Abstract:Aflatoxin M1 (AFM1) contamination in dairy products is a growing concern. In Iran, where dairy consumption is integral to daily life, understanding the extent of AFM1 exposure is crucial. This study presents a comprehensive human health risk assessment (HHRA) of AFM1 in six popular dairy products: yogurt, kashk, ice cream, doogh, butter, and tarkhineh through a systematic review. This study used Monte Carlo simulations to estimate the dietary exposure to AFM1 across different age groups. Other HHRA parameters, including liver cancer risk (LCR), hazard index (HI), and margin of exposure (MoE), were also used to explore safety concerns arising from AFM1 contamination. In addition, statistics, uncertainty, and sensitivity analysis for AFM1 in dairy products across various ages were performed. Our findings reveal varying levels of AFM1 contamination, with 67.54% of the products being contaminated with AFM1, of which 12.49% exceeded the regulatory limits set by the European Commission (EC) and Institute of Standards & Industrial Research of Iran (ISIRI) regulations. The results highlight the importance of strict monitoring and control measures to minimize AFM1 contamination and mitigate the associated health risks. This research is a valuable resource for food safety authorities and the dairy industry to enhance the safety and quality of dairy products in Iran.

Abstract:Background: Milk and dairy products are essential for global nutrition, and ensuring their safety is crucial. Aflatoxin M1 (AFM1), a carcinogen from contaminated animal feed, poses a significant threat to consumers. Objectives: This “health policy brief" highlights the critical need for meticulous and constant monitoring of Iran's animal feed quality and storage conditions to minimize AFM1 contamination in dairy products. Methods: Comprehensive review papers on AFM1 were reviewed in Iranian dairy products, drawing evidence from various databases. Results: The prevalence of AFM1 contamination in Iranian dairy products, particularly risky for children due to extensive consumption, exceeds safety limits, with livestock feed identified as the primary source. Public health demands rigorous monitoring practices, such as stricter feed quality and storage regulations, regular AFM1 testing, stakeholder collaboration, and public awareness. There are many benefits associated with these measures, including lowering AFM1 levels in dairy products, improving food safety standards, minimizing AFM1 exposure risks, especially for children, and increasing consumer confidence in the integrity of the Iranian dairy industry. Conclusions: Continuous monitoring of animal feed is crucial to safeguard public health and ensure the safety of Iranian dairy products, particularly for children. This comprehensive approach, collaboration, and public awareness are vital to mitigating AFM1 contamination and promoting Iran's safe and robust dairy industry.
The aflatoxin B1 content of peanut-based foods in Iran: a systematic review

Abstract:Abstract Backgrounds One of the common consumed snacks among Iranian is nuts. The aim of this systematic review was to determine the rate of peanut contamination with aflatoxin B1 (AfB1) in different provinces of the Iran. Materials and methods The research studies with keywords “aflatoxin B1”, “peanut”, “peanut butter”, “peanut oil”, “coated peanut”, “roasted peanut”, “snack peanut” were searched in PubMed, Scopus, Science Direct, Google Scholar and scientific information databases (SID), regardless of publication time. A total of 43 studies were obtained and only six articles were finally selected according to exclusion and inclusion criteria. Margin of Exposure (MOE) and Hazard Quotient (HQ) were also calculated to evaluate the oral exposure of AfB1 through peanuts and peanut-based products. Results The contamination of AFB1 in peanut was high in Mashhad and Tehran compared with the other cities. The value of MOE was calculated less than 10,000. The results of MOE indicate that there are chances of the risk of developing cancer and these products may not be safe. Therefore, AFB1 levels should be measured regularly in peanut products in large cities.

Abstract:Food contamination with aflatoxins is the focus of current concern. The i ntermittent incidence of these toxins in agricultural commodities has negative effects on the economy of the affected regions where harvest and postharvest techniques for the prevention of mold growth are seldom practiced. Aflatoxins are highly hepatotoxic, hepatocarcinogenic,teratogenic, and mutagenic in nature and they contaminate a wide variety of important agricultural commodities, such as peanuts, maize, rice and cottonseed, before, during and after harvest in various environmental conditions/settings. The biosynthesis of aflatoxins in nature depends upon the various factors such as carbon, nitrogen, temperature, water activity,pH, developmental stages, oxidative stress, and plant metabolites. These toxins can cause several ailments and can be controlled by blocking and interfering with the absorption and disturbing the metabolic enzymes. The present article overviews the biosynthesis, infection, transport, metabolism and toxicity of aflatoxins. Detailed studies on their biosynthesis,mechanism of toxicity, structure-function relationship, metabolism and transport will provide deep insight into the technology involved in the development of an effective control measures for aflatoxins.

Abstract:IntroductionAgricultural commodities contaminated by molds and mycotoxins can be considered as public health problems in less developed countries, particularly in Iran. Hence the main purpose of this study was to identify mold fungi and molecular analysis of the most important species of aflatoxin-B1-producing Aspergillus species in some dried nuts and grains in local markets in Tehran.Materials and methodsTwo hundred fifty samples of wheat, rice, corn, pistachios, and peanuts were collected from the five different locations of Tehran between January 2018 and January 2019. The samples were analyzed by using direct seed inoculation method and grain crushing method. Fungal strains were identified as Aspergillus spp. on the basis of morphological characters and further confirmed by using of β-tubulin gene sequencing. To differentiate between aflatoxigenic and non-aflatoxigenic Aspergillus spp., the isolates were screened for the presence of aflatoxigenic genes (nor-1, ver-1, omtA, and aflR).ResultsOne-handed forty-eight aflatoxigenic Aspergillus isolates (144 A. flavus and 4 A. parasiticus) were identified and aflR gene was the most frequent gene in these species. Five isolates (4 A. flavus, 1 A. parasiticus) had quadruplet pattern, 64 isolates (63 A. flavus, 1 A. parasiticus) had more than 1 gene and 39 isolates (38 A. flavus,1 A. parasiticus) did not have any genes.ConclusionAccording to the contamination of dried nuts and grains by some aflatoxigenic fungi, an extensive surveillance is necessary to provide a wider view on these products. Moreover, effective and efficient aflatoxin control program requires identifying and managing key elements that are effective in reducing mycotoxin production at farm level or in storage conditions.

Abstract:In this study, the average level of aflatoxin M1 in various types of milk from 107 articles (297 studies with 16,274 milk samples) were meta-analyzed using random-effect model based on the milk varieties (animal species and heating processes), geographical regions, seasons, detection techniques and dairy farming subgroups. Studies on milk contamination with aflatoxin M1 in Iran were collected using universal and Persian databanks from January 1974 to the end of November 2021. The overall aflatoxin M1 mean concentration and prevalence in milk samples of Iran were 39.65 ng/l (95% CI: 36.00–43.30) and 80% (95% CI: 76–85%), respectively. The rank order of importance of various variables in mean levels of aflatoxin M1 in milk samples included milk type (animal species) > geographical regions > detection techniques > dairy farming types > milk types (heating processes) > seasons. Findings revealed that the overall content of aflatoxin M1 in milk samples of Iran was lower than that allowed by the European Union, Institute of Standards and Industrial Research of Iran, and the USA, possibly due to the milk monitoring by the Iranian regulatory systems.

Abstract:Cheese is popular in Iran because of its high nutritional value; therefore, it is necessary to control this product regarding health risk factors, particularly aflatoxin M1 (AFM1). This research reviewed AFM1 in various varieties of cheese in Iran to assess the potential health risks associated with consuming these products for different age groups. In this regard, all accessible papers from different databases were screened between June 27, 2000 and October 10, 2022 b y systematic research and then considering the selection criteria of the studies; finally, 22 articles were selected for the current review. The amount and prevalence of AFM1 were calculated and separated based on the cheese variety, and the sampling location; health risk assessment (HRA), statistical, uncertainty, and sensitivity analysis for AFM1 of cheese for different age groups were performed. The study results for 2143 samples showed that the overall average AFM1 for cheese is 160 ± 175 ng/kg, below the European Commission (EC) regulation (250 ng/kg). AFM1 contaminated 72.42% of all cheese samples, and 13% of these contaminated samples had a higher AFM1 than the EC regulation. Cheese varieties were ranked based on average levels of AFM1 as white pasteurized > traditional > creamy > probiotic > Lighvan, and this ranking was obtained based on sampling locations as market > dairy factories > livestock farms. Based on the HRAs, from the perspective of the liver cancer risk (LCR), the margin of exposure (MOE), and the hazard index (HI) approach, it can be concluded that cheese produced in Iran, in terms of AFM1, particularly for children, poses serious health risks. Accordingly, it is imperative to carefully consider implementing suitable management methods to inhibit the growth of aflatoxin B1 (AFB1) in livestock fodder, and training in sanitary production and processing of dairy products according to world standards is suggested for industrial and traditional cheese producers across Iran.
Quantitative analysis and carcinogenic/non-carcinogenic risk assessment of aflatoxin M1 in milk-based baby food and infant formula milk – a case study in Iran

Abstract:In this study, solid-phase extraction (SPE) combined with the dispersive liquid–liquid microextraction based on novel hydrophobic deep eutectic solvent (DLLME − DES) has been developed as an ultra-pre-concentration technique for the extraction of aflatoxin M 1 (AFM1) in milk-based baby food (MBBF) and infant formula milk (IFM) samples followed by HPLC combined with fluorescence detection (HPLC − FL). In addition, carcinogenic and non-carcinogenic risk assessment was performed by health-related risk factors including liver cancer risk (LCR), margin of exposure (MOE) and target hazard quotient (THQ) were calculated using the mean of AFM1 in different infant food samples. The results of the study showed that the mean of AFM1 was statistically significant different between various brands and types of IFM and MBBF. The results of the study showed that the percentage of positive samples higher than the allowable limit of AFM1 in 36 samples of domestic infant formula milk (DIFM), 24 samples of imported infant formula milk (IIFM), 36 samples of domestic milk-based baby food (DMBBF) and 18 samples of imported milk-based baby food (IMBBF) were 41.6, 12.5, 66.7 and 33.3%, respectively. In addition, estimated values for health risk-related factors including LCR, MOE and THQ indicated that for most infants less than one-year-old were higher than the acceptable levels. Based on the results, it can be concluded that the quality of IFM and MBBF consumed in Iran in terms of AFM1 is poor. Therefore, it is necessary to take appropriate measures to reduce the amount of AFM1 in DIFM and DMBBF, and in addition, the IIFM and IMBBF should be controlled qualitatively before supplying the market.

Abstract:Aflatoxins (AFs) are the most important toxic, mutagenic, and carcinogenic fungal toxins that routinely contaminate food and feed. While more than 20 AFs have been identified to date, aflatoxin B1 (AFB1), B2 (AFB2), G1 (AFG1), G2 (AFG2), and M1 (AFM1) are the most common. Over 25 species of Aspergillus have been shown to produce AFs, with Aspergillus flavus, Aspergillus parasiticus, and Aspergillus nomius being the most important and well-known AF-producing fungi. These ubiquitous molds can propagate on agricultural commodities to produce AFs in fields and during harvesting, processing, transportation, and storage. Countries with warmer climates and that produce foods susceptible to AF contamination shoulder a substantial portion of the global AF burden. Pakistan's warm climate promotes the growth of toxigenic fungi, resulting in frequent AF contamination of human foods and animal feeds. The potential for contamination in Pakistan is exacerbated by improper storage conditions and a lack of regulatory limits and enforcement mechanisms. High levels of AFs in common commodities produced in Pakistan are a major food safety problem, posing serious health risks to the population. Furthermore, aflatoxin contamination contributes to economic losses by limiting exports of these commodities. In this review, recent information regarding the fungal producers of AFs, prevalence of AF contamination of foods and feed, current regulations, and AF prevention and removal strategies are summarized, with a major focus on Pakistan.

Abstract:Aflatoxin B1 (AFB1) is widespread and seriously threatens public health worldwide. This study aimed to investigate AFB1 in imported hazelnut samples in northwest of Iran (Eastern Azerbaijan Province) using High-Performance Liquid Chromatography with a Fluorescent Detector (HPLC-FLD). In all tested samples AFB1 was detected. The mean concentration of AFB1 was 4.20 μg/kg and ranged from 3.145 to 8.13 μg/kg. All samples contained AFB1 levels within the maximum acceptable limit except for one sample. Furthermore, the human health risk assessment of AFB1 from consuming imported hazelnuts by Iranian children and adults was evaluated based on the margin of exposure (MoE) and quantitative liver cancer risk approaches. The MoE mean for children was 2529.76, while for adults, it was 8854.16, indicating a public health concern. The present study found that the risk of developing liver cancer among Iranian children was 0.11100736 per 100,000 people, and in the Iranian adult population was 0.0314496 cancers per 100,000 people. Since environmental conditions potentially affect aflatoxin levels in nuts, countries are advised to monitor aflatoxin contents in imported nuts, especially from countries with a conducive climate for mold growth.

Abstract:A comparison of experimental data and two-dimensional numerical simulations of polysilicon thin-film transistors (TFTs) is presented. It is shown that avalanche multiplication causes both the kink effect in the output characteristics and the reduction of threshold voltage in short-channel device. It is shown that exactly the same physical model for avalanche multiplication gives very good agreement between simulations and experimental data for both these effects. It is demonstrated that it is the presence of grain boundaries or traps in the polysilicon that causes avalanche effects to be much greater than in comparable single-crystal silicon devices.<<ETX>>

Abstract:Pistachio nuts are among the commodities with the highest risk of aflatoxin contamination in Iran. Aflatoxin B1 (AFB1) is one of the most hazardous mycotoxins for humans and livestock. In nature, there are microorganisms which are capable of reducing aftatoxins contamination in food and feed products. In this study, Bacillus subtilis strain UTBSP1 was isolated from pistachio nuts and studied for the degradation of AFB1. The AFB1 contents were determined by the use of HPTLC and HPLC as well as multiple reactions monitoring (MRM) method in LC-MS/MS. The results indicated B. subtilis uTBSP1 could considerably remediate AFB1 from nutrient broth culture and pistachio nut by 85.66% and 05%, respectively. Cell free supernatant fluid caused an apparent 78.39% decrease in AFB1 content. The optimal conditions for AFB1 degradation by cell free supernatant appeared at 35-40 degrees C, during 24 h. Furthermore, the results indicated that AFB1 degradation is enzymatic and responsible enzymes are extracellular and constitutively produced. The destructive AFB1 differed from standard AFB1 chemically, and lost a fluorescence property. (C) 2011 Elsevier Ltd. All rights reserved.
Impacts of Dehydroacetic Acid and Ozonated Water on Aspergillus flavus Colonization and Aflatoxin B1 Accumulation in Iranian Pistachio

Abstract:Background: Aflatoxins (AFs) are the most prevalent carcinogenic mycotoxins, produced mainly by Aspergillus flavus and A. parasiticus. The general purpose of the present study was to use Dehydroacetic Acid (DHA) and Ozonated Water (OW) to control A. flavus growth and accumulation of Aflatoxin B1 (AFB1) in Iranian pistachios. Methods: Three treatment pistachio groups were separately immersed in DHA (1 N) and OW (10 mg/L), and also combination of DHA and OW. Then, each group was contaminated with 1×10 3 A. flavus cell suspensions. Also, sterile distilled water as well as cyclohexamide solutions were considered as negative and positive control, respectively. The solution inhibitory characteristics were assessed by enumeration of viable as well as cultivable fungal conidia on the samples. High Performance Liquid Chromatography (HPLC) technique was used for AFB1 determination. Data were analyzed by One way ANOVA and Tukey T-tests using SPSS, Inc, Chicago, IL software (v. 16.0). Results: DHA and combinations of DHA and OW groups which exhibited no growth of A. flavus, had the most antifungal activity (p<0.05) in comparison with the other groups. The highest AFB1 contamination content was seen in negative control samples (6.19±0.33 μg/kg) and also OW treated ones (6.1±0.32 μg/kg). The least AFB1 level was detected in DHA treated group (1.5±0.21 μg/kg) and combinations of DHA and OW (1.59±0.21 μg/kg). However, no AFB1 was found in positive control samples. Conclusion: We found that DHA showed antifungal and inhibitory activities more than OW in Iranian pistachios. Application of DHA is recommend for Iranian food industries as a potential mean for prevention of the fungi growth and AFB1 accumulation in pistachios nuts.

Abstract:Abstract Mycotoxins are secondary metabolites produced by several fungal species and molds. Under favorable conditions like high temperature and moisture, they contaminate a large number of food commodities and regional crops during pre and post-harvesting. Aflatoxin is the main mycotoxin that harm animal and human health due to its carcinogenic nature. Aflatoxins are mainly released by Aspergillus flavus and Aspergillus parasiticus . AFB1 constitutes the most harmful type of aflatoxins and is a potent hepato-carcinogenic, mutagenic, teratogenic and it suppresses the immune system. To maintain food safety and to prevent aflatoxin contamination in food crops, combined approaches of using resistant varieties along with recommended farming practices should be followed. This review concentrates on various aspects of mycotoxin contamination in crops and recent methods to prevent or minimize the contamination.

Abstract:Aflatoxins (AFs) are secondary metabolites produced by fungi, and they are deemed the most perilous mycotoxin and food safety predicament. The exposure of humans to mycotoxins transpires either directly through the consumption of contaminated agricultural commodities or indirectly through the ingestion of items derived from animals that have been nourished with tainted substances of animal origin. To ensure the detoxification of AFs in animal and plant food products and to mitigate the risks they pose to public health and the economy, diverse techniques (physical, chemical, and biological) have been subject to scrutiny. By altering and eradicating the molecular structure of the toxin, all of these approaches impede its transmission to the digestive system and potentially diminish the accessibility of toxins to the target tissue, ultimately eliminating them. Given the pervasive predicaments attributed to the contamination of foods and feeds by AFs, it is of utmost importance to urgently devise cost-effective and appropriate strategies to combat this hazard. This review highlights the concept of AFs, definitions, and benefits of postbiotics and their biological role in the detoxification of AFs, as well as their benefits in the food-pharmaceutical industry.

Abstract:Aflatoxins (AFTs) are toxic products of fungal metabolism, associated with serious health consequences and substantial economic losses to agriculture, livestock and poultry sectors, particularly in the developing countries. This review outlines the current information on AFTs in terms of historical background, classification, relative occurrence and co-existence with other mycotoxins in various food commodities. The phenomenon of aflatoxin (AFT) biosynthesis has been elucidated with reference to molecular basis, genetic regulation and factors affecting the AFT production. Moreover, the in vivo disposition kinetics, toxicological action and toxico-pathological consequences of AFTs have also been highlighted. Currently employed strategies for the detection and detoxification of AFTs, biomarkers of exposure assessment, potential economic impact and regulatory considerations regarding the AFTs have been emphasized.

Abstract:Rice (Oryza sativa L.) is one of the most consumed cereals that along with several important nutritional constituents typically provide more than 21% of the caloric requirements of human beings. Aflatoxins (AFs) are toxic secondary metabolites of several Aspergillus species that are prevalent in cereals, including rice. This review provides a comprehensive overview on production factors, prevalence, regulations, detection methods, and decontamination strategies for AFs in the rice production chain. The prevalence of AFs in rice is more prominent in African and Asian than in European countries. Developed nations have more stringent regulations for AFs in rice than in the developing world. The contamination level of AFs in the rice varied at different stages of rice production chain and is affected by production practices, environmental conditions comprising temperature, humidity, moisture, and water activity as well as milling operations such as de-husking, parboiling, and polishing. A range of methods including chromatographic techniques, immunochemical methods, and spectrophotometric methods have been developed, and used for monitoring AFs in rice. Chromatographic methods are the most used methods of AFs detection followed by immunochemical techniques. AFs decontamination strategies adopted worldwide involve various physical, chemical, and biological strategies, and even using plant materials. In conclusion, adopting good agricultural practices, implementing efficient AFs detection methods, and developing innovative aflatoxin decontamination strategies are imperative to ensure the safety and quality of rice for consumers.
Natural inhibitors: A sustainable way to combat aflatoxins

Abstract:Among a few hundred mycotoxins, aflatoxins had always posed a major threat to the world. Apart from A. flavus, A. parasiticus, and A. nomius of Aspergillus genus, which are most toxin-producing strains, several fungal bodies including Fusarium, Penicillium, and Alternaria that can biosynthesis aflatoxins. Basically, there are four different types of aflatoxins (Aflatoxin B1 (AFB1), Aflatoxin B2 (AFB2), Aflatoxin G1 (AFG1), Aflatoxin G2 (AFG2)) are produced as secondary metabolites. There are certainly other types of aflatoxins found but they are the by-products of these toxins. The fungal agents generally infect the food crops during harvesting, storing, and/or transporting; making a heavy post-harvest as well as economic loss in both developed and developing countries. And while ingesting the crop products, these toxins get into the dietary system causing aflatoxicosis, liver cirrhosis, etc. Therefore, it is imperative to search for certain ways to control the spread of infections and/or production of these toxins which may also not harm the crop harvest. In this review, we are going to discuss some sustainable methods that can effectively control the spread of infection and inhibit the biosynthesis of aflatoxins.

Abstract:Food contamination is a common phenomenon in the production, distribution and consumption of processed and agricultural commodities all over the world. Food safety is now taking the frontal stage in food production, processing and distribution. This study assessed the presence of aflatoxigenic fungi and mycotoxins in foods, occurrence, control, socio-economic and health implications. This study also gave comprehensive information on the safety and mycological quality of foods as well the effect on the people and economy of various countries. The review revealed the various aflatoxigenic fungi in foods, their distribution in agricultural produce and their effects on reduction in yield and quality with attendant consumer health implications and resulting significant economic losses. In developing countries, majority of the people do not know the inherent dangers of consuming mouldy produce or food contaminated with fungi and moulds with possible contamination by mycotoxigenic fungi because of lack of awareness about the danger involved. In view of this, there is need for general and public education to sensitise the people on the economic and health hazards posed by mycotoxins. Control measures such as good agricultural practices, crop selection, proper washing and cooking practices of food commodities should be emphasized to the people. Regulatory control, fast and effective analyses and detection, good produce handling and storage should be encouraged as this will assist in mitigating the side effects of mycotoxins in foods particularly in the tropical and sub-tropical countries, and in Africa nations where there is enabling environment that promotes fungal growth, shortage of food and drought of modern storage and processing infrastructures.

Abstract:Transcription factor IID (TFIID) recognizes the TATA element of promoters transcribed by RNA polymerase II (RNAPII) and serves as the base for subsequent association by other general transcription factors and RNAPII. The carboxyl-terminal domain of TFIID is highly conserved and contains an imperfect repetition of a 60-amino acid sequence. These repeats are separated by a region rich in basic amino acids. Mutagenesis of the lysines in this region resulted in a conditioned phenotype in vivo, and the mutant proteins were defective for interactions with transcription factor IIA in vitro. Binding of TFIID to DNA was unaffected. These results suggest that the basic domain of TFIID is important for protein-protein interactions.
