Abstract:In this work, we present six new zinc(II) complexes with the general formulas [Zn 2 (oligopyridine) 2 L 4 ] and [Zn(oligopyridine) 2 L 2 ], where L – = 5-(4-chlorophenyl)-1H-tetrazolate anion, and oligopyridine = 1,10-phenanthroline or 2,2′-bipyridine derivatives (dmphen – 4,7-dimethyl-1,10-phenanthroline, phen – 1,10 phenanthroline, dmbipy – 2,2′-bi-4-picoline, bipy – 2,2′-bipyridine), which have been synthesized with high yield and characterized by IR spectroscopy, elemental, thermogravimetric, single crystal and powder X-ray analyses. A distorted square-pyramidal and octahedral geometry of zinc(II) ion have been demonstrated by single crystal X-ray diffraction analysis. The tetrazolate ligand exhibits two types of coordination: monodentate, and bidentate-bridging via the N(2), N(3) or N(1), N(2) atoms. The behavior of all complexes in solution has been investigated by UV–Vis and the 1 H, 13 C NMR spectroscopy, conductometry, and mass spectrometry. The antimicrobial study of the compounds has been carried out against E. coli , St. aureus , P. italicum , and C. steinii, and [Zn 2 (dmbipy) 2 L 4 ] has been shown to possess significant protistocidal properties. According to the cytotoxicity study,all complexes don't have cytotoxic properties in 1–100 μM concentration range against tumor human cell lines (larynx carcinoma Hep2, hepatocellular carcinoma HepG2 and breast adenocarcinoma MCF-7) with the exception of complex 5 which has displayed significant cytotoxic effect (LC 50 = 19.8 ± 2.4 μM) against Hep2 cells. However, obtained compounds have a pronounced cytostatic effect on all tumor cell lines.

Abstract:Two zinc(II) complexes with dimethyl 2,2′-bipyridine-4,5-dicarboxylate (py-2py) of the general formula [Zn(py-2py)X2], X = Cl− (1) and Br− (2) were synthesized and characterized by NMR, IR and UV-Vis spectroscopy and single-crystal X-ray diffraction analysis. Complexes 1 and 2 are isostructural and adopt a slightly distorted tetrahedral geometry with values of tetrahedral indices τ4 and τ’4 in the range of 0.80–0.85. The complexes were evaluated for their in vitro antimicrobial activity against two bacterial (Pseudomonas aeruginosa and Staphylococcus aureus) and two fungal strains (Candida albicans and Candida parapsilosis), while their cytotoxicity was tested on the normal human lung fibroblast cell line (MRC-5) and the model organism Caenorhabditis elegans. Complex 1 showed moderate activity against both Candida strains. However, this complex was twofold more cytotoxic compared to complex 2. The complexes tested had no effect on the survival rate of C. elegans. Complex 2 showed the ability to inhibit filamentation of C. albicans, while complex 1 was more effective than complex 2 in inhibiting biofilm formation. The interactions of complexes 1 and 2 with calf thymus DNA (ct-DNA) and bovine serum albumin (BSA) were studied to evaluate their binding affinity toward these biomolecules.
Zinc and Cadmium Complexes of Pyridyl-Benzotriazole: Syntheses, Structures and Supramolecular Interaction

Abstract:Five novel complexes with two pyridine substituted benzotriazole ligands, 1-(2-pyridyl)benzotriazole (L1) and 1-(4-pyridyl)benzotriazole (L2), [Zn(L-1)(2)Cl-2](1), [{Zn(L-1)(2)Cl-2} (C)(L1)2](2), [Zn(L-2)(2)Cl-2](3), [{Zn(L2)(H2O)(3)(mu(2)-SO4)}center dot H2O] (4), and [{Cd(L2)(H2O)(3)(mu(2)-SO4)}center dot H2O](5) were synthesized. The details of the structures were characterized by X-ray single crystal analysis, revealing that these complexes were assembled together via supramolecular interaction, such as, hydrogen bonding and pi-pi interactions. Abstract:Four new Zn(II) and Cu(II) coordinated polymers ([ZnL2N3]ClO4 (1), [Cu2L2(CH3CN)]Cl4 (2) [CuL](NO3)2 (3), [Cu(H2O)L](SO4) (4) L=2-((4-phenyl-2H-1,2,3-triazol-2-yl)methyl) pyridine (ptmp)) have been reported. All the compounds have been characterized by IR spectrum, elemental analyses and X-ray crystallography diffraction. Single-crystal X-ray diffraction analyses show that one-dimensional polymers are formed in these four complexes. Chain-like structures are formed in complex 1, 2 and 3, which are connected by azide, chloride and nitrate anions, respectively. In complex 4, one-dimensional left-handed polymer is formed by a μ2-SO4 bridge. The fluorescent and electrochemical properties of these four complexes were investigated. It was found that these three Cu(II) complexes displayed a quenching of fluorescence, while Zn(II) complex exhibited a clear enhanced fluorescence.

Abstract:Zn-based MOFs, [Zn(formato)2(4,4´-bipy)] (4,4′-bipy = 4,4′-bipyridine) (1), had been successfully synthesized and characterized by elemental analysis (CHN), single crystal X-ray analysis and Single crystal X-ray diffraction (XRD), thermogravimetric analysis (TGA) and Fourier Transform Infrared Spectroscopy (FTIR). Then, the antimicrobial activities of complex 1 against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) were evaluated by measuring growth inhibition curves and the zones of inhibition. The experimental data showed that 1 exhibited superior antibacterial activities in comparison with the zinc ion and the formic acid ligand as well as the 4,4′-bipy ligand, and the mechanism of action might be associated with the generation of reactive oxygen species (ROS).

Abstract:Preparation and crystal structure of {4-chloro-2-[(2-morpholinoethylimino)methyl]phenolato} methanolchlorozinc(II) are reported. The X-ray structure reveals highly distorted square pyramidal geometry around zinc, binding to one phenolate O and two imine N atoms of the Schiff base, one methanol and one chloride. The complex and its ligand were tested in vitro for antibacterial and cytotoxic activity with a wide range of bactericidal activity and significant cytotoxic activity.

Abstract:Three new zinc(II) complexes, [ZnCl2L]·CH3OH (1), [ZnClL(NCS)]·2CH3OH·0.5H2O (2), and [ZnL(NCS)2]·CH3OH·H2O (3), where L is the zwitterionic form of 5-bromo-2-(((2-piperazin-1-yl)ethyl)imino)methyl)phenol, NCS is thiocyanate anion, were facile prepared by reaction of different molar ratio of L, zinc chloride and ammonium thiocyanate in methanol. The complexes were characterized by IR and UV-Vis spectroscopy. Detailed structures of the three complexes were confirmed by single crystal X-ray determination. The Zn atoms in the complexes are in tetrahedral coordination. The Schiff base ligand coordinates to Zn atom through phenolate oxygen atom and amino and imino nitrogen atoms. The remaining two sites are occupied by two Cl for 1, one Cl and one NCS for 2, and two NCS for 3. The compounds show significant antimicrobial activities.

Complex 1 is a mononuclear zinc(II) compound, with the Zn(II) in tetrahedral coordination. Complexes 2 , 3 and 4 are acetate, azide and dicyanamide bridged polymeric zinc(II) compounds, respectively, with the Zn(II) in distorted trigonal bipyramidal coordination. Complex 5 is an azide bridged dinuclear nickel(II) compound, with the Ni(II) ion having octahedral coordination. The compounds were evaluated for their antibacterial activities on Bacillus subtilis , Staphylococcus aureus , Escherichia coli and Pseudomonas fluorescens and antifungal activities on Candida albicans and Aspergillus niger . Complexes 1 and 2 showed the most effective activity against B. subtilis and E. coli , with MIC values of 1.2‚Äď4.7‚ÄČ őľ M.

Abstract:In an attempt to increase the biological activity of the 1,2,4-triazolo[1,5-a]pyrimidine scaffold through complexation with essential metal ions, the complexes trans-[Cu(mptp)2Cl2] (1), [Zn(mptp)Cl2(DMSO)] (2) (mptp: 5-methyl-7-phenyl-1,2,4-triazolo[1,5-a]pyrimidine), [Cu2(dmtp)4Cl4]·2H2O (3) and [Zn(dmtp)2Cl2] (4) (dmtp: 5,7-dimethyl-1,2,4-triazolo[1,5-a]pyrimidine), were synthesized and characterized as new antiproliferative and antimicrobial species. Both complexes (1) and (2) crystallize in the P21/n monoclinic space group, with the tetrahedral surroundings generating a square-planar stereochemistry in the Cu(II) complex and a tetrahedral stereochemistry in the Zn(II) species. The mononuclear units are interconnected in a supramolecular network through π–π interactions between the pyrimidine moiety and the phenyl ring in (1) while supramolecular chains resulting from C-H∙∙∙π interactions were observed in (2). All complexes exhibit an antiproliferative effect against B16 tumor cells and improved antibacterial and antifungal activities compared to the free ligands. Complex (3) displays the best antimicrobial activity against all four tested strains, both in the planktonic and biofilm-embedded states, which can be correlated to its stronger DNA-binding and nuclease-activity traits.

Abstract:4-Benzoyl-3-methyl-1-phenyl-5-pyrazolone (HPMBP) was synthesized via the Friedel–Crafts reaction and characterized through 1 H-NMR spectroscopy, Infrared spectroscopy and Electrospray ionization time-of-flight(ESI-TOF). The Cu(II) and Zn(II) coordination complexes, namely, [Cu(HPMBP)(2,2′-bpy)(Cl)]·H 2 O(1) and [Zn(HPMBP)(2,2′-bpy)(Cl)]·H 2 O(2), were synthesized in CH 3 OH(HPMBP:2,2′-bipyridine:M=1:1:1, M=Cu, Zn). Single crystal X-ray diffraction(SCXRD) studies show that Cu(II) and Zn(II) exhibit a distorted square pyramidal configuration through two oxygen atoms of HPMBP, two cis-nitrogen atoms of 2,2′-bipyridine and monodentate chloride ion. Moreover, the Hirshfeld surface analysis of the in complexes was performed using crystallographic calculations. Furthermore, the antibacterial activities of 1 and 2 are more potent than HPMBP, in particular, the antimicrobial properties of 2 are better than those of 1. Scanning electron microscopy (SEM) indicated that 1 had a regular strip shape, while 2 had an irregular circular shape. Our work provides a theoretical basis for studying the antibacterial activity of transition metal ion complexes.

Tris-(2-pyridyl)-pyrazolyl Borate Zinc(II) Complexes: Synthesis, DNA/Protein Binding and In Vitro Cytotoxicity Studies

Abstract:Zn(II) complexes bearing tris[3-(2-pyridyl)-pyrazolyl] borate (Tppy) ligand (1–3) was synthesized and examined by spectroscopic and analytical tools. Mononuclear [TppyZnCl] (1) has a Zn(II) centre with one arm (pyrazolyl-pyridyl) dangling outside the coordination sphere which is a novel finding in TppyZn(II) chemistry. In complex [TppyZn(H2O)][BF4] (2) hydrogen bonding interaction of aqua moiety stabilizes the dangling arm. In addition, solution state behaviour of complex 1 confirms the tridentate binding mode and reactivity studies show the exogenous axial substituents used to form the [TppyZnN3] (3). The complexes (1–3) were tested for their ability to bind with Calf thymus (CT) DNA and Bovine serum albumin (BSA) wherein they revealed to exhibit good binding constant values with both the biomolecules in the order of 104–105 M−1. The intercalative binding mode with CT DNA was confirmed from the UV-Visible absorption, viscosity, and ethidium bromide (EB) DNA displacement studies. Further, the complexes were tested for in vitro cytotoxic ability on four triple-negative breast cancer (TNBC) cell lines (MDA-MB-231, MDA-MB-468, HCC1937, and Hs 578T). All three complexes (1–3) exhibited good IC50 values (6.81 to 16.87 μM for 24 h as seen from the MTS assay) results which indicated that these complexes were found to be potential anticancer agents against the TNBC cells.

Abstract:The (thiazol-2-yl)hydrazone derivatives from 2-, 3-pyridine named (E)-4-(pyridin-2-yl)-2-(2-(pyridin-2-ylmethylene)hydrazinyl)thiazole (L1), (E)-4-(pyridin-3-yl)-2-(2-(pyridin-3- ylmethylene)hydrazinyl)thiazole (L2), and their three transition metal complexes named [Cd(L1)(NO3)2] (1), [Ni(L1)2(NO3)2] (2) and [Zn(L2)2(PF6)2] (DMF) 12H2O (3) were synthesized and characterized by elemental analysis (EA), infrared spectra (IR) and single-crystal diffraction. Both complex 1 and 2 featured a mononuclear molecular structure, while complex 3 possessed an intricate 3D metal-organic framework. The antibacterial and antitumor activity of the ligands and complexes were tested. The results showed some compounds had absolute specificity for certain bacteria or cancer cell line, which implied a good application prospect in pharmaceutical use.

Zinc(II) coordination compound with N'-(pyridin-2-ylmethylene)nicotinohydrazide: Synthesis, crystal structure, computational and cytotoxicity studies

Abstract:In this work, we report on the synthesis of a novel zinc(II) coordination compound [ZnL2] (1), which was readily obtained from the reaction of Zn(OAc)·2H2O and N'-(pyridin-2-ylmethylene)nicotinohydrazide (HL) in methanol. Recrystallization of 1 from dimethylformamide under ambient conditions allowed to produce yellow block-like crystals of 1·H2O. Complex 1·H2O was characterized by FT-IR and 1H NMR spectroscopy, while its optical properties were studied by UV-vis and spectrofluorimetry in methanol. The crystal structure of the title complex was revealed by single crystal X-ray diffraction and further explored in detail by the Hirshfeld surface analysis. Theoretical investigations based on the DFT calculations have also been applied to show the electronic properties of complex 1. The antitumor activities of the parent ligand HL and complex 1 were studied using Dalton's lymphoma malignant cancer model. Both compounds were found to induce concentration-dependent cytotoxicity and apoptotic cell death, leading to a decrease in cell viability, body weight, and tumor volume in mice with the superior activity of complex 1 over HL. Mice treated with complex 1 demonstrated an increase in life span with a survival period of 23 days. Finally, using a molecular docking approach, we have probed complex 1 to inhibit the recombinant mouse tumor-necrosis factor alpha (mTNF).

Abstract:Two Schiff base complexes [Cu(L)(2)] (1) and [Zn(L)(2)]center dot 3H(2)O (2) of (E)-4-fluoro-2-((3-morpholino propylimino)methyl)phenol (HL) have been synthesized and characterized by elemental analysis and single-crystal X-ray diffraction. Crystal data for 1: triclinic, space group P (1) over bar with a = 4.8000(10), b = 11.109(2), c = 13.921(3) angstrom, alpha = 66.55(3), beta = 80.35(3), gamma = 81.09(3)degrees, V = 668.2(2) angstrom(3), Z = 1, D-c = 1.477 g/cm(3), F(000) = 311, and the final R = 0.0744 and wR = 0.1601 for 2301 observed reflections with I > 2 sigma(I). And those for 2: monoclinic, space group P2(1)/c with a = 14.789(3), b = 11.713(2), c = 18.846(4) angstrom, beta = 107.85(3)degrees, V = 3107.4(11) angstrom(3), Z = 4, D-c = 1.389 g/cm(3), F(000) = 1368, and the final R = 0.0749 and wR = 0.1348 for 5423 observed reflections with I > 2 sigma(I). Both 1 and 2 are four-coordinated complexes, with a square-planar geometry of copper(II) in 1 and a slightly distorted tetrahedral geometry of zinc(II) in 2. An asymmetric unit consists of only one half of a Cu ion and one L ligand in 1, while an asymmetric unit is composed of one Zn ion, two L ligands and three water molecules in 2. Non-classical C-H center dot center dot center dot O and C-H center dot center dot center dot F hydrogen bonds play important roles in the formation of a 1D chain of 1, but O-H center dot center dot center dot O and O-H center dot center dot center dot N hydrogen bonds play dominant roles in the self-assembly of a 3D network of 2. The antibacterial activities of 1 and 2 against B. subtilis, S. aureus, S. faecalis, P. aeruginosa, E. coli and E. cloacae have been evaluated by MTT method.

It crystallizes in the monoclinic system, space group P2(1)/c with a = 9.0103(11), b = 17.0276(18), c = 9.2288(11) angstrom, beta = 101.309(6)degrees, M-r = 346.51, V = 1388.4(3) angstrom(3), Z = 4, D-c = 1.658 g/cm(3), F(000) = 696, mu = 2.143 mm(-1), the final R = 0.0492 and wR = 0.1236 for 2545 observed reflections with I > 2 sigma(I). The Zn atom is four-coordinated by two N and two Cl atoms, forming a slightly distorted tetrahedral geometry. Each zigzag chain links its adjacent symmetry-related equivalents via multiple pi-pi interactions, which are not frequently observed in the previously reported architectures.

Abstract:A new dizinc(II) complex with 6,20-bis(2-hydroxyethyl)-3,6,9,17,20,23-hexaaza-tricyclo [23,3,1,111,15]-triaconta-1(29),11(30),12,14,25,27-hexaene (L) was synthesized and characterized. The crystal structure of [Zn2H−2L](ClO4)2 has indicated that both Zn2+ ions are in penta-coordination environment with distorted trigonal bipyramid consisting of three N atoms from diethylenetriamine moieties, one internal O atom of the hydroxyethyl pendant, one intermolar O atom from another hydroxyethyl pendant, and the separation of Zn⋯Zn is 3.05 Å. The two aromatic rings are parallel to each other, with the centroid–centroid distance 9.21 Å. ES–MS spectra showed the main species of [Zn2H−2L]2+, [Zn2H−2L(H2O)2]2+, and [Zn2H−2L(ClO4)]+. The hydrolysis of 4-nitrophenyl acetate catalyzed by [Zn2H−2L]2+ in 10% (v/v) CH3CN at 25 °C, with I=0.10 (NaNO3) and pH 9.0, has shown a second-order rate constant of 0.021M−1s−1.

Abstract:Two polynuclear zinc(II) complexes, [Zn2(L1)2(OH2)2] (1) and [ZnL2(μ1,1-N3)]n (2), where L1 is the dianionic form of N,N'-bis(3,5-difluoro-2-hydroxybenzylidene)-1,3-diaminopropane, and L2 is the monoanionic form of 2-[(2-dimethylaminoethylimino)methyl]-4,6-difluorophenol, have been prepared and structurally characterized by elemental analyses, IR and UV-Vis spectroscopy, as well as single-crystal X-ray diffraction. In complex 1, the Zn atom is in octahedral coordination, with the donor atoms of the Schiff base ligands L1 and one water O atom. In complex 2, the Zn atom is in trigonal-bipyramidal coordination, with the three donor atoms of the Schiff base ligand L2 and two azido N atoms. The complexes have strong antibacterial activity against B. subtilis and S. aureus.

Abstract:A new macrocyclic dinuclear Zn(II) complex is synthesized by the cyclocondensation between 3,3'-(ethane-1,2-diylbis (benzylazanediyl))bis(methylene) bis (2-hydroxy-5-methyl-benzaldehyde) and 1,3-diaminopropane and the subsequent with zinc salts. The as-prepared complex is characterized by elemental analysis, FT-IR, ES-MS and single crystal X-ray diffraction. The complex consists of one independent molecular unit of [Zn2(μ-OAC)L]+ ClO4− and two water molecules with one on a symmetry element. Four of these units collectively form two pairs of dinuclear complex units of [Zn2(μ-OAc)L]+ with one unit sharing two perchlorate ions and one water molecule and the other unit sharing two perchlorate ions and one symmetry equivalent water molecule. Each zinc(II) ion exhibits a slightly distorted square pyramidal geometry. Two Zn atoms are bridged by two oxygen atoms of an acetate anion and two phenolic O atoms. The Zn–Zn distance is 2.951 Å. An ordered two-dimensional network is formed by weak intermolecular interactions in the crystal structure of the complex. The phosphate ester hydrolysis activity and the antibacterial activity of the complex are studied. The results showed that the synthesized complex has an efficient catalytic activity of phosphoester bond cleavage and the potential to inhibit the growth of E. coli. The optimum catalytic rate constant (kcat) at pH 7.8 for the hydrolysis of 4-nitrophenyl phosphate disodium salt hexahydrate (pNPP) by the synthesized complex is 2.98 × 10−4 s−1, and 105 times faster than that the spontaneous hydrolysis of the phosphate monoester.

Abstract:A series of Zn(II) complexes, [Zn(MTS)2(tmeda)] (1), [Zn(MTS)2(2,2-bipy.)] (2), [Zn(MTS)2(1,10-phen.)] (3) and [Zn(MTS)2(4,4-bipy.)2]n (4), (where MTS = methylthiosalicylate, and tmeda = tetramethyl ethylenediamine, bipy.= bipyridyl, phen=phenanthroline ), have been synthesized and characterized by single crystal X-ray analysis and other spectroscopic techniques. The geometry of complex 3 is distorted square pyramidal around the Zn center, while complex 2 has distorted trigonal bipyramidal geometry. Complexes 1 and 4 show distorted tetrahedral geometry in which complex 4 has a zig-zag polymeric structure. Due to differences in geometries, these Zn complexes are expected to exhibit differences in their biological activities. We have carried out studies on DNA and protein binding, anticancer activity as well as cytotoxicity of these complexes. The anticancer activities were cell-line dependent, however, the complex 4 was found to be most active.
