Abstract:Tumor accurate imaging can effectively guide tumor resection and accurate follow-up targeted therapy. The development of imaging-stable, safe, and metabolizable contrast agents is key to accurate tumor imaging. Herein, ultra-small and metabolizable dual-mode imaging probe Au/Gd@FA NCs is rationally engineered by a simple hydrothermal method to achieve accurate FL/MRI imaging of tumors. The probes exhibit ultra-small size (2.5–3.0 nm), near-infrared fluorescence (690 nm), high quantum yield (4.4%), and a better T1 nuclear magnetic signal compared to commercial MRI contrast agents. By modifying the folic acid (FA) molecules, the uptake and targeting of the probes are effectively improved, enabling specific fluorescence imaging of breast cancer. Au/Gd@FA NCs with good biosafety were found to be excreted in the feces after imaging without affecting the normal physiological metabolism of mice. Intracellular reactive oxygen species (ROS) increased significantly after incubation of Au/Gd@FA NCs with tumor cells under 660 nm laser irradiation, indicating that Au/Gd@FA NCs can promote intracellular ROS production and effectively induce cell apoptosis. Thus, metabolizable Au/Gd@FA NCs provide a potential candidate probe for multimodal imaging and tumor diagnosis in clinical basic research. Meanwhile, Au/Gd@FA NCs mediated excessive intracellular production of ROS that could help promote tumor cell death.

Abstract:We developed a facile strategy to obtain a new kind of mesoporous core-shell structured up-conversion nanoparticles (mUCNPs), composed of a NaYbF4:2%Er core and a mesoporous NaGdF4 shell. This mesoporous shell not only enhanced the up-conversion luminescence but also endowed many other functionalities of the nanoparticles such as drug delivery and bio-imaging capabilities. Moreover, after being conjugated with polyethylenimine (PEI) and folic acid (FA), core-shell mUCNPs exhibited good water dispersibility, enhanced drug delivery efficiency, and remarkable targeting ability to cancer cells. To certify the folate receptors (FR)-mediated targeted drug delivery, cell viability assay, cell up-conversion luminescence imaging and flow cytometry analysis were carried out. Furthermore, apart from the application for targeted drug delivery, the as-prepared core-shell mUCNPs could also be employed as the contrast agents for X-ray computed tomography (CT) and magnetic resonance (MR) imaging, because of the strong X-ray attenuation ability of Yb and high longitudinal molar relaxivity (r1) of Gd in the nanoparticles, providing the potential for simultaneously bio-imaging and cancer-targeting therapy.

Abstract:Detection of early malignant tumors remains clinically difficult; developing ultrasensitive imaging agents is therefore highly demanded. Owing to the unusual magnetic and optical properties associated with f-electrons, rare-earth elements are very suitable for creating functional materials potentially useful for tumor imaging. Nanometer-sized particles offer such a platform with which versatile unique properties of the rare-earth elements can be integrated. Yet the development of rare-earth nanoparticle-based tumor probes suitable for imaging tiny tumors in vivo remains difficult, which challenges not only the physical properties of the nanoparticles but also the rationality of the probe design. Here we report new approaches for size control synthesis of magnetic/upconversion fluorescent NaGdF4:Yb,Er nanocrystals and their applications for imaging tiny tumors in vivo. By independently varying F(-):Ln(3+) and Na(+):Ln(3+) ratios, the size and shape regulation mechanisms were investigated. By replacing the oleic acid ligand with PEG2000 bearing a maleimide group at one end and two phosphate groups at the other end, PEGylated NaGdF4:Yb,Er nanoparticles with optimized size and upconversion fluorescence were obtained. Accordingly, a dual-modality molecular tumor probe was prepared, as a proof of concept, by covalently attaching antitumor antibody to PEGylated NaGdF4:Yb,Er nanoparticles through a "click" reaction. Systematic investigations on tumor detections, through magnetic resonance imaging and upconversion fluorescence imaging, were carried out to image intraperitoneal tumors and subcutaneous tumors in vivo. Owing to the excellent properties of the molecular probes, tumors smaller than 2 mm was successfully imaged in vivo. In addition, pharmacokinetic studies on differently sized particles were performed to disclose the particle size dependent biodistributions and elimination pathways.

Abstract:This work reports the novel microwave-assisted solvothermal synthesis and structural, topographic, spectroscopic characterization of NaYF(4):Yb,Er upconversion nanoparticles (UCNPs) as well as their application in the labeling of HeLa cells. The nanoparticles were prepared in ethylene glycol, with rare earth acetates as precursor and NH(4)F and NaCl as the fluorine and sodium sources. X-ray diffraction, transmission electron microscopy, and luminescence spectroscopy were applied to characterize the nanoparticles. Experimental results showed that the microwave-assisted solvothermal method is an effective approach to create highly crystalline, strongly luminescent UCNPs at a lower temperature (160 °C) and within a significantly shortened reaction time (only 1 h) compared to the traditional methods. The effect of fluorine source on the optical properties of UCNPs was investigated by using NH(4)F, NH(4)HF(2), NaF, and 1-butyl-3-methylimidazolium tetrafluoroborate (BmimBF(4)) as different fluorine sources; NH(4)F proved to be the best one, making the luminescent intensity increase at least 2 orders of magnitude. The UCNPs with four different colors (green, yellow, orange, and cyan) were successfully obtained. After being modified with amino groups and coupled with CEA-8 antibody, the obtained nanoparticles were successfully applied in the specific fluorescent immunolabeling and imaging of HeLa cells to further verify their function as a marker in immunolabeling.

Abstract:Abstract Colon cancer is one of the world’s most deadly diseases. Because of its internal location, it is necessary to obtain faster and more efficient diagnostic tools for this organ site. In this context, we studied the development of new luminescent nanoprobes (LNPs) as an alternative diagnostic apparatus for detecting this disease. The nanoparticles examined herein are lanthanide-doped sodium yttrium fluoride (NaYF4:Ln) and have shown to be promising as investigative devices. However, significant problems with the use of LNPs are the lack of biocompatibility and the targeting of the system to tumor regions. One of the strategies to bypass these problems is to increase of the particle lipophilicity modifying their surfaces with organic compounds that present high similarity to the biological system. In this work, we synthesized six new materials for use in bioimaging techniques obtained from the combination of nanoparticles of NaYF4:5%Eu with organic aromatic compounds covalently bonded. The materials were characterized structurally and morphologically using XRD and TEM, techniques, which showed the identification of the crystallographic phase β-NaYF4:5%Eu and its nanometric size (particles smaller than 50 nm). The conjugation process was confirmed by FT-IR spectra analysis and from the TGA profile. Excitation and emission spectra allowed the evaluation of the optical properties of the synthesized compounds. The interaction and cellular uptake was confirmed when HT-29 colon cancer cells were exposed to LNPs, indicating that the developed system has promising applications in bioimaging procedures. Abstract:5nm intense green emission NaYF4:Yb/Er upconversion nanoparticles (UCNPs) with pure β phase was synthesized with a simple "green" strategy for the first time. Traditional organic solvothermal method is often applied to prepare the high-quality and uniform UCNPs, but the preparation of lanthanide-oleate complexes is laborious as heating and multistep post-treatment for purification are often required. The water-alcohols solvothermal method is environmentally friendly, but the fabricated UCNPs have big size, poor biocompatibility and high cytotoxicity, which limited their application for cell imaging. Herein, NaYF4:Yb/Er UCNPs were prepared with rare-earth nitrates RE(NO3)3 (RE=Y0.80 Yb0.18 Er0.02) as precursors and diethylene glycol (DEG)/ethylene glycol (EG)/water as the solvent. A facile green solvothermal method with the temperature being controlled at 300°C was developed. The as-prepared NaYF4:Yb/Er UCNPs were characterized and were found to have enhanced UC emission and controllable particle size. The as-prepared UCNPs were further functionalized via folic acid coating for the targeted imaging and improved bio- compatibility. It was made the UCNPs potential for upconversion bioimaging of living cells by the strong upconversion luminescence, the excellent biocompatibility, and the super-small size. The good colloidal stability and low cell cytotoxicity of the as-prepared UCNPs and the developed synthesis protocol might advance both the fields of UCNPs and biomolecule-based nanotechnology for future studies.

Abstract:New core-shell structured NaYF(4):Yb(3+),Tm(3+)@Fe(x)O(y) nanocrystals, with 20 nm Yb(3+),Tm(3+)-co-doped NaYF(4) nanocrystals as a core and 5 nm Fe(x)O(y) nanocrystals as a shell, have been synthesized and characterized by TEM and XRD analysis. These core-shell nanocrystals exhibit excellent near-infrared upconversion luminescence (UCL) emission at 800 nm under excitation by a continuous-wave 980 nm laser and superparamagnetic properties with a saturation magnetization (Ms) of ∼12 emu/g. Water-soluble nanocrystals were obtained by surface ligand exchange of oleic acid-coated precursor nanocrystals, and their internalization within living cells has been investigated by laser scanning UCL microscopy. Furthermore, the obtained core-shell nanocrystals have been applied in dual-modality T(2)-enhanced magnetic resonance (MR) and UCL imaging in vivo of the lymphatic system. Moreover, the toxicity of NaYF(4):Yb(3+),Tm(3+)@Fe(x)O(y) nanocrystals has also been evaluated by MTT assay, IC50 values, and histological analysis of lymphatic node sections.

Abstract:Lanthanide-doped fluoride up-converting nanoparticles (UCNPs) represent the new class of imaging contrast agents which hold great potential for overcoming existing problems associated with traditionally used dyes, proteins and quantum dots. In this study, a new kind of hybrid NaYF4:Yb,Er/PLGA nanoparticles for efficient biolabeling were prepared through one-pot solvothermal synthesis route. Morphological and structural characteristics of the as-designed particles were obtained using X-ray powder diffraction (XRPD), scanning and transmission electron microscopy (SEM/TEM), energy dispersive spectroscopy (EDS), Fourier transform infrared (FTIR) and photoluminescence (PL) spectroscopy, while their cytotoxicity as well as up-conversion (UC) labeling capability were tested in vitro toward human gingival cells (HGC) and oral squamous cell carcinoma (OSCC). The results revealed coexistence of the cubic (Fm-3m) and hexagonal (P63/m) phase in spherical and irregularly shaped nanoparticles, respectively. PLGA [Poly(lactic-co-glycolic acid)] ligands attached at the surface of UCNPs particles provide their enhanced cellular uptake and enable high-quality cells imaging through a near-infrared (NIR) laser scanning microscopy (λex = 980 nm). Moreover, the fact that NaYF4:Yb,Er/PLGA UCNPs show low cytotoxicity against HGC over the whole concentration range (10-50 μg/mL) while a dose dependent viability of OSCC is obtained indicates that these might be a promising candidates for targeted cancer cell therapy.

Abstract:Background Gastric cancer is 2th most common cancer in China, and is still the second most common cause of cancer-related death in the world. Successful development of safe and effective nanoprobes for in vivo gastric cancer targeting imaging is a big challenge. This study is aimed to develop folic acid (FA)-conjugated silica coated gold nanoclusters (AuNCs) for targeted dual-modal fluorescent and X-ray computed tomography imaging (CT) of in vivo gastric cancer cells. Method AuNCs were prepared, silica was coated on the surface of AuNCs, then folic acid was covalently anchored on the surface of AuNCs, resultant FA-conjugated AuNCs@SiO 2 nanoprobes were investigated their cytotoxicity by MTT method, and their targeted ability to FR(+) MGC803 cells and FR(−) GES-1 cells. Nude mice model loaded with MGC803 cells were prepared, prepared nanoprobes were injected into nude mice via tail vein, and then were imaged by fluorescent and X-ray computed tomography (CT) imaging. Results FA-conjugated AuNCs@SiO 2 nanoprobes exhibited good biocompatibility, and could target actively the FR(+) MGC-803 cells and in vivo gastric cancer tissues with 5 mm in diameter in nude mice models, exhibited excellent red emitting fluorescence imaging and CT imaging. Conclusion The high-performance FA-conjugated AuNCs@SiO 2 nanoprobes can target in vivo gastric cancer cells, can be used for fluorescent and CT dual-mode imaging, and may own great potential in applications such as targeted dual-mode imaging of in vivo early gastric cancer and other tumors with FR positive expression in near future.

Abstract:Nanoparticulate contrast agents have attracted a great deal of attention along with the rapid development of modern medicine. Here, a binary contrast agent based on PAA modified BaYbF5:Tm nanoparticles for direct visualization of gastrointestinal (GI) tract has been designed and developed via a one-pot solvothermal route. By taking advantages of excellent colloidal stability, low cytotoxicity, and neglectable hemolysis of these well-designed nanoparticles, their feasibility as a multi-modal contrast agent for GI tract was intensively investigated. Significant enhancement of contrast efficacy relative to clinical barium meal and iodine-based contrast agent was evaluated via X-ray imaging and CT imaging in vivo. By doping Tm(3+) ions into these nanoprobes, in vivo NIR-NIR imaging was then demonstrated. Unlike some invasive imaging modalities, non-invasive imaging strategy including X-ray imaging, CT imaging, and UCL imaging for GI tract could extremely reduce the painlessness to patients, effectively facilitate imaging procedure, as well as rationality economize diagnostic time. Critical to clinical applications, long-term toxicity of our contrast agent was additionally investigated in detail, indicating their overall safety. Based on our results, PAA-BaYbF5:Tm nanoparticles were the excellent multi-modal contrast agent to integrate X-ray imaging, CT imaging, and UCL imaging for direct visualization of GI tract with low systemic toxicity.

Abstract:A new facile approach, namely chemical-assisted sol-gel growth (CASGG), was successfully developed to induce the formation of fine CaF2:Yb3+,Tm3+ nanocrystals within the pore channels of mesoporous silica (mSiO(2)) nanoparticles. A series of upconversion photoluminescent crystalline CaF2:Yb3+,Tm3+@mSiO(2) nanospheres with controlled diameters from similar to 65 nm to similar to 290 nm were fabricated. All nanospheres presented sound cyto-compatibility and unique ratiometric spectral monitoring functionalities for drug release kinetics. The nanospheres with smallest dimension (UCNP-2.5, similar to 65 nm) induced the most sustained DOX release kinetics. More importantly, the in vitro study demonstrated that the DOX loaded UCNP-2.5 nanospheres presented the strongest anti-cancer efficacy to MCF-7 human breast cancer cells due to its stronger penetration ability to cell nuclei due to the size effect.
Upconversion NaYF4:Yb3+/Er3+@silica-TPGS Bio-Nano Complexes: Synthesis, Characterization, and In Vitro Tests for Labeling Cancer Cells.

Abstract:Fluorescence imaging is an important technique used for early diagnosis and effective treatment of some incurable diseases including cancer. Herein, we report novel NaYF4:Yb3+/Er3+@silica-TPGS bio-nano complexes for labeling cancer cells. The NaYF4:Yb3+/Er3+ nanoparticles have been successfully synthesized via a hydrothermal route, further coated with a silica shell, and functionalized with d-α-tocopheryl polyethylene glycol 1000 succinate (TPGS). The experimental results indicate that NaYF4:Yb3+/Er3+@silica-TPGS emits stronger upconversion luminescence than NaYF4:Yb3+/Er3+ under an excitation of 980 nm. More significantly, the NaYF4:Yb3+/Er3+@silica-TPGS bio-nano complexes could strongly label MCF-7 breast cancer cells for in vitro experiments detected by a fluorescence microscope. On the other hand, the complex could not typically probe healthy cells, which are HEK-293A human embryonic kidney cells, under the same experimental conditions. Because of their strong upconversion luminescence, good dispersibility, and biocompatibility, NaYF4:Yb3+/Er3+@silica-TPGS bio-nano complexes can be a promising candidate/probe for biomedical labeling and diagnostics.

Abstract:Molecular imaging modalities provide a wealth of information that is highly complementary and rarely redundant. To combine the advantages of molecular imaging techniques, F-18-labeled Cd3+/Yb3+/Er3+ co-doped NaYF4 nanophosphors (NPs) simultaneously possessing with radioactivity, magnetic, and upconversion luminescent properties have been fabricated for multimodality positron emission tomography (PET), magnetic resonance imaging (MRI), and laser scanning upconversion luminescence (UCL) imaging. Hydrophilic citrate-capped NaY0.2Gd0.6Yb0.18Er0.02F4 nanophosphors (cit-NPs) were obtained from hydrophobic oleic acid (OA)-coated nanoparticles (OA-NPs) through a process of ligand exchange of OA with citrate, and were found to be monodisperse with an average size of 22 x 19 nm. The obtained hexagonal cit-NPs show intense UCL emission in the visible region and paramagnetic longitudinal relaxivity (r(1) = 0.405 s(-1) -(mM)(-1)). Through a facile inorganic reaction based on the strong binding between Y3+ and F-, F-18-labeled NPs have been fabricated in high yield. The use of cit-NPs as a multimodal probe has been further explored for T-1-weighted MR and PET imaging in vivo and UCL imaging of living cells and tissue slides. The results indicate that F-18-labeled NaY0.2Gd0.5Yb0.18Er0.02 is a potential candidate as a multimodal nanoprobe for ultra-sensitive molecular imaging from the cellular scale to whole-body evaluation. Sub-10 nm hexagonal lanthanide-doped NaLuF 4 upconversion nanocrystals for sensitive bioimaging in vivo

Abstract:By thermal decomposition in the presence only of oleylamine, sub-10 nm hexagonal NaLuF4-based nanocrystals codoped with Gd3+, Yb3+, and Er3+ (or Tm3+) have been successfully synthesized. Sub-10 nm beta-NaLuF4: 24 mol % Gd3+, 20 mol % Yb3+, 1 mol % Tm3+ nanocrystals display bright upconversion luminescence (UCL) with a quantum yield of 0.47 +/- 0.06% under continuous-wave excitation at 980 nm. Furthermore, through the use of beta-NaLuF4: Gd3+,Yb3+,Tm3+ nanocrystals as a luminescent label, the detection limit of <50 nanocrystal-labeled cells was achieved for whole-body photoluminescent imaging of a small animal (mouse), and high-contrast UCL imaging of a whole-body black mouse with a penetration depth of similar to 2 cm was achieved.
Shape, Size, and Phase-Controlled Rare-Earth Fluoride Nanocrystals with Optical Up-Conversion Properties

Abstract:High-quality rare-earth fluorides, alpha-NaMF(4) (M=Dy, Ho, Er, Tm, Y, Yb, and Lu) nanocrystals and beta-NaMF(4) (M=Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Y, Yb, and Lu) nanoarrays, have been synthesized by using oleic acid as a stabilizing agent through a facile hydrothermal method at 130-230 degrees C. The phase, shape, and size of the products are varied by careful control of synthetic conditions, including hydrothermal temperature and time, and the amounts of reactants and solvents. Tuning the hydrothermal temperature, time, and the amount of NaOH can cause the transformation from the cubic alpha-NaMF(4) to hexagonal phase beta-NaMF(4). Upon adjustment of the amount of NaOH, NaF, M(3+), and ethanol, the morphologies for the beta-NaMF(4) nanoarrays can range from tube, rod, wire, and zigzagged rod, to flower-patterned disk. Simultaneously, the size of the rare-earth fluoride crystals is variable from 5 nm to several micrometers. A combination of "diffusion-controlled growth" and the "organic-inorganic interface effect" is proposed to understand the formation of the nanocrystals. An ideal "1D growth" of rare-earth fluorides is preferred at high temperatures and high ethanol contents, from which the tube- and rodlike nanoarrays with high aspect ratio are obtained. In contrast, the disklike beta-NaMF(4) nanoarrays with low aspect ratios are produced by decreasing the ethanol content or prolonging the reaction time, an effect probably caused by "1D/2D ripening". Multicolor up-conversion fluorescence is also successfully realized in the Yb(3+)/Er(3+) (green, red) and Yb(3+)/Tm(3+) (blue) co-doped alpha-NaYF(4) nanocrystals and beta-NaYF(4) nanoarrays by excitation in the NIR region (980 nm).
Multimodal-luminescence Core-Shell Nanocomposites for Targeted Imaging of Tumor Cells.

Abstract:Uniform silica-coated NaYF(4): 20 mol % Yb, 2 mol % Er nanocomposites with good dispersibility, containing organic dye incorporated in the silica shell and folic acid conjugated on the surface of the shell, were prepared and characterized. The core-shell nanocomposites are 20-22 nm in size, water soluble, and buffer stable, with good photostability and biocompatibility. Folic acid (FA) offers a means of targeting human cells that greatly overexpress the folate receptor (FR). By the use of confocal microscopy and quantitative flow cytometry analysis, we demonstrate the receptor-mediated delivery of FA-conjugated nanocomposites targeting FR-positive cell lines, such as KB cells. The receptor-mediated targeting was confirmed by a comparison with the uptake of these nanocomposites in FR-negative cell lines, such as MCF-7. These results show that the silica-coated upconverting nanophosphor (UCNP) nanocomposites prepared by our strategy can potentially be useful as multimodal bioimaging agents.

Abstract:Nanocrystalline infrared-to-visible up-conversion phosphors, ytterbium and erbium co-doped sodium yttrium fluoride, were synthesized. Spherical particles with narrow size distribution were prepared by a co-precipitation method in the presence of ethylenediaminetetraacetic acid (EDTA). Particles of controlled size in the range of 37 to 166 nm diameter were obtained by adjusting the molar ratio of EDTA to total lanthanides. Although the as-prepared nanoparticles emit very weak up-conversion fluorescence when excited by infrared light, the emission was enhanced by up to 40-fold after they were annealed at temperatures between 400 and 600 degreesC. By comparison with the bulk phosphor, the luminescence efficiency of the nanoparticle was estimated to be 1%. Factors affecting the particle size and their up-conversion fluorescence intensity were investigated by various microscopic and spectroscopic techniques. Preliminary results demonstrated the nanoparticles as promising up-converting fluorescence labels in the detection of biological interactions.

Abstract:As a matrix material, NaYbF4 has attracted more and more attention due to its large absorption cross section and high energy transfer efficiency. In this study, lanthanide doped NaYbF4 upconversion (UC) nano-/micro-crystalline was carefully synthesized by hydrothermal method. The influence of kinds of external conditions, including reaction time, initial reaction solution pH value, organic solvents codoping, and Na+ concentration on crystalline phase, size, morphology and especially luminescent property are systematically studied and the possible formation mechanism based on PH values is emphatically proposed. The products of NaYbF4 with various morphologies range from nanospheres, microprisms, microdisks, hexagonal crown-capped shapes, hexagonal clam-like shapes and hexagonal flower-like shapes were successfully prepared. The results show that the up-conversion luminescence property is positively correlated with the slenderness ratio. Solvents codoping and Na+ concentration can greatly improve the luminescence property, especially for red emission. Impressively, the emission colors and intensities might be finely tuned through finely tuning the PH values, solvents co-doping and Na+ dosage. Furthermore, this study will be expected to contribute to the synthesis of various morphologies and improving the UC intensity of lanthanide doped crystals.
