Abstract:The spherical hydroxyapatite nanoparticles (nHAP) were synthesized by using surfactant hexadecyl (cetyl)trimethyl ammonium bromide(CTAB) as surface modifier and stabilizer. The physical-chemical properties of nHAP were charactered using TEM/SAED, XRD, and contact angle measurements. The different effects on growth and osteogenic differentiation of bone marrow derived mesenchymal stem cells (BMSCs) between the nHAP particles and the conventional HAP crystallites (cHAP) were studied. The results showed that the nHAP particles were more uniformed in shape and size and therefore, its thin films presented smoother and better-formed surface. BMSCs seeded on nHAP thin films showed enhanced attachment and proliferation, increased osteogenic marker expression such as alkaline phosphates (ALP) and osteocalcin (OC) compared with those on the cHAP thin films.

Abstract:Silk sericin, a water-soluble glue-like protein, is extensively used as a biomaterial due to its biocompatibility, hydrophilicity, biodegradability, and adequate resource. In addition, hydroxyapatite-based drug carriers are functionally efficient for drug or gene delivery due to their biodegradability, biocompatibility and easy metabolism in vivo. Herein, for the first time, this study used sericin, from a wild silkworm called Antheraea pernyi (A. pernyi), as a template to nucleate hydroxylapatite (HAp) nano-needles and form porous sericin-HAp nanocomposite microspheres as an anticancer drug carrier. Specifically, A. pernyi sericin (AS) was incubated in 1.5× simulated body fluid to induce the formation of porous AS/HAp microspheres in situ. Doxorubicin (DOX) loading and release assays proved that the microspheres exhibited pH-dependent controlled and sustained release of DOX. In particular, the microspheres can selectively release DOX at a higher rate at the acidic conditions typical for tumor microenvironment than at the physiological conditions typical for normal tissues, which will potentially reduce the side effect of the cancer drugs in normal tissues. Cancer cell toxicity assay, cancer cell imaging and intracellular DOX distribution assay provided further evidence to support the pH-dependent controlled and sustained release of DOX to cancer cells from the microspheres. Our work has demonstrated a biomimetic strategy for the design and synthesis of silk protein-based drug carriers that can be potentially employed in drug delivery and regenerative medicine.

Abstract:Calcium phosphate biomaterials are very promising for various biomedical applications owing to their excellent biocompatibility and biodegradability. Calcium phosphate nanostructured materials with a porous and hollow structure are excellent drug carriers due to their advantages such as high biocompatibility, large specific surface area, nanosized channels for drug loading and release, high drug loading capacity and pH-responsive drug release behavior. In this work, porous hollow microspheres of amorphous calcium phosphate have been successfully prepared by the microwave-assisted hydrothermal method using adenosine triphosphate disodium salt, CaCl2 and soybean lecithin in aqueous solution. This preparation method is facile, rapid, energy-saving and environment friendly. The effects of microwave hydrothermal temperature and concentrations of the reactants on the morphology and structure of the product were investigated. The as-prepared samples were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray powder diffraction (XRD) and Fourier-transform infrared (FTIR) spectroscopy. The as-prepared porous hollow microspheres of amorphous calcium phosphate are efficient for drug loading and release, and the drug delivery system shows a pH-responsive drug release behavior and high ability to damage tumor cells. Thus, the as-prepared porous hollow microspheres of amorphous calcium phosphate are promising for the applications in various biomedical fields.

Abstract:Ultralong hydroxyapatite (HAP) nanowires are promising for various biomedical applications owing to their chemical similarity to the inorganic constituent of bone, high biocompatibility, good flexibility, excellent mechanical properties, etc. However, it is still challenging to control the formation of ultralong HAP nanowires because of the presence of free PO43− ions in the reaction system containing the inorganic phosphate source. In addition, it takes a long period of time (usually tens of hours) for the synthetic process of ultralong HAP nanowires. Herein, for the first time, we have developed an eco-friendly calcium oleate precursor microwave hydrothermal method using biocompatible adenosine 5′-triphosphate (ATP) as a bio-phosphorus source and water as the only solvent for the rapid synthesis of ultralong HAP nanowires. The controllable hydrolysis of ATP can avoid the premature formation of calcium phosphate nuclei and uncontrollable crystal growth. Microwave heating can significantly shorten the synthetic time from tens of hours required by the traditional heating to 1 h, thus achieving high efficiency, energy saving and low cost. The as-prepared ultralong HAP nanowires with high flexibility have lengths of several hundred micrometers and diameters of 10~20 nm, and they usually self-assemble into nanowire bundles along their longitudinal direction. The as-prepared ultralong HAP nanowire/chitosan porous scaffold has excellent bioactivity, good biodegradation and cytocompatibility owing to the bioactive adenosine adsorbed on the surface of ultralong HAP nanowires. It is expected that ultralong HAP nanowires will be promising for various applications in the biomedical fields, such as bone defect repair, skin wound healing, and as a drug nanocarrier.

A highly linear voltage-to-time converter with variable conversion gain for time-based ADCs

Abstract:This paper presents a linear voltage-to-time converter (VTC) in 40nm CMOS technology. An inverter-based amplifier is utilized to improve the linearity of the proposed VTC. A digital controlled switched charge pump is used to generate four different current so that this VTC can operate with variable conversion gain. The VTC is designed for time-based ADCs. The simulation results shows that SFDR of the VTC can be greater than 60dB and THD can be lower than −54dB for all working modes. The VTC's conversion gain is above 2ns/V in all operating modes.

Abstract:In this paper, we report that hydroxyapatite (HAP) nanocrystals with various shape and size have been successfully prepared by a dual template-assisted hydrothermal synthesis approach in isopropanol/water mixed solvents. Herein, we chose 4-aminobenzenesulfonic acid (ABSA) and polyethylene glycol 4000 (PEG-4000) as structure-directed templates, respectively. By gradually changing the mass ratios (R) of PEG-4000 to ABSA, the obtained HAP samples can undergo distinct morphological evolution accordingly. A class of unique sheet-like HAP sample could be formed when only using ABSA as template. Moreover, when the R value was changed to 1/4, uniform and high-yielding HAP plate-shaped structure can be obtained. When only using PEG-4000 as a template, almost novel and wedge-shaped HAP samples can be obtained. The results demonstrated that the obtained plate-like HAP sample has single-crystalline behavior and uniform distribution in both size and shape. Additionally, we perform and evaluate the bioactivity of HAP in simulated body fluid (SBF), indicating that the two obtained HAP samples can possess apparently improved bioactivity compared to previous literature reports, and bone-like apatite could be readily formed on the HAP surface through different soaking periods in SBF. Notably, for the first time, a novel method for evaluating the in vitro bioactivity of HAP has been proposed. Therefore, the presented synthetic route for HAP is well-controlled and facilitated, which could be extended to the preparation of other biomaterials with specific morphologies and architectures.

Abstract:The use of polymer particles as organic templates for producing inorganic materials is an intriguing approach, because it offers the synthetic feasibility of organic-inorganic hybrid materials which may promise a variety of interesting applications. Here, inspired by mussel-adhesion phenomena in nature, we combined the use of the active surface of polydopamine (PDA) with biomimetic mineralization of hydroxyapatite (HAP) to synthesize a novel HAP-based, organic-inorganic hybrid material. Monodisperse micro-sized polystyrene (PS) particles were first prepared as inert polymer templates via dispersion polymerization, which, subsequently, were coated with PDA through the self-polymerization of dopamine in a weakly alkaline aqueous environment (pH = 8.5). This chemical modification activated the inert surface of PS particles to become bioactive. Thus, these synthesized PS/PDA composite particles were ideal templates for the biomimetic mineralization of HAP when they were incubated in a simulated body fluid (SBF) solution in aid of the biocompatible PDA layer. During the PDA-assisted biomimetic mineralization, the PDA layer interacted with the mineral ions introduced, and anchored the mineralization of HAP exclusively onto the surfaces of the composite particles. The detailed structures and morphologies of HAP-based hybrid materials (PS/PDA/HAP hybrid materials) were characterized. These HAP-based hybrid materials did not show a significant in vitro cytotoxicity against HeLa cells, indicating their potential biocapacity for drug delivery, protein and gene transfer, and as coating materials for bioimplants or as the injectable scaffold materials in bone tissue engineering for defect filling and regeneration.

Abstract:Hydroxyapatite (HAp), a major inorganic and essential component of normal bone and teeth, is a promising biomaterial due to its excellent biocompatibility, bioactivity, and osteoconductivity. Therefore, synthetic HAp has been widely used as a bone substitute, cell carrier, and delivery carrier of therapeutic genes or drugs. Mesoporous materials have attracted considerable attention due to their relatively high surface area, large pore volume, high porosity, and tunable pore size. Recently, mesoporous HAp has also been successfully synthesized by the traditional template-based process and has been demonstrated to possess better drug-loading and release efficiencies than traditional HAp. It is widely accepted that cell adhesion and most cellular activities, including spreading, migration, proliferation, gene expression, surface antigen display, and cytoskeletal functioning, are sensitive to the topography and molecular composition of the matrix. The native extracellular matrix is a porous, nanofibrous structure. The major focus of this study is the fabrication of porous hydroxyapatite-CaO composite nanofibers (p-HApFs) and the investigation of its drug-release property. In this study, nanofibers were prepared by the sol-gel route and an electrospinning technique to mimic the three-dimensional structure of the natural extracellular matrix. We analyzed the components of fibers using X-ray diffraction and determined the morphology of fibers using scanning and transmission electron microscopy. The average diameter of the nanofibers was approximately 461 ± 186 nm. The N2 adsorption–desorption isotherms were type IV isotherms. Moreover, p-HApFs had better drug-loading efficiency and could retard the burst release of tetracycline and maintain antibacterial activity for a period of 7 days. Hence, p-HApFs have the potential to become a new bone graft material.

Abstract:Controlled synthesis of hierarchical hydroxyapatite materials is a hot research topic because of the excellent biocompatibility and bioactivity of the materials. In this study, flower-like hydroxyapatite spheres (FHAPS) were facile synthesized in one pot using Al(OH)(3) as a structure-directing agent. The prepared FHAPS comprised nanosheets possessing a uniform diameter of approximately 4 mm. Notably, the FHAPS can be degraded in solutions with a pH of 5.5 for 144 h or incubated with cells for 48 h. In addition, the FHAPS have rough surfaces, which exhibit high loading contents for the anticancer drug doxorubicin (DOX, 9.1%) and siRNA (2.0%). Thus, the FHAPS can effectively deliver DOX into drug-resistant breast cancer cells to exert an excellent killing effect compared with free DOX and transfect siRNA into tumor cells to interfere with the expression of the target protein. Taken together, this work successfully prepared FHAPS via a convenient synthesis route that shows high delivery efficiency for anticancer drugs and siRNA. (C) 2020 Elsevier Inc. All rights reserved.

Abstract:A suitable microenvironment provided by artificial granule scaffold plays a critical regulatory role in bone repairing progression. Till now, it is still a challenge to prepare large hydroxyapatite granules (millimetre level) which provide beneficial physical and chemical stimulation for bone reconstruction. Herein, we developed a facile synthetic strategy for synthesizing hydroxyapatite granules by a repaid gelling method. Also, the satisfying drug-releasing behavior of these granules further proves their potential prospect for bone tissue engineering as bone filler.

Abstract:This paper reports a biomolecule-assisted hydrothermal method to synthesize hydroxyapatite (HAP) nanocrystals. The as-synthesized samples were characterized by X-ray diffraction (XRD), Raman, transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), and Fourier transform infrared (FT-IR) spectroscopy. The data showed that the growth process of the HAP nanocrystals was composed of nucleation and growth processes. HAP nanocrystals can inhibit the proliferation of human HeLa cells in dose- and time-dependent manners, suggesting the promising potential of HAP nanocrystals in cancer therapy. Also, HAP nanocrystals own excellent mechanical properties, suggesting promising applications in dental restoration. This approach opens up vast opportunities for the synthesis and potential applications of nanostructures in the biomedical field.

Abstract:Hydroxyapatite (HA) is promising in various biomedical applications owing to its similar chemical composition, structure and properties to the inorganic component in natural hard tissues. Herein, we report a DNA-templated microwave-assisted hydrothermal strategy for the preparation of HA nanostructured materials. As a kind of natural biomacromolecule, DNA molecules open up a new way to the synthesis of HA nanostructured materials with well-defined structures and morphologies. The HA nanostructured materials with a nanosheet-assembled hierarchical structure and a HA nanorod ordered structure are successfully prepared. The important roles of DNA molecules and pH values in the formation of HA nanostructured materials are investigated, and a possible formation mechanism is proposed. The as-prepared HA nanostructured materials exhibit a relatively high adsorption ability for chicken immunoglobulin Y (IgY) protein and a sustained protein release behavior. The as-prepared HA nanostructured materials after loading the IgY protein show a high antimicrobial activity. Thus, the HA nanostructured materials prepared by the DNA-templated microwave hydrothermal method are promising for the applications in various areas such as the prevention and treatment of dental caries.

Abstract:We report here a facile strategy to fabricate three-dimensional (3D) hydroxyapatite (HA) architectures with well-defined long continuous interconnected pores by using electrospinning and biomimetic mineralization. To this end, a polymeric nanofiber (NF) scaffold with well-defined architecture was fabricated by electrospinning, and bone morphogenetic protein 2 (BMP2) was then adsorbed onto the chemically modified NFs through bio-conjugation. The 3D nanoporous HA architecture was finally fabricated by biomimetic mineralization of the NF–BMP2 hybrid in simulated body fluids and subsequent dissolution of NFs in hexafluoroisopropanol. The formation of NF–BMP2 hybrid was identified by confocal laser scanning microscopy analysis. The crystal structure of HA crystals formed on NFs was examined by X-ray diffraction. The chemical composition and interconnected porous structure of the created 3D HA architectures were measured by X-ray photoelectron spectroscopy, focused ion beam scanning electron microscopy, and transmission electron microscopy, respectively. This bottom-up strategy based on electrospinning and biomimetic mineralization opens up a new way to prepare diverse porous HA-based hybrid materials and shows great potential in drug delivery, gene transfer and tissue engineering.

Abstract:The ability to manipulate and control the surface properties of hydroxyapatite (HA) nanoparticles is of crucial importance for the design of HA-based carriers of therapeutic agents. In this work, surface-initiated atom transfer radical polymerization (ATRP) of (2-dimethyl amino) ethyl methacrylate (DMAEMA) is first employed to tailor the functionality of HA surfaces in a well-controlled manner and to produce a series of new cationic hybrids (termed as HA-PDM). The HA parts of HA-PDM were coated by different lengths of PDMAEMA chains. The HA-PDM exhibited a good ability to condense plasmid DNA (pDNA) with suitable particle size and a zeta potential for gene transfection. Most importantly, in comparison with PDMAEMA homopolymers, the HA-PDM displayed considerably enhanced buffering capacity, and exhibited much higher gene transfection efficiencies in different cell lines, including osteoblast MC3T3 and osteosarcoma MG63 cells. In addition, the HA-PDM/pDNA complexes also could largely enhance the differentiation of preosteoblast cells. Such well-defined HA-PDM nanohybrids possess great potential applications as new drug-delivery vectors in bone tissue engineering.
Synthesis of Hollow Amorphous Calcium Phosphate for Drug Delivery

Abstract:The self-assembly of nonionic surfactants Tween 80 and PEG 6000 in aqueous solution leads to the formation of hollow nanoscale niosomes. Calcium phosphate nanoparticales can be formed using the vesicles as templates. The calcium phosphate spheres were characterised by TEM, SEM, XRD and FTIR. The results indicated that the calcium phosphate particles were hollow and amorphous with the size in the range of 100 similar to 150 nm. This system had a good biocompatibility and could be used as drug carriers. In this work, we also used bovine serum albumin as model protein to study the drug content of the nanoparticles and the controlled release behaviour in vitro, and we found that the hollow calcium phosphate system had a higher drug content and release rate compared to traditionary hydroxyapatite particles.
