Abstract:PURPOSE: To review the literature on the long-term health effects of exposure to diethylstilbestrol (DES) among women prescribed DES during pregnancy (DES mothers), among their children exposed inutero to the drug (DES sons and daughters) and among the progeny of these exposed sons and daughters (DES grandchildren).DATA SOURCES: English-language articles were identified through MEDLINE and CANCERLIT searches and through review of the bibliographies of identified articles.STUDY SELECTION: All human studies relevant to long-term health effects of exposure to DES were reviewed.DATA EXTRACTION: Descriptive data on existing DES cohorts were extracted from early publications. Risk estimates for health effects were extracted from published reports.DATA SYNTHESIS: An estimated 5 to 10 million Americans received DES during pregnancy or were exposed to the drug in utero. Exposure to DES has been associated with an increased risk for breast cancer in DES mothers (relative risk, < 2.0) and with a lifetime risk of clear-cell cervicovaginal cancer in DES daughters of 1/1000 to 1/10,000. The association between DES exposure and testicular cancer in DES sons remains controversial. Exposure to DES has also been linked to reproductive tract abnormalities in DES sons and daughters that consist of immune system disorders and psychosexual effects. No evidence for transgenerational effects currently exists. Recommendations for screening persons exposed to DES are reviewed.CONCLUSIONS: Further research is needed to define long-term health effects related to DES exposure. Such research would provide a basis for counseling persons exposed to DES and would further understanding of environmental and pharmacologic compounds similar to DES.

Abstract:Objective: Lead exposure is one of major public health problems. The aim of this study was to examine in vitro the effect of lead on lymphocyte intracellular Ca2+ ([Ca2+]i). Methods: Lymphocytes were separated from mice spleen and treated with three doses of PbCl2 (1, 10 and 100 mu mol/L), [Ca2+]i was measured in a Thermo Labsystems Fluoroskan Ascent after loading the cells with Fura2-AM probe. Results: The lymphocyte [Ca2+]i in 10 and 100 mu mol/L PbCl2 groups increased to the highest level after 10 min of lead exposure (P<0.05). After I h of lead exposure, however, lymphocyte [Ca2+]i levels were not statistically different from that in control group cells (P>0.05). Removal of Ca2+ from external solution did not significantly affect the PbCl2-induced lymphocyte [Ca2+]i, PbCl2 increased the [Ca2+]i under both normal Ca2+ and Ca2+-free conditions. With pretreatment of calmodulin (CaM) antagonist W7, lymphocyte [Ca2+]i levels were still high, but [Ca2+]i levels were not as high as those in the absence of W7. Conclusion: When the lymphocytes were exposed to PbCl2 ranging from 1 to 100 mu mol/L, the lymphocyte [Ca2+]i level was increased, but the increase appeared reversible CaM may play a role in the process of the effect of lead on the lymphocyte [Ca2+]i. Abstract:Diethylstilbestrol (DES) is used as a kind of animal feed additive and affects people’s health through the food chain. The purpose of this study is to detect the residue level of DES in 576 human urine samples directly. DES-BSA was used to immunize Balb/c mice. The monoclonal antibody was produced by hybridoma that was screened through cell fusion techniques. Finally, we developed the indirect competitive ELISA method to analyze 576 human urine samples from Zhejiang Province, China. The IC50 of this method was 3.33 ng/mL. The LOD and LOQ were 0.16 and 0.54 ng/mL. Linear range of the standard curve was from LOD to 12.50 ng/mL. There was no cross-reactivity with two kinds of estrogens and two structural analogs with DES. Five hundred seventy-six urine samples were analyzed by the indirect competitive ELISA method, and the detection rate was 98.78%. The mean concentration and geometric mean were 4.70 and 3.50 ng/mL. The indirect competitive ELISA method based on monoclonal antibody was sensitive and reliable for the detection of DES in human urine samples. The results warned us to pay more attention to human health and food safety.
Effects of diethylstilbestrol on the expression of thyroid stimulating hormone receptor in mice lymphoid nodes

Abstract:Objective To investigate the expression of thyroid stimulating hormone receptor (TSHR) in lymphoid nodes of BALB/c mice and in the mice treated with diethylstilbestrol ( DES) during the neonatal period, and to explore the possible sources of thyroid stimulating hormone ( TSH ) expressed in the lymphoid nodes and the possible mechanism of the effect of DES on the expression of TSH. Methods Neonatal BALB/c mice whose weight discrepancies were less than 5% were divided randomly into a control group and a DES group. A total of 120 mice (30 pairs of females and 30 pairs of males) were chosen. Within 24 hours after birth, the mice in the DES group were injected subcutaneously at the cervixbackside with DES at intervals of 24 hours for 5 times, and the mice in the control group were injected with germfree bean oil using the same method. Mice were killed on postnatal day 7, 14, 21, 35, 49 and 63.5 pairs of female mice and 5 pairs of male mice at each time point. Deep cervical lymph nodes were taken out, embedded in paraffin and sliced serially in a thickness of 4μm. The expression of TSHR in the lymphoid nodes was detected by immunohistochemistry technique. Results In both control and DES groups, TSHR positive cells were present in lymphoid nodes from postnatal day 7 to day 63. In female mice, the expression of TSHR reached peak on day 49 and in male mice on day 35. At each the same time point, the average absorbance (AA) and the integrate absorbance (I4) of TSHR expression were higher in the DES group than those in the control group. In female mice, statistic differences were showed in both AA and I4 between the two groups at all the time points, and in male mice statistic differences were showed in AA on day 14, 21, 35 and 49 and in 1A on day 14, 21 , 35, 49 and 63 between the two groups. Conclusion TSHR positive cells are present in all the lymphoid nodes of both female and male BALB/c mice at all the detected time points. This suggests that the lymphocytes in lymphoid nodes of BALB/c mice during the postnatal period may have the receptor foundation of combing TSH which comes from antehypophysis. Expossure to DES during the neonatal period may lead an increased TSHR expression in lymphoid nodes of BALB/c mice, particularly in female mice. The effect of DES on the expression of TSHR in lymphocytes of both female and male mice may last up to adulthood of the mouse at least.
The effect of diethylstilbestrol as measured by host resistance and tumor susceptibility assays in mice

Abstract:As part of a program to develop and validate methodology to measure chemically induced immunotoxicity, the effect of DES on resistance of adult B6C3F1 female mice to various microorganisms and to challenge with syngeneic tumor cells was evaluated. The mice received sc injections of 50 microliter corn oil alone or of corn oil containing the equivalent of 0.2, 1, and 4 mg DES/kg X d for 14 d. Three days later they were challenged with Listeria monocytogenes, Streptococcus sp. influenza virus, herpes virus, Trichinella spiralis, or B16-F10 tumor cells. Host resistance parameters were mortality for the bacterial and viral systems, expulsion of adult parasites from the gut for T. spiralis, and lung weights for the B16-F10 tumor-cell model. Host resistance to L. monocytogenes, herpes virus, and T. spiralis was significantly decreased following DES exposure. Resistance to Streptococcus sp. was decreased, but not at a statistically significant level following these doses of DES. However a dose of DES at 8 mg/kg X d resulted in a highly significant decrease in resistance to the organism. Resistance to influenza virus was unaffected by the DES. In contrast to the above, host resistance to iv-administered B16-F10 tumor cells was significantly increased as a consequence of DES exposure. These model systems for measuring alterations in host resistance have been indicated to hold potential for the routine screening of drugs, chemicals, and environmental agents for their possible immune effects, both adverse and potentiating. The results indicate the importance of selecting the appropriate assay for evaluating a particular agent. They also stress the necessity for including host resistance assays along with assays to measure specific immune aspects, in order to assess in the intact animal the overall effect of complex immune interactions following exposure to a test agent.

Abstract:The synthetic hormone diethylstilbestrol (DES) was widely prescribed between 1943 and 1971 to minimize pregnancy complications. It has caused serious physical and psychological damage to the women who took it and to their offspring. DES-exposed mothers may suffer a higher incidence of breast cancer, their exposed daughters are at risk for reproductive tract cancers and infertility, and their exposed sons are more likely to have genital abnormalities and reproductive dysfunction. Psychiatric disorders among DES-exposed persons are reportedly twice as common as for nonexposed persons, with anger, anxiety, low self-worth, identity confusion, and guilt the most frequent symptoms. The author describes therapeutic interventions designed to alleviate these problems.

Abstract:Placental growth and function are of biological significance in that placental tissue promotes prenatal life and the maintenance of pregnancy. Exposure to synthetic estrogens causes embryonic mortality and placental growth restriction in mice. The aim of the present study was to examine the effects of diethylstilbestrol (DES) on placenta in mice. DES at 1, 5, 10 or 15 µg kg(-1) day(-1) , or 17β-estradiol (E2 ) at 50 µg kg(-1) day(-1) , was administered orally to ICR mice on days 4 through to 8 of gestation. Expression of ERα, ERβ, ERRβ or ERRγ mRNA in the junctional or labyrinth zone of the placentas on day 13 was assessed using RT-PCR, as well as the embrynic mortality, embryonic and placental weight, histological changes of labyrinth and ultrastructural changes of the trophoblast giant cells (TGCs). Embryo mortalities in the DES 10 and 15 µg kg(-1) day(-1) groups were markedly increased. No significant changes in embryonic and placental weight were observed in any DES- or E2 -exposed groups. Expression of ERα mRNA in the junctional zone with male embryos in the 5 µg kg(-1) day(-1) group was significantly higher than that in the control, whereas expression was not determined in the 15 µg kg(-1) day(-1) group. Histological observation revealed that the placentas exposed to DES at 10 µg kg(-1) day(-1) lacked the developing labyrinth. Ultrastructural observation of the TGCs showed poor rough-surfaced endoplasmic reticulum in the DES 10 µg kg(-1) day(-1) group. The present data suggest that developmental changes induced by DES may be related to interference with the nutrition and oxygen exchange between mother and embryo or decreased protein synthesis, resulting in a high frequency of embryo mortality.

Abstract:[Objective]To investigate the effect of diethylstilbestrol administered externally to neonatal female BALB/c mice on the quantity and distribution of their splenic lymphocytes,and on the percentage of T cell subsets.[Methods]Neonatal brood BALB/c mice whose weight discrepancy was less than 5% were divided randomly into control groups and DES groups,and 25 pairs of female mice in all were chosen for use.Mice in DES groups were injected subcutaneously on nape with DES within 24 hours after birth at intervals of 24 hours for 5 times,and mice in control groups were injected with axenic bean oil with the same method.Mice were killed on the d 7,d 14,d 21,d 35 and d 49 after birth separately;the spleens were taken out,imbedded with paraffin and sliced serially in 5 μm.CD3 and CD20 were detected with immunohistochemical technique.Mononuclearcells were separated with Ficoll delamination fluid and CD4+,CD8+ and CD4+CD8+(DP)cells were detected with flow cytometry.[Results]In DES groups the average optical density(AOD)and integrated optical density(IOD)of both CD3+ cells and CD20+ cells on the d 7,d 14,d 21,d 35 and d 49 after birth were all significantly lower than in control groups.In DES groups,the percentage of CD4+ cells was significantly decreased on d 35 after birth and significantly increased on d 49 after birth;the percentage of DP cells was significantly increased on d 7 and d 35 after birth and significantly decreased on d 49 after birth;there were not marked changes were detected on the percentage of CD8+ cells.Distribution of CD3+ and CD20+ cells in spleens of DES groups was similar to that in control groups.[Conclusion]DES can significantly decrease the number of T and B cells and the expressional intensity of specific surface antigen on T and B cells in spleens of female BALB/c mice,significantly change the percentage of CD4+ cells and DP cells,but can not effect the distribution of T and B cells in spleen.
iTRAQ-based proteomics analysis of diethylstilbestrol induced dysfunction of testicular gubernaculum development in mouse

Abstract:BackgroundDiethylstilbestrol (DES) has been shown to disrupt the morphology and proliferation of gubernaculum testis cells.ObjectiveThis study aimed to elucidate the mechanism underlying DES induced damage to gubernaculum testis based on proteome analysis.ResultsNeonatal mice were exposed to DES or control vehicle and the gonads were collected for isobaric tags for relative and absolute quantitation based proteomics analysis. We found that at the early and late stages of gubernaculum development, treatment with different concentrations of DES upregulated the expression of proteins which were generally involved in several function categories including single-organism process, cellular process, binding, catalytic activity, cell part and organelle. KEGG pathway analysis showed that cardiac muscle contraction, oxidative phosphorylation, calcium signaling and cGMP-PKG signaling pathway were the most dysregulated pathways associated with DES in the late stage of gubernaculum development, while steroid biosynthesis was the only enriched pathway in the early stage of gubernaculum development.ConclusionProteomics profiling and functional analysis reveal new clues on how DES interferes with cellular processes and disrupts the development of gubernaculum. These findings have potential applications in the treatment of genitourinary diseases such as cryptorchidism.

Abstract:Objectives Di(2-ethylhexyl) phthalate (DEHP) is a common endocrine-disrupting chemical, which has potential reproductive toxicity. This study aimed to explore the effects of DEHP exposure in women with polycystic ovary syndrome (PCOS) undergoing in vitro fertilization. Methods In this case-control study, DEHP levels in follicular fluid (FF) of women with PCOS (n = 56) and controls (n = 51) were measured. The in vitro effects of DEHP exposure on primary-cultured human granulosa cells (GCs) and a steroidogenic human granulosa-like tumor cell line (KGN cells) were analyzed. Results Concentrations of DEHP in FF were significantly higher in women with PCOS than in controls. The clinical pregnancy rate was significantly lower in women with PCOS with high levels of DEHP than in controls. The levels of androgens produced by human GCs were significantly increased following DEHP exposure. Compared with controls, DEHP-treated human GCs and KGN cells showed significantly lower viability, cell cycle arrest, higher apoptosis, and altered expression of apoptosis-related genes. Conclusion Women with PCOS are exposed to increased levels of DEHP in follicles, which may be associated with pregnancy loss following in vitro fertilization. DEHP may disrupt steroid production, balance in cellular proliferation, and apoptosis in human granulosa cells.
Diethyl Sulfate Induced Cdk2-dependent Centrosome Amplification in CHL Cells.

Abstract:The function of centrosome that serves as the main microtubule organizing center is essential to ensure the genomic integrity during the cell division cycle. Centrosome abnormalities are frequently observed in many tumors and cells exposed to genotoxic agents. Here, we investigated the centrosome abnormalities induced by diethyl sulfate (DES) in Chinese hamster lung (CHL) fibroblasts and the underlying molecular mechanisms. Our results showed that DES exposure at 0.3, 1 and 3mM for 48 h caused centrosome amplification in a dose dependent manner. This effect was associated with transient S and G2/M phase delay and up-regulating of Cdk2, Cyclin A expressions. Furthermore, inhibition of Cdk2 activities reversed the centrosome amplification induced by DES. These results reveal that centrosome is one of the key subcellular targets of DES. Centrosome abnormalities might be important mechanisms behind the aneuploidy induction and carcinogenicity of DES.
Exposure to Di-(2-ethylhexyl) Phthalate Reduces Secretion of GDNF Via Interfering with Estrogen Pathway and Downregulating ERK/c-fos Signaling Pathway in Astrocytes.

Abstract:Di-(2-ethylhexyl) phthalate (DEHP) is a typical endocrine-disrupting chemical (EDC) that can increase the risk of central nervous system disease. This study aimed to investigate the in vitro and in vivo effects of DEHP exposure on GDNF secretion and the underlying mechanisms. Pregnant Wistar rats were randomly assigned into four groups and administered 0, 30, 300, or 750 mg/kg DEHP daily by oral gavage. In addition, primary astrocytes were exposed to mono-(2-ethylhexyl) phthalate (MEHP), the main metabolite of DEHP. Our results showed that DEHP exposure reduced GDNF levels and downregulated the ERK/c-fos signaling pathway in the cerebral cortex of male, but not female, offspring. Moreover, exogenous estrogen could overcome the decreased GDNF levels in astrocytes caused by MEHP exposure. MEHP also decreased p300 levels and downregulated the ERK/c-fos signaling pathway in primary astrocytes. Honokiol restored GDNF levels following MEHP exposure by activating the ERK/c-fos signaling pathway, while the inhibitor U0126 further reduced the GDNF levels. These results suggested that DEHP exposure could interfere with the normal effects of estrogen in the brain and downregulate the ERK/c-fos signaling pathway to decrease the GDNF secretion from astrocytes in the cerebral cortex.
Diethyl Sulfate-Induced Cell Cycle Arrest and Apoptosis in Human Bronchial Epithelial 16HBE Cells

Abstract:In this study, we investigated the effects of diethyl sulfate (DES) on cell proliferation, cell cycle progression and apoptosis in human bronchial epithelial 16HBE cells. Cells were treated with various doses of DES (0, 0.5, 1.0, 2.0, 4.0 or 8.0mM) for 12, 24 or 36h. Cell proliferation and apoptosis were determined by MTT assay and flow cytometer, respectively. The results showed that DES inhibited cell proliferation in a dose- and time-dependent manner, and induced significant apoptosis in 16HBE cells. Apoptosis related proteins measurement results revealed that DES-induced apoptosis was concurrent with the increasing of Bax and cleavage fragment caspase-3 and the decreasing of Bcl-2 and full length procaspase-3. When cells were incubated with 2.0mM of DES for several time intervals, S and G2/M phase accumulation was observed. Further analysis indicated that both DES-induced G1/S transition acceleration and S arrest resulted in S phase accumulation, and that DES-induced G2/M arrest resulted in G2/M phase accumulation. Western blotting results demonstrated that after DES treatment p-chk1 (Ser345) and p-chk2 (Thr68) levels decreased in G1 cells, and increased in S and G2/M cells. In addition, the increasing of chk1 and chk2 were also induced by DES treatment. With the increase in the dose of DES, p53 levels first increased (0.5-4.0mM) and then decreased (8.0mM). Down-regulation of p53 by RNA interference increased 4.0mM of DES-induced apoptosis but did not affect 2.0mM DES-induced cell cycle arrest. In conclusion, DES inhibits 16HBE cells proliferation in a dose- and time-dependent behavior. Within the sublethal dose, DES induces S and G2/M arrest through activating DNA damage checkpoints. Within the lethal dose, DES induces apoptosis through evoking apoptosis programs. p53 might play an important role in the transition between evoking cell cycle arrest/pro-survival and apoptosis programs upon DES exposure.
In utero exposure to diethylstilbestrol and blood DNA methylation in adult women: Results from a meta-analysis of two cohort studies

Abstract:Background: Prenatal exposure to diethylstilbestrol (DES) is associated with several adverse health outcomes. Animal studies have shown associations between prenatal DES exposure and DNA methylation. Objective: The aim of this study was to explore blood DNA methylation in women exposed and unexposed to DES in utero. Methods: Sixty women (40 exposed and 20 unexposed) in the National Cancer Institute's Combined DES Cohort Study and 199 women (99 exposed and 100 unexposed women) in the Sister Study Cohort were included in this analysis. Within each study, robust linear regression models were used to assess associations between DES exposure and blood DNA methylation. Study-specific associations were combined using fixed-effect meta-analysis with inverse variance weights. Our analysis focused on CpG sites located within nine candidate genes identified in animal models. We further explored whether in utero DES exposure was associated with age acceleration. Results: Blood DNA methylation levels at 10 CpG sites in six of the nine candidate genes were statistically significantly associated with prenatal DES exposure (P < 0.05) in this meta-analysis. Genes included EGF, EMB, EGFR, WNT11, FOS, and TGFB1, which are related to cell proliferation and differentiation. The most statistically significant CpG site was cg19830739 in gene EGF, and it was associated with lower methylation levels in women prenatally exposed to DES compared with those not exposed (P < 0.0001; false discovery rate<0.05). The association between prenatal DES exposure in utero and age acceleration was not statistically significant (P = 0.07 for meta-analyzed results). Conclusions: There are few opportunities to investigate the effects of prenatal DES exposure. These findings suggest that in utero DES exposure may be associated with differential blood DNA methylation levels, which could mediate the increased risk of several adverse health outcomes observed in exposed women. Our findings need further evaluation using larger data sets.
Clinical study of diethylstilbestrol in hormone refractory prostate cancer

Abstract:Objective To summarize the clinical outcome and adverse events of estrogen therapy for hormone refractory prostate cancer.Methods A total of 32 patients with hormone refractory prostate cancer received diethylstilbestrol (DES) 2 mg daily at our institute.The data of PSA (prostate-specific antigen)change, time to progression, overall survival rate, disease-specific survival rate and adverse events were collected and analyzed.Results The data of 29 patients were complete.The mean duration of DES dosing was 8.6 ±0.9 months.Among them, 8 (27.5% ) patients achieved a PSA response with a 50% decrement of PSA or more.Seven (24.1% ) patients had a stable level of PSA(50% ＜ PSA ＜ 125% ) while 14 of 29 (48.3% ) maintained a PSA progression with a 25% increment of PSA or more.The overall median time to progression was 4 ( 1 - 12) months.And the median time to progression was 6 (5 - 12) months in the PSA response group.The overall survival rate was 48.3% and disease-specific survival rate 55.2%.The main adverse events were gynecomastia ( 10/29, 34.5% ) and deep vein thrombosis ( 3/29, 10.3% ).Conclusion When used for the treatment of hormone refractory prostate cancer, dethylstilbestrol at a daily dose of 2 mg can achieve a PSA response in 27.5% patients and a PSA stability in 24.1% patients.And the median time to progression is 4 months.Estrogen is efficacious for the patients with hormone refractory prostate cancer.
Effects of combined medication of epimedium pubescens flavonoids and diethylstibestrol on uterus and bones in ovariectomized rats

Abstract:Objective:To observe the effects of combined medication of epimedium pubescens flavonoids (EF) and diethylstilbestrol (DES) on uterus and bones in ovariectomized (OVX) rats.Methods:Fifty female SD rats were divided into 5 groups:Sham-OVX group,OVX group,DES group,EF group and combined medication of EF and DES group (EF+DES group).After an experiment period of 12 weeks,the side-effects on uterus was analyzed by measuring the wet weight of uterus and the endometrium epithelial thickness(EET) and the changes of bone mass with quantitative bone histomorphometry were observed.Results:The percent of bone trabecala area (%Tb.Ar),uterine wet weight and EET in DES group were significantly increased as compared with OVX group.EF+DES group showed increase in%Tb.Ar and decrease in uterine weight and EET,as compared with DES group.There was no significant difference between EF+DES group and sham-OVX group.Conclusion:EF could inhibit the DES's stimulative side effect on uterus and increase its anti-osteoporosis effects.

Abstract:Environmental estrogen is a substance that functions as an endocrine hormone in organisms and can cause endocrine system disruption. A typical environmental estrogen, diethylstilbestrol (DES), can affect normal sexual function and organism development. However, even though the effects of different exposure stages of DES on the endocrine system and gonadal development of zebrafish juveniles are unknown, sex determination is strongly influenced by endocrine-disrupting chemicals (EDCs). From 10-90 days post fertilization (dpf), juvenile zebrafish were exposed to DES (100 and 1000 ng/L) in three different stages (initial development stage (IDS), 10-25 dpf; gonadal differentiation stage (GDS), 25-45 dpf and gonadal maturity stage (GMS), 45-60 dpf). Compared with that of IDS and GMS, the growth indicators (body length, body weight, and others) decreased significantly at GDS, and the proportion of zebrafish females exposed to 100 ng/L DES was significantly higher (by 59.65%) than that of the control; in addition, the zebrafish were biased towards female differentiation. The GDS is a critical period for sex differentiation. Our results show that exposure to environmental estrogen during the critical gonadal differentiation period not only affects the development of zebrafish, but also affects the population development.

Abstract:Background/Aims . Diabetic kidney disease (DKD) is a leading cause of end-stage renal disease. Calcium dobesilate (CaD) is widely used to treat diabetic retinopathy. Recent studies have demonstrated that CaD exerts protective effects against diabetic nephropathy. The aim of this study was to elucidate the molecular and cellular mechanisms underlying the protective effects of CaD. Methods . Human umbilical vein endothelial cells (HUVECs) were cultured with different D-glucose concentrations to determine the effects of high glucose on HUVEC gene expression. HUVECs were also incubated with CaD (25 μ M, 50 μ M, and 100 μ M) for 3 days to determine the effects of CaD on HUVEC viability. db/db mice were treated with CaD. 2-[(Aminocarbonyl)amino]-5-(4-fluorophenyl)-3-thiophenecarboxamide (TPCA-1) blocked the nuclear factor- κ B (NF- κ B) pathway in HUVECs. A pentraxin 3 (PTX3) small interfering RNA (siRNA) intervention experiment was performed in the cells. An adenovirus-encapsulated PTX3 siRNA intervention experiment was performed in db/db mice. Western blot and real-time PCR analyses were used to detect PTX3, p-IKBa/IKBa (I-kappa-B-alpha), and p-eNOS/eNOS (endothelial nitric oxide synthase) expression in mice and HUVECs. Hematoxylin-eosin (HE) staining and periodic acid-Schiff (PAS) staining were used to observe renal tissue damage in mice. PTX3 expression was observed by immunohistochemical staining. Results . CaD downregulated the expression of PTX3 and p-IKBa/IKBa and upregulated the expression of p-eNOS/eNOS in vitro. When TPCA-1 was used, high glucose induced high PTX3 expression, and the expression of p-eNOS/eNOS increased. After PTX3 gene silencing, the expression of p-eNOS/eNOS also increased. In vivo, CaD reduced the expression of PTX3 and p-IKBa/IKBa in the kidneys of db/db mice and increased the expression of p-eNOS/eNOS. After PTX3 gene silencing, the urine protein and renal function of db/db mice were ameliorated, the glomerular extracellular matrix was decreased, and the expression of p-eNOS/eNOS was increased. Conclusions . Our results suggested that CaD may inhibit the expression of PTX3 by altering the IKK/IKB/NF- κ B pathway, thereby improving endothelial dysfunction in HUVECs. PTX3 may be a potential therapeutic target for DKD.

Abstract:The idea that susceptibility to breast cancer is determined not only through inherited germline mutations but also by epigenetic changes induced by alterations in hormonal environment during fetal development is gaining increasing support. Using findings obtained in human and animal studies, this review addresses the mechanisms that may explain why daughters of mothers who took synthetic estrogen diethylstilbestrol (DES) during pregnancy have two times higher breast cancer risk than women who were not exposed to it. The mechanisms likely involve epigenetic alterations, such as increased DNA methylation and modifications in histones and microRNA expression. Further, these alterations may target genes that regulate stem cells and prevent differentiation of their daughter cells. Recent findings in a preclinical model suggest that not only are women exposed to DES in utero at an increased risk of developing breast cancer, but this risk may extend to their daughters and granddaughters as well. It is critical, therefore, to determine if the increased risk is driven by epigenetic alterations in genes that increase susceptibility to breast cancer and if these alterations are reversible.

Abstract:Millions of women and their fetuses were exposed to the toxic pregnancy drug diethylstilbestrol (DES) from the 1940s into the 1970s, a time when the medical profession had little knowledge about potential developmental consequences of fetal drug exposures. Pathological consequences of DES exposure to the pregnant mothers and their offspring are well documented, but now generational research is finding that the grandchildren of women given DES in pregnancy are also at risk. This commentary summarizes presentations on this subject from the Beyond Genes panel "Heritable Impacts of Diethylstilbestrol (DES)."
