Abstract:Canonical analysis has long been the primary analysis method for studies of phase transitions. However, this approach is not sensitive enough if transition signals are too close in temperature space. The recently introduced generalized microcanonical inflection-point analysis method not only enables the systematic identification and classification of transitions in systems of any size, but it can also distinguish transitions that standard canonical analysis cannot resolve. By applying this method to a generic coarse-grained model for semiflexible polymers, we identify a mixed structural phase dominated by secondary structures such as hairpins and loops that originates from a bifurcation in the hyperspace spanned by inverse temperature and bending stiffness. This intermediate phase, which is embraced by the well-known random-coil and toroidal phases, is testimony to the necessity of balancing entropic variability and energetic stability in functional macromolecules under physiological conditions.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is about the structural phase - transition behavior of semi - flexible polymers under different temperature and bending - stiffness conditions. Specifically, the authors used a new statistical analysis method - the generalized microcanonical inflection - point analysis method - to systematically identify and classify these phase transitions, especially in cases where the subtle phase - transition signals cannot be resolved by the standard canonical analysis method. The paper points out that the traditional canonical analysis method is not sensitive enough when studying phase transitions, especially when the phase - transition signals are very close in the temperature space. The newly introduced generalized microcanonical inflection - point analysis method can not only systematically identify and classify the phase transitions of any - sized systems, but also distinguish those phase transitions that cannot be distinguished by the standard canonical analysis method. By applying this method to a general coarse - grained model of semi - flexible polymers, the authors identified a mixed - structure phase dominated by secondary structures (such as hairpins and loops). This phase transition originates from the bifurcation in the hyperspace composed of the inverse temperature and bending stiffness. This intermediate phase is located between the known random - coil phase and the looped phase, demonstrating the importance of functional macromolecules in balancing entropy change and energy stability under physiological conditions. In short, this research aims to reveal the complex phase - transition behavior of semi - flexible polymers under specific conditions through advanced statistical analysis methods, especially those phase transitions that are difficult to detect by traditional methods. This helps to gain a deeper understanding of the folding mechanisms of biomacromolecules in cells and the ways in which their functions are realized.
