00%), indicating that the rice inoculum was the most effective, followed by the peanut meal fertilizer and peanut coating. In 2019, the mean aflatoxin content of freshly harvested peanuts in untreated fields was 19.35 µg/kg higher than that in the fields treated with 7.5 kg/ha rice inoculum, which was 1.37 µg/kg. Moreover, no aflatoxin was detected in the two other plots treated with 10 and 15 kg/ha rice inoculum. This study showed that the native Chinese non-aflatoxigenic strain of A. flavus (18PAsp-zy1) had the potential to reduce aflatoxin contamination in peanuts. In addition, peanut meal can be used as an alternative substrate to replace traditional grains, reducing the cost of biocontrol products.
Another look at the nephrotic syndrome in adult Nigerians: pathological and immunological findings.

Abstract:One hundred and twenty adult Nigerians with the nephrotic syndrome were subjected to similar pathological investigations which included light microscopy and immunoperoxidase study of renal biopsy specimens, serum immunoglobulins and C3c levels. Histological findings showed a wide range of changes with the minimal change type of lesion and proliferative glomerulonephritis accounting each for 33.3 percent of cases studied. Focal changes accounted for 26.3 percent of cases. Glomerular immune deposits were present in 78.6 percent of cases showing non-minimal change glomerular lesions. Serum IgG and IgA were significantly depressed when compared with matched controls (P less than 0.05 and P less than 0.01 respectively) while IgM levels were significantly increased, (P less than 0.001). There was no significant difference between the mean values of serum C3c in the nephrotic group and matched control group. This study shows the high incidence of minimal change nephropathy in our adult population. Although some immunoglobulin (especially IgM) may be identified in occasional glomeruli and cause uncertainty as to the diagnosis, such minor deposition of IgM which is often non specific does not preclude a diagnosis of minimal change nephropathy.
Resistance Induction Against White Rot of Tuber Mustard Using Chitosans and Acetyl Salicylic Acid

Abstract:Introduction: White rot, caused by Sclerotinia sclerotiorum, has recently become a serious threat to tuber mustard cultivation in Hangzhou, China. The objective of this study was to evaluate the inhibitory effect of acetyl salicylic acid (ASA) and three different chitosans (A, B and C) against mustard white rot. The degree of N-deacetylation and the molecular weight of chitosans A, B and C were 85%-1129 kDa, 95%-521 kDa and 75%-607 kDa, respectively. Materials and Methods: The inhibitory effect of chitosans with different concentrations against the mycelia growth and sclerotia formation of 3 isolates of the pathogen was determined in vitro. In addition, the efficacy of chitosans and ASA against mustard white rot was assessed during in vivo tests. After protein extraction, effects of chitosans and ASA on resistance related enzymes including chitinase, β-1,3-glucanase, phenylalanine ammonia lyase, polyphenol oxidase (PPO) and peroxidase (POD) were evaluated. Results: The chitosans reduced the mycelia growth and sclerotia formation of the pathogen. The chitosans showed significant antifungal effect against the disease in vivo. The chitosans and ASA markedly reduced the severity of the disease over time. Moreover, the chitosans and ASA markedly enhanced the level of most of the resistant related enzymes after 3 and 6 days. The chitosan B was found to have the best effect against tested pathogen isolates. Conclusions: The chitosan with the lowest molecular weight was found to be more effective against the disease. In addition, chitosans and ASA were able to significantly increase resistance-related enzymes over time indicating that they can be considered as resistant inducers against mustard white rot.
Characterisation and antifungal activity of extracellular chitinase from a biocontrol fungus, Trichoderma asperellum PQ34

Abstract:Trichoderma species were known as biological control agents against phytopathogenic fungi because they produce a variety of chitinases. Chitinases are hydrolytic enzymes that break down glycosidic bonds in chitin, a major component of the cell walls of fungi. The present study shows that extracellular chitinase activity reached a maximum value of approximately 22 U/mL after 96 h of T. asperellum PQ34 strain culture. The optimal temperature and pH of enzyme are 40°C and 7, respectively, whereas the thermal and pH stability range from 25°C to 50°C and 4 to 10, respectively. Chitinase at 60 U/mL inhibited nearly completely in vitro growth of Colletotrichum sp. (about 95%) and Sclerotium rolfsii (about 97%). In peanut plants, 20 U/mL of chitinase significantly reduced the incidence of S. rolfsii infection compared to controls. The fungal infection incidence of seeds before germination and 30 days after germination was only 2.22% and 2.38%, while the control was 13.33% and 17.95%. Besides, chitinase from T. asperellum PQ34 can also prevent anthracnose that is caused by Colletotrichum sp. on both mango and chilli fruits up to 72 h after enzyme pre-treatment at 40 U/mL. In mango and chilli fruits infected with anthracnose, 40 U/mL dose of chitinase inhibited the growth of fungi after 96 h of treatment, the diameter of lesion was only 0.88 cm for mango and 1.45 cm for chilli, while the control was 1.67 cm and 2.85 cm, respectively.

Abstract:Aspergillus flavus is a ubiquitous facultative pathogen that routinely infects important crops leading to formation of aflatoxins during crop development and after harvest. Corn and peanuts in warm and/or drought-prone regions are highly susceptible to aflatoxin contamination. Controlling aflatoxin using atoxigenic A. flavus is a widely adopted strategy. However, no A. flavus genotypes are currently approved for use in China. The current study aimed to select atoxigenic A. flavus endemic to Guangxi Zhuang Autonomous Region with potential as active ingredients of aflatoxin biocontrol products. A total of 204 A. flavus isolates from corn, peanuts, and field soil were evaluated for ability to produce the targeted mycotoxins. Overall, 57.3% could not produce aflatoxins while 17.15% were incapable of producing both aflatoxins and CPA. Atoxigenic germplasm endemic to Guangxi was highly diverse, yielding 8 different gene deletion patterns in the aflatoxin and CPA biosynthesis gene clusters ranging from no deletion to deletion of both clusters. Inoculation of corn and peanuts with both an aflatoxin producer and selected atoxigenic genotypes showed significant reduction (74 to 99%) in aflatoxin B1 (AFB1) formation compared with inoculation with the aflatoxin producer alone. Atoxigenic genotypes also efficiently degraded AFB1 (61%). Furthermore, atoxigenic isolates were also highly efficient at reducing aflatoxin concentrations even when present at lower concentrations than aflatoxin producers. The use of multiple atoxigenics was not always as effective as the use of a single atoxigenic. Effective atoxigenic genotypes of A. flavus with known mechanisms of atoxigenicity are demonstrated to be endemic to Southern China. These A. flavus may be utilized as active ingredients of biocontrol products without concern for detrimental impacts that may result from introduction of exotic fungi. Field efficacy trials in the agroecosystems of Southern China are needed to determine the extent to which such products may allow the production of safer food and feed.

Synergistic effect of acetic acid and chitosan against Aspergillus flavus

Abstract:The mycotoxin-producing fungi contamination of foodstuffs and agricultural commodities is a serious problem. In this study, the efficacy of acetic acid (AcA), chitosan (CS), and CS with extra AcA (CS/extra AcA) against Aspergillus flavus (A. flavus) in vitro and in vivo were investigated. Results showed that CS/extra AcA strongly inhibited the mycelial growth and aflatoxins production of A. flavus at lower concentrations than CS (MIC > 16.0 mg/mL) or AcA (MIC: 0.31 %). Significant inhibition was achieved in the presence of 4.0/0.12 and 8.0/0.08 CS (mg/mL)/extra AcA (%). CS/extra AcA exhibited remarkable antifungal activities based on the results of spore germination inhibition, abnormalities of spore morphology, disruption of cell membrane, mitochondrial dysfunction, and induction of apoptosis of hyphae. Moreover, pathogenicity test in peanut kernels showed that 8.0/0.16 CS (mg/mL)/extra AcA (%) could effectively inhibit the fungal infection and aflatoxin B1 production. The efficacy of CS/extra AcA is possibly due to the synergy of the antifungal property of CS, and undissociated AcA. The findings referred that CS/extra AcA is safe, efficient, and economical, and it could be used as a promising way to control A. flavus contamination in agro-foods preservation.

Abstract:Abstract Background Peanut stem rot is a serious plant disease that causes great economic losses. At present, there are no effective measures to prevent or control the occurrence of this plant disease. Biological control is one of the most promising plant disease control measures. In this study, Pseudomonas chlororaphis subsp. aurantiaca strain zm-1, a bacterial strain with potential biocontrol properties isolated by our team from the rhizosphere soil of Anemarrhena asphodeloides , was studied to control this plant disease. Methods We prepared extracts of Pseudomonas chloroaphis zm-1 extracellular antibacterial compounds (PECEs), determined their antifungal activities by confrontation assay, and identified their components by UPLC-MS/MS. The gene knockout strains were constructed by homologous recombination, and the biocontrol efficacy of P. chlororaphis zm-1 and its mutant strains were evaluated by pot experiments under greenhouse conditions and plot experiments, respectively. Results P. chlororaphis zm-1 could produce extracellular antifungal substances and inhibit the growth of Sclerotium rolfsii , the main pathogenic fungus causing peanut stem rot. The components of PECEs identified by UPLC-MS/MS showed that three kinds of phenazine compounds, i.e., 1-hydroxyphenazine, phenazine-1-carboxylic acid (PCA), and the core phenazine, were the principal components. In particular, 1-hydroxyphenazine produced by P. chlororaphis zm-1 showed antifungal activities against S. rolfsii , but 2-hydroxyphenazine did not. This is quite different with the previously reported. The extracellular compounds of two mutant strains, Δ phzH and Δ phzE , was analysed and showed that Δ phzE did not produce any phenazine compounds, and Δ phzH no longer produced 1-hydroxyphenazine but could still produce PCA and phenazine. Furthermore, the antagonistic ability of Δ phzH declined, and that of Δ phzE was almost completely abolished. According to the results of pot experiments under greenhouse conditions, the biocontrol efficacy of Δ phzH dramatically declined to 47.21% compared with that of wild-type P. chlororaphis zm-1 (75.63%). Moreover, Δ phzE almost completely lost its ability to inhibit S. rolfsii (its biocontrol efficacy was reduced to 6.19%). The results of the larger plot experiments were also consistent with these results. Conclusions P. chlororaphis zm-1 has the potential to prevent and control peanut stem rot disease. Phenazines produced and secreted by P. chlororaphis zm-1 play a key role in the control of peanut stem rot caused by S. rolfsii . These findings provide a new idea for the effective prevention and treatment of peanut stem rot.

Biocontrol Effect Against Tomato Fusarium Wilt of Pseudomonas Aeruginosa Strain FJAT-346

Abstract:A study on the biocontrol efficacy and mechanism of Pseudomonas aeruginosa strain FJAT-346 was performed for evaluating the application significance of the strain for the biological control of plant diseases.The results showed that FJAT-346 exhibited strong antagonism against F.oxysporum f.sp.cubense,Fusarium oxysporum f.sp.radicis lycopersici,F.oxysporum f.sp.niveum,F.oxysporum f.sp.nelonis,Botrytis cinerea and Sclerotinia sclerotiorura in vitro test.It exhibited 81.57% of control effectiveness in pot experiment.The strain strongly inhibited mycelium growth of F.oxysporum f.sp.radicis lycopersici,but no inhibitory effect was detected on spore germination.The results of observation under scanning electron microscope showed that the terminal mycelia were distorted and swollen,and the cell wall was ruptured after treated with FJAT-346 filtrate culture.Antagonistic substance of FJAT-346 was obtained by 70% ammonium sulphate precipitation.The results implied that the antagonistic substance was a kind of proteins.FJAT-346 also promoted the growth of tomato significantly in the pots.
The inhibitory effect of Bacillus megaterium on aflatoxin and cyclopiazonic acid biosynthetic pathway gene expression in Aspergillus flavus

Abstract:Aspergillus flavus is one of the major moulds that colonize peanut in the field and during storage. The impact to human and animal health, and to the economy in agriculture and commerce, is significant since this mold produces the most potent known natural toxins, aflatoxins, which are carcinogenic, mutagenic, immunosuppressive, and teratogenic. A strain of marine Bacillus megaterium isolated from the Yellow Sea of East China was evaluated for its effect in inhibiting aflatoxin formation in A. flavus through down-regulating aflatoxin pathway gene expression as demonstrated by gene chip analysis. Aflatoxin accumulation in potato dextrose broth liquid medium and liquid minimal medium was almost totally (more than 98 %) inhibited by co-cultivation with B. megaterium. Growth was also reduced. Using expression studies, we identified the fungal genes down-regulated by co-cultivation with B. megaterium across the entire fungal genome and specifically within the aflatoxin pathway gene cluster (aflF, aflT, aflS, aflJ, aflL, aflX). Modulating the expression of these genes could be used for controlling aflatoxin contamination in crops such as corn, cotton, and peanut. Importantly, the expression of the regulatory gene aflS was significantly down-regulated during co-cultivation. We present a model showing a hypothesis of the regulatory mechanism of aflatoxin production suppression by AflS and AflR through B. megaterium co-cultivation.

Abstract:The climate-change-coupled fungal burden in crop management and the need to reduce chemical pesticide usage highlight the importance of finding sustainable ways to control Aspergillus flavus. This study examines the effectiveness of 50 Pseudomonas isolates obtained from corn rhizospheres against A. flavus in both solid and liquid co-cultures. The presence and quantity of aflatoxin B1 (AFB1) and AFB1-related compounds were determined using high-performance liquid chromatography–high resolution mass spectrometry analysis. Various enzymatic- or non-enzymatic mechanisms are proposed to interpret the decrease in AFB1 production, accompanied by the accumulation of biosynthetic intermediates (11-hydroxy-O-methylsterigmatocystin, aspertoxin, 11-hydroxyaspertoxin) or degradation products (the compounds C16H10O6, C16H14O5, C18H16O7, and C19H16O8). Our finding implies the upregulation or enhanced activity of fungal oxidoreductases and laccases in response to bacterial bioactive compound(s). Furthermore, non-enzymatic reactions resulted in the formation of additional degradation products due to acid accumulation in the fermented broth. Three isolates completely inhibited AFB1 or any AFB1-related compounds without significantly affecting fungal growth. These bacterial isolates supposedly block the entire pathway for AFB1 production in the fungus during interaction. Apart from identifying effective Pseudomonas isolates as potential biocontrol agents, this work lays the foundation for exploring new bacterial bioactive compounds.

Bacillus siamensis 3BS12-4 Extracellular Compounds as a Potential Biological Control Agent against Aspergillus flavus

Abstract:Aspergillus flavus, the primary mold that causes food spoilage, poses significant health and economic problems worldwide. Eliminating A. flavus growth is essential to ensure the safety of agricultural products, and extracellular compounds (ECCs) produced by Bacillus spp. have been demonstrated to inhibit the growth of this pathogen. In this study, we aimed to identify microorganisms efficient at inhibiting A. flavus growth and degrading aflatoxin B1. We isolated microorganisms from soil samples using a culture medium containing coumarin (CM medium) as the sole carbon source. Of the 498 isolates grown on CM medium, only 132 bacterial strains were capable of inhibiting A. flavus growth. Isolate 3BS12-4, identified as Bacillus siamensis, exhibited the highest antifungal activity with an inhibition ratio of 43.10%, and was therefore selected for further studies. The inhibition of A. flavus by isolate 3BS12-4 was predominantly attributed to ECCs, with a minimum inhibitory concentration and minimum fungicidal concentration of 0.512 g/ml. SEM analysis revealed that the ECCs disrupted the mycelium of A. flavus. The hydrolytic enzyme activity of the ECCs was assessed by protease, β-1,3-glucanase, and chitinase activity. Our results demonstrate a remarkable 96.11% aflatoxin B1 degradation mediated by ECCs produced by isolate 3BS12-4. Furthermore, treatment with these compounds resulted in a significant 97.93% inhibition of A. flavus growth on peanut seeds. These findings collectively present B. siamensis 3BS12-4 as a promising tool for developing environmentally friendly products to manage aflatoxin-producing fungi and contribute to the enhancement of agricultural product safety and food security.

Abstract:Aspergillus flavus and the produced aflatoxins cause great hazards to food security and human health across all countries. The control of A. flavus and aflatoxins in grains during storage is of great significance to humans. In the current study, bacteria strain YM6 isolated from sea sediment was demonstrated effective in controlling A. flavus by the production of anti-fungal volatiles. According to morphological characteristics and phylogenetic analysis, strain YM6 was identified as Pseudomonas stutzeri. YM6 can produce abundant volatile compounds which could inhibit mycelial growth and conidial germination of A. flavus. Moreover, it greatly prevented fungal infection and aflatoxin production on maize and peanuts during storage. The inhibition rate was 100%. Scanning electron microscopy further supported that the volatiles could destroy the cell structure of A. flavus and prevent conidia germination on the grain surface. Gas chromatography/mass spectrometry revealed that dimethyl trisulfide (DMTS) with a relative abundance of 13% is the most abundant fraction in the volatiles from strain YM6. The minimal inhibitory concentration of DMTS to A. flavus conidia is 200 μL/L (compound volume/airspace volume). Thus, we concluded that Pseudomonas stutzeri YM6 and the produced DMTS showed great inhibition to A. flavus, which could be considered as effective biocontrol agents in further application.

Abstract:A new alkalophilic low-molecular-mass chitinase of 14 kD from the potent biocontrol agent Bacillus subtilis JN032305 was partially purified and enzymology of the chitinase was studied. The enzyme showed optimal pH of 9.0 and temperature of 50°C. The enzyme was found stable during the 60-min incubation at 50 °C. The chitinase was inhibited by group specific agents like IAA, DAN, TLCK, and SDS and metal ions Mg(2+), Ca(2+), Fe(2+), Mn(2+), Ba(2+), and Hg(2+), whereas Zn(2+) did not show significant inhibitory effect against the chitinase. PMSF partially inhibited the enzyme. Substrates specificity tests indicated that the enzyme showed 75% of relative activity on glycol chitin, 58% on carboxymethylcellulose (CMC), 33% on chitin flakes, and 166% laminarin compared to that on colloidal chitin. The enzyme also hydrolyzed 4-methylumbelliferyl-N-acetyl-D-glucosaminide, indicating its chitobiase activity. The chitinase of this study has broad specificity, which could hydrolyze not only the glycosidic bond in GlcNAc-GlcNAc but also that of related carbohydrates with glycosidic linkages. The partially purified chitinase not only showed antifungal activity against Rhizoctonia solani and Colletotrichum gloeosporioides, two potent phytopathogens of chilli, but also increased the germination of chilli seeds when infected with the two potent phytopathogenic fungi.
