Abstract:Aiming at exploring the excellent structural performance of the vein-stiffening membrane structure of dragonfly hind wings, we analyzed two planar computational models and three 3D computational models with cambered corrugation based on the finite element method. It is shown that the vein size in different zones is proportional to the magnitude of the vein internal force when the wing structure is subjected to uniform out-of-plane transverse loading. The membrane contributes little to the flexural stiffness of the planar wing models, while exerting an immense impact upon the stiffness of the 3D wing models with cambered corrugation. If a lumped mass of 10% of the wing is fixed on the leading edge close to the wing tip, the wing fundamental frequency decreases by 10.7%∼13.2%; if a lumped mass is connected to the wing via multiple springs, the wing fundamental frequency decreases by 16.0%∼18.0%. Such decrease in fundamental frequency explains the special function of the wing pterostigma in alleviating the wing quivering effect. These particular features of dragonfly wings can be mimicked in the design of new-style reticulately stiffening thin-walled roof systems and flapping wings in novel intelligent aerial vehicles.

Abstract:Motor neurones in the thoracic ganglia of the locust were impaled with microelectrodes and identified according to the muscle they innervated and their other physiological properties. They were then labeled by intracellular injection of horseradish peroxidase and processed for electron microscopy. The nature and distribution of synapses on each motor neurone was examined and, by the use of reconstruction from serial sections, their spatial relationships revealed. The metathoracic fast extensor tibiae and mesothoracic tergosternal flight motor neurones have both output and input synapses on their neuropilar branches. These synapses are involved in serial, reciprocal, and recurrent relationships showing that the structural equivalent of a physiological synapse may be complex. The metathoracic slow extensor tibiae and anterior fast flexor tibiae motor neurones apparently have only input synapses within the neuropile.

Abstract:The internal fine structure of the wing veins is explored and the relationships between the structures and the functions of dragonfly wing veins are revealed. SEM photographs of the cross-sections of dragonfly wing veins have shown that: (a) the micro/nano structures vary along the axis of a vein, i.e. different cross-sections have different micro/nano structures. - (b) In a given cross-section, the micro/nano structures are at multi-levels and multi-scales. - (c) At a large scale, the structures of the veins are of diversities and disorders. The larger the size scale, the more complicated the structures and the higher are the diversities and disorders. The smaller the size scale, the simpler are the structures, and the higher are the unifications and orders. - (d) At nano scale, we may induce an unified assembling mode for the vein's structures, i.e. "nano fibres -> nano layers (or nano bunches)". - (e) Both the mechanical and the biological functions of the micro/nano structures of the veins are optimized.

Abstract:The morphologies of the motoneurons of the dorsolongitudinal and the three dorsoventral flight power muscles (DLM, DVM 1-3) of Calliphora were investigated by means of cobalt backfills and intracellular biocytin stainings. The DLM is innervated by four prothoracic motoneurons supplying the four ventral muscle fibers and one mesothoracic motoneuron supplying the two dorsal fibers. The three fibers of the DVM 1 and the two fibers of the DVM 2 are innervated by five mesothoracic motoneurons, whereas the two fibers of the DVM 3 are innervated by two prothoracic motoneurons. In general, the motoneurons of each muscle have a common ventral soma cluster located in a characteristic position on the ipsilateral side of the thoracic ganglion, show similar dendritic arborizations in the mesothoracic wing neuropil, and have the same axon pathway. Only the soma of the common motoneuron of two dorsal fibers of the DLM is situated dorsally in the contralateral hemiganglion. The motoneurons of each muscle were found to be strongly dye coupled with each other, indicating that they are connected by gap junctions. In addition, the motoneurons of each muscle establish characteristic coupling patterns with the motoneurons of the other flight power muscles on both sides of the thorax and with two bilateral groups of local mesothoracic interneurons. The revealed coupling patterns are assumed to be of major relevance for the generation the characteristic, rhythmic flight activity of the motoneurons described in previous studies.

Abstract:Animal movement is controlled by motor neurons (MNs), which project out of the central nervous system to activate muscles 1 . MN activity is coordinated by complex premotor networks that facilitate the contribution of individual muscles to many different behaviours 2,3,4,5,6 . Here we use connectomics 7 to analyse the wiring logic of premotor circuits controlling the Drosophila leg and wing. We find that both premotor networks cluster into modules that link MNs innervating muscles with related functions. Within most leg motor modules, the synaptic weights of each premotor neuron are proportional to the size of their target MNs, establishing a circuit basis for hierarchical MN recruitment. By contrast, wing premotor networks lack proportional synaptic connectivity, which may enable more flexible recruitment of wing steering muscles. Through comparison of the architecture of distinct motor control systems within the same animal, we identify common principles of premotor network organization and specializations that reflect the unique biomechanical constraints and evolutionary origins of leg and wing motor control.

Abstract:In most animals, the brain controls the body via a set of descending neurons (DNs) that traverse the neck. DN activity activates, maintains or modulates locomotion and other behaviors. Individual DNs have been well-studied in species from insects to primates, but little is known about overall connectivity patterns across the DN population. We systematically investigated DN anatomy in Drosophila melanogaster and created over 100 transgenic lines targeting individual cell types. We identified roughly half of all Drosophila DNs and comprehensively map connectivity between sensory and motor neuropils in the brain and nerve cord, respectively. We find the nerve cord is a layered system of neuropils reflecting the fly's capability for two largely independent means of locomotion -- walking and flight -- using distinct sets of appendages. Our results reveal the basic functional map of descending pathways in flies and provide tools for systematic interrogation of neural circuits.
Peripheral neuropathy in HIV patients: Treatment by decompression of peripheral nerves

Abstract:Peripheral neuropathy is the most common nerve disorder in human immunodeficiency virus (HIV) patients. Distal symmetrical sensory polyneuropathy (DSP) affects roughly one third of HIV patients. With the introduction of antiretrovirals, more patients are surviving longer, and chronic complications are surfacing. Three consecutive patients with at least a 5‐year history of HIV presented during the period from 2007 to 2009. All three patients were on antiretrovirals and had no other comorbid conditions such as spinal pathology or diabetes. All patients had symptoms of pain, numbness, and weakness. Quantitative sensory testing and/or electromyography/nerve conduction testing (EMG/NCT) were performed preoperatively and correlated with the presence of Tinel signs. Targeted nerve releases were performed in four extremities, for a total of 18 nerves. All three patients had symptomatic improvement at 1 year with a visual analog scale (VAS) reduction in pain by at least five points. Quantitative sensory testing showed improvement, as did two EMG/NCTs obtained postoperatively. This showed improvement in conduction velocity at the fibular tunnel and posterior tibial nerve at the tarsal tunnel. This is the first report of nerve decompressions in the lower and upper extremity of HIV patients in the literature outside of the median nerve in the carpal tunnel. © 2011 Wiley‐Liss, Inc. Hierarchical Dragonfly Wing: Microstructure-Biomechanical Behavior Relations

Abstract:The dragonfly wing, which consists of veins and membrane, is of biological hierarchical material. We observed the cross-sections of longitudinal veins and membrane using Environmental Scanning Electron Microscopy (ESEM). Based on the experiments and previous studies, we described the longitudinal vein and the membrane in terms of two hierarchical levels of organization of composite materials at the micro- and nano-scales. The longitudinal vein of dragonfly wing has a complex sandwich structure with two chitinous shells and a protein layer, and it is considered as the first hierarchical level of the vein. Moreover, the chitinous shells are concentric multilayered structures. Clusters of nano-fibrils grow along the circumferential orientation embedded into the protein layer. It is considered as the second level of the hierarchy. Similarly, the upper and lower epidermises of membrane constitute the first hierarchical level of organization in micro scale. Similar to the vein shell, the membrane epidermises were found to be a paralleled multilayered structure, defined as the second hierarchical level of the membrane. Combining with the mechanical behavior analysis of the dragonfly wing, we concluded that the growth orientation of the hierarchical structure of the longitudinal vein and membrane is relevant to its biomechanical behavior.

Abstract:Ground penetrating radar (GPR) is a promising technique for buried threat detection which provides a complimentary phenomenology to electro-magnetic induction (EMI) based sensing. However, many successful GPR-based buried threat detection algorithms require data collected both before and after an object of interest is encountered to make a declaration (typically this data is used to perform background normalization, or to adequately characterize the object's shape). Samples taken past an object of interest, but before a decision is made, constitute an algorithm's “spatial latency”. For vehicular mounted antennae arrays, where vehicle stopping distance is a function of vehicle dynamics, driver responsiveness, and algorithmic spatial latency, reducing an algorithm's spatial latency can increase overall system safety and help keep operators out of harm's way. In this work we propose a stochastic sampling algorithm that can help reduce spatial latency for a wide range of GPR-based buried threat detection algorithms.

Abstract:The ultrastructure of locust muscles with different function is examined: the pleuroaxillary flight steering muscle is compared with a typical flight (power muscle) and a typical leg muscle, in particular with respect to sarcomere length, tracheation, mitochondria, and sarcoplasmatic reticulum. The pleuroaxillary muscle exhibits some features characteristic of flight muscles but most of the ultrastructure resembles that of leg muscles. This is in agreement with the innervation of this muscle by an octopaminergic neuron, which also innervates leg muscles but no other flight muscles. It also supports the hypothesis that octopaminergic neurons are important metabolic regulators and that the above muscle types exhibit important differences in energy metabolism.

An Organic Junction Between the Vein and Membrane of the Dragonfly Wing

Abstract:A dragonfly wing consists of membranes and both longitudinal and cross veins. We observed the microstructure cross-section at several locations in the dragonfly wing using environmental scanning electron microscopy (ESEM). The organic nature of the junction between the vein and the membrane was clearly identifiable. The membrane was divided into two layers, the upper epidermis and the lower epidermis. These layers extend around the sandwich structure vein, and combine with the adjacent membrane at a symmetrical location along the vein. Thus, we defined this as an organic junction between the vein and the membranes. The organic junction is able to form a tight corrugation angle, which dramatically increases both the warping rigidity and the strength of the wing, but not the torsional rigidity. The torsional deformation is primarily controlled by the microstructure of the longitudinal veins, and is based on the relative rotation angle between the epidermal layer and the inner layer of the vein that forms the zigzag section.

Abstract:The motions that make up animal behavior arise from the interplay between neural circuits and the mechanical parts of the body. Therefore, in order to comprehend the operational mechanisms governing behavior, it is essential to examine not only the underlying neural network but also the mechanical characteristics of the animal's body. The locomotor system of fly larvae serves as an ideal model for pursuing this integrative approach. By virtue of diverse investigation methods encompassing connectomics analysis and quantification of locomotion kinematics, research on larval locomotion has shed light on the underlying mechanisms of animal behavior. These studies have elucidated the roles of interneurons in coordinating muscle activities within and between segments, as well as the neural circuits responsible for exploration. This review aims to provide an overview of recent research on the neuromechanics of animal locomotion in fly larvae. We also briefly review interspecific diversity in fly larval locomotion and explore the latest advancements in soft robots inspired by larval locomotion. The integrative analysis of animal behavior using fly larvae could establish a practical framework for scrutinizing the behavior of other animal species.

Abstract:At the onset of each flight bout in flies, neural circuits in the CNS must rapidly integrate multimodal sensory stimuli and synchronously engage hinges of the left and right wings for coordinated wing movements. Whereas anatomical and physiological investigations of flight have been conducted on larger flies, molecular genetic studies in Drosophila have helped identify neurons that mediate various levels of flight control. However, neurons that might mediate bilateral coordination of wing movements to precisely synchronize left and right wing engagement at flight onset and maintain their movement in perfect coordination at rapid frequencies during flight maneuvers remain largely unexplored. Wing coordination could be directly modulated via bilateral sensory inputs to motoneurons of steering muscles and/or through central interneurons. Using a Ca(2+)-activity-based GFP reporter, we identified three flight-activated central dopaminergic interneurons in the ventral ganglion, which connect to and activate motoneurons that innervate a pair of direct-steering flight muscles. The activation of these newly identified dopaminergic interneurons is context specific. Whereas bilateral wing engagement for flight requires these neurons, they do not control unilateral wing extension during courtship. Thus, independent central circuits function in the context of different natural behaviors to control the motor circuit for Drosophila wing movement.
Representation of Drosophila Larval Behaviors by Muscle Activity Patterns

Abstract:How muscle actions are coordinated to realize animal movement is a fundamental question in behavioral study. To obtain the overall muscular activity patterns accompanying behaviors at high spatiotemporal resolution is technically difficult. In this work, we used light sheet microscopy to simultaneously image and analyze the activity, length and orientation of Drosophila larval muscles across body segments at single muscle resolution in nearly free behaviors. For typical behavioral modes such as peristalsis, head cast and turning, larval muscles showed behavioral mode specific activity patterns. Unexpectedly, reorientation of larval head involves muscle tone in the apparently motionless posterior segments. With a STGCN(spatial temporal graph convolution neural network)-Generator model, sequence of larval behavioral poses outlined by morphological patterns of muscles could be accurately predicted based on the time series of ventral but not dorsal muscle activities, and vice versa. Laser ablation of ventral but not dorsal muscles interrupted peristaltic wave and undermined head cast in both frequency and amplitude. Our results provide a simplified muscle activity representation of soft body motion that can be used for probing the key components of animal motor control. ### Competing Interest Statement The authors have declared no competing interest.

Abstract:In this paper, the natural structures of a dragonfly wing, including the corrugation of the chordwise cross-section, the sandwich microstructure veins, and the junctions between the vein and the membrane, have been investigated with experimental observations, and the morphological parameters of these structural features are measured. The experimental result indicates that the corrugated angle among the longitudinal veins ranges from 80° to 150°, and the sandwiched microstructure vein mainly consists of chitin and protein layers. Meanwhile, different finite element models, which include models I and I* for the planar forewings, models II and II* for the corrugated forewings, and a submodel with solid veins and membranes, are created to investigate the effects of these structural features on the natural frequency/modal, the dynamical behaviors of the flapping flight, and the deformation mechanism of the forewings. The numerical results indicate that the corrugated forewing has a more reasonable natural frequency/modal, and the first order up-down flapping frequency of the corrugated wing is closer to the experimental result (about 27.00 Hz), which is significantly larger than that of the planar forewing (10.94 Hz). For the dynamical responses, the corrugated forewing has a larger torsional angle than the planar forewing, but a lower flapping angle. In addition, the sandwich microstructure veins can induce larger amplitudes of torsion deformation, because of the decreasing stiffness of the whole forewing. For the submodel of the forewing, the average stress of the chitin layer is much larger than that of the protein layer in the longitudinal veins. These simulative methods assist us to explain the flapping flight mechanism of the dragonfly and to design a micro aerial vehicle by automatically adjusting the corrugated behavior of the wing.

Abstract:This paper aims to reveal the multi-optimal mechanisms for dynamic control in dragonfly wings. By combining the Arnold circulation with such micro/nano structures as the hollow inside constructions of the pterostigma, veins and spikes, dragonfly wings can create variable mass, variable rotating inertia and variable natural frequency. This marvelous ability enables dragonflies to overcome the contradictory requirements of both light-weight-wing and heavy-weight-wing, and displays the multi-optimal mechanisms for the excellent flying ability and dynamic control capacity of dragonflies. These results provide new perspectives for understanding the wings’ functions and new inspirations for bionic manufactures.
Effects of Dragonfly Wing Structure on the Dynamic Performances

Abstract:The configurations of dragonfly wings, including the corrugations of the chordwise cross-section, the microstructure of the longitudinal veins and membrane, were comprehensively investigated using the Environmental Scanning Electron Microscopy (ESEM). Based on the experimental results reported previously, the multi-scale and multi-dimensional models with different structural features of dragonfly wing were created, and the biological dynamic behaviors of wing models were discussed through the Finite Element Method (FEM). The results demonstrate that the effects of different structural features on dynamic behaviors of dragonfly wing such as natural frequency/modal, bending/torsional deformation, reaction force/torque are very significant. The corrugations of dragonfly wing along the chordwise can observably improve the flapping frequency because of the greater structural stiffness of wings. In updated model, the novel sandwich microstructure of the longitudinal veins remarkably improves the torsional deformation of dragonfly wing while it has a little effect on the flapping frequency and bending deformation. These integrated structural features can adjust the deformation of wing oneself, therefore the flow field around the wings can be controlled adaptively. The fact is that the flights of dragonfly wing with sandwich microstructure of longitudinal veins are more efficient and intelligent.
Co-ordinating interneurones of the locust which convey two patterns of motor commands: their connexions with ventilatory motoneurones

Abstract:1. The interneurones which make widespread connexions with flight motoneurones also synapse upon ventilatory motoneurones so that in all 50 motoneurones receive synapses. They influence three aspects of ventilation; (a) the closing and opening movements of the thoracic spiracles, (b) some aspects of abdominal pumping movements and (c) the recruitment of some motoneurones controlling head pumping. 2. The two closer motoneurones of a particular thoracic spiracle receive the same excitatory synaptic inputs (EPSPs) during expiration. The EPSPs match those in appropriate flight motoneurones. 3. The closer motoneurones of each thoracic spiracle whose somata are in the pro-, meso- or metathoracic ganglia all receive the same excitatory synaptic inputs. These inputs are an adequate explanation of the pattern of spikes in the closer motoneurones. Both the slow ventilatory and fast rhythms of synaptic potentials are expressed as spikes; the slow as the overall expiratory burst of spikes and the fast as the groups of spikes within that burst. This establishes a ventilatory function for the interneurones. All thoracic closer motoneurones therefore receive the same excitatory commands which will tend to synchronize the movements of each spiracle. 4. Spiracular opener motoneurones are inhibited during expiration, their IPSPs matching the EPSPs in flight or closer motoneurones. Therefore the interneurones have reciprocal effects on the antagonistic motoneurones of the spiracles. 5. The interneurones synapse upon some motoneurones which control the pumping movements of the abdomen and which have their somata in the metathoracic or first unfused abdominal ganglion. Motoneurones in four separate ganglia therefore receive inputs from these interneurones. 6. The interneurones also synapse upon motoneurones which control an auxiliary form of ventilation, head pumping.
