Abstract:Based on the special electrical and optical properties resulted from periodic nanostructure, localized surface plasmon resonance (LSPR) sensors have been widely used for various chemical and biological detections. Utilizing uniform alignment nanocups, an electrophoresis-enhanced LSPR sensor was designed for sensitive thrombin detection. The nanocup array was composed of nano-scaled funnel shaped cups with nanoparticles along the side walls. Through self-assembly, polyethylene glycol, thrombin-specific peptide, and bovine serum albumin were successively immobilized on the nanocup array. The results demonstrated that the synchronous implementation of electrophoresis and LSPR measurement could lead to obvious peak shifts in optical transmission spectra for detecting thrombin. The detection limit was as low as 10−11M. With the high sensitivity and well linearity, the electrophoresis-enhanced LSPR sensing offered a novel design perspective for chemical and biological sensors with specific modifications on the nanosensor surfaces.

Abstract:Thrombin is an important kind of proteases that can cleavage specific peptide bonds and plays a crucial role in numerous diseases, which leads to large amounts of efforts on its detection. In this study, a novel peptide self-assembled biosensor was developed based on gold nanoparticles functionalized graphene oxide (GO) for specific detection of thrombin. Through sulfhydryl design, peptides containing thrombin cleavage sites could self-assemble with the nanocomposites of the graphene material and gold particles, which brought the biosensor unique optoelectronic properties and high biological sensitivity to trans duce bio-molecular interactions into optical signals. Spectroscopic measurements were carried out in the presence of thrombin to observe an increase of the absorption peak at similar to 528 nm anda decrease of the absorbance at wavelength larger than 600 nm. The biosensor showed a linear dose-dependence and stable response to thrombin. And with a 528-680 nm absorbance ratio as the sensitive detection parameter, the self-assembled biosensor exhibited high sensitivity and specificity for the thrombin detection. Thus, it revealed convenient and sensitive detection abilities for protease of thrombin, indicating a promising prospect for other related proteases, such as matrix metalloproteinases to cancer related diseases. Abstract:A photoelectrochemical (PEC) sensor for the sensitive detection of thrombin (TB) was established. Co-sensitized combination of TiO2 nanoparticles combined with modified cadmium sulfide and cadmium telluride quantum dots (CdS/CdTe QDs) was utilized as a photoactive material. Successful growth of CdS/CdTe quantum dots on mesoporous TiO2 films occured by successive ion-layer adsorption and reaction. This interesting formation of co-sensitive structure is conducive to enhancing the photocurrent response by improving the use rate of light energy. Additionally, the step-level structure of CdS/CdTe QDs and TiO2 NPs shows a wide range of visible light absorption, facilitating the dissociation of excitons into free electrons and holes. Consequently, the photoelectric response of the PEC analysis platform is significantly enhanced. This constructed PEC aptasensor shows good detection of thrombin with a low detection limit (0.033 pM) and a wide linear range (0.0001-100 nM) in diluted actual human serum samples. In addition, this PEC aptasensor also has the characteristics of good stability and good reproducibility, which provides a novel insight for the quantitative measurement of other similar analytes.
The role of time-factor and RBE for the induction of osteosarcomas by incorporated short-lived bone-seekers.

Abstract:In a large series of experiments, fractionated injections of short-lived bone-seekers have been shown in many cases to cause a remarkable increase of the osteosarcoma incidence compared with a single administration of the same total skeletal dose. This effect has been observed with both alpha- and beta-emitters. In addition the latency period was shortened by protracting the dose. The total skeletal doses investigated ranged between 0.9 and 20 Gy for alpha-emitters (224Ra and 227Th) and between 28 and 112 Gy for the beta-emitter (177Lu). In all cases the protracted dose had higher or at least equal effects when compared with a single application. Reference experiments with long-lived alpha- and beta-emitting bone-seeking nuclides (226Ra and 90Sr) showed that the incidence of osteosarcomas per Gy was sometimes lower than that observed when the same skeletal dose was applied by protraction of short-lived radionuclides. The dependence of osteosarcoma incidence on dose-time distribution, duration of internal irradiation, and radiation quality is discussed. In this context the possibility that the critical initial dose rate may be related to the initiating event within the multi-stage hypothesis of carcinogenesis is considered.
A Novel Electrochemical Aptasensor for Thrombin Detection Based on the Hybridization Chain Reaction with Hemin/g-Quadruplex DNAzyme-signal Amplification.

Abstract:In this work, a novel signal amplification electrochemical aptasensor for the sensitive and selective detection of thrombin was successfully fabricated. The amplification method was based on the hybridization chain reaction (HCR) and a pseudobienzyme electrocatalytic system. HCR-based double-stranded DNA (dsDNA) polymers not only constructed an effective carrier for anchoring larger amounts of electron mediator methylene blue (MB) into the DNA duplexes to produce a strong differential pulse voltammetry (DPV) signal, but also resulted in the formation of hemin/G-quadruplex DNAzymes nanowires by intercalating hemin into two induced single-stranded DNA (ssDNA). With the addition of NADH into the electrolytic cell, the hemin/G-quadruplex acting as an NADH oxidase and HRP-mimicking DNAzyme for the pseudobienzyme amplifying system could in situ biocatalyze the formation of H2O2 with local concentrations and low transfer loss resulting in dramatic signal enhancements. The binding event can be detected by a decrease in the integrated charge of MB which electrostatically absorbed onto dsDNA polymers. In the presence of thrombin, the dsDNA polymers associated with MB and hemin/G-quadruplex structures were removed from the electrode surface, leading to a significant decrease of redox current. DPV signals of MB provided quantitative measures of the concentrations of thrombin, with a linear calibration range of 0.01-50 nM and a detection limit of 2 pM. Moreover, the resulting aptasensor also exhibited good specificity, acceptable reproducibility and stability, indicating that the present strategy was promising for broad potential application in clinic assay and various protein analyses.

Abstract:In most aptamer based stimulus response mesoporous silica nanoparticles (MSN) systems, the aptamer is modified on the MSN via electrostatic interaction, however leakage might exist after a certain time in the system and hence the stability is not good. In this study, the pores of MSN were capped by aptamer through click chemistry reaction for the first time and the system was then employed to develop a fluorescence biosensor. Specifically, the aptamer of the target (thrombin in this study) was hybridized with its complementary DNA (which was initially modified with alkyne at the terminal) to form a double strand DNA (dsDNA) firstly, and then this dsDNA was modified on N3 modified MSN via Cu(I) catalyzed alkyne-azide cycloaddition reaction. The guest molecules (fluorescein) were blocked in the pores of the MSN with high efficiency and nearly no leakage was detected. Upon the introduction of thrombin, thrombin specifically recognized its aptamer, so aptamer released from the MSN; and the single strand DNA(ssDNA) left could not cap the pores of the MSN efficiently and hence caused the releasing of fluorescein into the solution. The enhanced fluorescence intensity of the system has a good linear relationship with the thrombin concentration in the range of 50-1000ngmL-1 with a detection limit of 28.46ngmL-1. The proposed biosensor has been successfully applied to detect thrombin in serum samples with high selectivity. The same strategy can be applied to develop biosensors for different targets by changing the adopted aptamer.

Abstract:A sensitive aptamer-based sandwich-type sensor is presented to detect human thrombin using quantum dots as electrochemical label. CdSe quantum dots were labeled to the secondary aptamer, which were determined by the square wave stripping voltammetric analysis after dissolution with nitric acid. The aptasensor has a lower detection limit at 1 pM, while the sample consumption is reduced to 5 μl. The proposed approach shows high selectivity and minimizes the nonspecific adsorption, so that it was used for the detection of target protein in the human serum sample. Such an aptamer-based biosensor provides a promising strategy for screening biomarkers at ultratrace levels in the complex matrices.

Abstract:Thrombin plays a pivotal role in blood coagulation, wound healing, and tumor metastasis. In this study, we developed a fluorescent probe (Apt15@QDs) by functionalizing quantum dots (QDs) with an aptamer 15 to specifically recognize thrombin. The aptamer Apt15 binds specifically to thrombin by adopting a G-quadruplex conformation, resulting in the formation of a binary complex that quenches the fluorescence of the probe. Analysis using the classical fluorescence quenching equations Stem-Volmer and Lineweaver-Burk revealed that the mechanism of quenching is static in nature. The established method was successfully applied for detecting thrombin in human serum samples. As shown from the results, the established method exhibited high sensitivity, with a limit of detection (LOD) of 8 nM, and possessed a wide linear range within the concentration span of 20 to 200 nM. Additionally, this method circumvented the need for intricate experimental procedures. Furthermore, it offered a short analysis time, obviated the requirement for additional reagents, and proved suitable for high-throughput analysis of biological samples. This study presents a sensitive, straightforward, and rapid approach for detecting thrombin in complex biological matrices.

Abstract:A novel signal-off photoelectrochemical (PEC) biosensing platform based on hybridization chain-doped manganese porphyrin quenching on PEC signal of CdSe quantum dots (QDs) coupling with cyclic amplification was developed for thrombin detection. Target thrombin firstly induced DNA enzyme-aided cyclic amplification reaction and produced abundant DNA I. Then DNA I hybridized with capture DNA on the TiO2/CdSe QDs modified electrode to form DNA bridge, which triggered a linear chain hybridization reaction with S4 and S5. Numerous manganese porphyrin (MnPP) were doped in the long double-stranded DNA chain, which hindered electron transfer and resulted in obvious decrease of QDs PEC signal, achieving a sensitive detection of thrombin. This PEC biosensor exhibits excellent performance with a wide linear range from 1.0 f. to 100.0 nM and detection limit of 0.331 fM, and the recovery value ranged from 95.5% to 103.5%. The results suggest that this PEC biosensor has high sensitivity and precision, showing great application prospect in microbiological analysis of biomolecules and clinical medicine.

Abstract:We fabricated a novel fluorescence biosensor for the selective detection of thrombin by using bovine serum albumin-capped CdS quantum dots (BSA-CdS QDs). Two kinds of designed DNA (DNA1 and DNA2) could bind to CdS QDs through the electrostatic interaction between DNA and Cd(2+) on the surface of CdS QDs. The obtained DNA/BSA-CdS QDs kept stable in the solution with the fluorescence intensity obviously enhanced. Hairpin structure of DNA1contained two domains, one is the aptamer sequence of thrombin and the other is the complementary sequence of DNA2. When thrombin was added, it would bind to DNA1 and induce the hairpin structure of DNA1 changed into G-quadplex structure. Meanwhile, DNA2 would transfer from the surface of CdS QDs to DNA1 via hybridization, which resulted in the removal of DNA1 and DNA2 from the surface of CdS QDs, and led to the fluorescence intensity of CdS QDs reduced. Thus, the determination of thrombin could be achieved by monitoring the change of the fluorescence intensity of CdS QDs. The present method is simple and fast, and exhibits good selectivity for thrombin over other proteins. We have successfully detected thrombin in human serum samples with satisfactory results.

Abstract:This work demonstrates an aptasensor for ultrasensitive electrochemiluminescence (ECL) detection of thrombin based on an “off–on–off” approach. The system is composed of an Eu3+-doped CdS nanocrystals (CdS:Eu NCs) film on glassy carbon electrode (GCE) as ECL emitter. Then gold nanoparticles (AuNPs) labeled hairpin-DNA probe (ssDNA1) containing thrombin-binding aptamer (TBA) sequence was linked on the NCs film, which led to ECL quenching (off) as a result of Förster-resonance energy transfer (FRET) between the CdS:Eu NC film and the proximal AuNPs. Upon the occurrence of hybridization with its complementary DNA (ssDNA2), an ECL enhancement (on) occurred owing to the interactions of the excited CdS:Eu NCs with ECL-induced surface plasmon resonance (SPR) in AuNPs at large separation. Thrombin could induce ssDNA1 forming a G-quadruplex and cause the AuNPs to be close to CdS:Eu NCs film again, which resulted in an enhanced ECL quenching (off). This “off–on–off” system showed a maximum 7.4-fold change of ECL intensity due to the configuration transformation of ssDNA1 and provides great sensitivity for detection of thrombin in a wide detection range from 50aM to 1pM.

Abstract:A novel biosensor for the detection of thrombin was developed by using QDs electrochemiluminescence (ECL) technique. The thiol-terminated aptamer with 15 nucleotides (probe I) was first immobilized on Au electrode, and then thrombin was imported to form the aptamer–thrombin bioaffinity complexes. Another 5′-biotin modified aptamer (29 nucleotides, probe II) was next hybridized with the combined thrombin to form a sandwich type structure. Streptavidin modified QDs (avidin–QDs) were bound to probe II via the biotin–avidin-system. The QDs ECL signal was responsive to the amount of probe II, which was indirect proportional to the combined thrombin. The ECL intensity of the biosensor increased with the increase of thrombin concentration in the range of 0–20μgmL−1. In addition, the biosensor exhibited the excellent selectivity responses and good stability toward the target analyte.

Abstract:A photoelectrochemical (PEC) aptasensor for thrombin detection was rationally designed based on the photoanode of one-dimensional hematite nanorods (α-Fe 2 O 3 NRs) with several steps of modifications. Uniform α-Fe 2 O 3 NRs were grown vertically on the surface of fluorine-doped tin oxide (FTO) conductive glass through a one-step hydrothermal method; then Ag was grown on the surface of α-Fe 2 O 3 NRs through a photoreduction method followed by a partial in-situ transformation into Ag 2 S, conferring an improvement on the initial photocurrent. Two main critical factors, namely, the steric hindrance of thrombin, benzoquinone (BQ) precipitation oxidized by H 2 O 2 under the catalysis of G-quadruplexes/hemin, contributed to the sensitive signal-down response toward the target. Photocurrent signals related with thrombin concentration was established for thrombin analysis due to the non-conductive complex as well as their competitive consumption of electron donors and irradiation light. The excellent initial photocurrent was combined with the signal-down amplification in the design of the biosensor, conferring a limit of detection (LOD) as low as 40.2 fM and a wide linear range from 0.0001 nM to 50 nM for the detection of thrombin. The proposed biosensor was also assessed in terms of selectivity, stability, and applicability in human serum analyses, which provided an appealing maneuver for the specific analysis of thrombin in trace amount.

Abstract:The design of a label-free aptamer for separation of recognition sequence from signal reporter is significant to ensure the high-efficiency affinity between aptamer and target. This work develops a label-free triple-helix aptamer (THA) as sensing platform for "signal-on" fluorescent detection of thrombin. THA was composed of aptamer sequence and help DNA 1 (H1), which contained the complementary sequence of hexachloro-fluorescein (HEX) labeled help DNA 2 (H2). The specific recognition event between aptamer and thrombin triggered the dismission of THA to release H1. The released H1 then reacted with the signal probe of H2/graphene oxide (GO) nanocomposite to form H1-H2 duplex, leading to the fluorescence recovery of H2 due to the detachment of H1-H2 duplex from the surface of GO. With employment of THA as a signal transducer and GO as a "superquencher", this method shows a sensitive response to thrombin with a wide concentration range from 5 to 1200 nM. The limit of detection is 1.8 nM (S/N=3) with excellent selectivity. Considering the universality of THA, the proposed aptasensor would provide a platform for homogeneous fluorescent detection of a wide range of analytes.

Abstract:In this paper, we have synthesized hyperbranched polyester microspheres with carboxylic acid functional groups (HBPE-CA) and developed a label-free electrochemical aptamer biosensor using thrombin-binding aptamer (TBA) as receptor for the measurement of thrombin in whole blood. The indium tin oxide (ITO) electrode surface modified with HBPE-CA microspheres was grafted with TBA, which has excellent binding affinity and selectivity for thrombin. Binding of the thrombin at the modified ITO electrode surface greatly restrained access of electrons for a redox probe of [Fe(CN)6](3-/4-). Moreover, the aptamer biosensor could be used for detection of thrombin in whole blood, a wide detection range (10fM-100nM) and a detection limit on the order of 0.90fM were demonstrated. Control experiments were also carried out by using bull serum albumin (BSA) and lysozyme in the absence of thrombin. The good stability and repeatability of this aptamer biosensor were also proved. We expect that this demonstration will lead to the development of highly sensitive label-free sensors based on aptamer with lower cost than current technology. The integration of the technologies, which include anticoagulant, sensor and nanoscience, will bring significant input to high-performance biosensors relevant to diagnostics and therapy of interest for human health.

Abstract:arbohydrate restriction and rigid meal patterns are no longer the rule for diabetic patients. In addition, differences have been noted in dietary approaches for insulin-dependent diabetes mellitus versus noninsulin-dependent diabetes mellitus. An insulin-dependent person, for example, may need to plan to have extra food when he exercises, while a non-insulin-dependent person does not. The current standards of the American Diabetes Association (ADA) allow up to 60 percent'of daily calories to come from carbohydrate(l). This recommendation is based on clinical studies demonstrating that higher carbohydrate diets may improve glucose tolerance(2). It also reflects a concern that to restrict carbohydrate means to allow more fat-which is not advisable in view of the high incidence of arteriosclerosis among individuals with diabetes. Because of this, ADA recommends that fat intake be limited to 30 percent or less of calories.

Abstract:Thrombin facilitates the aggregation of platelet in hemostatic processes and participates in the regulation of cell signaling. Therefore, the development of thrombin sensors is conducive to comprehending the role of thrombin in the course of a disease. Biosensors based on aptamers screened by SELEX have exhibited superiority for thrombin detection. In this review, we summarized the aptamer-based sensors for thrombin detection which rely on the specific recognitions between thrombin and aptamer. Meanwhile, the unique advantages of different sensors including optical and electrochemical sensors were also highlighted. Especially, these sensors based on electrochemistry have the potential to be miniaturized, and thus have gained comprehensive attention. Furthermore, concerns about aptamer-based sensors for thrombin detection, prospects of the future and promising avenues in this field were also presented.

To realize this purpose, a dense monolayer of 16-mercaptohexadecanoic acid was modified on the gold electrode. In the presence of thrombin, SWNTs were controllably assembled on this insulating monolayer, which could mediate efficient electron transfer between the electrode and eletroactive species to generate a larger redox current. Through detecting the redox signal mediated by SWNTs, this strategy could present significant signal amplification and a detection limit of 50pM thrombin was achieved. Such an aptamer–SWNT based biosensor opens a rapid, selective and sensitive route for thrombin detection and offers a promising strategy for specific protein detection.

Abstract:Herein, a label-free and ultra-sensitive sensing strategy was proposed for the colorimetric detection of thrombin. A catechol oxidase-mimicking nanozyme, MOF-818, was in-situ uniformly synthesized on carbon cloth (CC) fibers surface by hydrothermal method, which did not aggregate during the colorimetric sensing and showed high catalytic activity to the oxidation of faint yellow colored substrate 3,5-Di-tert-butylcatechol (3,5-DTBC) to luminous yellow colored 3,5-di-tert-butyl-obenzoquinone (3,5-DTBQ). After the conjugation of aptamer on MOF-818/CC, the nanozyme composite can effectively recognize and bind thrombin, resulting in the lighter color of substrate solution and the decrease of absorbance at 414 nm. Thus, the inhibition of the nanozyme activity by the bound thrombin was employed to detect thrombin in a wide linear range and a low limit of detection of 1.4 × 10−10M − 1.4 × 10−5M and 6.4 pM, respectively, which was superior to the most of reported methods including fluorescence and electrochemical sensing, Raman spectroscopy. Meanwhile, the nanozyme composite can be easily separated from the substrate solution by a tweezers instead of the conventional centrifugation, which greatly simplified the operation process and facilitated the on-site detection. The nanozyme composite has been successfully applied for the determination of thrombin in real samples with satisfactory results. Consequently, this work is not only important for a better understanding of the unique properties of MOF-818, but also has great potential for medical diagnosis and treatment of human health.
