Abstract:Magnesium alloy is a potential biodegradable material and is widely used in medicine. However, its rapid degradation causes many negative effects and limits its application, so it is essential to improve corrosion resistance. In this study, a chitosan/polylactic acid composite coating was designed on the AZ31 alloy for medical applications. The composite coating sealed the pores of the chitosan layer to provide strong protection for the AZ31 alloy, with a reduced corrosion current density by 4 orders of magnitude. The excellent biocompatibility of the composite coating was proved by an increased cell viability test and morphology of cells adhered to the chitosan/polylactic acid coated sample. The composite coating provides a feasible method to enhance the corrosion resistance and biocompatibility of magnesium alloys.

Abstract:In order to solve the problem of excessive degradation rate and insufficient biocompatibility of magnesium‐based bone implants, a polyphenol (EGCG) induced hydroxyapatite (HA) coating was prepared on the surface of AZ31 alloy. The physical and chemical properties and corrosion resistance of the coating were analysed in depth, and its biocompatibility was preliminarily explored in vitro. The results showed that the polyphenol (EGCG) conversion coating constructed on the AZ31 could successfully induce the formation of HA by complexing the phenolic hydroxyl group with calcium ions. The electrochemical and long‐term immersion experiments showed that the corrosion resistance of EGCG/HA composite coating was significantly improved. The self‐corrosion current density, hydrogen evolution and the increase of pH value of AZ31‐EGCG/HA were significantly lower than those of AZ31. On the basis of inhibiting the excessive corrosion of the substrate, the composite coating significantly improves the compatibility of pre‐osteoblasts, supports the adhesion and spreading and effectively reduces the haemolysis rate to less than 5%. The preparation method of the coating is simple, low cost and suitable for complex shape surfaces, which can significantly improve the corrosion resistance and biocompatibility of the AZ31 substrate. It is expected to provide a solution for the surface modification of magnesium‐based bone implants.

Abstract:To slow down the corrosion rate and increase the bioactivity of magnesium alloys, biodegradable hydroxyapatite/magnesium (HA/Mg) composite coating was deposited onto AZ91D alloy substrate by high-velocity suspension flame spray technique using nano-sized HA and micron-sized Mg powders. Microstructures and phase constitutions of HA/Mg composite coating were analyzed by scanning electron microscopy and x-ray diffraction. Corrosion resistance of composite coating was also evaluated by potentiodynamic polarization test and electrochemical impedance spectroscopy in Hanks’ balanced salt solution (HBSS). The result shows that the HA/Mg composite coating mainly consisted of Mg and HA in addition to a few of MgO phases. The coating showed a rough surface morphology with homogeneous elemental distribution and a lamellar structure in cross section as well as well-bonded coating/substrate interface. Mg particles presented superior deposition during spraying due to their higher melting degree compared to HA particles. AZ91D alloy substrate was significantly protected from corrosion in HBSS by the HA/Mg composite coating with Mg dissolution on surfaces of the composite coating. This strategy can offer a new route to fabricate HA/Mg composite coatings on other metal substrates for use in biodegradable implant fields.

Engineering a Bioactive Hybrid Coating for In Vitro Corrosion Control of Magnesium and Its Alloy

Abstract:Magnesium (Mg) and its alloys are promising biodegradable metallic implant materials. However, their clinical applications are limited by their fast corrosion rate in the biological environment. In this work, with an outlook to improve the in vitro corrosion resistance of Mg and WE43 Mg alloy, a layer-by-layer interfacially engineered anticorrosive and bioactive coating consisting of a natural oxide lower layer, hydroxyapatite (HA) middle layer, and silk fibroin (SF) top layer was fabricated and investigated. Anodization was used to create natural oxide layer induced microroughness on substrates. The electrochemically deposited HA layer improved the surface microroughness and microhardness but significantly decreased Mg ion release, hydrogen gas evolution, and weight loss in simulated body fluid. The spin-coated SF layer further decreased hydrophilicity, in vitro degradation, and corrosion rate. The nonspecific and specific intermolecular interactions between fabricated layers along with their mechanical interlocking interface contributed to improved adhesion strength and integrity of the coating. The SF+HA-coated samples showed enhanced degradation and corrosion resistance due to a synergistic effect of the underlying HA layer, hindering the ingress of aggressive ions and the top hydrophobic SF layer, preventing the ingress of corrosive solution. The SF+HA-coated Mg and WE43 Mg alloy samples exhibited 50 and 26 times decreased corrosion rate, respectively, compared to uncoated samples. Moreover, in vitro cytotoxicity and cell culture studies using a mouse fibroblast cell showed that the SF+HA hybrid coating improved the cell viability, attachment, and proliferation, with cells exhibiting elongated morphology on coated samples as compared to a round shape on uncoated samples.

Abstract:The biodegradability of magnesium (Mg) metal implants necessitates surface modification to mitigate the high corrosion rate in the body's physiological environment. This study focuses on synthesizing and applying a nanocomposite coating of polydimethylsiloxane-nanosized bioactive glass (PDMS-nBG) on the surface of a plasma electrolytic oxidation (PEO)-modified AZ91 Mg alloy. The PDMS-1 wt% nBG nanocomposite coating was deposited on the PEO-treated substrate using the dip-coating method. Surface and cross-sectional morphologies of the nanocomposite coatings were examined using field emission scanning electron microscopy (FESEM). Corrosion behavior of the AZ91 substrate, PEO coating, and double-layer PEO/PDMS-nBG coating was investigated using electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization (PDP) tests. Results showed that the corrosion resistance and corrosion current density of Mg substrates increased and decreased from 4815 Ω.cm 2 and 8.96 μA.cm − 2 to 18 MΩ.cm 2 and 0.00016 μA.cm − 2 for AZ91 and PEO/PDMS-nBG coating, respectively. Additionally, the morphology and adhesion of cultured human mesenchymal cells on the PEO-modified AZ91 substrates with a single PDMS polymer coating and a PDMS-nBG nanocomposite coating were investigated using fluorescent light microscopy and FESEM. The PEO/PDMS-nBG bilayer coating effectively addressed the limitations of magnesium alloys and holds potential for biomedical applications, particularly in orthopedic implants, due to the enhanced biocompatibility and bioactivity of Mg alloys.

Abstract:Magnesium (Mg) alloys have been promised for biomedical implants in orthopedic field, however, the fast corrosion rate and mode challenge their clinical application. To push Mg alloys materials into practice, a composite coating with biodegradable and high compatible components to improve anticorrosion property of an Mg alloy (i.e., AZ31) is designed and fabricated. The inner layer is micro-nano structured Mg(OH)2 through hydrothermal treatment. Then stearic acid (SA) is introduced to modify Mg(OH)2 for better reducing the gap below a surface-degradation polymer layer of poly(1,3-trimethylene carbonate). Benefited by the SA modification effect, this sandwiched coating avoids corrosive medium penetration via enhancing the adhesion strength at the interface between outer and inner layers. Both in vitro and in vivo tests indicate that the composite coating modified AZ31 perform a better anticorrosion behavior and biocompatibility compared to bare AZ31. Strikingly, a 1.7-fold improvement in volume of newly formed bone is observed surrounding the composite coating modified implant after 12 week implantation. The sandwiched biocompatible coating strategy paves a hopeful way for future translational application of Mg alloys orthopedic materials in clinics.
Hydroxyapatite/palmitic acid superhydrophobic composite coating on AZ31 magnesium alloy with both corrosion resistance and bacterial inhibition

Abstract:The coating-modified magnesium (Mg) alloys exhibit controllable corrosion resistance, but the insufficient antibacterial performance limits their clinical applications as degradable implants. Superhydrophobic coatings show excellent performance in terms of both corrosion resistance and inhibition of bacterial adhesion and growth. In this work, a hydroxyapatite (HA)/palmitic acid (PA) superhydrophobic composite coating was fabricated on the Mg alloy by the hydrothermal technique and immersion treatment. The HA/PA composite coating showed superhydrophobicity with a contact angle of 153° and a sliding angle of 2°. The coated Mg alloy exhibited excellent corrosion resistance in the simulated body fluid, with high polarization resistance (77.10 kΩ·cm 2 ) and low corrosion current density ((0.491 ± 0.015) μA·cm −2 ). Meanwhile, the antibacterial efficiency of the composite coating was over 98% against E. coli and S. aureus in different periods. The results indicate that the construction of such superhydrophobic composite coating (HA/PA) on the Mg alloy can greatly improve the corrosion resistance of Mg alloy implants within the human body and avoid bacterial infection during the initial stages of implantation.

Abstract:A new biodegradable nanocomposite coating to control the biocompatibility and anticorrosion property of Mg is reported in this work. The key feature of this strategy is to equip the Mg surfaces with poly(gamma-glutamic acid)-g-7-amino-4-methylcoumarin/hydroxyapatite (gamma-PGA-g-AMC/HA) hybrid nanoparticles via electrophoretic deposition in ethanol. The microstructures of the resulting nanocomposite coating are characterized by Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscopy. The results of standard electrochemical measurements along with immersion tests indicate that the nanocomposite coating has preferable in vitro degradation and corrosion resistance behavior than bare Mg. In addition, cytocompatibility is conducted using NIH3T3 cells and the coated sample shows better cell viability and cell adhesion than pure Mg substrate over the whole incubation period. The favorable anticorrosion behavior and cytocompatibility of the nanocomposite coating suggest that the newly developed gamma-PGA-g-AMC/HA biodegradable nanocomposite coating may have a potential to improve the biological performance of Mg-based biomedical implants.
Mg-MOF-74/MgF2 Composite Coating for Improving the Properties of Magnesium Alloy Implants: Hydrophilicity and Corrosion Resistance

Abstract:Surface modification on Mg alloys is highly promising for their application in the field of bone repair. In this study, a new metal-organic framework/MgF₂ (Mg-MOF-74/MgF₂) composite coating was prepared on the surface of AZ31B Mg alloy via pre-treatment of hydrofluoric acid and in situ hydrothermal synthesis methods. The surface topography of the composite coating is compact and homogeneous, and Mg-MOF-74 has good crystallinity. The corrosion resistance of this composite coating was investigated through Tafel polarization test and immersion test in simulated body fluid at 37 °C. It was found that Mg-MOF-74/MgF₂ composite coating significantly slowed down the corrosion rate of Mg alloy. Additionally, Mg-MOF-74/MgF₂ composite coating expresses super-hydrophilicity with the water contact angle of nearly 0°. In conclusion, on the basis of MgF₂ anticorrosive coating, the introduction of Mg-MOF-74 further improves the biological property of Mg alloys. At last, we propose that the hydrophilicity of the composite coating is mainly owing to the large number of hydroxyl groups, the high specific surface area of Mg-MOF-74, and the rough coating produced by Mg-MOF-74 particles. Hence, Mg-MOF-74 has a great advantage in enhancing the hydrophilicity of Mg alloy surface.

Abstract:Magnesium (Mg) and its alloys have exhibited great potential for orthopedic applications; however, their poor corrosion resistance and potential cytotoxicity have hindered their further clinical applications. In this study, we prepared a calcium phosphate (Ca-P) coating with a micro-nanofibrous porous structure on the Mg alloy surface by a chemical conversion method. The morphology, composition, and corrosion performance of the coatings were investigated by scanning electron microscope (SEM), energy-dispersive spectrometer (EDS), X-ray diffraction (XRD), immersion tests, and electrochemical measurements. The effects of the preparation temperature of the Ca-P coatings were analyzed, and the results confirmed that the coating obtained at 60 °C had the densest structure and the best corrosion resistance. In addition, a systematic investigation into cell viability, ALP activity, and cell morphology confirmed that the Ca-P coating had excellent biocompatibility, which could effectively promote the proliferation, differentiation, and adhesion of osteoblasts. Hence, the Ca-P coating demonstrates great potential in the field of biodegradable Mg-based orthopedic implant materials.
Evaluation of Corrosion Performance of AZ31 Mg Alloy in Physiological and Highly Corrosive Solutions

Abstract:Resorbable Mg and Mg alloys have gained significant interest as promising biomedical materials. However, corrosion of these alloys can lead to premature reduction in their mechanical properties, and therefore their corrosion rate needs to be controlled. The aim of this study is to select an appropriate environment where the effects of coatings on the corrosion rate of the underlying Mg alloy can be discerned and measured in a relatively short time period. The corrosion resistance of uncoated AZ31 alloy in different solutions [Hank's Balanced Salt Solution, 1× phosphate buffered solution (PBS), 4× PBS, 0.9%, 3.5%, and 5 M sodium chloride (NaCl)] was determined by measuring the weight loss over a 2 week period. Upon exposure to physiological solutions, the uncoated AZ31 alloys exhibited a variable weight increase of 0.4 ± 0.4%. 3.5% and 5 M NaCl solutions led to 0.27 and 9.7 mm/year corrosion rates, respectively, where the compositions of corrosion products from AZ31 in all saline solutions were similar. However, the corrosion of the AZ31 alloy when coated by electrochemical oxidation with two phosphate coatings, one containing fluorine (PF) and another containing both fluorine and silica (PFS), showed 0.3 and 0.25 mm/year corrosion rates, respectively. This is more than 30 times lower than that of the uncoated alloy (7.8 mm/year), making them promising candidates for corrosion protection in severe corrosive environments. Cross-sections of the samples showed that the coatings protected the alloy from corrosion by preventing access of saline to the alloy surface, and this was further reinforced by corrosion products from both the alloy and the coatings forming an additional barrier. The information in this paper provides a methodology for evaluating the effects of coatings on the rate of corrosion of magnesium alloys.

Abstract:Manipulating the degradation rate of biomedical magnesium alloys poses a challenge. The characteristics of a microarc oxidation (MAO), prepared in phytic acid, and poly(L-lactic acid) (PLLA) composite coating, fabricated on a novel Mg-1Li-1Ca alloy, were studied through field emission scanning electron microscopy (FE-SEM), electron probe X-ray microanalysis (EPMA), energy dispersive X-ray spectroscopy (EDS) and X-ray diffraction (XRD). The corrosion behaviors of the samples were evaluated via hydrogen evolution, potentiodynamic polarization and electrochemical impedance spectroscopy in Hanks' solution. The results indicated that the MAO/PLLA composite coatings significantly enhanced the corrosion resistance of the Mg-1Li-1Ca alloy. MTT and ALP assays using MC3T3 osteoblasts indicated that the MAO/PLLA coatings greatly improved the cytocompatibility, and the morphology of the cells cultured on different samples exhibited good adhesion. Hemolysis tests showed that the composite coatings endowed the Mg-1Li-1Ca alloys with a low hemolysis ratio. The increased solution pH resulting from the corrosion of magnesium could be tailored by the degradation of PLLA. The degradation mechanism of the composite coatings was discussed. The MAO/PLLA composite coating may be appropriate for applications on degradable Mg-based orthopedic implants.

Abstract:Orthopaedic implants often make use of magnesium and its alloys. These alloys are more mechanically compatible with bone tissue than other alloys, such as titanium, cobalt, and nickel, but a loss of implant mechanical strength can occur due to their low corrosion resistance. The plasma electrolytic oxidation (PEO) method was used to modify the surface of AZ31 magnesium alloy in this study. The effects of varying concentrations of hydroxyapatite (HA) nanoparticles (1, 3, and 5 g/l) on the morphology and corrosion characteristics of coatings were examined using phosphate-based electrolytes. The elemental composition was determined using Energy dispersive spectroscopy (EDS), elemental map analysis (MAP) and phase X-ray diffraction analysis (XRD). Field emission-SEM was employed to investigate the surface morphology and cross-section of the coatings. The corrosion characteristics of all samples were evaluated by electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization (PDP) in Ringer's solution. The sample with 3 g/l HA showed the best anti-corrosion performance, as it had the smallest average pore size (8.7 μm), the thickest coating (22.5 μm) and the lowest corrosion current density (0.034 μA/cm 2 ) among all the samples tested.
Deposition and corrosion performance of phosphate-polylactic acid composite coatings on WE43 magnesium alloy

Abstract:The degradation profile of Mg alloys in biomedical environments can be controlled by applying various types of surface coatings. Phosphate chemical conversion coatings (CaP) and polylactic acid (PLA) biopolymer coatings are biocompatible and promising for the corrosion protection of Mg alloys. The aim of this work was to investigate the physicochemical properties and corrosion performance of inorganic, organic, and mixed inorganic-organic coatings. Three types of surface modification, CaP, PLA, and a combined CaP-PLA coating were obtained on the surface of the magnesium alloy WE43. Scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, and infrared spectroscopy were employed to characterize the surface and interfacial morphology, along with the composition of the coatings before and after corrosion experiments. The protective performance of obtained coatings was examined by electrochemical impedance spectroscopy and immersion testing in Hank's solution. Coated samples showed significantly improved corrosion resistance. The values of polarization resistance based on impedance measurements were 164; 8688; 69,410; and 228,369 Ω cm 2 for WE43 alloy, CaP, PLA, and CaP-PLA coatings, respectively. The results allowed to propose the mechanism of the coatings growth and their further corrosion degradation. A two-component CaP-PLA coating provided reliable corrosion resistance in Hank's solution for 14 days.

Surface Modification by Natural Biopolymer Coatings on Magnesium Alloys for Biomedical Applications

Abstract:Natural biopolymers possess good biocompatibility and exhibit excellent bioactivity due to the components of the extracellular cell matrix, which are abundant in the biomass and are easily accessible from a range of sources, such as forest products, grasses, tunicates, crustacean, and stalks. In this chapter, magnesium alloys modified with a natural biopolymer coating to improve their biocompatibility and degradation properties are introduced. Some natural biopolymers, such as chitosan, phytic acid, collagen, and stearic acid have been studied and coated on the surface of magnesium alloy for biomedical applications. Enhanced corrosion resistance, improved biocompatibility, and a reduced hemolysis ratio of WE43 was achieved after modification with phytic acid solution with different pH values of the conversion coating, especially when the pH value of the modified solution was 5. Various chitosan dip-coated Mg-1Ca alloys also displayed greater corrosion resistance in simulated body fluid. Calcium phosphate/chitosan composite films coated on the surface of micro-arc oxidized (MAO)-AZ91D alloy through electrophoretic deposition or on the surface of AZ31 alloy by aerosol deposition were adopted to enhance the bond strength between the magnesium substrate and hydroxyapatite. Stearic acid coating on hydrothermal-treated Mg samples enhanced the corrosion resistance of Mg. A drug release system on MAO-WE42 was established by a cross-linked gelatin/PLGA particle loaded with paclitaxel. Finally, a comparatively smooth surface of magnesium coated with albumin attached via linker groups was employed for better corrosion property, taking advantage of the presence of an Mg(OH)2-terminated MgO layer via silane coupling chemistry. Layered double hydroxide/hydroxyapatite-ciprofloxacin composite coating on AZ31 magnesium alloy: Corrosion resistance, antibacterial, osteogenesis

Abstract:Magnesium alloys have received more attention as orthopedic repair materials, while their rapid degradation and susceptibility to bacterial infections limit their clinical medical applications. In this study, the layered double hydroxide/hydroxyapatite (LDH/HAp) composite coating was first prepared by hydrothermal method, and then ciprofloxacin (CIP) was loaded on LDH/HAp by the traditional immersion method. Composite coatings were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), and scanning electron microscopy (SEM) techniques. The LDH/HAp-CIP coating has a more excellent inhibitory effect on the activity of Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). At the same time, composite coatings showed higher cell compatibility to MC3T3-E1 cells. Composite coatings promoted the expression of alkaline phosphatase (ALP) and upregulated the expression level of related osteogenic genes. Overall, these results are conducive to the advancement of the clinical application of magnesium alloys as orthopedic restorative materials.Graphical abstract

Abstract:Human endogenous retrovirus (HERV) K is present in about 50 copies in the human genome and transcription of HERV-K has previously been detected in several tumor cell lines as well as in peripheral blood lymphocytes from healthy donors. By means of immunoblotting we investigated the presence of antibodies recognizing recombinant HERV-K-envelope outer membrane constructs in different serum collectives. A total of 12.6% of sera obtained from normal blood donors was found positive. In serum collectives from breast carcinoma patients, HIV-1-positive individuals, and persons with cytomegalovirus infections no significant difference from the normal blood donor serum collective could be observed. Only a group of persons with a repeatedly raised serum neopterin concentration (> 10 nmol/liter) of unknown cause (HIV and hepatitis B and C virus infections were excluded) showed a significant higher percentage of HERV-K-outer membrane envelope-positive sera (21%). Furthermore we could observe a parallel HIV-1/HERV-K seroconversion, which probably is not due to an HIV-1/HERV-K-outer membrane envelope cross-reactivity. Possible implications of these findings are discussed.
Corrosion Behavior of Zrn/Zr Coated Biomedical Az91 Magnesium Alloy

Abstract:Magnesium alloys are potential materials in biodegradable hard tissue implants. However, their fast degradation rates in a physiological environment constitute the main limitation for their applications. In this work, a ZrN/Zr bilayered coating is deposited on AZ91 magnesium alloy using a filtered cathodic arc deposition system. The ZrN/Zr bilayered structure buffers the serious mismatch between the substrate and ZrN coating thus improving film adhesion. The composition of the coating is evaluated by X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). The electrochemical behavior of the coated alloy is systematically studied by means of potentiodynamic polarization test and electrochemical impedance spectroscopy (EIS). Our results disclose that the corrosion resistance of the coated alloy is significantly improved.

Abstract:Mg alloys can be used as bioresorsable metallic implants. However, the high corrosion rate of magnesium alloys has limited their biomedical applications. Although Mg ions are essential to the human body, an excess may cause undesirable health effects. Therefore, surface treatments are required to enhance the corrosion resistance of magnesium parts, decreasing its rate to biocompatible levels and allowing its safe application as bioresorbable metallic implants. The application of biocompatible silane coatings is envisaged as a suitable strategy for retarding the corrosion process of magnesium alloys. In the current work, a new glycidoxypropyltrimethoxysilane (GPTMS) based coating was tested on AZ31 magnesium substrates subjected to different surface conditioning procedures before coating deposition. The surface conditioning included a short etching with hydrofluoric acid (HF) or a dc polarisation in alkaline electrolyte. The silane coated samples were immersed in Hank's solution and the protective performance of the coating was studied through electrochemical impedance spectroscopy (EIS). The EIS data was treated by new equivalent circuit models and the results revealed that the surface conditioning process plays a key role in the effectiveness of the silane coating. The HF treated samples led to the highest impedance values and delayed the coating degradation, compared to the mechanically polished samples or to those submitted to dc polarisation.

Abstract:Magnesium alloy is emerging as a leading choice for biodegradable orthopedic implants, thanks to its superior biocompatibility and mechanical characteristics that align with those of natural bone. Nonetheless, its swift corrosion rate poses a challenge to its use in clinical settings. In this study, two methods were used to apply Cerium and Hydroxyapatite (HA) coatings on Mg AZ31 implants, specifically a one-step process (HA + Ce) and a two-step process (HA + Ce/CeCC), with the aim of improving their resistance to corrosion. The susceptibility of the samples to corrosion and the efficiency of the coatings in a physiological media were evaluated using Electrochemical Impedance Spectroscopy (EIS) and Direct Current (DC) polarization tests in a Simulated Body Fluid (SBF) solution. In the HA + Ce sample, the effect of immersion time was also examined. The Field Emission Scanning Electron Microscope (FE-SEM) results showed that after 15 min of coating process, a very weak and uneven coating is formed on the surface. However, at 30 and 60 min, the structure of the coating changes, forming a more crystalline and denser coating on the surface, which also has greater corrosion resistance. The results of the electrochemical tests showed that the sample prepared using the two-step method (HA + Ce/CeCC sample) had the highest resistance to both corrosion and biocorrosion. The morphology and composition of the coatings were inspected using FE-SEM and X-ray diffraction (XRD), confirming the formation of HA crystals and an amorphous layer of Cerium. Moreover, the HA + Ce/CeCC sample demonstrated the highest level of corrosion resistance in an Simulated Body Fluid (SBF) media over an extended duration of submersion.
