Age-Specific Levels of Diabetes-Related GAD and IA-2 Antibodies in Healthy Children and Adults

Abstract:Glutamic acid decarboxylase (GAD) and tyrosine phosphatase IA-2 antibody levels were measured in 375 healthy children and adults and in 187 children with newly diagnosed type 1 diabetes mellitus (DM). GAD antibody levels were determined by radioimmunoassay, and IA-2 antibody levels by immunoprecipitation. Healthy children had higher GAD antibody levels than adults (p < 0.001). The 98th percentile was 1.60 U/ml in children and 1.16 U/ml in adults. IA-2 antibody levels did not differ between these two cohorts. Children with DM had higher GAD and IA-2 antibody levels than healthy children (p <0.001). Based on receiver operating characteristics (ROC) analysis, elevated levels of these antibodies showed high specificity rates (+/- SE) of 0.968 +/- 0.14 to 1.00 +/- 0.00. The sensitivity ranged between 0.439 +/- 0.037 (both antibodies elevated) and 0.850 +/- 0.027 (at least one antibody elevated). These data emphasize the importance of establishing age-specific reference values for DM-related antibodies in the background population before applying them for screening and intervention studies.
[Comparison of processed and crude Polygoni Multiflori Radix induced rat liver injury and screening for sensitive indicators].

Abstract:To investigate the difference of liver injury in rats gavaged with crude and processed Polygoni Multiflori Radix. The 75% ethanol extract of crude and processed Polygoni Multiflori Radix (50 g · kg(-1) crude medicine weight/body weight) were continuous oral administered to rats for 6 weeks. Serum biochemical indicators were dynamically detected, the change of liver histopathology was assessed 6 weeks later. Principal component analysis (PCA) was adopted to screen sensitive indicator of the liver damage induced by polygoni multiflori radix. Biochemical tests showed that the crude Polygoni Multiflori Radix group had significant increase of serum ALT, AST, ALP, DBIL and TBIL (P < 0.01 or P < 0.05) and significant decreases of serum IBIL and TBA (P < 0.01 or P < 0.05), while the processed Polygoni Multiflori Radix group showed no obvious changes, compared to the untreated normal group. Histopathologic analysis revealed that crude Polygoni Multiflori Radix group exhibited significant inflammatory cells infiltration in portal area around the blood vessels, tissue destruction and local necrosis of liver cells. There were not obvious pathological changes in processed Polygoni Multiflori Radix group. The results demonstrated that the injury effect of processed Polygoni Multiflori Radix on liver injury of rats was significantly lower than that of unprocessed, and that processing can effectively reduce the hepatotoxicity of Polygoni Multiflori Radix. Traditional transaminase liver function indicators were not sensitive for crude Polygoni Multiflori Radix induced liver damage. The serum content of DBIL and TBIL can reflect the liver damage induced by crude Polygoni Multiflori Radix early and can be sensitive indicators for clinical monitoring the usage of it.
Effects of Polygoni Multiflori Radix and Its Three Ingredients on Human Normal Hepatocyte Apoptosis

Abstract:Objective To observe the effects of Polygoni Multiflori Radix on toxicity and apoptosis of human normal hepatocyte;To discuss the possible substances and channels of Polygoni Multiflori Radix to hepatic injury.Methods L02 cells were set as evaluation model. Cellular morphology in each group after medication was observed through Giemsa dyeing. Apoptosis rate and cell cycle of each group after medication were detected by FCM.Results Compared with blank control group, the 70% alcohol extracts of Polygoni Multiflori Radix (1, 2 and 4 mg raw materials per mL) had almost no effects on cellular morphology or cell cycle of L02 cells. Apoptosis rates after treatment with extracts of Polygoni Multiflori Radix were 9.51%, 10.3%, and 13.2% respectively. When L02 cells were treated with different dosages of emodin, rhein and gallic acid, three ingredients of Polygoni Multiflori Radix, apoptosis was obvious. The proportion of G1 phase cells significantly decreased, the S phase and G2 phase increased, which presented the concentration dependent effect. Apoptosis rates of three high dosages (100μmol/L) treated groups after treatment with emodin, rhein and gallic acid were 19.8%, 12.0%, and 13.7% respectively. Conclusion Polygoni Multiflori Radix has apoptosis-inducing effects on L02 cells. Apoptosis-inducing effects of its ingredient of emodin is stronger than rhein and gallic acid.
Protective Effects of Extracts from Polygonum Hydropiper L on Ethanol-Induced Acute Gastric Mucosal Lesions

Abstract:Introduction: Free radicals and oxidative stress play a central role in the generation and development of acute gastric mucosal lesions (AGML).Traditional Chinese medicine has its unique effect in removing oxygen free radicals and reducing gastric mucosa injury.Polygonum hydropiper L is extensively used as a traditional Chinese herbal medicine for treating gastrointestinal diseases in China, the reports shows that it possess various pharmacological activities, especially antioxidant and anti-inflammatory properties.Objective: This study was to investigate the protective effect of extracts from Polygonum hydropiper on experimentally-induced AGML in mice. Methods:The AGML mice model was induced by ethanol, Glutathione peroxidase (GSH-Px), malondialdehyde (MDA) and superoxide dismutase (SOD) were used to measure the antioxidant activity.The nitric monoxide (NO) and PGE2 contents were tested. Results:The findings demonstrated that the extracts enhanced the activities of antioxidative enzymes SOD and GSH-Px and decreased the level of MDA in the serum and gastric tissues.The higher-concentration extract (1.78 g kg -1 ) had higher antioxidative activities than the lower-concentration extract (0.89 g kg -1 ).Both extracts increased the production of PGE2and decreased the NO level in gastric tissues.In addition, oral administration of absolute ethanol to mice significantly produced extensive hemorrhagic lesions of gastric mucosa, whereas animals pretreated with extract or ranitidine significantly reduced the formation of gastric lesions compared to control group.The extract could decrease distinctly the acute gastric mucosal damage index compared to the model group.The high-dose extract treatment obviously reduced injuries in the gland region after administration of ethanol compared to the model group. Conclusion:The results obtained from this study showed that extracts from Polygonum hydropiper L exhibited significant protective effects against ethanol-induced AGML, and these are closely associated with antioxidant effect.
Influence of Extracting Solvent on Hepatocytes Toxicity of Polygonum Multiflorum

Abstract:Objective: The difference of extracting solvent to hepatocytes toxicity of Polygonum multiflorum was researched to probe the influence of different usages to the safety of P. multiflorum. Method: The inhibition rate of P. multiflorum samples to human hepatocytes( L02 cell line) with different extracting methods was detected by CCK-8 method. The dose of all the samples was 5 g·L- 1counted on the quantity of crude material. The influence of extracting solvent,the amount of solvent and extracting duration on hepatocytes toxicity was investigated by orthogonal test. UPLC fingerprints of the P. multiflorum samples were acquired and then analyzed with relationship to hepatocytes toxicity by partial least squares( PLS) analysis. Result: The method to detect the hepatocytes toxicity of P. multiflorum was established based on the CCK-8 colorimetry. The hepatocytes toxicity of the sample extracted with 75% ethanol was much greater than that of the sample extracted with water.The concentration of ethanol investigated in orthogonal test revealed significant influence to the hepatocytes toxicity,and the toxicity sequence of the samples was: 50% ethanol 75% ethanol 95% ethanol. According PLS analysis there are two contents relative to hepatocytes toxicity. Conclusion: The extracting solvent showed significant influence on the hepatocytes toxicity of P. multiflorum and the sample extracted with 50% ethanol was of much toxic potential. It should be cautious about the high risk of liver damage of P. multiflorum,considering people usually used the herb by soaking with wine around 50 degree.
Anti-Tumor Effect of Polysaccharides Isolated from Taraxacum Mongolicum Hand-Mazz on Mcf-7 Human Breast Cancer Cells

Abstract:Purpose: To optimize the extraction conditions for the ultrasound-assisted extraction of polysaccharides from T. mongolicum (PTM) and investigate their anti-tumor effect on human breast cancer MCF-7 cells.Methods: To optimize the extraction conditions of PTM, response surface methodology (RSM) was performed. The effects of extraction temperature, liquid-solid ratio and extraction time on the yield of PTM were investigated using a Box-Behnken design (BBD). The in vitro anti-tumor effect of PTM on MCF-7 cells was investigated by methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay, while the mechanism of PTM-induced apoptosis was assessed by evaluating the expressions of p53, Bax and Bcl-2 proteins using western blot analysis. Furthermore, the in vivo anti-tumor effect of PTM on MCF-7 cells was studied in mice.Results: The optimal conditions for the extraction of PTM were as follows: extraction temperature, 58.2 degrees C; liquid-solid ratio, 15 mL/g; and extraction time, 44.12 min. Under these optimal conditions, the yield of PTM was 4.84 +/- 0.13 %. PTM showed significant anti-tumor effect on MCF-7 cells in vitro. The expressions of pro-apoptotic proteins, p53 and Bax, were significantly upregulated (p < 0.05), while the expression of anti-apoptotic protein, Bcl-2, was significantly down-regulated (p < 0.05) after treatment with PTM. PTM also showed significant inhibitory effect (p < 0.05) on MCF-7 cells in vivo in a dosedependent manner.Conclusion: RSM is effective in optimizing the extraction conditions of PTM by ultrasonic extraction. PTM possesses significant anti-tumor effect on MCF-7 human breast cancer cells, both in vitro and in vivo.

Abstract:It was shown that different extracts had significant differences in the toxicity of Polygonum multiflorum. In this study, the effect of sample preparation on components and liver toxicity of different extracts from P. multiflorum were determined. Hepatoxic components were discovered based on biomembrane extraction. Comparative chemistry and toxicology between ethanol and water extracts were also performed. The results showed that ethanol extract had much stronger hepatotoxicity, the content of emodin-8-O-β-d-glucopyranoside, physcion-8-O-β-d-glucopyranoside, emodin and physcion was significantly higher in ethanol extract than in water extract, while the human hepatocytes extraction showed that 2,3,5,4'-tetrahydroxystilbene-2-O-β-d-glucopyranoside, emodin-8-O-β-d-glucopyranoside, physcion-8-O-β-d-glucopyranoside, emodin and physcion had interaction with human hepatocytes. The hepatotoxic effect of these components was investigated on human hepatocytes LO2 cells and emodin-8-O-β-d-glucopyranoside, physcion-8-O-β-d-glucopyranoside, emodin and physcion were finally confirmed to be, at least partial, hepatotoxic components. The results showed that sample preparation has significant effect on components in extracts of P. multiflorum especially the components related to hepatotoxicity. Water extract, the conventional administration form of Chinese herbs, is prefer for phytotherapy before well understanding their chemistry and biological activities.

Antitumor Effects and Related Mechanisms of Ethyl Acetate Extracts of Polygonum Perfoliatum L.

Abstract:Polygonum perfoliatum L. belongs to the genus Polygonaceae and has a long history to be used as a Chinese medicinal herb to reduce swelling, control body temperature, and promote detoxification. However, its anticancer activity and mechanisms of action have not been evaluated yet. In the present study, we used several cell lines and xenograft models from different cancers to demonstrate the broad-spectrum anticancer activity of P. perfoliatum L as well as its underlying mechanisms of action in vitro and in vivo. The ethyl acetate extract of P. perfoliatum L showed good anticancer activity and was further fractioned to obtain five active components, including PEA to PEE. Among these fractions, PEC showed the strongest cytotoxicities against various cancer cell lines. It was further observed that PEC inhibited cancer cell growth, arrested cells at G2 phase, and induced apoptosis in vitro and suppressed tumor growth and angiogenesis in vivo in a dose- and time-dependent manner. Furthermore, PEC decreased the expression of vascular endothelial growth factor (VEGF) and micro-vascular density (MVD) in tumor tissues in vivo. It also promoted the proliferation of T and B lymphocytes, increased the activities of natural killer (NK) cells and cytotoxic T lymphocytes (CTLs), enhanced the secretion of interleukin 2 (IL-2) by spleen cells, and raised the levels of IgG, IgG2a, and IgG2b antibodies in tumor-bearing mice in vivo, which were at least partially responsible for the anticancer activity of PEC. In summary, PEC has shown broad-spectrum anticancer activities without causing any host toxicity in vitro and in vivo and may be developed as a preventive and therapeutic agent against human cancer. Further studies are urgently needed to determine the anticancer compounds in PEC and their detailed molecular mechanisms.

Abstract:By observing the cytotoxic effects of anthraquinones on HepG2 cell and using the precision-cut liver slices technique to authenticate the cytotoxic constituents, the paper aims to explore the material basis of Polygonum multiflorum root to cause liver toxicity. Firstly, MTT method was used to detect the effect of 11 anthraquinone derivatives on HepG2 cell. Then, the clear cytotoxic ingredients were co-cultured with rat liver slices for 6h respectively, and the liver tissue homogenate was prepared. BCA method was used to determine the content of protein in the homogenate and continuous monitoring method was used to monitor the leakage of alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamine amino transpeptidase (GGT) and lactate dehydrogenase (LDH). The toxic effect of these ingredients on liver tissue was tested by calculating the leakage rate of the monitored enzymes. As a result, rhein, emodin, physcion-8-O-β-D-glucopyranoside and physcion-8-O-(6'-O-acetyl)-β-D-glucopyranoside showed cytotoxic effects on HepG2 cell and their IC₅₀ values were 71.07, 125.62, 242.27, 402.32 μmol•L⁻¹ respectively, but the other 7 compounds are less toxic and their IC₅₀ values can not be calculated. The precision-cut liver slices tests showed that rhein group of 400 μmol•L⁻¹ concentration significantly increased the leakage rate of ALT, AST and LDH (P<0.01), and the rhein group of 100 μmol•L⁻¹ concentration only increased the leakage rate of LDH (P<0.05). With the increase of rhein concentration, the protein content in liver slices decreased significantly (P<0.05) with a certain range of does. Emodin group of 400 μmol•L⁻¹ concentration significantly increased the leakage rate of ALT, GGT and LDH (P<0.01). Physcion-8-O-β-D-glucopyranoside group of 800 μmol•L⁻¹ concentration also significantly increased the leakage rate of ALT, AST and LDH (P<0.01 or P<0.05), but the group of 200 μmol•L⁻¹ concentration only significantly increased the LDH leakage (P<0.05). Along with the increase of the concentration of physcion-8-O-β-D-glucopyranoside, the leakage rate of ALT, AST and LDH showed a trend of increase, but the protein content in liver slices was in decline. Furthermore, MTT reduction ability of liver slices significantly decreased (P<0.01) in the physcion-8-O-β-D-glucopyranoside group of 800 μmol•L⁻¹ concentration. The results suggested that rhein, emodin and physcion-8-O-β-D-glucopyranoside at high concentrations (≥400 μmol•L⁻¹) can produce some damage to the liver tissue. However, the exposure levels of these constituents are very low, so to reach the toxic concentration (400 μmol•L⁻¹ or 800 μmol•L⁻¹) an adult of 65 kg body weight will need at least a single oral 4 898 g, 339 g and 5 581 g of P.multiflorum root respectively, which is far from the statutory dose of crude P. multiflorum root (3-6 g) or its processed product (6-12 g). Therefore, the conclusion that anthraquinones are the prime constituents of the hepatotoxicity of P. multiflorum root are still not be proved.
Protective Mechanism of Polygonum perfoliatum L. Extract on Chronic Alcoholic Liver Injury Based on UHPLC-QExactive Plus Mass Spectrometry Lipidomics and MALDI-TOF/TOF Mass Spectrometry Imaging

Abstract:Polygonum perfoliatum L. has a long history of medicinal and edible applications. Studies have shown that it can significantly protect liver injury, but the mechanism is unclear. The purpose of this study was to explore the protective mechanism of P. perfoliatum on chronic alcoholic liver injury from the perspective of lipid metabolism. After 8 weeks of alcohol exposure in male Wister mice, the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) in serum were significantly increased, and the activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) in liver were significantly decreased. Meanwhile, pathological changes of liver tissue in mice were observed by histopathology. Then, Ultra-High Performance Liquid Chromatography (UHPLC) QExactive Plus Mass Spectrometer lipidomics and matrix-assisted laser desorption/ionization time-of-flight/time -of-flight (MALDI-TOF/TOF) mass spectrometry imaging methods were established to analyze lipid metabolism in mice. Ten different lipids were identified by statistical analysis, including Fatty Acyls, Glycerophospholipids, Prenol lipids and Sphingomyelins. After intervention with P. perfoliatum extracts at different doses (25 to 100 mg/kg), levels of AST, ALT, ALP in serum, and activities of ADH and ALDH in liver were significantly corrected. The hepatic cord structure was clear, and the liver cells were closely arranged without other obvious abnormalities. Non-target lipidomics analysis showed that P. perfoliatum extract could regulate the metabolic disorders of the 10 different lipids caused by continuous alcohol exposure. Pathway analysis suggested that the mechanism of P. perfoliatum extract on chronic alcoholic liver injury may be related to the regulation of linoleic acid and α-linolenic acid.

Exploration of components and mechanisms of Polygoni Multiflori Radix-induced hepatotoxicity using siRNA -mediated CYP3A4 or UGT1A1 knockdown liver cells

Abstract:Ethnopharmacological relevance: Polygoni Multiflori Radix, the dried root of Polygonum multiflorum Thunb., and its processed products have been used as restoratives for centuries in China. However, the reports of Polygoni Multiflori Radix-induced liver injury (PMR-ILI) have received wide attention in recent years, and the components and mechanism of PMR-ILI are not completely clear yet. Our previous studies found that the PMR-ILI was related to the down-regulation of some drug metabolism enzymes (DME). Aim of the study: To explore the effect of the inhibition of CYP3A4 or UGT1A1 on PMR-ILI, screen the relevant hepatotoxic components and unveil its mechanism. Methods: RT-qPCR was used to detect the effects of water extract of Polygoni Multiflori Radix (PMR) and its main components on the mRNA expression of CYP3A4 and UGT1A1 in human hepatic parenchyma cell line L02. High-performance liquid chromatography (HPLC) was employed to detect the content of major components in the PMR. And then, the stable CYP3A4 or UGT1A1 knockdown cells were generated using short hairpin RNAs (shRNA) in L02 and HepaRG cells. Hepatotoxic components were identified by cell viability assay when PMR and its four representative components, 2,3,5,4'-tetrahydroxy stilbene glycoside (TSG), emodin (EM), emodin-8-O-β-D-glucoside (EG), and gallic acid (GA), acted on CYP3A4 or UGT1A1 knockdown cell lines. The PMR-ILI mechanism of oxidative stress injury and apoptosis in L02 and HepaRG cells were detected by flow cytometry. Finally, the network toxicology prediction analysis was employed to excavate the targets of its possible toxic components and the influence on the metabolic pathway. Results: PMR and EM significantly inhibited the mRNA expression of CYP3A4 and UGT1A1 in L02 cells, while TSG and GA activated the mRNA expression of CYP3A4 and UGT1A1, and EG activated CYP3A4 expression while inhibited UGT1A1 expression. The contents of TSG, EG, EM and GA were 34.93 mg/g, 1.39 mg/g, 0.43 mg/g and 0.44 mg/g, respectively. The CYP3A4 or UGT1A1 knockdown cells were successfully constructed in both L02 and HepaRG cells. Low expression of CYP3A4 or UGT1A1 increased PMR cytotoxicity remarkably. Same as PMR, the toxicity of EM and GA increased in shCYP3A4 and shUGT1A1 cells, which suggested EM and GA may be the main components of hepatotoxicity in PMR. Besides, EM not only inhibited the expression of metabolic enzymes but also reduced the cytotoxicity threshold. EM and GA affected the level of ROS, mitochondrial membrane potential, Ca2+ concentration, and dose-dependent induced hepatocyte apoptosis in L02 and HepaRG cells. The network toxicology analysis showed that PMR-ILI was related to drug metabolism-cytochrome P450, glutathione metabolism, and steroid hormone biosynthesis. Conclusion: The inhibition of mRNA expression of CYP3A4 or UGT1A1 enhanced hepatotoxicity of PMR. EM and GA, especially EM, may be the main hepatotoxic components in PMR. The mechanism of PMR, EM and GA induced hepatotoxicity was proved to be related to elevated levels of ROS, mitochondrial membrane potential, Ca2+ concentration, and induction of apoptosis in liver cells.

Abstract:Ethnopharmacological relevance: The liver damage associated with Polygonum multiflorum Thunb. (P. multiflorum) and its preparations have aroused widespread concern. Opinions on the toxicity mechanisms and targets of P. multiflorum vary, and the toxic components are even more controversial. However, based on the current research results, we believed that any single component in P. multiflorum could not directly lead to liver injury, but may be the synergistic effect of multiple components. In addition, the toxicity mechanism also involved multiple targets. Aim of the study: This study aimed to elucidate the mechanism and target of the hepatotoxicity of P. multiflorum. Materials and methods: In this study, the manifestations of liver injury triggered by P. multiflorum and the associated metabolic enzymes/transporters in the metabolic pathways of bilirubin and bile acid were investigated to elucidate the mechanism and target of the hepatotoxicity of P. multiflorum and related components. First, the hepatotoxicity and potential effect of P. multiflorum on both metabolic pathways were studied in rats administered P. multiflorum extracts (in 70% ethanol) for 42 days. Then, in vitro cultured hepatocyte spheroids were used to determine the hepatotoxicity of monomer components. Results: This revealed that P. multiflorum could simultaneously block bilirubin(BIL) and bile acid(BA) metabolism pathways, subsequently leading to liver damage. The targets and modes of action include reducing the activity of UGT1A1, the only metabolic enzyme of BIL, downregulating BIL and BA uptake transporters NTCP, OATP1B1, OATP1B3, efflux transporters MRP2, and BSEP, and upregulating efflux transporter MRP3. Furthermore, our data indicated that 2,3,5,4'-tetrahydroxystilbene-2-O-β-glucoside (TSG) and emodin-8-O-β-D-glucoside (EG) are the main toxic components in P. multiflorum. TSG accounts for 3.71% of the total content of P. multiflorum. In addition to markedly downregulating UGT1A1, TSG can upregulate OATP1B1/3 and promote the uptakes of bilirubin and bile acid, producing synergistic toxicity. EG accounts for 0.29% of the total content and demonstrates direct hepatotoxicity and extensive substrate overlap with bilirubin and bile acids. It can affect these two metabolic pathways simultaneously, promoting the accumulation of both bilirubin and bile acid for further toxic effects. Emodin is other major component, accounting for 0.01% of the total content, and its hepatotoxicity mechanisms include direct toxicity and inhibitory effects on bilirubin metabolizing enzymes. However, emodin is mainly distributed in the kidneys, so its hepatotoxicity risk is relatively low. Conclusion: The simultaneous blockade of bilirubin and bile acid metabolic pathways as the critical toxic mechanism of P. multiflorum-induced liver injury, and potential toxic components were TSG and EG.

Abstract:To observe the effect of processed Polygonum multiflorum on mRNA expression levels of five subtypes of CYP450 enzymes in rat liver. SD rats were randomly divided into the normal control group, processed P. multiflorum high dose and low dose groups (5.40 g•kg⁻¹ and 1.08 g•kg⁻¹). The rats in administration groups were continuously given with processed P. mutiflorum for 7 days by ig administration, and the rats in normal control group were given with the same volume of distilled water. After successive administration of 7 days, the serum biochemical indications were detected, and Real-time quantitative PCR technology was used to detect the mRNA expression levels of five subtypes of CYP450 enzymes in rat liver. Experimental results showed that AST was decreased significantly in both low and high dose groups. ALT was significantly decreased in low dose group and significantly increased in high dose group. The mRNA expression levels of five subtypes of CYP450 enzymes in rat liver were decreased in high dose and low dose groups in a dose-dependent manner. Especially the high dose processed P. multiflorum could significantly inhibit CYP1A2 and CYP2E1 mRNA expression levels in rats. The study showed that high dose P. multiflorum water extract had hepatotoxicity, and the degree of liver damage was increased with the increase of dose. It shall be noted that 5.40 g•kg⁻¹ water extract of P. multiflorum could significantly inhibit CYP1A2 and CYP2E1 mRNA expression levels in the liver of rats.
Protective effect of saponins extracted from Panax japonicus on ethanol-induced hepatic cells L-O2 injury

Abstract:OBJECTIVE: To study the protective effect of saponins from Panax japonicus against ethanol-induced hepatic cells L-O2 injury and their possible mechanisms. METHODS: Saponins from P. japonicus was purified by HPLC-evaporation light scattering detection (ELSD). The survival rate of hepatic cells was determined by MTT assay. The level of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in L-O2 cells were measured by spectrophotometer. RESULTS: Six major saponins from P. japonicus were identified by HPLC-ESI-MS: ginsenosides Rg 1, Re, Rf, F 3, Rg 2 and Rd. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 could promote normal cells L-O2 proliferation (P < 0.01), at the rate of 22.7%, 34.8% and 28.5%, respectively. Rd 0.16 g·L -1 and F 3 1.28 g·L -1 showed evident inhibition to normal cells L-O2 (P < 0.01), at the rate of 49.7% and 43.3%. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 also showed significantly protection against ethanol-induced hepatic cells L-O2 injury (P < 0.01). The rate of inhibition decreased to 23.3%, 26.9% and 26.6% compared with the ethanol-induced model control, the inhibition rate of which was 50.4%. Rd 0.16 g·L -1 and F 3 1.28 g·L -1 showed more evident toxicity to ethanol-induced hepatic cells L-O2. The inhibition rate was 83.2% and 64.8% (P < 0.01), respectively. Meanwhile, the content of MDA and the activity of SOD and GSH-Px in ethanol-induced L-O2 cells were (1.35 ± 0.05) mmol·L -1, (26.4 ± 2.8) kU·L -1 and (67.0 ± 4.3) kU·L -1, respectively. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 could decrease the level of intracellular MDA to (1.17 ± 0.04), (1.21 ± 0.03) and (1.21 ± 0.05) mmol·L -1 (P < 0.01), increase the activity of intracellular SOD and GSH-Px activity to 38.3 ± 4.8, 35.6 ± 4.4 and (39.0 ± 3.3) kU·L -1 (P < 0.01), and 86.3 ± 5.7, 78.4 ± 3.5 and (84.2 ± 4.4) kU·L -1 (P < 0.01), respectively. CONCLUSION: Rg 1, Re and Rf have evidently protective effect against ethanol-induced hepatic cells L-O2 injury. The possible mechanism may be related to the decline of MDA content and increase in antioxidant enzymes such as SOD and GSH-Px activities.

Abstract:Polygonum multiflorum Thunb. (PM) is a traditional Chinese medicine, commonly used to treat a variety of diseases. However, the hepatotoxicity associated with PM hampers its clinical application and development. In this study, we refined the zebrafish hepatotoxicity model with regard to the following endpoints: liver size, liver gray value, and the area of yolk sac. The levels of alanine aminotransferase, aspartate transaminase, albumin, and microRNAs-122 were evaluated to verify the model. Subsequently, this model was used to screen different extracts, components, and constituents of PM, including 70 % EtOH extracts of PM, four fractions from macroporous resin (components A, B, C, and D), and 19 compounds from component D. We found that emodin, chrysophanol, emodin-8-O-β-D-glucopyranoside, (cis)-emodin-emodin dianthrones, and (trans)-emodin-emodin dianthrones showed higher hepatotoxicity compared to other components in PM, whereas polyphenols showed lower hepatotoxicity. To the best of our knowledge, this study is the first to identify that dianthrones may account for the hepatotoxicity of PM. We believe that these findings will be helpful in regulating the hepatotoxicity of PM.

Abstract:The present study is to explore the optimal extraction parameters and liver protective effect of the polygonatum polysaccharides in vivo. The order of factor effects on polysaccharides production was found to be extraction time (min, A)>ratio of solvent to solid (C)>extraction temperature (°C, B)>extraction number (D). The results show that the effects of extraction time (min, A) and ratio of solvent to solid (C) were more significant than those of the other factors. Optimal extraction parameters were as followings: extraction time 120 min, extraction temperature 100 °C, ratio of solvent to solid 5, and extraction number 4. Polygonatum polysaccharides was administered orally at doses of 150, 300 and 450 mg/(kg day) to carbon tetrachloride (CCl(4))-treated rats. Results showed that administration of polygonatum polysaccharides could increase rats' final body weight, liver antioxidant enzymes activities (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and glutathione reductase (GR)), decrease serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) activities and liver malondialdehyde (MDA) level. The liver sections obtained from animals supplemented with polygonatum polysaccharides extract demonstrated reduced pathological damages, supporting that polygonatum polysaccharides extract could effectively decrease the toxicity of CCl(4). It can be concluded that polygonatum polysaccharides treatment may prevent CCl(4)-induced liver oxidative injury in experimental rats.

Abstract:Polygonum perfoliatum L. is an herbal medicine that has been extensively used in traditional Chinese medicine to treat various health conditions ranging from ancient internal to surgical and gynecological diseases. Numerous studies suggest that P. perfoliatum extract elicits significant anti-tumor, anti-inflammatory, anti-bacterial, and anti-viral effects. Nevertheless, the underlying mechanisms of its anti-liver cancer effects remain poorly understood. Our study suggests that P. perfoliatum stem extract (PPLA) has a favorable safety profile and exhibits a significant anti-liver cancer effect both in vitro and in vivo. We identified that PPLA activates the cGMP-PKG signaling pathway, and key regulatory genes including ADRA1B, PLCB2, PRKG2, CALML4, and GLO1 involved in this activation. Moreover, PPLA modulates the expression of genes responsible for the cell cycle. Additionally, we identified four constituents of PPLA, namely taxifolin, myricetin, eriodictyol, and pinocembrin, that plausibly act via the cGMP-PKG signaling pathway. Both in vitro and in vivo experiments confirmed that PPLA, along with its constituting compounds taxifolin, myricetin, and eriodictyol, exhibit potent anti-cancer activities and hold the promise of being developed into therapeutic agents.
