Computational and Mathematical Methods in Medicine

Abstract:Increasingly wider availability of biomedical imaging modalities, such as X-ray computed tomography (CT), magnetic resonance imaging (MRI), ultrasonic imaging, positron emission tomography (PET), single-photon emission computed tomography (SPECT), and optical imaging, have led to significant progresses in both biomedical research and clinical practice. Advancements in imaging technology and image informatics have facilitated and enabled quantitative understanding of the biological structures, functional organisms, and pathological mechanisms. This special issue presents 14 latest research contributions in computational biomedical imaging. The focuses have been on the computational principles, strategies, methods, and algorithms that create, extract, and understand imaging data of biomedical significance. The first group of papers has showcased the development and applications of rigorous optimization techniques in biomedical imaging. The work based on dual unscented Kalman filtering provides a reliable and efficient platform to understand the BOLD signals from fMRI. In the other paper, the robust discrete particle swarm optimization algorithm is adopted to solve the branch-cut phase unwrapping problem of MRI data. Without the need for a regularization penalty term, a semigreed method for bioluminescence tomography reconstruction is fast and stable for small animal imaging. The second group of papers has addressed image segmentation and tissue characterization problems, ever popular among image analysis researchers. The work of L. Li et al. applies machine learning techniques to characterize lymph node metastasis in gastric cancer from gemstone spectral imaging with much improved outcomes, while G. V. Sanchez. Ferrero et al. make new advances in ultrasonic speckle characterization using generalized Gama distribution and generalized Gama mixture model. In recent years, physically meaningful modeling and simulation have become integral components for model-constrained imaging and model-based image understanding. Thus, the third group of papers has continued the stride in this direction. The propagation of myocardial electrical activation is simulated using a monodomain model, without the need of explicit mesh constraints. In similar spirit, one work performs forward modeling of 3D optical molecular imaging with simplified spherical harmonics approximation using extended finite element method. The final group of papers has highlighted some new imaging modalities and their potential use in clinical practice. The further development of ultrasound-based quantitative imaging of Young's modulus with high accuracy is presented in this issue. And the novel Diffusion Kurtosis Imaging shows that more detailed classification of brain tissues is clinical significance. We appreciate the many high quality submissions to this special issue from leading researchers in computational biomedical imaging. Because of space limitation, we can only accept a small number of most exciting works. We sincerely hope that the remaining interesting research outcomes will be soon published in other venues.

Abstract:In this study,we summarize the advancements in high-resolution vascular imaging technology and its applications in the biomedical field.In particular,we focus on quantitative characterization methods applicable to high-resolution vascular images.The quantification process of vascular images generally comprises three main steps:image preprocessing,vascular image reconstruction and quantitative characterization acquisition,and statistical analysis of quantitative parameters.We provide a detailed explanation of the algorithm pipeline,accuracy assessment,and potential optimization directions for the methods employed in each step.Furthermore,we explore the significance of extracting biological information from various vascular and blood parameters for clinical reference.We also discuss the robustness of multiparametric models in distinguishing different stages of disease development within specific disease contexts.These advancements not only reflect the potential value of high-resolution vascular imaging technology and the application of quantitative characterizations but also provide new insights into their promising prospects for advancing fundamental biomedical research and clinical diagnosis.

Abstract:Molecular imaging is a newly emerging and rapidly developing biomedical imaging field in which the modern tools are being married to depict noninvasive in vivo cellular and molecular processes sensitively and specifically, such as monitoring multiple molecular events, cell trafficking and targeting. The goals of this field are to develop technologies and instruments for studying biological and medical processes as well as diagnosing and managing diseases better. Although rapid progress in the fundamentals and applications make molecular imaging become an important tool for biomedical research in recent years, many difficult problems and challenges remain. Discussing the problems and challenges in detail and illustrating recent progress and future direction, the special issue collects the high-quality, peer-reviewed, original research papers in the area of molecular imaging. Novel molecular imaging theories and algorithms, new molecular probes, multimodality molecular imaging prototype systems and experiments, and final clinical applications are introduced mainly in this special issue. In molecular imaging theories and algorithms, genetic algorithm-based optimization tool is used to improve the accuracy of the diffusion model in strongly absorbing media by adjusting the optical parameters. Furthermore, a penalized linear and nonlinear combined conjugate gradient method, a fast preiteration algorithm based on the generalized inverse matrix, and a Monte-Carlo-based network method are also proposed for light source reconstruction in optical tomography. Considering the importance of molecular probes, synthesis and bioconjugation of gold nanoparticles as potential molecular probes for light-based imaging techniques are described in this special issue, and the nitroimidazole-based thioflavin-T derivatives as cerebral ischemia markers are evaluated in vivo. In order to test the feasibility and effectiveness of imaging theories, algorithms and probes, molecular imaging prototype systems should be designed, constructed and employed for small animal or phantom imaging. Thus, multimodality fusion near-infrared optical tomography Systems with highly sensitive CCD camera and photomultiplier tube can be consulted respectively in this special issue. Furthermore, a novel confocal optical system design and a dual laser confocal scanner have been developed for molecular imaging applications of biochip. In clinical applications, the advantage of PET and CT integration in examination of lung tumors is analyzed. Moreover, several innovative processing methods of molecular image are also presented in this special issue. In conclusion, this special issue covers recent important advances in molecular imaging field.

Abstract:Recent advancements on interventional molecular imaging aimed to further complement the advantages of two imaging fields, namely, interventional radiology and molecular imaging. Interventional molecular imaging significantly improved the early diag-nosis of cancer, local treatment, and monitoring of tumor treatments. Interventional radiology has continuously extended the capabili-ties of the currently available molecular imaging techniques to (i) obtain deep-seated targets;(ii) thoroughly examine small targets;(iii) precisely guide the delivery of non-targeted imaging tracers or therapeutics;and (iv) selectively enhance the effectiveness of targeted imaging and treatment with high accuracy. Molecular imaging has been used to guide interventional therapies and assess the thera-peutic efficacies of medical interventions. The continuous efforts on interventional molecular imaging extend molecular imaging from benches or small animal laboratories to large animal suites and ultimately to certain clinical applications in humans and enhance the diagnosis and treatment of cancer.

Abstract:Cancer is a serious health problem which increasingly causes morbidity and mortality worldwide. It causes abnormal and uncontrolled cell division. Traditional cancer treatments include surgery, chemotherapy, radiotherapy and so on. These traditional therapies suffer from high toxicity and arouse safety concern in normal area and have difficulty in accurately targeting tumour. Recently, a variety of nanomaterials could be used for cancer diagnosis and therapy. Nanomaterials have several advantages, e.g. , high concentration in tumour via targeting design, reduced toxicity in normal area and controlled drug release after various rational designs. They can combine with many types of biomaterials in order to improve biocompatibility. In this review, we outlined the latest research on the use of bioresponsive nanomaterials for various cancer imaging modalities (magnetic resonance imaging, positron emission tomography and phototacoustic imaging) and imaging-guided therapy means (chemotherapy, radiotherapy, photothermal therapy and photodynamic therapy), followed by discussing the challenges and future perspectives of this bioresponsive nanomaterials in biomedicine.

Abstract:Different tissues and diseases have specific biological signals. The development of magnetic resonance imaging (MRI) contrast agents responding to these signals can not only improve the sensitivity of disease detection by MRI, but also increase the accuracy of disease diagnosis through molecular imaging of these disease signals. This paper gives a review of MRI contrast agents responding to albumin, pH, metal ion, enzyme, redox and other biological signals with the focus on the latest developments and perspectives.

Abstract:Cancer remains one of the most pressing diseases in the world. Traditional treatments, including surgery, chemotherapy, and radiotherapy still show certain limitations. Recently, numerous cancer treatments have been proposed in combination with novel materials, such as photothermal therapy, chemodynamic therapy, immunotherapy, and a combination of therapeutic approaches. These new methods have shown significant advantages in reducing side effects and synergistically enhancing anti-cancer efficacy. In addition to the above approaches, early diagnosis and in situ monitoring of lesion areas are also important for reducing side effects and improving the success rate of cancer therapy. This depends on the decent use of bioimaging technology. In this review, we mainly summarize the recent advances in porous framework materials for bioimaging and cancer therapy. In addition, we present future challenges relating to bioimaging and cancer therapy based on porous framework materials.

Abstract:The papers in this special issue focus on biomedical Big Data. Biomedical imaging is an essential component in various fields of biomedical research and clinical practice. The study of biologists requires continuous monitoring of cell behavior under microscope. Neuroscientists detect regional metabolic brain activity from positron emission tomography (PET), functional magnetic resonance imaging (MRI), and magnetic resonance spectrum imaging (MRSI) scans. During these researching process, large amount of biomedical data will be produced for processing. The development of advanced imaging equipment and diverse applications also have driven the generation of biomedical big data. The main challenge and bottleneck for the related research is the conversion of “biomedical big data” into interpretable information and hence discoveries. Computer vision theory has a huge potential in many aspects for automated understanding of biomedical data and has been used successfully to speed up and improve applications such as large-scale cell image analysis (image preconditioning, cell segmentation and detection, cell tracking, and cell behavior identification), image reconstruction and registration, organ segmentation and disease classification. Considering the recent advance in machine learning technique, deep learning has revolutionized multiple fields of computer vision, significantly pushing the state of arts of computer vision systems in a broad array of high-level tasks. Hopefully these technique advance will help to deal problems in biomedical big data.

Abstract:With the development of basic science, the imaging medicine which is based on the basic subject have gained constant progress over the past several years. The imaging medicine had received great changes in the imaging way and imaging target, which has developed from the classic morphological diagnosis to the combination of form and function nowadays, from macro to micro, and from the tissues and organs to the cellular and molecular level. The molecular imaging arises at the historic moment and it is gradually becoming a mainstream technology. Molecular imaging is a frontal subject which intercross and fusion of cell biology, molecular biology, pharmaceutical chemistry and image technology. It can show lesions from the cellular and molecular level, and is a kind of specificity, new living imaging method. Molecular imaging can help us to understand the occurrence, development of diseases, which can reduce invasive examinations. In addition, it can detect the diseases at their early stages, identify the qualitative evaluation and treatment, which can help to improve the level of early diagnosis and treatment, thereby improving the prognosis of diseases. In this paper, we stated the research on the status and problems of MR molecular imaging.

Abstract:Fluorescence imaging has been recognized as a powerful tool for the real-time detection and specific imaging of biomarkers within living systems, crucial for early diagnosis and treatment evaluation. Over the years, significant advancements in this field have been achieved, particularly with the development of novel fluorescent probes and advanced imaging technologies such as NIR-II imaging, super-resolution imaging, and 3D imaging. These technologies have enabled deeper tissue penetration, higher imaging contrast, and more accurate of disease-related biomarkers. Despite these advancements, challenges such as improving probe specificity, enhancing imaging depth and resolution, and optimizing signal-to-noise ratios remain. Artificial intelligence (AI) has shown potential in optimizing probe design and improving imaging techniques, offering new tools for more precise disease diagnosis. This review will not only discuss chemical modifications of classic fluorophores and in-situ visualization of vrious biomarkers including metal ions, reactive species, and enzymes, but also share with some breakthroughs on AI-driven fluorescence imaging, aiming to provide a comprehensive understanding of these advancements. Future prospects of fluorescence imaging for biomarkersimaginginluding the potential impact of AI in this rapidly evolving field are also highlighted.

Abstract:Timely and accurate assessment and diagnosis are extremely important and beneficial for all diseases, especially for some of the major human disease, such as cancers, cardiovascular diseases, infectious diseases, and neurodegenerative diseases. Limited by the variable disease microenvironment, early imperceptible symptoms, complex immune system interactions, and delayed clinical phenotypes, disease diagnosis and treatment are difficult in most cases. Molecular imaging (MI) techniques can track therapeutic drugs and disease sites in vivo and in vitro in a non-invasive, real-time and visible strategies. Comprehensive visual imaging and quantitative analysis based on different levels can help to clarify the disease process, pathogenesis, drug pharmacokinetics, and further evaluate the therapeutic effects. This review summarizes the application of different MI techniques in the diagnosis and treatment of these major human diseases. It is hoped to shed a light on the development of related technologies and fields.

Abstract:Medical image analysis is performed in order to facilitate medical research and ultimately provide better healthcare. It is critical to the advancement of imaging-based medical research, for example, using magnetic resonance (MR) imaging to probe brain structural and functional changes related to a disease or cognitive process. This special issue highlights new methods of signal and image processing, computer vision, machine learning, and statistical analysis and their application in medical image analysis. It is organized into two groups of papers: predictive modeling and image processing. In the first group, we include a set of papers for imaging-derived biomarker detection and clinical decision support systems. The overall theme is how to detect biomarkers characterizing a disease and to build decision support systems using computer algorithms. B. Tay et al. propose a classification scheme for identifying healthy individuals and patients with spinal cord injuries based on fractional anisotropy values obtained from diffusion tensor imaging data. H. Liu et al. address cirrhosis classification based on multimodality imaging and propose a new method to extract texture features for multilabel classification (normal, early, middle, and advanced stages). G.-P. Liu et al. employ deep learning and multilabel learning to construct the syndrome diagnostic model for chronic gastritis. M. Yang et al. describe a method to automatically detect corticospinal tract damage in chronic stroke patients and demonstrate that the detected biomarker is associated with motor impairment. Y. Liu et al. describe a novel machine learning algorithm which combines tract-based spatial statistics and Bayesian data mining to quantify white matter changes in mild traumatic brain injury. The second group of papers consists of a wide range of medical image processing algorithms, including segmentation, visualization, and enhancement. For brain tissue segmentation, S. Ji et al. describe a multistage method based on superpixel and fuzzy clustering for brain MRI segmentation. Y. Zhong et al. propose a method for regularization of fMRI data to address the limitations of traditional spatial independent component analysis. J. Gao et al. describe a global search algorithm based method to separate MR images blindly. Y. P. Du et al. develop a method to reduce partial volume effect with voxel-shifted interpolation and this algorithm substantially improves the detection of BOLD signal in fMRI. Z. Jin et al. use a high-pass filter based method to enhance the visibility of the venous vasculature and reduce the artifacts in the venography. A framework for tracking left ventricular endocardium through 2D echocardiography image sequence is proposed by H. Ketout and J. Gu. E. Bengtsson and P. Malm review automated analysis of the cell samples to screen for cervical cancer. This special issue is selective. Among 27 submissions, 12 were selected. Abstract:Systematic imaging can be broadly defined as the systematic identification and characterization of biological processes at multiple scales and levels. In contrast to “classical” diagnostic imaging, systematic imaging emphasizes on detecting the overall abnormalities including molecular, functional, and structural alterations occurring during disease course in a systematic manner, rather than just one aspect in a partial manner. Concomitant efforts including improvement of imaging instruments, development of novel imaging agents, and advancement of artificial intelligence are warranted for achievement of systematic imaging. It is undeniable that scientists and radiologists will play a predominant role in directing this burgeoning field. This article introduces several recent developments in imaging modalities and nanoparticles-based imaging agents, and discusses how systematic imaging can be achieved. In the near future, systematic imaging which combines multiple imaging modalities with multimodal imaging agents will pave a new avenue for comprehensive characterization of diseases, successful achievement of image-guided therapy, precise evaluation of therapeutic effects, and rapid development of novel pharmaceuticals, with the final goal of improving human health-related outcomes.

Abstract:A variety of imaging techniques are available for detecting biological processes with sufficient penetration depth and temporal resolution. However, inflammation, cardiovascular, and cancer-related disorders might be difficult to diagnose with typical bioimaging methods because of the lack of resolution in the imaging of deep tissues. Therefore, nanomaterials are the most promising candidate to overcome this hurdle. This review is on the utilization of carbon-based nanomaterials (CNMs), ranging from zero-dimension (0D) to three-dimension (3D), in the development of fluorescence (FL) imaging, photoacoustic imaging (PAI), and biosensing for the early detection of cancer. Nanoengineered CNMs, such as graphene, carbon nanotubes (CNTs), and functional carbon quantum dots (QDs), are being further studied for multimodal biometrics and targeted therapy. CNMs have many advantages over conventional dyes in FL sensing and imaging, including clear emission spectra, long photostability, low cost, and high FL intensity. Nanoprobe production, mechanical illustrations, and diagnostic therapeutic applications are the key areas of focus. The bioimaging technique has facilitated a greater understanding of the biochemical events underlying multiple disease etiologies, consequently facilitating disease diagnosis, evaluation of therapeutic efficacy, and drug development.
