Abstract:Eumelanin is responsible for photoprotection in living organisms. It is made of 5,6-dihydroxyindole (DHI) oligomers. However, lack of detailed structural knowledge limits understanding its function and exploiting its potential in material science. To uncover the relationship between structural stability and optical properties, we have studied a virtual library of 830 DHI dimers. We find a preference for oxidized, polycyclic structures which speaks in favor of graphite-like structures for the larger oligomers, and propose an electrocyclic formation mechanism. Besides widely considered quinone oxidation patterns, also structures with interfragment double bonds and zwitterionic resonance structures are stable. Future theoretical melanine models will have to cover this diversity, and we introduce a new representative set of 49 stable dimers. Some stable oxidized dimers have absorption energies as low as 1.3 eV. They may be present as substructures in the naturally found oligomers and contribute to the absorption spectrum of the biopolymer.

Abstract:Melanin is responsible, in Nature, for photoprotection, for this reason it is expected to be poorly photoreactive. However, the photo‐reactivity of melanin and related materials is well documented. Here we discuss some relevant recent examples to demonstrate that, indeed, the actual mechanism of interaction of melanin with light is complex and still not completely understood. Photochemical and photothermal processes are involved, giving a contribution that strongly depends on light wavelength and intensity. Moreover, some interesting experiments demonstrated that photochemical reactivity of melanin related compounds is likely to be indirect, in the sense that the effect of light is to increase the number of radical species rather than creating photoreactive excited state. These suggestions open‐up new perspectives in the interpretation of the role of melanin in photoprotection and in the design of new melanin based photoactive materials for energy conversion, environmental remediation, and nanomedicine. Further complication is given by the role of atmospheric oxygen and humidity in the photoinduced processes. Beside this complexity of behavior makes it difficult a systematic understanding of the interaction of melanin with light, it surely strongly contributes to make the properties of melanin and related materials unique.

Abstract:Melanin is the dominant light absorber in retinal pigment epithelium (RPE). The loss of RPE melanin is a sign of ocular senescence and is both a risk factor and a symptom of age-related macular degeneration (AMD). Quantifying the RPE melanin concentration provides insight into the pathological role of RPE in ocular aging and the onset and progression of AMD. The main challenge in accurate quantification of RPE melanin concentration is to distinguish this ten-micrometer-thick cell monolayer from the underlying choroid, which also contains melanin but carries different pathognomonic information. In this work, we investigated a three-dimensional photoacoustic microscopic (PAM) method with high axial resolution, empowered by broad acoustic detection bandwidth, to distinguish RPE from choroid and quantify melanin concentrations in the RPE ex vivo. We first conducted numerical simulation on photoacoustic generation in the RPE, which suggested that a PAM system with at least 100-MHz detection bandwidth provided sufficient axial resolution to distinguish the melanin in RPE from that in choroid. Based on simulation results, we integrated a transparent broadband micro-ring resonator (MRR) based detector in a homebuilt PAM system. We imaged ex vivo RPE-choroid complexes (RCCs) from both porcine and human eyes and quantified the absolute melanin concentrations in the RPE and choroid, respectively. In our study, the measured melanin concentrations were 14.7 mg/mL and 17.0 mg/mL in human and porcine RPE, and 12 mg/mL and 61 mg/mL in human and porcine choroid, respectively. This study suggests that broadband PAM is capable of quantifying the RPE melanin concentration from RCCs ex vivo.

New Aspects Regarding the Fluorescence Spectra of Melanin and Neuromelanin in Pigmented Human Tissue Concerning Hypoxia

Abstract:Melanin is a crucial pigment in melanomagenesis. Its fluorescence in human tissue is exceedingly weak but can be detected through advanced laser spectroscopy techniques. The spectral profile of melanin fluorescence distinctively varies among melanocytes, nevomelanocytes, and melanoma cells, with melanoma cells exhibiting a notably "red" fluorescence spectrum. This characteristic enables the diagnosis of melanoma both in vivo and in histological samples. Neuromelanin, a brain pigment akin to melanin, shares similar fluorescence properties. Its fluorescence can also be quantified with high spectral resolution using the same laser spectroscopic methods. Documented fluorescence spectra of neuromelanin in histological samples from the substantia nigra substantiate these findings. Our research reveals that the spectral behavior of neuromelanin fluorescence mirrors that of melanin in melanomas. This indicates that the typical red fluorescence is likely influenced by the microenvironment around (neuro)melanin, rather than by direct pigment interactions. Our ongoing studies aim to further explore this distinctive "red" fluorescence. We have observed this red fluorescence spectrum in post-mortem measurements of melanin in benign nevus. The characteristic red spectrum is also evident here (unlike the benign nevus in vivo), suggesting that hypoxia may contribute to this phenomenon. Given the central role of hypoxia in both melanoma development and treatment, as well as in fundamental Parkinson's disease mechanisms, this study discusses strategies aimed at reinforcing the hypothesis that red fluorescence from (neuro)melanin serves as an indicator of hypoxia.

Abstract:Melanins are a group of complex pigments of biological origin, widely spread in all species from fungi to man. Among diverse types of melanins, the human melanins, eumelanins,are brown or black nitrogen-containing pigments, mostly known for their photoprotective properties in human skin. We have undertaken theoretical studies aimed to understand absorption spectra of eumelanins and their chemical precursors. The structure of the biopigment is poorly defined, although it is believed to be composed of cross-linked heteropolymers based on indolequinones. As a basic model of the eumelanin structure, we have chosen pentamers containing hydroquinones (HQ) and/or 5,6-indolequinones (IQ) and/or semiquinones (SQ) often listed as structural melanin monomers. The eumelanin oligomers have been constructed as random compositions of basic monomers and opitimized for the energy of bonding. Absorption spectra of model assemblies have been calculated within the semiempirical intermediate neglect of differential overlap (INDO) approximation. Model spectrum of eumelanin has been further obtained by sum of independent spectra of singular polymers. By comparison with experimental data it is shown that the INDO/CI method manages to reproduce well characteristic properties of experimental spectrum of synthetic eumelanins.

Abstract:For large scale applications, hybrid pixel detectors, in which sensor and read-out IC are separate entities, constitute the state of the art in pixel detector technology to date. They have been developed and start to be used as tracking detectors and also imaging devices in radiography, autoradiography, protein crystallography and in X-ray astronomy. A number of trends and possibilities for future applications in these fields with improved performance, less material, high read-out speed, large radiation tolerance, and potential off-the-shelf availability have appeared and are momentarily matured. Among them are monolithic or semi-monolithic approaches which do not require complicated hybridization but come as single sensor/IC entities. Most of these are presently still in the development phase waiting to be used as detectors in experiments. The present state in pixel detector development including hybrid and (semi-)monolithic pixel techniques and their suitability for particle detection and for imaging, is reviewed.

Abstract:Efficient π–π stacking together with better electronic delocalization of non‐functionalized melanins allowed for more significant electrochemical responses than sulfonated melanins. Melanins are macromolecular pigments widely spread in many living organisms, with unique physical and chemical properties. Specifically, their conductive properties have drawn attention for applications in different devices; however, the electrochemical response is equally necessary for designing technologies in sustainable bioelectronics. In this sense, we report a comparative study of the redox electrochemical properties of non‐functionalized and sulfonated melanins in different pH environments. The electrochemical response was investigated using cyclic voltammetry, electrochemical impedance spectroscopy, dielectric permittivity and AC/DC conductivity at pH 3, 5 and 7. The voltammetric currents were higher at low pH, in agreement with the known proton transport properties of melanins. The effect of pH on electrochemical properties was slightly more significant in non‐functionalized pigments. Melanins with a higher 5,6‐dihydroxyindole carboxylic acid/5,6‐dihydroxyindole ratio showed high DC current and low impedance. No significant difference was observed in the dielectric relaxation process between the different samples. © 2024 Society of Chemical Industry.

The peculiarities of dynamic current-voltage and charge-voltage loops and transient currents of melanin under pulse hydration

Abstract:Melanin is a biopolymer with unique set of applicable physical and chemical properties. The impact of a moisture-saturated air pulse (~1 s) on dynamic current-voltage (I-V-), charge-voltage (Q-V-) and transient (I-t-) characteristics of fungal melanin thin layers was investigated. The pulse hydration gives rise to sharp increase of conductance and capacitance (by I-V-loops and I-t-curves swing), transferred charge (by Q-V-loops swing) and causes the appearance of the following peculiarities: hump-like on I-V-loops, knee-like on I-V-curves and step-like on I-t-curves. General shape of I-V-loops was modelled under low hydration by linear series-parallel RC-circuit and under high hydration by the circuit with anti-series Zener diodes as nonlinear elements. By processing of bipolar I-V-loops and I-t-curves allowing for unipolar ones, the maxima of displacement current - voltage loops were revealed and their similarity to current maxima observed under ionic space charge transfer in metal-insulator-semiconductor structures and under polarization reversal in ferroelectrics is discussed. Corresponding processing of bipolar Q-V-loops evidences general shape of obtained dielectric displacement - voltage loops characteristic to ion-conducting materials. As a reason of observed transformations, the appearance of temporal polar media with reversible ferroelectric-like polarization and ionic space charge transfer is considered. Obtained results disclose the functional capabilities of melanin based sensing devices operating in dynamic mode for both hydration and driving voltage.

Quantitative photoluminescence of broad band absorbing melanins: A procedure to correct for inner filter and re-absorption effects

Abstract:We report methods for correcting the photoluminescence emission and excitation spectra of highly absorbing samples for re-absorption and inner filter effects. We derive the general form of the correction, and investigate various methods for determining the parameters. Additionally, the correction methods are tested with highly absorbing fluorescein and melanin (broadband absorption) solutions; the expected linear relationships between absorption and emission are recovered upon application of the correction, indicating that the methods are valid. These procedures allow accurate quantitative analysis of the emission of low quantum yield samples (such as melanin) at concentrations where absorption is significant.

Abstract:A rapid, continuous assay for calcium-activated neutral protease activity is described. This assay is based on monitoring the elevation in fluorescence intensity that occurs upon calpainolytic digestion of dichlorotriazinylamino-fluorescein-labeled microtubule-associated protein 2. Tedious separation of peptide products from the protein substrate in this rapid assay is unnecessary, which thus offers two remarkable advantages over conventional caseinolytic assay procedures: (i) it raises sensitivity of detection by about three orders of magnitude, allowing the quantitative determination of calpain in the high picogram range in 10 min; and (ii) it permits a continuous detection of activity, which may prove invaluable in enzyme-mechanism studies that require pre-steady-state measurements. Other features and advantages of the assay, along with its limitations, are discussed in detail.

Abstract:The melanin is a group of biological pigments commonly found in living beings, it can be classified in three groups: eumelanin human beings, pheomelanins in animals, and allomelanins in the vegetal kingdom. There is a special interest in the eumelanin because this biopolymer exhibits the typical properties of a semiconductor. Non-invasive visualization of melanin and melanocytes by reflectance-mode confocal microscopy

Abstract:In vivo visualization of epidermal melanin was performed by reflectance-mode confocal microscopy (RCM). Firstly, we examined the distribution of epidermal melanin in pigmented animals and compared with that of the human skin. Melanocytes in the skin of pigmented animals were found to accumulate a large amount of melanin that can be easily visualized because of its brightness. Their RCM images correlated well with the Fontana-Masson-stained sections for melanin. In contrast, in the human skin, typical dendritic melanocytes were hardly observed even in pigmented lesions, although supranuclear melanin caps were easily visible. These results suggested that human melanocytes rapidly transfer the produced melanin to keratinocytes and do not accumulate it. Secondly, to elucidate the production of melanin by human melanocytes, we evaluated the changes of melanin after a single ultraviolet (UV) exposure. The melanin-accumulating melanocytes were clearly visualized during the skin pigmentation process. The RCM images showed the brightness because of melanin gradually increased from day 4, then dendrite-elongated melanocytes appearing from day 8, and finally melanin caps formed from day 29. In conclusion, RCM successfully evidenced the difference in melanin distribution between the pigmented animals and humans, and the UV-induced pigmentation process in vivo as well.
Enhanced photochemical activity and ultrafast photocarrier dynamics in sustainable synthetic melanin nanoparticle-based donor-acceptor inkjet-printed molecular junctions

Abstract:Melanin is a stable, widely light-absorbing, photoactive, and biocompatible material viable for energy conversion, photocatalysis, and bioelectronic applications. To achieve multifunctional nanostructures, we synthesized melanin nanoparticles of uniform size and controlled chemical composition (dopamelanin and eumelanin) and used them with titanium dioxide to fabricate donor-acceptor bilayers. Their size enhances the surface-to-volume ratio important for any surface-mediated functionality, such as photocatalysis, sensing, and drug loading and release, while controlling their chemical composition enables to control the film's functionality and reproducibility. Inkjet printing uniquely allowed us to control the deposited amount of materials with minimum ink waste suitable for reproducible materials deposition. We studied the photochemical characteristics of the donor-acceptor melanin-TiO2 nanostructured films via photocatalytic degradation of methylene blue dye under selective UV-NIR and Vis-NIR irradiation conditions. Under both irradiation conditions, they exhibited photocatalytic characteristics superior to pure melanin and, under UV-NIR irradiation, superior to TiO2 alone; TiO2 is photoactive only under UV irradiation. The enhanced photocatalytic characteristics of the melanin-TiO2 nanostructured bilayer films, particularly when excited by visible light, point to charge separation at the melanin-TiO2 interface as a possible mechanism. We performed ultrafast laser spectroscopy to investigate the photochemical characteristics of pure melanin and the melanin-TiO2 constructs and found that their time-resolved photoexcited spectral patterns differ. We performed singular value decomposition analysis to quantitatively deconvolute and compare the dynamics of photochemical processes for melanin and melanin-TiO2 heterostructures. This observation supports electronic interactions, namely, interfacial charge separation at the melanin and TiO2 interface. The excited-state relaxation in melanin-TiO2 increases markedly from 5 ps to 400 ps. The results are remarkable for the future intriguing application of melanin-based constructs for bioelectronics and energy conversion.

Abstract:Four melanin polymers prepared from different precursors (5,6‐dihydroxyindole, 5,6‐dihydroxyindole 2‐carboxylic acid, 1,8‐dihydroxynaphthalene and dopamine) have been adopted as photocatalysts in the reductive dehalogenation of a series of model a‐halogen carbonyl derivatives. The best performing melanin was that obtained from 5,6‐dihydroxyindole 2‐carboxylic acid, which allowed to dehalogenate efficiently both bromomalonate esters and phenacyl bromide; on the other hand, chloromalonate esters were almost unreactive under the same reaction conditions. Electrochemical and control experiments enabled to elucidate the reaction mechanism, demonstrating the key role of electrons as charge carriers in the observed photocatalytic process.

Radiative Relaxation Quantum Yields for Synthetic Eumelanin

Abstract:We report absolute values for the radiative relaxation quantum yield of synthetic eumelanin as a function of excitation energy. These values were determined by correcting for pump beam attenuation and emission re-absorption in both eumelanin samples and fluorescein standards over a large range of concentrations. Our results confirm that eumelanins are capable of dissipating >99.9% of absorbed UV and visible radiation through non-radiative means. Furthermore, we have found that the radiative quantum yield of synthetic eumelanin is excitation energy dependent. This observation is supported by corrected emission spectra, which also show a clear dependence of both peak position and peak width upon excitation energy. Our findings indicate that photoluminescence emission in eumelanins is derived from ensembles of small chemically distinct oligomeric units which can be selectively pumped. This hypothesis lends support to the theory that the basic structural unit of eumelanin is oligomeric rather than hetero-polymeric.

Abstract:Abstract Choroidal melanocytes (HCMs) are melanin-producing cells in the vascular uvea of the human eye (iris, ciliary body and choroid). These cranial neural crest-derived cells migrate to populate a mesodermal microenvironment, and display cellular functions and extracellular interactions that are biologically distinct to skin melanocytes. HCMs (and melanins) are important in normal human eye physiology with roles including photoprotection, regulation of oxidative damage and immune responses. To extend knowledge of cytoplasmic melanins and melanosomes in label-free HCMs, a non-invasive ‘fit-free’ approach, combining 2-photon excitation fluorescence lifetimes and emission spectral imaging with phasor plot segmentation was applied. Intracellular melanin-mapped FLIM phasors showed a linear distribution indicating that HCM melanins are a ratio of two fluorophores, eumelanin and pheomelanin. A quantitative histogram of HCM melanins was generated by identifying the image pixel fraction contributed by phasor clusters mapped to varying eumelanin/pheomelanin ratio. Eumelanin-enriched dark HCM regions mapped to phasors with shorter lifetimes and longer spectral emission (580–625 nm) and pheomelanin-enriched lighter pigmented HCM regions mapped to phasors with longer lifetimes and shorter spectral emission (550–585 nm). Overall, we demonstrated that these methods can identify and quantitatively profile the heterogeneous eumelanins/pheomelanins within in situ HCMs, and visualize melanosome spatial distributions, not previously reported for these cells.

Optical studies of melanin films as a material for solar light absorbers

Abstract:This study investigates optical and electrical properties of thin melanin films self-organized grown from water solutions of eumelanin extracted from edible plants. The melanin films exhibit the characteristics of a transparent low conductive polymer with stable in time electrical parameters. The films demonstrate indirect allowed optical transitions, an optical band gap E_g = 1.7 eV and a phonon energy {\hbar \omega} = 0.185 eV as well as a high absorption coefficient in the ultraviolet range, which highlight the film potential for applications as light-absorbing layers in photovoltaic solar energy converters. Analysis of infrared transmission spectra evidences presence of a significant amount of OH groups in the films, pointing to their hydrophilicity, which may have an effect on the film electrical conductivity. It is hypothesized as well that the ratio of sp^3 to sp^2 hybridized CH_n bonds influences the films' optical and electrical properties and that higher concentration of the sp^2 bonds may increase the conductivity due to the enhanced {\pi}-electron delocalization.

Abstract:We discuss recent progress towards the establishment of important structure-property-function relationships in eumelanins-key functional bio-macromolecular systems responsible for photo-protection and immune response in humans, and implicated in the development of melanoma skin cancer. We focus on the link between eumelanin's secondary structure and optical properties such as broad band UV-visible absorption and strong non-radiative relaxation; both key features of the photo-protective function. We emphasise the insights gained through a holistic approach combining optical spectroscopy with first principles quantum chemical calculations, and advance the hypothesis that the robust functionality characteristic of eumelanin is related to extreme chemical and structural disorder at the secondary level. This inherent disorder is a low cost natural resource, and it is interesting to speculate as to whether it may play a role in other functional bio-macromolecular systems.
Melanin-dependent tissue interactions induced by a 755-nm picosecond-domain laser: complementary visualization by optical imaging and histology

Abstract:Fractional picosecond-domain lasers (PSL) induce optical breakdown, which correlates histologically to vacuolization in the epidermis and dermis. In this ex vivo porcine study, we sought to establish a framework for the investigation of laser-tissue interactions and their dependence on melanin density. Light- (melanin index: 24.5 [0-100]), medium- (58.7), and dark-pigmented (> 98) porcine skin samples were exposed to a 755-nm fractional PSL and examined with dermoscopy, line-field confocal optical coherence tomography (LC-OCT), conventional OCT, and subsequently biopsied for digitally stained ex vivo confocal microscopy (EVCM) and histology, using hematoxylin and eosin (HE) and Warthin-Starry (WS) melanin staining. Dermoscopy showed focal whitening in medium- and dark-pigmented skin. Similarly, LC-OCT and OCT visualized melanin-dependent differences in PSL-induced tissue alterations. Vacuoles were located superficially in the epidermis in dark-pigmented skin but at or below the dermal-epidermal junction in medium-pigmented skin; in light-pigmented skin, no vacuoles were observed. Histology confirmed the presence of vacuoles surrounded by areas void of WS staining and disrupted stratum corneum in darker skin. The combined use of optical imaging for multiplanar visualization and histological techniques for examination of all skin layers may mitigate the effect of common artifacts and attain a nuanced understanding of melanin-dependent laser-tissue interactions.
