Abstract:Background: Due to the pathogenic role of opportunistic fungi in immunodeficiency patients, many efforts have been made for developing effective treatment strategies to augment current practice standards. Nystatin, as one of the treatment candidates, is characterized by antifungal effects. In this study, we tried to use liposomal formulation as a nystatin carrier to increase its antifungal efficacy. Materials and methods: A total of 87 positive culture samples of yeast agents were applied to the study. Yeast species were identified by culturing on CHROMagar medium (HiMEDIA), culturing on NigerSide agar medium, and Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Characterization of nanoparticles was examined by the size, zeta potential (ZP), scanning electron microscope (SEM), drug loading, and drug release rate. The standard method of broth microdilution according to CLSI M27-A and the quality control standard of Candida parapsilosis ATCC 22019 were used to evaluate the minimum inhibitory concentration (MIC) of nystatin and nystatin nanoliposomes. Results: The particle size for liposomes containing nystatin was 100.8 ± 17.3 nm. Moreover, the ZP for liposomal formulation of nystatin was 21.14 ± 0.92 -mV. The formulation of nystatin in nanoparticles markedly increased the susceptibility of Candida species to nystatin at lower doses, which was statistically significant compared to free nystatin (P ≤ 0.05). Conclusion: Our results showed that liposomal formulation improves the efficiency of nystatin against albicans species. This formulation can be used to develop new antifungal agents to improve the delivery and absorption of hydrophobic drugs.
Fungus-mediated Biosynthesis of Gold Nanoparticles with Synergistic Antifungal Activity Against Multidrug-Resistant Candida Albicans

Abstract:The widespread prevalence of antifungal resistance results in the ineffective treatment of Candida-related infections since current approaches still heavily rely on antifungal drugs such as azoles. Adjuvant therapy is an alternative approach to alleviate this crisis that can re-sensitize multidrug-resistant (MDR) fungi to antifungal drugs. Herein, we report a synergistic strategy to restore antifungal activity of azoles against MDR Candida albicans (C. albicans) through nanotechnology. C. albicans-mediated biosynthetic gold nanoparticles (Ca_AuNPs) exhibit a significant potentiating effect (16-32 folds) on azoles (including fluconazole, itraconazole, and voriconazole) against MDR C. albicans. Mechanistic studies demonstrate that Ca_AuNPs can promote the intracellular accumulation of fluconazole and trigger the biochemical processes including cell structure destruction, membrane potential dissipation, intracellular ROS generation, and ATP level reduction to overcome the fungal intrinsic resistance. We demonstrate that the adjuvant therapy significantly reduces fungal viability and enhances vaginal mucosa regeneration when treating Candida vaginitis-infected mice. This study reveals the potential of biosynthetic nanoparticles as novel adjuvants to extend the lifespan of existing antifungal drugs for the treatment of MDR pathogen-induced infections.
Evaluation of the in vitro Activity and in vivo Efficacy of Anidulafungin-Loaded Human Serum Albumin Nanoparticles Against Candida albicans

Abstract:Recent decades have seen a significant increase in invasive fungal infections, resulting in unacceptably high mortality rates. Anidulafungin (AN) is the newest echinocandin and appears to have several advantages over existing antifungals. However, its poor water solubility and burdensome route of administration (i.e., repeated, long-term intravenous infusions) have limited its practical use. The objective of this study was to develop anidulafungin-loaded Human Serum Albumin (HSA) nanoparticles (NP) so as to increase both its solubility and antifungal efficacy. HSA was reduced using SDS and DTT, allowing liberation of free thiols to form the intermolecular disulfide network and nanoassembly. Reduced HSA was then added to MES buffer (0.1 M, pH 4.8) and magnetically stirred at 350 rpm and 25°C with AN (m/m 50:1) for 2 h to form nanoparticles (AN NP). We next performed routine antifungal susceptibility testing of Candida strains ( n = 31) using Clinical and Laboratory Standards Institute (CLSI) methodologies. Finally, the in vivo efficacy of both AN and AN NP was investigated in a murine model of invasive infection by one of the most common fungal species— C. albicans. The results indicated that our carrier formulations successfully improved the water solubility of AN and encapsulated AN, with the latter having a particle size of 29 ± 1.5 nm with Polymer dispersity index (PDI) equaling 0.173 ± 0.039. In vitro AN NP testing revealed a stronger effect against Candida species ( n = 31), with Minimum Inhibitory Concentration (MIC) values 4- to 32-fold lower than AN alone. In mice infected with Candida and having invasive candidiasis, we found that AN NP prolonged survival time ( P < 0.005) and reduced fungal burden in kidneys compared to equivalent concentrations of free drug ( P < 0.0001). In conclusion, the anidulafungin nanoparticles developed here have the potential to improve drug administration and therapeutic outcomes for individuals suffering from fungal diseases.

Abstract:Phytotherapeutics is widely used nowadays as an alternative to the current antifungal drugs to reduce their side effects. Curcumin, with its wide therapeutic array as antioxidant and anti-inflammatory agent, is one of the natural compounds that ha..s an antifungal effect, especially when being used at nanoscale to increase its bioavailability. Our research aimed to evaluate clinically and microbiologically the effect of using topical nanocurcumin suspension to treat oral candidiasis. After 4 days from induction of oral candidiasis (baseline), we randomly divided 39 female BALB/c mice into three groups of 13 animals; nanocurcumin, nystatin, and sham groups. All animals in nanocurcumin and nystatin groups received topical treatment twice daily for 10 days. Then, we performed clinical and microbiological evaluations at baseline, day 5, and day 10. By the end of treatment, our results revealed that nanocurcumin promoted a significant reduction in the number of candida colonies. There was no statistically significant difference neither clinically nor microbiologically between nanocurcumin and nystatin groups. In conclusion, nanocurcumin has a good antifungal effect as nystatin, however, its therapeutic efficacy takes a longer time to appear than nystatin. The enhanced bioavailability of curcumin at the nanoscale qualifies this nano-herb as a promising alternative therapy for oral candidiasis, evading nystatin-associated morbidity.
Efficacy of Nystatin for the Treatment of Oral Candidiasis: A Systematic Review and Meta-Analysis

Abstract:Objective: To systematically review and assess the efficacy, different treatment protocols (formulation, dosage, and duration), and safety of nystatin for treating oral candidiasis.Methods: Four electronic databases were searched for trials published in English till July 1, 2015. Randomized controlled trials comparing nystatin with other antifungal therapies or a placebo were included. Clinical and/or mycological cure was the outcome evaluation. A meta-analysis or descriptive study on the efficacy, treatment protocols, and safety of nystatin was conducted.Results: The meta-analysis showed that nystatin pastille was significantly superior to placebo in treating denture stomatitis. Nystatin suspension was not superior to fluconazole in treating oral candidiasis in infants, children, or HIV/AIDS patients. The descriptive investigations showed that administration of nystatin suspension and pastilles in combination for 2 weeks might achieve a higher clinical and mycological cure rate, and using the nystatin pastilles alone might have a higher mycological cure rate, when compared with using nystatin suspensions alone. Nystatin pastilles at a dose of 400,000 IU resulted in a significantly higher mycological cure rate than that administrated at a dose of 200,000 IU. Furthermore, treatment with nystatin pastilles for 4 weeks seemed to have better clinical efficacy than treatment for 2 weeks. Descriptive safety assessment showed that poor taste and gastrointestinal adverse reaction are the most common adverse effects of nystatin.Conclusion: Nystatin pastille was significantly superior to placebo in treating denture stomatitis, while nystatin suspension was not superior to fluconazole in treating oral candidiasis in infants, children, or HIV/AIDS patients. Indirect evidence from a descriptive study demonstrated that administration of nystatin pastille alone or pastille and suspension in combination is more effective than that of suspension alone; prolonged treatment duration for up to 4 weeks can increase the efficacy of nystatin. More well designed and high quality randomized control studies are needed to confirm these findings.

Abstract:The aim of this study was to improve the treatment of Candida albicans biofilms through the use of nystatin solid dispersions developed using maltodextrins as a hyperosmotic carrier. Characterization studies by differential scanning calorimetry, X-ray diffraction, dissolution studies, and particle size analysis were performed to evaluate changes in nystatin crystallinity. Antifungal activity and anti-biofilm efficacy were assessed by microbiological techniques. The results for nystatin solid dispersions showed that the enhancement of antifungal activity may be related to the high proportions of maltodextrins. Anti-biofilm assays showed a significant reduction (more than 80%) on biofilm formation with SD-N:MD [1:6] compared to the nystatin reference suspension. The elaboration process and physicochemical properties of SD-N:MD [1:6] could be a promising strategy for treatment of Candida biofilms.

Abstract:Background: The increasing prevalence of invasive fungal infections in immuno-compromised patients is a considerable cause of morbidity and mortality. With the rapid emergence of antifungal resistance and an inadequate pipeline of new therapies, novel treatment strategies are now urgently required. Methods: The antifungal activity of the alginate oligosaccharide OligoG in conjunction with nystatin was tested against a range of Candida spp. ( C. albicans , C. glabrata , C. parapsilosis , C. auris , C. tropicalis and C. dubliniensis ), in both planktonic and biofilm assays, to determine its potential clinical utility to enhance the treatment of candidal infections. The effect of OligoG (0-6%) ± nystatin on Candida spp. was examined in minimum inhibitory concentration (MIC) and growth curve assays. Antifungal effects of OligoG and nystatin treatment on biofilm formation and disruption were characterized using confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM) and ATP cellular viability assays. Effects on the cell membrane were determined using permeability assays and transmission electron microscopy (TEM). Results: MIC and growth curve assays demonstrated the synergistic effects of OligoG (0-6%) with nystatin, resulting in an up to 32-fold reduction in MIC, and a significant reduction in the growth of C. parapsilosis and C. auris (minimum significant difference = 0.2 and 0.12 respectively). CLSM and SEM imaging demonstrated that the combination treatment of OligoG (4%) with nystatin (1 μg/ml) resulted in significant inhibition of candidal biofilm formation on glass and clinical grade silicone surfaces ( p < 0.001), with increased cell death ( p < 0.0001). The ATP biofilm disruption assay demonstrated a significant reduction in cell viability with OligoG (4%) alone and the combined OligoG/nystatin (MIC value) treatment ( p < 0.04) for all Candida strains tested. TEM studies revealed the combined OligoG/nystatin treatment induced structural reorganization of the Candida cell membrane, with increased permeability when compared to the untreated control ( p < 0.001). Conclusions: Antimicrobial synergy between OligoG and nystatin against Candida spp. highlights the potential utility of this combination therapy in the prevention and topical treatment of candidal biofilm infections, to overcome the inherent tolerance of biofilm structures to antifungal agents.

Abstract:Oral candidiasis is the most common opportunistic fungal infection caused by commensal Candida species. Since there are various local and systemic predisposing factors for the disease, the treatment also varies from topical to systemic antifungal agents. Nystatin is a common antifungal agent used topically. The aim of this systematic review was to evaluate and compare the efficacy of different antifungal agents and the safety of nystatin in the treatment of oral candidiasis. Three electronic databases were searched for randomized controlled trials comparing nystatin with other anti-fungal therapies or placebo. Clinical and/or mycological cure was the outcome evaluation. A meta-analysis and descriptive study on the efficacy, treatment protocols, and safety of nystatin was also conducted. The meta-analysis included five studies, which compared the efficacy of nystatin suspensions with photodynamic therapy. A significant difference in the colony-forming units per milliliters (CFU/mL) of Candida species was observed at 60 days intervals for both palatal mucosa and denture surfaces, with both groups favoring nystatin with low heterogeneity at a 95% confidence interval. Nystatin and photodynamic therapy were found to be equally effective for the clinical remission of denture stomatitis as well as a significant reduction of CFU/mL of Candida species from dentures and palatal surfaces of the patients.

Abstract:Miconazole is a Biopharmaceutics Classification System (BCS) class II drug, thus limiting its oral bioavailability. Nanosponges can improve the oral bioavailability of poorly soluble drugs and sustain their release. The work was aimed at formulating miconazole nanosponges to enhance its solubility and consequently, oral bioavailability. Batches of nanosponges incorporating different concentrations (1, 2 and 3 %) of miconazole nitrate were formed using ethyl cellulose (EC), polyvinyl alcohol (PVA), ethanol and Kolliphor® 188. They were characterized using percentage yield, encapsulation efficiency, Differential Scanning Calorimetry (DSC), Xray Diffraction (XRD), particle sizes, polydispersity index (PDI) and Scanning Electron Microscopy (SEM). In vitro antifungal evaluation against Candida albicans was assessed while in vitro release using simulated gastric fluid was done. Furthermore, in vivo release studies in rats were carried out using the bioassay method. The percentage yield of the nanosponges were between 50 to 60 %. Encapsulation efficiencies ranged between 77 and 98 %. Particle sizes ranged between 266.5 and 781.1 nm. Particles were porous and spongy in appearance. Antifungal evaluation of the nanosponges against Candida albicans revealed activity like that of the pure drug. In vitro release studies showed a sustained release pattern for the nanosponges up till 12 hours whereas the unformulated drug peaked at 2 h. In vivo evaluation showed a concentration dependent increase in blood concentration of the drug with NS4 having the highest release. Nanosponges containing miconazole nitrate were successfully formulated and evaluated and are a promising oral delivery system for miconazole nitrate.

Abstract:Abstract Engineered nanoparticles have enabled the development of novel uses, particularly in disease management. In this investigation, we synthesized and studied three distinct nanomaterials: solid lipid nanoparticles (SLNPs), chitosan nanoparticles (CSNPs), and carbon nanotubes (CNTs), either alone or loaded with two antifungals, nystatin, and fluconazole. The purpose of this study is to investigate the different properties of the produced nanomaterials, either alone or in combination with antifungals. Drug release studies revealed that about 55% from SLNPs, 43% from CSNPs and 97% from CNTs of nystatin drug were released at the longest time point assessed (12 h). In addition, about 89% from SLNPs, 84% from CSNPs and 81% from CNTs of fluconazole drug were released at the longest time point assessed (12 h). This research will expand the understanding of nanomaterials as a viable technique for the management of different fungal diseases that harm several agricultural crops.

Nystatin pastilles and suspension in the treatment of oral candidosis

Abstract:Clinical audit revealed that the treatment of oral candidosis was more successful with nystatin pastilles than with nystatin suspension. The purpose of this investigation was to determine the reasons for this observation. The concentration of nystatin needed to kill 49 consecutive clinical isolates of Candida albicans was measured. The isolates varied in cidal concentrations from 1.875 to 30 U/ml. The time taken to kill these isolates at their cidal concentrations was found to vary from 120 to 300 min. Volunteer studies showed that antifungal activity in the oral cavity was eliminated rapidly after the use of nystatin suspension. In contrast, the polyene could be detected for at least 5 hours after use of the nystatin pastille. The nystatin pastille can be expected to be more effective at killing Candida albicans than the suspension due to its persistent effects

Abstract:Ponderal and linear growth of children has been widely studied; however, epidemiologic evidence of a relationship between the two is inconsistent. Child undernutrition in the form of low height for age and low weight for height continues to burden the developing world. A downward shift in the distribution of height for age in the first 2 y of life is commonly observed in many developing countries and is usually summarized as the percentage stunted (height for age Z-score <-2). Similar shifts are seen in weight for height; however, weight-for-height shifts are often less extreme, perhaps because weight for height is more tightly biologically controlled. Low height for age and low weight for height in childhood share some common factors, including food insecurity, infectious diseases, and inappropriate feeding practices. Reductions in weight for height, generally seen as a short-term response to inadequate dietary intake or utilization, are thought to precede decreases in height for age; however, given an adequate diet and no further insults, catch-up linear growth can occur. Serial instances of decreased weight for height, however, are thought to limit the degree of catch-up growth attained, contributing to linear growth retardation. Additional research is needed to identify the factors associated with recovery of linear growth after a child experiences decreased weight for height. Although the direct relationship between weight for height and height for age is likely limited, each of these measurements indicates important information about the general health of children and their risk of the development of illness or dying; therefore, eliminating the downward shift of height for age and weight for height in developing countries should be prioritized as a public policy.
The Effectiveness of Silver Nanoparticles Mixed with Calcium Hydroxide against Candida albicans: An Ex Vivo Analysis

Abstract:Introduction: The purpose of this study was to assess the antifungal activity of silver nanoparticles (AgNPs) in combination with calcium hydroxide (Ca(OH)2) against Candida albicans (C. albicans). Methods: AgNPs was mixed with pure Ca(OH)2 powder in an aqueous base. A standard suspension (1 × 108 bacterial cells/mL) of C. albicans was prepared in a 96-well plate and incubated on shaker at 37 °C in 100% humidity to allow fungal biofilm formation in infected dentin slices (n = 98). The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of AgNPs alone or with Ca(OH)2 were determined. The samples were separately placed in 24-well tissue culture plates and divided into three experimental groups (0.03, 0.04, and 0.06) and three control groups; negative (saline) and positive chlorhexidine gel and Ca(OH)2. Quantitative measurements of fungal activity by XTT colorimetric assay and qualitative measurements using confocal laser microscopy and scanning electron microscopy were performed. Results: The cell viability of C. albicans in the experimental groups was significantly reduced compared to the negative control group. The combination of (AgNPs (0.04%) and Ca(OH)2) was the most potent against C. albicans. Conclusions: The findings demonstrated that combining silver nanoparticles with Ca(OH)2 was more effective against C. albicans biofilm compared to Ca(OH)2 alone, suggesting a combing effect.

Comparative evaluation of the antifungal efficacy of sodium hypochlorite, chlorhexidine, and silver nanoparticles against Candida albicans

Abstract:Candida albicans is a microorganism that causes root canal infections. However, it cannot be eliminated with conventional irrigation solutions. Recently, silver nanoparticles (AgNPs) have become popular for their superior antimicrobial effects. The purpose of this study was to evaluate the antifungal effect of AgNPs to C. albicans comparing with 5.25% sodium hypochlorite (NaOCl) and 2% chlorhexidine (CHX). Silver nanoparticles were synthesized by chemical reduction method. Minimum inhibitory concentration and minimum fungicidal concentration of AgNPs against C. albicans strain were determined according to microdilution method reported by Clinical Laboratory Standards Institute. C. albicans biofilm layer was formed on the dentin blocks for 10 days. The biofilm structure was observed by scanning electron microscopy. Statistical analysis was performed with one way analysis of variance analysis and group comparisons were performed with Tukey test. AgNPs showed the highest antifungal effect among the groups. 5.25% NaOCl showed the lowest antifungal effect among the groups. While the 2% CHX solution had a statistically lower antifungal effect than AgNPs, it was found to have a higher effect than NaOCl (p < .016). Nanoparticles present a wide research field as an alternative irrigation solution in root canal treatment. The antifungal effect of AgNPs against C. albicans was confirmed in this study. Further in vivo studies should evaluate the conditions of use and long-term prognosis of AgNPs.

Abstract:Naringenin (NRG) inhibits the fungal 17β-hydroxysteroid dehydrogenase accountable for ergosterol synthesis in Candida albicans ( C. albicans ), a causative agent for cutaneous candidiasis. In present research, NRG was complexed with ZnO nanomaterial (NRG-Zn 2+ ) to synthesize NRG-Zn 2+ nanocomposites. The particle size and ζ-potential of NRG-Zn 2+ nanocomposites were respectively estimated to be 180.33 ± 1.22-nm and − 3.92 ± 0.35-mV. In silico data predicted the greater affinity of NRG-Zn 2+ nanocomposite for 14α-demethylase and ceramide in comparison to NRG alone. Later, NRG-Zn 2+ nanocomposites solution was transformed in to naringenin-zinc oxide nanocomposites loaded chitosan gel (NRG-Zn-CS-Gel) with viscosity and firmness of 854806.7 ± 52386.43 cP and 698.27 ± 10.35 g, respectively. The ex-vivo skin permeation demonstrated 70.49 ± 5.22% skin retention, significantly greater ( P < 0.05) than 44.48 ± 3.06% of naringenin loaded chitosan gel (NRG-CS-Gel) and 31.24 ± 3.28% of naringenin solution (NRG Solution). NRG-Zn-CS-Gel demonstrated 6.71 ± 0.84% permeation of NRG with a flux value of 0.046 ± 0.01-μg/cm 2 /h. The MIC 50 of NRG-Zn-CS-Gel against C. albicans was estimated to be 0.156-μg/mL with FICI (fractional inhibitory concentration index) of 0.018 that consequently exhibited synergistic efficacy. Further, NRG-Zn-CS-Gel demonstrated superior antifungal efficacy in C. albicans induced cutaneous candidiasis infection in Balb/c mice. The fungal burden in NRG-Zn-CS-Gel treated group was 109 ± 25 CFU/mL, significantly lower ( P < 0.05) than positive control (2260 ± 446 CFU/mL), naringenin loaded chitosan gel (NRG-CS-Gel; 928 ± 127 CFU/mL) and chitosan gel (CS-Gel; 2116 ± 186 CFU/mL) treated mice. Further, histopathology examination and cytokine profiling of TNF-α, IL-1β and IL-10 revealed the healing of skin and inflammation associated with cutaneous candidiasis infection. In conclusion, NRG-Zn-CS-Gel may be a potential candidate for translating in to a clinical viable topical nanotherapeutic. Graphical

The Antifungal Activity of Chitosan Nanoparticle-Incorporated Probiotics Against Oral Candidiasis

Abstract:Background: Candida species, especially Candida albicans, cause oral candidiasis, also known as oral thrush. It affects the elderly, newborns, patients under antibiotics, chemotherapeutic agents, and patients with weaker immune systems. Although successful, traditional antifungal medications are available, they may have negative effects and do not prevent recurrence. Conventional antifungal medications have a restricted range of effectiveness; hence, the need for a novel antifungal agent arises. Chitosan, a natural polymer made from chitin found in crustaceans, kills fungal cells by damaging membranes. Excellent biocompatibility and biodegradability make it a promising medical material. Probiotics, live bacteria, compete with pathogenic microbes for adhesion sites and nutrients, produce antimicrobials such as lactic acid, hydrogen peroxide, and bacteriocins, and modulate the host's immunological response. Adding probiotics to chitosan nanoparticles (CSNPs) boosts their antifungal activity against Candida species and increases their stability and transport to the infection site. Methods: The antifungal activity was evaluated through in vitro experiments utilizing Candida strains that are significant to oral candidiasis. Different concentrations of CSNPs and formulations loaded with probiotics were examined to assess their effectiveness. The antifungal properties were assessed using microbiological assays, specifically agar diffusion and minimum inhibitory concentration (MIC). Results: The study demonstrated a notable fungicidal impact of CSNPs combined with probiotics against oral candidiasis. Furthermore, the MIC values were lower for the probiotic-loaded CSNPs than for chitosan alone or probiotics alone. Conclusion: The combination of CSNPs and probiotics shows potential in effectively combating oral candidiasis by inhibiting fungal growth. This novel method offers a promising strategy for creating therapeutic treatments for oral candidiasis by utilizing the combined benefits of chitosan and probiotics to combat fungal infections effectively.
Formulation and evaluation of Fluconazole Nanosuspensions: In vitro characterization and transcorneal permeability studies

Abstract:The aim in this study was to develop and evaluate a nanofluconazole (FLZ) formulation with increased solubility and permeation rate using nanosuspensions. The FLZ nanosuspensions were stabilized using a variety of stabilizing agents and surfactants in various concentrations. The FLZ nanosuspension was characterized in vitro using particle size, zeta potential, X-ray powder diffraction (XRPD), and solubility. In addition, the ex vivo ocular permeation of FLZ through a goat cornea was analyzed. The results showed that the particle size of all nanosuspension formulations was in the nanometer range from 174.5 ± 1.9 to 720.2 ± 4.77 nm; that of the untreated drug was 18.34 μm. The zeta potential values were acceptable, which indicated suitable stability for formulations. The solubility of the nanosuspensions was up to 5.7-fold higher compared with that of the untreated drug. The results of the ex vivo ocular diffusion of the FLZ nanosuspensions showed the percentage of FLZ penetrating via the goat cornea increased after using Kollicoat to stabilize the nanosuspension formulation. Consequently, when using a nanosuspension formulation of Kollicoat, the antifungal activity of the drug strengthens.

Continuous dosing of nikkomycin Z against systemic candidiasis, in vivo and in vitro correlates

Abstract:Background: Candidiasis is the most common cause of fungal sepsis, and new agents are of interest to ameliorate current deficiencies in therapy. Nikkomycin Z (NIKZ) is an inhibitor of chitin synthase, interfering with fungal cell wall development. Objectives/methods: We studied NIKZ therapy of disseminated murine candidiasis, via continuous drug exposure, in drinking water, to compensate for rapid clearance of the drug. Results: Drinking, and thus drug intake in the NIKZ groups, as well as body weight, was affected by the degree of illness. NIKZ effect on survival, despite reduced drinking initially after infection, was highly efficacious and dose-related, and comparable to fluconazole, though neither were curative with the regimens employed. The challenge was rapidly lethal to all untreated animals, whereas NIKZ groups achieved >50% survival. Assays of residual fungal infection were consistent with impressions of efficacy based on survival. Although NIKZ MIC for Candida albicans appeared unpromising, mycelial formation assays more closely correlated with in vivo observations. Conclusions: In vitro-in vivo disparity may be explained by NIKZ tissue concentration in the target tissue and/or by enhanced NIKZ action on mycelial formation, a morphological change in vivo wherein chitin synthesis is more critical, compared to NIKZ activity in inhibiting planktonic growth. A sustained release oral form of NIKZ in drug development for humans could hold promise, possibly also in future exploring previously demonstrated synergy in vitro with other antifungals.

Abstract:High-resolution FTIR data of carbonyl fluoride in the region of the C-O stretch nu1 fundamental have been analyzed. Satisfactory interpretation of the spectral data required a rigorous analysis that involved the inclusion of Fermi resonance with the 2nu2 state and Coriolis interaction with the 2nu3 + nu6 dark state. Deperturbed molecular constants for the three interacting states as well as perturbation parameters that fit the data within the limits of the experimental accuracy of approximately 0.00035 cm-1 have been obtained. Abstract:In CCD based imaging systems operating in multiple output taps (channels), electronic channel crosstalk is a major issue that limits the image quality. We propose characterisation of crosstalk in a high speed image acquisition system that employs multi-tap CCD as the imaging sensor. The characterisation is performed by analysing imaged test patterns in frame mode. These test patterns are formed based on the analog signal flow architecture of the sensor and image acquisition system. Test patterns imaged from a high resolution monochrome monitor are analysed to observe and quantify crosstalk on every channel. This pattern image based approach formulated in a hierarchical manner can be applied to characterise crosstalk for any multi-port, multi-plane sensor configuration of high speed imaging systems.
