Abstract:Triterpenoids are crucial active ingredients of Ganoderma lucidum (G. lucidum) with various health benefits. Yet the low biosynthesis greatly restricts their industrial applications, novel metabolic engineering strategies are needed to further enhance Ganoderma triterpenoids production. Transcription factors play vital roles in the metabolic regulation of terpenoids, which are still scarce to study in G. lucidum. Herein, a transcription factor GlbHLH5 (GenBank No. ) potential for metabolic regulation of Ganoderma triterpenes was identified for the first time. MeJA could increase Ganoderma triterpenoids biosynthesis, and GlbHLH5 significantly responded to MeJA induction, suggesting GlbHLH5 is a new target for Ganoderma triterpenoids overproduction. The regulatory effect of the newly identified target was further validated by homologous gene overexpression and silence in G. lucidum. It's demonstrated that overexpression of GlbHLH5 significantly increased triterpenoids accumulation and the key enzyme genes transcription in the biosynthetic pathway, while silencing it displayed the opposite effect, indicating GlbHLH5 could positively regulate the triterpenoids biosynthesis by activating the synergistic expression of key enzyme genes in the biosynthetic pathway. Consequently, GlbHLH5 was identified as a positive regulator and novel metabolic target for Ganoderma triterpenoids biosynthesis, it sheds new lights on the regulatory effect regulation and synthetic biology of Ganoderma triterpenoids.
A New Strategy to Improve Ganoderma Polysaccharides Production by Symbiotic Fungi Elicitors Through Activating the Biosynthetic Pathway

Abstract:Ganoderma lucidum polysaccharides (GLP) attract growing attention due to their remarkable bioactivities, but the low content in raw materials remains a bottleneck severely restricting their application. We previously found a higher polysaccharides accumulation in Ganoderma lucidum cultured in continuous cropping soil, and soil symbiotic fungi are presumed as the key among many factors. Herein, 33 symbiotic fungi were isolated from the soil, and fungal elicitors were prepared to investigate their biotic eliciting effect on GLP biosynthesis. Most elicitors were found to significantly improve GLP production, among which the NO.16 molecularly identified as Penicillium citrinum, exhibited the optimum eliciting effect with GLP yield increasing by 3.4 times. Differences in the biosynthetic pathway genes expressions and the monosaccharide components of GLP were further analyzed. The transcriptions of the main genes of GLP biosynthetic pathway were up-regulated under PCE treatments, suggesting it improves GLP production by activating transcriptions of the biosynthetic pathway genes. Moreover, PCE eliciting significantly altered the monosaccharide compositions of GLP with Gal, Man, GalA, GlcA, and Fuc increasing by 8.17 %, 5.68 %, 5.41 %, 2.66 %, and 1.51 % respectively, but Glc decreased by 23.43 %, which may result in the activity change. It can serve as a new strategy to improve GLP production.
Roles of Α-1,3-glucosyltransferase in Growth and Polysaccharides Biosynthesis of Ganoderma Lucidum

Abstract:Ganoderma lucidum polysaccharides are valuable natural compounds possessing significant biological activity, with glycosyltransferases playing a crucial role in their biosynthesis. Although the function of (3-1,3-glucosyltransferase in polysaccharides production is well understood, the role of alpha-1,3-glucosyltransferase in edible fungi remains unclear. In this study, over-expression of the alpha-1,3-glucosyltransferase gene in G. lucidum ( glagt ) was found to suppress the growth, with the maximum biomass and mycelial growth rate decreasing by 21.78 % and 79.61 %, respectively, a behavior distinct from (3-1,3-glucosyltransferase. The fungal pellet diameter decreased by 38 % and the cell-wall thickness by 32.44 %, whereas intracellular and extracellular polysaccharides production increased by 27.58 % and 66.08 %, respectively. In the transcription level, overexpressing the glagt gene i) downregulated the citrate synthase and isocitrate dehydrogenase gene in the TCA cycle, disrupting energy metabolism and fungal growth; ii) upregulated key enzymes involved in UDP-glucose synthesis and glycosyltransferases ( gl24465 , gl24971, , and gl22535); ); and iii) universally increased the transcriptional level of glucosidases gl21451, , gl30087, , and gl24581 by 22 %-397 %, contributing to cell-wall thinning to facilitate polysaccharides export. Conversely, the glagt gene downregulation promoted G. lucidum growth and decreased polysaccharides production. The results elucidate the roles of GLAGT and are expected to inspire in-depth exploration of polysaccharides biosynthesis pathways.
Development of a 2A peptide-based multigene expression system and its application for enhanced production of ganoderic acids in Ganoderma lucidum

Abstract:Ganoderma has received much attention for its medicinal value, but the manipulation of multiple genes remains a challenge, hindering the genetic engineering of this species for the development of cell factories. Here, we first showed that the presence of an intron is necessary for the efficient expression of the endogenous cDNA of carboxin-resistant gene (cbx) in G. lucidum. Then, the self-cleaving function of 2 A peptide was investigated in G. lucidum by linking cbx cDNA to the codon-optimized hygromycin B-resistant gene (ophph) using the 2A-peptide sequence. The results showed that cbx cDNA and ophph can be successfully expressed in G. lucidum in a bicistronic manner from a single transcript. Moreover, the expression of both genes was not affected by the order within the 2 A cassette. In addition, simultaneous expression of cbx cDNA, ophph, and codon-optimized yellow fluorescent protein gene (opyfp) was conducted for the first time in G. lucidum using the 2 A peptide-based approach. The developed method was successfully applied to express both cDNA of the 3-hydroxy-3-methylglutaryl coenzyme A reductase (hmgr) and squalene epoxidase gene (se) for enhanced production of ganoderic acids (GAs) in G. lucidum. The engineered strain produced the maximum content of GA-Mk, GA-T, GA-S, and GA-Me were 26.56±3.53,39.58±3.75, 16.54±2.16, and 19.1±1.87 μg/100 mg dry weight, respectively. These values were 3.85-, 4.74-, 3.65-, and 3.23-fold higher than those produced by the control strain. The developed method will be useful for the manipulation of complex metabolic or regulatory pathways involving multiple genes in Ganoderma.

Abstract:Ganoderic acid (GA), a triterpenoid from the traditional Chinese medicinal higher fungus Ganoderma lucidum, possesses antitumor and other significant pharmacological activities. Owing to the notorious difficulty and immaturity in genetic manipulation of higher fungi as well as their slow growth, biosynthesis of GAs in a heterologous host is an attractive alternative for their efficient bioproduction. In this study, using Saccharomyces cerevisiae as a host, we did a systematic screening of cytochrome P450 monooxygenase (CYP450) gene candidates from G. lucidum, which may be responsible for the GA biosynthesis from lanosterol but have not been functionally characterized. As a result, overexpression of a CYP450 gene cyp5150l8 was firstly found to produce an antitumor GA, 3-hydroxy-lanosta-8, 24-dien-26 oic acid (HLDOA) in S. cerevisiae, as confirmed by HPLC, LC-MS and NMR. A final titer of 14.5 mg/L of HLDOA was obtained at 120 hr of the yeast fermentation. Furthermore, our in vitro enzymatic experiments indicate that CYP5150L8 catalyzes a three-step biotransformation of lanosterol at C-26 to synthesize HLDOA. To our knowledge, this is the first report on the heterologous biosynthesis of GAs. The results will be helpful to the GA biosynthetic pathway elucidation and to future optimization of heterologous cell factories for GA production.

Abstract:Ganoderma lucidum has been a well-known species of basidiomycetes for a long time, and has been widely applied in the fields of food and medicine. Based on the simulation results of model iZBM1060 in our previous research, the effect of L-phenylalanine on G. lucidum extracellular polysaccharides (EPSs) was investigated in this study. EPS production reached 0.91 g/L at 0.4 g/L L-phenylalanine after a 24 h culture, which was 62.5% higher than that of control (0.56 g/L). Transcriptome and genome analysis showed that L-phenylalanine deaminase and benzoate 4-hydroxylase (related to L-phenylalanine metabolism) were significantly up-regulated, while the cell wall mannoprotein gene was down-regulated. Transmission electronic microscopy (TEM) and atomic force microscopy results showed that the cell wall thickness decreased by 58.58%, and cell wall porosity increased in cells treated with L-phenylalanine, which probably contribute to the increasing EPS production. This study provides an efficient strategy for fungal polysaccharide production with high output and low cost.
Determination of compounds in mycelium of Ganoderma lucidum in submerged fermentation

Abstract:The annotation of whole genome from Ganoderma lucidum provides the molecular basis for the establishment of its genome-scale metabolic network.In order to obtain the detailed data of the metabolic and biomass components necessary for constructing metabolic network of G.lucidum,different biomass compounds were analyzed during submerged fermentation of G.lucidum in a 200-L fermentor.The highest production of endopolysaccharides (IPS) (2.57±0.08) g/L,exopolysaccharides (EPS) (0.55±0.01) g/L and biomass (8.45±0.23) g/L were obtained on day 6.Meanwhile,various compounds in mycelium of G.lucidum were determinated and showed as follows:total protein (34.02±2.34)%,crude fibre (8.33±0.10)%,total lipid (10.10±0.20)%,RNA (3.76±0.01)%,DNA (1.11±0.01)%,and ash (9.35±0.10)%.The quantitation of amino acids and fatty acids (FAs) were done by HPLC and GC-MS,respectively.Based on obtained data,the contents of Gln,Asn and eight nucleotides were predicted after combining with G.lucidum genome.Gln,Asn and eight nucleotides content were predicted and the results in this study will be beneficial to provide favorable data support for the subsequent construction,refining and analysis of global metabolic model based on systems bioinformatics,and for new strategies for metabolic control of bioactive substances in G.lucidum.

Abstract:Abstract Background Ganoderma lucidum , a well-known medicinal mushroom, has received wide attention as a promising cell factory for producing bioactive compounds. However, efficient expression of heterologous genes remains a major challenge in Ganoderma , hindering metabolic regulation research and molecular breeding of this species. Results We show that the presence of glyceraldehyde-3-phosphate dehydrogenase gene ( gpd ) intron 1 at the 5′ end of, the 3′ end of, or within the heterologous phosphinothricin-resistant gene ( bar ) is efficient for its expression in G. lucidum . The enhanced expression of bar is exhibited by the higher accumulation of mRNA and increased amounts of protein. Moreover, the insertion of the gpd intron 1 in the β-glucuronidase gene ( gus ) elevates its mRNA accumulation and enzyme activity, which facilitates the use of this reporter gene in Ganoderma . Conclusions This study has demonstrated the importance of the introduction of gpd intron 1 for the efficient expression of bar and gus in G. lucidum . The presence of the gpd intron 1 in heterologous genes increases levels of mRNA accumulation and protein expression in basidiomycete Ganoderma . The developed method may be utilized in upregulating the expression of other heterologous genes in Ganoderma .

Abstract:Ganoderma lucidum is a well-known and important medicinal mushroom, but its genetic modification has not been reported. We developed an efficient procedure for isolation and regeneration of protoplasts from G. lucidum . To construct a vector for high-level expression of heterologous genes in G. lucidum , the 1.4-kb regulatory region of the glyceraldehyde-3-phosphate dehydrogenase gene ( GPD ) was isolated from the genomic DNA of Lentinus edodes , and the GPD promoter was fused to the β-glucuronidase ( GUS ) and bialaphos resistance ( bar ) genes. Using the resulting construct, p301-bG1, an efficient transformation system based on electroporation was established for G. lucidum. GUS expression was observed among transformants conferring bialaphos resistance, indicating that the L. edodes GPD promoter can be used for expression of exogenous genes in G. lucidum . We also studied green fluorescent protein ( GFP ) as another reporter for transformation of G. lucidum . The L. edodes GPD promoter was fused respectively to the GFP and bar genes, and the resulting construct, p301-bg, was introduced into G. lucidum . Stable GFP expression in transformants was detectable by fluorescence microscopy, suggesting the suitability of GFP as a reporter system in transformation of this mushroom. This is the first report of an efficient transformation system for G. lucidum using different reporters, paving the way for genetic modification of this famous medicinal mushroom.
Enhanced production of polysaccharides and triterpenoids in Ganoderma lucidum fruit bodies on induction with signal transduction during the fruiting stage

Abstract:Ganoderma lucidum is a medicinal mushroom that has been widely used in East Asia for the treatment of various diseases. The pharmacological activity of this fungus is primarily attributable to the polysaccharides and triterpenoids. In this study, to obtain the fruit bodies with improved content of active constituents, we examined the effect of salicylic acid (SA) and calcium ion on the biosynthesis of polysaccharides and triterpenoids by spraying the chemicals during the fruiting. To explore the underlying mechanisms for the variation, the transcripts of related genes involved in the polysaccharide and triterpenoid biosynthesis were measured. Results showed that Ca2+ had no effect on production of polysaccharides and triterpenoids, whereas SA increased triterpenoid content by 23.32%, compared to the control, but it had little influence on polysaccharide production. Interestingly, the combined induction increased polysaccharide and triterpenoid content by 9.02% and 13.61%, respectively, compared to the control. Under Ca2+ induction, the transcript of ugp gene in the polysaccharide biosynthetic pathway up-regulated in all three stages (mycelium, primordium, and fruit body), while pgm and gls gave no response in the mycelium and primordium stages, and up-regulated in the fruit body stage. Differently, six key triterpenoid biosynthetic genes including hmgr, hmgs, mvd, fps, sqs, and ls did not respond to the induction. In the case of SA and combined induction, pgm and ugp were up-regulated in all three stages, while gls showed an increased expression in the primordium stage and no response in other stages. The six triterpenoid biosynthetic genes were up-regulated in all three stages. The present study provides a useful approach to producing G. lucidum fruit bodies with high polysaccharide and triterpenoid content. This is important to the G. lucidum industry.

Abstract:Basidiomycetes can produce various valuable secondary metabolites with bioactivities. However, the low titer of metabolites has greatly limited their application. Currently, shortage of mutant complementation techniques, especially in-situ complementation, impeded the elucidation of their metabolites biosynthesis pathways. In this work, Ganoderma lucidum, a traditional Chinese medicinal basidiomycete, which can produce various bioactive ganoderic acids (triterpenoids), was used; and a clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein-9 nuclease (Cas9) mediated in-situ complementation system for two functional genes was established. For a disruptant of gl26097, which encodes a Cys2-His2 (C2H2)-type zinc finger transcription factor involved in calmodulin-calcineurin signaling, one in-situ complemented strain of 8 transformants was obtained,concluded by sequencing confirmation. For the function of gl26097, both the cell growth and content of four identified individual ganoderic acids (GAs) were restored in the complemented strain compared to the gl26097 disruptant. For a disruptant of cyp5150l8, which is responsible for transforming lanosterol to a GA, one in-situ complemented strain from 8 transformants was obtained as confirmed by sequencing. The titer of four identified GAs increased significantly in the complemented strain compared to the cyp5150l8 disruptant. To the best of our knowledge, this is the first research report on applying CRISPR-Cas9 to complement mutants in-situ in basidiomycetes. The CRISPR-Cas9 assisted gene complementation technology will be helpful to study gene functions and GA biosynthesis pathway in G. lucidum, which may be also applied to other basidiomycetes.

Abstract:Ganoderma lucidum is an important medicinal fungus in Asian countries. Ganoderic acid (GA) is the major variety of bioactive and medicative components in G. lucidum . Biosynthesis of secondary metabolites is usually associated with cell differentiation and development. However, the mechanism underlying these phenomena remain unclear. Transcription factors play an essential regulatory role in the signal transduction pathway, owing to the fact that they represent the major link between signal transduction and expression of target genes. In the present study, we performed transcriptome and metabolome analyses to identify transcription factors involved in GA biosynthesis during development of G. lucidum . Transcriptome data revealed differentially expressed genes between mycelia and primordia, as well as between mycelia and the fruiting body. Results from gene ontology enrichment analysis and metabolome analyses suggested that GAs and flavonoids biosynthetic process significantly changed during fungal development. The analysis of predicted occurrences of DNA-binding domains revealed a set of 53 potential transcription factor families in G. lucidum . Notably, we found homeobox transcription factor and velvet family protein played important role in GA biosynthesis. Combined with previous studies, we provided a model diagram of transcription factors involved in GA biosynthesis during fruiting body formation. Collectively, these results are expected to enhance our understanding into the mechanisms underlying secondary metabolite biosynthesis and development in fungi.

Influence of different cultivation methods on ganoderic acid synthesis and solid-state fermentation condition optimization

Abstract:Compared shake culture,two-stage culture process by combining shake culture with static cultureand solid-state fermentationof Ganoderma lucidum,analyzed the influence of different cultivation methods on total ganoderic acid,ganoderic acid Mk,ganoderic acid S,ganoderic acid T and ganoderic acid Me synthesis.Corn was used as the main medium to produce ganoderic acids,Plackett-Burman(PB)design was applied to screen potential key factors associated with total ganoderic acid content.Then,the steepest ascent experiment,central composite design and response surface methodology were applied to optimize the significant factors.The final results were as follows:39.0 g solid substrate (moisture content 38.0％) shake flask,1.08％ silkworm chrysalis powder,0.86％ sucrose,0.08％tri-ammonium citrate,0.22％ KH2PO4,0.13％ MgSO4,0.02％ VB1,culture temperature 29.5 ℃,and fermentation time 10 days.Under this condition,the total ganoderic acids content was 24.93 mg every gram of the ganoderma lucidum mycelium and corn powder,and ganoderic acid Mk,ganoderic acid S and ganoderic acid content reached 0.86,0.69 and 3.69 mg/g,respestively.Production of three ganodericacids by Ganoderma lucidum solid-state fermentation has good application prospect.
Breeding a new Ganoderma lucidum strain with increased contents of individual ganoderic acids by mono–mono crossing of genetically modified monokaryons

Abstract:Ganoderic acids (GAs) are major functional components of Ganoderma lucidum . The study aimed to breed a new G. lucidum strain with increased contents of individual GAs. Two mating-compatible monokaryotic strains, G. 260125 and G. 260124, were successfully isolated from the dikaryotic G. lucidum CGMCC 5.0026 via protoplast formation and regeneration. The Vitreoscilla hemoglobin gene ( vgb ) and squalene synthase gene ( sqs ) were overexpressed in the monokaryotic G. 260124 and G. 260125 strain, respectively. Mating between the G. 260124 strain overexpressing vgb and the G. 260125 strain overexpressing sqs resulted in the formation of the new hybrid dikaryotic G. lucidum strain sqs-vgb. The maximum contents of ganoderic acid (GA)-T, GA-Me, and GA-P in the fruiting body of the mated sqs-vgb strain were 23.1, 15.3, and 39.8 μg/g dry weight (DW), respectively, 2.23-, 1.75-, and 2.69-fold greater than those in G. lucidum 5.0026. The squalene and lanosterol contents increased 2.35- and 1.75-fold, respectively, in the fruiting body of the mated sqs-vgb strain compared with those in the G. lucidum 5.0026. In addition, the maximum expression levels of the sqs and lanosterol synthase gene ( ls ) were increased 3.23- and 2.13-fold, respectively, in the mated sqs-vgb strain. In summary, we developed a new G. lucidum strain with higher contents of individual GAs in the fruiting body by integrating genetic engineering and mono–mono crossing.

Abstract:BackgroundGanoderic acids (GAs), derived from the medicinal mushroom Ganoderma lucidum, possess anticancer and other important pharmacological activities. To improve production of GAs, a homologous farnesyl diphosphate synthase (FPS) gene was overexpressed in G. lucidum. Moreover, the influence of FPS gene overexpression on GA production was investigated by developing the corresponding mathematical models.ResultsThe maximum levels of total GAs and individual GAs (GA-T, GA-S, and GA-Me) in the transgenic strain were 2.76 mg/100 mg dry weight (DW), 41 ± 2, 21 ± 5, and 28 ± 1 μg/100 mg DW, respectively, which were increased by 2.28-, 2.27-, 2.62-, and 2.80-folds compared with those in the control. Transcription levels of squalene synthase (SQS) and lanosterol synthase (LS) genes during GA biosynthesis were upregulated by 2.28- and 1.73-folds, respectively, in the transgenic G. lucidum. In addition, the developed unstructured models had a satisfactory fit for the process of GA production in submerged cultures of G. lucidum. Analysis of the kinetic process showed that FPS gene overexpression had a stronger positive impact on GA production compared with its influence on cell growth. Also, FPS gene overexpression led to a higher non-growth-associated-constant β (1.151) over the growth-associated-constant α (0.026) in the developed models.ConclusionsFPS gene overexpression is an effective strategy to improve the production of GAs in G. lucidum. The developed mathematical models are useful for developing a better GA production process in future large-scale bioreactors.

Abstract:Mitochondrial pyruvate carriers (MPCs), located in the inner membrane of mitochondria, are essential carriers for pyruvate to enter mitochondria. MPCs regulate a wide range of intracellular metabolic processes, such as glycolysis, the tricarboxylic acid cycle (TCA cycle), fatty acid metabolism, and amino acid metabolism. However, the metabolic regulation of MPCs in macrofungi is poorly studied. We studied the role of MPCs in Ganoderma lucidum ( Gl MPC) on ganoderic acid (GA) biosynthesis regulation in G. lucidum . In this study, we found that the mitochondrial/cytoplasmic ratio of pyruvate was downregulated about 75% in GlMPC1 - and GlMPC2 -silenced transformants compared with wild type (WT). In addition, the GA content was 17.72 mg/g and increased by approximately 50% in GlMPC1 - and GlMPC2 -silenced transformants compared with WT. By assaying the expression levels of three key enzymes and the enzyme activities of isocitrate dehydrogenase (IDH) and α-ketoglutarate dehydrogenase (α-KGDH) of the TCA cycle in GlMPC1 - and GlMPC2 -silenced transformants, it was found that the decrease in Gl MPCs activity did not significantly downregulate the TCA cycle rate, and the enzyme activity of IDH increased by 44% compared with WT. We then verified that fatty acid β-oxidation (FAO) supplements the TCA cycle by detecting the expression levels of key enzymes involved in FAO. The results showed that compared with WT, the GA content was 1.14 mg/g and reduced by approximately 40% in co-silenced transformants.

Abstract:Type II ganoderic acids (GAs) produced by the traditional medicinal mushroom Ganoderma are a group of triterpenoids with superior biological activities. However, challenges in the genetic manipulation of the native producer, low level of accumulation in the farmed mushroom, the vulnerabilities of the farming-based supply chain, and the elusive biosynthetic pathway have hindered the efficient production of type II GAs. Here, we assemble the genome of type II GAs accumulating G. lucidum accession, screen cytochrome P450 enzymes (CYPs) identified from G. lucidum in baker's yeast, identify key missing CYPs involved in type II GAs biosynthesis, and investigate the catalytic reaction sequence of a promiscuous CYP. Then, we engineer baker's yeast for bioproduciton of GA-Y ( 3 ) and GA-Jb ( 4 ) and achieve their production at higher level than those from the farmed mushroom. Our findings facilitate the further deconvolution of the complex GA biosynthetic network and the development of microbial cell factories for producing GAs at commercial scale.

Abstract:A hydroxymethylglutaryl-CoA synthase gene, designated as GlHMGS (GenBank accession No. JN391469) involved in ganoderic acid (GA) biosynthesis pathway was cloned from Ganoderma lucidum. The full-length cDNA of GlHMGS (GenBank accession No. JN391468) was found to contain an open reading frame of 1,413 bp encoding a polypeptide of 471 amino acid residues. The deduced amino acid sequence of GlHMGS shared high homology with other known hydroxymethylglutaryl-CoA synthase (HMGS) enzymes. In addition, functional complementation of GlHMGS in a mutant yeast strain YSC1021 lacking HMGS activity demonstrated that the cloned cDNA encodes a functional HMGS. A 1,561 bp promoter sequence was isolated and its putative regulatory elements and potential specific transcription factor binding sites were analyzed. GlHMGS expression profile analysis revealed that salicylic acid, abscisic acid and methyl jasmonate up-regulated GlHMGS transcript levels over the control. Further expression analysis revealed that the developmental stage and carbon source had significant effects on GlHMGS transcript levels. GlHMGS expression peaked on day 16 before decreasing with prolonged culture time. The highest mRNA level was observed when the carbon source was maltose. Overexpression of GlHMGS enhanced GA content in G. lucidum. This study provides useful information for further studying this gene and on its function in the ganoderic acid biosynthetic pathway in G. lucidum.

Abstract:Ganoderic acids (GAs), mainly from the traditional Chinese medicinal Ganoderma lucidum , possess antitumor, anti-metastasis and other significant pharmacological activities. Due to the difficulty to purify the GAs monomer with low yield and similar structure, the pharmacological activities of most of GA have not been clarified, which hindered their widespread application. In this study, we expressed our previously identified two CYPs, CYP5150L8 and CYP5139G1 in Saccharomyces cerevisiae to generate two plasmid-containing yeast strain Y2P, which can produce 3,28-dihydroxy-lanosta-8,24-dien-26-oic acid (DHLDOA) at concentration of 0.6 mg/L. To improve the production of DHLDOA, we transfer CYP partner, cytochrome P450 reductase, into the CYP5150 and CYP5139G1 co-expression Y2P strain to obtain 3 plasmids co-transformation strain Y3P, and used 3 antibiotics to avoid plasmid loss during cell cultivation. The DHLDOA production of S. cerevisiae Y3P reached to 2.9 and 11.1 mg/L without or with adding hygromycin, G418, and zeocin, respectively. In addition, it was found that DHLDOA was effective against non-small cell lung cancer but had low toxicity to normal cells. This study may facilitate the wide-spread application of GA-DHLDOA and further improvement of DHLDOA production by the yeast fermentation.

Abstract:The heme oxygenase gene has antioxidant and cytoprotective effects in organisms, but no related research has been conducted in Ganoderma lucidum. For the first time, we cloned the HMX1 gene in G. lucidum. The CDS is 1092 bp in length and encodes 363 amino acids. The HMX1 protein was prokaryotically expressed and purified, and the enzyme activity of the purified protein was measured. The value of Km was 0.699 μM, and Vm was 81.9 nmol BV h−1 nmol−1 protein. By constructing the silencing vector pAN7‐dual‐HMX1i, the transformants HMX1i1 and HMX1i2 were obtained. Compared with the wild‐type (WT), the average growth rate of HMX1i1 and HMX1i2 decreased by 31% and 23%, respectively, and the mycelium biomass decreased by 53% and 48%, respectively. Compared with the WT, the extracellular polysaccharide content of HMX1i1 and HMX1i2 increased by 59% and 51%, and the intracellular polysaccharide content increased by 24% and 22%, respectively. These results indicate that the HMX1 gene affects mycelial growth and polysaccharide synthesis in G. lucidum.
