Abstract:Water ecosystems face enormous, variable anthropogenic stressors that contribute to water degradation. This study explores the effects of three substances frequently found in high concentrations in natural environments linked to agricultural areas on plankton abundance, composition, and trophic interactions. We conducted a microcosm experiment where we exposed a natural plankton assemblage to excess copper (Cu), atrazine (At), and dissolved organic matter (OM), along with a control group encompassing all possible treatment combinations. Each treatment was replicated three times ( n = 24), and the experiment lasted 96 h. At the beginning and end of the experiment, relevant limnological variables, including inorganic dissolved nutrients, were measured. We used one-way ANOVA analyses with total abundances and species richness as response variables for data analysis. Results showed that the response of each plankton group to these substances was unequal, with some benefiting at tested concentrations. For example, Cu was beneficial for Chlorophyceae and Euglenophyceae, while CuOM was beneficial for Cyanobacteria and Cryptophyceae. Regarding zooplankton, Copepoda density was depleted in all treatments where stressors were combined and when exposed to OM compared to the control. When richness was considered, phytoplankton showed depletion in CuOMAt compared to OM and AtCu. No treatments affected zooplankton richness. Zooplankton:phytoplankton ratios showed significant effects for total zooplankton and Copepoda in all treatments compared to the control. These results indicate that the effects of contaminants on natural assemblages strongly depend on the plankton group and the combinations of contaminants, making predictions and extrapolations a challenging task that requires further study.

Oxidative stress and toxicology of Cu2+ based on surface areas in mixed cultures of green alga and cyanobacteria: The pivotal role of H2O2

Abstract:The toxicity of heavy metals in algal monocultures is well studied and is mediated by reactive oxygen and nitrogen species (ROS/RNS). However, little is known about the toxicity of heavy metals and the mechanisms involved in mixed cultures. Here we examine the oxidative stress and toxic effects of Cu2+ on the green alga Dunaliella salina (DS) and the cyanobacteria Synecochoccus elongatus (SE) in both mono- and mixed cultures. We find that both species benefit in mixed cultures and acquire higher resistance to Cu2+ toxicity, with a particularly marked effect on SE. DS has a larger surface area than SE, so increases in the number of DS cells compared to SE diminishes the proportion of SE surface area exposed to Cu2+, and contributes to increasing cyanobacterial resistance in mixed cultures. However, these mixed cultures also display as an unexpected property an increased resistance of DS in mixed cultures. SE and DS cells showed significant differences on the kinetics of H2O2 production and antioxidant capacities. The integrated (overall) redox response of mixed cultures, in terms of total amount of H2O2 produced, was proportional to the total surface area of algal species exposed to Cu2+, independent of algal composition in mixed systems. However, mixed cultures display emergent properties, as the time course of H2O2 accumulation is not a simple function of the composition of the mixed cultures. Emergent properties are also observed in the speed of membrane lipid oxidation by the two species, as measured using mixed cultures in which only one of the two species is labeled using the membrane oxidation indicator C11-BODIPY581/591. We suggest that, in addition to H2O2¸ other redox signals (e.g. NO) and allelochemicals (auxins, cytokinins, etc.) may be used to construct a complex inter-species communication network. This could allow mixed algal systems, whatever their composition, to integrate their cellular responses and perform as a coherent unit against toxic Cu2+ ions.

Abstract:The physical and chemical properties of silver nanoparticles (AgNPs) have led to their increasing use in various fields such as medicine, food, and industry. Evidence has proven that AgNPs cause adverse effects in aquatic ecosystems, especially when the release of Ag is prolonged in time. Several studies have shown short-term adverse effects of AgNPs on freshwater phytoplankton, but few studies have analysed the impact of long-term exposures on these populations. Our studies were carried out to assess the effects of AgNPs on growth rate, photosynthesis activity, and reactive oxygen species (ROS) generation on the freshwater green algae Scenedesmus armatus and the cyanobacteria Microcystis aeruginosa , and additionally on microcystin (MC-LR) generation from these cyanobacteria. The tests were conducted both in single-species cultures and in phytoplanktonic communities exposed to 1 ngL -1 AgNPs for 28 days. The results showed that cell growth rate of both single-species cultures decreased significantly at the beginning and progressively reached control-like values at 28 days post-exposure. This effect was similar for the community-cultured cyanobacteria, but not for the green algae, which maintained a sustained decrease in growth rate. While gross photosynthesis (Pg) increased in both strains exposed in single cultures, dark respiration (R) and net photosynthesis (Pn) decreased in S. armatus and M. aeruginosa , respectively. These effects were mitigated when both strains were exposed under community culture conditions. Similarly, the ROS generation shown by both strains exposed in single-species cultures was mitigated when exposure occurred in community cultures. MC-LR production and release were significantly decreased in both single-species and community exposures. These results can supply helpful information to further investigate the potential risks of AgNPs and ultimately help policymakers make better-informed decisions about their utilization for environmental restoration.

Abstract:Heavy metals (HMs) and polycyclic aromatic hydrocarbons (PAHs) are toxic components of atmospheric particles. These pollutants induce a wide variety of responses in plants, leading to tolerance or toxicity. Their effects on plants depend on many different environmental conditions, not only the type and concentration of contaminant, temperature or soil pH, but also on the physiological or genetic status of the plant. The main detoxification process in plants is the accumulation of the contaminant in vacuoles or cell walls. PAHs are normally transformed by enzymatic plant machinery prior to conjugation and immobilization; heavy metals are frequently chelated by some molecules, with glutathione, phytochelatins and metallothioneins being the main players in heavy metal detoxification. Besides these detoxification mechanisms, the presence of contaminants leads to the production of the reactive oxygen species (ROS) and the dynamic of ROS production and detoxification renders different outcomes in different scenarios, from cellular death to the induction of stress resistances. ROS responses have been extensively studied; the complexity of the ROS response and the subsequent cascade of effects on phytohormones and metabolic changes, which depend on local concentrations in different organelles and on the lifetime of each ROS species, allow the plant to modulate its responses to different environmental clues. Basic knowledge of plant responses toward pollutants is key to improving phytoremediation technologies.

Abstract:Bullfrog tadpoles, Aquarana catesbeiana, were exposed to settleable particulate matter (SePM), (1 g L −1 , 96 h) and their organs were collected for analysis of metal/metalloid, oxidative stress and neurotoxicity in liver, muscle, kidney and brain. The SePM water of the exposed groups contained 18 of the 28 metals/metalloids detected in ambient particulate matter (APM). Fe 56 and Al were those that presented the highest concentrations, Cr, Mn, Pb and Cu increased from 10 to 20 times and Ti, V, Sr, Rb, Cd, Sn and Ni increased from 1 to 3 times compared to the control. Bioaccumulation of metals/metalloids in the exposure water varied significantly between organs, with the muscle and liver showing the highest concentrations of metals, followed by the brain. Lipoperoxidation and malondialdehyde increased only in muscle, while carbonyl proteins increased only in the liver and brain. Regarding nitric oxide synthase, there was an increase in the liver and brain in the group exposed to SePM. Catalase activity decreased in the liver and muscle, while the activity of glutathione peroxidase, increased in the liver and kidney and decreased in muscle. Glutathione S -transferase, which is mainly responsible for detoxification, increased in the liver and decreased in muscle and the kidney. Cholinesterase activity increased only in the muscle. The results indicate oxidative stress, due to oxidation catalyzed by metals, components of SePM. Thus, the results contribute to the understanding that SePM has a deleterious effect on the aquatic environment, negatively affecting bullfrog tadpoles, in different ways and levels in relation to the analyzed organs.

Abstract:Relations among polycyclic aromatic hydrocarbons (PAHs), biomarkers of oxidative stress (lipid peroxidation, glutathione, and glutathione S-transferase activity), and the possible influence of environmental factors (temperature, pH, and salinity) were assessed in situ for specimens of Ramnogaster arcuata, a native estuarine fish. PAH levels found in the muscular tissue of R. arcuata ranged from 0.7 to 293.4 ng g-1 wet weight with petrogenic and pyrolytic inputs. Lipid peroxidation in the liver showed positive correlations (P < 0.05) with total PAHs (r = 0.66), 3-ring (r = 0.66) and 4-ring PAHs (r = 0.52) and glutathione in muscle (r = 0.58). Significant positive correlations (P < 0.05) were also evidenced between muscular glutathione with total (r = 0.62) and 3-ring PAHs (r = 0.75). Hepatic glutathione S-transferase negatively correlated with 4-ring PAHs (r = -0.58). These correlations suggest that lipid peroxidation and muscular glutathione could be good biomarkers for complex mixtures of PAHs, and hepatic glutathione S-transferase could be a suitable biomarker for 4-ring PAHs. Furthermore, significant correlations (P < 0.05) of environmental factors with PAH levels and biomarkers were observed, especially pH with 3-ring PAHs (r = -0.65), lipid peroxidation (r = -0.6), glutathione in the liver (r = -0.73) and muscle (r = -0.75); and temperature with 2-ring PAHs (r = -0.75) and glutathione in muscle (r = 0.51). The data suggest an influence of physicochemical parameters which could be driving a shift in PAH toxicity in R. arcuata. These results are essential for an integrated understanding of ecotoxicology and could help to predict environmental effects in present and future scenarios of ocean warming and acidification.

Abstract:The wide-ranging use of heavy metals and pesticides worldwide and their irreversible accumulation in aquatic ecosystems is a major concern. As the range of household and agricultural chemicals increases, water pollution is trending from the toxic effects of a single agent to complex agent pollution that threatens aquatic ecosystems. The aim of this study was to investigate the effects of pesticides (imidacloprid, IMI) and metals (copper sulfate, CuSO4) on oxidative stress biomarkers, antioxidant enzymes, and biomolecular parameters. The present study on the individual and combined effects of Oreochromis niloticus copper sulfate (CuSO4; 1 ppm), imidacloprid (IMI; 10 and 50 ppm), and IMI + CuSO4 (IMI10 + CuSO4, IMI50 + CuSO4) groups for 14 days. In this context, oxidative stress/antioxidant markers (SOD, CAT, GST, and GSH) and biomolecular markers including HSP70, 8-OHdG, PC, and TBARS levels were examined in fish liver and kidney tissues, which are detoxification organs. The results indicated that IMI and CuSO4 toxicity alone and in combination altered oxidative stress/antioxidant markers and biomolecular parameters; moreover, 14 days of exposure to the combination of CuSO4 and imidacloprid in particular exhibited a synergistic effect and caused oxidative toxicity. These findings highlighted the importance of evaluating mixtures of pesticides and metals and that the results show a remarkably synergistic effect. It can be concluded that these biomarkers are important indicators of physiological changes in living organisms.
A Long‐Term Study on the Effect of Cyanobacterial Crude Extracts from Lake Chapultepec (Mexico City) on Selected Zooplankton Species

Abstract:Many urban lakes in Mexico City, such as Lake Chapultepec, are infested with high densities of cyanobacteria, particularly Microcystis. We tested the effect of cyanotoxins from cyanobacterial crude extracts on the demographic variables of zooplankton. The rotifers Brachionus havanaensis and Brachionus calyciflorus, and the cladocerans Ceriodaphnia dubia and Moina macrocopa were used for the assays. Temperature effects on the response of B. calyciflorus and two clones of M. macrocopa were tested. We hypothesized that with an increase in cyanotoxin concentration and temperature there would be an increase in the adverse effect on the test species and that the clone of Moina previously exposed to cyanobacteria from Lake Chapultepec would be more resistant to the cyanotoxins. Demography experiments showed that B. havanaensis was more sensitive than C. dubia. The negative effect of the cyanobacterial crude extract on B. calyciflorus was greater at 30ºC than at 20º or 25ºC. The strain of M. macrocopa isolated from Lake Chapultepec was more resistant to the cyanotoxins than the strain which had not been previously exposed to the cyanobacteria. Our study indicated that cyanobacteria in Lake Chapultepec are highly toxic and, considering the recreational use of this lake, should be controlled.This article is protected by copyright. Abstract:Heavy metals can cause a significant damage to submerged macrophytes and affect its periphyton biofilms in aquatic environments. This study investigated the effects of heavy metals such as copper (Cu), lead (Pb), cadmium (Cd) and their mixture on physiological and biochemical responses and ultrastructure characteristics of Vallisneria natans (V. natans). Furthermore, differences in structures of microbial communities were observed in biofilms. The results showed that Cu2+, Pb2+, Cd2+ and their mixture could destroy cell structure and photosynthetic system, and directly caused oxidative damage to submerged macrophyte and induced antioxidant enzyme system. In general, biomass and total chlorophyll content of V. natans noticeably decreased, while the activities of superoxide dismutase, peroxidase and catalase were enhanced by heavy metal stress inducement in restricted range, and the malondialdehyde content increased with the aggravation of the damage. The single heavy metal stress played a negative impact, however, the combined stress was not always synergistic effects on plants. High-throughput sequencing analysis suggested that heavy metals changed the abundance and structure of the microbial biofilm community. Proteobacteria and Bacteroidete were the dominant bacteria under heavy metal stress and other species and abundance of bacteria such as Firmicute, Cyanobacteria, Chloroflexi, Actinobacteria, Verrucomicrobia, Acidobacteria, Deinococcus-Thermus, Chlamydiae were also present. These findings provided useful information for further understanding about submerged macrophytes and periphyton biofilms responsed to heavy metal stress in aquatic environments in the future.

Preparation of Lithium Niobate Nanoparticles by High Energy Ball Milling and their Characterization

Abstract:We present investigations on the preparation of lithium niobate, LiNbO3 nanoparticles using high energy ball-milling. Stoichiometric composition of LiNbO3 powder was prepared by solid-state reaction method and used for ball-milling. Various milling parameter were optimized to get required particle sizes. Synthesized nanoparticles were characterized for their structure and particle sizes using X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and dynamic light scattering (DLS) techniques. The UV-Visible transmission shows the blue shift in cutoff, which indicate nearly stoichiometric composition of the prepared material. It has been observed that the sizes of particles decreases with increasing milling speed and time. Observed particle sizes were found in the range ~30-60 nm.

Abstract:Metals such as copper (Cu) enter marine environments from natural and anthropogenic sources, causing changes in the biodiversity of marine microalgae and cyanobacteria. Cu plays a dual role as either a micronutrient or toxicant depending on the environmental concentration. Many studies have summarized the potential of Cu to become more toxic to microalgae under environmental stress (for instance climate change). Most of the data available on Cu toxicity concerning microalgae and cyanobacteria have been produced using single-species laboratory tests, and there is still a significant gap in the information concerning the behavior of a group of algae exposed to environmental stressors. Thus, the objective of this study was to evaluate the toxicity of Cu at two concentrations (C1 = 2 μg L −1 and C2 = 5 μg L −1 ) in multispecies bioassays using three phytoplankton species (one cyanobacteria, Synechococcus sp., and two microalgae, Chaetoceros gracilis and Pleurochrisys cf. roscoffensis ). Combinations of two temperatures (20 and 23 °C) and two salinities (33 and 36 PSU), were applied in a 96 h study using flow cytometry analysis (FCM). Algal growth and reactive oxygen species (ROS) production by 2′7′-dichlorofluorescein (DCFH) were monitored by FCM. The results indicated that Synechococcus sp. was more sensitive than C. gracilis and P. roscoffensis to Cu stress at a temperature 23 °C and salinity of 36 PSU under both concentrations of Cu. Chlorophyll a fluorescence showed a significant decrease ( p < 0.05) in Synechococcus sp. under 5 μg L −1 of Cu in the combined treatment of 20 °C and 33 PSU; however, there was a significant increase in P. roscoffensis in all combinations at C2 = 5 μg L −1 compared to the control with no Cu, indicating a potential hormetic response to Cu for P. roscoffensis . ROS levels were triggered in a combination of 23 °C and 33 PSU and 5 μg L −1 of Cu, which was higher than all the other combinations studied. Our study resulted in data concerning the potential impacts caused by possible future climate change scenarios in aquatic habitats chronically exposed to metals.

Abstract:Two of the largest water reservoirs in the Metropolitan Region of São Paulo, Brazil (MRSP), named Billings and Guarapiranga, are facing high levels of anthropic impact. This is evidenced by the presence of contaminants and pollutants, which are deteriorating their water quality. Therefore, this study evaluated antioxidant defense enzymes, lipoperoxidation and genotoxicity, in adult females of a native species, Astyanax altiparanae from the Guarapiranga and Billings reservoirs. The study also aimed to evaluate these biomarkers during two different periods of the year, the rainy (summer) and dry (winter) seasons. The oxidative stress was evaluated by the activity of enzymes such as glutathione peroxidase, glutathione S-transferases, superoxide dismutase, and catalase in the gills and liver, and the occurrence of lipoperoxidation was also evaluated in both organs. The genotoxicity was assessed by performing comet assay, micronucleus, and nuclear abnormality tests on blood samples. The results showed that fish from both reservoirs are subjected to oxidative stress and genotoxic damage, mainly during winter, but fish living in Billings showed greater alterations than fish from Guarapiranga. Likewise, the results of the principal component analysis suggested that caffeine, nitrogenous compounds, and some metals might be triggering these toxic effects in fish.

Abstract:A neutral polysaccharide fraction (WGPN) prepared from Panax ginseng C.A. Meyer by hot water extraction and DEAE-cellulose chromatography was tested for its anticancer activity alone and in combination with 5-fluorouracil (5-FU) in Sarcoma-180 (S180) tumor-bearing mice by intragastric administration. WGPN alone inhibited S180 tumor growth in a bell-shaped dose-response curve, and the combination with 5-FU showed a synergistic effect. Studies of various immunological activities in S180-bearing mice revealed that WGPN stimulated the proliferation of lymphocytes, increased natural killer cell cytotoxicity, enhanced the phagocytosis and nitric oxide production by macrophages, and increased the level of tumor necrosis factor-alpha in serum. In combination with 5-FU, WGPN mitigated damage to the immune system caused by 5-FU in S180-bearing mice. These results suggest that WGPN might be a potential adjuvant for chemotherapeutic drugs.
Evaluation of oxidative stress biomarkers in Aiolopus thalassinus (Orthoptera: Acrididae) collected from areas polluted by the fertilizer industry

Abstract:The waste products of the fertilizer industry such as heavy metals, but especially phosphates and sulphates, are a serious problem that influences the structure and functioning of ecosystems. The levels of Cd, Pb, Zn, Cu, sulphates and phosphates were measured in soil samples from four sites: a control and sites that were 1, 3 and 6 km (sites A-C) away from the Abu-Zaabal Fertilizer Company. Oxidative stress markers (protein carbonyls, lipid peroxides), antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), polyphenoloxidase (PPO) and ascorbate peroxidase (APOX)) were evaluated in the tissues of Aiolopus thalassinus, collected from the corresponding sites. The highest concentrations of Cu and Zn were found in the soil from site A. The level of protein carbonyls in the brain, thoracic muscles and gut of the males and females from sites A, B and C were 11.82, 4.38, 5.97 (males) and 19.04, 16.65, 7.79 (females) times higher, respectively, compared to the individuals from the control site. Lipid peroxides levels in both sexes were significantly correlated with the distance from the source of the contamination. In the brain, thoracic muscles and gut of the males and females collected from site A, the level of lipid peroxides were 15.41, 23.49, 11.50 (males) and 25.36, 11.34, 15.37 (females) times higher compared to the values of the control animals. The activities of SOD, PPO, CAT and APOX were significantly affected by the environmental pollutants. The enzymatic and non-enzymatic oxidative markers in the Aiolopus thalassinus, a common insect species that inhabits various ecosystems, can also be used as a relevant biomarker of the pollution that is caused by the fertilizer industry.
Antioxidant enzyme activities and lipid peroxidation in the freshwater cladoceran Daphnia magna exposed to redox cycling compounds

Abstract:Contaminant-related changes in antioxidative processes in the freshwater crustacea Daphnia magna exposed to model redox cycling contaminant were assessed. Activities of key antioxidant enzymes including catalase, superoxide dismutase, glutathione peroxidase and glutathione S-transferases and levels of lipid peroxidation measured as thiobarbituric acid-reactive substances (TBARS) and lipofucsin pigment content were determined in D. magna juveniles after being exposed to sublethal levels of menadione, paraquat, endosulfan, cadmium and copper for 48 h. Results denoted different patterns of antioxidant enzyme responses, suggesting that different toxicants may induce different antioxidant/prooxidant responses depending on their ability to produce reactive oxygen species and antioxidant enzymes to detoxify them. Low responses of antioxidant enzyme activities for menadione and endosulfan, associated with increasing levels of lipid peroxidation and enhanced levels of antioxidant enzyme activities for paraquat, seemed to prevent lipid peroxidation, whereas high levels of both antioxidant enzyme activities and lipid peroxidation were found for copper. For cadmium, low antioxidant enzyme responses coupled with negligible increases in lipid peroxidation indicated low potential for cadmium to alter the antioxidant/prooxidant status in Daphnia. Among the studied enzymes, total glutathione peroxidase, catalase and glutathione S-transferase appeared to be the most responsive biomarkers of oxidative stress.

Relationships between high-concentration toxic metals in sediment and evolution of microbial community structure and carbon–nitrogen metabolism functions under long-term stress perspective

Abstract:Microorganisms are highly sensitive to toxic metal pollution and play an important role in the material cycling and energy flow of the water ecosystem. Herein, 13 sediment samples from Junchong Reservoir (Guangxi Province, China) were collected in December 2021. The spatial distribution of pollution levels for toxic metals and the effects of toxic metals on the composition, functional characteristics, and metabolism of microorganisms were investigated. The results demonstrated that the area is a proximate area to industrial zones with severity of toxic metal pollution. Their mean concentrations of As, Cu, Zn, and Pb were up to 128.79 mg/kg, 57.62 mg/kg, 594.77 mg/kg, and 97.12 mg/kg respectively. There was a strong correlation between As, Cu, Zn, and Pb, with the highest correlation coefficient reaching 0.94. As the level of toxic metal pollution increases, the diversity and abundance of microorganisms gradually decrease. Compared to those with lower pollution levels, the Shannon index in regions with higher pollution levels decreases by up to 0.373, and the Chao index decreases by up to 143.507. However, the relative abundance of Bacteroidota , Patescibacteria , and Chloroflexi increased by 23%, 20%, and 5%, respectively, indicating their higher adaptability to toxic metals. Furthermore, microbial carbon and nitrogen metabolism were also affected by the presence of toxic metals. FAPROTAX analysis demonstrated an abundant reduction of ecologically functional groups associated with carbon and nitrogen transformations under high toxic metal pollution levels. KEGG pathway analysis indicated that carbon fixation and nitrogen metabolism pathways were inhibited with increasing toxic metal concentrations. These findings would contribute to a better understanding of the effects of toxic metal pollution on sediment microbial communities and function, shedding light on the ecological consequences of toxic metal contamination.

Abstract:Butyl Xanthate (BX) is a typical flotation reagent used to extract non-ferrous nickel ores, discharged into the surrounding environment of mining areas in large quantities. However, few studies have focused on the toxicity of combined pollution of BX and nickel (Ni) on aquatic plants, especially phytoplankton, the main producer of aquatic ecosystems. The toxicity and potential mechanism of single and combined pollution of BX and Ni at different concentrations (0-20 mg L-1) on typical freshwater algae (Chlorella pyrenoidosa) were studied. BX slightly stimulated the growth of C. pyrenoidosa on the first day, but Ni and Ni/BX mixture significantly inhibited it during incubation. Results showed that the inhibition rate (I) of the pollutants on the growth of C. pyrenoidosa followed the order: Ni/BX mixture > Ni > BX. The 96-h 20% effective inhibitory concentrations (96h-EC20) of Ni and BX on C. pyrenoidosa growth were 3.86 mg L-1 and 19.25 mg L-1, respectively, indicating C. pyrenoidosa was sensitive to pollutants. The content of total soluble protein (TSP) and chlorophyll a (Chl-a) changed significantly, which may be caused by the damage of pollutants to cell structures (cell membranes and chloroplasts). In addition, the I of pollutants on C. pyrenoidosa growth was related to dose, superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA). The increasement of reactive oxygen species (ROS), antioxidant enzymes (SOD and CAT), and MDA content, suggested C. pyrenoidosa suffered from oxidative stress, leading to lipid oxidation. These results will help to understand the toxicity mechanism of pollutants in typical mining areas and assess the environmental risks of pollutants to primary producers in aquatic ecosystems.

Abstract:Microalgal metabolism is affected by the surrounding environment and nutrients such as phosphorus (P) and nitrogen (N) are essential for optimal metabolism, as well as trace amounts of essential metals such as zinc (Zn); although in higher doses than required, Zn can be toxic. Lead (Pb) is a non-essential metal that can harm organisms from different trophic levels. In the environment, algae are exposed to several stressors simultaneously and adapt their metabolism. In the present study, we evaluated P limitation combined with environmental concentrations of Zn or Pb to the freshwater microalga Raphidocelis subcapitata regarding growth, pigments production, and photosynthetic parameters. Our results indicate that P limitation affected the growth, pigments production, relative maximum electron transport rate (rETR max ), and saturation irradiance; while Pb altered growth, pigments production, and maximum quantum yield; and Zn affected pigment production, photochemical and non-photochemical quenching, and rETR max . However, the combination of metal and P limitation resulted in synergistic responses, i.e., higher damages than the isolated stressors, in growth, maximum and effective quantum yield, and in the rapid light curve parameters. On the other hand, antagonism, i.e., lower damages than isolated stressors, was observed in pigments production and non-photochemical quenching, suggesting that algae activated defense mechanisms to cope with both stressors simultaneously. In addition, our results indicate an algal metabolism adjustment to P limitation and highlight the importance of considering physicochemical water characteristics when defining regulations of acceptable levels of metals in aquatic ecosystems.

Usefulness of oxidative stress biomarkers in native species for the biomonitoring of pesticide pollution in a shallow lake of the Austral Pampas, Argentina

Abstract:Pesticide contamination and its adverse effects on native freshwater species continue to be a worldwide major concern, mainly in developing countries. Passive biomonitoring of pesticide pollution in shallow lakes may be achieved by the simultaneous use of fish and wetland plants. Thus, the present study aimed to evaluate the occurrence of current-use pesticides in the surface water of a shallow lake of the Austral Pampas region (Buenos Aires Province, Argentina) surrounded by intensive agricultural activities and its relationship with a battery of biomarkers, including oxidative stress and genotoxicity, in two native species, the fish Oligosarcus jenynsii and the macrophyte Bidens laevis . A total of 26 pesticide residues were analyzed, and the main ones detected were glyphosate and its metabolite aminomethylphosphonic acid (AMPA), chlorpyrifos, and imidacloprid. In O. jenynsii , hydrogen peroxide (H 2 O 2 ) content in the liver increased with chlorpyrifos occurrence, while malondialdehyde (MDA) levels in the brain and liver increased with the presence of both chlorpyrifos and glyphosate. In B. laevis , H 2 O 2 and MDA levels in leaves and roots increased with AMPA occurrence. Also, leaf H 2 O 2 contents and root MDA levels increased with chlorpyrifos concentration. In contrast, catalase and peroxidase activities in roots decreased with AMPA and chlorpyrifos occurrence. In both species, mainly H 2 O 2 and MDA levels demonstrated their sensitivity to be used as biomarkers in the biomonitoring of current-use pesticide pollution in shallow lakes. Their use may provide information to plan strategies for environmental conservation by government institutions or decision-makers, and to assess the biota health status.

Abstract:Heavy metals can cause a significant damage to submerged macrophytes and affect its periphyton biofilms in aquatic environments. This study investigated the effects of heavy metals such as copper (Cu), lead (Pb), cadmium (Cd) and their mixture on physiological and biochemical responses and ultrastructure characteristics of Vallisneria natans (V. natans). Furthermore, differences in structures of microbial communities were observed in biofilms. The results showed that Cu2+, Pb2+, Cd2+ and their mixture could destroy cell structure and photosynthetic system, and directly caused oxidative damage to submerged macrophyte and induced antioxidant enzyme system. In general, biomass and total chlorophyll content of V. natans noticeably decreased, while the activities of superoxide dismutase, peroxidase and catalase were enhanced by heavy metal stress inducement in restricted range, and the malondialdehyde content increased with the aggravation of the damage. The single heavy metal stress played a negative impact, however, the combined stress was not always synergistic effects on plants. High-throughput sequencing analysis suggested that heavy metals changed the abundance and structure of the microbial biofilm community. Proteobacteria and Bacteroidete were the dominant bacteria under heavy metal stress and other species and abundance of bacteria such as Firmicute, Cyanobacteria, Chloroflexi, Actinobacteria, Verrucomicrobia, Acidobacteria, Deinococcus-Thermus, Chlamydiae were also present. These findings provided useful information for further understanding about submerged macrophytes and periphyton biofilms responsed to heavy metal stress in aquatic environments in the future.
