Abstract:SUMMARY Poison frogs in the anuran family Dendrobatidae use bright colors on their bodies to advertise toxicity. The species Dendrobates pumilio Schmidt 1858, the strawberry poison frog, shows extreme polymorphism in color and pattern in Panama. It is known that females of D. pumiliopreferentially choose mates of their own color morph. Nevertheless, potential predators must clearly see and recognize all color morphs if the aposematic signaling system is to function effectively. We examined the ability of conspecifics and a model predator to discriminate a diverse selection of D. pumilio colors from each other and from background colors. Microspectrophotometry of isolated rod and cone photoreceptors of D. pumilio revealed the presence of a trichromatic photopic visual system. A typical tetrachromatic bird system was used for the model predator. Reflectance spectra of frog and background colors were obtained, and discrimination among spectra in natural illuminants was mathematically modeled. The results revealed that both D. pumilio and the model predator discriminate most colors quite well, both from each other and from typical backgrounds, with the predator generally performing somewhat better than the conspecifics. Each color morph displayed at least one color signal that is highly visible against backgrounds to both visual systems. Our results indicate that the colors displayed by the various color morphs of D. pumilio are effective signals both to conspecifics and to a model predator.

Abstract:Abstract Aposematism is the use of warning signals to advertise unpleasant or dangerous defences to potential predators. As the effectiveness of this strategy depends on predator learning, little variation is expected in aposematic warning signals, as similar signals facilitate predator learning. However, warning signals are frequently variable in aposematic species. Such variability could arise as a result of geographic variation in the interpretation that local predators give warning signals. We tested this divergent learning hypothesis in the polytypic poison frog Andinobates bombetes (Anura: Dendrobatidae), focusing on visual predators. Our study was conducted in two populations of this species located in the Western Andes of Colombia, where individuals at some localities exhibit red dorsolateral stripes, while those in others exhibit yellow dorsolateral stripes. We deployed paraffin models imitating both forms of A. bombetes in size and colouration, as well as dull‐coloured controls, at sites inhabited by either red‐striped or yellow‐striped frogs. Red and yellow models were attacked at similar rates at both sites, and brown models were attacked more frequently at one of the sites. These results suggest that red and yellow colourations function as similarly effective aposematic signals for primarily visual predators, regardless of the form previously experienced by these predators. Therefore, our results do not support the hypothesis of divergent predator learning as a driver of the polytypism present in this species. Finally, we discuss other mechanisms that may be involved in the evolution and maintenance of this polytypism.

Abstract:In this manuscript, we investigate color variances between aposematic and cryptic color morphs of two Oophaga species and under the visual system of three different predators. No significant variations was observed in color and luminance contrasts between species. Animals that are toxic often advertise their unprofitability to potential predators through bright aposematic colors while cryptic ones blend in with their natural background to avoid predators. In the poison dart frogs, Oophaga pumilio and O. granulifera, some populations in Costa Rica and Panama display cryptic green and aposematic red color morphs. We herein used reflectance spectra from the dorsum of red and green morphs of these frogs to estimate their perception by the visual systems of three potential predators (birds, lizards, and crabs) against three natural backgrounds (leaves, trunks and leaf litter). Statistical analyses revealed no strong differences in color contrast against backgrounds between the two frog species. However, strong effects of frog morph, predator, background, and their interactions were observed. When viewed against diverse backgrounds, red frogs of both Oophaga species are more color conspicuous to birds and Anoline lizards than to crabs. A strong effect of species was observed on luminance contrast. Concerning the latter, green frogs particularly in O. granulifera appear more conspicuous than red frogs, while birds perceive higher brightness contrasts than lizards or crabs. Our results further support the importance of birds and lizards as Oophaga predators and provide a first quantitative comparison of conspicuousness between these two frog species.

Detectability of a poison frog and its Batesian mimic depends on body posture and viewing angle

Abstract:Aposematic signals warn predators that prey should be avoided due to dangerous secondary defences. However, as warning signals do not always produce avoidance, warning colors may evolve as a trade-off balancing detectability against signal saliency. For Batesian mimics, which display salient signals but lack secondary defenses, the costs of predator encounters are greater, potentially increasing the benefit of crypsis. This raises the question of whether imperfect mimicry may reduce detectability while retaining mimetic efficacy. We tested this hypothesis with the poisonous frog Ameerega bilinguis and undefended Batesian mimic Allobates zaparo, using computational visual modeling and screen-based detection trials with human participants. We found that both species incorporate camouflage into their warning colors, but to different degrees depending on viewing angle and behavior. Contrary to expectation, we found differences in detectability between model and mimic that do not adhere to the hypothesized cryptic mimetic phenotype. To aerial observers, we found the mimic to be more detectable than the model. To terrestrial observers, likely owing to the model's bright ventral color, we found the model more detectable in viewing angles that highlight the ventral coloration, whereas the mimic was more detectable in viewing angles that highlight the dorsal coloration. Consequently, we suggest that in addition to being the result of perceptual or developmental constraints, imperfect mimicry may also evolve as an adaptive strategy which balances camouflage with different signaling functions. Our findings complement the emerging view that aposematic signals may evolve in response to a multitude of selection pressures beyond aversion alone.

Abstract:Aposematic organisms rely on their bright conspicuous coloration to communicate to potential predators that they are toxic and unpalatable. These aposematic phenotypes are strongly tied to survival and therefore make excellent opportunities to investigate the genetic underpinning of coloration. The genus Ranitomeya includes phenotypically diverse members of Neotropical aposematic poison frogs native to South America. Significant progress has been made in elucidating the molecular mechanisms responsible for aposematic coloration in poison frogs, which have paved the way for future studies to test hypotheses of the evolution of coloration across aposematic vertebrates. However, very little is known about whether these color related genes are under positive selection. We assembled transcriptomes from publicly available data reads sets for 9 different color morphs of poison frogs in the Ranitomeya genus that display bright conspicuous coloration (four morphs of R. imitator , two morphs of R. variabilis , two morphs of R. fantastica , one morph of R. summersi ) to identify protein-coding genes responsible for color production that are under positive selection. Our results show that there are multiple genes under strong positive selection that are predicted to play roles in melanin synthesis ( dct, tyrp1, irf4 ), iridophore development ( fhl1 ), keratin metabolism ( ovol1 ), pteridine synthesis ( prps1 , xdh ), and carotenoid metabolism ( adh1b, aldh2 ). The identification of positive selection affecting candidate color-pattern genes is consistent with the possibility that these genes mediate (in part) the molecular evolution of coloration. This may be attributed to aposematic phenotypes being directly tied to survival and reproduction in poison frogs.

Abstract:Aposematic signals visually advertise underlying anti-predatory defences in many species. They should be detectable (e.g. contrasting against the background) and bold (e.g. using internal pattern contrast) to enhance predator recognition, learning and memorization. However, the signalling function of aposematic colour patterns may be distance-dependent: signals may be undetectable from a distance to reduce increased attacks from naïve predators but bold when viewed up close. Using quantitative colour pattern analysis, we quantified the chromatic and achromatic detectability and boldness of colour patterns in 13 nudibranch species with variable strength of chemical defences in terms of unpalatability and toxicity, approximating the visual perception of a triggerfish ( Rhinecanthus aculeatus ) across a predation sequence (detection to subjugation). When viewed from an ecologically relevant distance of 30 cm, there were no differences in detectability and boldness between well-defended and undefended species. However, when viewed at closer distances (less than 30 cm), well-defended species were more detectable and bolder than undefended species. As distance increased, detectability decreased more significantly than boldness for defended species. For undefended species, boldness and detectability remained comparatively consistent, regardless of viewing distance. We provide evidence for distance-dependent signalling in aposematic nudibranchs and highlight the importance of distinguishing signal detectability from boldness in studies of aposematism.

Abstract:Amphibians are ideal for studying visual system evolution because their biphasic (aquatic and terrestrial) life history and ecological diversity expose them to a broad range of visual conditions. Here, we evaluate signatures of selection on visual opsin genes across Neotropical anurans and focus on three diurnal clades that are well-known for the concurrence of conspicuous colors and chemical defense (i.e., aposematism): poison frogs (Dendrobatidae), Harlequin toads (Bufonidae: Atelopus ), and pumpkin toadlets (Brachycephalidae: Brachycephalus ). We found evidence of positive selection on 44 amino acid sites in LWS , SWS1 , SWS2 , and RH1 opsin genes, of which one in LWS and two in RH1 have been previously identified as spectral tuning sites in other vertebrates. Given that anurans have mostly nocturnal habits, the patterns of selection revealed new sites that might be important in spectral tuning for frogs, potentially for adaptation to diurnal habits and for color-based intraspecific communication. Furthermore, we provide evidence that SWS2 , normally expressed in rod cells in frogs and some salamanders, has likely been lost in the ancestor of Dendrobatidae, suggesting that under low-light levels, dendrobatids have inferior wavelength discrimination compared to other frogs. This loss might follow the origin of diurnal activity in dendrobatids and could have implications for their behavior. Our analyses show that assessments of opsin diversification in across taxa could expand our understanding of the role of sensory system evolution in ecological adaptation.

Abstract:Abstract Camouflage and warning signals are different antipredator strategies, which offer an excellent opportunity to study the evolutionary forces acting on prey appearance. Edible prey often escape detection via camouflage, which usually leads to apostatic selection favoring rare morphs. By contrast, defended prey often display conspicuous coloration acting as warning signals to predators, which usually leads to positive frequency dependence and signal uniformity. However, when two morphs of the same species vary greatly in conspicuousness, the maintenance of both cryptic and conspicuous forms in profitable prey populations remains enigmatic. Using the white and melanic morphs of the invasive box tree moth (Cydalima perspectalis) presented at three different frequencies, we investigate (a) the palatability of caterpillars and adult moths to birds, (b) predation rates on the less conspicuous melanic morph, and (c) the role of frequency dependence in balancing morph frequencies. Our results show that caterpillars are distasteful for birds but not adult moths that are fully palatable. We found that the less conspicuous, melanic morph, benefits from reduced predation due to its lower detectability. The more conspicuous, white morph, instead, is more predated and is best off when common, suggesting positive frequency dependence. These results offer new insights into the evolution of color polymorphism and prey defenses in a polymorphic moth species. Further investigation is required to understand the role of different predation regimes on the maintenance of the polymorphism in this species and test whether additional selection pressures operate in natural populations.

Abstract:While there are many studies documenting female mating preferences across taxa, male mate choice remains relatively understudied. Male mate choice often develops when there is variation in female quality and thus the fitness benefits of mating with particular females. Specifically, males tend to prefer females with traits that confer direct fitness benefits such as large body size, which may be linked with high fecundity. Prior work has shown that females of the strawberry poison frog, Oophaga pumilio , prefer males bearing certain coloration (most often the female's own color), and that this preference can be learned through maternal imprinting. Females have been shown to prefer larger males as well. Here we test whether similar mate preferences for color and size exist in males of this species using two-way choice tests on captive bred male O. pumilio . In each test focal males were placed in an arena with two stimulus females: either both of the same size but differing in color, or both of the same color but differing in size. We found only weak evidence for behavioral biases toward particular colors and no evidence for biases toward larger females, suggesting that males of O. pumilio do not predictably choose mates based on these female traits. Despite several aspects of their natural history that suggest males have reasons to be choosy, our findings suggest that the cost of mate rejection may outweigh any fitness benefits derived from being selective of mates. Studies of additional populations, ideally conducted on wild individuals, are needed to better understand the range of conditions under which males may exhibit mate choice and the types of traits on which they base these choices.

Abstract:BackgroundColor and pattern phenotypes have clear implications for survival and reproduction in many species. However, the mechanisms that produce this coloration are still poorly characterized, especially at the genomic level. Here we have taken a transcriptomics-based approach to elucidate the underlying genetic mechanisms affecting color and pattern in a highly polytypic poison frog. We sequenced RNA from the skin from four different color morphs during the final stage of metamorphosis and assembled a de novo transcriptome. We then investigated differential gene expression, with an emphasis on examining candidate color genes from other taxa.ResultsOverall, we found differential expression of a suite of genes that control melanogenesis, melanocyte differentiation, and melanocyte proliferation (e.g., tyrp1, lef1, leo1, and mitf) as well as several differentially expressed genes involved in purine synthesis and iridophore development (e.g., arfgap1, arfgap2, airc, and gart).ConclusionsOur results provide evidence that several gene networks known to affect color and pattern in vertebrates play a role in color and pattern variation in this species of poison frog.

Pattern Matters in the Aposematic Colouration of Papilio polytes Butterflies

Abstract:Many toxic animals display bright colour patterns to warn predators about their toxicity. This sometimes leads other sympatric palatable organisms to evolve mimetic colour patterns to also evade predation. These mimics, however, are often imperfect, and it is unclear how much their colour patterns can vary away from the model before they become ineffective. In this study, we investigated how predation risk of the palatable Common Mormon butterfly (Papilio polytes) is affected by two alterations of its wing pattern that make it progressively more distinct from its model, the Common Rose (Pachliopta aristolochiae). We deployed butterfly paper models in the field, where all models displayed the same colours but had different patterns. In the first modification from the Wildtype pattern, we exchanged the position of the red and white colour patches but kept the overall pattern constant. In the second modification, we created an eyespot-like shape from the pre-existing pattern elements by moving their positions in the wing, altering the overall wing pattern. Both modifications increased attack risk from predators relative to Wildtype patterns, with the eyespot-like modification having the highest predation risk. Our results show that avian predators can distinguish between all three patterns tested, and that pattern is important in aposematic signals. Predators learn to avoid aposematic colours, not in isolation, but as part of specific patterns.
Surviving the serenade: how conflicting selection pressures shape the early stages of sexual signal diversification

Abstract:Abstract Understanding how the early stages of sexual signal diversification proceed is critically important because these microevolutionary dynamics directly shape species trajectories and impact macroevolutionary patterns. Unfortunately, studying this is challenging because signals involve complex interactions between behavior, morphology, and physiology, much of which can only be measured in real-time. In Hawaii, male Pacific field cricket song attracts both females and a deadly parasitoid fly. Over the past two decades, there has been a marked increase in signal variation in Hawaiian populations of these crickets, including novel male morphs with distinct mating songs. We capitalize on this rare opportunity to track changes in morph composition over time in a population with three novel morphs, investigating how mate and parasitoid attraction (components of sexual and natural selection) may shape signal evolution. We find dramatic fluctuation in morph proportions over the three years of the study, including the arrival and rapid increase of one novel morph. Natural and sexual selection pressures act differently among morphs, with some more attractive to mates and others more protected from parasitism. Collectively, our results suggest that differential protection from parasitism among morphs, rather than mate attraction, aligns with recent patterns of phenotypic change in the wild.

Abstract:Intraspecific variation in body size, both among populations and between sexes, is an important factor influencing life-history strategies. This variation might be the response to different environmental conditions, as well as natural and sexual selection, and can result in differences in behavior and reproductive strategies among populations. Here, we use the dyeing poison frog ( Dendrobates tinctorius ) as a model to investigate how interpopulation variation in body size and sexual size dimorphism affects reproductive strategies. As body size increased, sexual size dimorphism also increased, i.e., females were larger than males, and more so in populations with overall larger frogs. This indicates that there is a stronger selection for body size in females than in males, likely as a response to divergent reproductive investment between the sexes. Females from larger-bodied populations produced larger clutches, but the overall number of froglets produced per clutch did not differ among populations. We discuss potential causes and mechanisms that might be responsible for the observed divergence in body size, sexual size dimorphism, and reproductive strategies among populations that likely represent local adaptations. Our findings demonstrate the importance of cross-population studies, cautioning against drawing general conclusions about a species' ecology without accounting for intraspecific variation.

Abstract:Assortative or disassortative aggression – when individuals display more aggression towards conspecifics with similar or different phenotypic characteristics – can either maintain polymorphisms or facilitate gene flow between populations depending on which direction the aggression is aimed. Deciphering which factors elicit or prevent aggression is crucial to improving our knowledge of the origin and maintenance of reproductive barriers and subsequent speciation. The Peruvian mimic poison frog, Ranitomeya imitator , is a monogamous and territorial species that has evolved into four distinct color-pattern morphs in a mimetic radiation. Here we use historical landscape genetic data and competition trials between male individuals sourced from different populations and color-pattern morphs to show that the level of aggression between individuals is not associated with color morph or body size but rather with source population. Individuals spent more time in combat with individuals from their own deme (genetically homogeneous population), irrespective of color morph or size. These findings indicate that genotypic similarity is correlated with increased aggression in R. imitator , though the mechanism by which R. imitator males identify conspecifics as territorial threats remains unclear. As body size and color morph were not significantly associated with aggression levels, this study emphasizes the necessity of further research to identify whether other phenotypic traits are influencing territorial behavior between male frogs, and if these factors play a role in increasing gene flow, or conversely, the formation of reproductive barriers between populations.

Context-dependent coloration of prey and predator decision making in contrasting light environments

Abstract:A big question in behavioral ecology is what drives diversity of color signals. One possible explanation is that environmental conditions, such as light environment, may alter visual signaling of prey, which could affect predator decision-making. Here, we tested the context-dependent predator selection on prey coloration. In the first experiment, we tested detectability of artificial visual stimuli to blue tits (Cyanistes caeruleus) by manipulating stimulus luminance and chromatic context of the background. We expected the presence of the chromatic context to facilitate faster target detection. As expected, blue tits found targets on chromatic yellow background faster than on achromatic grey background whereas in the latter, targets were found with smaller contrast differences to the background. In the second experiment, we tested the effect of two light environments on the survival of aposematic, color polymorphic wood tiger moth (Arctia plantaginis). As luminance contrast should be more detectable than chromatic contrast in low light intensities, we expected birds, if they find the moths aversive, to avoid the white morph which is more conspicuous than the yellow morph in low light (and vice versa in bright light). Alternatively, birds may attack first moths that are more detectable. We found birds to attack yellow moths first in low light conditions, whereas white moths were attacked first more frequently in bright light conditions. Our results show that light environments affect predator foraging decisions, which may facilitate context-dependent selection on visual signals and diversity of prey phenotypes in the wild.
Anti-predator defences are linked with high levels of genetic differentiation in frogs

Abstract:Predator–prey interactions have been suggested as drivers of diversity in different lineages, and the presence of anti-predator defences in some clades is linked to higher rates of diversification. Warning signals are some of the most widespread defences in the animal world, and there is evidence of higher diversification rates in aposematic lineages. The mechanisms behind such species richness, however, are still unclear. Here, we test whether lineages that use aposematism as anti-predator defence exhibit higher levels of genetic differentiation between populations, leading to increased opportunities for divergence. We collated from the literature more than 3000 pairwise genetic differentiation values across more than 700 populations from over 60 amphibian species. We find evidence that over short geographical distances, populations of species of aposematic lineages exhibit greater genetic divergence relative to species that are not aposematic. Our results support a scenario where the use of warning signals could restrict gene flow, and suggest that anti-predator defences could impact divergence between populations and potentially have effects at a macro-evolutionary scale.

Zinc(II)-coordinated oligotyrosine: a new class of cell penetrating peptide.

Abstract:A new series of cell penetrating peptides (CPPs) are described. The peptides are oligomers of Tyr-ZnDPA, a tyrosine derivative with an appended 2,2'-dipicolylamine unit that forms a very stable coordination complex with a zinc (II) cation. This in turn allows reversible association with a chelating oxyanion such as a carboxylate or phosphate derivative. The peptide oligomers (Tyr-ZnDPA) n where n = 1, 2, 4, 8, are highly water soluble, but upon association with fatty acids or phospholipids they partition into an organic octanol phase. Furthermore, a fluorescent, fluorescein-labeled version of the octamer, (Tyr-ZnDPA) 8-Fl, can enter living mammalian cells via endocytosis and a biotin derivative can deliver fluorescein-labeled streptavidin. Fluorescence microscopy and flow cytometry experiments show that cell uptake is diminished by conditions that inhibit endocytosis. Additionally, uptake of (Tyr-ZnDPA) 8-Fl is greater than fluorescein labeled octaarginine (Arg 8-Fl) in all cell lines tested (CHO, COS-7, HeLa). Another difference with Arg 8-Fl is that cell uptake of (Tyr-ZnDPA) 8-Fl does not require the presence of heparan sulfate proteoglycans on the cell surface. This difference may eventually be of practical value because drug delivery systems that employ alternative endocytic mechanisms may be optimal for different cell lines or they may deliver selectively to different organelles within a cell.
Sex-specific ventral dichromatism and melanization in harlequin toads (Atelopus): a common but overlooked character of unknown function

Abstract:Hidden colors are a widespread phenomenon in the animal kingdom, particularly in anurans. In some cases, hidden colors are suddenly exposed during defensive displays to startle predators, others seemingly remain hidden—particularly from researchers. Amazonian species of Neotropical harlequin toads (genus Atelopus ) show striking and consistent ventral sexual dichromatism where females show and males generally lack melanization. Inspired by these observations we undertook a deeper inquiry across this species-rich genus. We collected data on ventral sexual dichromatism in Atelopus species and scored expression of sex-specific ventral melanization (i.e. black, brown and/or grey coloration). Ventral sexual dichromatism was present throughout the entire range of the genus and in almost all phylogenetic groups. However, there was a clear geographic signal with this trait being most common and widespread in Amazonian Atelopus species. Ventral melanization was correlated with temperature and elevation. Focusing on the Amazonian species, we present hypotheses on potential functions of sexually dimorphic ventral patterns and sex-specific ventral melanization as a baseline to further investigate the dynamics of sexual and natural selection as potential drivers of these traits. Selective pressures on less exposed body parts, such as ventral sides, likely differ considerably from those on dorsal appearance. Given the amount of research on amphibian coloration, it is remarkable how little we know about the evolution, function and underlying mechanisms of ventral appearance. We hope our work will spark more interest in the flip side of amphibians, thereby broadening our understanding of animal coloration.

Abstract:To avoid predation, many animals mimic behaviours and/or coloration of dangerous prey. Here we examine potential sex-specific mimicry in the jumping spider Habronattus pyrrithrix . Previous work proposed that males' conspicuous dorsal coloration paired with characteristic leg-waving (i.e. false antennation) imperfectly mimics hymenopteran insects (e.g. wasps and bees), affording protection to males during mate-searching and courtship. By contrast, less active females are cryptic and display less leg-waving. Here we test the hypothesis that sexually dimorphic dorsal colour patterns in H. pyrrithrix are most effective when paired with sex-specific behaviours. We manipulated spider dorsal coloration with makeup to model the opposite sex and exposed them to a larger salticid predator ( Phidippus californicus ). We predicted that males painted like females should suffer higher predation rates than sham-control males. Likewise, females painted like males should suffer higher predation rates than sham-control females. Contrary to expectations, spiders with male-like coloration were attacked more than those with female-like coloration, regardless of their actual sex. Moreover, males were more likely to be captured, and were captured sooner, than females (regardless of colour pattern). With these unexpected negative results, we discuss alternative functional hypotheses for H. pyrrithrix colours, as well as the evolution of defensive coloration generally.

Abstract:Variation of visual signal coloration has been predicted among closely related species or populations by the sensory drive hypothesis and tested in fishes, reptiles, or birds. However, despite the tremendous diversity of insect colorations and habitat preferences, sensory drive constrained by habitat transmission through different light environments has not been investigated in this group. Here, we study in a strictly crepuscular horned beetle (Coprophanaeus lancifer) the relationship between body coloration, ambient light spectra, and conspicuousness to conspecifics. We estimate brightness contrast of the mean coloration, as it is seen at relatively long distance, and color contrast of the horn seen in front of the pronotum, as it is seen during interindividual contests occurring at short distance, using a physiological model of crepuscular horned beetle vision. As predicted, brightness contrast of the mean coloration is optimal in the dusk ambient light spectrum compared with diurnal light environments. Similarly, color contrast of the horn seen in front of the pronotum is maximal in dusk light compared with other forest light environments. Dusk light favors both chromatic and achromatic detection of C. lancifer by conspecifics, which is critical for pair formation during this restricted time window. We created varying reflectance spectra and calculated their conspicuousness under different light environments: maximal color contrast is obtained for the combination beetle signal–dusk light environment. This quantitative example of adaptation of body coloration to ambient light spectrum is the first insight into sensory drive through habitat-mediated transmission of a color signal in an insect species.
