Abstract:Jasmonic acid (JA) is thought to be involved in plant responses to cadmium (Cd) stress, but the underlying molecular mechanisms are poorly understood. Here, we show that Cd treatment rapidly induces the expression of genes promoting endogenous JA synthesis, and subsequently increases the JA concentration in Arabidopsis roots. Furthermore, exogenous methyl jasmonate (MeJA) alleviates Cd-generated chlorosis of new leaves by decreasing the Cd concentration in root cell sap and shoot, and decreasing the expression of the AtIRT1, AtHMA2 and AtHMA4 genes promoting Cd uptake and long-distance translocation, respectively. In contrast, mutation of a key JA synthesis gene, AtAOS, greatly enhances the expression of AtIRT1, AtHMA2 and AtHMA4, increases Cd concentration in both roots and shoots, and confers increased sensitivity to Cd. Exogenous MeJA recovers the enhanced Cd-sensitivity of the ataos mutant, but not of atcoi1, a JA receptor mutant. In addition, exogenous MeJA reduces NO levels in Cd-stressed Arabidopsis root tips. Taken together, our results suggest that Cd-induced JA acts via the JA signaling pathway and its effects on NO levels to positively restrict Cd accumulation and alleviates Cd toxicity in Arabidopsis via suppression of the expression of genes promoting Cd uptake and long-distance translocation.
Role of Jasmonic Acid in Improving Tolerance of Rapeseed ( Brassica Napus L.) to Cd Toxicity

Abstract:The well-known detrimental effects of cadmium (Cd) on plants are chloroplast destruction, photosynthetic pigment inhibition, imbalance of essential plant nutrients, and membrane damage. Jasmonic acid (JA) is an alleviator against different stresses such as salinity and drought. However, the functional attributes of JA in plants such as the interactive effects of JA application and Cd on rapeseed in response to heavy metal stress remain unclear. JA at 50 μmol/L was observed in literature to have senescence effects in plants. In the present study, 25 μmol/L JA is observed to be a “stress ameliorating molecule” by improving the tolerance of rapeseed plants to Cd toxicity. JA reduces the Cd uptake in the leaves, thereby reducing membrane damage and malondialdehyde content and increasing the essential nutrient uptake. Furthermore, JA shields the chloroplast against the damaging effects of Cd, thereby increasing gas exchange and photosynthetic pigments. Moreover, JA modulates the antioxidant enzyme activity to strengthen the internal defense system. Our results demonstrate the function of JA in alleviating Cd toxicity and its underlying mechanism. Moreover, JA attenuates the damage of Cd to plants. This study enriches our knowledge regarding the use of and protection provided by JA in Cd stress.
The Alleviation of Cadmium Toxicity in Oilseed Rape (brassica Napus) by the Application of Salicylic Acid

Abstract:To assess the potential role of salicylic acid (SA) in plants under cadmium (Cd) stress, a study was conducted on three different oilseed rape (Brassica napus) genotypes. Gas exchange parameters, photosynthetic pigments, antioxidant enzymes activities, mineral nutrients concentration, and ultrastructural analysis were carried out for assessment. Interestingly, cadmium treatment reduced gas exchange parameters, photosynthetic pigments, mineral elements [calcium (Ca), magnesium (Mg), and iron] and the activity of catalase (CAT) enzyme. Whereas, a pronounced increase was observed in malondialdehyde content and antioxidant enzymes, cadmium and zinc accumulation. Impressively, SA played its role as an alleviatory agent by reducing the damage to all parameters caused by Cd except internal CO2 concentration which was further decreased by SA. Even more, Cd and SA showed synergistic effects by increasing superoxide dismutase, peroxidases, ascorbate peroxidase, guaiacol peroxidase, glutathione reductase and decreasing CAT. Apart from affecting the enzyme activities of oilseed rape, both Cd and SA had the same inhibitory effects on Ca and Mg concentrations. Damage caused by Cd to chloroplast and other internal organelles were almost rectified by SA. Effective role of SA, in alleviating Cd toxicity, could be attributed to the SA-induced improvement of photosynthetic activities, enhancement of antioxidant enzymes activities, reduction in lipid peroxidation and Cd uptake. These findings reflect the possible role of SA as a potential inhibitor of cadmium toxicity by strengthening the internal immunity in oilseed rape.
Root Defense in Salicylic Acid-Altering Arabidopsis Plants in Responses to Cadmium Stress

Abstract:The regulatory role of salicylic acid (SA) has been extensively reported in plants subjected to cadmium (Cd) stress. However, the underlying mechanisms still remain to be elucidated. This study provided evidence that root biological processes played crucial roles in SA-altering Arabidopsis plant responses to Cd stress. The nahG (naphthalene hydroxylase G)-transformed plants (with a significantly reduced endogenous SA) displayed a stronger tolerance, while the mutant snc1 (suppressor of nonexpressor of PR gene, constitutive 1; with high endogenous SA) was more sensitive, compared with wild-type (WT) plants under Cd stress. Correspondingly, a stronger antioxidative defense and a weaker oxidative stress were detected in nahG roots, while a more severe oxidative damage occurred in snc1 roots. The expression of a set of redox-related genes was examined, and mostly presented a higher level in nahG roots. A higher content of total flavonoids was observed in nahG roots under Cd stress, which was positively correlated with the expression of flavonoid biosynthesis-related genes. The metal-chelating and -sequestrating genes presented higher expression levels in nahG than in other two genotypes following Cd exposure. Noticeably, the pectin methylesterase inhibitor-encoding genes displayed dramatically high basal expression levels in nahG roots, and further induced under Cd stress. Accordingly, the pectin methylesterase activity was the least in nahG under both control and Cd stress. These results suggest that nahG-conferred Cd tolerance is associated with the root antioxidation defense, flavonoid biosynthesis, metal chelation and sequestration, and cell wall modification.

Abstract:In higher plants, Zn nutritional imbalance can affect growth, physiology and response to stress, with effect variable depending on host–pathogen interaction. Mechanisms through which Zn operates are not yet well known. The hormone salicylic acid (SA) can affect plant ion uptake, transport and defence responses. Thus, in this study the impact of Zn imbalance and SA co-supply on severity of infection with the necrotrophic fungal pathogen B. cinerea or the biotroph G. cichoracearum was assessed in A. thaliana Col-0.Spectrophotometric assays for pigments and malondialdehyde (MDA) content as a marker of lipid peroxidation, plant defensin 1.2 gene expression by semi-quantitative PCR, callose visualization by fluorescence microscopy and diseases evaluation by macro- and microscopic observations were carried out.Zinc plant concentration varied with the supplied dose. In comparison with the control, Zn-deficit or Zn-excess led to reduced chlorophyll content and PDF 1.2 transcripts induction. In Zn-deficient plants, where MDA increased, also the susceptibility to B. cinerea increased, whereas MDA decreased in G. cichoracearum. Zinc excess increased susceptibility to both pathogens. Co-administration of SA positively affected MDA level, callose deposition, PDF 1.2 transcripts and plant response to the two pathogens.The increased susceptibility to B. cinerea in both Zn-deficient and Zn-excess plants could be related to lack of induction of PDF 1.2 transcripts; oxidative stress could explain higher susceptibility to the necrotroph and lower susceptibility to the biotroph in Zn-deficient plants. This research shows that an appropriate evaluation of Zn supply according to the prevalent stress factor is desirable for plants.

Abstract:AimsPhytoremediation is an emerging strategy for the removal of heavy metal contaminants. However, one of the prerequisite is to understand adequately plant resistant mechanisms. The present study was performed to assess the role of endogenous SA in plant response to Pb or Cd using wild-type (wt) Arabidopsis and its SA-accumulating mutant snc1, SA-reducing transgenic line nahG, SA signal-blocking npr1-1, and snc1/nahG (i.e. expression of nahG in snc1 plant) with a comparable level of SA to the wt.MethodsPlants were grown hydroponically in controlled conditions. For heavy metal exposure, Pb2+ or Cd2+ at final concentrations of 50 μM, 100 μM, and 150 μM, respectively, was added to the culture solution. Unless otherwise indicated, samples were harvested after 7 d of exposure, and used for analyses.ResultsCompared to the wt level, the high endogenous SA significantly potentiated Pb- and Cd-induced plant growth retardation, whereas SA deficiency decreased the growth inhibition, and SA signaling blockage also had some protective effect. The expression of nahG in snc1 plant mitigated effectively the growth inhibition. The SA-related mechanism was involved in redox homeostasis, photosynthetic process, and soluble matter accumulation.ConclusionsThese results suggest that Pb- or Cd-induced phytotoxicity in Arabidopsis was intensified by elevated endogenous SA, whereas ameliorated by reduced SA.

Abstract:Soil cadmium (Cd) pollution is escalating, necessitating effective remediation strategies. This study investigated the effects of exogenous jasmonic acid (JA) on Sedum alfredii Hance under Cd stress , aiming to enhance its phytoextraction efficiency. Initially, experiments were conducted to assess the impact of various concentrations of JA added to environments with Cd concentrations of 100, 300, and 500 μmol/L. The results determined that a concentration of 1 μmol/L JA was optimal. This concentration effectively mitigated the level of ROS products by enhancing the activity of antioxidant enzymes. Additionally, JA fostered Cd absorption and accumulation, while markedly improving plant biomass and photosynthetic performance. In further experiments, treatment with 1 μmol/L JA under 300 μmol/L Cd stress was performed and transcriptomic analysis unveiled a series of differentially expressed genes (DEGs) instrumental in the JA-mediated Cd stress response. These DEGs encompass not only pathways of JA biosynthesis and signaling but also genes encoding functions that influence antioxidant systems and photosynthesis, alongside genes pertinent to cell wall synthesis, and metal chelation and transport. This study highlights that JA treatment significantly enhances S. alfredii 's Cd tolerance and accumulation, offering a promising strategy for plant remediation and deepening our understanding of plant responses to heavy metal stress.

Abstract:Abstract Background The elemental defense hypothesis states a new defensive strategy that hyperaccumulators defense against herbivores or pathogens attacks by accumulating heavy metals. Brassica juncea has an excellent ability of cadmium (Cd) accumulation. However, the elemental defense effect and its regulation mechanism in B. juncea remain unclear. Results In this study, we profiled the elemental defense effect and the molecular regulatory mechanism in Cd-accumulated B. juncea after Alternaria brassicicola infection. B. juncea treated with 180 mg Kg − 1 DW CdCl 2 2.5H 2 O exhibited obvious elemental defense effect after 72 h of infection with A. brassicicola . The expression of some defense-related genes including BjNPR1 , BjPR12, BjPR2 , and stress-related miRNAs (miR156, miR397, miR398a, miR398b/c, miR408, miR395a, miR395b, miR396a, and miR396b) were remarkably elevated during elemental defense in B. juncea . Conclusions The results indicate that Cd-accumulated B. juncea may defend against pathogens by coordinating salicylic acid (SA) and jasmonic acid (JA) mediated systemic acquired resistance (SAR) and elemental defense in a synergistic joint effect. Furthermore, the expression of miRNAs related to heavy metal stress response and disease resistance may regulate the balance between pathogen defense and heavy metal stress-responsive in B. juncea . The findings provide experimental evidence for the elemental defense hypothesis in plants from the perspectives of phytohormones, defense-related genes, and miRNAs.

Abstract:Cadmium (Cd) is a nonessential element in plants and has adverse effects on the growth and development of plants. However, the molecular mechanisms of Cd phytotoxicity, tolerance and accumulation in hyperaccumulators Solanum nigrum L. has not been well understood. Here, physiology, transcriptome, and metabolome analyses were conducted to investigate the influence on the S. nigrum under 0, 25, 50, 75 and 100 μM Cd concentrations for 7 days. Pot experiments demonstrated that compared with the control, Cd treatment significantly inhibited the biomass, promoted the Cd accumulation and translocation, and disturbed the balance of mineral nutrient metabolism in S. nigrum , particularly at 100 μM Cd level. Moreover, the photosynthetic pigments contents were severely decreased, while the content of total protein, proline, malondialdehyde (MDA), H 2 O 2 , and antioxidant enzyme activities generally increased first and then slightly declined with increasing Cd concentrations, in both leaves and roots. Furthermore, combined with the previous transcriptomic data, numerous crucial coding-genes related to mineral nutrients and Cd ion transport, and the antioxidant enzymes biosynthesis were identified, and their expression pattern was regulated under different Cd stress. Simultaneously, metabolomic analyses revealed that Cd treatment significantly changed the expression level of many metabolites related to amino acid, lipid, carbohydrate, and nucleotide metabolism. Metabolic pathway analysis also showed that S. nigrum roots activated some differentially expressed metabolites (DEMs) involved in energy metabolism, which may enhance the energy supply for detoxification. Importantly, central common metabolism pathways of DEGs and DEMs, including the "TCA cycle", "glutathione metabolic pathway" and "glyoxylate and dicarboxylate metabolism" were screened using conjoint transcriptomics and metabolomics analysis. Our results provide some novel evidences on the physiological and molecular mechanisms of Cd tolerance in hyperaccumulator S. nigrum plants.

Abstract:To evaluate the role of endogenous SA in plant response to Cd stress,Arabidopsis wild type(Columbia)and its SA-altering mutants snc1 (suppressor of npr1-1, constitutive) with high SA level, nahG(tansgenic line)with low SA level and npr1-1(non-expressor of PR gene)with SA signaling blockage were used in this study. Results showed that a greater growth inhibition occurred in snc1,while a less inhibition was observed in nahG and npr1-1 plants. Although the anti-oxidative enzymes SOD and POD increased upon Cd exposure,they were insufficient to remove oxidative stress,especially in snc1 plants. The accumulations of soluble sugar and proline in the tested plants were positively related to their tolerance to Cd stress.
The Effects of the Endophytic Bacterium Pseudomonas fluorescens Sasm05 and IAA on the Plant Growth and Cadmium Uptake of Sedum alfredii Hance.

Abstract:Endophytic bacteria have received attention for their ability to promote plant growth and enhance phytoremediation, which may be attributed to their ability to produce indole-3-acetic acid (IAA). As a signal molecular, IAA plays a key role on the interaction of plant and its endomicrobes. However, the different effects that endophytic bacteria and IAA may have on plant growth and heavy metal uptake is not clear. In this study, the endophytic bacterium Pseudomonas fluorescens Sasm05 was isolated from the stem of the zinc (Zn)/cadmium (Cd) hyperaccumulator Sedum alfredii Hance. The effects of Sasm05 and exogenous IAA on plant growth, leaf chlorophyll concentration, leaf Mg2+ C-ATPase and Ca2+C-ATPase activity, cadmium (Cd) uptake and accumulation as well as the expression of metal transporter genes were compared in a hydroponic experiment with 10 mu M Cd. The results showed that after treatment with 1 mM IAA, the shoot biomass and chlorophyll concentration increased significantly, but the Cd uptake and accumulation by the plant was not obviously affected. Sasm05 inoculation dramatically increased plant biomass, Cd concentration, shoot chlorophyll concentration and enzyme activities, largely improved the relative expression of the three metal transporter families ZRT/IRT-like protein (ZIP), natural resistance associated macrophage protein (NRAMP) and heavy metal ATPase (HMA). Sasm05 stimulated the expression of the SaHMAs (SaHMA2, SaHMA3, and SaHMA4), which enhanced Cd root to shoot translocation, and upregulated SaZIP, especially SaIRT1, expression to increase Cd uptake. These results showed that although both exogenous IAA and Sasm05 inoculation can improve plant growth and photosynthesis, Sasm05 inoculation has a greater effect on Cd uptake and translocation, indicating that this endophytic bacterium might not only produce IAA to promote plant growth under Cd stress but also directly regulate the expression of putative key Cd uptake and transport genes to enhance Cd accumulation of plant.
The endophytic bacterium Sphingomonas SaMR12 alleviates Cd stress in oilseed rape through regulation of the GSH-AsA cycle and antioxidative enzymes

Abstract:Abstract Background Microbes isolated from hyperaccumulating plants have been reported to be effective in achieving higher phytoextraction efficiency. The plant growth-promoting bacteria (PGPB) SaMR12 from the cadmium (Cd)/zinc hyperaccumulator Sedum alfredii Hance could promote the growth of a non-host plant, oilseed rape, under Cd stress. However, the effect of SaMR12 on Brasscia juncea antioxidative response under Cd exposure was still unclear. Results A hydroponic experiment was conducted to study the effects of Sphingomonas SaMR12 on its non-host plant Brassica juncea (L.) Czern. under four different Cd treatments. The results showed that SaMR12 could colonize and aggregate in the roots and then move to the shoots. SaMR12 inoculation promoted plant growth by up to 71% in aboveground biomass and 81% in root biomass over that of the non-inoculated plants. SaMR12-inoculated plants significantly enhanced root Cd accumulation in the 10 and 20 μM Cd treatments, with 1.72- and 0.86-fold increases, respectively, over that of the non-inoculated plants. SaMR12 inoculation not only decreased shoot hydrogen peroxide (H 2 O 2 ) content by up to 38% and malondialdehyde (MDA) content by up to 60% but also reduced proline content by 7–30% in shoots and 17–32% in roots compared to the levels in non-inoculated plants. Additionally, SaMR12 inoculation promoted the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) and facilitated the relative gene expression levels of dehydroascorbate reductase ( DHAR ) and glutathione reductase ( GR ) involved in the glutathione (GSH)-ascorbic acid (AsA) cycle. Conclusions The results demonstrated that, under Cd stress, SaMR12 inoculation could activate the antioxidative response of B. juncea by decreasing the concentrations of H 2 O 2 , MDA and proline, increasing the activities of antioxidative enzymes, and regulating the GSH-AsA cycle. These results provide a theoretical foundation for the potential application of hyperaccumulator endophytic bacteria as remediating agents to improve heavy metal tolerance within non-host plant species, which could further improve phytoextraction efficiency. Abstract:Cd accumulation in plants not only affects the plant growth, but also further endangers human healthy by plant-derived food process. Brassica juncea L. has been appreciated as the potential Cd hyperaccumulator plant as it is suitable for bioremediation purposes. The previous studies revealed that B. juncea biomass and metabolism were severely affected by Cd stress. However, the relationship between Cd accumulation and micronutrient accumulation in B. juncea remains unknown. Cd shares chemical similarity to Zn, Fe, and Mn and can thus replace them in the ironic related groups of many proteins in plant cells. In the present study, the micronutrients Mn, Zn, Fe, and Cu contents and Cd contents in B. juncea were detected under gradient Cd treatment condition. The result showed that Zn, Mn and Cu accumulation in roots were significantly affected by Cd stress. The main metal and heavy metal transporter family candidates were then identified according to the phylogenetic analysis of related homologues in Arabidopsis thaliana, B. napus and B. juncea. We further investigated the expression level of BjNramp, BjHMA, BjCET and BjMTPs genes under 1 and 5 mu M Cd treatment condition by qRT-PCR assays. The result revealed that BjNramp1;4, BjNramp3;5, BjNramp3;10, BjNramp4;4, BjHMA3, and BjHMA4 might be involved in the uptake of Cd, Cu, Zn and Mn by the roots under low (1 mu M) Cd treatment. The present study is important to understand the relation between micronutrient and heavy metal uptake, and provides essential information for the phytoremediation on soil heavy metal contamination to coordinate the detoxification of heavy metal and micronutrient accumulation. Abstract:With the acceleration of industrialization, Cd pollution has emerged as a major threat to soil ecosystem health and food safety. Hyperaccumulating plants like Sedum alfredii Hance are considered to be used as part of an effective strategy for the ecological remediation of Cd polluted soils. This study delved deeply into the physiological, transcriptomic, and metabolomic responses of S. alfredii under cadmium (Cd) stress when treated with exogenous salicylic acid (SA). We found that SA notably enhanced the growth of S. alfredii and thereby increased absorption and accumulation of Cd, effectively alleviating the oxidative stress caused by Cd through upregulation of the antioxidant system. Transcriptomic and metabolomic data further unveiled the influence of SA on photosynthesis, antioxidant defensive mechanisms, and metal absorption enrichment pathways. Notably, the interactions between SA and other plant hormones, especially IAA and JA, played a central role in these processes. These findings offer us a comprehensive perspective on understanding how to enhance the growth and heavy metal absorption capabilities of hyperaccumulator plants by regulating plant hormones, providing invaluable strategies for future environmental remediation efforts.

Abstract:Metal hyperaccumulating plant species are an interesting example of natural selection and environmental adaptation but they may also be useful to developing new technologies of environmental monitoring and remediation. Noccaea caerulescens and Arabidopsis halleri are both Brassicaceae and are known metal hyperaccumulators. This study evaluated tolerance, uptake and translocation of zinc sulfide quantum dots by N. cearulescens and cadmium sulfide quantum dots by A. halleri in direct comparison with the non-hyperaccumulator, genetically similar T. perfoliatum and A. thaliana. Growth media were supplied with two different concentrations of metal in either salt (ZnSO4 and CdSO4) or nanoscale form (ZnS QDs and CdS QDs). After 30 days of exposure, the concentration of metals in the soil, roots and leaves was determined. Uptake and localization of the metal in both nanoscale and non-nanoscale form inside plant tissues was investigated by Environmental Scanning Electron Microscopy (ESEM) equipped with an X-ray probe. Specifically, the hyperaccumulators in comparison with the non-hyperaccumulators accumulate ionic and nanoscale Zn and Cd in the aerial parts with a BCF ratio of 45.9 for Zn ion, 49.6 for nanoscale Zn, 2.64 for Cd ion and 2.54 for nanoscale Cd. Results obtained with a differential extraction analytical procedure also showed that a significant fraction of nanoscale metals remained inside the plants in a form compatible with the retention of at least a partial initial structure. The molecular consequences of the hyperaccumulation of nanoscale materials are discussed considering data obtained with hyperaccumulation of ionic metal. This is the first report of conventional hyperaccumulating plants demonstrating an ability to hyperaccumulate also engineered nanomaterials (ENMs) and suggests a potential novel strategy for not only understanding plant-nanomaterial interactions but also for potential biomonitoring in the environment to avoid their entering into the food chains.
Differential responses to Cd stress induced by exogenous application of Cu, Zn or Ca in the medicinal plant Catharanthus roseus

Abstract:Cd(II) is one of the most widespread and toxic heavy metals and seriously threatens plant growth, furthermore negatively affecting human health. For survival from this metal stress, plants always fight with Cd(II) toxicity by themselves or using other external factors. The effects of second metals copper (Cu(II)), zinc (Zn(II)) and calcium (Ca(II)) on the Cd(II)-affected root morphology, Cd(II) translocation and metabolic responses in Catharanthus roseus were investigated under hydroponic conditions. We found that the Cd-stressed plants displayed the browning and rot root symptom, excess H2O2 content, lipid peroxidation and Cd(II) accumulation in plants. However, the supplement with second metals largely alleviated Cd-induced toxicity, including browning and rot roots, oxidative stress and internal Cd(II) accumulation. The amended effects at metabolic and transcriptional levels involved in different second metals share either common or divergent strategies. They commonly repressed Cd uptake and promoted Cd(II) translocation from roots to shoots with divergent mechanisms. High Zn(II) could activate MTs expression in roots, while Cu(II) or Ca(II) did not under Cd(II) stress condition. The presence of Ca(II) under Cd stress condition largely initiated occurrence of lateral roots. We then grouped a metabolic diagram integrating terpenoid indole alkaloid (TIA) accumulation and TIA pathway gene expression to elucidate the metabolic response of C. roseus to Cd(II) alone or combined with second metals. The treatment with 100 Cd(II) alone largely promoted accumulation of vinblastine, vindoline, catharanthine and loganin, whereas depressed or little changed the expression levels of genes detected here, compared to 0 Cd(II) control. In the presence of Cd(II), the supplement with second metals displayed specific effect on different alkaloid. Among them, the metal Ca(II) is especially beneficial for serpentine accumulation, Zn(II) mainly promoted tabersonine production. However, the addition of Cu(II) commonly depressed accumulation of most alkaloids detected here. Generally, we presented different mechanisms by which the second metals used to alleviate Cd (II) toxicity. This plant has potential application in phytoremediation of Cd(II), due to relatively substantial accumulation of biomass, as well as secondary metabolites TIAs used as pharmaceutical materials when facing Cd stress.

Abstract:Cadmium (Cd) is highly toxic to plants and limits crop growth and productivity; it also affects the uptake and distribution of nutrients, accelerates oxidative damage, and disrupts metabolic pathways in plants. Salicylic acid (SA) is an important stress-signaling molecule that has been reported to ameliorate the deleterious effects of Cd in many plant species. In this study, the ability of 100 µM SA application to alleviate the effects of 5 µM Cd stress was investigated in ErJiuNan1 (EJN1, an early-indica rice genotype) under hydroponic culture. The results showed that the addition of SA alleviated Cd toxicity by increasing the accumulation of nutrients (Mn, Fe), increasing the activity of antioxidative defense system (e.g., superoxide dismutase, peroxidase, and soluble protein), and decreasing lipid peroxidation. SA addition in Cd-stressed rice plants upregulated the expression of Cd stress tolerance genes (OsHMA3 and OsNRAMP5), which enhanced Cd tolerance, reduced the accumulation of Cd, and affected Mn accumulation. The expression of Fe-inducible genes (OsIRT1, OsNRAMP1, OsNAS3, and OsYSL15) was upregulated in response to Cd exposure but significantly down-regulated by exogenous SA treatment. Furthermore, co-application of Cd and SA upregulated the expression of the SA biosynthesis-associated genes OsICS1 and OsWRKY6. Thus, SA application might alleviate Cd toxicity by reversing the negative effects of plant growth stemming from Cd exposure and improve Cd tolerance by modulating the expression of uptake/transport-related genes.

Abstract:Salicylic acid (SA), as an enigmatic signalling molecule in plants, has been intensively studied to elucidate its role in defence against biotic and abiotic stresses. This review focuses on recent research on the role of the SA signalling pathway in regulating cadmium (Cd) tolerance in plants under various SA exposure methods, including pre-soaking, hydroponic exposure, and spraying. Pretreatment with appropriate levels of SA showed a mitigating effect on Cd damage, whereas an excessive dose of exogenous SA aggravated the toxic effects of Cd. SA signalling mechanisms are mainly associated with modification of reactive oxygen species (ROS) levels in plant tissues. Then, ROS, as second messengers, regulate a series of physiological and genetic adaptive responses, including remodelling cell wall construction, balancing the uptake of Cd and other ions, refining the antioxidant defence system, and regulating photosynthesis, glutathione synthesis and senescence. These findings together elucidate the expanding role of SA in phytotoxicology.

Abstract:Metal hyperaccumulation, in which plants store exceptional concentrations of metals in their shoots, is an unusual trait whose evolutionary and ecological significance has prompted extensive debate. Hyperaccumulator plants are usually found on metalliferous soils, and it has been proposed that hyperaccumulation provides a defense against herbivores and pathogens, an idea termed the 'elemental defense' hypothesis. We have investigated this hypothesis using the crucifer Thlaspi caerulescens, a hyperaccumulator of zinc, nickel, and cadmium, and the bacterial pathogen Pseudomonas syringae pv. maculicola (Psm). Using leaf inoculation assays, we have shown that hyperaccumulation of any of the three metals inhibits growth of Psm in planta. Metal concentrations in the bulk leaf and in the apoplast, through which the pathogen invades the leaf, were shown to be sufficient to account for the defensive effect by comparison with in vitro dose-response curves. Further, mutants of Psm with increased and decreased zinc tolerance created by transposon insertion had either enhanced or reduced ability, respectively, to grow in high-zinc plants, indicating that the metal affects the pathogen directly. Finally, we have shown that bacteria naturally colonizing T. caerulescens leaves at the site of a former lead-zinc mine have high zinc tolerance compared with bacteria isolated from non-accumulating plants, suggesting local adaptation to high metal. These results demonstrate that the disease resistance observed in metal-exposed T. caerulescens can be attributed to a direct effect of metal hyperaccumulation, which may thus be functionally analogous to the resistance conferred by antimicrobial metabolites in non-accumulating plants.

Abstract:Chelate induction of organic acids has been recognized to enhance metal uptake and translocation in plants, but the underlying mechanism remains unclear. In this study, seedlings of Salix variegata were hydroponically exposed to the combinations of Cd (0 and 50 μM) and three exogenous organic acids (100 μM of citric, tartaric, or malic acid). Plant biomass, antioxidant enzymes, non-protein thiol compounds (NPT) content, and the expression of candidate genes associated with Cd accumulation and tolerance were determined. Results showed that Cd significantly inhibited plant biomass but stimulated the activity of antioxidant enzymes in the roots and leaves, while the lipid peroxidation increased as well. Respective addition of three organic acids greatly enhanced plant resistance to oxidative stress and reduced the lipid peroxidation induced by Cd, with the effect of malic acid showing greatest. The addition of organic acids also significantly increased the content of glutathione in the root, further improving the antioxidant capacity and potential of phytochelatin biosynthesis. Moreover, Cd induced the expression level of candidate genes in roots of S. variegata. The addition of three organic acids not only promoted the expression of candidate genes but also drastically increased Cd accumulation in S. variegata. In summary, application of citric, tartaric, or malic acid alleviated Cd-imposed toxicity through the boost of enzymatic and non-enzymatic antioxidants and candidate gene expression, while their effects on Cd tolerance and accumulation of S. variegata differed.
