Abstract:Cell adhesion factor (CAF) is a major external glycoprotein expressed on the surface of a variety of adherent cells, but it is absent from the plasma membrane of many,'-4 but not a11,5.6 transformed cells. We have previously demonstrated7 the requirement for a component in partially purified C A F during fibroblast attachment and spreading on collagen-coated dishes pretreated with 8 M urea. Physicochemical evidence indicated that the active factor was CAF. I n the present study, we have developed a reproducible isolation procedure for obtaining highly purified C A F from human plasma and have obtained direct evidence that C A F is responsible for mediating the attachment and spreading of fibroblasts on ureatreated collagen. Furthermore, inhibition of attachment by antisera specific for C A F has been used to provide additional support for these conclusions. Pretreatment of collagen-coated dishes with 8 M urea denatures the triple helical structure of collagen' and allows a high efficiency of CAF binding to the primary structure.' To more closely mimic the in vivo situation, assay of the CAF dependence of fibroblast attachment to undenatured collagen has been attempted following incorporation of C A F into the collagen gel both during and after the induction of gelation by the elevation of pH. Although poor cellular spreading on the native collagen substratum was demonstrated, cellular attachment, albeit slower, remained C A F dependent. There is evidence that cellular attachment and spreading on plastic tissue culture dishes involve at least two biochemically distinct processes. The attachment process necessitates divalent and intact sulfhydryl groups,"*" and cellular spreading requires intact microtubules and microfilaments. In the present study, these requirements were also demonstrated to be important on collagen-coated dishes. On the collagen substratum, cellular spreading, but not attachment, can be inhibited by amino-containing compounds7; both attachment and spreading require ATP. We have utilized pharmacologic agents to determine structural and metabolic requirements for normal and transformed cells during attachment and spreading on collagen in the presence of exogenously supplied CAF. In addition, normal and transformed cells were compared to ascertain whether previously described alterations in cytoskeletal structures following transformation affected CAFmediated attachment and spreading. Requirements for adhesion and spreading were studied as separate events, because it has already been established that cellular attachment can be measured independently of ~ p r e a d i n g . ~ I n our experiments, we were unable to demonstrate any qualitative difference between BHK and PyBHK cells in cellular adhesion or spreading within all experimental variations.
