Abstract:This study investigated the effect of co-substrate amendments on EDB biodegradation under aerobic conditions. Microcosms were established using contaminated soil and groundwater samples and maintained under in situ conditions to determine EDB degradation rates, and the diversity and abundance of EDB degrading indigenous bacteria. After 100days of incubation, between 25% and 56% of the initial EDB was degraded in the microcosms, with added jet fuel providing highest degradation rates (2.97±0.49yr(-1)). In all microcosms, the quantity of dehalogenase genes did not change significantly, while the number of BTEX monooxygenase and phenol hydroxylase genes increased with jet fuel amendments. These results indicate that EDB was not degraded by prior dehalogenation, but rather by cometabolism with adapted indigenous microorganisms. This is also reflected in the history of the plume, which originated from an aviation gasoline pipeline leak. This study suggests that biostimulation of EDB is possible at aerobic groundwater sites.
Electrical Stimulation Accelerated Phenanthrene Biodegradation Coupling with Nitrate Reduction in Groundwater

Abstract:Phenanthrene (PHE) and nitrate co-contamination widely exists in the groundwater. Anaerobic bioremediation of the two suffered from the slow removal rate and limited electron transfer within microbes. Herein electrical stimulation (ES) was proved to stimulate the simultaneous removal of PHE and nitrate anaerobically. High removal of PHE and nitrate (90.94 % and 100 %, respectively) was obtained within 24 h in the ES-bioreactor, two times higher than the control anaerobic bioreactor (32.16 %). ES-bioreactor exhibited a good PHE removal performance within initial PHE ranging from 5 to 25 mg/L. In ES-bioreactor, extracellular polymeric substances increased PHE bioavailability and acted as protective capsules. Nicotinamide adenine dinucleotide (NADH) and cytochrome C (cyt-C) facilitated the extracellular and intracellular electron exchange during PHE biotransformation. ES significantly selected Acetobacterium, PHE-degraders (f_Bacteroidetesvadin HA17, Limno-bacter and Stenotrophomonas) and electrogens (Clostrium_sensu_stricto_13) to participate in PHE decomposition. Acetobacterium decomposed complex organics to acetate, proving accessible substrates for electrogens to main-tain an efficient electron flow. Enzymes regulating bacterial chemotaxis, flagellar assembly and carbohydrate metabolism were up-regulated with ES, suggesting the indispensable role of such enzymatic processes in ES-bioreactor. Overall, this work demonstrates ES can promote anaerobic bioremediation of PHE and nitrate in groundwater and, thus, opens up a new bioelectrochemistry application scenario.
Metagenomic insights into phenanthrene biodegradation in electrical field-governed biofilms for groundwater bioremediation

Abstract:Electrical fields (EFs)-assisted in-situ bioremediation of petroleum-contaminated groundwater, such as polycyclic aromatic hydrocarbons, has drawn increasing attention. However, the long-term stability, the EFs influence, and metabolic pathways are still poorly understood, hindering the further development of robust technology design. Herein, a series of EFs was applied to the phenanthrene-contaminated groundwater, and the corresponding system performance was investigated. The highest removal capacity of phenanthrene (phe) (7.63 g/(m 3 ·d)) was achieved with EF_0.8 V biofilm at a hydrolytic retention time of 0.5 d. All the biofilms with four EFs exhibited a high removal efficiency of phe over 80% during a 100-d continuous-flow operation. Intermediates analysis revealed the main pathways of phe degradation: phthalate and salicylate via hydroxylation, methylation , carboxylation, and ring cleavage steps. Synergistic effects between phe-degraders ( Dechloromonas ), fermentative bacteria ( Delftia ), and electroactive microorganisms ( Geobacter ) were the main contributors to the complete phe mineralization. Genes encoding various proteins of methyl-accepting ( mcp ), response regulator ( cheABDRY ), and type IV pilus ( pilABCMQV ) were dominant, revealing the importance of cell motility and extracellular electron transfer . Metagenomics analysis unveiled phe-degrading genes, including ring reduction enzymes ( bamBCDE ), carboxylase of aromatics ( ubiD ), and methyltransferase protein ( ubiE , pcm ). These findings offered a molecular understanding of refractory organics' decompositions in EFs-governed biotechnology.

Radiolytic Degradation of BDE-209 in Rice-Vegetable Rotation Soils Induced by Electron Beam Irradiation.

Abstract:Deca-brominated diphenyl ether (BDE-209) is shown to be persistent in soil and it's urgent to find an effective method to remove BDE-209 from contaminated soil. In this study, the investigation of BDE-209 degradation in three different rice-vegetable rotation soils induced by electron beam (E-beam) irradiation under flooded and non-flooded conditions was conducted. The reductive efficiency of 14C-BDE209 reached the highest level at 50 kGy and the values in flooded soils of rice-eggplant rotation soil (RES), rice-peanut soil (RPS), and rice-chili pepper soil (RCS) were 93.5%, 87.2%, and 73.8%, respectively. The reductive efficiencies in non-flooded soils of RES, RPS, and RCS were 73.4%, 81.0%, and 78%, respectively. The D0.5 values (dose required for reducing 50% BDE-209) of BDE-209 in non-flooded soils were lower than those in flooded soils, suggesting greater degradation efficiency of BDE-209 in non-flooded soils than in flooded soils. The BDE-209 was degraded into higher-brominated PBDEs and lower-brominated PBDEs by E-beam irradiation. The results demonstrate that BDE-209 in the soil can be degraded by E-beam irradiation, non-flooded condition is better than flooded condition for the removal of BDE-209, and the main degradation mechanism of BDE-209 by E-beam irradiation is debromination. This study provides a rapid and effective method for degrading BDE-209 that is persistent in soils, and has important implications for the remediation of soil contaminated by PBDEs in and around E-waste dismantling areas.
Transportation and Degradation of Decabrominated Diphenyl Ether in Sequential Anoxic and Oxic Crop Rotation.

Abstract:This work evaluated the debromination and uptake of C-14-labeled BDE-209 in rice cultivars grown in anoxic soil for 120 days (d) followed by cultivation of vegetables (peanut, eggplant and pepper) in oxic soil (120 d). Degradation of BDE-209 to lower polybrominated diphenyl ethers (PBDEs) occurred in cultivated soils, and more metabolites were released in oxic soil than in anoxic soil. The crop rotation from anoxic to oxic greatly enhanced the dissipation of BDE-209 in the soil (P < 0.05), in which the dissipation in anoxic soil planted with Huanghuazhan (HHZ, indica) and Yudao 1 (YD1, indica) were 6.8% and 2.4%, respectively, while in oxic soil with peanut and pepper were increased to 25.8% and 21.7%, respectively. The crop rotation also enhanced the degradation of BDE-209 in the soil, the recovered BDE-209 in soil after 120 d anoxic incubation with YD1 was 81.1%, but it decreased to 47.8% and 45.8% after another 120 d oxic incubation. Bioconcentration factors were between 0.23 and 0.36 for rice, eggplant and pepper but reached to 0.5 in peanut, which contains more lipids in the edible portion than the other test crops. The estimated daily intake for vegetables was 0.01-0.07 mu g BDE-209-equivalent kg(-1) bw day(-1), which is at least two orders of magnitude below the maximum acceptable oral dose (7 mu g kg(-1) bw day(-1)). Our work confirms that crop rotation from rice to vegetable enhanced the dissipation and debromination of BDE-209 in the soil, and indicate that sequential anoxic-oxic rotation practice is considered to be effective in remediation of environmental pollutants. Abstract:A remediation approach which uses pump and treatment (PAT) to enhance the biodegradation of organic contaminants by increasing dispersive mixing between plumes and groundwater was evaluated for a phenol-contaminated aquifer, using a reactive transport model which simulates kinetic reactions between an electron donor (ED) in the plume and electron acceptor (EA) in the groundwater. The influence of system design and operation was examined in six modelling scenarios. Injection or extraction of groundwater increases biodegradation above no action and the location, pumping rate, and distance between well(s) are important variables which influence biodegradation. An increase in pumping rate, distance of the wells from the plume centreline, and changing the flow direction increase dispersive mixing between the plume and groundwater. This increases plume spreading and the plume fringe interface, providing a greater flux of dissolved EAs for biodegradation. In general, injection of groundwater containing natural EAs enhances biodegradation more than extraction. The enhancement of biodegradation is sensitive to the relative fluxes of ED and EA, as controlled by the arrangement of the wells. In the best performing scenario, biodegradation was enhanced by 128%, compared with no action.

Abstract:Polybrominated diphenyl ethers (PBDEs), one kind of persistent organic pollutants, were widely detected in coastal wetlands. Microbial reductive debromination is one of the most important attenuation processes for PBDEs in anaerobic environment, whereas the underlying reaction mechanisms remain elusive. Dual-element stable isotope analysis was recently recognized to distinguish different reaction mechanism for degradation of organic pollutants. In this study, the dual carbon-bromine isotope effects associated with the anaerobic microbial degradation were first investigated to characterize the reaction mechanisms for BDE-47 and BDE-153. Presence of lower brominated congeners indicated stepwise debromination as the main degradation pathway, with the preferential removal of bromine in para position > meta/ortho position. The pronounced isotope fractionation was observed for both carbon and bromine, with similar carbon (εC) and bromine isotope enrichment factor (εBr) between BDE-47 (εC = -5.98‰, εBr = -2.44‰) and BDE-153 (εC = -5.57‰, εBr = -2.06‰) during the microbial degradation. Compared to εC and εBr, the correlation of carbon and isotope effects (ΛC/Br = Δδ81Br/Δδ13C) was almost the same between BDE-47 (0.436) and BDE-153 (0.435), indicating the similar reaction mechanism. The calculated carbon and bromine apparent kinetic isotope effects (AKIEC and AKIEBr) were 1.0773 and 1.0098 for BDE-47 and 1.0716 and 1.0125 for BDE-153, within range reported for degradation of halogenated compounds following nucleophilic substitution. Combination analysis of degradation products, ΛC/Br and AKIE, all the results pointed to that the anaerobic reductive debromination of BDE-47 and BDE-153 followed the nucleophilic aromatic substitution, with the addition of cofactor to the benzene ring concomitant with dissociation of carbon-bromine bond via the inner-sphere electron transfer, and the cleavage of C-Br bond was the rate-determining step. This study contributed to the development of dual carbon-bromine isotope analysis as a robust approach to probe the fate of PBDEs in contaminated sites.

Abstract:Abstract Understanding anaerobic biodegradation of ether oxygenates beyond MTBE in groundwater is important, given that it is replaced by ETBE as a gasoline additive in several regions. The lack of studies demonstrating anaerobic biodegradation of ETBE, and its product TBA, reflects the relative resistance of ethers and alcohols with a tertiary carbon atom to enzymatic attack under anoxic conditions. Anaerobic ETBE- or TBA-degrading microorganisms have not been characterized. Only one field study suggested anaerobic ETBE biodegradation. Anaerobic (co)metabolism of ETBE or TBA was reported in anoxic microcosms, indicating their biodegradation potential in anoxic groundwater systems. Non-isotopic methods, such as the detection of contaminant loss, metabolites, or ETBE- and TBA-degrading bacteria are not sufficiently sensitive to track anaerobic biodegradation in situ . Compound- and position-specific stable isotope analysis provides a means to study MTBE biodegradation, but isotopic fractionation of ETBE has only been studied with a few aerobic bacteria (εC −0.7 to −1.7‰, εH −11 to −73‰) and at one anoxic field site (δ 2 H-ETBE +14‰). Similarly, stable carbon isotope enrichment (δ 13 C-TBA +6.5‰) indicated TBA biodegradation at an anoxic field site. CSIA and PSIA are promising methods to detect anaerobic ETBE and TBA biodegradation but need to be investigated further to assess their full potential at field scale. Graphical abstract

Enhanced bioremediation of RDX and Co-Contaminants perchlorate and nitrate using an anaerobic dehalogenating consortium in a fractured rock aquifer

Abstract:The potential neurotoxic and carcinogenic effects of the explosives compound RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) on human health requires groundwater remediation strategies to meet low cleanup goals. Bioremediation of RDX is feasible through biostimulation of native microbes with an organic carbon donor but may be less efficient, or not occur at all, in the presence of the common co-contaminants perchlorate and nitrate. Laboratory tests compared biostimulation with bioaugmentation to achieve anaerobic degradation of RDX, perchlorate, and nitrate; a field pilot test was then conducted in a fractured rock aquifer with the selected bioaugmentation approach. Insignificant reduction of RDX, perchlorate, or nitrate was observed by the native microbes in microcosms, with or without biostimulation by addition of lactate. Tests of the RDX-degrading ability of the microbial consortium WBC-2, originally developed for dehalogenation of chlorinated volatile organic compounds, showed first-order biodegradation rate constants ranging from 0.57 to 0.90 per day (half-lives 1.2 to 0.80 days). WBC-2 sustained degradation without daughter product accumulation when repeatedly amended with RDX and lactate for a year. In microcosms with groundwater containing perchlorate and nitrate, RDX degradation began without delay when bioaugmented with 10% WBC-2. Slower RDX degradation occurred with 3% or 5% WBC-2 amendment, indicating a direct relation with cell density. Transient RDX daughter compounds included methylene dinitramine, MNX, and DNX. With WBC-2 amendment, nitrate concentrations immediately decreased to near or below detection, and perchlorate degradation occurred with half-lives of 25-34 days. Single-well injection tests with WBC-2 and lactate showed that the onset of RDX degradation coincided with the onset of sulfide production, which was affected by the initial perchlorate concentration. Biodegradation rates in the pilot injection tests agreed well with those measured in the microcosms. These results support bioaugmentation with an anaerobic culture as a remedial strategy for sites contaminated with RDX, nitrate, and perchlorate.
Vertical Distribution of Archaeal Communities Associated with Anaerobic Degradation of Pentabromodiphenyl Ether (BDE-99) in River-Based Groundwater Recharge with Reclaimed Water.

Abstract:When groundwater is recharged with reclaimed water, the presence of trace amounts of biorefractory pentabromodiphenyl ether (PBDE, specifically BDE-99) might cause potential groundwater pollution. A laboratory-scale column was designed to investigate the distribution of the community of archaea in this scenario and the associated anaerobic degradation of BDE-99. The concentration of BDE-99 decreased significantly as soil depth increased, and fluorescence in situ hybridization (FISH) analysis suggested that archaea exerted significant effects on the biodegradation of PBDE. Through 454 pyrosequencing of 16s rRNA genes, we found that the distribution and structure of the archaeal community associated with anaerobic degradation of BDE-99 in the river-based aquifer media changed significantly between different soil depths. The primary debrominated metabolites varied with changes in the vertically distributed archaeal community. The archaea in the surface layer were dominated by Methanomethylovorans, and the middle layer was mainly composed of Nitrososphaera. Nitrosopumilus and Nitrososphaera were equally abundant in the bottom layer. In addition, Methanomethylovorans abundance depended on the depth of soil, and the relative abundance of Nitrosopumilus increased with increasing depth, which was associated with the oxidation-reduction potential and the content of intermediate metabolites. We propose that Nitrososphaera and Nitrosopumilus might be the key archaeal taxa mediating the biodegradation of BDE-99.
Dynamic changes in microbial community structure and function in phenol-degrading microcosms inoculated with cells from a contaminated aquifer

Abstract:Contamination of aquifers by organic pollutants threatens groundwater supplies and the environment. In situ biodegradation of organic pollutants by microbial communities is important for the remediation of contaminated sites, but our understanding of the relationship between microbial development and pollutant biodegradation is poor. A particular challenge is understanding the in situ status of microorganisms attached to solid surfaces, but not accessible via conventional sampling of groundwater. We have developed novel flow-through microcosms and examined dynamic changes in microbial community structure and function in a phenol-degrading system. Inoculation of these microcosms with a complex microbial community from a plume in a phenol-contaminated aquifer led to the initial establishment of a population dominated by a few species, most attached to the solid substratum. Initially, phenol biodegradation was incomplete, but as the microbial community structure became more complex, phenol biodegradation was more extensive and complete. These results were replicated between independent microcosms, indicating a deterministic succession of species. This work demonstrates the importance of examining community dynamics when assessing the potential for microbial biodegradation of organic pollutants. It provides a novel system in which such measurements can be made readily and reproducibly to study the temporal development and spatial succession of microbial communities during biodegradation of organic pollutants at interfaces within such environments.
Identification of Aerobic ETBE‐Degrading Microorganisms in Groundwater Using Stable Isotope Probing

Abstract:A limited number of microorganisms have been identified with the capability to degrade ethyl tert‐butyl ether (ETBE) in the environment. Knowledge of the identity and distribution of ETBE‐degrading microorganisms is important for the implementation of management measures such as natural attenuation and bioremediation at ETBE‐release sites. In this study, DNA‐stable isotope probing (SIP) was used to identify microorganisms able to aerobically degrade 13C‐labeled ETBE in laboratory microcosms constructed with groundwater and aquifer material from an ETBE‐release site. Microorganisms in the Class γ‐proteobacteria, Order β‐proteobacteriales, Family Burkholderiaceae, and classified as Methylibium and Leptothrix, respectively, were identified as primary ETBE degraders. Comparisons with ETBE‐responsive microorganisms (those which increased in abundance after the addition of ETBE), identified by high‐throughput sequencing of microcosms established from the same site, showed that only a small proportion of the ETBE‐responsive organisms were primary degraders as determined by SIP. ETBE degraders were taxonomically related to microorganisms able to degrade other gasoline components, but not ETBE, implying that this functionality results from acquisition of the eth gene cluster by these organisms. These ETBE degraders could also be identified at ETBE‐release sites, but at low relative abundance and generally only in those locations from which the microcosms had been established. Therefore, we recommend that molecular investigations of ETBE‐contaminated sites focus on functional genes (i.e., the eth gene cluster) rather than specific taxa.

Efficient and Complete Detoxification of Polybrominated Diphenyl Ethers in Sediments Achieved by Bioaugmentation with Dehalococcoides and Microbial Ecological Insights

Abstract:Polybrominated diphenyl ethers (PBDEs) are prevalent environmental pollutants, but bioremediation of PBDEs remains to be reported. Here we report accelerated remediation of a penta-BDE mixture in sediments by bioaugmentation with Dehalococcoides mccartyi strains CG1 and TZ50. Bioaugmentation with different amounts of each Dehalococcoides strain enhanced debromination of penta-BDEs compared with the controls. The sediment microcosm spiked with 6.8 × 106 cells/mL strain CG1 showed the highest penta-BDEs removal (89.9 ± 7.3%) to diphenyl ether within 60 days. Interestingly, co-contaminant tetrachloroethene (PCE) improved bioaugmentation performance, resulting in faster and more extensive penta-BDEs debromination using less bioinoculants, which was also completely dechlorinated to ethene by introducing D. mccartyi strain 11a. The better bioaugmentation performance in sediments with PCE could be attributed to the boosted growth of the augmented Dehalococcoides and capability of the PCE-induced reductive dehalogenases to debrominate penta-BDEs. Finally, ecological analyses showed that bioaugmentation resulted in more deterministic microbial communities, where the augmented Dehalococcoides established linkages with indigenous microorganisms but without causing obvious alterations of the overall community diversity and structure. Collectively, this study demonstrates that bioaugmentation with Dehalococcoides is a feasible strategy to completely remove PBDEs in sediments.

Degradation of decabromodiphenyl ether (BDE-209) in microcosms mimicking sediment environment subjected to comparative bioremediation strategies

Abstract:The aim of this study was to examine bioremediation strategies for BDE-209 contaminated sediments. Sediment microcosms were established to observe anaerobic debromination of BDE-209 under conditions representing three bioremediation strategies: biostimulation, bioaugmentation and natural attenuation. To simulate biostimulation, a defined mineral medium containing both a carbon source (sodium formate) and electron donor (ethanol) was added into sediments. Bioaugmentation was established by enrichment of the sediments using a culture of Dehalobium chlorocoercia strain DF-1, previously shown to dechlorinate polychlorinated biphenyls, to sediments. No amendments were made to the third set in order to represent natural attenuation. The biostimulation, bioaugmentation and natural attenuation strategies resulted in 55.3%, 40.2% and 30.9% reductions in BDE-209, respectively, after 180 days. Nona- through tri-BDEs were observed as products, with 17 PBDE congeners detected in 25 different proposed debromination pathways. At the end of the 180 day incubation period, the products for bioaugmentation, biostimulation and natural attenuation were tri-BDEs, tetra-BDEs and penta-BDEs, respectively. The proposed pathways revealed that meta- and ortho-Br removal were favored in sediments, and that debromination regiospecificity varied with each bioremediation strategy applied. Lastly, pseudo-first-order rate constants for BDE-209 reduction were calculated as 0.0049 d-1, 0.0028 d-1, 0.0025 d-1 for biostimulation, bioaugmentation and natural attenuation, respectively.
Enhanced in situ bioremediation of chlorinated ethenes: from in situ microcosms to full-scale application

Abstract:Concentrations greater than 20 mg/L of chlorinated volatile organic compounds (cVOCs) including tetrachloroethene (PCE), trichloroethene (TCE), and cis-1,2-dichloroethene (cDCE) have been present in site groundwater for more than four decades. To promote a faster clean-up time, an in situ bioremediation approach was evaluated using In-Situ Microcosms ® (ISMs) followed by a full-scale in situ bioremediation approach. The ISM study evaluated slow-release versus quick-release carbon substrates with and without bioaugmentation using the chlorinated ethene degrading culture, SDC-9TM. After a three-month incubation period, the ISMs were retrieved. The ISMs amended with a carbon source with or without bioaugmentation displayed greater than a 93% reduction in TCE, which corresponded to an increase in cDCE in all the ISMs. The Dehalococcoides population and gene abundances associated with chlorinated ethene biodegradation ( tceA , bvcA , vcrA ) increased three orders of magnitude in the bioaugmented ISMs over the natural attenuation ISM and carbon source only amended ISMs. Additionally, the SDC-9TM and AquaBupH ® , an enhanced emulsified oil substrate (EOS ® ) and buffer, amended ISM unit showed the highest level of vinyl chloride and similar level of ethene to the SDC-9TM and EHC ® , a controlled-release, organo-iron substrate, ISM. However, the AquaBupH and SDC-9TM ISM displayed the highest level of acetate, demonstrating active fermentation processes. The results of the ISM study indicated that a combined approach of biostimulation along with bioaugmentation effectively promoted conditions conducive to reductive dechlorination. The full-scale in situ bioremediation system coupled biostimulation with bioaugmentation using SDC-9TM to successfully reduce chlorinated ethenes in groundwater. The slow-release carbon substrate, EOS-100 ® served as a sustained carbon source along with CoBupH (buffering agent similar to AquaBupH), facilitating the production of hydrogen, through volatile fatty acid fermentation. This led to the reduction of chlorinated ethenes over the subsequent three years, showcasing minimal rebound in contaminant levels. Two rounds of bioaugmentation notably increased the Dehalococcoides population, accelerating the biodegradation processes, which is setting up the site for monitored natural attenuation. This study shows that using ISMs to guide full-scale bioremediation design resulted in an effective cVOC biodegradation system that led to quicker and more sustainable clean-up.

Abstract:Pentabromodiphenyl ether (BDE-99) pollution can affect groundwater that has been recharged with treated municipal wastewater. To properly investigate the effect of biochar on the anaerobic degradation of BDE-99 by an archaeal community, we established three lab-scale percolation columns to simulate the recharge process. BDE-99 content decreased with increasing soil depth, especially in the biochar-amended column (BCA), which demonstrated high adsorption capacity and biodegradability. The essential biodegradation of BDE-99 by archaea was confirmed during the recharge process by fluorescence in situ hybridization (FISH) analysis. The effect of biochar on the vertical distribution of typical archaea taxa associated with BDE-99 degradation in the river-based aquifer media was confirmed by 454 pyrosequencing the archaeal 16S ribosomal RNA (16S rRNA) genes. As well as affecting the concentration of polybrominated diphenyl ethers, biochar improved the biodiversity of the archaea and had a critical influence on the majority of archaeal communities. We observed large populations of Nitrosopumilus, Nitrososphaera, and Methanomethylovorans, which are associated with the anaerobic degradation of BDE-99, and the populations of some novel archaea genera, such as Halorhabdus and Methanospirillum, increased markedly owing to the effect of biochar. We suggest that biochar could play a positive role in the anaerobic degradation of BDE-99, and could provide a cost-effective means of mitigating the migration of pollutants.

Abstract:Polybrominated diphenyl ethers (PBDEs) are persistent pollutants that may undergo microbial-mediated debromination in anoxic environments, where diverse anaerobic microbes such as methanogenic archaea co-exist. However, current understanding of the relations between PBDE pollution and methanogenic process is far from complete. To address this knowledge gap, a series of anaerobic soil microcosms were established. BDE-47 (2, 2′, 4, 4′-tetrabromodiphenyl ether) was selected as a model pollutant, and electron donors were supplied to stimulate the activity of anaerobes. Debromination and methane production were monitored during the 12 weeks incubation, while obligate organohalide-respiring bacteria (OHRBs), methanogenic, and the total bacterial communities were examined at week 7 and 12. The results demonstrated slow debromination of BDE-47 in all microcosms, with considerable growth of Dehalococcoides and Dehalogenimonas over the incubation observed in most BDE-47 spiked treatments. In addition, the accumulation of intermediate metabolites positively correlated with the abundance of Dehalogenimonas at week 7, suggesting potential role of these OHRBs in debromination. Methanosarcinaceae were identified as the primary methanogenic archaea, and their abundance were correlated with the production of debrominated metabolites at week 7. Furthermore, it was observed for the first time that BDE-47 considerably enhanced methane production and increased the abundance of mcrA genes, highlighting the potential effects of PBDE pollution on climate change. This might be related to the inhibition of reductive N- and S-transforming microbes, as revealed by the quantitative microbial element cycling (QMEC) analysis. Overall, our findings shed light on the intricate interactions between PBDE and methanogenic processes, and contribute to a better understanding of the environmental fate and ecological implication of PBDE under anaerobic settings.

Abstract:The ubiquitous environmental contaminants, polycyclic aromatic hydrocarbons (PAHs), can be aerobically biodegraded. Strategies for biodegradation of PAHs are needed for the persisted character of it in anoxic environments. In current study, we obtained a highly enriched anaerobic, PAHs-degrading co-culture DYM1, from petroleum-polluted soil. DYM1 significantly degrades a range of PAHs in 4 days without supplementary terminal electron acceptors. Co-culture DYM1 is consists of two microorganisms (a degrading bacterium Paracoccus sp. strain PheM1 and an aceticlastic methanogen Methanosaeta concilii.) that utilize different carbon sources in a syntrophic metabolic process of phenanthrene. About 93% of phenanthrene (104.5 μM) has been removed under methanogenic conditions after incubation with co-culture DYM1 for 4 d, and produced 33.68 μmol CH4. Carboxylation, which is catalyzed by UbiD-like carboxylase, was proposed as the initial steps of methanogenic phenanthrene-degrading pathway based upon the detection of 2-phenanthroic acid and 4-phenanthrene acid. Reduction and hydration of the benzene rings were followed by the initial reaction. Hydrated phenanthroic acid metabolites were newly detected and characterized under anaerobic conditions. Anaerobic degradation of phenanthrene without terminal electron acceptor addition not only sheds light on a poorly understood and environmentally relevant biological process, but also supply a novel approach to recover the energy of toxic pollutant in forms of methane.
Combined Anaerobic/Aerobic Biostimulation for Remediation of Rail Yards Contaminated by Diesel Engine Repair and Maintenance

Abstract:Perchloroethylene (PCE) and trichloroethylene (TCE) have been commonly used in the repair and maintenance of diesel engine locomotives. Improper handling, storage, and disposal lead to contamination of rail yard soils and groundwater with chlorinated ethylenes. Benzene, toluene, ethyl-benzene, and xylene (BTEX) are also common contaminants at rail yards because of the leakage of diesel fuel storage tanks and spills of diesel fuel. Co-contamination of groundwater with BTEX and chlorinated ethylenes allows for the application of anaerobic/aerobic bioremediation to achieve mineralization of both types of compounds. Bench scale laboratory experiments were run to select and enrich for an undefined, mixed, microbial consortium able to mediate PCE dechlorination to dichloroethylene (DCE), mineralization of the DCE, and mineralization of aromatic compounds. A periodically operated suspended culture reactor created alternating anaerobic/aerobic environments. When glucose was added to the reactor as the sole electron donor, the mixed culture dechlorinated PCE to cis-1,2-dichloroethylene (cDCE) in 24 h. When phenol and glucose were added to the reactor, the mixed culture dechlorinated PCE to cDCE, metabolized the influent phenol, and oxidized 90 percent of the cDCE produced in 24 h. Because both phenol and toluene can induce the enzymes necessary for mineralization of TCE and DCE, an anaerobic/aerobic bioremediation approach has potential application for remediation of groundwater at rail yards co-contaminated with diesel fuel and chlorinated ethylenes.

Abstract:Polybrominated diphenyl ethers (PBDEs) are a group of organic pollutants that have attracted much concerns of scientific community over the ubiquitous distribution, chemical persistence and toxicological risks in the environment. Though a great number of aerobic bacteria have been isolated for the rapid removal of PBDEs, the knowledge about biodegradation characteristics and mechanism is less provided yet. Herein, the congener-specificity of aerobic biodegradation of PBDEs by typical bacteria, i.e. B. xenovorans LB400 was identified with the different biodegradation kinetics, of which the changes were largely hinged on the bromination pattern. The more bromination isomerically at ortho -sites other than meta -sites or the single bromination at one of aromatic rings might always exert the positive effect. The biodegradation of PBDEs should be thermodynamically constrained to some extent because the calculated Gibbs free energy changes of initial dioxygenation by quantum chemical method increased with the increase of bromination. Within the transition state theory, the high correlativity between the apparent biodegradation rates and Gibbs free energy changes implied the predominance and rate-limiting character of initial dioxygenation, while the regioselectivity of dioxygenation at the ortho / meta -sites was also manifested for the more negative charge population. The molecular binding with the active domain of dioxygenase BphA1 in aerobe was firstly investigated using docking approach. As significantly illustrated with the positive relationship, the higher binding affinity with BphA1 should probably signify the more rapid biodegradation. Besides the edge-on π-π stacking of PBDEs with F227 or Y277 and π-cation formulation with histidines (H233, H239) in BphA1, the reticular hydrophobic contacts appeared as the major force to underpin the high binding affinity and rapid biodegradation of PBDEs. Overall, the experimental and theoretical results would not only help understand the aerobic biodegradation mechanism, but facilitate enhancing applicability or strategy development of engineering bacteria for bioremediation of PBDEs in the environment.
